The problem of plasmon resonance in spherical metal nanoparticles incorporated into an anisotropic dielectric medium is studied theoretically. Analytic solution is obtained in long-wavelength approximation for the case of weak uniaxial anisotropy. It is shown that medium anisotropy causes shifts of plasmon frequency from its position in an isotropic medium, which are different for plasmons with dipole momenta parallel and perpendicular to the axis of the medium. For the parallel and perpendicular orientations, which are determined by polarization of incident light, plasmon shifts differ by a factor of 4/3. Analytic expressions for field enhancement in the vicinity of a metal nanoparticle is obtained and analyzed for the cases of noble metals. The perspectives of experimental check of the obtained results and possible applications of plasmon anisotropy in plasmonics and sensorics are discussed.  相似文献

We present simple physical and chemical procedures that allow tuning and modification of the topography of gratings present in optical storage discs into geometries optimal for grating coupled plasmon resonance excitation. After proper metal coating, the tuned surfaces exhibit sharp plasmon resonances that can be excited at wavelengths ranging from 260 nm to over 2.7 μm with relatively high quality factors. As an immediate exemplary application, use of such optimized gratings in aqueous medium for refractive index measurement is demonstrated.  相似文献

Genetic variability is an important component of the phenotypic variation within populations; however, there are often many other contributing factors, which receive little attention. Randomization techniques or grouping factors known to contribute to variability can do much to isolate, or at least accommodate, this variability. The European Centre for the Validation of Alternative Methods addressed some of these approaches in 1998, and others are considered in this article. However, laboratory animals are living beings that respond to scientific procedures, or indeed to subtle variations in husbandry conditions, which our senses do not equip us to comprehend readily. These variations may either bias our experimental results or introduce sufficient background noise to mask differences arising from the scientific procedures. In planning an experiment, it is important to devote adequate time considering such factors and developing appropriate strategies to handle them.  相似文献

We present a study of the near-field pattern created by chains of gold nanodiscs situated on a gold thin film and illuminated at oblique incidence. Each disc generates surface plasmon polaritons that propagate on the gold surface. The created waves interfere between them and with the illuminating beam. We observed that when the discs are separated by a distance smaller than the half wavelength, the chain behaves like a continuous ridge. When the discs separation increases, a complex periodic pattern appears and extends up to several wavelengths from the chain. For some specific separation distances, a directional emission of surface plasmon is also observed. The experimental results are in good agreement with numerical simulations performed by considering each disk as an independent dipole-like surface plasmon source.  相似文献

The anticrossing behavior of dispersion curves of the surface plasmon polaritons supported by metal-insulator-metal structures are studied experimentally and theoretically. Samples consisting of a poly(methyl methacrylate) layer sandwiched by Ag films are prepared and their angle- and wavelength-scan attenuated total reflection spectra are measured. From an analysis of the angle-scan spectrum, the coupled-mode nature of the surface plasmon polariton modes is suggested. The dispersion relations obtained from the wavelength-scan spectra exhibit clearly the anticrossing behavior that arises from the coupling of the modes. The experimental dispersion relations are in good agreement with theoretical ones.  相似文献

Preparation of animals is important for optimization of animal welfare as well as to minimize interanimal variation, thereby strengthening the quality of data for in vivo studies. These issues are important in the work of institutional animal care and use committees (IACUCs), but they pose many challenges as well. This article provides IACUC members with a resource for use in determining whether and how preparation of animals for research affects the IACUC's responsibilities. The topics addressed are intended to serve as a starting point for consideration and discussion. Questions related to subject selection and acclimation of subjects to experimental housing and procedures are emphasized and should provide IACUC members with a framework for discussion of relevant questions. Guidelines are provided for promoting the acclimation of a number of species to experimental settings. Additional, potentially controversial points are also raised, including the effects on longitudinal data sets of changing subject preparation procedures. The roles of the IACUC in the research endeavor are multifaceted and continuously evolving. As empirical data are produced that affect additional aspects of animal care and use, it is important for these committees to be able to evaluate and, when appropriate, stimulate the implementation of improved procedures and strategies.  相似文献

Analysis of urine for endogenous thiols and thiol drugs content by HPLC with ultraviolet detection is addressed. Other methodologies for detection and determination of thiols in urine are only mentioned. Outline of metabolism, role of main biological thiols in physiological and pathological processes and their reference concentrations in urine are presented. In particular, urine sample preparation procedures, including reduction of thiol disulfides, chemical derivatization and reversed-phase HPLC separation steps are discussed. Some experimental details of analytical procedures for determination of endogenous thiols cysteine, cysteinylglycine, homocysteine, N-acetylcysteine, thioglycolic acid; and thiol drugs cysteamine, tiopronin, d-penicillamine, captopril, mesna, methimazole, propylthiouracil and thioguanine are reviewed.  相似文献

α-Acetal, ω-alkyne poly(ethylene oxide) was synthesized as building block of glycoconjugated poly(ε-caprolactone)-graft-poly(ethylene oxide) (PCL-g-PEO) copolymers. The alkyne group is indeed instrumental for the PEGylation of a poly(α-azido-ε-caprolactone-co-ε-caprolactone) copolymer by the Huisgen's 1,3 dipolar cycloaddition, i.e., a click reaction. Moreover, deprotection of the acetal end-group of the hydrophilic PEO grafts followed by reductive amination of the accordingly formed aldehyde with an aminated sugar is a valuable strategy of glycoconjugation of the graft copolymer, whose micelles are then potential. A model molecule (fluoresceinamine) was first considered in order to optimize the experimental conditions for the reductive amination. These conditions were then extended to the decoration of the graft copolymer micelles with mannose, which is a targeting agent of dendritic cells and macrophages. The bioavailability of the sugar units at the surface of micelles was investigated by surface plasmon resonance (SPR). The same question was addressed to nanoparticles stabilized by the graft copolymer. Enzyme linked lectin assay (ELLA) confirmed the availability of mannose at the nanoparticle surface.  相似文献

We present decomposition procedures for scheduling semiconductor testing facilities. These facilities are characterized by the presence of different types of work centers, some of which have sequence-dependent setup times and some parallel identical machines. We exploit the structure of the routings in semiconductor testing to develop tailored decomposition procedures that decompose the shop into a number of work centers that are scheduled using specialized procedures. Extensive computational experiments show that these procedures significantly outperform existing methods in reasonable CPU times. These results indicate that decomposition methods can be successfully applied to complex scheduling problems of the type addressed in this paper, as well as the classical job shop problems addressed in previous research.  相似文献

Surgical procedures for hernia surgery are usually performed using prosthetic meshes. In spite of all the improvements in these biomaterials, the perfect match between the prosthesis and the implant site has not been achieved. Thus, new designs of surgical meshes are still being developed. Previous to implantation in humans, the validity of the meshes has to be addressed, and to date experimental studies have been the gold standard in testing and validating new implants. Nevertheless, these procedures involve long periods of time and are expensive. Thus, a computational framework for the simulation of prosthesis and surgical procedures may overcome some disadvantages of the experimental methods. The computational framework includes two computational models for designing and validating the behaviour of new meshes, respectively. Firstly, the beam model, which reproduces the exact geometry of the mesh, is set to design the weave and determine the stiffness of the surgical prosthesis. However, this implies a high computational cost whereas the membrane model, defined within the framework of the large deformation hyperelasticity, is a relatively inexpensive computational tool, which also enables a prosthesis to be included in more complex geometries such as human or animal bodies.  相似文献

Surface plasmon resonance-based biosensors are now acknowledged as robust and reliable instruments to determine the kinetic parameters related to the interactions between biomolecules. These kinetic parameters are used in screening campaigns: there is a considerable interest in reducing the experimental time, thus improving the throughput of the surface plasmon resonance assays. Kinetic parameters are typically obtained by analyzing data from several injections of a given analyte at different concentrations over a surface where its binding partner has been immobilized. It has been already proven that an iterative optimization approach aiming at determining optimal analyte injections to be performed online can significantly reduce the experimentation time devoted to kinetic parameter determination, without any detrimental effect on their standard errors. In this study, we explore the potential of this iterative optimization approach to further reduce experiment duration by combining it with the simultaneous injection of two analytes.  相似文献

The collective electronic excitations in thin Ag films deposited onto the Ni(111) surface were studied by high-resolution electron energy loss spectroscopy. A broad loss peak at 7.7 eV was assigned to the Ag multipole plasmon, in excellent agreement with calculations based on s-d polarization model. Ag multipole plasmon was excited only at grazing incidence. Furthermore, a strong dependence on the impinging energy exists. Multipole plasmon could be measured only for a very strict range of primary electron beam energies and it was excited by electrons scattered at a reflection plane located just underneath the jellium edge. Such mode was found to be dramatically more sensible to the state of the surface with respect to ordinary surface plasmon. Moreover, we report experimental evidence of interference effects in surface plasmon excitation.  相似文献

Functionalized colloidal gold is widely used for qualitative and quantitative detection of specific analytes. We report here a novel modification of gold nanoparticles by digitonin, a glycoside used for precipitating membrane cholesterol. The specific molecular recognition of cholesterol by digitonin gold nanoparticles (DGNP), make it an attractive alternative to the existing enzymatic methods for cholesterol sensing. To enable cholesterol binding, we modified mercapto modified GNPs with digitonin, by a simple esterification reaction. The blue shift in the plasmon absorption spectra of DGNP with different cholesterol concentrations accompanied by a decrease in the absorbance is the principle applied here for the estimation. The observed size reduction followed by cholesterol binding is reasoned due to the enhanced hydophobicity of the surface which in turn expels the water layers associated with the particles prior to cholesterol binding. The method exhibited linearity between concentration of cholesterol and the corresponding absorbance of the plasmon peak, in the range of 160-600 ng/mL with a detection limit of 100±9 ng/mL. Other steroids did not show any binding affinity towards DGNP. The method depicted here has potential for development as an enzyme free sensor for cholesterol although many factors need to be addressed to transform it for assaying samples like blood.  相似文献

In this paper we have addressed the problem of analysing Next Generation Sequencing samples with an expected large biodiversity content. We analysed several well-known 16S rRNA datasets from experimental samples, including both large and short sequences, in numbers of tens of thousands, in addition to carefully crafted synthetic datasets containing more than 7000 OTUs. From this data analysis several patterns were identified and used to develop new guidelines for experimentation in conditions of high biodiversity. We analysed the suitability of different clustering packages for these type of situations, the problem of even sampling, the relative effectiveness of Chao1 and ACE estimators as well as their effect on sampling size for a variety of population distributions. As regards practical analysis procedures, we advocated an approach that retains as much high-quality experimental data as possible. By carefully applying selection rules combining the taxonomic assignment with clustering strategies, we derived a set of recommendations for ultra-sequencing data analysis at high biodiversity levels.  相似文献

New method of experimental determination of local field enhancement at metal nanoparticles is suggested. It uses surface plasmon as a probe. Alternating-sign shift of surface plasmon resonance in copper nanoparticles incorporated in silica matrix has been observed under irradiation by intense femtosecond laser pulse. The red shift of plasmon observed during the action of pump pulse is interpreted as a result of change of dielectric constant of silica matrix due to optical Kerr effect in electric field of pump pulse enhanced in a vicinity of metal nanoparticles. The field enhancement factor is estimated from the value of the observed red shift of plasmon resonance.  相似文献

Dictyostelium allows some of the general problems of eukaryotic biology to be addressed by using molecular genetic tools that are more normally associated with yeast. The genome project, now underway, marks an important increase in the attractiveness of Dictyostelium as an experimental organism and will invite increased 'species hopping' by experimenters. CD2 is a cell adhesion molecule found on the plasma membrane of T-lymphocytes. Its counter-receptor in rat is the structurally related CD48. This interaction is believed to contribute to the adhesion of T-cells to other cells such as cytotoxic targets and antigen presenting cells. Cell-cell adhesion involves the formation of multiple cell adhesion molecule complexes at the cell surface and if cell-cell de-adhesion is to occur, these complexes need to be disrupted. The affinities of cell adhesion molecule interactions are suggested to be relatively weak to allow this de-adhesion of cell-cell interactions. The CD2/CD48 interaction has been studied using recombinant extracellular proteins and the affinity of the interaction of soluble recombinant rat CD2–CD48 has been determined (at 37°C) using surface plasmon resonance (and shown to be weak), with the dissociation constant Kd=60–90 μm. The values determined by surface plasmon resonance results could be affected by the immobilisation of the ligand on the chip and any self-association on the chip. We used three different analytical ultracentrifuge procedures which each allowed the interaction to be studied in free solution without the need for an immobilisation medium. Both sedimentation equilibrium (using direct analysis of the concentration distribution and also modelling of molecular weight versus concentration data) and sedimentation velocity at 5°C yielded dissociation constants in the range of 20– 110 μm, supporting the surface plasmon resonance findings showing that binding between these cell adhesion molecules is relatively weak. These studies also ruled out the presence of any significant self-association of the reactants which could lead to systematic error in the surface plasmon resonance results. In a recent study, we reported the results of a rapid high-throughput expression analysis of the affinity-tagged proteins present in total cell lysates, using a surface plasmon resonance (SPR) imaging protein chip system. In this paper, we describe a novel method, which is able to sequentially carry out a recombinant Escherichia coli culture, as well as the detection and purification of the expressed proteins on a single microwell chip, fabricated on a two-dimensional thin gold film. Following the induction of the protein on the microwell chip, the E. coli cells were lysed on the chip via the addition of lysozymes, and the expressed glutathione S-transferase-fused green fluorescent protein (GST–GFP) was then purified on the chip via affinity interaction with the glutathionylated gold surface of the chip. Finally, the expressed protein was directly detected using the surface plasmon resonance (SPR) imaging system. This system saves a substantial amount of time, experimental resources, and labor, by allowing for the complicated and labor-intensive procedures inherent to the production of recombinant proteins to be conducted on a single microwell chip, simply and economically.M. The light propagation through 1D metallic nanowires in strong light–matter interaction regimes have been analyzed theoretically. The theoretical calculations are based on differential formalism using curvilinear coordinate transformation and Fourier modal methods, and its comparison in the case of nanowires with rectangular cross section was performed. The transformation of local plasmon into surface plasmon polariton at an increasing metal filling factor while changing the width of rectangular nanowires was predicted theoretically. The essential enhancement of local plasmon oscillator strength at transformation to surface plasmon polariton was obtained too.  相似文献

The propagation of a surface plasmon polariton wave at the interface of a metal and an ambichiral nanostructured medium was theoretically investigated in the Kretschmann configuration using transfer matrix method. The dependence of optical absorption linear polarization on structural parameters was reported. The results were compared with those obtained from the interface of a metal and a chiral dielectric medium as a reference structure. We found that multiple plasmon modes are excited at the interface of metal and ambichiral dielectric medium. Our calculations revealed that there exist five plasmon modes for chiral, trigonal, and tetragonal structures; three plasmon modes for pentagonal structure; two plasmon modes for hexagonal structure; and one plasmon mode for dodecagonal structure that propagate with different phase speeds. The obtained results showed that only one plasmon mode occurs at all pitches, while other modes exist at some of the pitches of anisotropic chiral and ambichiral dielectric mediums. The time-averaged Poynting vector versus the thickness of metal film confirmed that the energy of photons of incident light is transferred to surface plasmon polariton quasiparticles and the surface plasmon polariton wave is localized at the interface of metal and ambichiral dielectric medium.  相似文献
