Conformation changes in rRNA induced by electric impulses   总被引:1,自引:0,他引:1  
We have investigated the effects of electric field pulses (of about 44 kV/cm magnitude and 30 μsec duration) on E. coli ribosomal RNA in solution (5 × 10?4 M sodium cacodylate buffer, pH 7, 20° C). The time course of changes in ultraviolet absorbance and electric dichroism can be resolved into contributions from orientational and conformational changes. The fast part of the relaxation spectrum (completed in less than 1 μs) is attributed to alignment of double-helical segments of rRNA with the external electric field; the slower part (relaxation times in the ms and μs range) is shown to be due to changes in orientation of the entire τRNA molecule, concomitant with electric field-induced helix-coil transitions of oligomeric base-paired regions.  相似文献

Mechanisms that guide directional migration of neuroblasts from the subventricular zone (SVZ) are not well understood. We report here that endogenous electric currents serve as a guidance cue for neuroblast migration. We identify the existence of naturally occurring electric currents (1.5±0.6 μA/cm2, average field strength of ~3 mV/mm) along the rostral migration path in adult mouse brain. Electric fields of similar strength direct migration of neuroblasts from the SVZ in culture and in brain slices. The purinergic receptor P2Y1 mediates this migration. The results indicate that naturally occurring electric currents serve as a new guidance mechanism for rostral neuronal migration.  相似文献

Electric impulses of high field intensity (2 × 105 to 3 × 106 Vm?1, 1 to 20 μs duration) cause transient changes in the optical absorbance of suspended purple membranes of Halobacterium halobium. The electric dichroism at 1 mm NaCL, pH ≈ 6 and at 293K is dependent on field strength, pulse duration and wavelength of the monitoring, plane-polarized light in the range 400 to 650 nm. The optically detected processes are, however, independent of bacteriorhodopsin concentration, of ionic strenght and of the intensity of the monitoring light. These data together with the analysis of time course ands steady state of the reduced dichroism, suggest electric field-sensitive, intramemembraneous structural changes which lead to restricted orientation changes of the chromophore. A thoretical analysis of restricted orientation is developed and applied to the electro-optic data. As a result, it is found that the electric dichroism of purple membrane is associated with a large polarizability anisotropy of 2.4 × 10?30 Fm2 (2.2 × 10?14 cm3); the electric permanent dipole moment which is involved amounts to 4.7 × 10?28 Cm(140 Debye). The kinetic data suggest a cyclic reaction scheme with at least five different conformations. The high polarizability is probably due to displaceable ionic groups within the cooperative lattice of bacteriorhodopsin molecules in purple membranes.  相似文献

The process of controlling bacterial adhesion using an electric current deserves attention because of its ease of automation and environmentally friendly nature. This study investigated the role of electric currents (negative, positive, alternating) for preventing adhesion of Pseudomonas aeruginosa and achieving bacterial inactivation. Indium tin oxide (ITO) film was used as a working electrode to observe adhesion and inactivation under electric polarization. Electric current types were classified into negative, positive, and alternating current. The working electrode acted as a cathode or anode by applying a negative or positive current, and an alternating current indicates that the negative current was combined sequentially with the positive current. The numbers of adhered cells were compared under a flow condition, and the in situ behavior of the bacterial cells and the extent of their inactivation were also investigated using time-lapse recording and live/dead staining, respectively. The application of a negative current prevented bacterial adhesion significantly (~81% at 15.0 μA cm?2). The positive current did not significantly inhibit adhesion (<20% at 15.0 μA cm?2), compared to the nonpolarized case. The alternating current had a similar effect as the negative current on preventing bacterial adhesion, but it also exhibited bactericidal effects, making it the most suitable method for bacterial adhesion control.  相似文献

Cell electrofusion has been widely used in the induction of tetraploidy in mammals, but little attention has been paid in molluscs. This work pursued the establishment of fusion medium (ionic vs. non-ionic) and electric parameters in the electrofusion of Pacific oyster zygotes (prior to the completion of the first mitotic division), minimizing all deleterious effects possible to D-larval stage. The tested combinations of electric field intensity (Vcm−1) and number of square DC pulses applied (for 50 μs each) were (Voltage × N° pulses): 400 × 1, 400 × 2, 400 × 3 and 600 × 1, 600 × 2, 600 × 3.When pulses were applied for first time, it was determined that an ionic fusion medium (microfiltered seawater) offered better conditions than the non-ionic fusion media previously used (0.6 M sucrose or 0.6 M mannitol) in terms of embryo survival and lysis rates. In this fusion medium, two different combinations of electric parameters (3 square DC pulses of 400 Vcm−1 for 50 μs each at 26 °C and 1 square DC pulse of 600 Vcm−1 for 50 μs at 26 °C) offered the best technical results of fusion (57 and 79% respectively) and survival until D-larva (44 and 41% respectively). In conclusion, these electric parameters could be established, using seawater as electrofusion medium, for further approaches to evaluate individual ploidy and survival beyond spat.  相似文献

The thermographic method for determining specific absorption rate (SAR) in animals and models of tissues or bodies exposed to electromagnetic fields was applied to the problem of quantifying the current distribution in homogeneous bodies of arbitrary shape exposed to 60-Hz electric fields. The 60-Hz field exposures were simulated by exposing scale models of high electrical conductivity to 57.3-MHz VHF fields of high strength in a large 3.66 × 3.66 × 2.44-m TE101 mode resonant cavity. After exposure periods of 2–30 s, the models were quickly disassembled so that the temperature distribution (maximum value up to 7 °C) along internal cross-sectional planes of the model could be recorded thermographically. The SAR, W′, calculated from the temperature changes at any point in the scale model was used to determine the SAR, W, for a full-scale model exposed to a 60-Hz electric field of the same strength by the relation W = (60/ f2 · (σ′/σ) · W′ where f′ is the model exposure frequency, σ′ is the conductivity of the scale model at the VHF exposure frequency, and σ is the conductivity of the full-scale subject at 60 Hz. The SAR was used to calculate either the electric field strength or the current density for the full-scale subject. The models were used to simulate the exposure of the full-scale subject located either in free space or in contact with a conducting ground plane. Measurements made on a number of spheroidal models with axial ratios from 1 to 10 and conductivity from 1 to 10 s/m agreed well with theoretical predictions. Maximum current densities of 200 nA/cm2 predicted in the ankles of man models and 50 nA/cm2 predicted in the legs of pig models exposed to 60-Hz fields at 1kV/m agreed well with independent measurements on full-scale models.  相似文献

How might electric fish determine, from patterns of transdermal voltage changes, the size, shape, location, and impedance of a nearby object? I have investigated this question by measuring and simulating electric images of spheres and ellipsoids near an Apteronotus leptorhynchus. Previous studies have shown that this fish's electric field magnitude, and perturbations of the field due to objects, are complicated nonliner functions of distance from the fish. These functions become much simpler when distance is measured from the axes of symmetry of the fish and the object, instead of their respective edges. My analysis suggests the following characteristics of high frequency electric sense and electric images. 1. The shape of electric images on the fish's body is relatively independent of a spherical object's radius, conductivity, and rostrocaudal location. 2. An image's relative width increases linearly with lateral distance, and might therefore unambiguously encode object distance. 3. Only objects with very large dielectric constants cause appreciable phase shifts, and the degree of shift depends strongly on water conductivity. 4. Several parameters, such as the range of electric sense, may depend on the rostrocaudal location of an object. Large objects may be detectable further from the head than the tail, and conversely, small objects may be detectable further from the tail than head. 5. Asymmetrical objects produce different electric images, correlated with their cross-sections, for different orientations and phases of the electric field. 6. The steep attenuation with distance of the field magnitude causes spatial distortions in electric images, somewhat analogous to the perspective distortion inherent in wide angle optical lenses.  相似文献

Rats, given the choice, avoid exposure to alternating current (ac) 60-Hz electric fields at intensities ? 75 kV/m. This study investigated the generality of this behavior by studying the response of rats when exposed to high voltage direct current (HV dc) electric fields. Three hundred eighty male Long Evans rats were studied in 9 experiments with 40 rats per experiment and in one experiment with 20 rats to determine 1) if rats avoid exposure to HVdc electric fields of varying field strengths, and 2) if avoidance did occur, what role, if any, the concentration of air ions would have on the avoidance behavior. In all experiments a three-compartment glass shuttlebox was used; either the left or right compartment could be exposed to a combination of HVdc electric fields and air ions while the other compartment remained sham-exposed. The third, center compartment was a transition zone between exposure and sham-exposure. In each experiment, the rats were individually assessed in 1-h sessions where half of the rats (n = 20) had the choice to locomote between the two sides being exposed or sham-exposed, while the other half of the rats'(n = 20) were sham-exposed regardless of their location, except in one experiment where there was no sham-exposed group. The exposure levels for the first six experiments were 80, 55, 42.5, 30, ?36, and ?55 kV/m, respectively. The air ion concentration was constant at 1.4 × 106 ions/cc for the four positive exposure levels and ?1.4 × 106 ions/cc for the two negative exposure levels. Rats having a choice between exposure and non-exposure relative to always sham-exposed control animals significantly reduced the amount of time spent on the exposed side at 80kV/m (P < .002) as they did at both 55 and ?55 kV/m (P < .005). No significant differences between groups were observed at 42.5, 30, or -36 kV/m. To determine what role the air ion concentration might have had on the avoidance behavior at field strengths of 55 kV/m or greater, four additional experiments were conducted. The HVdc exposure level was held constant at either ?55 kV/m (for three experiments) or -55 kV/m (for 1 experiment) while the air ion concentration was varied between experiments at 2.5 × 105 ions/cc, 1.0 × 104 for two of the experiments and was below the measurement limit (< ± 2 × 103 ions/cc) for the other two experiments at 55 and ?55 kV/m. The exposed rats significantly reduced the amount of time spent on the exposed side at 55 and ?55 kV/m, relative to the sham-exposed rats regardless of air ion concentration (all at P < .005). Thus, HVdc electric fields of ? + or ?55 kV/m are sufficient to produce avoidance behavior in rats. The finite-difference time-domain (FDTD) method has previously been used to calculate induced currents in anatomically based models of the human body at frequencies ranging from 20 to 915 MHz and resolutions down to about 1.25 cm. Calculations at lower frequencies and higher resolutions have been precluded by the huge number of time steps that would be needed in these simulations. This paper describes a method used to overcome this problem and efficiently calculate induced currents in an MRI-based, 6-mm-resolution model of the human under a high-voltage transmission line. This model is significantly higher resolution than the 1.31-cm-resolution model previously used; therefore, it can be used to pinpoint locations of peak current densities in the body. Proposed safety guidelines would allow external electric fields of 10 kV/m and 25 kV/m for exposure to 60 Hz fields of the general public and workers, respectively. For this external electric field exposure of 10 kV/m, local induced current densities as high as 20 mA/m2 are found in the head and trunk with even higher values (above 150 mA/m2) in the legs. These currents are considerably higher than the 4 or even 10 mA/m2 that have been suggested in the various safety guidelines, thus indicating an inconsistency in the proposed guidelines. In addition, several ratios of E/H typical of power line exposures were examined, and it was found that the vertical electric field couples strongly to the body, whereas the horizontal magnetic field does not. Proliferative scarring is a human disease with neither available effective treatment nor relevant animal model. One of the hypotheses for scar formation involves deregulation of fibroblast signaling and delayed apoptosis. Here, we introduce a new chemical‐free method for fibroblast density control in culture by intermittently delivered pulsed electric fields (IDPEF), which cause irreversible damage to cell membranes. Using 5–100 pulses with electric field strength of 150 V/mm, pulse duration 70 µs, and frequency of 1 Hz, we investigated the effects of PEF application on growth, death, and regeneration of normal human dermal fibroblasts in culture. We found that the fraction of fibroblasts that survive depends on the number of pulses applied and follows a Weibull distribution. We have successfully developed an IDPEF protocol that controls fibroblasts density in culture. Specifically, through application of IDPEF every 72 h for 12 days, we maintain a normal human dermal fibroblast density in the 3.1 ± 0.2 × 105–1.4 ± 0.2 × 105 cell/mL range. Our results suggest that IDPEFs may prove useful as a non‐chemical method for fibroblast density control in human wound healing. Biotechnol. We conducted a pilot study to assess magnetic field levels in electric compared to gasoline‐powered vehicles, and established a methodology that would provide valid data for further assessments. The sample consisted of 14 vehicles, all manufactured between January 2000 and April 2009; 6 were gasoline‐powered vehicles and 8 were electric vehicles of various types. Of the eight models available, three were represented by a gasoline‐powered vehicle and at least one electric vehicle, enabling intra‐model comparisons. Vehicles were driven over a 16.3 km test route. Each vehicle was equipped with six EMDEX Lite broadband meters with a 40–1,000 Hz bandwidth programmed to sample every 4 s. Standard statistical testing was based on the fact that the autocorrelation statistic damped quickly with time. For seven electric cars, the geometric mean (GM) of all measurements (N = 18,318) was 0.095 µT with a geometric standard deviation (GSD) of 2.66, compared to 0.051 µT (N = 9,301; GSD = 2.11) for four gasoline‐powered cars (P < 0.0001). Using the data from a previous exposure assessment of residential exposure in eight geographic regions in the United States as a basis for comparison (N = 218), the broadband magnetic fields in electric vehicles covered the same range as personal exposure levels recorded in that study. All fields measured in all vehicles were much less than the exposure limits published by the International Commission on Non‐Ionizing Radiation Protection (ICNIRP) and the Institute of Electrical and Electronics Engineers (IEEE). A computational fluid dynamics (CFD) model describing the flow, electric field and temperature distribution of a laboratory‐scale pulsed electric field (PEF) treatment chamber with co‐field electrode configuration was developed. The predicted temperature increase was validated by means of integral temperature studies using thermocouples at the outlet of each flow cell for grape juice and salt solutions. Simulations of PEF treatments revealed intensity peaks of the electric field and laminar flow conditions in the treatment chamber causing local temperature hot spots near the chamber walls. Furthermore, thermal inactivation kinetics of lactoperoxidase (LPO) dissolved in simulated milk ultrafiltrate were determined with a glass capillary method at temperatures ranging from 65 to 80°C. Temperature dependence of first order inactivation rate constants was accurately described by the Arrhenius equation yielding an activation energy of 597.1 kJ mol?1. The thermal impact of different PEF processes on LPO activity was estimated by coupling the derived Arrhenius model with the CFD model and the predicted enzyme inactivation was compared to experimental measurements. Results indicated that LPO inactivation during combined PEF/thermal treatments was largely due to thermal effects, but 5–12% enzyme inactivation may be related to other electro‐chemical effects occurring during PEF treatments. © 2012 American Institute of Chemical Engineers Biotechnol. Aims: The objective was to evaluate the relation of sublethal injury in the outer membrane of Enterobacter sakazakii to the inactivating effect of the combination of pulsed electric fields (PEF) treatments and citral. Methods and Results: The occurrence of sublethal injury in the outer membrane was measured using selective recovery media containing bile salts. Loss of membrane integrity was measured by the increased uptake of the fluorescent dye propidium iodide (PI). PEF caused nonpermanent and permanent envelope permeabilization of Ent. sakazakii at pH 4·0. After PEF, most surviving cells showed transient cell permeabilization and sublethal injury in their outer membranes. The simultaneous application of a mild PEF treatment (100 pulses, 25 kV cm?1) and 200 μl l?1 of citral to cells suspended in pH 4·0 buffer at a final concentration of 107 cells per ml showed an outstanding synergistic lethal effect, causing the inactivation of more than two extra log10 cycles. Conclusions: Our results confirm that the detection of sublethal injury in the outer membrane after PEF may contribute to the identification of the treatment conditions under which PEF may act synergistically with hydrophobic compounds such as citral. Significance and Impact of the Study: Knowledge about the mechanism of microbial inactivation by PEF will aid the establishment of successful combined preservation treatments.  相似文献

The occupational exposure to electric and magnetic fields during various work tasks at seven 110 kV substations in Finland's Tampere region was studied. The aim was to investigate if the action values (10 kV/m for the E‐field and 500 µT for the B‐field) of the EU Directive 2004/40/EC were exceeded. Electric and magnetic fields were measured during the following work tasks: (1) walking or operating devices on the ground; (2) working from a service platform; (3) working around the power transformer on the ground or using a ladder; and (4) changing a bulb from a man hoist. In work task 2 “working from a service platform” the measured electric field (maximum value 16.6 kV/m) exceeded 10 kV/m in three cases. In the future it is important to study if the limit value (10 mA/m2) of Directive 2004/40/EC is exceeded at 110 kV substations. We have studied the dichroism of various samples of calf thymus DNA (of molecular weight from 3 × 105 to 7 × 106) in pulsed electric fields. The results may be summarized as follows:
  • 1 We find that calf thymus DNA behaves in electrical orientation as if it possessed a large permanent dipole moment. This apparent moment is sensitive to such effects as Mg++ binding which lower the net charge on DNA.
  • 2 The limiting dichroism at infinite field corresponds to an angle of at least 80% between the transition moments at 265 nm and the helix axis, and could be consistant with a number of known forms of DNA. This result is independent of DNA molecular weight. There is evidence that the conformation may be different in 80% ethanol.
  • 3 The dichroism relaxation curves contain a component with a relaxation time of about 8 μsec, which is nearly independent of molecular weight, and a longest component which behaves either according to the Broersma theory for low-molecular-weight samples, or the Zimm-Rouse theory at high molecular weights.

The effect of sinusoidal electric fields on the cytosolic free [Ca2+]i concentration in differentiated HL-60 cells was measured. The calcium concentration was measured in a fluorescence spectrometer using the fluorescence sample fluo-3. In the fluorescence spectrometer two samples can be measured simultaneously, one as the sham-exposed control and the other as the field-exposed sample. The effects of an external field, applied using two capacitor plates outside the cuvettes, and a field applied directly to the medium, using two platinum electrodes inside the cuvettes, were measured at selected frequencies between 0 and 100 Hz and field strengths from 1 to 2000 Vpp/m (external field) and from 0.1 to 1000 Vpp/m (in medium). No significant effects of the fields on the cytosolic free [Ca2+]i concentration in HL-60 cells have been observed at the measured frequencies and field strengths. Purple membrane fragments from Halobacterium halobium were oriented by a static electric field in a water suspension. It was found that an electric field of approx. 20 V/cm is sufficient to achieve practically complete orientation; the purple membranes have a permanent electric dipole moment of (6 ±1)· 10?23 C · m, the orientation of the retinal transition moment relative to the direction of the electric dipole moment, θ, is (59 ± 1)0, and the purple membrane rotational diffusion constant Drot = 0.65 s?1. It was found that because of the electrophoretic movement of the particles a hydrodynamic velocity gradient builds up which also orients the purple membranes.  相似文献

A method for calculating the electron kinetics in dense gases in strong electric fields is developed. The method differs from the forward-backward approximation proposed by Ra?zer and Shne?der for “high-energy” electrons in that it introduces the effective cosines of the scattering angles with respect to the electric force, μ+(?, E) and μ?(?, E), which are different from +1 and ?1, as in the forward-backward approximation. The method was implemented numerically for atmospheric-pressure helium and molecular nitrogen for fields in the ranges 10–200 and 50–800 kV/cm, respectively. The cosines μ+(?, E) and μ?(?, E), the frequency of “fatal” collisions making high-energy electrons to pass from an acceleration regime to a deceleration one, and the rate at which the electrons leave the low-energy reservoir with energies of ≤15 eV for nitrogen and of ≤20 eV for helium are calculated.  相似文献

Measurements of the flux density and spectra of magnetic fields (MFs) generated by several types of electric bed heaters (EBH) were made in order to characterize the MFs to which the fetus may be exposed in utero from the mother's use of these devices. Data on MFs were gathered from more than 1,300 in-home and laboratory spot measurements. In-home measurements taken at seven different positions 10 cm from the EBHs determined that the mean flux density at the estimated position of the fetus relative to the device was 0.45 μT (4.5 mG) for electric blankets and 0.20 μT (2.0 mG) for electrically heated water beds. A rate-of-change (RC) metric applied to the nighttime segment of 24 h EMDEX-C personal-dosimeter measurements, which were taken next to the bed of volunteers, yielded an approximate fourfold to sixfold higher value for electric blanket users compared to water-bed heater users. These same data records yielded an approximate twofold difference for the same measurements when evaluated by the time-weighted-average (TWA) MF exposure metric. Performance of exposure meters was checked against standard fields generated in the laboratory, and studies of sources of variance in the in-home measurement protocols were carried out. Spectral measurements showed that the EBH's measured produced no appreciable high-frequency MFs. Data gathered during this work will be used in interpreting results from a component of the California Pregnancy Outcome Study, which evaluates the use of EBHs as a possible risk factor in miscarriage. © 1996 Wiley-Liss, Inc.  相似文献
