The zonal organization of the corticonuclear and the olivocerebellar climbing fiber projections to the vermis of the cerebellum of the rat was compared to the pattern of zebrin-positive and zebrin-negative bands in material double-stained for zebrin II and for different anterograde tracers injected in subnuclei of the inferior olive, or retrograde tracers injected in the cerebellar and vestibular target nuclei of the Purkinje cells of the vermis. Projection zones A1, AX, X, B, CX in the vermis and A2 (accessory A zone) and C2 in the hemisphere were defined by their efferent corticonuclear and their afferent climbing fiber connections, and were found to share the same topographical framework with the zebrin pattern.  相似文献

目的:探讨小脑顶核对淋巴细胞功能的调节作用及其作用途径。方法:用海人酸(KA)损毁大鼠双侧小脑顶核,术后第8d用血细胞计数法和酶联免疫吸附试验(ELISA)分别检测动物外周血中淋巴细胞数和血清中抗绵羊红细胞(SRBC)特异性IgM抗体水平。用电损毁小脑上脚交叉中顶核投射至下丘脑的神经纤维,检测动物淋巴细胞数和抗SRBC特异性IgM抗体水平的变化。结果:KA注入双侧小脑顶核后第8d,在Nissl染色的小脑切片,呈现双侧顶核内神经元胞体破坏。作为对照,在生理盐水注入顶核的动物脑片上,可见正常的Nissl小体。小脑顶核损毁后第8d,动物外周血中淋巴细胞数占白细胞总数的百分比以及血清中抗SRBC特异性IgM抗体水平均明显高于顶核注射生理盐水的对照动物。电损毁小脑上脚交叉处顶核投射至下丘脑的神经纤维后第8d,外周血中淋巴细胞的百分比及抗SRBC特异性IgM抗体水平均明显高于假损毁小脑上脚交叉的对照动物。结论:小脑顶核的神经元胞体损毁导致淋巴细胞功能增强,小脑顶核投射至下丘脑的神经纤维损毁同样引起淋巴细胞功能增强,这些结果提示小脑顶核至下丘脑的神经投射参与介导小脑顶核对淋巴细胞功能的调节作用。  相似文献

Projections of the central cerebellar nuclei to the intralaminar thalamic nuclei were studied in cats with the use of light and electron microscopy. Almost all intralaminar nuclei were shown to obtain cerebello-thalamic projections. The entire complex of the central cerebellar nuclei serves as a source of such projections; yet, involvement of different nuclei is dissimilar. Destruction of the central and, especially, caudal regions of the fastigial nucleus evoked in the intralaminar thalamic nuclei degenerative changes in the nerve fibers (from swelling and development of varicosities up to total fragmentation). Pathological phenomena could be noticed in the most caudal regions of the above thalamic nuclear group, including the medial dorsal nucleus. Projections of the cerebellar interpositus nucleus were directed toward nearly the same regions of the intralaminar nuclei; degeneration was more intensive (covered thecentrum medianum) when posterior regions of the interpositus nucleus were destroyed. Destruction of the lateral cerebellar nucleus evoked a similar pattern of pathological changes, but degeneration was also observed in some structures of the ventral and anterior nuclear groups of the thalamus. Electron microscopic examination showed that degeneration of dark and light types developed in the fiber preterminals and terminals. It can be concluded that the central cerebellar nuclei project not only to the ventral complex of the thalamic nuclei, but also to the anterior, medial, and intralaminar nuclear groups (rostral and caudal portions).  相似文献

3H-glycine was applied to the cat cerebellar cortex under resting conditions and during inferior olive stimulation which activated the climbing fiber system on a restricted area. Electric recording was made. The autoradiograms showed, that under resting condition labelled glycine was incorporated mainly in granule, Golgi and basket cells and only a few Purkinje and stellate cells were active. Also cerebellar glomeruli remained without labelling. On climbing fiber stimulation Purkinje cells became activated singly and grouped, also Golgi and stellate cells increased in number. Granule cells were totally inhibited. 3H-glycine, when applied to the rat hippocampus, the dentate gyrus, CA1 and CA4 fields showed labelling at low frequency stimulation. When 400 Hz high frequency stimulation periods were interposed, long-term potentiation ensued. The overall labelling of each hippocampal region was intensified significantly, indicating that glycine incorporation may be a sign not only of excitation but also of long-term potentiation. 3H-glycine was applied to frog spinal cord during rest and dorsal root stimulation. Interneurons and motor neurons excited by the afferent fibers showed intensive glycine uptake. It is concluded that the glycine labelling method is suitable for detecting neural excitation in the structures dealt with in this paper.  相似文献

Summary The localization of serotonin (5-HT)-immunoreactive nerve fibers in the cerebellum of the rat and cat was investigated by means of the peroxidase-anti-peroxidase (PAP) method using highly specific antibodies to 5-HT.Serotonin-containing nerve fibers were distributed throughout the entire cerebellum including the deep cerebellar nuclei, while 5-HT-positive neuronal somata were not detected in the cerebellum of either species. A different pattern of 5-HT innervation was found among the three layers of the cerebellar cortex. There were also interspecific differences in the pattern of distribution of 5-HT. In the rat, the pool of 5-HT nerve fibers mainly consisted of tangential elements, which were predominant in the molecular layer, while in the cat only a few 5-HT fibers were found in the molecular layer of the cerebellar cortex; dense networks of 5-HT nerve fibers were present in the granular layer. 应用辣根过氧化物酶(HRP)做逆行标记,以四甲基联苯胺(TMB),为呈色剂,对成年大鼠下橄榄复合体到小脑分级球和线球的纤维投射进行了详细的研究。结果显示旁线球和线球接受对侧下橄榄复合体的纤维投射。其中,绒球只接受背帽(DC)和内侧副橄榄核(MAO)嘴段的投射;旁绒球接受MAO、橄榄主核(PO)及少量腹外延伸(VLO)的投射。但是,未观察到由下橄榄复合体的背侧副橄榄核(DAO)到旁绒球或绒球的投射。此外,对大鼠橄榄旁绒球和橄榄绒球投射的带型以及下橄榄与绒球和旁线球的功能做了相关的讨论。  相似文献

The distribution of alpha-melanocyte-stimulating hormone-like immunoreactive structures was studied in the brainstem of the cat using an indirect immunoperoxidase technique. Immunoreactivity was observed in several brainstem nuclei of the cat in which no immunoreactivity had been previously reported. Immunoreactive fibres were observed in the following; the inferior central nucleus; the pontine gray nuclei; the K?lliker-Fuse nucleus; the motor trigeminal nucleus, the anteroventral cochlear nucleus; the abducens nucleus; the retrofacial nucleus; the superior, lateral, inferior, and medial vestibular nuclei; the lateral nucleus of the superior olive; the external cuneate nucleus; the nucleus of the trapezoid body; the postpyramidal nucleus of the raphe; the medial accessory inferior olive; the dorsal accessory nucleus of the inferior olive; the nucleus ambiguus; the principal nucleus of the inferior olive; the preolivary nucleus; the nucleus ruber; the substantia nigra; and in the area postrema. Our results point to a more widespread distribution of alpha-melanocyte-stimulating hormone-like immunoreactive structures in the cat brainstem than that reported in previous studies carried out in the same region of the cat, rat and humans.  相似文献

Climbing fiber afferents to the cerebellum, from the inferior olivary complex, have a powerful excitatory effect on Purkinje cells. Changes in the responsiveness of olivary neurons to their afferent inputs, leading to changes in the firing rate or pattern of activation in climbing fibers, have a significant effect on the activation of cerebellar neurons and ultimately on cerebellar function. Several neuropeptides have been localized in both varicosities and cell bodies of the mouse inferior olivary complex, one of which, calcitonin gene related peptide (CGRP), has been shown to modulate the activity of olivary neurons. The purpose of the present study was to investigate the synaptic relationships of CGRP-containing components of the caudal medial accessory olive and the principal olive of adult mice, using immunohistochemistry and electron microscopy. The vast majority of immunoreactive profiles were dendrites and dendritic spines within and outside the glial boundaries of synaptic glomeruli (clusters). Both received synaptic inputs from non-CGRP labeled axon terminals. CGRP was also present within the somata of olivary neurons as well as in profiles that had cytological characteristics of axons, some of which were filled with synaptic vesicles. These swellings infrequently formed synaptic contacts. At the LM level, few, if any, CGRP-immunoreactive climbing fibers, were seen, suggesting that CGRP is compartmentalized within the somata and dendrites of olivary neurons and is not transported to their axon terminals. Thus, in addition to previously identified extrinsic sources of CGRP, the widespread distribution of CGRP within olivary somata and dendrites identifies an intrinsic source of the peptide suggesting the possibility of dendritic release and a subsequent autocrine or paracrine function for this peptide within olivary circuits.  相似文献

Cerebellar climbing fibers have a unique relationship with the dendritic tree of cerebellar Purkinje cells and have been proposed as a key input in establishing long-term plastic changes in the cerebellar cortex. Although both glutamate and aspartate and a number of neuropeptides have been implicated as climbing fiber-released neurotransmitters/neuromodulators, the in vivo release of these substances during climbing fiber stimulation remains to be demonstrated. In the present study, climbing fibers were activated with harmaline and rats or mice were implanted with a microdialysis probe or a microperfusion probe, respectively, to measure amino acid or peptide release. Additional rats were euthanized at various timepoints post-harmaline injection and Fos immunocytochemistry was used to visualize the activation pattern of the inferior olive, cerebellar cortex and deep nuclei over time. Fos expression was first detected in the inferior olive at 15 min post-harmaline injection followed by expression in the deep cerebellar nuclei (30 min) and then in the cerebellar cortex (1 h). Between 2 and 6 h Purkinje cells expressing Fos were found in variable numbers in both the vermal and paravermal regions and there was a distinct parasagittal-banding pattern in the vermal region. Of several amino acids measured following harmaline administration only glutamate and aspartate levels increased significantly in the first dialysate sample compared to preharmaline levels and their release was blocked by prior lesion of the inferior olive. Citrulline also increased following climbing fiber stimulation, but this occurred in the second and third dialysate samples and may reflect nitric oxide production. Four peptides were examined in cerebellar microperfusates following climbing fiber stimulation. Only corticotropin releasing factor (CRF), calcitonin gene related peptide (CGRP) and bradykinin were significantly increased compared to pre-harmaline levels. These results suggest that glutamate, aspartate, CRF and CGRP are released from climbing fibers during activation of the olivocerebellar system.  相似文献

Evoked response (field potentials) were recorded in all cerebellar nuclei when stimulating the locus ceruleus (LC). Response occurring in the dentate and fastigial nuclei were of considerably greater amplitude and were repeated at a higher rate than in the interstitial nucleus. Blockade of beta-adrenoreceptors led to clear-cut depression of response in the dentate and fastigial nuclei and increased response in the interstitial nucleus. Neuronal response to LC stimulation in the nuclei mainly took the form of inhibition of spontaneous discharges. A less pronounced response was observed in the interstitial nucleus. The findings obtained would indicate the presence of noradrenergic afferents reaching the cerebellar nuclei, where they are unevenly distributed.Kh. Volumes of medial, interposed, and lateral cerebellar nuclei (MCN, ICN, and LCN) were measured in Insectivora, Scandentia, and Primates, including man. The relative size of the nuclei was expressed in size indices. Insectivora had by far the smallest cerebellar nuclei. The simians, in general, had larger cerebellar nuclei than the prosimians, but there was considerable overlap. From Insectivora to man, the MCN was the least progressive and the LCN the most progressive. The indices are expected to reflect the relative size of the three longitudinal zones of the cerebellum (vermis/MCN, pars intermedius/ICN, hemisphere/LCN). They, together with those of the ventral pons and cerebellum (part I), are discussed in relation to the predominant locomotor pattern of a species, and with reference to evolutionary trends in primate phylogeny.  相似文献

The serotoninergic innervation of the inferior olivary complex of the rat was studied using a specific immunohistochemical technique. Fibers and varicosities positive for serotonin were present throughout the nucleus. The densest varicosities were found in the lateral portion of the dorsal accessory olive and the caudal portion of the medial accessory olive. The rostral medial accessory olive and the principal olive were sparsely populated with labeled elements. Ultrastructurally, labeled profiles were found in close opposition to small dendrites and to olivary cell bodies, but they did not display any synaptic specialization. Labeled perikaria were found in the periolivary regions, some of them located laterally to the olivary complex are responsible for the serotoninergic innervation of the dorsal accessory olive; some others located dorsally and medially in the nucleus raphe obscurus and raphe pallidus were responsible for the innervation of the medial accessory olive.  相似文献

The correlation of the activity of neurons in the interposed and dentate nuclei of the cerebellum with conditioned movements of the nictitating membrane was investigated using linear systems analysis. The activity of single deep cerebellar nuclear cells was assumed to be the input to a linear system that produced nictitating membrane movement. Data were initially analyzed with a causal model to assess the degree to which past neural activity predicted the conditioned response. 55 of 165 cells had correlation coefficients of 0.50 or greater between the model's moment-to-moment output and the actual output, with two interpositus cells having correlation coefficients of greater than 0.90. Double-sided impulse responses indicated that afference from the face and efference copy probably affect deep cerebellar neural activity. Nonlinearities were also found in the relationship between neuronal activity and conditioned movement. It was concluded that cerebellar deep nuclear firing is highly correlated with future nictitating membrane movements but that the firing-movement relationship contains noncausal and nonlinear components.  相似文献

Under nembutal narcosis on cats, intracellular electrophysiological studies have been made of common properties and differences in the neuronal organization of the nucleus reticularis tegmenti pontis and intrinsic pontine nuclei. Reciprocal connections of all the nuclei with the cerebellum and sensorimotor cortex were revealed. It was shown that the cerebellar peduncles are involved to a different extent in the transmission of cerebellofugal signals from the nucleus reticularis tegmenti pontis and the pontine nuclei; all these nuclei have different projections to the central cerebellar nuclei and receive different descending cortical impulsation. Functionally, the investigated cortico-cerebellar pontine relay structures supplement and duplicate each other.  相似文献

The occurrence and distribution of retrogradely labeled cells in the perihypoglossal nuclei of the monkey were mapped after injections of horseradish peroxidase in various cerebellar cortical regions. In general the findings are in accord with those made in the cat. The flocculus receives a heavy bilateral projection from the nucleus prepositus, particularly from its caudoventral part, and from the nucleus of Roller. There is an apparently scanty projection from the nucleus intercalatus. The uvula receives a rather similar projection, but in the prepositus the cells projecting to the uvula are on the whole situated more dorsally and rostrally than those supplying the flocculus. The projection to lobules VII-VIII is distinct. More scanty projections have been found to the paramedian lobule and the anterior lobe. The different but partially overlapping sites of origin in the prepositus of fibers to the flocculus and uvula indicate the presence of a topical pattern within the perihypoglosso-cerebellar projection, as in the cat (34). In the monkey the two regions of origin appear to coincide with two particular cell collections in the prepositus (12). Both small and middle sized cells project to the cerebellum, as they do in the cat (9, 48). The nucleus supragenualis nervi facialis in the macaque is morphologically different from the corresponding nucleus in most other mammalian species (12), but it contains labeled cells after injections in the flocculus, uvula and other cerebellar regions. A considerable number of cells in the abducent nucleus are labeled after injections in the flocculus and the posterior vermis.  相似文献

In the cat the possible contribution of cerebellar afferents from the accessory oculomotor nuclei was investigated by means of retrograde transport of the wheat germ agglutinin-horseradish peroxidase complex. A sparse bilateral projection from the interstitial nucleus of Cajal was demonstrated. The fibres reach cerebellar cortical as well as nuclear regions. No retrogradely labelled cells were found in the nucleus of Darkschewitsch and the nucleus of the posterior commissure.  相似文献

The organization of the projection from the pretectal region to the inferior olive in the cat was studied with autoradiographic and horseradish peroxidase (HRP) methods. After injections of HRP into the olive in six cats, cells were labeled ipsilaterally in the anterior pretectal nucleus (NPA), the posterior pretectal nucleus (NPP), the nucleus of the optic tract (NOT), and the dorsal terminal nucleus of the accessory optic tract (DTN). In three experiments, tritiated amino acids were injected into those parts of the pretectal region which contained labeled cells in the HRP experiments, and the projections to the olive were plotted. Both NPA and NPP projected to the rostral half of the dorsal accessory olive, the rostromedial margin of the ventral lamella, and the lateral part of the ventrolateral outgrowth. NOT projected to the caudal half of the dorsal cap, while DTN projected to both the dorsal cap and nucleus beta. The projections are entirely ipsilateral.  相似文献

Location within the brain of HP-labeled neurons (origins of projections to the lateral vestibular nucleus) was investigated by iontophoretic injection of this enzyme. Bilateral projections to the following midbrain structures were revealed: the field of Forel, interstitial nuclei of Cajal, oculomotor nerve nuclei, and the red nucleus — to all parts of the lateral vestibular nucleus. Bilateral projections were also shown from more caudally located structures, viz. the superior, medial and inferior (descending) vestibular nuclei, Y groups of the vestibular nuclear complex, facial nucleus and hypoglossi, nucleus prepositus nervi hypoglossi and caudal nuclei of the trigeminal tract; ipsilateral projections from crus IIa of lobulus ansiformus of the cerebellar hemisphere; contralateral projections from the bulbar lateral reticular nucleus and Deiter's nucleus. A tonic organization pattern of afferent inputs from a number of brainstem formations to the dorsal and ventral lateral vestibular nucleus is revealed and trajectories of HP-labeled fiber systems projecting to Deiter's nucleus described.L. This study investigated the retinal projections of the adult Formosan rock monkey by monocular injection of radioactive proline and fucose. We found that the retinofugal fibers terminated bilaterally in the suprachiasmatic, pregeniculate, lateral geniculate, pretectal complex, pulvinar nucleus, superior colliculus, dorsal and lateral terminal nuclei of the accessory optic system. More crossed retinal terminations were observed, with the exception that the suprachiasmatic nucleus received almost equally of both retinal projections. The existence of the retinal projection to the medial terminal nucleus of the accessory nucleus was in doubt. In the geniculate nucleus, the retinal fibers terminated contralaterally in layers 1, 4 and 6; and ipsilaterally in 2, 3 and 5. In the superior colliculus, most retinal fibers were aggregated superficially in a band located in the contralateral striatum griseum superficialis of the superior colliculus, and had few gaps on the ipsilateral one. The present investigation shows that the Formosan rock monkey has a similar pattern of optic fiber distribution to that of other macaques.  相似文献

Silver staining of nucleolus organizer regions was used for studying their structural alterations during inhibition of ribosomal RNA synthesis by actinomycin D and after its removal in pig kidney cells. Three nuclear types were revealed from the pattern of restored silver grains distribution in the nuclei. When the inhibitor was removed, nuclear multitype gradually disappeared for 24 hours and nuclear RNA synthesis became normal.  相似文献
