This study investigated the effects of pelvic suspension and slaughter age on longissimus thoracis et lumborum (LTL) from 40 heifers with at least 75% Angus breeding. A total of 20 heifers were slaughtered directly from pasture at 18 months of age, and carcass sides were hung either by the Achilles tendon or the pelvic bone. The other 20 heifers were assigned to an additional winter housing period and slaughtered at 22 months of age; carcass sides were hung only by Achilles suspension. All carcasses were electrically stimulated and assessed according to the EUROP carcass classification system. In addition, the LTL muscles were aged for 7 or 14 days before meat quality was evaluated for intramuscular fat (IMF), drip loss, colour, shear force, compression and sensory analysis. The 22-month-old heifers were heavier, fatter and had more IMF than 18-month-old heifers. Conformation scores (muscling) did not differ between the two slaughter groups. Pelvic suspension reduced both between- and within-animal variation for peak force, total energy and compression peak force. For the 18-month-old heifers, pelvic suspension also decreased peak force, total energy and compression variables for the LTL muscles from both ageing periods, whereas Achilles-suspended samples had lower shear force values only at day 14. Sensory analysis showed that pelvic-suspended sides had greater tenderness, lower bite resistance, less threadiness, higher juiciness and meat flavour and less visible marbling than meat from Achilles-suspended sides. Pelvic-suspended sides at 18 months of age were similar in peak force and total energy values to the 22-month-old heifers. The importance of ageing the Achilles-suspended sides was more obvious for samples from 18-month-old heifers than from the 22-month-old animals. The correlations between the different instrumental measurements and sensory tenderness were considerably higher for carcasses suspended by the Achilles tendon (r = −0.55 to 0.20) than for those hung by the pelvic bone (r = −0.25 to 0.19). More correlations between sensory-evaluated tenderness and shear variables were significant after 7 days (n = 6) of ageing than after 14 days (n = 4) of ageing. This study clearly shows the benefits of pelvic suspension, which reduces the need for additional feeding after pasture.  相似文献

In practice cattle may be slaughtered at different combinations of age and weight. As each of these factors could affect meat quality traits, the present work aimed to identify which combination can be expected to increase overall meat quality of m. rectus abdominis of Charolais heifers. Totally, 40 heifers were slaughtered either at 26±1 or at 36±1 months of age. Young heifers were sampled at two different carcass weights (349±12 and 394±8 kg). Old heifers were also sampled at two different carcass weights (397±6 and 451±9 kg). The m. rectus abdominis was excised 24 h postmortem to determine metabolic enzyme activities, myosin heavy-chain isoform proportions, lipid contents, collagen content and collagen solubility. Shear force measurements were evaluated on raw and broiled meat after 14 days of ageing. Meat quality traits scored between 0 and 10 by sensory analysis. Increasing slaughter age from 26 to 36 months had no impact on either raw/broiled shear force (0.31⩽P⩽0.47) and/or meat quality traits (0.62⩽P⩽0.91) or on physicochemical properties of heifer’s meat samples. Increasing carcass weight for a similar slaughter age of 26 months had also impact neither on meat quality traits (0.52⩽P⩽0.91) nor on muscular properties. On the contrary, increasing carcass weight for a similar slaughter age of 36 months had induced a decrease of muscular shear force (raw muscle; P=0.009) and a concomitant decrease of total collagen content (P=0.03). Nevertheless, no significant impact on meat quality traits was revealed by the sensorial panel (0.13⩽P⩽0.49). Metabolic enzyme activities (0.13⩽P⩽0.86) and myosin heavy-chain proportions (0.13⩽P⩽0.96) were not significantly impacted by slaughter age and carcass weight. Thus, the impact of increasing carcass weight and/or slaughter age in young Charolais heifers has a limited impact on meat quality traits and associated muscular characteristics. Modulating heifer’s cycles (age and/or carcass weight in the studied range) appears to be a way to answer to the numerous marketing chains, without penalising meat quality traits.  相似文献

We analyzed single nucleotide polymorphisms in calpain, leptin, leptin receptor, and growth hormone receptor genes and their association with color, drip and cooking losses of longissimus muscle at 7, 14 and 21 days postmortem in 638 purebred Nellore bulls slaughtered between 22 and 26 months of age. Meat samples were vacuum-packed and aged at 4°C. The single nucleotide polymorphisms T945M, GHR2, E2FB, and CAPN4751 were evaluated. All genotypic classes were observed; however, the T/T genotype of T945M and E2FB was found at a low frequency. A significant association of E2FB with drip loss (a measure of water-holding capacity) was detected at seven days of meat aging. CAPN4751 had an additive effect on red and yellow color intensities. The T allele of CAPN4751 was found to be positively associated with improved meat color, but not with meat tenderness, differing from a previous report indicating that it is associated with meat tenderness. We conclude that the potential for use of CAPN4751 as a marker for these meat quality traits requires further research.  相似文献

The objective of this study was to determine the levels of catecholamines and their relationship to beef quality in Nguni steers fed on Acacia karroo leaves. A total of 30 19-month-old steers were randomly assigned to A. karroo leaves (AK), sunflower cake (SF) and the control with no supplement (CN) diets. The AK and SF diets provided the steers with an additional 150 g of protein per day for 60 days. Catecholamine levels were determined from urine samples collected from each steer before and after slaughter. The Musculus longissimus thoracis et lumborum was sampled for selected meat quality measurements. Nguni steers on the CN diet had higher (P < 0.05) concentrations of post-mortem urinary norepinephrine and dopamine compared with those that received the AK and SF diets. Norepinephrine was negatively linearly related (P < 0.05) to the Warner-Bratzler shear force value of meat aged for 21 days and cooking loss of meat aged for 2 days (CL2) in steers that were given the SF diet. Meat pH and drip loss values were inversely related (P < 0.05) to epinephrine concentration in steers that received the AK diet. Dopamine concentration was negatively linearly related (P < 0.05) to water holding capacity and CL2 for steers on the CN diet. For steers on the CN diet, lightness (L*) values increased (P < 0.05) with increase in dopamine concentration. It was concluded that stress responsiveness and its relationship to certain beef quality attributes could be positively manipulated by supplementation with A. karroo leaves.  相似文献

Cattle breeds indigenous to Africa (Sanga) compare favourably to Bos indicus breeds with regard to adaptation to harsh environments. This study compared the meat quality of three Sanga breeds (Nguni, Tuli and Drakensberger), a Sanga-related breed (Bonsmara) and a B. indicus breed (Brahman) and supported these results with biochemical and histological measurements on the M. longissimus lumborum. Twelve young grain-fed steers of each breed were slaughtered and carcasses were electrically stimulated. All Sanga (and related) breeds, with the exception of the Tuli, had lower Warner-Bratzler shear force (SF) values at 2 and 21 days post mortem compared with the BR (P < 0.05). Measurements related to the calcium-dependent protease system and myofibrillar fragmentation explained the bulk of the variation among breeds, whereas variation in fibre type, sarcomere length and connective tissue properties gave less convincing support. With the exception of the Tuli, Sanga and Sanga-related breeds produced more tender (according to SF) meat than BR, mainly due to favourable calpain-to-calpastatin ratios. Data of Merino sheep descended from two divergent selection lines for (H line) and against (L line) multiple rearing ability were collected in three different trials to compare response to stressful situations between lines. Trial 1: the behaviour of 138, 20-month-old sheep born in 2001 and 124, 8-month-old sheep born in 2002 were observed in an arena test. Experimental animals amounted to 111 and 101 for the H line in the respective years. Corresponding numbers for the L line were 27 and 23, respectively. Progeny born in 2001 were accustomed to handling, as the tests were performed shortly after routine performance recording. Lambs born in 2002 were unaccustomed to handling when tested. Progeny from the H line born in 2002 allowed a smaller (P < 0.01) minimum distance between them and the human operator seated between them and a group of similar aged sheep than L line contemporaries. Progeny born in the L line during 2002 defecated more (P < 0.05) frequently than H line contemporaries. No line differences were found in progeny born during 2001. Trials 2 and 3 were designed to test whether the putative line difference in stress response would impact on physical meat quality. Trial 2: a total of 103, 18–20-month-old progeny from the two selection lines were slaughtered during 2001, 2002–2003. Mean slaughter weight, dressing percentage and carcass weight of H line animals were between 5 and 13% higher (P < 0.05) than that of L line animals. There were no differences in cooking loss, drip loss and tenderness between the two lines. As pertaining to the meat colour, L line progeny had less vivid red meat, as seen by their lower (P < 0.05) a* colour reading and chroma values. The L line animals had lower (P < 0.01) initial pH values as measured in the Musculus longissimus dorsi. These results are indicative of susceptibility to acute, short-term stress. Trial 3: 20 progeny of the two selection lines were slaughtered and their muscle pH measured at 11 intervals from 45 min to 48 h post slaughter. As in Trial 1, the initial pH of the L line animals were lower (P < 0.05) than that of H line animals. An opposite tendency was observed by 48 h post slaughter. The obtained results support a hypothesis that divergent selection for ewe multiple ability resulted in adaptations to cope with unfamiliar conditions.  相似文献

Single nucleotide polymorphisms (SNPs) in the calpain 1 (CAPN1) and calpastatin (CAST) genes were studied to determine their effects on meat tenderness in Bos taurus cattle. Strip loins (M. longissimus dorsi) were removed from cattle in four resource populations after slaughter (n = 1042), aged under controlled conditions until fixed times after rigor mortis, cooked and measured using a tenderometer. Animals were genotyped for the CAPN1 SNP c.947C>G (p.Ala316Gly; AF252504) and for the CAST SNP c.2959A>G (AF159246). Frequencies of CAPN1 C alleles ranged from 23% to 68%, and CAST A alleles from 84% to 99.5%. From all data combined, the CAPN1 CC genotype (compared with the GG genotype) was associated with a 20.1 +/- 1.7% reduced average shear force at intermediate stages of ageing (P < 0.001) and with a 9.5 +/- 1.3% reduction near ultimate tenderness (P < 0.001). The heterozygote was intermediate. For CAST, corresponding values for AA compared with AG genotypes were reductions of 8.6 +/- 2.0% and 5.1 +/- 1.6% respectively (both P < 0.001), but there were too few GG genotypes for comparison. There were small interactions between the CAPN1 and CAST genotypes. For the CAPN1 and CAST genotypes combined, the maximal genotype effect in average shear force was 25.7 +/- 5.5% (P < 0.001) at intermediate stages and 15.2 +/- 4.8% near ultimate tenderness (P < 0.01).  相似文献

The aim of the present study was to evaluate the effect of age on embryogenic competence of oocytes recovered from Bos indicus crossbred calves and heifers. Cumulus-oocyte complexes (COCs) were collected from 4- to 7-month-old calves (experiment 1) and from 9- to 14-month-old heifers (experiment 2) during processing at an abattoir. In both experiments cow COCs were used as control. COCs were in vitro matured and fertilized, and the presumptive zygotes co-cultured with cumulus cells until 224 h post insemination (hpi). In experiment 1, the development rate during the first 68-72 hpi was similar (P > 0.05) between embryos derived from calves and cows. Fewer embryos from calves developed to the blastocyst stage, resulting in a lesser blastocyst production as well as lesser hatching rate (P < 0.05). The embryo development after blastocyst stage was, nevertheless, similar (P > 0.05) between blastocysts derived from calves and cows, suggesting that the development after blastocoele formation is not compromised in embryos derived from calves. In experiment 2, there were no differences (P > 0.05) on cleavage, blastocyst and hatching rates between embryos derived from prepubertal heifers and cows. The rate of blastocyst development until hatching was also similar (P > 0.05). These results indicate that oocytes from 9- to 14-month-old B. indicus crossbred heifers have the same developmental competence as oocytes derived from cows, while ocytes derived from 4- to 7-month-old B. indicus crossbred calves are less competent in developing to the blastocyst stage in vitro. It suggests that oocyte competence in B. indicus crossbred cattle is achieved around 9-14 months of age.  相似文献

In Exp. I oxytocin (60 micrograms/100 kg/day) was infused into the jugular vein of 3 heifers on Days 14-22, 15-18 and 16-19 of the oestrous cycle respectively. In Exp. II 5 heifers were infused with 12 micrograms oxytocin/100 kg/day from Day 15 of the oestrous cycle until clear signs of oestrus. Blood samples were taken from the contralateral jugular vein at 2-h intervals from the start of the infusion. The oestrous cycle before and after treatment served as the controls for each animal. Blood samples were taken less frequently during the control cycles. In Exp. III 3 heifers were infused with 12 micrograms oxytocin/100 kg/day for 50 h before expected oestrus and slaughtered 30-40 min after the end of infusion for determination of oxytocin receptor amounts in the endometrium. Three other heifers slaughtered at the same days of the cycle served as controls. Peripheral concentrations of oxytocin during infusion ranged between 155 and 641 pg/ml in Exp. I and 18 and 25 pg/ml in Exp. II. In 4 our of 8 heifers of Exps I and II, one high pulse of 15-keto-13,14-dihydro-prostaglandin F-2 alpha (PGFM) appeared soon after the start of oxytocin infusion followed by some irregular pulses. The first PGFM pulse was accompanied by a transient (10-14 h) decrease of blood progesterone concentration. High regular pulses of PGFM in all heifers examined were measured between Days 17 and 19 during spontaneous luteolysis. A non-surgical system of embryo recovery is described. It consists of a rigid probe of small diameter (4 mm) for recovering the embryos from young heifers (10 to 12 days after estrus). An average of 6.35 eggs were recovered per donor from 64 heifers 15 to 22 month-old having more than two palpable corpora lutea after superovulation. Forty donors were slaughtered after recovery to determine the number of ovulations, the state of the uterus and to do a post-mortem perfusion. The average recovery rate of embryos was 56.4 % ; an additional 9 % were recovered after slaughter. We compared two recovery methods differing in mode of liquid return ; no significant differences were found. Twenty-four animals not slaughtered after recovery returned to heat a mean 23.8 days after induced estrus (control cycle).  相似文献

Reproductively normal crossbred beef heifers were individually offered a diet of barley straw and concentrate supplemented with one of four levels of a fish oil (FO) enriched supplement. Following oestrous cycle synchronisation, blood samples were collected at appropriate intervals for the measurement of progesterone (P(4)), oestradiol (E(2)), fatty acids, insulin-like growth factor 1 (IGF-1) and metabolites. On days 15 and 16 of the cycle, oxytocin was administered intravenously and the prostaglandin F(2alpha) (PGF(2alpha)) response was measured as venous concentrations of 13,14-dihydro-15-keto PGF(2alpha) (PGFM). The heifers were slaughtered on days 17 or 18 of the oestrous cycle and endometrial tissue, rumen fluid and follicular fluid were collected for determination of fatty acid concentrations. In general there was no effect (P>0.05) of diet on plasma P(4) or E(2) concentrations. Increasing FO supplementation increased CL diameter on day 7 post-oestrus (P<0.0001) but had no effect on diameter on day of slaughter (P>0.05). On day 15, PGFM concentration was greater on the highest level of FO supplementation compared to controls (P<0.05), however, there were no differences between other diet comparisons (P>0.05). There was no effect of diet on PGFM concentration on day 16 (P>0.05). There was a strong positive relationship between plasma and uterine endometrial concentrations of both EPA (R(2)=0.86; P<0.0001) and total n-3 PUFA (R(2)=0.77; P<0.0001). IGF-1 concentrations increased on all diets and were greatest at the highest level of n-3 PUFA supplementation (P<0.05).  相似文献

A total of 200 (Landrace × Large White dam × Pietrain × Large White sire) gilts of 50 ± 3 days of age (23.3 ± 1.47 kg BW) were used to investigate the effects of castration (intact gilt, IG v. castrated gilt, CG) and slaughter weight (SW; 106 v. 122 kg BW) on productive performance, carcass and meat quality. Four treatments were arranged factorially and five replicates of 10 pigs each per treatment. Half of the gilts were ovariectomized at 58 days of age (8 days after the beginning of the trial at 29.8 ± 1.64 kg BW), whereas the other half remained intact. The pigs were slaughtered at 106 or 122 kg BW. Meat samples were taken at Musculus longissimus thoracis at the level of the last rib and subcutaneous fat samples were taken at the tail insertion. For the entire experimental period, CG had higher (P < 0.05) BW gain and higher (P < 0.001) backfat and Musculus gluteus medius fat thickness than IG. However, IG had higher (P < 0.05) loin and trimmed primal cut yields than CG. Meat quality was similar for IG and CG but the proportion of linoleic acid in subcutaneous fat was higher (P < 0.001) for IG. Pigs slaughtered at 122 kg BW had higher (P < 0.001) feed intake and poorer feed efficiency than pigs slaughtered at 106 kg BW. An increase in SW improved (P < 0.001) carcass yield but decreased (P < 0.05) trimmed primal cut yield. Meat from pigs slaughtered at the heavier BW was redder (a*; P < 0.001) and had more (P < 0.01) intramuscular fat and less thawing (P < 0.05) and cooking (P < 0.10) loss than meat from pigs slaughtered at the lighter BW. In addition, pigs slaughtered at 122 kg BW had less (P < 0.01) linoleic acid content in subcutaneous fat than pigs slaughtered at 106 kg BW. Castration of gilts and slaughtering at heavier BW are useful practices for the production of heavy pigs destined to the dry-cured industry in which a certain amount of fat in the carcass is required. In contrast, when the carcasses are destined to fresh meat production, IG slaughtered at 106 kg BW is a more efficient alternative.  相似文献

Trichomonas cultures taken from 2389 bulls showed that approximately 4.7% of them were infected. Correlation of these data with the ranches from which diagnostic samples were obtained indicated that in the period of 1984 through 1987 26.7 to 44.1% of ranches had at least one infected bull. Thirty-four 18-month-old Holstein heifers were assigned to one of three groups, controls n = 12 animals, soluble vaccine n = 11 animals, and whole vaccine n = 11 animals to determine the effect of Tritrichomonas foetus vaccines on the reproductive performance of T . foetus infected animals. Heifers were bred with T . foetus infected bulls beginning two weeks after the second T . foetus vaccination. All immunized animals developed antibody titers of at least 1:1000 following vaccination. In addition, all control and immunized animals became infected with T . foetus . However, the duration of infection was approximately two weeks shorter in immunized animals. Approximately 42% (5 of 12) of control heifers remained infected with T . foetus for the duration of the experiment, while only 18% (2 of 11) of each of the vaccine groups remained infected for the duration of the experiment. Finally, 27% (3 of 11) of heifers in each of the vaccine groups were pregnant at slaughter, while none of the control heifers were pregnant at slaughter. Therefore, both vaccine formulations appeared to protect heifers (P<0.05) from fetal loss due to trichomoniasis.  相似文献

Ractopamine hydrochloride (RH) alters protein metabolism and improves growth performance in Bos taurus cattle with high carcass fat. Our objective was to evaluate the effects of RH, dietary CP and RH×CP interaction on performance, blood metabolites, carcass characteristics and meat quality of young Nellore bulls. A total of 48 bulls were randomly assigned to four treatments in a 2×2 factorial arrangement. The factors were two levels of dietary CP (100% and 120% of metabolizable protein requirement, defined as CP100 and CP120, respectively), and two levels of RH (0 and 300 mg/animal·per day). Treated animal received RH for the final 35 days before slaughter. Animals were weighed at the beginning of the feedlot period (day 63), at the beginning of ractopamine supplementation (day 0), after 18 days of supplementation (day 18) and before slaughter (day 34). Animals were slaughtered and hot carcass weights recorded. After chilling, carcass data was collected and longissimus samples were obtained for determination of meat quality. The 9–11th rib section was removed for carcass composition analysis. Supplementation with RH increased ADG independently of dietary CP. There was a RH×CP interaction on dry matter intake (DMI), where RH reduced DMI at CP120, with no effect at CP100. Ractopamine improved feed efficiency, without RH×CP interaction. Ractopamine had no effect on plasma creatinine and urea concentration. Greater dietary CP tended to increase blood urea, and there was a RH×CP interaction for plasma total protein. Ractopamine supplementation increased plasma total protein at CP120, and had no effect at CP100. Ractopamine also decreased plasma glucose concentration at CP100, but had no effect at CP120. Ractopamine increased alkaline phosphatase activity at CP120 and had no effect at CP100. There was a tendency for RH to increase longissimus muscle area, independently of dietary CP. Ractopamine did not alter fat thickness; however, fat thickness was reduced by greater CP in the diet. Supplementation with RH decreased meat shear force, but only at day 0 of aging, having no effect after 7, 14 or 21 days. Greater dietary protein increased meat shear force after 0 and 7 days of aging, with no effect after 14 or 21 days. These results demonstrate for the first time the efficacy of ractopamine supplementation to improve gain and feed efficiency of intact Bos indicus males, with relatively low carcass fat content. Ractopamine effects were not further improved by increasing dietary protein content above requirements.  相似文献

We investigated the influence of Swedish recommended vitamins A, D3 and E supplementation levels on muscle tenderness and fatty acid (FA) composition under indoor or outdoor finishing programmes. Swedish Red breed steer calves were divided into vitamin supplemented (n = 12) and non-supplemented (n = 15) groups while on pasture prior to the finishing period. This trial began at the beginning of the winter housing period during which the steers were fed a 55 : 45 dry matter barley : grass silage diet indoors. The indoor finished group was comprised of vitamin supplemented (n = 6) and non-supplemented (n = 8) steers slaughtered after about 155 days on feed. Vitamin supplemented steers were provided with 100 g mineral supplement providing 400 000 IU vitamin A, 100 000 IU D3 and 3000 IU E daily as recommended for Swedish production practices. In spring, outdoor finished vitamin supplemented (n = 6) and non-supplemented (n = 7) steers grazed semi-natural grassland for an additional 120 days before slaughter. During pasture, vitamin supplemented steers had free-choice access to a mineral supplement containing vitamins A, D3 and E. The mineral supplement for the non-supplemented steers did not contain vitamins A, D3 and E and was provided at the same amount as the vitamin supplemented steers. Shear force values were similar between vitamin supplemented and non-supplemented steers after ageing 2, 7 and 14 days within indoor and outdoor finishing programmes. The shear force values had decreased by 14 days of ageing within all programmes. The μ- and m-calpain activity did not differ between vitamin supplemented and non-supplemented steers for either the indoor or outdoor finishing programmes. The calpastatin activity was higher for the indoor, vitamin supplemented steers. Indoor finished vitamin supplemented steers had a greater proportion of C18:1c-9 and total monounsaturated fatty acids, whereas the non-supplemented steers had a greater proportion of total saturated fatty acids. We concluded that the meat quality from steers not receiving vitamin supplementation was similar to that of steers receiving vitamins A, D3 and E supplementation at Swedish recommended levels under indoor and outdoor finishing programmes.  相似文献

Sixty-eight 12- to 14-month-old crossbred beef heifers averaging 285 kg were assigned at random to treated (n = 35) and control (n = 33) groups to evaluate the use of repetitive norgestomet treatments. Treated heifers received an ear implant containing 6 mg norgestomet on two occasions 16 days apart. Injections of 3 mg norgestomet and 5 mg estradiol valerate (EV) were given intramuscularly the same day as first implantation (Syncro-Mate-B). Implants were removed after eight days. Four bulls were then placed in each of two pastures containing half of the treated and half of the control heifers for 24 days after the time of the first implant removal. Progesterone concentrations from blood samples collected prior to the first treatment were used to determine reproductive status. The overall pregnancy rate 64 days after first implant removal for treated anestrous heifers (61%; 14 23 ) was similar (p > 0.25) to untreated (73%; 11 15 ) and treated (75%; 9 12 ) cyclic heifers, but higher (p < 0.1) than for untreated anestrous heifers (33%; 6 18 ). This treatment advantage resulted from an increased (p < 0.01) pregnancy rate after the second implant removal. In summary, repetitive norgestomet treatments enhanced pregnancy rate in anestrous heifers within a 24-day breeding season.  相似文献

The objectives were to examine the effects of dietary energy and protein density on age and body composition at puberty, and on ovarian follicular dynamics during the pre- and peripubertal periods in Holstein heifers. In Phase 1, heifers were randomly allotted (n=10 per diet) at 100 kg body weight (BW) to diets with either low (P1L), medium (P1M) or high (P1H) energy and protein formulated for an average daily gain (ADG) of 0.5, 0.8 or 1.1 kg per day, respectively. During Phase 2 (P2), all heifers were fed ad libitum a common diet formulated for an ADG of 0.8 kg per day. Half the animals within the high (n=5) and low groups (n=5) entered P2 either at 12 months of age (P2H-12; P2L-12) or at 330 kg BW (P2H-330; P2L-330). Heifers fed P1H, P1M, P1L, and P2L-12 diets attained puberty at approximately 9, 11, 16, and 14 months of age, respectively (P<0.01). Urea space estimates of body fat and protein percent, and back-fat thickness, were lower in P1L heifers compared to P1H or P1M heifers at similar chronological ages (P<0.05) but did not differ at puberty (P>0.10). Compared to P1L heifers, P1H heifers had high amplitude LH pulses at 8 months, and high frequency low amplitude LH pulses at 10 months of age (P<0.05). The mean diameter (mm) of the dominant follicle was smaller (P<0.05) in P1L heifers (10.6) compared to P1H (12.8) or P1M (12.2) heifers at 8 months. Maximum size and growth rate of the nonovulatory dominant follicle increased with age (P<0.05) but did not differ between P1H and P1M heifers at puberty. The diameter (mm) of the nonovulatory dominant follicle, and the first and second ovulatory follicles were larger in P2L-12 heifers (14.0, 14.7, and 14.9) compared to P1M heifers (13.1, 12.5, and 11.9), while the peak progesterone levels and CL growth were lower (P<0.05) in the first cycle. In conclusion, dairy heifers attained puberty at a constant body weight and body composition independent of dietary manipulation, the size of dominant follicles increased with age in association with increased LH support, and heifers realimented from a low energy diet developed larger first ovulatory follicles and smaller CL with lower peak progesterone concentrations in the first cycle.  相似文献

Thirty-two beef heifers were induced to superovulate by the administration of follicle stimulating hormone-porcine (FSH-P). All heifers received 32 mg FSH-P (total dose) which was injected twice daily in decreasing amounts for 4 d commencing on Days 8 to 10 of the estrous cycle. Cloprostenol was administered at 60 and 72 h after the first injection of FSH-P. Heifers were observed for estrus every 6 h and were slaughtered at known times between 48 to 100 h after the first cloprostenol treatment. The populations of ovulated and nonovulated follicles in the ovaries were quantified immediately after slaughter. Blood samples were taken at 2-h intervals from six heifers from 24 h after cloprostenol treatment until slaughter and the plasma was assayed for luteinizing hormone (LH) concentrations. The interval from cloprostenol injection to the onset of estrus was 41.3 +/- 1.25 h (n = 20). The interval from cloprostenol injection to the preovulatory peak of LH was 43.3 +/- 1.69 h (n = 6). No ovulations were observed in animals slaughtered prior to 64.5 h after cloprostenol (n = 12). After 64.5 h, ovulation had commenced in all animals except in one animal slaughtered at 65.5 h. The ovulation rate varied from 4 to 50 ovulations. Approximately 80% of large follicles (> 10 mm diameter) had ovulated within 12 h of the onset of ovulation. Triploid suspensions generally grew more vigorously in modified MS medium with 2,4-D than those of diploids. The embryogenic potential of 26-month-old auxin-dependent suspension cultures depended on the line. Neither triploid nor diploid BOR (Borszczagowski line) were able to produce somatic embryos. Similarly, 12–20-month-old cytokinin-dependent suspensions from the same triploid line were not capable of regeneration. Only aggregates from 26-month-old auxin-dependent suspension of triploid line 603 differentiated into somatic embryos. In contrast, 18-month-old diploid and triploid liquid cultures of meristematic clumps (LMC) of BOR retained their regeneration potential. The ploidy level of triploid and diploid auxin-dependent suspension cultures was stable during the first 8 months. However, the ploidy level of triploids remained stable over 26 months of culture, whereas 66.7% of diploid cultures underwent chromosome doubling. No ploidy changes were observed among plants regenerated from 18-month-old LMC. Our data suggest that loss of embryogenic potential in suspension culture was independent of ploidy level.  相似文献

Twenty Assaf lambs fed barley straw plus a concentrate alone (CONTROL group) or enriched with naringin (1.5 g/kg DM, NARINGIN group) were used to assess the effect of this polyphenolic compound on meat quality attributes. Serum samples were collected for 7 weeks, then the animals were slaughtered and the livers and longissimus thoracis et lumborum muscles extracted for analysis. Triacylglycerol levels in the serum samples tended to show (P = 0.087) lower average values for the NARINGIN group when compared with the CONTROL, but no differences were observed when the meat was analysed for the intramuscular fat content. Lower thiobarbituric acid-reactive substances procedure (TBARS) values (P < 0.001) in the liver of the NARINGIN group were detected, probably as a consequence of naringenin accumulation in this organ. No significant differences were observed in the meat samples concerning TBARS or colour evolution during refrigerated storage, as not enough naringenin would have reached the muscle. Independent of naringin administration, the low levels of the most atherogenic oxysterols must be highlighted as the most important quality score in the lamb meat samples studied.  相似文献
