Nitrite accumulates during biological denitrification processes when carbon sources are insufficient. Acetate, methanol, and ethanol were investigated as supplementary carbon sources in the nitrite denitrification process using biogranules. Without supplementary external electron donors (control), the biogranules degraded 200 mg l?1 nitrite at a rate of 0.27 mg NO2–N g?1?VSS h?1. Notably, 1,500 mg l?1 acetate and 700 mg l?1 methanol or ethanol enhanced denitrification rates for 200 mg l?1 nitrite at 2.07, 1.20, and 1.60 mg NO2–N g?1?VSS h?1, respectively; these rates were significantly higher than that of the control. The sodium dodecyl sulfate polyacrylamide gel electrophoresis of the nitrite reductase (NiR) enzyme identified three prominent bands with molecular weights of 37–41 kDa. A linear correlation existed between incremental denitrification rates and incremental activity of the NiR enzyme. The NiR enzyme activity was enhanced by the supplementary carbon sources, thereby increasing the nitrite denitrification rate. The primary objective of this study was to clarify the influence of crop plants on atmospheric methane (CH4) in an agriculture-dominated landscape in the Upper Midwest of the United States. Measurements were carried out at two contrasting scales. At the plant scale, CH4 fluxes from soybean and corn plants were measured with a laser-based plant chamber system. At the landscape scale, the land surface flux was estimated with a modified Bowen ratio technique using measurements made on a tall tower. The chamber data revealed a diurnal pattern for the plant CH4 flux: it was positive (an emission rate of 0.4?±?0.1 nmol m?2 s?1, average of soybean and corn, in reference to the unit ground area) during the day, and negative (an uptake rate of ?0.8?±?0.8 nmol m?2 s?1) during the night. At the landscape scale, the flux was estimated to be 14.8 nmol m?2 s?1 at night and highly uncertain during the day, but the available references and the flux estimates from the equilibrium methods suggested that the CH4 flux during the entire observation period was similar to the estimated nighttime flux. Thus, soybean and corn plants have a negligible role in the landscape-scale CH4 budget.  相似文献

This work investigated the effect of a 6-day heat acclimation (HA) protocol on myotube metabolic responses at baseline and in response to a subsequent lipopolysaccharide (LPS) challenge. C2C12 myotubes were incubated for 2 h/day at 40 °C for 6 days (HA) or maintained at 37 °C (C). Following 24-h recovery, myotubes were challenged with 500 ng/ml LPS for 2 h, then collected for analysis of protein markers of mitochondrial biogenesis and macronutrient storage. Functional significance of these changes was confirmed with mitochondrial respiration and glycolytic measurements on a Seahorse XF-96 analyzer. HA stimulated mitochondrial biogenesis and increased indicators of mitochondrial content [SIRT1 (+?62%); PGC-1α (+?57%); NRF-1 (+?40%); TFAM (+?141%); CS (+?25%); CytC (+?38%); all p?<?0.05]. Altered lipid biosynthesis enzymes [p-ACCa:ACC (+?59%; p?=?0.04) and FAS (??86%; p?<?0.01)] suggest fatty acid generation may have been downregulated, whereas increased GLUT4 (+?69%; p?<?0.01) and LDH-B (+?366%; p?<?0.01) suggest aerobic glycolytic capacity may have been improved. Mitochondrial biogenesis signaling in HA myotubes was suppressed by 500 ng/ml LPS (PGC-1α, NRF-1, TFAM; all p?> 0.05) but increased LDH-B (+?30%; p?=?0.02) and CPT-1 (+?55%; p?<?0.01) suggesting improved catabolic function. Basal respiration was increased in HA myotubes (+?8%; p?<?0.01) and HA myotubes maintained elevated basal respiration during LPS challenge (+?8%; p?<?0.01). LPS reduced peak respiration in C myotubes (??6%; p?<?0.01) but did not impair peak respiration in HA myotubes (p?>?0.05). Oxidative reliance was elevated in HA over that in control (+?25%; p?<?0.01) and in HA?+?LPS over C?+?LPS (+?30%; p?<?0.01). In summary, HA stimulated mitochondrial biogenesis in C2C12 myotubes. HA myotubes exhibited (1) elevated basal/peak mitochondrial respiration capacities; (2) greater oxidative reliance; and (3) protection against LPS-mediated respiration impairment. Collectively, these data suggest HA may improve aerobic metabolism in skeletal muscle and protect against LPS-mediated energy deficit.  相似文献

(S)-1-Phenylethanol derivatives, which are the precursors of many pharmacological products, have also been used as anti-Alzheimer drugs. Bioreduction experiments were performed in a batch and packed-bed bioreactor. Then, the kinetics constants were determined by examining the reaction kinetics in the batch system with free and immobilized carrot cells. Also, the effective diffusion coefficient (De) of acetophenone in calcium alginate-immobilized carrot cells was investigated. Kinetics constants for free cells, which are intrinsic values, are reaction rate Vmax?=?0.052?mmol?L?1?min?1, and constants of the Michaelis–Menten KM?=?2.31?mmol?L?1. Kinetics constants for immobilized cells, which are considered apparent values, are Vmax, app?=?0.0407?mmol?L?1 min?1, KM, app?=?3.0472?mmol?L?1 for 2?mm bead diameter, and Vmax, app?=?0.0453?mmol?L?1 min?1, KM, app?=?4.9383?mmol?L?1 for 3?mm bead diameter. Average value of effective diffusion coefficient of acetophenone in immobilized beads was determined as 1.97?×?10?6?cm2?s?1. Using immobilized carrot cells in an up-flow packed-bed reactor, continuous production of (S)-1-phenylethanol through asymmetric bioreduction of acetophenone was performed. The effects of the residence time and concentrations of substrate were investigated at pH 7.6 and 33°C. The hydrolytic activity of a recombinant β-glycosidase from Dictyoglomus turgidum that specifically hydrolyzed the xylose at the C-6 position and the glucose in protopanaxatriol (PPT)-type ginsenosides followed the order Rf > Rg1 > Re > R1 > Rh1 > R2. The production of aglycone protopanaxatriol (APPT) from ginsenoside Rf was optimal at pH 6.0, 80 °C, 1 mg ml?1 Rf, and 10.6 U ml?1 enzyme. Under these conditions, D. turgidum β-glycosidase converted ginsenoside R1 to APPT with a molar conversion yield of 75.6 % and a productivity of 15 mg l?1 h?1 after 24 h by the transformation pathway of R1 → R2 → Rh1 → APPT, whereas the complete conversion of ginsenosides Rf and Rg1 to APPT was achieved with a productivity of 1,515 mg l?1 h?1 after 6.6 h by the pathways of Rf → Rh1 → APPT and Rg1 → Rh1 → APPT, respectively. In addition, D. turgidum β-glycosidase produced 0.54 mg ml?1 APPT from 2.29 mg ml?1 PPT-type ginsenosides of Panax ginseng root extract after 24 h, with a molar conversion yield of 43.2 % and a productivity of 23 mg l?1 h?1, and 0.62 mg ml?1 APPT from 1.35 mg ml?1 PPT-type ginsenosides of Panax notoginseng root extract after 20 h, with a molar conversion yield of 81.2 % and a productivity of 31 mg l?1 h?1. This is the first report on the APPT production from ginseng root extract. Moreover, the concentrations, yields, and productivities of APPT achieved in the present study are the highest reported to date.  相似文献

The roles of plant growth regulators (PGRs) in plants are well documented. However, there is a little information regarding their roles in alleviating salt stress in plants, particularly peppermint. This necessitated the evaluation of the efficiency of three selected PGRs in counteracting the ill effect of salt stress by conducting a pot experiment on peppermint (Lamiaceae). Three uniform size suckers were transplanted in each pot containing proper nutrients. Thirty day old plants were subjected to 4 levels of salinity, viz. 0, 50, 100 or 150 mM NaCl. Salt stress was given at 30 days after their transplantation (DAT). Plants were sprayed twice, i.e., at 60 and 75 DAT with 10?6 M each of gibberellic acid (GA3), salicylic acid (SA) or triacontanol (Tria). The sampling was made at 100 DAT and harvesting at 120 DAT. The graded levels of salinity decreased growth, photosynthesis, carbonic anhydrase (CA) activity, NPK content, peltate glandular trichome (PGT) density, essential oil (EO) and menthol content and herb, EO and menthol yield, but increased catalase (CAT), peroxidase (POX) and superoxide dismutase (SOD) activities and proline content linearly. Spray of PGRs particularly SA improved all parameters under both salt and salt free conditions. The maximum values for yields of herb, EO and menthol were noted with 0 mM NaCl?×?SA. However, antioxidants, proline content, PGT density and EO content were found to be maximum with 150 mM NaCl?×?SA.  相似文献

Prolonged swimming performances of two as yet unnamed species of three‐spined stickleback, Gasterosteus spp., were compared. The two fishes (not yet formally described, referred to here as benthic and limnetic) inhabit different niches within Paxton Lake, Texada Island, British Columbia, Canada, and are recent, morphologically distinct species. Limnetics had longer endurance during prolonged swimming than did benthics. The mean regression of the log10 of fatigue time (Ft, s) on swimming speed (U, standard length, LS s?1) for limnetics (log10Ft = 7·03 ? 0·46U) had a similar slope, but a significantly higher intercept than that for benthics (log10Ft = 5·55 ? 0·43U). Adult benthics were larger, heavier and deeper‐bodied fish than limnetics. Limnetics, however, had a significantly greater pectoral fin edge:base ratio (mean ± s .e .: limnetics, 4·58 ± 0·43; benthics, 3·63 ± 0·27). In addition, limnetics had significantly lower drag coefficients (CD) than benthics (limnetics, log10CD = ?0·49log10Re + 0·66; benthics, log10CD = ?0·26log10Re ? 0·30) where Re is the Reynolds number [(LSU?1), where U and ν are swimming velocity and the kinematic viscosity of the water, respectively]. Compared to their ancestral form, the anadromous three‐spined stickleback Gasterosteus aculeatus L., limnetics and benthics had significantly longer and shorter endurance times, respectively. In addition, both these fishes had significantly higher fast‐start velocities than their ancestral form. Selection due to differential resource use may have lead to divergence of body form, and, therefore, of steady swimming performance. Therefore predation may be the selective force for the similar high escape performance in these two fishes.  相似文献

A sandy culture experiment was conducted to investigate the effects of exogenous CaCl2 on the indole alkaloid accumulation in Catharanthus roseus under salt stress. One-month seedlings of C. roseus were treated with the different concentrations of NaCl (0, 50, and 100 mmol l? 1) and 7.5 mmol l? 1 CaCl2. The plant samples were analyzed after 7 days of the treatments. The NaCl-stressed plants showed decrease of fresh and dry weight and increase of malondialdehyde (MDA) content compared to control. Tryptophan decarboxylase (TDC) activity increased significantly under 50 mmol l? 1 NaCl without CaCl2 addition, 50 mmol l? 1 NaCl with 7.5 mmol l? 1 CaCl2, and CaCl2 treatment without NaCl addition. There was a significant increase in peroxidase activity under NaCl stress compared to control. The vindoline, catharanthine, vincristine, and vinblastine contents increased under salt stress (especially with 50 mmol l? 1 NaCl treatment with or without CaCl2). Addition of CaCl2 to NaCl-stressed plants increased biomass, TDC activity, vindoline, and catharanthine contents and lowered MDA and vincirstine contents compared to the plants without CaCl2. The plants treated with CaCl2 alone showed higher TDC activity, vindoline, catharanthine, and vinblastine content when compared to control. The results showed that exogenous CaCl2 could promote the indole alkaloid metabolism under salt stress.  相似文献

Pistachio is a tree of the arid and semi-arid regions where salinity and boron (B) toxicity can be major environmental stresses. In this study, individual and combined effects of different concentrations of NaCl (0, 800, 1600, 2400 and 3200 mg kg?1 soil) and B (0, 2.5, 5.0, 10.0 and 20.0 mg kg?1 soil) were studied on growth, gas-exchange and mineral composition of pistachio seedlings for a duration of 120 days. Excess amounts of salinity (> 1600 mg NaCl kg?1 soil) and B (20.0 mg kg?1 soil) significantly reduced the plant growth and CO2 assimilation, which was associated with accumulation of Na, Cl and B in leaves. There was also a decline in cell membrane stability index (MSI). Reduced stomatal conductance (g s) was the primary cause of inhibition of photosynthesis rate (P N) under mild to moderate salinity. However, under severe salt stress and B toxicity, non-stomatal effects contributed to the inhibition of CO2 assimilation in addition to the decline in g s value. Application of 5.0–10.0 mg B kg?1 soil significantly improved the plant growth and P N and also recovered the MSI as countermeasures against salt stress. These observations were related to the role of B in cell membrane structure and functioning which reduced the concentration of toxic ions in the leaves. However, cell membrane damages and chlorophyll loss in plants affected by salt were observed to be exacerbated when excess amounts of B were present. In conclusion, the results revealed that optimizing the B nutrition can improve the performance of pistachio seedlings under salt stress, and NaCl also showed a mitigating effect on B toxicity if its concentration in the soil is kept under the plant salt tolerance threshold.  相似文献

There has been considerable interest in cultivation of green microalgae (Chlorophyta) as a source of lipid that can alternatively be converted to biodiesel. However, almost all mass cultures of algae are carbon-limited. Therefore, to reach a high biomass and oil productivities, the ideal selected microalgae will most likely need a source of inorganic carbon. Here, growth and lipid productivities of Tetraselmis suecica CS-187 and Chlorella sp were tested under various ranges of pH and different sources of inorganic carbon (untreated flue gas from coal-fired power plant, pure industrial CO2, pH-adjusted using HCl and sodium bicarbonate). Biomass and lipid productivities were highest at pH 7.5 (320?±?29.9 mg biomass L?1 day?1and 92?±?13.1 mg lipid L?1 day?1) and pH 7 (407?±?5.5 mg biomass L?1 day?1 and 99?±?17.2 mg lipid L?1 day?1) for T. suecica CS-187 and Chlorella sp, respectively. In general, biomass and lipid productivities were pH 7.5?>?pH 7?>?pH 8?>?pH 6.5 and pH 7?>?pH 7.5?=?pH 8?>?pH 6.5?>?pH 6?>?pH 5.5 for T. suecica CS-187 and Chlorella sp, respectively. The effect of various inorganic carbon on growth and productivities of T. suecica (regulated at pH?=?7.5) and Chlorella sp (regulated at pH?=?7) grown in bag photobioreactors was also examined outdoor at the International Power Hazelwood, Gippsland, Victoria, Australia. The highest biomass and lipid productivities of T. suecica (51.45?±?2.67 mg biomass L?1 day?1 and 14.8?±?2.46 mg lipid L?1 day?1) and Chlorella sp (60.00?±?2.4 mg biomass L?1 day?1 and 13.70?±?1.35 mg lipid L?1 day?1) were achieved when grown using CO2 as inorganic carbon source. No significant differences were found between CO2 and flue gas biomass and lipid productivities. While grown using CO2 and flue gas, biomass productivities were 10, 13 and 18 %, and 7, 14 and 19 % higher than NaHCO3, HCl and unregulated pH for T. suecica and Chlorella sp, respectively. Addition of inorganic carbon increased specific growth rate and lipid content but reduced biomass yield and cell weight of T. suecica. Addition of inorganic carbon increased yield but did not change specific growth rate, cell weight or content of the cell weight of Chlorella sp. Both strains showed significantly higher maximum quantum yield (Fv/Fm) when grown under optimum pH.  相似文献

A greenhouse study was conducted to determine the effect of cadmium (Cd)-resistant Pseudomonas putida strain 710A and Commamonas aquatica strain 710B, used either alone or as a binary mixture on the toxicity of cadmium to mungbean [Vigna radiata (L.) wilczek] plants. The bacterial strains 710A and 710B, isolated from the Semra mines in Ranchi, India, were resistant to 0.5 mM and 1 mM, CdCl2 respectively. Moreover, both strains grew well at 10 °C and 30 °C. Of the two bacterial strains, strain 710B showed 129.6 and 83.5 times higher P solubilizing activity than strain 710A, when grown in liquid broth supplemented with cadmium at 10 °C and 30 °C, respectively. Furthermore, 16S ribosomal DNA (rDNA) sequencing identified 710A as a Pseudomonas putida strain and 710B as a Commamonas aquatica strain. The strain 710A significantly (p?<?0.05) stimulated the growth of roots (35.2 %) and shoots (30 %) of mungbean plants grown in soil treated with 110 μM CdCl2. However, the increase in the case of 710B was found to be 15.4 % and 5.17 %, respectively. The loss of chlorophyll content was replenished by up to 80 %, 66 %, and 77.3 % by inoculation of 710A, 710B and binary mixture, respectively. Moreover the strains were able to reduce Cd accumulation in roots and shoots. Most significantly, residual Cd concentration in soil was reduced in the presence of bioinoculants. However, in terms of efficiency, the strains were found to be in the following order: 710A?>?710A +710B?>?710B. The results suggest that P. putida 710A strain is a potentially effective candidate metal to be used for sequestering and as a growth-promoting bioinoculant in Cd polluted soil.  相似文献

PurposeTo investigate the biophysical meaning of Diffusion Kurtosis Imaging (DKI) parameters via correlations with the perfusion parameters obtained from a long Dynamic Contrast Enhanced MRI scan, in head and neck (HN) cancer.MethodsTwenty two patients with newly diagnosed HN tumor were included in the present retrospective study. Some patients had multiple lesions, therefore a total of 26 lesions were analyzed. DKI was acquired using 5b values at 0, 500, 1000,1500 and 2000 s/mm2. DCE-MRI was obtained with 130 dynamic volumes, with a temporal resolution of 5 s, to achieve a long scan time (>10 min). The apparent diffusion coefficient Dapp and apparent diffusional kurtosis Kapp were calculated voxel-by-voxel, removing the point at b value = 0 to eliminate possible perfusion effects on the parameter estimations. The transfer constants Ktrans and Kep, ve, and the histogram-based entropy (En) and interquartile range (IQR) of each DCE-MRI parameter were quantified. Correlations between all variables were investigated by the Spearman’s Rho correlation test.ResultsModerate relationships emerged between Dapp and Kep (Rho =  − 0.510, p = 0.009), and between Dapp and ve (Rho = 0.418, p = 0.038). En(Kep) was significantly related to Kapp (Rho = 0.407, p = 0.043), while IQR(Kep) showed an inverse association with Dapp (Rho = -0.422, p = 0.035).ConclusionsA weak to intermediate correlation was found between DKI parameters and both Kep and ve. The kurtosis was associated to the intratumoral heterogeneity and complexity of the capillary permeability, expressed by En(Kep).  相似文献

Ileal pouch-anal anastomosis (IPAA) is a classical surgery for ulcerative colitis patients. However, knowledge on trace element alteration in patients who had undergone this surgery is limited. This study was conducted to assess trace element alteration in patients with ulcerative colitis before and after ileal pouch-anal anastomosis. Preoperative (40) and postoperative (35) ulcerative colitis patients were studied. The dietary assessment of trace element intake was undertaken by a semiquantitative food frequency questionnaire. Patients' trace element status of zinc, copper, manganese, selenium, calcium, iron, and vitamin D3 was assessed by measuring their blood concentrations. We found that with the similar dietary intake, there was no statistical difference in the concentrations of plasma copper, iron, calcium, and vitamin D3 in the two groups (P?>?0.05). Compared with preoperative patients, postoperative patients had higher concentrations of plasma zinc (14.51?±?4.75 μmol/l) and manganese (0.21?±?0.11 μmol/l) and lower concentrations of plasma selenium (0.86?±?0.58 μmol/l). Both preoperative and postoperative mean concentrations of plasma calcium and vitamin D3 were below their reference range, respectively. We conclude that IPAA does not seem to alter patients' abnormal trace elements completely. It is important to monitor and supply some specified trace elements even in postoperative patients.  相似文献

Inappropriate use of essential oils may entail risks to human health due to mutational events, carcinogenic effects, genetic damages and sensitizing effect caused by generation of reactive oxygen species. In order to detect radicals that are expected to form during their oxidation, we measured the electron spin resonance (ESR) spectra of a standard reaction mixture (I) containing 25?μM flavin mononucleotide, 0.018% several essential oils (or 0.015% geraniol), 1.9 M acetonitrile, 20?mM phosphate buffer (pH 7.4), 0.1 M α-(4-pyridyl-1-oxide)-N-tert-butylnitrone (4-POBN) and 1.0?mM FeSO4(NH4)2SO4 irradiated with 436?nm visible light (7.8 J/cm2). The ESR peak heights of the standard reaction mixture (I) of the essential oils increased in the following order: tea tree?>?palmarosa?>geranium?>?clary sage?>?petitgrain?>?lavender?>?bergamot?>?frankincense?>?ravintsara?>?ylang ylang?>?lemongrass?>?niaouli?>?eucalyptus globulus?>?peppermint. The ESR peak height of the standard reaction mixture (I) of geraniol, a main component of palmarosa, was comparable to the one of palmarosa (97?±?19% of palmarosa). Furthermore, high performance liquid chromatography (HPLC)-ESR analyses of the standard reaction mixture (I) of palmarosa and geraniol gave the same peaks. The results suggest that the radicals formed in the standard reaction mixture (I) of palmarosa are derived from geraniol. HPLC-ESR-mass spectrometry analyses detected m/z 294 ions, 4-POBN/5-hydroxy-3-methyl-3-pentenyl radical adducts and m/z 320 ions, 4-POBN/C7O2H9 radical adducts in the standard reaction (I) of geraniol. The 5-hydroxy-3-methyl-3-pentenyl and C7O2H9 radicals may be implicated in the sensitizing effect of palmarosa.  相似文献

In this pot experiment, cucumbers (Cucumis sativus L.) were grown in a model soil contaminated by three different concentrations of cadmium (40, 160, and 320 mg.kg?1) with different accompanied anions (Cl?, SO4 2?). In all variants, the most Cd (90 %) was accumulated in the roots, but higher content in the case of Cl?. The distribution of Cd in various cucumber organs was as follows: root > stem > leaf > fruits. However, in variants with higher doses of Cd with SO4 2?, the ratio was changed as follows: root > leaf > stem > fruits. In all variants, least of Cd (max. 1 %) was found in fruits. Variants with the highest Cd doses were significantly different by comparison with all other variants, but higher content was in the case of Cl? anion. Stimulation effect on the biomass production and growth of aerial parts and roots of plants in all variants with Cd was observed. Toxicity symptoms, mainly in the presence of leaf chlorosis and yellowing, were more visible in the variants with Cl?, in comparison with SO4 2?. The amounts of phenol compounds in leaves rose almost in all variants. Only the variants with higher Cd content with SO4 2? showed slight reduction. One possible explanation of reduced content may be their bounding on Cd. The content of salicylic acid was reduced in all variants with Cd treatment. However, it is difficult to conclude their role in plant defence responses to heavy metal, because their actual defence mechanism is still unclear. However, from these results, we can suggest that the accompanying anion and the form in which Cd exists may have an impact on the involvement of various antioxidant systems.  相似文献

On the Hadean–Early Archean Earth, the first islands must have provided hot and dry environments for abiotically formed organic molecules. The heat sources, mainly volcanism and meteorite impacts, were also available on Mars during the Noachian period. In recent work simulating this scenario, we have shown that neat glycine forms a black, sparingly water-soluble polymer (“thermomelanoid”) when dry-heated at 200 °C under pure nitrogen. The present study explores whether relevant minerals and mineral mixtures can change this thermal behavior. Most experiments were conducted at 200 or 250 °C for 2 or 7 days. The mineral matrices used were phyllosilicates (Ca-montmorillonites SAz-1 and STx-1, Na-montmorillonite SAz-1-Na, nontronite NAu-1, kaolinite KGa-1), salts (NaCl, NaCl-KCl, CaCl2, artificial sea salt, gypsum, magnesite), picritic basalt, and three Martian regolith simulants (P-MRS, S-MRS, JSC Mars-1A). The main analytical method employed was high-performance liquid chromatography (HPLC). Glycine intercalated in SAz-1 and SAz-1-Na was well protected against thermomelanoid formation and sublimation at 200 °C: after 2 days, 95 and 79 %, respectively, had either survived unaltered or been transformed into the cyclic dipeptide (DKP) and linear peptides up to Gly6. The glycine survival rate followed the order SAz-1?>?SAz-1-Na?>?STx-1?≈?NAu-1?>?KGa-1. Very good protection was also provided by artificial sea salt (84 % unaltered glycine after 200 °C for 7 days). P-MRS promoted the condensation up to Gly6, consistent with its high phyllosilicate content. The remaining matrices were less effective in preserving glycine as such or as peptides.  相似文献

To examine the effects of subsoil NaCl salinity in relation to water stress imposed at different growth stages, wheat was grown in a heavy texture clay soil (vertosol) under glasshouse conditions in polythene lined cylindrical PVC pots (100 cm long with 10.5 cm diameter) with very low salinity level (ECe 1.0 dS/m; ESP 1.0 and Cl 30 mg/kg soil) in top 10 cm soil (10–20 cm pot zone) and low salinity level (ECe 2.5 dS/m, ESP 5, and Cl 100 mg/kg soil) in top 10–20 cm soil (20–30 cm pot zone). The plants were exposed to three subsoil salinity levels in the 20–90 cm subsoil (30–100 cm pot zone) namely low salinity (ECe: 2.5 dS/m, ESP: 5, Cl: 100 mg/kg soil), medium salinity (ECe: 4.0 dS/m, ESP: 10, Cl: 400 mg/kg) and high salinity (ECe: 11.5 dS/m, ESP: 20, Cl: 1950 mg/kg) in the subsoil (20–90 cm soil layer: 30–100 cm pot zone). Watering of plants was withheld for 20 days commencing at either early booting or anthesis or mid grain filling, and then resumed until maturity, and these treatments were compared with no water stress. Water stress commencing at anthesis stage had the most depressing effect on grain yield and water use efficiency of wheat followed by water stress at grain filling stage and early booting stage. High subsoil salinity reduced grain yield by 39.1, 24.3%, and 13.4% respectively in plants water-stressed around anthesis, early booting, and mid grain filling compared with 36.6% in well-watered plants. There was a significant reduction in root biomass, rooting depth, water uptake and water use efficiency of wheat with increasing subsoil salinity irrespective of water regimes. Plants at high subsoil salinity had 64% of their root biomass in the top 0–30 cm soil and there was a marked reduction in subsoil water uptake. Roots also penetrated below the non-saline surface into salinised subsoil and led to attain high concentration of Na and Cl and reduced Ca/Na and K/Na ratio of flag leaf at anthesis stage. Results suggest that high subsoil salinity affects root growth and water uptake, grain yield and water use efficiency even in well water plants. Water stress at anthesis stage had the most depressing effect on wheat.  相似文献

The levels of essential (Ca, Mg, Mn, Fe, Zn, Cr, Cu, and Co) and non-essential (Cd and Pb) metal in six different varieties of Ethiopian khat (Catha edulis Forsk, an evergreen stimulant plant) commonly consumed in the country and exported to the neighboring countries were determined by flame atomic absorption spectrometry. Known weight of oven-dried khat samples were wet-digested using 2 mL of (69–72%) HNO3 and 2 mL of (70%) HClO4 for 2 h and 30 min at variable temperature (120–270°C). The mineral contents in the digests were analyzed using flame atomic absorption spectrometer. The following concentrations ranges in fresh-weight basis were recorded in decreasing order: Ca (1,038–2,173 µg/g)?>?Mg (478.2–812.3 µg/g)?>?Fe (53.95–82.83 µg/g)?>?Zn (5.18–9.40 µg/g)?>?Mn (6.98–8.66 µg/g)?>?Cu (1.85–5.53 µg/g)?>?Cr (0.66–3.47 µg/g)?>?Co (0.41–0.80 µg/g). A wide variation in the mineral contents of khat from different region of Ethiopia was noticed. The toxic metals (Pb and Cd) were not detected in all the samples analyzed.  相似文献

This work aimed to study the regulation of K+/Na+ homeostasis and the physiological responses of salt-treated sorghum plants [Sorghum bicolor (L.) Moench] grown with different inorganic nitrogen (N) sources. Four days after sowing (DAS), the plants were transferred to complete nutrient solutions containing 0.75 mM K+ and 5 mM N, supplied as either NO3 ? or NH4 +. Twelve DAS, the plants were subjected to salt stress with 75 mM NaCl, which was applied in two doses of 37.5 mM. The plants were harvested on the third and seventh days after the exposure to NaCl. Under the salt stress conditions, the reduction of K+ concentrations in the shoot and roots was higher in the culture with NO3 ? than with NH4 +. However, the more conspicuous effect of N was on the Na+ accumulation, which was severely limited in the presence of NH4 +. This ionic regulation had a positive influence on the K+/Na+ ratio and the selective absorption and transport of K+ in the plants grown with NH4 +. Under control and salt stress conditions, higher accumulation of free amino acids and soluble proteins was promoted in NH4 + grown roots than NO3 ? grown roots at both harvesting time, whereas higher accumulation of soluble sugars was observed only at 7 days of salt stress exposure. Unlike the NH4 + grown plants, the gas exchanges of the NO3 ? grown plants were reduced after 7 days of salt stress. These results suggest that external NH4 + may limit Na+ accumulation in sorghum, which could contribute to improving its physiological and metabolic responses to salt stress.  相似文献

Five isocaloric (430 kcal 100 g?1), isonitrogenous (40% CP) experimental diets were formulated with different concentrations of Bacillus licheniformis fb11 probionts (isolated from the gut of Chitala chitala) viz. Control (without probionts), 5 × 104 CFU g?1 (D1), 5 × 105 CFU g?1 (D2), 5 × 106 CFU g?1 (D3), 5 × 107 CFU g?1 (D4), 5 × 108 CFU g?1 (D5) to evaluate its efficiency in C. chitala juvenile. The best growth performance, feed utilisation, specific α-amylase, total protease and lipase activity were observed with the diet D3 (P < 0.05). The lowest Presumptive Pseudomonas Count, Motile Aeromonad Count, Total Coliform Count was observed for D3 (P < 0.05) on 90th day of trial. Two uppermost values were achieved in case of crude protein for D3 and D2 (P > 0.05). The highest lipid content (12.12 ± 0.4 g 100 g?1) was found for D5 (P < 0.05). The highest gross energy (18.75 ± 0.21 MJ 100 g?1) of carcass was recorded for D3. Thus B. licheniformis fb11 at the concentration 5 × 106 CFU g?1 as probiotic supplement promoted growth, digestion in C. chitala juvenile significantly by modulating intestinal microflora.  相似文献
