The Effects of Sulphur Dioxide on Phloem Transport in Two Cereals   总被引:1,自引:0,他引:1

In vivo investigations using 11C-labelled photosynthate revealedthat there is a change in the patterns of tracer profiles whencereal leaves are exposed to SO2. The change after exposureto SO2 was interpreted in terms of a decrease in lateral waterflow into the sieve tubes brought about by reduced phloem loadingalong the length of a leaf. Barley and tomato plants were cultured in nutrient solutionsincluding 0.15 mol m–3 H2PO4. The phosphate supplywas discontinued and the subsequent effects on growth, internalphosphorus concentrations, phosphate absorption and translocationwere measured at frequent intervals. Growth rates were at firstunchanged and the internal phosphorus concentration decreased.During this phase the rate of phosphate transport by the rootssometimes increased significantly. Growth slowed more in shootsthan in roots during a second phase of stress development andvisual symptoms of deficiency appeared in tomato but not inbarley. During this phase, enhancement of phosphate uptake capacityreached a maximum in both species. The subsequent decline inuptake capacity was associated with visible symptoms of deficiencydeveloping in barley and intensifying in tomato. When stressedplants were returned to a solution containing 0.15 mol m–3H2PO4 rapid absorption continued for several days afterthe internal phosphorus concentration had returned to the levelof the controls. Phosphate toxicity may have been the causeof leaf lesions and necrosis during the ‘recovery’phase. Stomatal conductance in tomato was decreased at an early stageof stress development. Foliar-applied phosphate was absorbedmore rapidly by P-stressed barley leaves than by their controlsand much larger amounts were translocated from the leaves tothe roots.  相似文献

The Uptake of Gaseous Ammonia by the Leaves of Italian Ryegrass   总被引:5,自引:0,他引:5  
Lockyer, D. R. and Whitehead, D. C. 1986. The uptake of gaseousammonia by the leaves of Italian ryegrass.—J. exp. Bot.37: 919–927. Plants of Italian ryegrass (Lolium multiflorum Lam.) grown insoil with two rates of added 15N-labelled nitrate were exposed,in chambers, for 40 d to NH3 in the air at concentrations of16, 118 and 520 µg m–3. At the highest concentrationof NH3, this source provided 47?3% of the total nitrogen inplants grown with the lower rate of nitrate addition (100mgN kg–1 dry soil) and 35?2% with the higher rate (200mgN kg–1 dry soil) At the intermediate concentration ofNH3, the contributions to total plant N were 19?6% and 10?8%,respectively, at low and high nitrate while, at the lowest concentrationof NH3, they were 5?1% and 32%. Most of the N derived from theNH3 remained in the leaves, but some was transported to theroots. The amount of N derived from the NH3 that was presentin the leaves was not reduced by washing the leaves in waterat pH 5?0 before harvesting, indicating that the N was assimilatedby the plant and not adsorbed superficially. Rates of uptakeof NH3 per unit leaf area ranged from 1?7 µg dm–2h–1 at a concentration of 16 µg m–3 to 29?0µg dm–2 h–1 at a concentration of 520 µgm–3 and with the lower rate of nitrate addition. Increasingthe supply of nitrate to the roots slightly reduced the rateof uptake of NH3 per unit leaf area. Bot. 38: 1937–1948. Associations between photosynthesis, the activation state ofRuBP carboxylase and the contents of photosynthetic intermediateswere compared in soybean and wheat leaves before and after exposureto photoinhibitory treatments in the presence of atmosphericO2. Exposing attached leaves to a supra-saturating irradiance(3 800 µmol quanta m– 2 s–1) for 2 h in CO2-freeair decreased carboxylation efficiency and the light-saturatedphotosynthetic rate in air by approximately 50%. Exposure tothe photoinhibitory treatment for periods in excess of 2 h didnot cause a further decrease of photosynthesis in soybean. Althoughphotosynthesis was reduced, the initial and total (fully-activated)activities of ribulose 1,5-bisphosphate carboxylase (RuBPCase)in leaf extracts were unaltered in each species by the photoinhibitorytreatment. This was true for leaves sampled under both air andat a rate-limiting intercellular CO2 partial pressure (Ci) of75 µPa Pa–1. The contents of ribulose l,5-bisphosphate(RuBP) and 3-phosphoglyceric acid (3-PGA) were reduced by thephotoinhibitory treatment in soybean leaves sampled in air andat a rate-limiting Ci, although the RuBP/3-PGA ratio was unaffected.The relative reduction of RuBP content in soybean leaves atrate-limiting C1 was similar to the corresponding reductionof carboxylation efficiency. For wheat,the relative reductionof RuBP content at rate-limiting Ci (–19%) caused by thephotoinhibitory treatment was considerably less than the correspondingdecrease of carboxylation efficiency (–49%).The RuBP/3-PGAratio of wheat was also increased significantly by the photoinhibitorytreatment The significance of these observations to the regulationof CO2-limited photosynthesis in leaves experiencing photoinhibitionunder atmospheric oxygen is discussed. Consideration is alsogiven to the previous contention that contemporary measurementsof initial activity in crude extracts may provide a spuriousindication of the amount of the enzyme-CO2-Mg2 + form of RuBPcarboxylase present in the leaf. The influence of salinity in the growing media on ribulose-1,5-bisphosphate (RuBP) carboxylase and on CO2 fixation by intactsugar beet (Beta vulgaris) leaves was investigated. RuBP carboxylase activity was mostly stimulated in young leavesafter exposure of plants for 1 week to 180 mM NaCl in the nutrientsolution. This stimulation was more effective at the higherNaHCO2 concentrations in the reaction medium. Salinity also enhanced CO2 fixation in intact leaves mostlyat rate-limiting light intensities. A 60 per cent stimulationin CO2 fixation rate was obtained by salinity under 450 µEm–2 s–1. At quantum flux densities of 150 µEm–2 s–1 (400–700 nm) this stimulation was280 per cent. Under high light intensities no stimulation bysalinity was found. In contrast, water stress achieved by directleaf desiccation or by polyethylene glycol inhibited enzymeactivity up to fourfold at –1.2 MPa. Sink-limited conditions, defined as treatment with continuousillumination, cause a reduction in the rate of photosyntheticfixation of CO2 in single-rooted leaves of soybean (Glycinemax. Merr. cv. Turunoko). We suggested previously that thisreduction is due to a deactivation of ribulose-1,5-bisphosphatecarboxylase (RuBPcase, E.C. 4.1.1.39 [EC] ) that is caused by a decreasein the level of Pi in the leaves [Sawada et al. (1989) PlantCell Physiol. 30: 691, Sawada et al. (1990) Plant Cell Physiol.31: 697]. In the present study, the mechanism of regulationof RuBPcase activity by Pi was examined. The activity of RuBPcasein the sink-limited leaves, exposed for 6 or 7 d to continuousillumination to alter the source/sink balance, was enhancedwith increasing concentrations of Pi, in a CO2-free preincubationmedium in the presence of 5 mM MgCl2 The maximum value [6.3µmole CO2 (mg Chl)–1 min–1] was obtained atapproximately 5 mM Pi after a 5 min incubation, being 3 timesof the activity without the preincubation. The activity of acrude preparation of RuBPcase that had been deactivated by removalof CO2 and Mg2+ ions by the gel filtration was 5.2–9.3nmole CO2 (mg protein)–1 min–1 and was also enhancedby Pi plus Mg2+ ions. The maximum value [147–151 nmoleCO2 (mg protein)–1 min–1] was attained at 5 mM Piafter a 5 min incubation. The cycle of activation and inactivationof deactivated crude RuBPcase was perfectly reversible by additionof Pi to the enzyme and removal of Pi from the enzyme. Levelsof free Pi and of esterified phosphate in the sink-limited leaveswere 69% and 31% of the total phosphate, respectively. By contrast,in the control leaves, these values were 87% and 13%, respectively.These results support our previously stated hypothesis and indicatean important role for free Pi in the regulation of RuBPcaseactivity, in particular in sink-limited plants. The effect of heat stress on processes related to carbohydratepartitioning was investigated in young bell pepper (Capsicumannum L. cv. Maor) plants in relation to abscission of theirreproductive organs at different stages of development. None of the reproductive organs abscised after 5 d in a normalday/night temperature regime (25/18°C). With a temperatureregime of 35°C day, 25°C night, abscission occurredin only a small portion of the flower buds and none of the flowersand fruitlets. However, when temperatures in the day and nightwere reversed (25/35°C, day/night) all the buds and someof the flowers abscised during that time period. The young fruitat the first node did not abscise under any temperature regime.The abscission rate of the flower buds was reduced under heatstress if the developing fruit at the first node had been removed. High temperature during either the light or dark periods reducedthe export of [14C]sucrose from the source leaf (fed for 48h with [14C]sucrose). Both heat stress and fruit presence reduced the relative amountof [14C]sucrose which was exported to the flower buds, flowersand roots. Likewise, these treatments reduced the concentrationof reducing sugars in the reproductive organs. Concomitantly,the heat stress and fruit presence on the first node reducedthe activity of soluble acid invertase in the flower buds andthe roots, but not in young leaves. Overall, the results show that heat stress causes alternationin sucrose distribution in the plant, but may also have specificeffects on metabolic activities related to sucrose import andutilization in flower buds and flowers which in turn may enhancetheir abscission. The extent of starch turnover in pepper leaves in the lighthas been estimated. After leaves were labelled with 14CO2 atconstant specific activity for 4–7 h, the irradiance wasreduced to a level which caused no net change in the starchcontent of the leaf, and the supply of 14CO2 was removed. Therewas no significant change in specific activity of starch overthe following 6–10 h, thus there was no exchange of 14C-starchwith 12C-assimilates entering the chloroplasts. Carbon Partitioning and Export in Mature Leaves of Pepper (Capsicum annuum)   总被引:1,自引:0,他引:1  
The partitioning of recently fixed carbon by mature pepper leaveshas been examined over a 10 h photoperiod using a constant specificradioactivity 14CO2 labelling technique. Changes in the ratesof carbon partitioning into export, starch, sucrose and hexoseswere examined following changes in irradiance during the photoperiod.Leaves grown under 80 W m–2 PAR were exposed to this irradiancefor the first 4 h of the photoperiod then the iiradiance wasdecreased. Leaves accumulated sufficient reserves in the first4 h to maintain export at the initial rate (approximately 20µg carbon cm–2 leaf h–1) over the following6 h of the photoperiod when the net photosynthesis rate (Pn)was decreased to 10% of the initial rate by the decreased irradiance.Export was initially maintained by the depletion of sucroseand hexose and then by carbon from the degradation of starchin the light. If leaves were exposed to low irradiance at the beginning ofthe photoperiod, then the export rate was linearly related tothe Pn during that period. When Pn exceeded that required tomaintain an export rate of approximately 20 µg carboncm–2 h–1, then more carbon was partitioned intostarch. At low initial irradiance, a greater proportion of photosynthatewas partitioned into export rather than starch and at high initialirradiancc the reverse occurred. There was a linear relationship between starch accumulationrate and Pn for all leaves but the relationship between Pn andexport rate was only significant for leaves with low levelsof reserve carbon. The results show that mature pepper leaves subjected to differentirradiances maintain constant export rates through alterationsof carbon partitioning. Export at low Pn is maintained at theexpense of sugar and starch reserves, with partitioning in highirradiance being predominantly to starch. Wolterbeek, H. Th. and De Bruin, M. 1986. The import and redistributionof several cations and anions in tomato leaves.—J. exp.Bot. 37: 331–340. The upward movements in the xylem and redistribution from theleaf of Na+ , K+ , Rb+, Cs+ and four anions were examined insub-systems of tomato plants (Lycopersicon esculentum, Mill.cv. Tiny Tim). There was a delay with respect to the redistributionof newly imported elements from the source leaf of about 16–20h for all four alkali ions. This is considerably less than theapparent delay for the anions Sb(SO4) WO42– Mo7O246–and AsO43– The prolonged delay for the anions is suggestedto be a consequence of metabolic transformation in the leaf.Reduction of the source-sink activity ratio did not decreasethe delay period from the source leaf, but apparently causedincreased Na+ transfer from the xylem. It is concluded thatthe application of a detailed mathematical descnption of upwardelement movement has considerable potential possibilities forunderstanding circulation of nutrients in the plant. It is known that phosphorus deficiency results in alterationsin the assimilation of nitrogen. An experiment was conductedto investigate mechanisms involved in altered 15NO3 uptake,endogenous 15N translocation, and amino acid accumulation insoybean (Glycine max L. Merrill, cv. Ransom) plants deprivedof an external phosphorus supply for 20 d in solution culture.Phosphorus deprivation led to decreased rates of 15NO3uptake and increased accumulation of absorbed 15N in the root.Both effects became more pronounced with time. Asparagine, theprimary transport amino acid in soybean, accumulated in largeexcess in roots and stems. In roots of phosphorus-deprived plants,concentrations of ATP and inorganic phosphate declined rapidly,but dry weight accumulation was similar to or above that ofthe control even after 20 d of treatment. Arginine accumulationin leaves was greatly enhanced, even though 15N partitioninginto the insoluble reduced-N fraction of leaves was unaffected.The results suggest that decreases in NO3 uptake in lowphosphorus plants could be caused by feedback control factorsand by limited ATP availability. The decline in endogenous Ntransport from the root to the shoot may be associated withchanges in membrane properties, which also result in paralleleffects on hydraulic conductance and the upward flow of waterthrough the plant. Plants of Lupinus albus L., cv. Ultra, were grown hydroponicallywith NO3-nutrition for 51 d under control (0.05 mol m–3Na+ and 10 mol m–3 Cl) and saline (40 mol m–3NaCI) conditions. Plants were harvested 41 and 51 d after germinationand analysed for content and net increment of C, N and the mineralcations K+, Na+, Mg2+, and Ca2+ and the anions Cl, NOJ,malate, phosphate, and SO42–. Roots, stem interaodes,petioles and leaflets were analysed separately. During the studyperiod net photosynthesis, respiratory losses of CO2 from shootand root and the composition of the spontaneously bleeding phloemsap and the root pressure xylem exudate were also determined.Using molar ratios of C over N in the transport fluids, incrementsof C and N, and photosynthetic gains as well as respiratorylosses of C, the net flows of C and N in the xylem and phloemwere then calculated as in earlier studies (Pate, Layzell andMcNeill, 1979a). Knowing the carbon flows, the ratios of ionto carbon in the phloem sap, and ion increments in individualorgans, net flows of K+, Na+, and Cl over the study periodwere also calculated. Salt stress led to a general decrease of all partial componentsof C and N partitioning indicating that inhibitions were notdue to specific effects of NaCI salinity on photosynthesis oron NO3 uptake. However, there were differences between variouslyaged organs, and net phloem export of nitrogenous compoundsfrom ageing leaves was substantially enhanced under saline conditions.In addition, NO3reduction in the roots was specificallyinhibited. Uptake and xylem transport of K+ was more severelyinhibited than photosynthetic carbon gain or NO3 uptakeby the root. K+ transport in the phloem was even more severelyrestricted under saline conditions. Na+ and Cl flowsand uptake, on the other hand, were substantially increasedin the presence of salt and, in particular, there were thenmassive flows of Na in the phloem. The results are discussedin relation to the causes of salt sensitivity of Lupinus albus.The data suggest that both a restriction of K+ supply and astrongly increased phloem translocation of Na+ contribute tothe adverse effects of salt in this species. Restriction ofK+ supply occurs by diminished K+ uptake and even more by reducedK+ cycling within the plant. Spinach chloroplasts catalyzed ATP formation from acetyl phosphateand ADP when exposed to light. No ATP formation was detectablein the dark. In the absence of ADP, chloroplasts did not hydrolyzeacetyl phosphate in the light or dark. Neither high-energy phosphatessuch as creatine phosphate and phosphoenol pyruvate nor inhibitorsof photophosphorylation competitive with Pi, such as ß-naphthylmonophosphate, phenyl phosphate and pyridoxal 5-phosphate, couldsubstitute for acetyl phosphate as a Pi donor. The apparentKm values for acetyl phosphate and Pi were 0.81 mM and 0.25mM, respectively. The maximal rate of ATP formation with acetylphosphate and Pi were 331 and 521 µmol ATP formed mg chl–1hr–1, respectively. The optimum pH value for acetyl phosphate-dependentATP formation was about 8.0. NH4Cl, dicyclohexylcarbodiimideand triphenyltin chloride inhibited the acetyl phosphate-dependentATP formation. Acid-base transition also could induce subsequentATP formation from acetyl phosphate and ADP. Osmotic stress temporarilyreverses the inhibitions of photosynthesis and stomatal conductanceby abscisic acid—evidence that abscisic acid induces alocalized closure of stomata in intact, detached leaves.—J.exp. Bot 39: 147–155. The influence of osmotic stress on whole leaf gas exchange wasmonitored in detached leaves of Glycine max supplied with anexogenous concentration (10–5 mol dm–3) of ±abscisicacid (ABA) sufficient to inhibit net photosynthesis and stomatalconductance by 60% and 70%, respectively, under a saturatingirradiance and normal air. Raising the osmotic (sorbitol) concentrationof the ABA solutions feeding leaves elicited rapid and synchronousreversals of the ABA-dependent inhibitions of net photosynthesisand conductance. These reversals reached a peak simultaneously,after which photosynthesis and conductance declined. The magnitudeof the transient stimulations at peak height was dependent uponthe sorbitol concentration of the ABA feeding solution, althoughthe time-course of the transients (half time, 4–6 min)was similar for the different osmotic concentrations applied.Irrespective of transient size the relative changes of photosynthesisand conductance were comparable; consequently the calculatedpartial pressure of CO2 in the substomatal space (Ci) remainedrelatively constant during the transient phase. In contrastto the ABA-treated leaves, elevating the osmotic concentrationof the distilled water supply feeding control leaves stimulatedconductance to a much greater relative extent than photosynthesis.The co-stimulations of photosynthesis and conductance inducedin ABA-treated leaves by osmotic shock were not due to a restrictionin the transpirational uptake of ABA and occurred irrespectiveof the source osmoticum applied. These data are consistent with the hypothesis that the ABA-dependentinhibition of photosynthesis at constant Ci is an artifact causedby the spatially heterogeneous closure of stomata in responseto ABA. Barley varieties are known to differ in the extent of Na+ andCl accumulation in leaves when grown in saline soil orhydroponic culture. In particular, the cv. Chevron accumulatesmore Na+ than the more salt-tolerant cv. CM67, and has lowerleaf K+ concentrations. When salt was applied as a spray tothe leaves, CM67 accumulated more Na+ than Chevron, and theselection Sinis 27 (from a landrace collected on the Sinis Peninsulaof Sardinia) accumulated more Na+ than Sinis 28. In some casesleaf K+ concentrations decreased in response to high concentrationsof salt sprayed on to the leaves. Accumulation of Na+ was greaterin the 4th leaf than in the flag leaf. Added CaCI2 had oppositeeffects when added to the salt applied to the soil or to thesaline spray. In the soil, CaCI2 reduced Na+ uptake; appliedto the leaf it increased Na+ uptake. Pre-wetting the leavesbefore the salt spray, or washing the leaves with non-salinewater 1 h after the salt spray, reduced the uptake of Na+ andCl. Avalon) was grown undersimulated autumn conditions for 4 weeks and exposed to a mixtureof SO2 and NO2. Biomass was measured after 2, 3 and 4 weeksand the flag leaves of sample plants were labelled with 14CO2.Biomass yields revealed an increase in shoot-to-root ratiosunder polluted conditions. The labelling experiments showedthat less assimilate was transported to the roots, whilst morewas allocated to the younger components of the plant. Tobacco shoots were grown in vitro for 35 d, in MS culture mediummodified to include various sources (nitrate-N, ammonium-N ora mixture) and levels (0–120 mM) of N, and in the presenceof 0–180 mM NaCI or iso-osmotic concentrations of mannitol.Growth of control plantlets was significantly inhibited whenNH4+-N was the sole N source, and at high (120 mM) NO3-N supply. Under conditions of salt stress (90 and 180 mM NaCI)growth was repressed, with roots being more severely affectedthan shoots. Salinity also inhibited root emergence in vitro.The only alleviation of the salt stress by nitrate nutritionobserved in this study was on shoot growth parameters of plantletsgrown on 60 mM NO3-N and 90 mM NaCI. Although both weresignificantly inhibited by NaCI, nitrate reduc-tase activitywas more severely affected than nitrate uptake. When mannitolreplaced NaCI in the culture medium, similar Inhibition of growth,nutrient uptake and enzyme activity were recorded. These observations,together with the relatively low recorded values for Na+ andCI uptake, indicate that under in vitro salt stress conditionsthe negative effects of NaCI are primarily osmotic. The levels of purine and pyrimidine nucleotides in suspensioncultures of Catharanthus roseus were determined 24 h after stationary-phasecells were transferred to fresh complete (‘+Pi’)or phosphate-deficient (‘–Pi’) Murashige-Skoogmedium. The levels of ATP, GTP, UTP and CTP were from approx.3 to 5-fold greater in the cells grown in ‘+Pi’medium than in the cells grown in ‘–Pi’ medium.The levels of almost all other nucleotides were slightly higherin the cells in ‘+Pi’ medium. The rates of de novoand salvage biosynthesis of purine and pyrimidine nucleotideswere estimated from the rates of incorporation of radioactivityfrom [14C]formate, [2–14C]glycine, NaH14CO3, [6–14C]orotate,[8–14C]adenine, [8–14C]adenosine, [2–14C]uraciland [2–14C]uridine. The results indicated that the activityof both the de novo and the salvage pathway was higher in thecells in ‘+Pi’ medium than in the cells in ‘–Pi’medium. The rate of degradation estimated from the rate of releaseof 14CO2 from labelled purines and pyrimidines indicated thatdegradation of uridine was significantly reduced in the cellsin ‘+Pi’ medium, but no significant difference wasfound in the degradation of adenine, adenosine and uracil. Thepossible role of Pi in the control of the biosynthesis of nucleotidesand in the degradation of uridine is discussed. Photosynthetic acclimation was examined by exposing third trifoliolateleaves of soybeans to air temperatures of 20 to 30°C andphotosynthetic photon flux densities (PPFD) of 150 to 950µmolphotons m–2 s–1 for the last 3 d before they reachedmaximum area. In some cases the environment of the third leafwas controlled separately from that of the rest of the plant.Photosynthesis, respiration and dry mass accumulation were determinedunder the treatment conditions, and photosynthetic capacity,and dry mass and protein content were determined at full expansion.Photosynthetic capacity, the light-saturated rate of net carbondioxide exchange at 25°C and 34 Pa external partial pressureof carbon dioxide, could be modified between 21 and 35 µmolCO2 m–2 s–1 by environmental changes after leaveshad become exporters of photosynthate. Protein per unit leafmass did not differ between treatments, and photosynthetic capacityincreased with leaf mass per unit area. Photosynthetic capacityof third leaves was affected by the PPFD incident on those leaves,but not by the PPFD on other leaves on the plant. Photosyntheticcapacity of third leaves was affected by the temperature ofthe rest of the plant, but not by the temperature of the thirdleaves. Photosynthetic capacity was linearly related to carbondioxide exchange rate in the growth regimes, but not to daytimePPFD. At high PPFD, and at 25 and 30°C, mass accumulationwas about 28% of the mass of photosynthate produced. At lowerPPFD, and at 20°C, larger percentages of the photosynthateproduced accumulated as dry mass. The results suggest that photosynthatesupply is an important factor controlling leaf structural growthand, consequently, photosynthetic acclimation to light and temperature.
