Enzymatic removal of hemicellulose from dissolving pulps   总被引:3,自引:0,他引:3  
Hemicellulases and an endoglucanase from seven different fungi were assessed for their potential to solubilze mannan and xylan from softwood sulfite dissolving pulps. A xylanase from Thermomyces lanuginosus and a mannanase from Sclerotium rolfsii acted synergistically on the pulp solubilizing 50% more mannan and 11% more xylan than did the individual enzymes. The addition of an endoglucanase further increased both the amount of xylan and mannan that was solubilized from the pulp. The aim of this work was to evaluate the response to biobleaching of steam exploded kraft pulps and to compare the results with the controls. For this end, a laccase-mediator treatment using commercial laccase (Novozyme 51003) and a natural mediator (acetosyringone) were assayed, followed by alkaline extraction and hydrogen peroxide stages.Our approach resulted in exploded biobleached pulps with lower kappa number and improved optical properties compared to controls, even after subjecting pulps to accelerated ageing. Additionally, use of hydrogen peroxide was reduced. The LMS (laccase-mediator system) had a smaller impact on the properties of the bleached pulps and on hydrogen peroxide consumption than the steam explosion process did.  相似文献

Anionic polymer sodium carboxymethylcellulose (CELLOGEN® HP-HS and/or HP-12HS) was investigated for its ability to influence the release of three model drugs propranolol hydrochloride, theophylline and ibuprofen from polyethylene oxide (POLYOX™ WSR 1105 and/or Coagulant) hydrophilic matrices. For anionic ibuprofen and non-ionic theophylline, no unusual/unexpected release profiles were obtained from tablets containing a mixture of two polymers. However, for cationic propranolol HCl, a combination of polyethylene oxide (PEO) with sodium carboxymethylcellulose (NaCMC) produced a significantly slower drug release compared to the matrices with single polymers. The potential use of this synergistic interaction can be a design of new extended release pharmaceutical dosage forms with a more prolonged release (beyond 12 h) using lower polymer amount, which could be particularly beneficial for freely water-soluble drugs, preferably for once daily oral administration. In order to explain changes in the obtained drug release profiles, Fourier transform infrared absorption spectroscopy was performed. A possible explanation for the more prolonged propranolol HCl release from matrices based on both PEO and NaCMC may be due to a chemical bond (i.e. ionic/electrostatic intermolecular interaction) between amine group of the cationic drug and carboxyl group of the anionic polymer, leading to a formation of a new type/form of the active (i.e. salt) with sustained release pattern.Key words: extended release, FT-IR, ibuprofen, matrix tablet, polyethylene oxide, polymer combination, propranolol hydrochloride, sodium carboxymethylcellulose, theophylline  相似文献

Use of biotechnology in pulp bleaching has attracted considerable attention and achieved interesting results in recent years. Enzymes of the hemicellulolytic type, particularly xylan-attacking enzymes, xylanases are now used commercially in the mills for pulp treatment and subsequent incorporation into bleach sequences. The aims of the enzymatic treatment depend on the actual mill conditions and may be related to environmental demands, reduction of chemical costs or maintenance or even improvement of product quality. The use of oxidative enzymes from white-rot fungi, that can directly attack lignin, is a second-generation approach, which could produce larger chemical savings than xylanase but has not yet been developed to the full scale. It is being studied in several laboratories in Canada, Japan, the U.S.A. and Europe. Certain white-rot fungi can delignify kraft pulps increasing their brightness and their responsiveness to brightening with chemicals. The fungal treatments are too slow but the enzyme manganese peroxidase and laccase can also delignify pulps and enzymatic processes are likely to be easier to optimize and apply than the fungal treatments. Development work on laccase and manganese peroxidase continues. This article presents an overview of developments in the application of hemicellulase enzymes, lignin-oxidizing enzymes and white-rot fungi in bleaching of chemical pulps. The basic enzymology involved and the present knowledge of the mechanisms of the action of enzymes as well as the practical results and advantages obtained on the laboratory and industrial scale are discussed.  相似文献

The ability of a crude enzyme preparation ofAureobasidium pullulans containing xylanase [61 units (U)/ml] and xylosidase (3 U/ml) activity to remove pentosans from unbleached sulphite pulps was investigated. Greater amounts of pentosans and reducing sugars were released from the pulp when the enzyme dosages and incubation times were increased. A combination of enzyme hydrolysis and alkali extraction resulted in a greater removal of pentosans than using the enzyme preparation alone. By treatment with a xylanase loading of 450 U/g pulp for 24 h followed by alkaline extraction, 35% of the pentosans were removed. The kappa number decreased up to 30% whereas viscosity was only slightly affected by these treatments. Enzymatic hydrolysis released mainly xylose whereas xylobiose was the main product liberated by alkaline extraction. Scanning electron micrographs indicated improved fibrillation and flexibility of the fibre structure by enzyme treatment.  相似文献

Fungal laccases in the presence of mediators are powerful biocatalysts to degrade lignin. Pycnoporus cinnabarinus laccase and 1-hydroxybenzotriazole (HBT) have been successfully used to delignify eucalypt kraft pulp once integrated in a totally chlorine-free bleaching sequence. Real time delignification of kraft pulp by laccase-HBT was verified in situ by monitoring the loss of lignin autofluorescence during the enzymatic treatment using confocal laser scanning microscopy. The highest delignification of pulp fibers occurred over a very short time-span (5 min). Moreover, we demonstrate the removal of sterols, responsible for pitch deposits in hardwood kraft pulps, as an additional effect of laccase-HBT. Spherical structures between pulp fibers localized by low temperature scanning electron microscopy were removed by laccase-HBT. The use of filipin, a specific stain, revealed the sterol nature of many of these structures. At the end of the enzyme-aided bleaching sequence, the fluorescent sterols-filipin signals were almost completely absent.  相似文献

Summary Anaerobic digestion of sugar beet pulps was studied in a 70 l digestor with sequential feeding, after enzymatic hydrolysis by Trichoderma harzianum cellulases. During the 130 days feeding, 3.6 m3 of biogas were produced with an average content of 58% CH4 from 270 l of hydrolysed pulps at 20 g VS/l. Average yield and production rate were respectively 0.67 m3/kg VS and 0.4 m3/ kg VS and 0.4 m3/m3 of digestor per day.  相似文献

A total of 179 non-spore-forming bacteria aerobically growing on Nutrient Agar, Plate Count Agar or in specific enrichment conditions for salmonella, campylobacteria, listeria, yersinia or staphylococci, were isolated from 16 untreated paper mill pulps. After phenotypical screening the isolates were characterised by automated ribotyping and partial sequencing of the 16S rRNA gene. They could be divided into seven taxonomical classes representing 63 taxa (species): actinobacteria (11 species), bacilli (7), flavobacteria (3) alphaproteobacteria (10), betaproteobacteria (5), gammaproteobacteria (25) and sphingobacteria (2). Most of the gammaproteobacteria were enterobacteria, mainly species of the genera Enterobacter (7 species, 7 samples/3 mills) and Klebsiella (5 species, 6 samples/3 mills). Other commonly occurring bacteria were most closely related to Microbacterium barkeri (7 samples/3 mills), Cloacibacterium normanense (6 samples/2 mills), Pseudoxanthomonas taiwanensis (5 samples/2 mills) and Sphingobacterium composti (5 samples/1 mill). Sporadic isolates of Listeria innocua, L. monocytogenes, Enterococcus casseliflavus and Staphylococcus warneri were detected, from which only L. monocytogenes is considered to be a food pathogen. No isolates of the genera Campylobacter, Salmonella or Yersinia were detected. The detected bacteria may be harmful in process control, but the load of food pathogens with recycled fibres to paper machines is insignificant. Faecal contamination of the pulp samples was not indicated.  相似文献

When purified xylanases from Trichoderma harzianum E58 or from a clone of Bacillus circulans were incubated with various low-yield wood pulps, little of the original enzyme activity could be detected in the filtrate at the end of the reaction. Partial bleaching of the pulps prior to enzymatic treatment generally resulted in an increased recovery of the xylanase activity. It appears that both nonspecific adsorption and soluble inhibitors may be responsible for the loss of much of the xylanase activity. However, xylanases from Aureobasidium pullulans and Schizophyllum commune were not as inhibited by the pulps, and the activity of the latter enzyme actually increased after incubation with several high-yield pulps. Although a lignin preparation from spent sulfite liquor at a concentration of 0.06 mg/mL could inhibit the xylanase activity of T. harzianum and B. circulans by 65% and 50%, respectively, xylanases from Thermoascus aurantiacus, S. commune, and A. pullulans were activated at similar lignin concentrations. At higher concentrations these latter xylanases were also inhibited. Water-soluble lignins extracted from a variety of pulps and used at a lignin concentration of 2.5 mug/mL resulted in inhibition of more than 65% of the original activity of the xylanase from T. harzianum. Kinetic studies showed that lignin from spent sulfite liquor resulted in noncompetitive inhibition of this enzyme.  相似文献

The delignification of chemical pulps with laccase and -hydroxybenzotriazole was explored employing a pre- and post-O2 delignified softwood draft pulp. The delignification properties of laccase were shown to be improved with -hydroxybenzotriazole was used as a mediator instead of 2,2′-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid). Analysis of the structure of residual lignin before and after laccase/ -hydroxybenzotriazole treatment indicated that the biobleaching system oxidizes the phenolic component of lignin and that the residual lignin is demethylated and significantly enriched in carboxylic acid groups.  相似文献

Sixteen strains of the new yeast species Candida sergipensis have been isolated from frozen pulps of the tropical fruits umbú ( Spondias tuberosa Avr. Cam.) and mangaba ( Hancornia speciosa Gom.). Candida sergipensis was one of the prevalent species in the yeast community of these substrates. The new asexual ascomycetous yeast is phylogenetically related to Candida spandovensis and Candida sorbophila, species belonging to the Wickerhamiella clade, as evidenced by the sequences of the D1/D2 domains of their large subunit ribosomal DNAs. The species C. sergipensis and C. spandovensis can be separated on the basis of growth on 50% glucose agar, xylose and succinate, negative for the first species and positive for the second. The type culture is strain UFMG-R188 (CBS 9567).  相似文献

A chemically defined media consisting of carboxymethylcellulose (CMC) was developed to maximize the production of antibiotics, hexaene H-85 and azalomycine, by Streptomyces hygroscopicus CH-7. The production of antibiotics by filamentous organisms is often dependent on the morphology and size distribution of the pellet population within the culture. By adding the polymer to the fermentation medium, the growth was changed from a single large glob to small reproducible pellets, and wall growth was diminished to a minimum. Maximum concentrations of hexaene H-85 (146.7 mg/dm(3)) and azalomycine (188.6 mg/dm(3)) were reached at 3.0% and 1.0% (w/v) CMC, respectively.  相似文献

Antimicrobial-modified sulfite pulps prepared by in situ copolymerization   总被引:1,自引:0,他引:1  
Grafting guanidine polymer (PHGH) onto cellulose fibers was conducted via in situ free-radical polymerization using ceric ammonium nitrate (CAN) as an initiator. The optimum reaction conditions were obtained, under which the grafting percentage and the grafting efficiency reached over 20% and 50%, respectively. Atomic force microscopy (AFM) images revealed that the grafted polymer tended to form grains with diameters ranging from 60 to 200 nm. AFM also enabled us to identify the location of the grafts on the surfaces of cellulose fibers by the measurements of the adhesion and attraction forces between a colloid probe and the samples. The cellulose fibers were rendered antimicrobial in the presence of 1.0% (wt) grafted polymer, and an excellent antimicrobial activity (over 99% inhibition) toward Escherichia coli was achieved. The AFM results also demonstrated that the antimicrobial mechanism of PHGH is to destroy the membrane of the cells.  相似文献

Activation of fibres by radical formation is the first step when aiming at oxidative coupling of new functional groups on the fibre bound lignin. In this work, factors affecting the amount of phenoxy radicals created to unbleached TMP, CTMP, softwood kraft and hardwood kraft pulp fibres in the laccase catalysed oxidation were determined by EPR. Laccase was able to catalyse the oxidation of all the pulps studied. The reactivity of the pulp was found to be affected by both the physical accessibility of lignin in the fibres and the chemistry of the surface lignin accessible to laccase. Laccase dosage, use of extra oxygen in the laccase catalysed radicalization reaction, treatment time and also the amount and type of low-molecular weight compounds (LMWC) present in the pulp were all found to contribute to the radical content of pulp fibres measured after the enzymatic reaction. It could not been excluded that two types of reactions take place during the radical formation in fibres. Within the fibre matrix there may be both fibre material bound and soluble lignin fragments differing with respect to accessibility, molecular weight or chemical structure which can be radicalized at various rates, and the formed radicals may also undergo cross-coupling reactions reducing the amount of the total radicals.  相似文献

The composition of pitch deposits occurring in pulp sheets and mill circuits during soda/anthraquinone pulping and elemental chlorine-free pulp bleaching of bast fibers of industrial hemp (Cannabis sativa) was studied. Pitch deposits were extracted with acetone, and the extracts analyzed by gas chromatography (GC) and gas chromatography/mass spectrometry (GC/MS). Acetone extracts (15-25% of pitch deposits) were constituted by the defoamers used at the mill and by lipophilic extractives from hemp fibers. Acetone-insoluble residues (75-85% of pitch deposits) were analyzed by pyrolysis-GC/MS in the presence and absence of tetramethylammonium hydroxide. These residues were constituted by salts of fatty acids (arising from hemp fibers) with calcium, magnesium, aluminum and other cations that were identified in the deposits. It was concluded that inappropriate use of defoamer together with the presence of multivalent ions seemed to be among the causes of hemp extractives deposition in the pitch problems reported here.  相似文献

A xylanase from Aspergillus kawachii, active at pH 2.0–2.5, was found to be suitable for improvement of bleachability. Due to the low working pH of the xylanase, the metal cations were also removed and thus the metal removal stage using chelating agents could be omitted. The use of acid-tolerant xylanase is especially beneficial prior to ozone or chlorine dioxide bleaching stages due to the minimization of pH adjustment steps during bleaching.  相似文献
