目的采用疏水相互作用层析分离重组人干扰素α2b,去除干扰素样品中的二聚体,得到高纯度的干扰素用于进一步的研究。方法首先采用阳离子交换层析纯化复性重组人干扰素α2b,去除了大部分的杂蛋白,然后采用疏水相互作用层析纯化重组人干扰素α2b,去除复性过程中产生的错误折叠体和二聚体,并考察盐浓度、pH值、流速和洗脱液中尿素对疏水相互作用层析纯化效果的影响。结果硫酸铵初始浓度1.2 mol/L、缓冲液pH值6.0、流速2.5 mL/min、洗脱液中添加尿素浓度为2 mol/L时疏水相互作用层析纯化效果最佳。最终得到的重组人干扰素α2b非还原型SDS-PAGE电泳均呈单一条带。结论确定了疏水层析纯化重组人干扰素α2b的最优条件,成功提取到具有高活性、高纯度的重组人干扰素α2b纯品。  相似文献

本文利用SDS-PAGE及蛋白质电泳印迹技术,从带有相应表达质粒的重组大肠杆菌裂解液中,将所表达的重组人嗜中性白细胞活化蛋白-1/白细胞介素-8(NAP-1/IL-8)转移至聚偏二氟乙烯膜上,直接进行N-末端15个氨基酸的序列分析,从而确证该目标蛋白得到高效表达和正确加工。随后采用Bio-Gel P30凝胶过滤层析和Mono-S阳离子交换层析对重组人NAP-1/IL-8进行了分离纯化,纯化产品达到SDS-PAGE纯。利用琼脂糖平板法测定了纯化产品的嗜中性白细胞趋化活性,推算其比活为2.8×10~5U/mg蛋白。又利用SDS-PAGE测出重组NAP-1/IL-8的分子量约为8.5kD,但根据凝胶过滤层析的洗脱时间推定,在溶液中确实存在分子量稍大于14.4kD的NAP-1/IL-8二聚体。  相似文献

Bovine transferrin (BTF) was fractionated from bovine whey using ganglioside affinity chromatography. After loading the immobilized matrix with a 2% whey solution, the matrix was washed with sodium acetate buffer at pH 4 containing 1 M NaCl before elution of BTF with sodium phosphate buffers at pH 7. Concanavalin-A affinity and ion exchange chromatography were used for further purification. The ganglioside column showed a 74.2% BTF recovery from whey and BTF was enriched to 61% purity with ion exchange chromatography. Bovine transferrin was identified by SDS-PAGE and western analysis. The Concanavalin-A affinity and ion exchange chromatography steps enriched BTF in the samples and removed other whey proteins from ganglioside purified fractions. These results indicate that immobilized ganglioside can be used to fractionate BTF from bovine whey. Our novel ganglioside affinity chromatography is rapid and efficient for the fractionation of BTF from whey.  相似文献

Apoplastic β-1, 3-glucanase was purified from leaves of tomato (Lycopersicon esculentum Mill. ) which were systematically infected by TMV (tobacco mosaic virus). The enzyme obtained through -20℃ acetone precipitation, CM-Sephadex C-25 ion exchange chromatography, DEAE-Sephadex A-25 ion exchange chromatography and Sephadex G-75 gel filtration, showed homogeneity in PAGE, and SDS-PAGE which had two isoenzymes of 27 kD and 36 kD. The enzyme hydrolysed laminarin at an optimum pH of 4.8--5.2 and was stable between pH 4--8 and at an optimum temperature between 30--40℃, and stable at 40℃ after 1 hour of incubation, It had a Km of 9. 2 mg/mL. SDS-PAGE profiles of the proteins in the tomato leaf intercellular fluid had the bands of 22 kD, 27 kD and 36 kD that were β-1, 3-glucanases.  相似文献

目的 探索有效的纯化大雁卵黄抗体及高效价二抗的方法。方法 应用了PEG沉淀,阴离子交换柱,硫酸铵分级沉淀纯化,免疫制备二抗,免疫电泳和免疫双扩散法及western blotting进行效价的测定。结果 改良后的硫酸铵沉淀法的纯度远远高于PEG沉淀,相对于离子交换法而言更加简单方便,经济省时。从免疫电泳看已达到了电泳纯,大雁卵黄IgY的相对分子质量为180×10^3,重链约为66×10^3,轻链约为24×103。免疫获得的二抗有较强的免疫活性。结论 用改良的硫酸铵沉淀法纯化大雁卵黄抗体,效率和纯度显著提高,值得推广。本实验为大雁卵黄抗体及其二抗的应用奠定了基础,也为雁形目其他种类卵黄抗体的纯化提供了参考。  相似文献

目的 建立高纯度、高活性的虎血清IgG纯化方法。方法 用饱和硫酸铵沉淀虎血清得到IgG粗品;结合Hitrap Protein A亲和层析预装柱及阴离子交换层析法对粗品IgG进一步分离纯化,采用PAGE电泳和Western-Blot免疫印迹法鉴定IgG纯度和免疫活性。结果 80 mL虎血清亲和纯化得到84 mg IgG,阴离子交换层析纯化得到30 mg虎的IgG纯品。结论 建立了简便快速、纯度高、活性好的虎血清IgG的分离纯化方法,为虎血清IgG二级抗体的制备提供了高纯度、活性好的一级抗体免疫原。  相似文献

Phycocyanin, a high value pigment was purified from diazotrophic cyanobacteria Anabaena variabilis CCC421 using a strategy involving ammonium sulfate precipitation, dialysis and anion exchange chromatography using DEAE-cellulose column. 36% phycocyanin with a purity of 2.75 was recovered finally after anion exchange chromatography. Purified phycocyanin was found to contain 2 subunits of 17 and 18 kDa which were identified as α and (β subunits by SDS-PAGE and MALDI-TOF. HPLC method using a C5 column coupled with fluorescence or photodiode-based detection was also developed to separate and detect the A. variabilis CCC421 phycocyanin subunits. The fluorescence method was more sensitive than photodiode one. The purified phycocyanin from A. variabilis CCC421 as well as its subunits was characterized with respect to absorption and IR spectra. Spectral characterization of the subunits revealed that α and β-subunits contained one and two phycocyanobilin groups as chromophores, respectively.  相似文献

We describe a one-step purification of hepatic metallothionein from the Amazon fish Colossoma macropomum injected with cadmium and from the copper-loaded metallothionein from the yeast Saccharomyces cerevisiae, performed by affinity chromatography through metal-chelating columns. Yeast metallothionein was purified from Cu2+-loaded resin and eluted by a continuous EDTA gradient whereas hepatic metallothionein extracted from fishes was purified by Ni2+-loaded resin and eluted by a continuous imidazol gradient. Purified metallothioneins were evaluated by SDS-PAGE and characterized by UV spectra of the apo- and Cd2+-loaded protein. This method allowed high purity and yield as well as rapid one-step extraction of both metal-loaded and apoprotein.  相似文献

Trypsin and papain proteinase inhibitors have been identified and purified from aqueous extracts of apple seeds (Malus domestica). Superdex G75 gel filtration chromatography identified a higher molecular weight (HMW) papain inhibitory fraction (22-26 kDa) and a lower molecular weight papain and trypsin inhibitory fraction (6-12 kDa). The lower molecular weight fraction was separated into a trypsin inhibitor (designated Trp1) and early (designated Pap1) and late (designated Pap2) eluting papain inhibitors after anion exchange (Hitrap SP) chromatography. For Pap2, two inhibitory peaks (designated Pap2-1 and Pap2-2) were identified after further anion exchange (Resource S) chromatography. Each of these lower molecular weight inhibitors was purified by reverse phase HPLC to homogeneity as determined by SDS-PAGE and by mass spectrometry. The HMW papain inhibitory fraction was purified further by anion-exchange (Hitrap Q followed by Resource Q) column chromatography where a minor inhibitor (HMWPap1) and major inhibitor (HMWPap2) fraction were identified. The relative abundance in seeds of apple and the spectrum of proteinase inhibition has been determined for all of these inhibitors. Reverse-phase HPLC separated HMWPap2 into a minor (HMWPap2-1) and a major (HMWPap2-2) inhibitory fraction, and SDS-PAGE and mass spectrometry confirmed that HMWPap2-2 was purified to homogeneity. Amino acid composition data were obtained from Trp1, Pap1, Pap2-2, and HMWPap2-2, and N-terminal sequence data from Trp1, Pap2-1, Pap2-2, and HMWPap2-2, with two of these sequences (Pap2-2 and HMWPap2-2) perfectly matching predicted protein sequences based on EST sequences from an apple database. The relationship of these inhibitors with those of other species is discussed.  相似文献

Bovine hemoglobin (bHb) was purified from bovine red blood cells (bRBCs) via anion exchange chromatography preceded by dialysis. This is a fast and effective way to obtain bHb from bRBCs using Q Sepharose XL, a strong anion exchange resin. This resin had double the binding capacity for bHb compared to three other anion exchange resins that were studied in this work. Methemoglobin levels remained below 2% with bHb concentrations between 0.7 and 1.7 mM. The high purity of bHb was confirmed via SDS-PAGE and size exclusion chromatography (SEC).  相似文献

Recombinant human interleukin-1 receptor antagonist (rHuIL-1ra) was produced in E. coli as an inclusion body. rHuIL-1ra was purified to Over 98% purity by anion exchange chromatography after on-column refolding. The optimized processes produced more than 2 g pure refolded rHuIL-1ra per 1 l culture, corresponding to a 44% recovery, without an intermediate dialysis step. Refolded rHuIL-1ra had full biological activity with the MTT assay. An intramolecular disulfide linkage in the oxidized recombinant protein was suggested by data from HPLC and non-reducing SDS-PAGE.  相似文献

通过盐溶液抽提、硫酸铵分级沉淀、CM 5 2纤维素阳离子交换层析、反相毛细管液相色谱 (re verse phasecapillaryliquidchromatography ,RP CLC)等步骤 ,从八棱丝瓜籽中分离到 2种单链核糖体失活蛋白luffaculin 1和luffaculin 2 .在SDS PAGE和IEF上均显示为单一条带 ,表观分子量均为 2 8kD ,其等电点分别为 8 86 (luffaculin 1)和 9 0 5 (luffaculin 2 ) .实验表明 ,它们具有RNAN 糖苷酶活性 .蛋白质合成抑制活性测试表明 ,它们对蛋白质合成具较强的抑制作用 .体外抑制肿瘤细胞生长活性检测表明 ,luffaculin 1和luffaculin 2对人白血病细胞株K5 6 2有较强的毒性 ,IC50 分别为 1 1×10 -6mol L和 2 0× 10 -7mol L .八棱丝瓜籽核糖体失活蛋白具有可以用于或构成免疫毒素治疗癌症的应用前景  相似文献

We developed (GLP-1A2G)2-HSA (GGH) analogs that are resistant to degradation and also show high serum glucose-reducing activity in vivo. Five analogs with N-terminal extensions were designed based on the protein GGH. Next, we constructed recombinant plasmids capable of expressing the five analogs in methylotrophic yeast Pichia pastoris GS115. Expression reached 150 mg/L in a small-scale incubation. Fusion proteins were successfully purified from the supernatant using ultrafiltration concentration, affinity absorption chromatography, hydrophobic chromatography, ion exchange chromatography and gel filtration. A single band was observed on SDS-PAGE and the purity was 97%. Activity test results suggested that both A-GGH and G-GGH showed better activity in vitro and that their cAMP levels were significantly increased by 10-fold compared to GGH without N-terminal extension. Additionally, A-GGH efficiently enhanced the glucoselowering effect, which was maintained after the administration for 24 h. A-GGH is a potential drug for treating type 2 diabetes.  相似文献

通过丙酮沉淀、DEAE-纤维素离子交换柱层析和Sephadex G-100凝胶过滤柱层析等分离纯化技术,对巴西橡胶树胶乳C-乳清磷脂酶A2进行分离纯化。用SDS-PAGE测定其亚基的相对分子量。测定该酶最适温度和pH,动力学常数Km和Vmax。并测定Ca2+和La3+对酶活性的影响。结果显示:该酶被纯化了49.47倍,产率为5.12%。SDS-PAGE检测为单一条带,其亚基相对分子量约43kDa。最适反应温度为37℃,最适反应pH为8.0,Km为0.44mmol·L-1,Vmax为7.22μmol.(mL.min)-1。最适Ca2+浓度为50μmol·L-1,稀土元素La3+离子对磷脂酶A2活性有抑制作用,但加入Ca2+后可缓解La3+对磷脂酶A2活性的抑制作用。胶乳C-乳清磷脂酶A2与其他植物磷脂酶A2在Ca2+的依赖性上存在差异。研究结果为今后探索橡胶树胶乳磷脂酶A2的催化机理、调节机理及生理功能等奠定了基础。  相似文献

A novel enzyme, the first metalloproteinase purified from a monocotyledonous plant, was extracted from the endosperm of sorghum seedlings and purified to homogeneity by ion exchange chromatography and size exclusion chromatography. SDS-PAGE analysis reveals a dimeric 17-kDa protein with two 8-kDa subunits linked by disulfide bond(s). The enzyme is 97% inhibited by 1 mM EDTA and is unaffected by inhibitors of aspartic, cysteine, and serine proteinases. Its pH optimum is 7.0 with hemoglobin as substrate.  相似文献

目的:以抗HER2抗体为模型,研究抗体在糖基工程酵母菌中的表达及工程菌发酵技术。方法:首先通过摇瓶试验分析诱导用甲醇浓度对抗体表达的影响,并用高表达HER2的SK-BR-3细胞分析抗HER2抗体的抗原结合活性。以此为基础,在5 L发酵罐中研究甲醇-山梨醇混合碳源流加诱导对抗HER2抗体表达水平的影响;收集发酵培养液,采用阳离子交换层析对目标产物进行纯化;利用SDS-PAGE、Western印迹、Lowry法对抗体的相对分子质量、浓度等进行分析。结果:摇瓶试验结果表明,甲醇浓度为0.5%时抗体表达量最高,且糖基工程毕赤酵母菌表达的抗HER2抗体具有与SK-BR-3细胞抗原结合的活性;在5 L发酵罐中,利用甲醇和山梨醇混合诱导方式发酵表达抗体,其表达量可提高至0.6 g/L,比摇瓶诱导表达的抗体产量提高了近10倍;非还原SDS-PAGE及Western印迹表明抗体相对分子质量为1.5×105,与商业化抗体Herceptin的大小一致;经过一步阳离子交换层析纯化,纯化后抗体浓度为0.365 g/L。结论:采用甲醇-山梨醇混合碳源诱导方式在5 L发酵罐中进行发酵表达,能够提高抗HER2抗体在糖基工程酵母菌中的表达量,本研究可为抗体在酵母中的规模发酵技术提供重要参考。  相似文献
