Dermatan sulfate and chondroitin 6-sulfate conformations   总被引:2,自引:0,他引:2  
X-ray diffraction patterns show that dermatan sulfate in oriented, crystalline films occurs as two or three or eight-fold helices. The two-fold helix has a greater axial rise per disaccharide residue [h = 9.6A?] than the corresponding chondroitin 6-sulfate helix [h = 9.3A?]. Three-fold dermatan sulfate and chondroitin 6-sulfate helices both have h = 9.5A?. Consequently the α-L-iduronate residues in dermatan sulfate helices have the C1 chair conformation like β-D-glucuronate in chondroitin 6-sulfate. Since the eight-fold dermatan sulfate helix has h = 9.3A? rather less than the eight-fold chondroitin 6-sulfate helix [h = 9.8A?] the possibility of α-L-iduronate 1C chairs cannot be ruled out for it. Computer methods have been used to produce molecular models. In these the polysaccharide chains are almost linear. Each backbone conformation can accommodate a variety of arrangements of charged side groups.  相似文献

A sulfatase, which liberates sulfate from UDP-N-acetylgalactosamine-6-sulfate (the nucleotide occurring in quail egg white at high concentration), has been isolated from quail oviduct. The tissue also contained sulfatase activities for UDP-N-acetylgalactosamine-4-sulfate and nitrocatechol sulfate but these activities were removed from the 6-sulfatase fraction during purification. The UDP-N-acetylgalactosamine-6-sulfate sulfatase appears to be very closely related to a sulfatase activity for the non-reducing N-acetylgalactosamine-6-sulfate end group in chondroitin sulfate, i.e. the two activities could not be separated from each other by various fractionation procedures and were affected in a parallel fashion by mild heating. The results, coupled with those of earlier studies on UDP-N-acetylgalactosamine-4-sulfate in hen oviduct, suggest that in avian oviducts a sulfation/desulfation system may exist wherein sulfated sugar nucleotides and sulfated glycosaminoglycans are involved as alternative or competitive substrates.  相似文献

Cartilage regeneration in the adult rabbit ear was examined with respect to glycosaminoglycan (GAG) synthesis at various stages of the regeneration process. Increased hyaluronic acid and chondroitin sulfate synthesis was first seen 31 days after wounding, when a metachromatic cartilage matrix could be distinguished from blastemal cells. Analysis of cartilage and the overlying skin separately showed that 90% of the labeled chondroitin sulfate was found in the cartilage being regenerated. DEAE-cellulose chromatography of GAG preparations from 35-day regenerating cartilages showed hyaluronic acid and chondroitin sulfate peaks eluting in the same position as those isolated from normal cartilages. The identity of the hyaluronic acid and chondroitin sulfate peaks was confirmed by their susceptibility to Streptomyces hyaluronidase and chondroitinase ABC, respectively. Although the degree of sulfation in normal and regenerated cartilages was similar, the ratio of chondroitin 6-sulfate to chondroitin 4-sulfate was increased in regenerated cartilages. GAG preparations from unlabeled cartilages were digested with chondroitinase ABC and the disaccharide digestive products were identified and quantitiated. Normal cartilage had a ΔDi-6SΔDi-4S ratio of 0.27; the same ratio for the regenerated cartilage was 1.58.  相似文献

Four patients with an unusual form of spondyloepiphyseal dysplasia excreted in the urine undersulfated chondroitin 6-sulfate (Biochem. Med. 7, 415–423, 1973). The sera of these patients show a low activity of PAPS — chondroitin sulfate sulfotransferase, while the undersulfated chondroitin sulfate present in their urine is a much better acceptor of 35SO4 than standard chondroitin sulfate when they are incubated with [35S]PAPS and normal sulfotransferases. These results suggest that in these patients the skeletal lesions are secondary to a defect in the synthesis of chondroitin sulfate involving specifically the sulfotransferase activity.  相似文献

The characterization of sulfated glycosaminoglycans (GAGs) in hematophagous arthropod vectors in general has been limited, with the exception of the studies in the triatomine Rhodnius prolixus. Heparan sulfate (HS) and chondroitin sulfate (CS) were previously identified and structurally characterized in extracts of whole bodies of fourth instar larvae of R. prolixus. Recently, we showed the expression of these two sulfated GAGs in specific body tissues of adult males and females and in embryos of R. prolixus. In the present work, we identified and compared the sulfated GAG composition in specific tissues of adult insects and in embryos of another triatomine species, Triatoma brasiliensis. Sulfated GAGs were isolated from the fat body, intestinal tract, and the reproductive tracts of adult insects and from embryos. Only HS and CS were found in the tissues analyzed. The present results extend the initial observations on the sulfated GAG composition in R. prolixus by showing that these molecules are widely distributed among internal organs of triatomines. These observations may be useful for future investigations aiming to evaluate the possible implication of these compounds in physiological events that take place in a specific organ(s) in these insects.  相似文献

[2H]Chondroitin sulfate was prepared by partial N-deacetylation of chondroitin sulfate (via hydrazinolysis) followed by treatment with [2H6]acetic anhydride. 2H NMR spectra of [2H]chondroitin sulfate in the presence of human plasma low density lipoprotein provide evidence for a soluble complex stoichiometry of 3 (and possibly 2) lipoproteins per polysaccharide molecule, and allow a rough estimation of the dissociation constant Kd.  相似文献

In order to study further the metabolism of neutral steroids in human fetal adrenal and liver tissue the fractions of unconjugated neutral steroids isolated from these tissues were analyzed by gas-liquid chromatography and gas chromatography — mass spectrometry. In the adrenals, pregnenolone and 17-hydroxypregnenolone, but no corticoids, were detected. In the liver, pregnenolone, 3α-hydroxy-5β-pregnan-20-one, 5β-pregnane-3α, 20α-diol and 3β, 16α-dihydroxy-5β-pregnan-20-one were found. Thus, all the free steroids detected were C21 compounds. From these results and those obtained earlier by the analysis of the sulfate-conjugated steroids present in these tissues it is concluded that in the fetal adrenals in situ both sulfated and unconjugated steroids are actively metabolized. Regarding the liver it is obvious that the conjugated metabolites of progesterone are rapidly eliminated from this tissue. Here, pregnenolone is present both in the free and sulfate conjugated form, whereas its metabolites are found only as sulfate conjugates.  相似文献

The β-lactamase (penicillinase) from Escherichia coli W3310 has been crystallized from 25% saturated ammonium sulfate solution at pH 6.9. An X-ray examination of the monoclinic crystals shows the space group is C2, with unit cell dimensions a = 47.2 A?, b = 76.9 A?, c = 73.3 A? and β = 98.6°. With Z = 4 and the molecular weight = 22,000 (Melling &; Scott, 1972), the Å3/dalton ratio is 2.98. The crystals are suitable for structure analysis to at least 2.4 Å resolution.  相似文献

On the basis of spectroscopic evidence, opening of a five-membered cyclic carbonate ring (1,3-dioxolan-2-one) in the C15-subunit of the previously determined partial structure 1 (Fig. 1) of the major neocarzinostatin chromophore (NCS-Chrom A), is proposed to account for its base-catalyzed methanolysis to NCS-Chrom C. NCS-Chrom B, apparently an authentic natural product present as a minor component in all preparations of NCS studied, was found to be formally equivalent to the hydrolysis/decarboxylation product of the cyclic carbonate functionality in NCS-Chrom A. The mercaptan-dependent DNA strand-scission activity, equivalent for NCS-Chrom A, B and C, is independent of the integrity of the cyclic carbonate ring system and implicates a secondary site in the C15-substructure for mercaptan activation.  相似文献

Spectroscopic evidence suggests the presence of a highly strained ether ring (Fig. 1) (possibly an epoxide) in the C12-subunit of the previously determined partial structure 2a (Fig. 2) of the major neocarzinostatin chromophore (NCS-Chrom A) which completes assignment of all the oxygens in the molecule. The main product from mercaptan treatment suggests opening of the ether ring involving the addition of one molecule of mercaptan as well as reduction of the C12-substructure, whereas a parallel two-step reduction occurs on NaBH4 treatment. Both reactions occur with rearrangement of the C12-substructure and the implication for the mechanism of action of NCS-Chrom A in DNA strand scission activity is discussed. The evidence suggests a downward revision of the molecular formula for NCS-Chrom A as well as minor components B and C by two protons.  相似文献

Activity levels of sulfotransferases, requisite for the sulfation of chondroitin sulfate proteoglycan, were measured in cell-free homogenates prepared from neonatal epiphyseal cartilage of normal C57B1/6J or homozygous brachymorphic mice. In the presence of [35S]-PAPS only or [35S]-PAPS plus an exogenous sulfate acceptor, comparable amounts of 35SO42? were incorporated into chondroitin sulfate by the normal and mutant types of cartilage. In contrast, the mutant cartilage catalyzed the conversion of only 30% of the 35SO42? into chondroitin sulfate as compared to normal mouse cartilage when synthesis was initiated from ATP and H235SO4. These results suggest that the production of an undersulfated proteoglycan which has previously been reported in brachymorphic mice (Orkin, R.W. etal. (1976) Devel. Biol. 50, 82–94) may result from a defect in the synthesis of the sulfate donor PAPS.  相似文献

Bacteriophage G4 has the same AB and DE overlapping gene systems as φX174 and together with the A and CK overlapping gene system (Shaw et al., 1978), 7 of the 11 G4 and φX174 genes are involved in overlaps. The nucleotide differences between G4 and φX174 in the overlapping portions of the A, C and D genes are 23%, 27% and 21%, respectively, compared with 32%, 36% and 34% in the non-overlapping portions of the same genes. The amino acid differences between the G4 and φX174 overlapping B, K and E proteins, are 44%, 39% and 44%, respectively, compared with 28%, 26% and 16% in the regions of genes A, A and C, and D which contain genes B, K and E. These results suggest that the nucleotide sequences of overlapping genes evolve at almost the same rate as in non-overlapping genes, and that this is made possible by a lower amino acid sequence stringency of one of the pairs of proteins. The overlapping DE and A and CK gene systems may have originated by taking advantage of a high incidence of T nucleotides in the second codon position to produce a hydrophobic protein and the AB gene system may have evolved by read-through of the A gene into the B gene. From the nucleotide sequences, other overlapping genes appear to be possible in these bacteriophages.  相似文献

An unstable epoxide, leukotriene A4 (5(S)-trans-5,6-oxido-7,9-trans-11,14-cis-eicosatetraenoic acid), was earlier proposed to be an intermediate in the conversion of arachidonic acid into the slow reacting substance (SRS), leukotriene C4. In the present work synthetic leukotriene A4 was incubated with human leukocytes or murine mastocytoma cells. A lipoxygenase inhibitor, BW755C, was added in order to prevent leukotriene formation from endogenous substrate. Leukotriene C4 and 11-trans-leukotriene C4 were the main products with SRS activity. It was not established whether the 11-trans-compound was formed by isomerization at the leukotriene A4 or C4 stage.  相似文献

The distribution of sulfated mucopolysaccharides in different tissues during growth and in cancer tissues is reported. It is shown that most of the tissues of 1 day-old rats and rabbits contain chondroitin sulfate A/C, chondroitin sulfate B and heparan sulfate in about the same proportions, whereas in adult animals chondroitin sulfate A/C decreases in concentration or disappears. Changes in the relative proportions of chondroitin sulfate B and heparan sulfate were also observed in most of the tissues. In rats, these changes occur in the first 25 days of extrauterine development. A great increase of chondroitin sulfate A/C was observed in human tumors of different origins when compared with the normal adjacent tissues. Changes in the relative proportions of chondroitin sulfate B and heparan sulfate were also observed in most of the tumors analysed. The possible role of chondroitin sulfate A/C in cell division is discussed in view of the present findings.  相似文献

It is shown that each tissue has a characteristic composition differing from each other regarding the relative amount, type and molecular size of chondroitin sulfate A/C, chondroitin sulfate B and heparan sulfate. It is also shown that the same tissue from different mammals has the same types and proportions of sulfated mucopolysaccharides, but with different molecular weights. Exception to this rule was observed for the distribution of heparin which was present only in a few tissues of the five mammals studied. The possible involvement of the sulfated mucopolysaccharides in cell recognition and/or adhesiveness is discussed in view of this characteristic distribution.  相似文献

The temperature-dependent relationship between K+ active influx, Mg2+-ATPase activity, transmembrane potential (ΔΨ) and the membrane lipid composition has been investigated in mycoplasma PG3. Native organisms were grown in a medium containing 10 μg/ml cholesterol and either oleic plus palmitic (chol (+), O + P) or elaidic (chol (+), E) acids. Adapted cells were grown in a medium free of exogenous cholesterol and supplemented with elaidic acid (chol (?), E).Arrhenius plots of 42K+ active influx gave a linear relationship for (chol (+), O + P) cells (EA = ?9 kcal). On the other hand, when oleic plus palmitic acids are replaced by elaidic acid, an upward discontinuity appears between 28 and 30°C, which is associated with a large increase in the apparent activation energy of the process (t > 30°C, EA = ?24 kcal; t < 30°C, EA = ?40 kcal).Finally, a biphasic response with a break at approx. 23°C (EA = ?7 kcal, t > 23°C; EA = ?44 kcal, t < 23°C) is observed for (chol (?), E) organisms. From the lack of correspondence between these effects on the K+ influx and the temperature dependence of both the Mg2+-ATPase activity and ΔΨ, it is suggested that changes in the membrane lipid composition affect the K+ transport at the level of the K+ carrier itself.Differential scanning calorimetry, steady-state fluorescence polarization of diphenylhexatriene and freeze-fracture electron microscopy experiments further suggest that the effect is largely due to modifications of the membrane microviscosity and that the K+ carrier is associated with the most fluid lipid species present in the membrane.  相似文献

Wheat germ agglutinin crystallizes in two monoclinic space groups, P21 and C2, under identical crystallization conditions. Unit cell dimensions are a = 73.8 A?, b = 51.2 A?, c = 90.8 A?, γ = 90 ° for P21; a = 51.31 A?, b = 73.35 A?, c = 91.45 A?, β = 97.75 ° for C2, both with eight subunit molecules in the unit cell. The C2 crystals were chosen as suitable for investigating the three-dimensional structure to high resolution, because of their smaller asymmetric unit (containing the dimer), and also because they display better diffraction patterns.  相似文献

The expression and core protein structure of two proteoglycans, the major cartilage proteoglycan isolated from a rat chondrosarcoma and a small molecular weight chondroitin sulfate proteoglycan isolated from a rat yolk sac tumor, have been compared. The cartilage proteoglycan was not detectable in the cartilage tissue of cartilage matrix deficient (cmdcmd) neonatal mice by immunofluorescence, but the cmd cartilage did react with antibodies against the core protein of the yolk sac tumor proteoglycan. Radioimmunoassays showed that the core proteins of these proteoglycans are not cross-reactive with each other. Analysis of the core proteins by sodium dodecyl sulfate/polyacrylamide gel electrophoresis after chondroitinase ABC treatment of the proteoglycan revealed a large difference in their sizes. The cartilage proteoglycan core protein had a molecular weight of about 200,000 while the yolk sac tumor proteoglycan core protein migrated with an apparent molecular weight of about 20,000. In addition, the cultured yolk sac tumor cells that make the small proteoglycan did not react with antiserum against the cartilage proteoglycan. These results indicate that the proteoglycan isolated from the yolk sac tumor is similar to the small chondroitin sulfate proteoglycan species found in cartilage and support the existence of at least two dissimilar and genetically independent chondroitin sulfate proteoglycan core proteins.  相似文献

The first chemical characterization of two polypeptides from human serum which stimulate the in vitro incorporation of 35S sulfate into chick cartilage is described. These two polypeptides, designated Somatomedin A1 and A2 have a molecular weight of approximately 7000. Although each peptide contains 1 cysteine residue and has asparagine as amino terminal residue, there are apparent differences in the amino acid composition. Administration of a Somatomedin A concentrate to hypophysectomized rats gave an increase in tibial width similar to that obtained with 20 μg human growth hormone.  相似文献
