The RAPD technique (Random Amplified Polymorphic DNA) was used to assess the level of genetic diversity in Bryum pseudotriquetrum from Southern Victoria Land, Antarctica. Isolates were collected from two transects, and from several other geographically distinct populations within 150 km of Ross Island. Moss growth in one transect, sampled down a small exposed meltstream channel at Cape Chocolate, was very sparse with no other moss colonies found within several hundred meters. Isolates from this transect showed low levels of genetic variation, with many moss clumps appearing identical; these were probably dispersed by water along the channel. In another transect analysed from Granite Harbour, the moss colonies were large, luxuriant and protected by boulders. These isolates showed considerably higher levels of genetic variability than the Cape Chocolate samples, and both within- and between-clump variation was observed. Samples from other sites showed varying levels of genetic diversity. It appears from these results that the degree of protection from the harsh Antarctic environment can influence not only the type of moss growth, but also the level of genetic diversity. The extent of genetic variation and dispersal mechanisms were investigated over short distances of 1–100 m, and up to 3 km, by the random amplified polymorphic DNA (RAPD) technique, for the moss Sarconeurum glaciale , at three locations on Ross Island, Antarctica. At Arrival Heights, genetic variation occurred within single colonies, and the relationships between clumps indicated that they were dispersed down small, meltwater drainage channels by water. The genetic similarities between the colonies from Arrival Heights and others from Scott Base and Crater Hill, a few km away, together with the prevailing wind direction and absence of this moss in the intervening snow-covered area, suggested longer-distance dispersal by wind. Overall, the Ross Island samples appeared to form a single, polymorphic population that was distinct from another population at Canada Glacier, 110 km distant. Somatic mutation, rather than immigration of genetically different propagules from elsewhere, appeared to be the most probable cause of genetic variability in these haploid, vegetatively reproducing Antarctic moss populations. Initiation of recolonization of S. glaciale across a dirt track at Arrival Heights was also investigated by RAPDs, to investigate how regrowth of mosses in disturbed areas occurred in the extreme environment of Antarctica.  相似文献

The moss Hennediella heimii constitutes a significant proportion of the terrestrial flora of East Antarctica, especially in the McMurdo Dry Valleys region of southern Victoria Land. This is the first investigation of the extent of genetic variation within a population of H. heimii. Random Amplified Polymorphic DNA has been used to study both genetic diversity and dispersal of this moss in Miers Valley, southern Victoria Land. The results showed that genetic variation does occur in H. heimii (possibly due to increased exposure to UV-B irradiation), even between shoots within single colonies, and that colonies collected from three transects along meltstreams within the valley constitute a single large population. However, comparison with samples of the same species from nearby valleys and sites up to 500 km distant showed that the Miers Valley population is distinct from these other populations. The results also indicate that wind plays a major role in dispersal of H. heimii in this region of Antarctica. Accepted: 2 September 1998  相似文献

Sixty-six isolates of the moss Sarconeurum glaciale were collected from sites in continental Antarctica at Ross Island, southern Victoria Land and the Vestfold Hills. Genetic variation within and among the populations was estimated using isozymes and random amplified polymorphic DNA (RAPD) technology. Isozyme results only reproducibly showed variation between the populations with one enzyme; RAPDs indicated significantly higher levels of genetic variability within and among the Vestfold Hills samples than in the Ross Sea region samples. A dendrogram produced from the RAPD bands suggested that the Ross Island and southern Victoria Land samples form one population, and those from the Vestfold Hills form a separate and more variable population, possibly resulting from separate colonisation events on the continent. The random amplified polymorphic DNA (RAPD) technique, and DNA sequencing of the conserved nuclear ribosomal DNA internal transcribed spacer region (ITS1-5.8S-ITS2), have been used to assess levels of genetic diversity in the moss Ceratodon purpureus from several locations in Australasia, subantarctic Heard and Macquarie Islands, and continental Antarctica. Populations from Heard and Macquarie Islands and from Antarctica maintain high levels of genetic variation. Both within- and among-colony variation were observed at these locations. DNA sequence analysis showed that samples from the Ross Sea region of Antarctica were most closely related to colonies from Casey and Macquarie Island, and that one colony from Heard Island was most closely related to one from Europe. DNA sequence data separated two Australian populations from the Antarctic and subantarctic group on a dendrogram. Detailed RAPD analysis of a single colony from continental Antarctica demonstrated that mutation probably causes the high variability observed in this moss. DNA sequencing and RAPD analysis are complementary techniques for genetic investigation of Antarctic moss populations.Jenny Ninham was an integral part of this research team for several years. We examined the phylogeography of the endemic Antarctic collembolan Gomphiocephalus hodgsoni using allozymes and mitochondrial DNA (mtDNA; COI) to determine if potentially limited dispersal and long-term habitat fragmentation have promoted regional genetic differentiation. Allozyme analyses showed that differentiation among 21 populations within the Ross Dependency was high (FST = 0.55) with two main groups each representing a distinct geographical region: (1) Ross Island and Beaufort Island; and (2) all continental sites. Ross Island populations showed low levels of differentiation (FST = 0.05) and no correlation with geographical distance, suggesting their derivation from a single glacial refuge. By contrast, continental regions revealed moderate levels of differentiation (FST = 0.27) and a strong correlation with geographical distance, indicating a much older history with several refugia likely. Two sympatric allozyme genotypes were found at three continental sites from Taylor Valley and were congruent with two mtDNA haplotypes, implying nonrandom breeding groups. Although haplotype sharing between one Ross Island site (Cape Bird) and one continental site (Granite Harbour) was identified, the clades showed mostly fragmented allopatric distributions. The extensive Pleistocene glaciations, in conjunction with limited dispersal opportunities, appear to have promoted isolation and divergence among the fragmented habitats. Furthermore, the McMurdo Sound appears to be an effective isolating barrier to dispersal. However, we suggest that the unaided dispersal capacity of G. hodgsoni is unlikely to account for the limited mixing of haplotypes across the McMurdo Sound and recent human- or bird-mediated dispersal is highly probable.  相似文献

Satellite telemetry was used to monitor the migratory movements of a single Adelie penguin (Pygoscelis adeliae) from Cape Hallett (72.31°S, 170.21°E) following the 1997/1998 breeding season. Locations were obtained using the ARGOS satellite system and compared with the migratory paths taken by two penguins from the Northern Colony at Cape Bird, Ross Island (77.22°S, 166.48°E) following the 1990/1991 breeding season. Although the sample sizes are small, if representative they would indicate that: (1) Adelie penguins breeding in the Ross Sea follow a common migratory path, (2) Adelie penguins breeding in the Ross Sea may travel to a common over-winter feeding ground west and north of the Balleny Islands, and (3) Adelie penguins breeding at 77°S on Ross Island travel nearly twice the distance during their over-winter migration as do those penguins breeding at Cape Hallett and colonies further north. While the Cape Hallett penguin was tracked successfully for 172 days, a record for Adelie penguins, the problem of long-term attachment of transmitters to penguins remains.  相似文献

Isolates of the moss Ceratodon purpureus were collected down a channel formed by a meltstream waterfall at Granite Harbour in Southern Victoria Land, Antarctica. The RAPD technique was used to analyse the extent of genetic variation within clumps, between clumps, and between this population and specimens of the same species from two other areas in Antarctica (one a few hundred metres away, the other at Edmonson Point, 300 km further north) and from Sydney, Australia. Genetic variation was detected within and among clumps, with some spatial structure to the population within the channel. Isolates from the nearby location were quite closely related, whereas those from Edmonson Point formed an outgroup on a phylogenetic tree of relatedness. This paper provides an analysis of genetic variability in local populations of the clonal moss Pleurochaete squarrosa, a Mediterranean moss typical of post-fire recovery, and characterised by asexual reproduction. ISSR (Internal Simple Sequence Repeats) primers and trnLUAA (intron of plastid gene for Leu tRNA) length polymorphism were employed to evaluate genetic structure in five southern Italy populations of this moss. Both molecular tools highlight high values of genetic diversity with geographic structure of sampled populations and a low gene flow among the investigated sites. Gene diversity is significant at every hierarchical level of sampling, and generally increases from lower to higher hierarchical levels. Among other factors, the ubiquitous genetic variability detected in P. squarrosa can be related to continuous, occasionally massive, short-range recruitment of propagules, and to the high degree of intermingling, both favoured by the modality of vegetative reproduction and growth occurring in the species.  相似文献

The population genetic structure and spatial genetic structure of the dioecious aquatic macrophyte Vallisneria americana was examined near Turkey Island in the Detroit River in late summer of 1991. The density and distribution of male, female, and vegetative ramets was determined for three transects parallel, and three transects perpendicular to the island, at mean water depths of 38–306 cm. A strong male-biased sex ratio was observed in shallow water, while sex ratios became increasingly female-biased, and plant density increased, at greater water depths. Cellulose acetate gel electrophoresis was used to characterize the allozyme phenotype of each ramet that had been screened for seven enzyme systems. Overall, 91 allozyme phenotypes were identified. A single allozyme phenotype accounted for 33%–55% of all the ramets (depending on the transect), indicating extensive regional vegetative growth. However, the population as a whole displayed high genetic diversity (HT = 0.3403), with most of the diversity occurring within transects (Hs = 0.3297) rather than between transects (DST = 0.0106). The possible factors accounting for the high small-scale genetic diversity of this clonal aquatic plant species are discussed.  相似文献

The gut of animals changes quickly from a totally sterile environment before birth to a numerous and highly diverse microbial community shortly after birth. However, few studies have examined the source of the bacteria colonizing the gut. We used a genetic based approach to investigate the distribution and acquisition of faecal microbial communities by Adelie penguins, Pygoscelis adeliae , breeding in the Ross Sea region of Antarctica by cloning a portion of the 16S rRNA gene and by automated ribosomal intergenic spacer analysis (ARISA). We hypothesized that bacteria were either acquired from the penguins' neighbours or inherited from their ancestors. Samples were collected from Adelie penguin breeding colonies at the north-western edge of the Ross Sea coast through to the southernmost Adelie penguin colonies on Ross Island. Most of the bacterial sequences we obtained were only distantly homologous with previously published sequences. Bacterial taxa appear to have a restricted distribution as the majority of 16S rRNA clones were isolated from only one or two hosts. Faecal bacterial community similarity was strongly negatively correlated with the genetic distance between hosts suggesting that bacterial communities are inherited. There was little support for a correlation between distance between collection sites and community similarity.  相似文献

We used mark-resight data and amplified fragment length polymorphism (AFLP) markers to assess movements and gene flow between Central Pacific breeding colonies of the great frigatebird, Fregata minor. Of 715 adult frigatebirds marked on Tern Island and Johnston Atoll, 21.3% were resighted at other frigatebird colonies at least 582 km away. Mark-resight data indicated regular movement of males and females between Tern Island and Johnston Atoll (873 km apart), and less frequent movements to other islands; no birds marked on Tern or Johnston were seen on Christmas Island, but one was seen in the Philippines, 7627 km from where it was marked. Despite the regular occurrence of interisland movements, Bayesian analyses of AFLP data showed significant genetic differentiation between Tern Island and Johnston Atoll, and more pronounced differentiation between these two islands and the more distant Christmas Island. The AFLP profiles of three birds breeding on Tern Island fell within the profile-cluster typical for Christmas Island birds, both in a nonmetric multidimensional scaling analysis and in a population assignment test, suggesting dispersal events from Christmas Island to Tern Island. Several factors could explain the persistence of genetic structure despite frequent movements between colonies: many movements occurred during the nonbreeding season, many breeding-season movements did not involve mate-acquisition behaviours and individuals that do disperse may be selected against, as suggested by morphometric differences between colonies. The persistence of genetic structure among breeding colonies despite significant interisland movements suggests limits to the effectiveness of migration as a homogenizing force in this broadly distributed, extremely mobile species.  相似文献

  • The moss Syntrichia caninervis is widely distributed in cool temperate and cold deserts where environmental pressures create a dependence on asexual reproduction (fragment reproduction). However, when compared to sporophyte‐producing mosses, there is a lack of evidence to support the capacity of drought‐tolerant mosses that predominantly fragment and produce protonema to disperse over long distances.
  • We used 20 microsatellite loci to study genetic variation and structure in six populations (five natural populations and one population from a regeneration site) in three contrasting and widely separated regions of China.
  • The genetic diversity and expected heterozygosity were lower in populations from the Tengger Desert than in populations from the other regions. Using PCoA, UPGMA and Structure analysis, the genetic grouping divided the three regions into three distinct groups. This may indicate that in regions where S. caninervis reproduces predominantly asexually, propagules are spread mainly by short‐distance dispersal. The genetic diversity of the population from the regeneration site in the Tengger Desert was slightly higher than that of the nearby, naturally occurring population, and included some input from the Pamir Plateau almost 2,300 km to the west, suggesting long‐distance dispersal of S. caninervis propagules across the region.
  • Predominantly asexually reproducing populations of S. caninervis are mainly dependent on short‐distance dispersal. Long‐distance dispersal of S. caninervis propagules across the region is difficult. Establishment of populations with dominant asexual reproduction will eventually result in genetic differentiation.

1. Coenagrion mercuriale is one of Europe's most threatened damselflies. There is concern for the long‐term persistence of many of its U.K. colonies because adult lifetime movement is limited, making isolated populations susceptible to extinction. 2. Using 14 microsatellite loci we characterised levels of genetic diversity, evidence for a recent decline and the spatial genetic structure for C. mercuriale population in Wales, U.K. 3. Spatial isolation is not an absolute predictor of low genetic diversity at either local or regional scales. 4. One population inhabiting a remote, edge of range site is genetically impoverished with levels of variability (at microsatellite loci) among the lowest reported for any insect species. 5. Agricultural land and high ground are physical barriers to dispersal by adults. 6. Consistent with work from elsewhere, movement by mature C. mercuriale in Pembrokeshire is sufficient to prevent significant genetic differentiation throughout a habitat matrix of some 3–4 km if the suitable habitat sites are <2 km apart and lack barriers to movement. Even within a good habitat matrix, however, genetic isolation by distance develops within 10 km.  相似文献

The genetic structure, diversity and phylogeography of the moss species Campylopus oerstedianus in Europe was studied, based on the ITS region of the nrDNA of nine selected European populations. Although this species is only known in sterile stage, long-distance dispersal and gene flow among populations seem to be present within Europe. High levels of genetic differentiation between the investigated Greek population and the western European populations indicate a long lasting isolation. The populations from France and Switzerland root together and supposedly have a common origin. From the French populations, the one from the Pyrenees is the most basal one and the populations in the Massif Central and the Vosges Mts can be derived from it. This indicates relatively recent dispersal of the species from the Mediterranean to the north, in spite of the lack of sporophytes and the fact that the type of the dispersed propagules and their vectors are not obvious.  相似文献

Diversity patterns of the dioecious haploid Mediterranean moss Pleurochaete squarrosa were analysed from Central and Northwest Europe using nuclear and chloroplast DNA sequencing and enzyme electrophoresis. Across 69 populations, 38 distinct haploid multilocus genotypes (MLGs) were detected, but nearly all populations were clonal. Only five MLGs occurred in more than two regions, and two diversity hotspots were detected. The Kaiserstuhl mountains in Southwest Germany harboured 34 MLGs, 25 being endemic within Central Europe. Levels of linkage disequilibrium and population structure in Kaiserstuhl populations were similar to levels and structure in sexually reproducing populations in the Mediterranean Basin. In the Moselle-Nahe area, some 250 km north, a comparably high allelic diversity, but no evidence of recombination, was detected. Genetic diversity measures were significantly lower than estimates obtained in the Mediterranean Basin and a G ST of 0.89 signified extreme population differentiation. Mantel tests identified a positive correlation on genetic and geographical distance for distances up to 50 km. Seven nrITS and three cpDNA haplotypes were detected, their geographical structure mirroring enzyme data set results. Comparative analysis with Mediterranean data demonstrated multiple recolonization of Central Europe from both the Iberian Peninsula and the Balkans. A suture zone of genotypes was detected along the border of Belgium/France and Germany. Despite P. squarrosa having haploid spore and/or vegetative propagules dispersal, we found patterns of postglacial recolonization of Central Europe comparable with those reported in flowering plants and animals. This study demonstrates the importance of comparative research on population genetics and phylogeography of a diverse range of organisms.  相似文献

A trial was undertaken with an electromagnetic position-tracking (Tracker) system by mapping a small section of a lichen habitat near Cape Royds, Ross Island, Antarctica. This method facilitated the simultaneous collection of precise information about both the topography of the site and the distribution of lichen species. These data were used to produce accurate maps and three-dimensional reconstructions of the mapped area. The technique offers considerable savings in time over conventional methods used in plant and lichen surveys, and has potential to enhance the information collected from broader-scale transect surveys. Effective management of reef corals requires knowledge of the extent to which populations are open or closed and the scales over which genetic exchange occurs, information which is commonly derived from population genetic data. Such data are sparse for Great Barrier Reef (GBR) corals and other organisms, with the studies that are available being mostly based on a small number of sampling locations spanning only part of the GBR. Using 11 microsatellite loci, we genotyped 947 colonies of the reef-building coral Acropora millepora from 20 sites spanning almost the full length of the GBR (~12° of latitude and ~1550 km). The results show a major divide between the southernmost central to southern offshore populations and all other sampled populations. We interpret this divide as a signature of allopatric divergence in northern and southern refugia during the Pleistocene glaciations, from which the GBR was subsequently recolonized. Superimposed on this pattern is a cross-shelf genetic division, as well as a separation of inshore populations south of the Cape Clifton Front at ~21.5-22°S. Most inshore populations north of this, as well as mid-shelf populations in the northern and far northern GBR, are open, exchanging recruits frequently. In contrast, inshore populations south of the Cape Clifton Front and offshore populations in the central and southern GBR are largely self-seeding, at least within the spatial resolution that was achieved given our sampling intensity. Populations that have been impacted by recent disturbance events causing extensive coral mortality show no evidence of reduced genetic diversity.  相似文献

The following study was the first to describe composition and structure of the peracarid fauna systematically along a latitudinal transect off Victoria Land (Ross Sea, Antarctica). During the 19th Antarctic expedition of the Italian research vessel “Italica” in February 2004, macrobenthic samples were collected by means of a Rauschert dredge with a mesh size of 500 μm at depths between 85 and 515 m. The composition of peracarid crustaceans, especially Cumacea was investigated. Peracarida contributed 63% to the total abundance of the fauna. The peracarid samples were dominated by amphipods (66%), whereas cumaceans were represented with 7%. Previously, only 13 cumacean species were known, now the number of species recorded from the Ross Sea increased to 34. Thus, the cumacean fauna of the Ross Sea, which was regarded as the poorest in terms of species richness, has to be considered as equivalent to that of other high Antarctic areas. Most important cumacean families concerning abundance and species richness were Leuconidae, Nannastacidae, and Diastylidae. Cumacean diversity was lowest at the northernmost area (Cape Adare). At the area off Coulman Island, which is characterized by muddy sediment, diversity was highest. Diversity and species number were higher at the deeper stations and abundance increased with latitude. A review of the bathymetric distribution of the Cumacea from the Ross Sea reveals that most species distribute across the Antarctic continental shelf and slope. So far, only few deep-sea records justify the assumption of a shallow-water–deep-sea relationship in some species of Ross Sea Cumacea, which is discussed from an evolutionary point of view.  相似文献

Mosses and other bryophytes are vital components of forests, because they sustain a tremendous diversity of invertebrates and influence significant ecological functions. There have been few studies on moss population diversity in Southeast Asia, despite the escalating deforestation in this region of rich biodiversity. The genetic diversity of the tropical moss Acanthorrhynchium papillatum (Harv.) Fleisch., collected from forested areas in Singapore and Peninsular Malaysia, was elucidated using eight microsatellite markers developed for this species. Significant levels of allelic and haplotypic diversity were observed among clumps of the moss. Differences in allelic richness and genotypic diversity among the populations were higher in less disturbed forests compared to the more disturbed areas, suggesting that genetic diversity is affected by habitat quality. Genetic diversity levels within the clumps studied were low, indicating that vegetative reproduction was more important within clumps than sexual reproduction. However, multilocus genotypes of samples within the clumps studied were not all alike, providing evidence of microsatellite mutation or of occasional sexuality. Despite the isolation of populations, A. papillatum can introduce genetic variability by mutation among vegetatively propagated individuals. This study provides baseline information on the genetic diversity of A. papillatum tropical rain forests.  相似文献
