从杭州地区呈现玉米矮花叶典型症状的玉米病组织中提纯得到大量线状病毒粒子,大多数长度为750nm。病组织中含有大量风轮状内含体和板状集结体,病毒外壳蛋白为33.6kD。病毒RNA1 3'端序列(1.8kb)与甘蔗茶花经叶病毒(SCMV)同源性最高,达71.5%-99.1%,与高梁花叶病毒(SrMV)同源性次之,为67.8%-68.5%,与玉米矮花叶病毒(MDMV)同泊性最低,仅为38.4%-48.4%,从而初步认为此病害由SCMV引起。根据已发表的SCMV外壳蛋白氨基酸序列作亲缘性分析,表明SCMV可分为美国、南非、澳大利亚、德国和中国三大类。  相似文献

测定了从浙江省呈现矮花叶病症状的玉米上分离得到的一个马铃薯Y病毒属病毒RNA的核苷酸全序列. 该病毒分离物的RNA基因组由9596个核苷酸组成(不包括polyA尾). 单一的ORF由9192个核苷酸组成, 编码一个分子量为346.1 ku的聚合蛋白. 该蛋白结构特征与高粱花叶病毒(SrMV)中国甘蔗分离物和一个玉米矮花叶病毒(MDMV)保加利亚分离物基因组编码的蛋白非常相似. 序列分析表明, 该病毒分离物与甘蔗花叶病毒(SCMV)各分离物(已报道的仅为基因组3′末端序列)同源性最高, 与SrMV和MDMV同源性次之, 而与约翰逊草花叶病毒(JGMV)同源性最低. 根据马铃薯Y病毒属区分不同病毒和株系的分类标准, 报道的玉米病毒分离物应当鉴定为SCMV的一个株系. 然而, 该分离物在HC-Pro, P3和CI蛋白区域和SrMV中国甘蔗分离物具有极高的氨基酸同源性.  相似文献

山东省玉米矮花叶病毒的生物学特性及基因组全序列测定   总被引:3,自引:0,他引:3  
对山东省玉米矮花叶病毒原分离物(SD)进行了寄主范围、血清学等普通生物学鉴定,测定了该病毒的基因组核苷酸全序列。该病毒基因组RNA由9596个核苷酸组成(不包括3’—polyA的长度),整个基因组按一个ORF编码一个3063个氨基酸的多聚蛋白。序列比较表明,该病毒分离物(SD)与玉米矮花叶病河南分离物(HN,EMBL登录号:AF94510)核苷酸全序列同源性最高,为98.2%,与已报道的甘蔗花叶病毒(SCMV)7个分离物同源性也高达79.5%-98.2%,但与玉米矮花叶病毒保加利亚分离物(MDMV—B8,AJ001691)和高梁花叶病毒萧山甘蔗分离物(SrMV—XoS,AJ310197)的同源性仅为67.8%和69.3%,与约翰逊草花叶病毒(226920,JGMV)差异最大。系统进化树分析也表明,该病毒与SCMV分离物位于同一进化簇,而与MDMV进化关系很远。  相似文献

黄瓜绿斑驳花叶病毒辽宁分离物全基因组序列测定   总被引:7,自引:0,他引:7  
以感病组织总RNA为模板,采用RT-PCR方法扩增并测定黄瓜绿斑驳花叶病毒(Cucumber green mottle mosaic virus,CGMMV)辽宁分离物(CGMMV-LN)的基因组全序列。CGMMV-LN基因组全长6 422 nt,5'非编码区(noncoding region,NCR)和3'NCR分别为59 nt和175 nt。CGMMV-LN编码的4个蛋白依次是186 kD和129kD的复制酶,29 kD的移动蛋白和17.4 kD的外壳蛋白。CGMMV-LN与其他4个CGMMV分离物基因组核苷酸序列同源性为97.6%~99.3%,与同属其他3种病毒基因组核苷酸序列同源性仅为61.7%~62.8%。基于186kD复制酶和外壳蛋白氨基酸序列的同源树显示:侵染葫芦科作物的烟草花叶病毒属病毒可分为2个亚组,亚组I包括所有CGMMV分离物,亚组II包括Kyuri绿斑驳花叶病毒(Kyuri green mottle mosaic virus,KGMMV)、黄瓜果实斑驳花叶病毒(Cucumber fruit mottle mosaic virus,CFMMV)和小西葫芦绿斑驳花叶病毒(Zucchini ...  相似文献

选取我国SCMV优势株系A株系的分离物SCMV-CA为材料,经过病毒和病毒RNA的提纯,反转录获得病毒cDNA,并克隆到载体pUC19的SmaI位点上,筛选得到多个重组质粒。选取其中一个克隆SCMV-CA54进行测序,得到一个全长为1296 bp的核苷酸序列。这段序列由一个长为1044 bp的开放阅读框架(ORF)和一个长279 bp的3’末端非编码区序列(3'-UTR)及poly(A)尾巴组成。这个ORF包括病毒完整的外壳蛋白(CP)及部分核内含体蛋白b(NIb)基因序列。将所得序列同已知SCMV亚组中各株系分离物的核苷酸和氨基酸进行同源性比较,结果表明该序列与其它株系分离物CP核苷酸序列的同源性介于63.7%~77.6%之间,氨基酸的同源性介于64%~89%之间。根据马铃薯Y病毒属的序列同源性划分标准,SCMV-CA与其它株系或分离物的同源性关系均介于种与株系划分标准之间。这是我国首次报道SCMVCP基因序列。  相似文献

从浙江省5个地区采集表现花叶症状的甘蔗病叶,用马铃薯Y病毒科简并引物做PCR扩增及测序鉴定.序列分析表明,5个甘蔗样品均含有高粱花叶病毒(SrMV),其中3个样品中还存在甘蔗花叶病毒(SCMV)的复合侵染.序列比较和系统进化树分析表明,浙江甘蔗样品中的SrMV序列彼此很相似,核苷酸同源性大于93%,与已报道的4个美国分离物在CP区域同源性很高,但是3′非编码区的同源性却仅为70%左右.SCMV欧洲和中国玉米分离物及美国、南非和澳大利亚甘蔗分离物分别形成两个远缘群体,浙江甘蔗分离物群体位于两者之间;群体间CP氨基酸序列同源性均大于80%.甘蔗和玉米上的SCMV差异明显,多为无义突变.  相似文献

大麦黄矮病毒GPV株系是我国特有的一个株系,与国外已报道的大麦黄矮病毒MAV,PAV,SGV,RPV和RMY在血清学上无反应.通过对GPV.外壳蛋白基因的序列分析,明确了病毒的外壳蛋白基因由603个核苷酸组成,编码201个氨基酸,分子量为22218ku.同MAV,PAV,RPV株系一样,在GPV外壳蛋白基因框架中(ORF)含有1个Vpg框架结构,分子量为17024ku.外壳蛋白基因序列同源性比较结果显示,GPV株系和RPV株系同源性较高,核苷酸和氨基酸的同源性分别为83.7%和77.5%.而与PAV和MAV株系同源性较低,核苷酸和氨基酸的同源性分别为56.9%,53.2%和44.1%,43.8%.按照GPV外壳蛋白基因序列,设计寡聚核苷酸引物,通过RT-PCR反应,得到GPV外壳蛋白基因全长cDNA,克隆到表达质粒,构建了高等植物表达载体pPPI1,pPPI2和pPPI5.  相似文献

根据已报道的洋葱黄矮病毒(OYDV)序列设计引物,扩增洋葱黄矮病毒(OYDV)六安分离物外壳蛋白(CP)基因.序列同源性分析结果表明,与目前已报道的OYDV不同分离物CP基因核苷酸序列同源性为84.0%~95.8%,相应推测的氨基酸序列同源性为86.8%~97.6%.将CP基因插入原核表达载体pSBET后在大肠杆菌BL21(DE3)Plys S中诱导表达.通过12%SDS-PAGE和5%~20%梯度SDS-PAGE两次制备电泳纯化诱导产物,免疫家兔获得抗CP血清,Western blot分析对CP具有高度特异性.硫酸铵沉淀初步IgG,然后用Protein A-Red Sepharose亲和层析进一步纯化IgG,IgG与甘油1: 1混合获得效价1: 4 800的一抗.将一抗、羊抗兔二抗和4-硝基苯基磷酸二钠组成OYDV检测试剂盒,可用于田间OYDV病样检测.  相似文献

测定了在中国分离得到由麦二叉蚜和麦长管蚜传播的大麦黄矮病毒GAV的基因组核苷酸全序列, 该病毒分离物的RNA由5685个核苷酸组成, 内含6个开放阅读框架(ORF)和4个非编码区(UTR), 基因组大小和结构与黄症病毒属(Luteovirus)的大麦黄矮病毒PAV(BYDV-PAV)和MAV(BYDV-MAV)相似. 序列分析表明, 它与BYDV-MAV的PS1分离物基因组序列的同源性最高. 在6个开放阅读框架中, 除ORF6核苷酸序列同源性为72.0%外, 其他ORF的核苷酸序列同源性均大于90%. 两者全基因组的同源性为90.4%. 推导的编码产物氨基酸序列同源性除P6和通读蛋白(RTP)分别为67.4%和87.4%外, 其他均大于90%, 其中外壳蛋白(CP)为95.5%. 根据与BYDV-MAV的相似性, BYDV-GAV应是一种与BYDV-MAV类似的病毒.  相似文献

由甘蔗花叶病毒引起的玉米矮花叶病是我国黄淮海地区玉米生产的重要病害,开发抗矮花叶病基因分子标记是开展抗病分子标记辅助育种的基础。本文基于玉米6.00-6.01区域的“一致性抗甘蔗花叶病毒QTL区间”寻找抗病基因的功能保守域,依据序列多态性开发出抗病分子标记InDel-130和InDel-110,在已知抗性的102份玉米自交系中进行验证。通过分析标记抗病带型和感病带型中的抗病和感病自交系数目,卡平方测验表明标记InDel-130在供试自交系中与抗病性的表现独立无关.而标记InDel-110与甘蔗花叶病毒抗性高度相关,为共显性标记,可用于玉米抗甘蔗花叶病毒种质筛选和分子标记辅助育种。  相似文献

During field surveys, three peanut green mosaic virus isolates differing in symptomatology on groundnut and a few other hosts were collected. Ultrathin sections of infected groundnut leaflets showed cytoplasmic inclusions with pin wheels and scrolls. In enzyme-linked immunosorbent assay they reacted strongly with antisera to peanut green mosaic and soybean mosaic virus antisera, and moderately with adzuki bean mosaic and peanut stripe virus antisera. All isolates also reacted positively with antisera to peanut eye spot, blackeye cowpea mosaic, pea seed-borne mosaic, potato virus Y and tobacco etch viruses, and did not react with antisera to peanut mottle, bean yellow mosaic, bean common mosaic, clover yellow vein and sugarcane mosaic viruses. SDS-PAGE analysis of purified virus preparations of the three isolates showed a single polypeptide with mol. wt. of 34,500 daltons. Based on these results, the three isolates are identified as biologically distinct strains of peanut green mosaic virus.  相似文献

Analyses of published host-range data for certain viruses reveal correlations with taxonomic groupings of grasses. Barley yellow dwarf virus (BYDV), cocksfoot mottle and phleum mottle viruses are found to have infected greater proportions of the festucoid grasses than of the non-festucoids to which they were inoculated. By contrast, all strains of sugarcane mosaic virus (SCMV) and of the closely related maize dwarf mosaic virus (MDMV) infected more non-festucoids than festucoids. In addition, infected plants from grass groups containing higher concentrations of genera susceptible to BYDV, SCMV and MDMV usually show clear symptoms, whereas infected plants from less susceptible groups are frequently symptomless. Some viruses, such as barley stripe mosaic, brome mosaic, cocksfoot streak and ryegrass mosaic, show no apparent preferences for particular grass groups. Samples of grasses employed in host-range studies are usually strongly biased towards festucoids. It is suggested that viruses ought to be adequately tested against genera from all the major groups, and a classified list of grass genera suitable for host-range studies is provided.  相似文献

Biological, serological and coat protein properties of a potyvirus (Poty-Rape) causing a mosaic disease of Brassica campestris and B. juncea in India were investigated. The virus readily infected 4 of the 5 plant species in the family Brassicaceae in which it induced severe systemic mosaic symptoms; it also induced chlorotic and necrotic local lesions in Chenopodium amaranticolor , but failed to infect 4 other species of Chenopodiaceae or 20 species of Amaranthaceae, Apiaceae, Canabinaceae, Compositae, Cucurbitaceae, Euphorbiaceae, Leguminosae and Solanaceae. The virus was transmitted in a non-persistant manner by Myzus persicae, Brevicoryne brassicae and Aphis gossypii. The Average size, of the virus particles in a purified preparation was 740 nm × 12 nm. SDS-PAGE analysis of the viral coat protein showed two major bands of approximately 37 kDa and 31 kDa, a pattern very similar to that of a reference isolate of turnip mosaic virus (TuMV) from the U.S. In Western-blot immunoassay, an antiserum to TuMV reacted with both the coat protein bands of the Poty-Rape islate and the reference TuMV, but not with the coat proteins of four other potyviruses. The high performance liquid chromatographic profile of tryptic peptides from the coat protein of Poty-Rape was found to be very similar to that of the reference TuMV, but differed substantially from those of four other potyviruses. The Poty-Rape isolate is considered to be a distinct strain of, TuMV.  相似文献

Maize dwarf mosaic is the most widespread virus disease affecting corn production in Hungary. In attempts to identify the causal virus by test plant reactions, enzyme‐linked immunosorbent assay (ELISA) and polymerase chain reaction (PCR), only Maize dwarf mosaic virus (MDMV) was detected. To further characterize Hungarian isolates of MDMV, one isolate from each of the sweet corn varieties Dallas, Royalty and GH23‐85 was selected for sequence analysis of its coat protein (CP) gene. The three Hungarian isolates shared CP amino acid sequence similarities of 95–98% not only with one another but also with MDMV isolates from other countries. However, the N‐terminus of the CP of the ‘Dallas’ isolate was unusual in containing a stretch of 13 additional amino acids. This is the first report of variation in the size of the N‐terminus of the MDMV CP.  相似文献

Degenerate Potyviridae primers were used to amplify and sequence the 3′‐terminal regions of viruses from traditional and modern cultivars of sugarcane with mosaic disease growing in different areas of Yunnan province, China. Seven samples contained Sugarcane mosaic virus (SCMV), 11 contained Sorghum mosaic virus (SrMV) and two contained both viruses. SCMV was only isolated from traditional cultivars. In a phylogenetic analysis of the partial NIb and complete coat protein coding regions, most SCMV isolates formed a distinctive phylogenetic cluster (named SO) that otherwise contained only three Vietnamese isolates. SCMV variation seems mostly related to host genotype. In the same analysis, the SrMV isolates formed three major groups, one of which is reported for the first time, but the significance of the grouping is unclear.  相似文献
