This study aimed to investigate whether fatigue-induced changes in synergistic muscle forces match their tendon elongation. The medial gastrocnemius muscle (MG) was fatigued by repeated electrical stimulation (1 min×5 times: interval 30 s, intensity: 20–30% of maximal voluntary plantar flexion torque) applied at the muscle belly under a partial occlusion of blood vessels. Before and after the MG fatigue task, ramp isometric contractions were performed voluntarily, during which tendon elongations were determined by ultrasonography, along with recordings of the surface EMG activities of MG, the soleus (SOL) and the lateral gastrocnemius (LG) muscles. The tendon elongation of MG and SOL in post-fatigue ramp was similar, although evoked MG forces dropped nearly to zero. In addition, for a given torque output, the tendon elongation of SOL significantly decreased while that of LG did not, although the activation levels of both muscles had increased. Results suggest that the fatigue-induced changes in force of the triceps surae muscles do not match their tendon elongation. These results imply that the tendons of the triceps surae muscles are mechanically coupled even after selective fatigue of a single muscle. 相似文献

While the impact of climate fluctuations on the demographic histories of species caused by changes in habitat availability is well studied, populations of species from systems without geographic isolation have received comparatively little attention. Vertebrate muscle Z-bands show zig-zag densities due to different sets of alpha-actinin cross-links between anti-parallel actin molecules. Their axial extent varies with muscle and fibre type: approximately 50 nm in fast and approximately 100 nm in cardiac and slow muscles, corresponding to the number of alpha-actinin cross-links present. Fish white (fast) muscle Z-bands have two sets of alpha-actinin links, mammalian slow muscle Z-bands have six. The modular structure of the approximately 3 MDa protein titin that spans from M-band to Z-band correlates with the axial structure of the sarcomere; it may form the template for myofibril assembly. The Z-band-located amino-terminal 80 kDa of titin includes 45 residue repeating modules (Z-repeats) that are expressed differentially; heart, slow and fast muscles have seven, four to six and two to four Z-repeats, respectively. Gautel et al. proposed a Z-band model in which each Z-repeat links to one level of alpha-actinin cross-links, requiring that the axial extent of a Z-repeat is the same as the axial separation of alpha-actinin layers, of which there are two in every actin crossover repeat. The span of a Z-repeat in vitro is estimated by Atkinson et al. to be 12 nm or less; much less than half the normal vertebrate muscle actin crossover length of 36 nm. Different actin-binding proteins can change this length; it is reduced markedly by cofilin binding, or can increase to 38.5 nm in the abnormally large nemaline myopathy Z-band. Here, we tested whether in normal vertebrate Z-bands there is a marked reduction in crossover repeat so that it matches twice the apparent Z-repeat length of 12 nm. We found that the measured periodicities in wide Z-bands in slow and cardiac muscles are all very similar, about 39 nm, just like the nemaline myopathy Z-bands. Hence, the 39 nm periodicity is an important conserved feature of Z-bands and either cannot be explained by titin Z-repeats as previously suggested or may correlate with two Z-repeats. 相似文献

To assess the extent of intra-species diversity and the links between phylogeny, lifestyle (habitat and pathogenicity) and phenotype, we assayed the growth yield on 95 carbon sources of 168 Escherichia strains. We also correlated the growth capacities of 14 E. coli strains with the presence/absence of enzyme-coding genes. Globally, we found that the genetic distance, based on multilocus sequence typing data, was a weak indicator of the metabolic phenotypic distance. Besides, lifestyle and phylogroup had almost no impact on the growth yield of non-Shigella E. coli strains. In these strains, the presence/absence of the metabolic pathways, which was linked to the phylogeny, explained most of the growth capacities. However, few discrepancies blurred the link between metabolic phenotypic distance and metabolic pathway distance. This study shows that a prokaryotic species structured into well-defined genetic and lifestyle groups can yet exhibit continuous phenotypic diversity, possibly caused by gene regulatory effects. 相似文献

In hostile environments diversity within a bacterial population may be beneficial for the fitness of the microbial community as a whole. Here we analysed the population diversity of Staphylococcus aureus in infecting and colonizing situations. In the study, performed independently in two German centres, the heterogeneity of the S. aureus population was determined by quantifying the occurrence of phenotypic variants (differences in haemolysis, pigmentation, colony morphology) in primary cultures from nose, oropharyngeal and sputum specimens from cystic fibrosis (CF) patients and in nose swabs from healthy S. aureus carriers. The proportion of heterogeneous samples, the number of clearly distinguishable isolates per sample and the qualitative differences between phenotypes was significantly higher in CF sputum specimens than in the other samples. The heterogeneity of the S. aureus population could be correlated with high bacterial densities in the sputum samples. In patients co-infected with Pseudomonas aeruginosa lower S. aureus bacterial loads and less heterogeneity in the S. aureus population were observed. Typing of all S. aureus isolates from heterogeneous samples by pulsed-field gel electrophoresis or spa typing revealed that the bacteria were polyclonal in 30%, monoclonal with minor genetic alterations in 25% or not distinguishable in 69% of the specimens. Some specimens harboured monoclonal and polyclonal variants simultaneously. Importantly, differences in antibiotic susceptibility were detected in phenotypic S. aureus variants within a single specimen. Diversification of a S. aureus population is highly favoured during chronic CF lung infection, supporting the general hypothesis that maintenance of intrahost diversity can be of adaptive value, increasing the fitness of the bacterial community. 相似文献

We examined whether the intense root competition in a rough fescue grassland plant community in central Alberta, Canada, was important in structuring plant species diversity or community composition. We measured competition intensity across gradients of species richness, evenness, and community composition, using pairs of naturally occurring plants of 12 species. One plant in each pair was isolated from neighbors to measure competition; community structure and environmental conditions were also measured at each pair. We used structural equation modeling to examine how competition influenced community structure. Competition intensity was unrelated to species richness and community composition, but increased competition intensity was associated with a slight decline in evenness. Size-symmetric root competition was probably unimportant in structuring this plant community because there are no feedback mechanisms through which size-symmetric competition can magnify small initial differences and eventually lead to competitive exclusion. In plant communities with little shoot competition, competition and community structure should be unlinked regardless of competition intensity. In more productive systems, we propose that interactions between root and shoot competition may indirectly structure communities by altering the overall asymmetry of competition. 相似文献

Studies of Vibrio cholerae diversity have focused primarily on pathogenic isolates of the O1 and O139 serotypes. However, autochthonous environmental isolates of this species routinely display more extensive genetic diversity than the primarily clonal pathogenic strains. In this study, genomic and metabolic profiles of 41 non-O1/O139 environmental isolates from central California coastal waters and four clinical strains are used to characterize the core genome and metabolome of V. cholerae. Comparative genome hybridization using microarrays constructed from the fully sequenced V. cholerae O1 El Tor N16961 genome identified 2,787 core genes that approximated the projected species core genome within 1.6%. Core genes are almost universally present in strains with widely different niches, suggesting that these genes are essential for persistence in diverse aquatic environments. In contrast, the dispensable genes and phenotypic traits identified in this study should provide increased fitness for certain niche environments. Environmental parameters, measured in situ during sample collection, are correlated to the presence of specific dispensable genes and metabolic capabilities, including utilization of mannose, sialic acid, citrate, and chitosan oligosaccharides. These results identify gene content and metabolic pathways that are likely selected for in certain coastal environments and may influence V. cholerae population structure in aquatic environments. 相似文献

Seasonal time constraints do not explain exceptions to the temperature size rule in ectotherms 总被引:1,自引:0,他引:1
A negative relationship between rearing temperature and adult size is widespread in ectotherms yet its causes remain poorly understood. It has been suggested that summer generations of some aquatic insects provide exceptions to this temperature size rule (TSR). The hypothesis that seasonal time constraints are responsible for these exceptions was tested using the mayfly Cloeon dipterum . Simulating early-season conditions by varying photoperiod and temperature, we found that this species followed the TSR. However, under simulated seasonal time constraints, the species omitted most of its summer adult generation (delaying completion of nymphal development till the next year), which limited the scope and applicability of the hypothesis for this species. Even those animals that completed nymphal development before winter showed no weakening of the negative temperature size relationship that was predicted by the hypothesis. Moreover, these animals did not increase rates of development or growth in response to simulated time constraints. We then analysed further the study that originally indicated aquatic insect exceptions to the TSR and suggest that gaps in the data and/or experimental design can explain the apparent exceptions. From these analyses and a survey of other exceptions, we conclude that time constraints do not account for any reported counter-TSR relationship. 相似文献

A major goal in evolutionary biology is to determine how phenotypic variation arises and is maintained in natural populations. Recent studies examining the morphological, physiological and behavioural differences among discrete colour morphotypes (morphs) have revealed several mechanisms that maintain discrete variation within populations, including frequency‐dependence, density‐dependence and correlational selection. For example, trade‐offs over resource allocation to morphological, physiological and behavioural traits can drive correlational selection for morph‐specific phenotypic optima. Here, we describe a ventral colour polymorphism in the wall lizard (Podarcis muralis) and test the hypothesis that morphs differ along multivariate axes defined by trade‐offs in morphological, physiological, and immunological traits. We show that ventral colour is a discrete trait and that morphs differ in body size, prevalence of infection by parasites and infection intensity. We also find that morphs differ along multivariate phenotypic axes and experience different multivariate selection pressures. Our results suggest that multivariate selection pressures may favour alternative optimal morph‐specific phenotypes in P. muralis. 相似文献

Endogenous levels of IAA, ABA and four types of CKs were analyzed in zygotic and indirect (ISE) and direct somatic embryogenesis of Acca sellowiana. Zygotic and somatic embryos at different developmental stages were sampled for morphological and hormonal analysis. Both embryo types showed substantial asymmetry in hormone levels. Zygotic embryos displayed a conspicuous peak of IAA in early developmental stages. The results outlined the hormonal variations occurring during zygotic and somatic embryogenesis regarding the timing, nature and hormonal status involved in both processes. The short transient pulse of IAA observed on the 3rd day in culture was suggested to be involved with the signaling for the induction of somatic embryogenesis. Fertilized ovule development was associated with increased IAA levels 21?C24?days after pollination, followed by a sharp decrease in the cotyledonary stage, both in zygotic and somatic embryos. There was a prominent increase in ABA levels in cultures which generated ISE 24?C30?days after pollination, a period that corresponds to the heart and torpedo stages. The levels of total CKs (Z, [9R]Z, iP and [9R]iP) were also always higher in zygotic than in somatic embryogenesis. While zygotic embryogenesis was dominated by the presence of zeatin, the somatic process, contrarily, was characterized by a large variation of the other cytokinin forms and amounts studied. The above results, when taken together, could be related to the previously observed high frequency formation of anomalous somatic embryos formed in A. sellowiana, as well as to their low germination ability. 相似文献

Copy number variations (CNVs) have provided a dynamic aspect to the apparently static human genome. We have analyzed CNVs
larger than 100 kb in 477 healthy individuals from 26 diverse Indian populations of different linguistic, ethnic and geographic
backgrounds. These CNVRs were identified using the Affymetrix 50K Xba 240 Array. We observed 1,425 and 1,337 CNVRs in the
deletion and amplification sets, respectively, after pooling data from all the populations. More than 50% of the genes encompassed
entirely in CNVs had both deletions and amplifications. There was wide variability across populations not only with respect
to CNV extent (ranging from 0.04–1.14% of genome under deletion and 0.11–0.86% under amplification) but also in terms of functional
enrichments of processes like keratinization, serine proteases and their inhibitors, cadherins, homeobox, olfactory receptors
etc. These did not correlate with linguistic, ethnic, geographic backgrounds and size of populations. Certain processes were
near exclusive to deletion (serine proteases, keratinization, olfactory receptors, GPCRs) or duplication (homeobox, serine
protease inhibitors, embryonic limb morphogenesis) datasets. Populations having same enriched processes were observed to contain
genes from different genomic loci. Comparison of polymorphic CNVRs (5% or more) with those cataloged in Database of Genomic
Variants revealed that 78% (2473) of the genes in CNVRs in Indian populations are novel. Validation of CNVs using Sequenom
MassARRAY revealed extensive heterogeneity in CNV boundaries. Exploration of CNV profiles in such diverse populations would

The taxonomy of the yeast genusMetschnikowia has undergone profound changes over the past century. Major developments, from the capacity to obtain pure cultures of parasitic species to progress associated with the extensive use of molecular biology tools in yeast systematics, are briefly reviewed. Results from past work and new data are combined to evaluate evolutionary relationships and clarify the classification of both terrestrial and aquatic species. Recent physiological studies, including the utilization of non-conventional carbon and nitrogen sources, and characteristics like lipolytic activity and maximum temperatures for growth, are presented. The assessment of the genetic diversity within the genus by restriction analysis of the mitochondrial DNA and by the production of specific DNA probes has been explored. Bacterial diversity in five Icelandic geothermal waters: temperature and sinter growth rate effects 总被引:1,自引:0,他引:1
The microbial ecology associated with siliceous sinters was studied in five geochemically diverse Icelandic geothermal systems.
Bacterial 16S rRNA clone libraries were constructed from water-saturated precipitates from each site resulting in a total
of 342 bacterial clone sequences and 43 species level phylotypes. In near-neutral, saline (2.6–4.7% salinity) geothermal waters
where sinter growth varied between 10 and ~300 kg year−1 m−2, 16S rRNA gene analyses revealed very low (no OTUs could be detected) to medium (9 OTUs) microbial activity. The most dominant
phylotypes found in these waters belong to marine genera of the Proteobacteria. In contrast, in alkaline (pH = 9–10), meteoric geothermal waters with temperature = 66–96°C and <1–20 kg year−1m−2 sinter growth, extensive biofilms (a total of 34 OTUs) were observed within the waters and these were dominated by members
of the class Aquificae (mostly related to Thermocrinis), Deinococci (Thermus species) as well as Proteobacteria. The observed phylogenetic diversity (i.e., number and composition of detected OTUs) is argued to be related to the physico-chemical
regime prevalent in the studied geothermal waters; alkaliphilic thermophilic microbial communities with phylotypes related
to heterotrophic and autotrophic microorganisms developed in alkaline high temperature waters, whereas halophilic mesophilic

Plantations of fast-growing hybrid trees, such as hybrid poplars and hybrid larch, are increasingly used for wood and timber production, but they are also believed to impair forest biodiversity. Most studies that have assessed how such plantations may alter the diversity and composition of understorey plants were established in agricultural landscapes or have compared tree plantations with old-growth natural forests. Moreover, many important aspects of biodiversity have been overlooked in previous studies, such as functional and beta-diversity. Here, we present results from a study that was aimed at quantifying alpha- and beta-diversity of understorey plant species and functional groups in hybrid poplar (9–10 years) and hybrid larch plantations (16 years) located within a forested landscape of Quebec, Canada. These hybrid plantations were compared to naturally regenerated secondary forests and to native plantations of black spruce of the same origin (clear cut) and similar age. Our results indicate that fast-growing hybrid plantations do not present lower taxonomic and functional alpha-biodiversity indices, but may harbour more diverse communities, in part through the introduction of plant species that are associated with open habitats. We provide further evidence that planted forests may be as heterogeneous as naturally regenerated forests in terms of understorey plant composition. Plant species and functional composition differed slightly between stand types (naturally regenerated forests, native and fast-growing hybrid plantations), with plantations offering a greater potential for colonisation by ruderal species, while being detrimental to species of closed forest habitats. Lastly, plantations of fast-growing hybrids do not induce greater changes in understorey vegetation relative to native plantations of black spruce, at least during the first stand rotation. 相似文献

How does genetic diversity within populations of plants develop during primary succession on alpine glacier forelands? Theory predicts that pioneer populations are characterized by low genetic diversity due to founder effects and that genetic diversity increases within populations as they mature and recurrent gene flow occurs. However, few genetic studies have so far been carried out on plants on glacier forelands. In this study, we analysed the development of genetic diversity with time for populations of Trifolium pallescens along successional series (chronosequences) on three parallel glacier forelands in the European Alps, using neutral amplified fragment length polymorphism. No general trend in the development of genetic diversity was observed with increasing population age: even pioneer populations harboured substantial genetic diversity. Assignment tests showed that the latter consist of a genetic sub-sample from several source areas, and not just from other populations on the glacier forelands. We also detected some long distances-that is, inter-valley gene flow events. However, gene flow was not spatially unrestricted, as shown by a weak isolation by distance pattern within glacier valleys. The actual patterns of genetic diversity along the chronosequences are a result of the combination of factors, such as gene flow and growth rate, influenced by site- and species-specific attributes. 相似文献
