Although species of Upogebiidae historically have been considered filter feeders, recent studies show that many species of this group also deposit feed. In this study, the degrees of trophic specialization of two species of this family, Upogebia omissa and Pomatogebia operculata , were analysed. Feeding mechanisms, stomach contents and morphology of the feeding appendages, with emphasis on setae, were examined. U. omissa , found in sandy substrate, is a generalistic feeder while P. operculata inhabits burrows inside corals, being more specialized in filter feeding. Only 21% of the 57 setal types described were common to both species. Setal types were clustered in three main categories: plumed, serrate and plumodenticulate. No simple setae were found. P. operculata has lower setal diversity, with higher ratio of plumodenticulate to serrate setal types than U. omissa. The 1st and 2nd pereiopods have an important role in collecting food. The mouthparts have two main functions: to brush and retain particles so that food can be transported from the pereiopods to the mouth. Generally in these appendages, the dactyli and basal endites are responsible for brushing particles and the meri and coxal endites for particle retention. The diversity of setal types and complexity of their distribution on the appendages may be related to the necessity to select and triturate particles prior to ingestion, reflecting the differences in trophic strategies utilized by U. omissa and P. operculata. Setal characters appear to be indicators of the relative importance of a specific feeding mode for species of this group.  相似文献

An investigation of the feeding behavior of the acorn worm Harrimania planktophilus suggests a novel form of enteropneust feeding with significant phylogenetic implications. H. planktophilus is a holoinfaunal worm that feeds on deposited sediments, and filter feeds on suspended particles in interstitial pore water. To visualize the particle retention behavior involved in filter feeding, adult animals were held in chilled seawater under low light and fed food coloring and fluorescent particles. The behavior was recorded by videography. Most particles ingested were drawn into the mouth by an incurrent flow created by cilia on the pharyngeal bars and without the aid of mucus. Particles that passed freely through the gill pores averaged 3.04 microm, whereas particles retained in the gut and defecated in the feces averaged 13.9 microm. Food coloring entered the mouth and was pumped through the pharynx at a rate of 0.5-2.0 mm/s. There is no evidence of an endostyle or mucus-net capture mechanism in H. planktophilus, but instead particles are filtered and manipulated by a dense covering of cilia on the pharyngeal gill bars. This study suggests that the filter-feeding pharynx is not an innovation of the chordates, but evolved prior to the evolutionary divergence of the hemichordate-echinoderm clade from the chordates.  相似文献

Herbivores that digest plant material in the fore-stomach can be divided in ruminants and non-ruminants. This study describes the distribution of feed particles (and inorganic material) and dry matter (DM) in the digestive tract of non-ruminant foregut fermenters. Results from passage trials led us to hypothesize that specific particle-sorting mechanisms, as observed in ruminants, are unlikely in non-ruminants. Therefore, no systematic particle size distribution effects (indicative of a sorting mechanism) should be evident in the fore-stomachs of these animals, but differences in fluid and particle retention suggest that differences in fluid concentration (measured as DM) could occur in the foregut of macropods and hippos. The gut content of eleven Bennett's wallabies (Macropus rufogriseus), six collared peccaries (Pecari tajacu), three pygmy hippos (Hexaprotodon liberiensis), two common hippos (Hippopotamus amphibius) and one two-toed sloth (Choloepus didactylus) were analyzed with an emphasis on the fore-stomach. The ventral and dorsal regions in sacciform compartments, and peripheral and central regions in tubular compartments, were examined. Results were not uniform across the species studied. A potential sedimentation mechanism was observed firstly by the accumulation of sand in the fore-stomach of the peccary and sloth, and secondly by the lower DM content in peripheral versus central and ventral versus dorsal regions of the fore-stomach of the wallabies and common hippos, respectively. However, pair-comparisons for different gut regions of wallabies and peccaries yielded no differences in mean particle size between fore-stomach regions. To conclude, some digesta fractionation does occur in the fore-stomach of the studied groups of non-ruminants, but not in a uniform manner, which in turn is in accordance with morphological dissimilarities of their respective foregut structures. The absence of systematic fractionation effects in non-ruminant foregut fermenters emphasizes the innovative character of the sorting mechanism in ruminants.  相似文献

Erosion-Resistant Surfaces Inspired by Tamarisk   总被引:2,自引:0,他引:2  
Tamarisk, a plant that thrives in arid and semi-arid regions, has adapted to blustery conditions by evolving extremely ef- fective and robust anti-erosion surface patterns. However, the details of these unique properties and their structural basis are still unexplored. In this paper, we demonstrate that the tamarisk surface only suffers minor scratches under wind-sand mixture erosion. The results show that the anti-erosion property of bionic sample, inspired by tamarisk surface with different surface morphologies, can be attributed to the flow rotating in the grooves that reduces the particle impact speed. Furthermore, the simulation and experiment on the erosion wear behavior of the bionic samples and bionic centrifugal fan blades show that the bionic surface with V-type groove exhibits the best erosion resistance. The bionic surface on centrifugal fan blades with opti- mum parameters can effectively improve anti-erosion property by 28.97%. This paper show more opportunities for bionic application in improving the anti-erosion performance of moving parts that work under dirt and sand particle environment, such as helicopter rotor blades, airplane propellers, rocket motor nozzles, and pipes that regularly wear out from erosion.  相似文献

1. Suspension feeding by caseless caddisfly larvae (Trichoptera) constitutes a major pathway for energy flow, and strongly influences productivity, in streams and rivers. 2. Consideration of the impact of these animals on lotic ecosystems has been strongly influenced by a single study investigating the efficiency of particle capture of nets built by one species of hydropsychid caddisfly. 3. Using water sampling techniques at appropriate spatial scales, and taking greater consideration of local hydrodynamics than previously, we examined the size‐frequency distribution of particles captured by the nets of Hydropsyche siltalai. Our results confirm that capture nets are selective in terms of particle size, and in addition suggest that this selectivity is for particles likely to provide the most energy. 4. By incorporating estimates of flow diversion around the nets of caseless caddisfly larvae, we show that capture efficiency (CE) is considerably higher than previously estimated, and conclude that more consideration of local hydrodynamics is needed to evaluate the efficiency of particle capture. 5. We use our results to postulate a mechanistic explanation for a recent example of interspecific facilitation, whereby a reduction of near‐bed velocities seen in single species monocultures leads to increased capture rates and local depletion of seston within the region of reduced velocity.  相似文献

The instability of nonviral vectors in aqueous suspensions has stimulated an interest in developing lyophilized formulations for use in gene therapy. Previous work has demonstrated a strong correlation between the maintenance of particle size and retention of transfection rates. Our earlier work has shown that aggregation of nonviral vectors typically occurs during the freezing step of the lyophilization process, and that high concentrations of sugars are capable of maintaining particle size. This study extends these observations, and demonstrates that glass formation is not the mechanism by which sugars protect lipid/DNA complexes during freezing. We also show that polymers (e.g., hydroxyethyl starch) are not capable of preventing aggregation despite their ability to form glasses at relatively high subzero temperatures. Instead, our data suggest that it is the separation of individual particles within the unfrozen fraction that prevents aggregation during freezing, i.e., the particle isolation hypothesis. Furthermore, we suggest that the relatively low surface tension of mono- and disaccharides, as compared to starch, allows phase-separated particles to remain dispersed within the unfrozen excipient solution, which preserves particle size and transfection rates during freezing.  相似文献

Morphological characteristics of the forestomach, as well as reports of a natural diet that mostly excludes monocots, suggest that dikdiks (Madoqua spp.), among smallest extant ruminants, should have a 'moose-type' forestomach physiology characterised by a low degree of selective particle retention. We tested this assumption in a series of feeding experiments with 12 adult Phillip's dikdiks (Madoqua saltiana phillipsi) on three different intake levels per animal, using cobalt-EDTA as a solute marker and a 'conventional' chromium-mordanted fibre (<2 mm; mean particle size 0.63 mm) marker for the particle phase. Body mass had no influence on retention measurements, whereas food intake level clearly had. Drinking water intake was not related to the retention of the solute marker. In contrast to our expectations, the particle marker was retained distinctively longer than the solute marker. Comparisons with results in larger ruminants and with faecal particle sizes measured in dikdiks suggested that in these small animals, the chosen particle marker was above the critical size threshold, above which particle delay in the forestomach is not only due to selective particle retention (as compared to fluids), but additionally due to the ruminal particle sorting mechanism that retains particles above this threshold longer than particles below this threshold. A second study with a similar marker of a lower mean particle size (0.17 mm, which is below the faecal particle size reported for dikdiks) resulted in particle and fluid retention patterns similar to those documented in other 'moose-type' ruminants. Nevertheless, even this smaller particle marker yielded retention times that were longer than those predicted by allometric equations based on quarter-power scaling, providing further support for observations that small ruminants generally achieve longer retention times and higher digestive efficiencies than expected based on their body size.  相似文献

随着流动沙丘的不断固定,不同地形部位的土壤机械组成、有机碳(SOC)和全氮(TN)含量的变化存在差异。为了探明固定沙丘的土壤机械组成与土壤养分空间变异特征,本研究以宁夏黄沙古渡固定沙丘为对象,分析了固定沙丘不同部位、不同土层的风沙土机械组成、SOC和TN特征。结果表明: 各土层土壤机械组成均以中沙和细沙为主。SOC和TN有明显的表聚作用,SOC和TN含量最大值分别为5.781和0.412 g·kg-1,分布在丘间地,脊线和背风坡含量最少。随着土层深度的增加,背风坡和迎风坡的SOC含量逐渐减少,丘间地的SOC含量先减少后增加。SOC和TN在小尺度上均表现出明显的空间异质性;SOC和TN与粉沙和极细沙含量呈显著正相关,与中沙和粗沙含量呈显著负相关。土壤机械组成对SOC和TN含量有明显影响,而且SOC和TN含量随着细颗粒含量的增加而增加,表明土壤细颗粒对有机质的吸附和积累起着重要作用。  相似文献

Virus retentive filters are a key product safety measure for biopharmaceuticals. A simplistic perception is that they function solely based on a size‐based particle removal mechanism of mechanical sieving and retention of particles based on their hydrodynamic size. Recent observations have revealed a more nuanced picture, indicating that changes in viral particle retention can result from process pressure and/or flow interruptions. In this study, a mechanistic investigation was performed to help identify a potential mechanism leading to the reported reduced particle retention in small virus filters. Myeloid dendritic cells (DCs) can capture HIV-1 via the receptor CD169/Siglec-1 that binds to the ganglioside, GM3, in the virus particle membrane. In turn, HIV-1 particles captured by CD169, an I-type lectin, whose expression on DCs is enhanced upon maturation with LPS, are protected from degradation in CD169+ virus-containing compartments (VCCs) and disseminated to CD4+ T cells, a mechanism of DC-mediated HIV-1 trans-infection. In this study, we describe the mechanism of VCC formation and its role in immune evasion mechanisms of HIV-1. We find HIV-1-induced formation of VCCs is restricted to myeloid cells, and that the cytoplasmic tail of CD169 is dispensable for HIV-1 trafficking and retention within VCCs and subsequent trans-infection to CD4+ T cells. Interestingly, introduction of a di-aromatic endocytic motif in the cytoplasmic tail of CD169 that results in endocytosis of HIV-1 particles, suppressed CD169-mediated HIV-1 trans-infection. Furthermore, super-resolution microscopy revealed close association of CD169 and HIV-1 particles in surface-accessible but deep plasma membrane invaginations. Intriguingly, HIV-1 particles in deep VCCs were inefficiently accessed by anti-gp120 broadly neutralizing antibodies, VRC01 and NIH45-46 G54W, and thus were less susceptible to neutralization. Our study suggests that HIV-1 capture by CD169 can provide virus evasion from both innate (phagocytosis) and adaptive immune responses.  相似文献

Dendritic cells (DCs) capture human immunodeficiency virus (HIV) through a non-fusogenic mechanism that enables viral transmission to CD4(+) T cells, contributing to in vivo viral dissemination. Although previous studies have provided important clues to cell-free viral capture by mature DCs (mDCs), dynamic and kinetic insight on this process is still missing. Here, we used three-dimensional video microscopy and single-particle tracking approaches to dynamically dissect both cell-free and cell-associated viral capture by living mDCs. We show that cell-free virus capture by mDCs operates through three sequential phases: virus binding through specific determinants expressed in the viral particle, polarized or directional movements toward concrete regions of the cell membrane and virus accumulation in a sac-like structure where trapped viral particles display a hindered diffusive behavior. Moreover, real-time imaging of cell-associated viral transfer to mDCs showed a similar dynamics to that exhibited by cell-free virus endocytosis leading to viral accumulation in compartments. However, cell-associated HIV type 1 transfer to mDCs was the most effective pathway, boosted throughout enhanced cellular contacts with infected CD4(+) T cells. Our results suggest that in lymphoid tissues, mDC viral uptake could occur either by encountering cell-free or cell-associated virus produced by infected cells generating the perfect scenario to promote HIV pathogenesis and impact disease progression.  相似文献

Many species of gekkotans possess adhesive subdigital pads that allow them to adhere to, and move on, a wide variety of surfaces. The natural surfaces exploited by these lizards may be rough, undulant and unpredictable and therefore likely provide only limited, patchy areas for adhesive contact. Here, we examine the microtopography of rock surfaces used by seven species of Rhoptropus and compare this to several rough and smooth artificial surfaces employed in previous studies of gekkotan adhesion. These data are considered in relation to the form, configuration, compliance and functional morphology of the setal fields of these species. Our results demonstrate that natural rock surfaces are rough and unpredictable at the scale of the setal arrays, with equal amounts of variation existing within and between the various types of rock surfaces examined. Such surfaces differ from smooth and rough artificial surfaces in the proportion of surface area available for attachment and the relative predictability of surface undulance. Generally, setal field characteristics of individual species are not relatable to specific substrates, but instead are configured to allow for sufficient attachment to a wide variety of unpredictable surfaces. Our findings provide insight into the evolution and microanatomy of the adhesive system of gekkotan lizards and its adaptive relationship to topographically unpredictable surfaces.  相似文献

Trap‐building antlion larvae detect their prey according to the substrate vibrations produced during movement of the prey on the sand surface. Although most studies are devoted to surface vibrational waves, in the present study, we determine the role of vibrations travelling through deeper sand layers. A behavioural experiment confirms that vibrational stimuli from prey insects on the surface of the sand stimulate the antlions buried in deeper sand layers to move towards the surface. Sand depth and particle size both have a strong effect on signal transmission. The damping coefficient (α10) varies from 0.49 dB to 3.30 dB cm?1 and depends on frequency (in the range from 100 to 300 Hz), particle size (from finest to coarse sand) and distance from the source of the vibrations. The deeper the sand, the narrower the frequency range of the signal becomes. Sand is a filter for higher frequencies. The smaller the sand particles, the more intense the filtering becomes. Fine sand with a mean sand particle size of 360 μm is a more efficient filter than coarse sand; consequently, high frequencies (> 2.5 kHz) are eliminated at a depth of 3 cm. Mean frequency depends on both depth and particle size. However, low frequency signals still propagate at a certain distance, which is biologically important in prey detection. Although the most efficient signal propagation appears to occur in coarse sand, it contains overly large particles that are inconvenient for relatively small antlion larvae. Predators seek a compromise between fine and coarse sand choosing medium sand.  相似文献

A local disruption of the metachronal wave always accompanies capture of algal cells by tentacles of Flustrellidra hispida (Fabricius). Beat changes for ≈0.2 s over ≈100μm of the ciliated band during capture of a 10-μm particle. The halted parcel of water is therefore larger than the particle of food but much smaller than the flow that continues past the tentacles elsewhere. These events are consistent with the hypothesis that an induced local reversal of beat concentrates particles for those suspension feeders that retain particles upstream from a band of simple cilia (adults or larvae of bryozoans, brachiopods, phoronids, hemichordates, and echinoderms). These events are not explained by other hypotheses that have been advanced for concentration of particles by these suspension feeders. Aerosol filtration models of direct interception are not applicable to this type of ciliary suspension feeder because retention depends on the magnitude of a stimulus and response to it. The stimulus will not be the same function of diameter of the food particle, and response is unlikely below a threshold stimulus.  相似文献

Current research efforts to improve immunoassay-biosensor functionality have centered on detection through the optimal design of microfluidic chambers, electrical circuitry, optical sensing elements, and so on. To date, little attention has been paid to the immunoassay-biosensor membrane surface on which interactions between antibodies and antigens must occur. For this reason, the objective of the present study was to manipulate the nanometer surface roughness of a model immunoassay-biosensor membrane to determine its role on sensitivity and specificity. It was hypothesized that surface roughness characteristics similar to those used by the body's own immune system with B-lymphocyte cell membranes would promote antigen-antibody interactions and minimize non-specific binding. To test this hypothesis, polystyrene 96-well plate surfaces were modified to possess similar topographies as those of B-lymphocyte cell membranes. This was accomplished by immobilizing Protein A conjugated gold particles and Protein A conjugated polystyrene particles ranging in sizes from 40 to 860 nm to the bottom of polystyrene wells. Atomic force microscopy results provided evidence of well-dispersed immunoassay-biosensor surfaces for all particles tested with high degrees of biologically inspired nanometer roughness. Testing the functionality of these immunosurfaces using antigenic fluorescent microspheres showed that specific antigen capture increased with greater nanometer surface roughness while nonspecific antigen capture did not correlate with surface roughness. In this manner, results from this study suggest that large degrees of biologically inspired nanometer surface roughness not only increases the amount of immobilized antibodies onto the immunosurface membrane, but it also enhances the functionality of those antibodies for optimal antigen capture, criteria critical for improving immunoassay-biosensor sensitivity and specificity.  相似文献

Climbing assisted by adhesive subdigital pads in gekkotan lizardshas been the subject of intrigue and study for centuries. Manyhypotheses have been advanced to explain the mechanism of adhesion,and recently this phenomenon has been investigated at the levelof individual setae. The ability to isolate, manipulate andrecord adhesive forces from individual setae has provided newinsights, not only into the mechanism of attachment, but alsointo the physical orientation of these structures necessaryto establish attachment, maximize adhesive force, and effectsubsequent release. This, in turn, has enabled a reassessmentof the overall morphology and mode of operation of the adhesivesystem. Digital hyperextension has often been noted as a behavioralcharacteristic associated with the deployment of the gekkotanadhesive system—this is now understandable in the contextof setal attachment and release kinematics, and in the contextof the evolution of this pattern of digital movement from theprimitive pattern of saurian digital kinematics. The perpendicularand parallel preloads associated with setal attachment are nowreconcilable with other morphological aspects of the gekkotanadhesive system—the lateral digital tendon complex andthe vascular sinus network, respectively. Future investigationsof the integrated adhesive system will help to further elucidatethe interdependence of its structural and functional components.  相似文献

Understanding the impact distribution of particles entering the human respiratory system is of primary importance as it concerns not only atmospheric pollutants or dusts of various kinds but also the efficiency of aerosol therapy and drug delivery. To model this process, current approaches consist of increasingly complex computations of the aerodynamics and particle capture phenomena, performed in geometries trying to mimic lungs in a more and more realistic manner for as many airway generations as possible. Their capture results from the complex interplay between the details of the aerodynamic streamlines and the particle drag mechanics in the resulting flow. In contrast, the present work proposes a major simplification valid for most airway generations at quiet breathing. Within this context, focusing on particle escape rather than capture reveals a simpler structure in the entire process. When gravity can be neglected, we show by computing the escape rates in various model geometries that, although still complicated, the escape process can be depicted as a multiplicative escape cascade in which each elementary step is associated with a single bifurcation. As a net result, understanding of the particle capture may not require computing particle deposition in the entire lung structure but can be abbreviated in some regions using our simpler approach of successive computations in single realistic bifurcations. Introducing gravity back into our model, we show that this multiplicative model can still be successfully applied on up to nine generations, depending on particle type and breathing conditions.  相似文献

Comparative phylogenetic and phylogeographic analyses have revealed a pervasive midpeninsular divergence in the mitochondrial genealogies of numerous vertebrate taxa distributed on the Baja California Peninsula. In this study, we extend the investigation of regional vicariance in Baja California to an arthropod taxon by examining patterns of phylogenetic and morphological divergence in the spider genus Homalonychus (Araneae, Homalonychidae). We analyzed data from two mtDNA genes (16S rRNA and NADH dehydrogenase subunit (1) and a nuclear gene (28S rRNA) using maximum parsimony and Bayesian phylogenetic analyses, and also conducted geometric morphometric analyses employing landmark data on male and female genitalia. Genes and morphology both reveal a deep split across the Colorado River and Gulf of California, separating Homalonychus selenopoides on the east side of river from its congener Homalonychus theologus on the west side of the river, including the Baja California Peninsula. Along the north-south axis of the Baja Peninsula, an apparently more recent midpeninsular phylogenetic break is evident within H. theologus in the mitochondrial genome and in female genitalia. However, there is no measurable divergence between northern and southern populations in either nuclear DNA or male genitalia. We suggest that this discordance between datasets reflects either a difference in rates of evolution between male versus female systems, or that male-based nuclear gene flow is obscuring a phylogenetic split that is fixed in the female-based systems. Our findings provide additional support for a midpeninsular Baja divergence event, although the timing and geological evidence for such an event remain elusive.  相似文献

Freeze-fracture and freeze-etch techniques have been employed to study the supramolecular structure of isolated spinach chloroplast membranes and to monitor structural changes associated with in vitro unstacking and restacking of these membranes. High-resolution particle size histograms prepared from the four fracture faces of normal chloroplast membranes reveal the presence of four distinct categories of intramembranous particles that are nonrandomly distributed between grana and stroma membranes. The large surface particles show a one to one relationship with the EF-face particles. Since the distribution of these particles between grana and stroma membranes coincides with the distribution of photosystem II (PS II) activity, it is argued that they could be structural equivalents of PS II complexes. An interpretative model depicting the structural relationship between all categories of particles is presented. Experimental unstacking of chloroplast membranes in low-salt medium for at least 45 min leads to a reorganization of the lamellae and to a concomitant intermixing of the different categories of membrane particles by means of translational movements in the plane of the membrane. In vitro restacking of such experimentally unstacked chloroplast membranes can be achieved by adding 2-20 mM MgCl2 or 100-200 mM NaCl to the membrane suspension. Membranes allowed to restack for at least 1 h at room temperature demonstrate a resegregation of the EF-face particles into the newly formed stacked membrane regions to yield a pattern and a size distribution nearly indistinguishable from the normally stacked controls. Restacking occurs in two steps: a rapid adhesion of adjoining stromal membrane surfaces with little particle movement, and a slower diffusion of additional large intramembranous particles into the stacked regions where they become trapped. Chlorophyll a:chlorophyll b ratios of membrane fraction obtained from normal, unstacked, and restacked membranes show that the particle movements are paralleled by movements of pigment molecules. The directed and reversible movements of membrane particles in isolated chloroplasts are compared with those reported for particles of plasma membranes.  相似文献

This study describes the morphology and distribution of setae on the lateral and medial flagella of the antennules of the spiny lobster Panulirus argus in an effort to identify antennular chemoreceptors in addition to the well-studied aesthetasc chemosensilla. Setae were examined using light and electron microscopy, and their distribution on flagellar annuli was analyzed. We identified ten setal types based on external morphology: hooded, plumose, short setuled, long simple, medium simple, short simple, aesthetasc, guard, companion, and asymmetric setae, with the last four types being unique to the "tuft" located on the distal half of the lateral flagellum. The three setal types whose ultrastructure was examined--hooded, long simple, and medium simple setae--had characteristics of bimodal (chemo-mechanoreceptive) sensilla. The antennules have four distinct annular types based on their setal complement, as shown by cluster analysis. This basic distribution of non-tuft setal types is similar for both lateral and medial flagella. Annuli in the tuft region have tuft setal types superimposed on a basic organization of non-tuft setal types. These results show that the antennules possess a diverse set of setae, that these setae have a highly ordered arrangement on the antennules, that at least four (and probably many more) of these setal types are chemosensilla, and suggest that most antennular chemosensilla are bimodally sensitive.  相似文献
