In Dunaliella tertiolecta, D. bioculata and D. viridis the activitiesof phosphoenolpyruvate carboxylase and carbonic anhydrase werehigher in the cells grown in ordinary air (low-CO2 cells) thanin those grown in air enriched with 1–5% CO2 (high-CO2cells), whereas in Porphyridium cruentum R-1 there was no differencein phosphoenolpyruvate carboxylase activity between these twotypes of cells. Apparent Km(NaHCO3) values for photosynthesisin low-CO2 cells of all species tested were smaller than thosein high-CO2 cells. Most of the 14C was incorporated into 3-phosphoglycerate,sugar mono- and di-phosphates during the initial periods ofphotosynthetic NaH14CO3 indicating that both types of cellsin D. tertiolecta are C3 plants. The effect of carbonic anhydrase (CA) on time courses of photosynthetic14C incorporation in the presence of 14CO2 or NaH14CO3 was studiedwith cells of Chlamydomonas reinhardtii which had been grownunder ordinary air (low-CO2 cells) or air enriched with 4% CO2(high-CO2 cells). Experimental data obtained at 20°C andpH 8.0 suggested that the major form of inorganic carbon utilizedby high-CO2 cells was CO2, while that utilized by low-CO2 cellswas HCO3. The cell suspension showed CA activity which was comparableto that observed in the sonicate of cells. Both activities werehigher in low-CO2 cells than in high-CO2 cells. Photosynthetic 14C fixation by Characean cells in solutionsof high pH containing NaH14CO3 gave a measure of the abilityof these cells to take up bicarbonate (H14CO3). Whereascells of Nitella translucens from plants collected and thenstored in the laboratory absorbed bicarbonate at 1–1.5µµmoles cm–2 sec–1, rates of 3–8µµmoles cm–2 sec–1 were obtained withN. translucens cells from plants grown in the laboratory. Influxesof 5–6 µµmoles cm–2 sec–1 wereobtained with Chara australis, 3–8 µµmolescm–2 sec–1 with Nitellopsis obtusa, and 1–5µµmoles cm–2 sec–1 with Tolypella intricata.It is considered that these influxes represent the activityof a bicarbonate pump, which may be an electrogenic process. In solutions of lower pH, H14CO3 uptake would be maskedby rapid diffusion of 14CO2 into the cells: the four Characeanspecies fixed 14CO2 at maximum rates of 30–40 µµmolescm–2 sec–1 (at 21° C).  相似文献

A study was made of the incorporation of 14C by intact leavesof Coffea arabica (cultivars Mundo Novo, Catuai, 1130–13,and H 6586–2) and Coffea canephora (cultivar Guarini)supplied with gas mixtures containing 14CO2 under controlledconditions. Samples of the leaves were combusted and the 14Cin the CO2 produced measured using a liquid scintillation counter.The results were used to estimate photosynthetic rates. Theeffects of changing the partial pressures of O2 and CO2 on thephotosynthetic rate were studied and estimates made of the CO2compensation point and photorespiration. The data obtained show differences between the mean net photosyntheticrates of the C. arabica cultivars (6·14 mg CO2 dm–2h–1) and the mean rate for the C. canephora cultivar (3·96mg CO2 dm–2 h–1). The cultivar of the latter speciesphotorespired more rapidly than the cultivar Catuai of C. arabica.Rates of photosynthesis in coffee measured using the 14CO2 methodwere similar to rates obtained by others using an infrared gasanalyser. The 14CO2 method proved to be reliable for photosyntheticmeasurements and the apparatus is suitable for use in fieldconditions.  相似文献

Photosynthetically competent chloroplasts were isolated fromcells of Euglena gracilis Z grown photoautotrophically in 1.5%CO2. The isolated chloroplasts were intact and substantiallyfree from cytosolic, mitochondrial and microbody materials.The effects of some compounds on the activity of photosynthetic14CO2 fixation were examined. The optimal pH and sorbitol concentrationwere 8.0 and 0.33 M, respectively. The chloroplasts requireda high level of P, (5 to 20 mM) for the maximal rate of photosynthesis.They were insusceptible to 10 mM of free Mg2+. ATP, ADP andAMP at 1 to 5 mM notably stimulated photosynthesis, althoughhigh concentrations of AMP were unfavorable. In the assay mediumdeveloped for this study, the chloroplasts exhibited photosyntheticactivity of 120µmoles-mg–1 Chl-h–1 at 30?C. Chloroplasts could also be isolated from cells grown under ordinaryair. The rate of photosynthetic 14CO2 fixation at 1 mM NaHl4CO3was higher in these chloroplasts than in those isolated fromcells grown in 1.5% CO2, whereas at 10 mM NaHl4CO3, the ratesof the two types of chloroplasts were nearly the same. Theseresults suggest that the CO2 concentration given during growthof the algal cells affects the affinity for dissolved inorganiccarbon at the chloroplast level. The levels of purine and pyrimidine nucleotides in suspensioncultures of Catharanthus roseus were determined 24 h after stationary-phasecells were transferred to fresh complete (‘+Pi’)or phosphate-deficient (‘–Pi’) Murashige-Skoogmedium. The levels of ATP, GTP, UTP and CTP were from approx.3 to 5-fold greater in the cells grown in ‘+Pi’medium than in the cells grown in ‘–Pi’ medium.The levels of almost all other nucleotides were slightly higherin the cells in ‘+Pi’ medium. The rates of de novoand salvage biosynthesis of purine and pyrimidine nucleotideswere estimated from the rates of incorporation of radioactivityfrom [14C]formate, [2–14C]glycine, NaH14CO3, [6–14C]orotate,[8–14C]adenine, [8–14C]adenosine, [2–14C]uraciland [2–14C]uridine. The results indicated that the activityof both the de novo and the salvage pathway was higher in thecells in ‘+Pi’ medium than in the cells in ‘–Pi’medium. The rate of degradation estimated from the rate of releaseof 14CO2 from labelled purines and pyrimidines indicated thatdegradation of uridine was significantly reduced in the cellsin ‘+Pi’ medium, but no significant difference wasfound in the degradation of adenine, adenosine and uracil. Thepossible role of Pi in the control of the biosynthesis of nucleotidesand in the degradation of uridine is discussed. The CO2 compensation point at 25 °C and 250 µEinsteinsm–2 s–1 wasmeasured for 27 bryo-phyte species, andwas found to be in the range of 45–160 µl CO2 I–1air. Under the same conditions Zea mays gave a value of 11 µlI–1 and Horde um vulgare 76 µI–1. The rate of loss of photosyntheticallyfixed 14CO2 in the light and dark in six bryophytes (three mosses,two leafy liverworts, one thalloid liverwort) was determinedin CO2-free air and 100% O2. The rate of 14CO2 evolution inthe light was less than that in the dark in CL2-free air, butin 100% O2 the rate in the light increased, so that in all butthe leafy liverworts it was greater than that in the dark. Raisingthe temperature tended to increase the rate of 14CO2 evolutioninto CO2-free air both in the light and dark, so that the light/dark(L/D) ratio did not greatly vary. The lower rate of loss of14CO2 in the light compared tothe dark could be due to partialinhibition of ‘dark respiration’ reactions in thelight, a low rate of glycolate synthesis and oxidation, or partialreassimilation of the 14CO2 produced, or a combination of someor all of these factors.  相似文献

Rates of CO2 and HCC3 fixation in cells of various Chlorellaspecies in suspension were compared from the amounts of 14Cfixed during the 5 s after the injection of a solution containingonly 14CO2 or H14CO3. Results indicated that irrespectiveof the CO2 concentration during growth, Chlorella vulgaris 11h and C. miniata mainly utilized CO2, whereas C. vulgaris C-3,C. sp. K. and C. ellipsoidea took up HCO3 in additionto CO2. Cells of C. pyrenoidosa that had been grown with 1.5%CO2 (high-CO2 cells) mainly utilized CO2, whereas those grownwith air (low-CO2 cells) utilized HCO3 in addition toCO2. Cells that utilized HCO3 had carbonic anhydrase(CA) on their surfaces. The effects of Diamox and CA on the rates of CO2 and HCO3fixation are in accord with the inference that HCO3 wasutilized after conversion to CO2 via the CA located on the cellsurface. CA was found in both the soluble and insoluble fractions;the CA on the cell surface was insoluble. Independent of the modes of utilization, the apparent Km (NaHCO3)for photosynthesis was much lower in low-CO2 cells than in high-CO2ones. Time courses of photosynthetic 14CO2 fixation and its simulationare presented for Chlorella cells grown under low CO2 concentration(low-CO2 cells) and subsequently exposed to 0.2 mM NaH14CO3or 130 ppm 14CO2 in the presence or absence of carbonic anhydrase(CA) in the suspending medium. It was shown that Chlorella cells utilized only free CO2 whenNaHCO3 was given in the presence or absence of CA, or when CO2was bubbled in the absence of CA. However, the present simulationindicated that both CO3 and HCO3 were utilized when CO2was given in the presence of CA. Based on these results, weconcluded that 1) Chlorella cells absorb only free CO2 and 2)this gas is provided to algal cells in two ways, i.e., by directand indirect CO2 supply. Usually, the dissolved CO2 is directlyutilized by the algal cells (direct supply of CO2). However,when the concentration of dissolved CO2 is extremely low andwhen there is CA, CO2 reconverted from HCO3 is also utilizedby Chlorella cells (indirect supply of CO2). The utilizationof HCO3 indicated by the above simulation was explainedby the indirect supply of CO2. We further assumed that the indirectsupply of CO2 to ribulose 1,5-bisphosphate carboxylase occursmainly in the chloroplasts of low-CO2 cells containing highCA. The rates of photosynthetic 14CO2 fixation by Chlorella vulgarisllh, grown under high CO2, were determined between 4 to 37°Cwith air containing from 300 to 13,000 ppm 14CO2. When the CO2level was increased, both the rate of photosynthesis and theoptimum temperature for maximum photosynthesis increased. Themaximum photosynthetic rate was reached at 12°C with 300ppm l4CO2. Among the photosynthetic products fromed at 300 ppm 14CO2, glycolatedecreased greatly when the temperature was raised from 20 to30°C. At 3,000 ppm 14CO2 an insignificant amount of glycolatewas formed at all temperatures, whereas 14C-incorporation intothe insoluble fraction, sucrose, and the lipid fraction wassignificantly higher than at 300 ppm 14CO2. Using NaH14CO3 (0·01-0·03 mol m–3) fed to5·0 mm of Nitella flexilis in artificial pond water at22–25°C, we have found that uptake in 5 min or in1 h varies with the streaming rate of the fed cell and is reducedby anoxia over the uptake position. We have also found thatthe %14C transported across the node between two internodalcells in tandem is highly sensitive to the streaming rate ineach cell, to the physiological state of the cells (summer versuswinter), to the method of feeding (5 min versus 1 h) and tocovering the node, particularly with winter cells or summercells preconditioned for 3 h in the dark and run in the dark.Transnodal transport by plasmodesmata is sensitive to anoxiaand seems to be at least partly active. Pascopyrum smithii (C3) andBouteloua gracilis (C4) are importantforage grasses native to the Colorado shortgrass steppe. Thisstudy investigated photosynthetic responses of these grassesto long-term CO2enrichment and temperature in relation to leafnonstructural carbohydrate (TNC) and [N]. Glasshouse-grown seedlingswere transferred to growth chambers and grown for 49 d at twoCO2concentrations (380 and 750 µmol mol-1) at 20 and 35°C, and two additional temperatures (25 and 30 °C) at750 µmol mol-1CO2. Leaf CO2exchange rate (CER) was measuredat a plant's respective growth temperature and at two CO2concentrationsof approx. 380 and 700 µmol mol-1. Long-term CO2enrichmentstimulated CER in both species, although the response was greaterin the C3,P. smithii . Doubling the [CO2] from 380 to 750 µmolmol-1stimulated CER ofP. smithii slightly more in plants grownand measured at 30 °C compared to plants grown at 20, 25or 35 °C. CO2-enriched plants sometimes exhibited lowerCER when compared to ambient-grown controls measured at thesame [CO2], indicating photosynthetic acclimation to CO2growthregime. InP. smithii , such reductions in CER were associatedwith increases in TNC and specific leaf mass, reductions inleaf [N] and, in one instance, a reduction in leaf conductancecompared to controls. InB. gracilis , photosynthetic acclimationwas observed more often, but significant changes in leaf metabolitelevels from growth at different [CO2] were generally less evident.Temperatures considered optimal for growth (C3: 20 °C; C4:35 °C) sometimes led to CO2-induced accumulations of TNCin both species, with starch accumulating in the leaves of bothspecies, and fructans accumulating only inP. smithii. Photosynthesisof both species is likely to be enhanced in future CO2-enrichedand warmer environments, although responses will sometimes beattenuated by acclimation. In hydroponically grown Lycopersicon esculentum (L.) Mill. cv.F144 the site of NO3 reduction and assimilation withinthe plant was shifted from the shoot to the root by salinity.Uptake of NO3 from the root solution was strongly inhibitedby salinization. Consequently, NO3 concentrations inthe leaf, stem and root tissues as well as the nitrate reductaseactivities of the leaves were lower in salinized than in controlplants. Lower NO3, but higher reduced-N, concentrationswere observed in the xylem sap as a result of the enhanced participationof the root in NO3 reduction in salinized plants. Lowerstem K+ concentrations and leaf malate concentrations were foundin salinized compared to control plants which indicates reducedfunctioning of the K+–shuttle in the salinized plants. Incorporation of inorganic carbon by the root was determinedby supplying a pulse of NaH14CO3 followed by extraction andseparation of the labelled products on ion exchange resins.The rate of H14CO3 incorporation was c. 2-fold higherin control than in salinized plants. In salinized plants theproducts of H14CO3 incorporation within the roots werediverted into amino acids, while the control plants divertedrelatively more 14C into organic acids. Products of inorganiccarbon incorporation in the roots of salinized plants providean anaplerotic source of carbon for assimilation of reducedNO3 into amino acids, while in control plants the productswere predominantly organic acids as part of mechanisms to maintainionic balance in the cells and in the xylem sap. This study investigated how CO2and temperature affect dry weight(d.wt) accumulation, total nonstructural carbohydrate (TNC)concentration, and partitioning of C and N among organs of twoimportant grasses of the shortgrass steppe,Pascopyrum smithiiRydb. (C3) andBouteloua gracilis(H.B.K.) Lag. ex Steud. (C4).Treatment combinations comprised two temperatures (20 and 35°C)at two concentrations of CO2(380 and 750 µmol mol-1),and two additional temperatures of 25 and 30°C at 750 µmolmol-1CO2. Plants were maintained under favourable nutrient andsoil moisture and harvested following 21, 35, and 49d of treatment.CO2-induced growth enhancements were greatest at temperaturesconsidered favourable for growth of these grasses. Comparedto growth at 380 µmol mol-1CO2, final d.wt of CO2-enrichedP.smithiiincreased 84% at 20°C, but only 4% at 35°C. Finald.wt ofB. graciliswas unaffected by CO2at 20°C, but wasenhanced by 28% at 35°C. Root:shoot ratios remained relativelyconstant across CO2levels, but increased inP. smithiiwith reductionin temperature. These partitioning results were adequately explainedby the theory of balanced root and shoot activity. Favourablegrowth temperatures led to CO2-induced accumulations of TNCin leaves of both species, and in stems ofP. smithii, whichgenerally reflected responses of above-ground d.wt partitioningto CO2. However, CO2-induced decreases in plant tissue N concentrationswere more evident forP. smithii. Roots of CO2-enrichedP. smithiihadgreater total N content at 20°C, an allocation of N below-groundthat may be an especially important adaptation for C3plants.Tissue N contents ofB. graciliswere unaffected by CO2. Resultssuggest CO2enrichment may lead to reduced N requirements forgrowth in C3plants and lower shoot N concentration, especiallyat favourable growth temperatures. A study has been made of photosynthetic 14CO2 fixation by isolated‘mature’ internodes of Nitella translucens. Experimentalconditions were similar to those used in studies of the ionicrelations of these cells. Maximum rates of photosynthesis were33–40µµmoles CO2, fixed per cm2 of surfacearea per second (equivalent to 12–15 /xmoles fixed permg chlorophyll per hour). l4CO2 fixation was inhibited to thedark level by 3(3,4,dichlorophenyl)-1, 1-dimethylurea (at 0-6µM or 10µM) and by the uncoupler carbonyl cyanide-m-chlorophenylhydrazone(SµM). The presence of imidazole or ammonium sulphate(both of which uncouple ATP production in vitro) did not resultin an inhibition of 14CO2 fixation. These results are discussedin relation to published work on solute uptake by Nitella translucens.During photosynthesis there was rapid movement of 14C-labelledorganic compounds out of the chloroplasts. 14C-labelled sucrose,ammo-acids, and sugar phosphates were found in samples of vacuolarsap.  相似文献

Inorganic carbon transport during photosynthesis of cyanobacteriumAnabaena variabilis grown under ordinary air was investigatedby supplying 14CO2 or H14CO3 solution to three differentstrains. Both CO2 and HCO3 were accumulated within thealgal cells. In the cell suspension from which dissolved inorganiccarbon had been depleted by pre-illumination, CO2 was transportedand accumulated faster than HCO3. When the concentrationof HCO3 injected into the cell suspension of A. variabilisM3 was 25 times as high as that of CO2 (the expected ratio atequilibrium at pH 7.8), the initial rates of fixation of bothinorganic carbon species were practically the same. On the otherhand, when 14CO2 or H14CO3 was added under steady statephotosynthetic conditions, both carbon species were transportedat similar rates. The ratio of fixed to transported carbon measuredafter the initial 5 s was only 23–27% regardless of thecarbon species supplied. A model is presented which compares the ratio of the two activitiesof the enzyme nbulose bisphosphate carboxylase/oxygenase asdetermined in vitro with the ratio of photosynthesis to photorespirationin leaves as determined from differential 14CO2/12CO2 uptakeor from CO2 compensation concentration. Discrepancies betweenmeasurements made in vitro and in vivo are attributed to theeffect of a CO2 concentrating system in the leaf cells. Interferencefrom dark respiration is discussed. A CO2 concentrating systemis postulated which is efficient mainly at low temperature andlow CO2 concentration. Photosynthetic assimilation of exogenous 14CO2 and H14CO3by the aquatic angiosperm Potamogeton lucens L. is reported.Equivalent maximum rates of assimilation (1.5 µmol s–1m–2) were obtained in the presence of saturating levelsof 14CO2 (1.0 mol m–3, pH 5.3) or H14CO3 (1.5 molm–3, pH, 9.2). Under subsaturating 14CO2 levels, bothgaseous diffusion and H14CO3 transport were shown tooperate simultaneously, such that maximal photosynthetic rateswere established. An induction lag of approximately 3 min was observed when exogenous14CO2 was assimilated. A longer lag of approximately 12 minwas required, however, before linear assimilation rates wereestablished when H14CO3 acted as the carbon source. The light-activatedH14CO3 transport system was found to be quite labile.A brief (5 min) dark treatment returned the system to the inactivestate. Bicarbonate transport was shown to be competitively inhibitedby CO32–ions. The possibility is discussed that this formof inhibition may be common to many HCO3 assimilators. Preliminary polar cation transport studies (from lower to upperleaf surface) indicated an almost exact one to one relationshipbetween the rates of Na+ influx and efflux and H14CO3assimilation. The possible relationship(s) between these transportprocesses and the requirement for electrical neutrality is brieflydiscussed.  相似文献

Perennial ryegrass (Lolium perenne L.) cv. S23 was exposed to0, 50, and400 µg m– 3 SO2 for a 29 d period, harvested,and then exposed under the same regime for a further 22 d periodof regrowth. Leaves from plants representing each exposure concentrationwere photosynthetically fed 14CO2 for 5 min at the end of eachperiod. A significant increase in photoassimilation of 14CO2and retention of I4C, concomitant with significant decreasesin [14C]glycine and [14C]serine with increasing SO2 concentration,implied that there was an inhibition of the photorespiratorypathway. At the second harvest, leaves from plants exposed to400 µg m– 3 SO2 also exhibited significant increasesin [14C]sucrose and [14C]fructose.  相似文献
