The infrared absorption properties of monodispersed, chemically and optically pure ‘amino-PEG’-bound linear homooligopeptides having the general formula t-Bx(l-Met)n NHPEG, n = 1–15 and H2+(l-Met)nNHPEG, n = 1–14 have been investigated in the solid state. The critical sizes for development of the β-structure and α-helix (n = 3 and n = 13, respectively), and the effect, on the latter, of the solvent from which the solid samples are cast, have been established.  相似文献

Poly(N-δ-trimethyl-l-ornithine), (Me3Orn)n, is usually not able to attain the α-helical conformation in aqueous solution independent of its pH value; however, it becomes α-helical at low concentrations of sodium perchlorate over a wide pH range according to the circular dichorism (c.d.) spectra. Cl?, SO42? and H2PO4? do not induce α-helix formation. One can conclude that a distinct topology of the anions bound by the side chains is responsible for the α-helix-inducing effect of some water-structure-breaking anions such as perchlorate. This means that the anions are inserted between the ?N+ of the side groups shielding the positive charges repelling one another. The insertion of the anions requires that the water molecules surrounding the ions can be stripped off, which is easily possible if they are water-structure-breaking ones. At higher perchlorate concentrations, the c.d. spectrum changes. It is characterized by a negative shoulder near 208 nm and a pronounced minimum at ≈ 226 nm. With increasing temperature, the c.d. spectrum of the α-helix occurs. Finally the α-helix undergoes a conformational change to the random coil. The apparent transition enthalpy ΔHvH is remarkably lower than that of the homologue (Me3Lys)n, obviously due to a lower cooperativity of the transition. In contrast to poly(l-ornithine), (Orn)n, the c.d. spectrum of (Me3Orn)n remains almost unchanged after adding anionic surfactants such as sodium octyl sulphate (SOS) or sodium dodecyl sulphate (SDS). In organic solvents like methanol or isopropanol, in contrast to (Orn)a and (Lys)n, no α-helix formation occurs. However, in mixtures of these alcohols or dioxane with water, α-helix formation is induced by perchlorate, as in pure water. The thermal stability of the α-helix in these systems is increased.  相似文献

15N-enriched poly(l-alanines) of various molecular weights were prepared from l-alanine-N-carboxyanhydride (l-Ala-NCA) and their helix/coil equilibrium in trifluoroacetic acid (TFA) investigated by means of 40.5 MHz 15N nuclear magneic resonance (n.m.r.), 22.3 MHz 13C n.m.r. and circular dichroism (c.d.) spectra. The 15N n.m.r. spectra exhibit at least three peaks, and the dependence of their intensities on molecular weight, molecular weight distribution and temperature, as well as dynamic nuclear Overhauser effect (NOE) measurements, indicate that the high-field peak represents the helix fraction. All three spectroscopic methods agree that a helix→coil transition takes place with decreasing concentration. Furthermore, poly(l-alanines) containing d-alanine or glycine in various mole ratios were synthesizsed by copolymerizations of N-carboxyanhydrides (NCAs). The 15N n.m.r. spetra demonstrate that one d-Ala unit per 100 l-Ala units suffices to affect significantly the helix/coil equilibrium in TFA. In other words, the helix content under equilibrium conditions is highly sensitive to racemization. Furthermore, 13 C n.m.r. cross-polarization/magic angle spinning (CP/MAS) spectra demonstrate that the presence of d-Ala units also affects the α-helix content in the solid state.  相似文献

13C n.m.r. CP/MAS spectra (50.3 and 75.4 MHz) of solid poly(l-lleucines) and poly(d-norvalines) measured with suitable acquisition parameters allow quantification of the composition of the secondary structure. The optimum acquisition parameters were found by systematic variation of the contact time by means of samples containing 5?0% α-helix structure. The polypeptides were prepared by primary or tertiary amine-initiated polymerizations of the corresponding amino acid NCAs and the average degrees of polymerization (DP) were determined by 1H n.m.r. endgroup analysis. The mole fraction of α-helices increases with increasing DP; it depends on the nature of the solvent and to a lesser degree on the polymerization temperature. When prepared under identical conditions, poly(d-norvaline) samples contain more β-sheet structure than poly(l-leucine. Reprecipitation increases the α-helix content, demonstrating that a part of the original β-sheet structure is thermodynamically unstable. The presence of oligomers of DP ?10 is mainly responsible for the thermodynamically stable part of the β-sheet structure. The chain growth mechanism is discussed.  相似文献

Previous studies support the validity of a linear thermodynamic formalism relating the rates of active Na+ transport and oxygen consumption Jr to the electrical potential difference ΔΨ an the affinity α (negative free energy) of the metabolic driving reaction. The formulation was further tested in paired control and experimental hemiskins by the use of two inhibitors of Na+ transport. Ouabain, a specific inhibitor of the Na+ pump, might be expected to diminish the dependence of Jr on ΔΨ without affecting α, whereas 2-deoxy-d-glucose, a competitive inhibitor of glucose metabolism, should be expected to diminish α. Both inhibitors were used at concentrations adequate to depress Na+ transport (i.e. short-circuit current Jo) to some 50°o of control level. Measurements were made of Io and dJrd(ΔΨ), and the apparent value of the affinity αapp was calculated according to the thermodynamic formulation. Ouabain depressed dJrd(ΔΨ) without affecting αapp whereas 2-deoxy-d-glucose depressed αapp without affecting dJrd(αΨ). The demonstration of these effects indicates the utility of the formalism.  相似文献

The nature of the hydrogen bonds formed between glutamic acid and histidine residues between aspartic acid and histidine residues is studied by i.r. spectroscopy. These studies were performed with (l-Glu)n+(l-His)n and with associates of monomeric Glu + His and Asp + His systems in solutions whereby these amino acids had protected α-amino and α-carboxylic groups. It is shown that the OH …N??O?H+N bonds are easily polarizable proton transfer hydrogen bonds. The residence time of the proton at the His is a little larger in the case of the Asp + His than in the case of the Glu + His systems. Polar environments shift this proton transfer equilibrium in favour of the proton limiting structure ?O?H+N, and less polar ones in favour of the structure OH?N. These results demonstrate that the large proton polarizability of the hydrogen bonded system in the active centre of chymotrypsin is responsible for the charge shift caused by the substrate, and thus for the increase in reactivity of the serine residue and the catalytic activity of the enzyme.  相似文献

The transport of [14C]Gly-Pro was examined using a mutant of Salmonella typhimurium (strain TN87) deficient in an X-Pro dipeptidase and an X-Pro-Y iminopeptidase. The dipeptide was taken up by one saturable transport system having a Km of 5.3 · 10?7M and a V of 1.4 nmol/mg dry wt cell per min. The uptake of Gly-Pro was not inhibited by amino acids or tripeptides and the transport system exhibited a rather broad side chain specificity for dipeptides. Dipeptides containing hydrophobic residues were the most potent inhibitors of this dipeptide transport system exhibiting Ki values between 10?8 and 10?7 M. In contrast, dipeptides containing glycine residues were particularly weak inhibitors. Finally, Gly-Pro was found to be in the intact form inside the cell and was concentrated more than 1000-fold.  相似文献

Proton and 13C magnetic resonance studies are reported on the synthetic polypentapeptide of elastin, HCO-(Val(1)-Pro(2)-Gly(3)-Val(4)-Gly(5))n-Val-OMe, where n ∼- 18. Temperature and solvent dependence of peptide NH chemical shift and solvent dependence of peptide carbonyl chemical shift were used to delineate these moieties preliminary to identification of secondary structure.Based on these studies it is proposed, for the organic solvents of dimethyl sulfoxide, methanol, and low-temperature trifluoroethanol, that dynamic hydrogen bonds form in order of decreasing frequency of occurrence between the Val(1)CO and the Val(4) NH (a β-turn), between the Gly(3) NH and the Gly(5)CO (an 11-atom, hydrogen-bonded ring), and a more limited interaction between the Gly(3)CO and the Gly(5) NH (a γ-turn).Arguments are presented that relate the conformational features proposed above to the coacervate, which is a filamentous state.  相似文献

The antibacterial activity and surfactant activity of the compounds trans-[Rh(L)4Cl2]Cl·nH2O increase in the order L = pyridine<4-methylpyridine<4-ethylpyridine<4-n-propylpyridine.As surfactants, the compounds are far more effective at reducing the interfacial tension, n-hexadecaneH2O, than the surface tension, H2Oair.The most effective and efficient surfactant in this series, trans-[Rh(4-n-propylpyridine)4Cl2]Cl·H2O, can cause the leakage of intracellular manganese ions from the gram-positive bacteria, Bacillus brevis ATCC 9999, at a concentration of 130 ppm but there is no observable effect on the retention of intracellular manganese ions at the minimum concentration required to prevent growth of this organism (~0.6 ppm at 23°C in nutrient broth).At 130 ppm, trans-[Rh(4-n-propylpyridine)4Cl2]Cl·H2O does not cause the loss of intracellular manganese ions from the gram-negative bacteria, Escherichia coli JS-1. In this case, a concentration of at least 63 ppm of this rhodium compound is required to prevent the growth of this organism in M9TUH medium at 35°C.On the basis of these results, it is suggested that gross membrane disruption effects caused by the surfactants trans-[Rh(L)4Cl2]Cl·nH2O are not directly responsible for their observed antibacterial action.  相似文献

Three chlorophyll-protein complexes of a Chroomonas species (Cryptophyceae) have been separated by sodium dodecyl sulphate polyacrylamide gel electrophoresis. The two bands at 100 and 42 kDa are Complex I (CP I) and Complex IV (CP IV), the ubiquitous chlorophyll a-proteins associated with Photosystems I and II, respectively. The third 55 kDa band, which had two peptide subunits (24 and 20 kDa), contained both chlorophyll a and chlorophyll c2 in a molar ratio of 1.4 chlorophyll a to 1 chlorophyll c2 (chlorophyll achlorophyll c2 ratio in whole cells = 4). A chlorophyll ac2 fraction with similar spectral and electrophoretic properties was isolated by digitonin-sucrose density gradient centrifugation. This fraction had no photochemical activity and contained only a single carotenoid species with absorbance maxima in methanol at 424, 448 and 476 nm. Efficient energy transfer from chlorophyll c2 to chlorophyll a occurred in the complex.  相似文献

The Michaelis-Menten parameters, JM and Km of the initial 1-min fluxes of uptake of l-phenylalanine and of α-aminoisobutyric acid were determined for extracellular concentrations of Na+ ranging from 0.5 to 110 mequiv/l for Ehrlich ascites tumor cells. The maximal initial flux, JM, decreased with decrease in extracellular Na+ for both α-aminoisobutyric acid and phenylalanine but the Km for α-aminoisobutyric acid increased markedly as the Na+ concentration fell whereas the Km for phenylalanine decreased. Cycloleucine behaved like phenylalanine.The data provides strong evidence that the Na+-independent flux of phenylalanine is an exchange diffusion flux that can be varied by changing the intracellular level of amino acids such as phenylalanine. For phenylalanine, cyclolcucine, and methionine this exchange diffusion flux appears to be additive with the Na+-dependent initial flux. α-Aminoisobutyric acid also has an exchange diffusion that is Na+-independent but it has a high Km and is not additive with the Na+-dependent flux.  相似文献

Binding of the chromogenic ligand p-nitrophenyl α-d-mannopyranoside to concanavalin A was studied in a stopped-flow spectrometer. Formation of the protein-ligand complex could be represented as a simple one-step process. No kinetic evidence could be obtained for a ligand-induced change in the conformation of concanavalin A, although the existence of such a conformational change was not excluded. The entire change in absorbance produced on ligand binding occurred in the monophasic process monitored in the stopped-flow spectrometer. The value of the apparent second-order rate constant (ka) for complex formation (ka = 54,000 s?1m? at 25 °C, pH 5.0, Γ/2 0.5) was independent of the protein concentration when the protein was in the range of 233–831 μm in combining sites and in excess of the ligand. The apparent first-order rate constant (k?a) for dissociation of the complex was obtained from the rate constant for the decomposition of the complex upon the addition of excess methyl α-d-mannopyranoside (k?a = 6.2 s?1 at 25 °C, pH 5.0, Γ/2 0.5). The ratio ka?a (0.9 × 104m?1) was in reasonable agreement with value of 1.1 ± 0.1 × 104m?1 determined for the equilibrium constant for complex formation by ultraviolet difference spectrometry. Plots of ln(kaT) and ln(kaT) vs 1T were linear (T is temperature) and were used to evaluate activation parameters. The enthalpies of activation for formation and dissociation of the complex are 9.5 ± 0.3 and 16.8 ± 0.2 kcal/mol, respectively. The unitary entropies of activation for formation and dissociation of the complex are 2.8 ± 1.1 and 1.3 ± 0.7 entropy units, respectively. These entropy changes are much less than those usually associated with substantial changes in the conformation of proteins.  相似文献

The effect of Mg2+ concentration and phosphorylation of light-harvesting chlorophyll ab-protein on various chlorophyll fluorescence induction parameters of isolated pea thylakoids has been studied. (1) Lowering the Mg2+ concentration from 3 to 0.4 mM decreases only the variable fluorescence (Fv) and the area above the induction curve while at the same time increasing the slow exponential component of the rise (βmax). (2) A further decrease in Mg2+ concentration from 0.4 to 0 mM decreases the initial (F0) fluorescence level such that the ratio FvFm increases slightly as does the area above the induction curve and βmax. (3) Thylakoid membranes, phosphorylated at 5 mM Mg2+, show an equal decrease in Fv and F0, no change in the area above the induction curve and an increase in βmax. At 2 mM Mg2+, however, phosphorylation induced a more extensive quenching of Fv so that the FvFm ratio was lowered and the area above the induction curve decreased while βmax increased. (4) When phosphorylated membranes were subsequently suspended in an Mg2+-free medium the effect on F0 due to phosphorylation was found to be additive to that due to the absence of Mg2+. The effect of membrane phosphorylation on fluorescence is discussed in relation to the control of excitation energy distribution and shows that different mechanisms operate depending on the background Mg2+ levels. At high Mg2+ the phosphorylation seems to affect the absorption cross-section of Photosystem II while at lower Mg2+ levels there is an additional effect of increased spillover from Photosystem II to I.  相似文献

Chicken liver dihydrofolate reductase purified to apparent homogeneity by affinity chromatography contains tightly bound dihydrofolate. The most effective method for removal of the bound substrate is by electrofocusing. This procedure also removes previously unsuspected contaminants. In addition, the isoelectric profile revealed as many as four distinct peaks of enzyme activity. The major peak (pI = 8.4) represents 60–75% of the total activity, is devoid of bound substrate, and exhibits an A280A260 ratio approaching 1.9 and a specific activity of 14 units/mg. The peak of activity at the isoelectric point of 7.4 contains bound dihydrofolate. The major isoelectric band is shown to be homogeneous by the usual criteria. Notable features of the amino acid composition include a single cysteine, three tryptophans, and an excess of acidic residues. The N-terminal residue is valine. The molecular weight as determined by sedimentation equilibrium is 22,474. The s20,w0 is 2.07. A frictional coefficient of 1.2 indicates that the enzyme approximates a sphere. Circular dichroism measurements suggest a low α-helical content and a high degree of β-structure. The molar extinction coefficient was determined to be 28,970.  相似文献

NMR titration curves have been recorded for all the 13C resonances of cis and transN-acetyl-dl-proline in 2H2O. the measured pK2H values are 3.4 ± 0.8 and 4.13 ± 0.08 respectively; the free energy of ionization for the trans isomer being (3.8 kJ/mole) greater than for the cis. The ionization shifts of the two isomers differ significantly only at the acetyl carbonyl and Cγ positions. It is suggested that these are related to conformational changes which stabilize the trans form at low p2H.  相似文献

Salivary glands of Chironomus thummi larvae were incubated in media composed of those NaClKCl, MgCl2NaClorMgCl2KCl combinations and at concentrations which they tolerated without visible damage. Resulting changes in puffing activity were recorded for three chromosomal segments. Within certain combinatorial ranges NaClKCl, MgCl2NaClandMgCl2KCl induced puffs in the three segments. Each inducing range is depicted as a ‘puff-inducing field’ for extracellular ion concentrations (IFe) in a two-dimensional lattice. The IFes are coherent, distinctly delineated and highly overlapping. At most places a transition from 0 to 100 % puff induction (induction of respective puff in each nucleus) depends on changes in media composition of 10–20 mM. Ion sensitivities of the three chromosomal segments were computed for NaCl/KCl/MgCl2 combinations and were found to conform to actual puff-inducing capacities of selected NaCl/KCl/MgCl2 media.  相似文献

An ATPase is demonstrated in plasma membrane fractions of goldfish gills. This enzyme is stimulated by Cl? and HCO3?, inhibited by SCN?.Biochemical characterization shows that HCO3? stimulation (Km = 2.5 mequiv./l) is specifically inhibited in a competitive fashion by SCN? (Ki = 0.25 mequiv./l). The residual Mg2+-dependent activity is weakly is weakly affected by SCN?.In the microsomal fraction chloride stimulation of the enzyme occurs in the presence of HCO3? (Kmfor chloride = 1 mequiv./l); no stimulation is observed in the absence of HCO3?. Thiocyanate exhibits a mixed type of inhibition (Ki = 0.06 mequiv./l) towards the Cl? stimulation of the enzyme.Bicarbonate-dependent ATPase from the mitochondrial fraction is stimulated by Cl?, but this enzyme has a relatively weak affinity for this substrate (Km = 14 mequiv./l).  相似文献
