Surveys of ixodoid ticks were performed in Novosibirsk Province (Novosibirsk and Toguchin Districts) and in the vicinity of Akademgorodok (Novosibirsk) in 2009–2010. The abundance and distribution of ticks were assessed in 8 types of habitats. Ixodes persulcatus (Schulze, 1930) was collected by flagging in Novosibirsk and Toguchin Districts, with the highest densities of 19 ind./km being observed in habitats with small-leaved trees. Three species of ticks: Ixodes persulcatus, I. pavlovskyi (subspecies I. pavlovskyi occidentalis Filip. et Pan., 1998), and Dermacentor reticulatus (Fabricius, 1794) were recorded in a recreational forest of Akademgorodok. A high abundance (22 ind./km) of I. pavlovskyi was observed in pine forests subjected to considerable recreational load. The abundance of I. persulcatus was the highest in aspen-birch and birch-aspen forests. D. reticulatus was captured in pine forests and fallow lands, its abundance varying from 0.2 to 2 ind./km.  相似文献

With the deployment of transgenic crops expressing δ‐endotoxins from Bacillus thuringiensis (Bt) for pest management, there is a need to generate information on the interaction of crop pests with their natural enemies that are important for regulation of pest populations. Therefore, we studied the effects of the Bt δ‐endotoxins Cry1Ab and Cry1Ac on the survival and development of the parasitoid Campoletis chlorideae Uchida (Hymenoptera: Ichneumonidae) reared on Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae) larvae fed on Bt toxin‐intoxicated artificial diet. The H. armigera larvae fed on artificial diet impregnated with Cry1Ab and Cry1Ac at LC50 (effective concentration to kill 50% of the neonate H. armigera larvae) and ED50 (effective concentration to cause a 50% reduction in larval weight) levels before and after parasitization resulted in a significant reduction in cocoon formation and adult emergence of C. chlorideae. Larval period of the parasitoid was prolonged by 2 days when fed on Bt‐intoxicated larvae. No adverse effects were observed on female fecundity. The observed effects appeared to be indirect in nature, because no Bt proteins were detected through enzyme‐linked immunosorbent assay in the C. chlorideae larvae, cocoons, or adults fed on Cry1Ab‐ or Cry1Ac‐treated H. armigera larvae. The effects of Bt toxin proteins on C. chlorideae were due to early mortality of H. armigera larvae, that is, before completion of parasitoid larval development.  相似文献

The vegetation of the aquatic and marshland habitats of the Orava region (north of Slovakia) was studied in 2009, using traditional phytosociological methods. Sixteen aquatic and eighteen marsh plant communities were described within 96 phytosociological relevés by using TWINSPAN with the application of the dominance principle. Three associations, Potametum alpini, Potametum zizii and Ranunculo-Juncetum bulbosi, were found as new communities for Slovakia. Myriophylletum verticillati, Potametum nodosi, Potametum graminei and Alisma gramineum community were recorded in the northernmost localities in Slovakia. Among marsh communities, Calletum palustris is very rare, both in the Orava region and in Slovakia as a whole. According to Ellenberg’s indicator values (EIV), moisture was evaluated as the main ecological gradient. Plant communities are ordered along the first Detrended Correspodence Analysis (DCA) axis in a typical hydroseries (Potametea → Lemnetea → Phragmition communis → Phalaridion arundinaceae, Oenanthion aquaticae and Sparganio-Glycerion → Magnocaricion elatae). The second DCA axis was most correlated with EIV for nutrients. Among the five directly measured ecological characteristics (temperature, pH and conductivity of water, water depth, and substrate type), conductivity of water (0.44, P < 0.01) and substrate type (0.32, P < 0.05) were the statistically significant variables explaining the variability along the first DCA axis.  相似文献

The cytomolecular discrimination of the Am- and A-genome chromosomes facilitates the selection of wheat-Triticum monococcum introgression lines. Fluorescence in situ hybridisation (FISH) with the commonly used DNA probes Afa family, 18S rDNA and pSc119.2 showed that the more complex hybridisation pattern obtained in T. monococcum relative to bread wheat made it possible to differentiate the Am and A chromosomes within homoeologous groups 1, 4 and 5. In order to provide additional chromosomal landmarks to discriminate the Am and A chromosomes, the microsatellite repeats (GAA)n, (CAG)n, (CAC)n, (AAC)n, (AGG)n and (ACT)n were tested as FISH probes. These showed that T. monococcum chromosomes have fewer, generally weaker, simple sequence repeat (SSR) signals than the A-genome chromosomes of hexaploid wheat. A differential hybridisation pattern was observed on 6Am and 6A chromosomes with all the SSR probes tested except for the (ACT)n probe. The 2Am and 2A chromosomes were differentiated by the signals given by the (GAA)n, (CAG)n and (AAC)n repeats, while only (GAA)n discriminated the chromosomes 3Am and 3A. Chromosomes 7Am and 7A could be differentiated by the lack of (GAA)n and (AGG)n signals on 7A. As potential landmarks for identifying the Am chromosomes, SSR repeats will facilitate the introgression of T. monococcum chromatin into wheat.  相似文献

Lead (Pb) exposure poses devastating effects on central nervous system development of children. To replicate aspects of this neurotoxicity, we examined the effect of lead on the expression of apoptosis and of apoptosis-related genes, XIAP (X chromosome-linked inhibitor of apoptosis protein) and Smac (second mitochondrial activator of caspase), in the hippocampus of developing rats. A total of 48 rats (30-day old) were randomly divided into four groups for intragastrical perfusion of lead acetate [Pb(Ac)2]: untreated, low (2 mg/kg/d), medium (20 mg/kg/d), and high (200 mg/kg/d) dose groups. Pb content was determined in blood, and the apoptosis indexes and XIAP and Smac gene expression were analyzed in the hippocampus. There was a significant difference in apoptosis indexes (AI) between the exposed and control groups (p < 0.01). AI was highest in the high exposure group. XIAP gene expression was reduced in the exposed groups and the expression was negatively correlated with blood lead levels (BLLs) (p < 0.05). But the four groups did not differ in the expression of Smac (p > 0.05). Our data indicate that exposure to Pb(Ac)2 caused a dose-dependent and significant increase of apoptosis in the hippocampus of developing rats through depressing the expression of the XIAP but not the Smac genes.  相似文献

The effect of foliar pretreatment by hydrogen peroxide (H2O2) at low concentrations of 0, 5, 10, and 15 mM on the chilling tolerance of two Zoysia cultivars, manilagrass (Zoysia matrella) and mascarenegrass (Zoysia tenuifolia), was studied. The optimal concentration for H2O2 pretreatment was 10 mM, as demonstrated by the lowest malondialdehyde (MDA) content and electrolyte leakage (EL) levels and higher protein content under chilling stress (7°C/2°C, day/night). Prior to initiation of chilling, exogenous 10 mM H2O2 significantly increased catalase (CAT), ascorbate peroxidase (APX), glutathione-dependent peroxidases (GPX), and glutathione-S-transferase (GST) activities in manilagrass, and guaiacol peroxidase (POD), APX, and glutathione reductase (GR) activities in mascarenegrass, suggesting that H2O2 may act as a signaling molecule, inducing protective metabolic responses against further oxidative damage due to chilling. Under further stress, optimal pretreatments alleviated the increase of H2O2 level and the decrease of turfgrass quality, and improved CAT, POD, APX, GR, and GPX activities, with especially significant enhancement of APX and GPX activities from the initiation to end of chilling. These antioxidative enzymes were likely the important factors for acquisition of tolerance to chilling stress in the two Zoysia cultivars. Our results showed that pretreatment with H2O2 at appropriate concentration may improve the tolerance of warm-season Zoysia grasses to chilling stress, and that manilagrass had better tolerance to chilling, as evaluated by lower MDA and EL, and better turfgrass quality, regardless of the pretreatment applied.  相似文献

The effects of various combinations of light intensity, oxygen concentration, and CO2 concentration on photosynthesis and growth in several algal types were studied. The results suggest the following. (1) Different algae show different responses to high oxygen concentrations and high light intensities. (2) Inhibition of photosynthesis (CO2 fixation and growth), if seen, increases with increasing oxygen concentration and with increasing light intensity (at light intensities greater than saturation). (3) The inhibition of net photosynthesis observed cannot be attributed to high light intensity alone. (4) The inhibition cannot be attributed to increased rates of excretion of organic materials under conditions of high oxygen concentration and high light intensity. (5) Increased concentrations of CO2 can decrease the effect of high oxygen and light in some algae. (6) The decrease in net photosynthesis observed is probably the result of photorespiration. (7) The effect of light intensity, oxygen concentration, or CO2 concentration on algal photosynthesis should not be studied without considering the effect of the other factors. Some implications of these results, as related to primary productivity measurements, are also discussed.  相似文献

An interactive effect of nitric oxide and 24-epibrassinolide (EBL) or 28-homobrassinolide (HBL) was investigated in Lycopersicon esculentum Mill. cv. K-21. Seeds prior to sowing were exposed to sodium nitroprusside (SNP) (0, 10−5 M or 1 M), a nitric oxide donor, for 8 h, and the resulting seedlings were subsequently sprayed with 10−8 M EBL or HBL at 30-day stage. The analysis of plant samples after 24 h of the EBL or HBL spray showed that the lower SNP concentration (10−5 M) favored the growth, pigment content, photosynthetic and enzymatic activities, and also improved the antioxidant system. However, 1 M SNP had inhibitory impact on most of the indices, except the antioxidant system. Brassinosteroids (EBL or HBL), on the other hand, had a stimulatory impact on the aforementioned indices. The inhibitory effect of SNP (1 M) was neutralized by the application of BRs, where EBL was more effective than HBL.  相似文献

Scavenging and anti-Staphylococcus aureus activities of extracts, fractions and subfractions from the in vitro mycelium of two strains of the edible mushroom Volvariella volvacea were determined. Chloroform subfractions of the ATCC62890 strain showed the highest inhibition percentage of the DPPH radical (over 80%) after 180 min. When chloroform and hexane subfractions of the R83 strain were combined they showed moderate (inhibition zone of 8.99 ± 0.78 mm) to high (inhibition zone of 13.06 ± 0.41 mm) activity against the Gram-positive bacteria Staphylococcus aureus, which are 74.4 and 51.2% of tetracycline (inhibition zone of 17.55 ± 0.11 mm). Partitioning suggested that the substances in the chloroform and hexane fractions of the R83 strain act synergistically to give the antimicrobial activity, while separating the substances of the ATCC62890 subfractions reduced their activity. In general, the R83 strain seems to be a source of antimicrobial substances, while the ATCC62890 strain appears to be an alternative source of antioxidants.  相似文献

(1) The serotonin1A receptor is a G-protein coupled receptor involved in several cognitive, behavioral, and developmental functions. It binds the neurotransmitter serotonin and signals across the membrane through its interactions with heterotrimeric G-proteins. (2) Lipid–protein interactions in membranes play an important role in the assembly, stability, and function of membrane proteins. The role of membrane environment in serotonin1A receptor function is beginning to be addressed by exploring the consequences of lipid manipulations on the ligand binding and G-protein coupling of serotonin1A receptors, the ability to functionally solubilize the serotonin1A receptor, and the factors influencing the membrane organization of the serotonin1A receptor. (3) Recent developments involving the application of detergent-based and detergent-free approaches to understand the membrane organization of the serotonin1A receptor under conditions of ligand activation and modulation of membrane lipid content, with an emphasis on membrane cholesterol, are described.  相似文献

Despite the veterinary and medical importance of horse flies, deer flies, and yellow flies, only a few trap types have been evaluated to monitor adult population dynamics. Currently, three trap types are being utilized (H‐trap, Horse Pal® (HP), and Nzi trap), but no head‐to‐head comparisons have been reported. Thus, we conducted comparative trapping studies in Florida and North Carolina. At two study sites in Florida, the efficacy of all three trap types was compared, but only the H‐trap and HP were compared in North Carolina. Although trap type was significant at all sites, the trap type which caught the most specimens was not the same. In Florida at the Lower Suwannee Wildlife Refuge (LSWR) site, the H‐trap caught the most specimens (2,006), followed in decreasing order by Nzi (938) and HP (541). At the Cedar Ridge Ranch site, the Nzi caught significantly more specimens (1,439) than the H‐trap (215) and HP (161), which were not significantly different from each other. In North Carolina, the H‐trap caught approximately twice as many specimens as the HP (1,458 vs 720). These trap comparison studies were followed up by a study on the efficacy of various bait combinations: (No Bait (NB), dry ice only (DI), Trap Tech Lure (TTL) only, and DI + TTL), which was conducted only at the two Florida sites with H‐traps. At both sites, bait combinations significantly affected trap collections. One pattern (DI +TTL > DI > TTL > NB) was recorded at the LSWR, while at the Cedar River Ranch the pattern was DI > DI +TTL > TTL > NB. Our data showed that trap type and bait combination significantly influence overall adult tabanid abundance as well as individual species composition.  相似文献

To investigate the effect of low CO2 on the expression and activity of ferredoxin-NADP+ oxidoreductase (FNR) and this enzyme-mediated cyclic electron flow around photosystem I (cyclic PSI), the activity staining, immunoblotting and initial rate of P700 + reduction were measured in high- or low-CO2-grown (H or L)-cells of wild-type Synechocystis sp. strain PCC 6803 (WT) and its ΔndhB mutant (M55). Major results were depicted as follows. (1) The protein levels and activity of FNR were remarkably stimulated in L-cells of both WT and M55 relative to that in their H-cells. (2) The rate of cyclic PSI was significantly increased in L-cells of WT, not M55, when compared to that in respective H-cells. (3) N-ethylmaleimide, an inhibitor of FNR, partially inhibited the increase in the rate of cyclic PSI induced by low CO2 in both WT and M55. These findings indicated that low CO2 enhanced the expression and activity of FNR and the cyclic PSI mediated by FNR. The contribution of FNR to cyclic PSI is shortly discussed.  相似文献

Previous studies document that PGE2 and adenosine suppress production of inflammatory cytokines. The present study demonstrates for the first time that (1) PGE2 and 2-chloroadenosine (CADO; a stable analog of adenosine) directly inhibit the cytolytic function of human tumor-infiltrating lymphocytes (TILs); (2) the combination PGE2 and CADO have additive suppressive effects; and (3) the cooperative immunosuppressive actions of PGE2 and CADO are mediated via EP2 receptors (EP2Rs) and A2A receptors (A2ARs) and are due to amplification of cAMP production, activation of protein kinase A (PKA) and T cell receptor (TCR) inhibitor Csk leading to inhibition of Lck, ZAP-70 and Akt phosphorylation. (4) During ex vivo expansion, TILs undergo three stages of differentiation converting from TILs with high cytotoxic activity and relative resistance to combined EP2R/A2AR suppression (stage I) to TILs retaining high cytotoxicity and gaining sensitivity to combined suppression (stage II) and then to TILS that are less cytotoxic and very sensitive to combined suppression (stage III). (5) Finally, we find that pretreatment of TILs with non-inhibitory concentrations of EP2R agonists (such as PGE2 or butaprost) or A2AR agonists (such as CADO or CGS21680) increases their cytotoxic activity and induces resistance to EP2R and A2AR inhibitory signaling (cross-resistance) due to homologous and heterologous desensitization and internalization of EP2Rs and A2ARs, thus preventing their inhibitory signaling. We conclude that inducing resistance of TILs to the suppressive effects of PGE2 and adenosine in the tumor microenvironment could represent a novel strategy for improving the efficacy of adoptive immunotherapy.  相似文献

Acetylcholine (ACh, 1 μM) stimulates the activity of contractile vacuole of the amoeba Amoeba proteus. The ACh action is not reproduced by ACh analogs carbacholine and 5-methylfurmethide that are not hydrolized by acetylcholinesterase (AChE). The ACh effect is not blocked by M-cholinolytics (atropine and metylone), but is suppressed by the N-cholynolytic tubocurarine (0.01 μM). The AChE inhibitors eserine (0.001 μM) and armine (0.01 μM) suppress action of ACh on the amoeba contractile vacuole. ACh does not affect the contractile vacuole activation produced by arginine-vasopressin (AVP, 1 μM), but blocks the contractile vacuole activation caused by the ligand of opioid receptors dynorphin A (1–13) at a concentration of 0.1 μM. Based on comparison of the obtained results with literature data, the conclusion is drawn that, in the described ACh effects, the enzyme AChE plays the role of synergist, but not of antagonist. Regulation of the contractile vacuole activity and, hence, the water-salt homeostasis of A. proteus is provided by three independent mechanisms through receptors of the AVP, ACh, and opioid systems.  相似文献

The hypothesis to be tested in this study is whether the introduction of the chloro group into diphenyl ditelluride molecule (p,p′-dichlorodiphenyl ditelluride, compound 1b) alters the antioxidant and scavenging activity of diphenyl ditelluride (compound 1a) in vitro. The results revealed that 1a and 1b had a potent antioxidant activity in vitro. However, the introduction of a functional group, chloro, into diphenyl ditelluride molecule (1b) did not cause great alterations in the antioxidant action of diphenyl ditelluride against lipid peroxidation, protein carbonyl, and scavenging of 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulfonate) and 2,2′-diphenyl-1-picrylhydrazyl radicals. Based on the in vitro results, different doses (0.25 and 0.75 μmol/kg) of 1a and 1b or vehicle (canola oil, 1 ml/kg) were administered to rats to investigate if the presence of chloro into diphenyl ditelluride molecule reduces its toxicity. The data demonstrate that the chloro group introduced into diphenyl ditelluride molecule did not alter the acute oral toxicity in rats. The administration of compound 1a in rats only altered the urea level, while compound 1b caused alterations in all toxicological parameters analyzed (alanine aminotransferase and aspartate aminotransferase activities, urea and creatinine levels) in plasma of rats. The results of the present investigation support similar antioxidant and scavenging activities of 1a and 1b in rat liver homogenate in vitro. Furthermore, the presence of chloro into diphenyl ditelluride molecule did not alter the mortality index but increased toxicity of diphenyl ditelluride in rats.  相似文献

Brown, Hill & Haywood (2020) offered a critical evaluation of the theory and methods of the Oscillayers approach and attempted to test its utility by reproducing global circulation model (GCM)‐based palaeoclimatic reconstructions of two variables (Bio1 and Bio12) for four different time points (130 ka, 787 ka, 3.2 Ma and 3.3 Ma). They concluded that Oscillayers show poor agreement with independent GCMs and thus do not provide a robust approximation of palaeoclimate throughout the Plio‐Pleistocene. Here, I demonstrate that the authors underestimated the ability of Oscillayers to reproduce independent GCMs by not taking into account inter‐framework differences between the models used to generate the Oscillayers and PaleoClim datasets. However, upon correcting this systematic error, differences in Bio1 and Bio12 between Oscillayers and PaleoClim GCMs are less than ± 1°C or ± 50 mm, on average, in 35.9% (range: 11.8–59.0%) or 46.7% (20.8–66.0%) of values, respectively. Thus, the agreement between Oscillayers and PaleoClim is, on average, c. 1.5–2 times higher than estimated by Brown et al. (2020) and mostly well above the inter‐model agreement between two commonly used GCMs (CCSM and MIROC) for the Last Glacial Maximum. Consequently, I conclude that the Oscillayers approach does provide reasonably robust approximations of palaeoclimate throughout the Plio‐Pleistocene. Some clarifications are given.  相似文献

Alternative splicing (AS) involving tandem acceptors that are separated by three nucleotides (NAGNAG) is an evolutionarily widespread class of AS, which is well studied in Homo sapiens (human) and Mus musculus (mouse). It has also been shown to be common in the model seed plants Arabidopsis thaliana and Oryza sativa (rice). In one of the first studies involving sequence-based prediction of AS in plants, we performed a genome-wide identification and characterization of NAGNAG AS in the model plant Physcomitrella patens, a moss.  相似文献

Earlier we have shown that regulation of rhythm and strength of the frog heart contractions, mediated by transmitters of the autonomic nervous system, is of the Ca2+-dependent character. In the present work, we studied chronoand inotropic effect of verapamil—an inhibitor of Ca2+-channels of the L-type, of nickel chloride-an inhibitor of Ca2+—channels of the T-type and of Na+,Ca2+exchangers as well as of adrenaline and acetylcholine (ACh) after nickel chloride. It has been found that the intracardially administered NiCh2 at a dose of 0.01 μg/kg produced a sharp fall of amplitude of action potential (AP) and an almost twofold deceleration of heart rate (HR). The intracardiac administration of NiCh2 (0.01 μg/kg) on the background of action of verapamil (6 mg/kg, i/m) led as soon as after 3 min to even more prominent HR deceleration and to further fall of the AP amplitude by more than 50% as compared with norm. An intracardiac administration of adrenaline (0.5 mg/kg) partly restored the cardiac activity. However, preservation of the myocardium electrical activity in such animals was brief and its duration did not exceed several minutes. Administration of Ni2+ on the background of acetylcholine (3.6 mg/kg) led to almost complete cessation of cardiac activity. As soon as 3 min after injection of this agent the HR decreased to 2 contractions/min. On electrograms (EG), the 10-fold fall of the AP amplitude was recorded. To elucidate role of extraand intracellular Ca2+ in regulation of strength of heart contractions, isometric contraction of myocardium preparations was studied in response to action of NiCl2 (10–200 μM), verapamil (70 μM), adrenaline (5 μM), and acetylcholine (0.2 μM) after NiCl2. It has been found that Ni2+ causes a dose-dependent increase of the muscle contraction amplitude. Minimal change of the contraction amplitude (on average, by 14.9% as compared with control) was recorded at a Ni2+ concentration of 100 μM. An increase of Ni2+ in the sample to 200 μM increased the cardiac contraction strength, on average, by 41%. The negative inotropic action of verapamil was essentially reduced by 100 μM Ni2+. Adrenaline added to the sample after Ni2+ produced stimulating effect on the cardiac muscle, with an almost twofold rise of the contraction amplitude. ACh (0.2 μM) decreased the cardiac contraction amplitude, on average, by 56.3%, whereas Ni2+ (200 μM) administered after ACh not only restored, but also stimulated partly the myocardial work. Within several parts of percent there was an increase of such isometric contraction parameters as amplitude of the effort developed by muscle, maximal rate, maximal acceleration, time of semirise and semifall. The obtained experimental results indicate that the functional activity of the frog pacemaker and contractile cardiomyocytes is regulated by Ca2+-dependent mechanisms. Structure of these mechanisms includes the potential-controlled Land T-channels of the plasma membrane as well as Na+,Ca2-exchangers characteristic exclusively of contractile cardiomyocytes. The existence of these differences seems to be due to the cardiomyocyte morphological peculiarities that appeared in evolution at the stage of the functional cell specialization.  相似文献
