BACKGROUND AND AIMS: Polyploidy is a central force structuring genetic diversity in angiosperms, but its ecological significance and modes of origin are not fully understood. This work investigated the patterns of coexistence and molecular relatedness of polyploids in the perennial herb, Arnica cordifolia. METHODS: The local- and broad-scale distributions of cytotypes were analysed using flow cytometry. Samples were collected from both roadside and understorey habitats to test the hypothesis of niche separation between triploids and tetraploids. The nuclear rDNA internal transcribed spacer (ITS) and plastid rpl16 spacer, trnL intron plus trnL-trnF spacer and trnK 3' intron regions were sequenced. KEY RESULTS: Broad-scale sampling established that both triploids and tetraploids were common throughout the range of the species, pentaploids were rare, and diploids were not found. Local-scale sampling revealed coexistence of both triploids and tetraploids within the majority of sites. Triploids and tetraploids were equally represented in the understorey and roadside habitat. Triploids were more variable than tetraploids, but both cytotypes shared polymorphisms in ITS. CONCLUSIONS: Coexistence of cytotypes appears to be the norm in A. cordifolia, but habitat differentiation (roadside vs. understorey) is not supported as a coexistence mechanism. Molecular analyses supported multiple events creating triploids but revealed a lack of variation in the tetraploids. Additionally, sequence polymorphisms in ITS suggested a hybridization event prior to polyploidization.  相似文献

Populations of apomictic seedlings of clones ofCitrus species,Citrus hybrids, andPoncirus in the sub-family Aurantioideae were examined for spontaneous tetraploids as a source of materials for use in breeding experiments. Diagnostic features found useful in identifying nucellar tetraploids were leaf shape, petiole blade shape, leaf blade thickness, leaf color, comparative size differences in leaf venation, oil glands, and stomata, stem thickness, and relative size and developmental pattern of the root system. In older or bearing-age plants, nucellar tetraploids may be identified by differences in growth habit, vigor, size, time of growth initiation and bloom, and flower and fruit characteristics. Data are given for tetraploid frequency in glasshouse-grown, first-year nucellar seedlings of 42 populations of 32 clones of different genetic and seed origin and for tetraploid frequency in commercialnursery nucellar seedlings of the Carrizo rootstock clone in two consecutive years. Comparative data are given for quantitative development of roots, stems, and leaves of tetraploids and similar diploid nucellar seedlings. The data suggest that the ability to produce tetraploid apomictic seedlings is a variable genetic trait present in all or nearly all clones able to reproduce by adventitious embryony. Aspects of tetraploid nucellar seedlings that might warrant their testing as tree-size-controlling rootstocks in commercial citrus growing are discussed.  相似文献

采用染色体压片技术对云贵高原17个多星韭居群,共412株个体进行染色体数目和核型研究。结合已有的细胞学资料,对云贵高原多星韭多倍体分布格局进行了研究。9个二倍体居群、6个二倍体与四倍体混生居群分布于云南中部至西北部,12个四倍体居群分布于整个云贵高原。二倍体除香格里拉d和保山居群核型类型为3A型外,其余均为2A型,四倍体除会泽和赫章居群为2 B型外,其余均为2A型。云南西北部四倍体核型不对称指数低于其它分布区,推测该地区四倍体可能起源较早。多星韭二倍体与四倍体核型类型和形态并无明显差异,表明多星韭四倍体可能为同源多倍体。  相似文献

以多胚系山金柑(Fortunella hindsii Swingle)为材料,采用“观根辨叶看油胞”形态初选法,从1289株实生后代筛选出疑似四倍体(双二倍体)8株,流式细胞仪检测和SSR分子鉴定表明它们均为同源四倍体,初选准确率100%,群体自然发生率0.62%。对其形态和初生代谢物进行检测,结果显示:山金柑四倍体株高、茎粗、节间数、节间长、气孔密度均显著低于二倍体,而叶片厚度、气孔大小显著高于二倍体;GC-MS分析鉴定到24种初生代谢物,四倍体叶片奎宁酸含量显著高于二倍体,肌醇、4-氨基丁酸和1-棕榈酸单甘油酯含量显著低于二倍体。  相似文献

Citrus limon (L.) Osbeck cultivated all over the world is a valuable source of aromatic essential oil. To develop tetraploids of C. limon, four different concentration of colchicine (0.025, 0.05, 0.1 and 0.2%; w/v) and three varied exposure time (12, 24 and 48 h) were employed. The ploidy level of diploids (2n = 2x = 18) and tetraploids (2n = 4x = 36) were determined by direct chromosome count and confirmed by flow cytometric analyses. Successful result with maximum tetraploidy frequency was observed in plantlets developed from seeds treated with 0.025% colchicine for 24 h. Morphological and stomatal characteristics indicated that tetraploids were taller with increased leaf and root length. On the other side, the leaves of tetraploids had fewer and larger stomata with a greater number of chloroplasts in guard cells in contrast with diploids. GC–GC/MS analyses showed cyclic monoterpene, limonene had increased significantly in tetraploids and was further confirmed by HPLC quantification. RT-PCR analyses revealed unaltered expression of monoterpene synthase, sesquiterpene synthase and flavone synthase and remarkable upregulation of genes such as limonene synthase, chalcone synthase and phenylalanine ammonia lyase in tetraploids. Antioxidant activity of essential oil of tetraploids was higher than diploids in all the five test systems studied. Overall, the findings of the present study prove that colchicine induced tetraploidy in C. limon could be a greater source of essential oil with improved composition and of economic significance.  相似文献

Photosynthetic rate (P N), SPAD value, specific leaf area (SLA), flag leaf area (FLA), and nitrogen content (LN) of genus Oryza were investigated and their correlation was analyzed to assess some of the main photosynthetic traits among different species in the genus Oryza. The results revealed wide variation in these traits. The species O. rufipogon and O. australiensis exhibited maximum photosynthetic rate. Comparison of different types of genomes (diploid: 2n=2x=24; tetraploid: 2n=4x=48) and growth habit (shade- or sun-grown) showed the species of diploid (with genome symbol EE; 2n=2x=24) genomes, with perennial and sun-grown species, had high apparent photosynthesis compared to others. The species with BB/BBCC, shade-grown and the tetraploids showed high SPAD value, and the flag leaf in sun-grown species and diploids were thicker (low SLA) compared with others. However, no significant difference could be noticed among the different types of genomes. Higher leaf area was noticed among the species of CC/CCDD genome, perennial shade-grown species and tetraploids than in others. The variety IR 36 exhibited highest leaf nitrogen concentration. Correlation analysis showed a strong relationship between P N and leaf nitrogen concentration while no marked relationships were observed among other characteristics. It implies that the species with thick and small leaves with high nitrogen concentration and high photosynthesis evolved better than others. O. rufipogon, with the same genome as O. sativa, could be one of the wild rice resources for elite crop improvement.  相似文献

The main aim of this work was to study the leaf secondary metabolite profiles of artificially induced tetraploids (2n=4x=48) of Solanum commersonii, a diploid (2n=2x=24) wild potato species. The tetraploid genotypes of S. commersonii were produced by oryzalin treatment. Both HPLC-UV and LC/MS analyses revealed that there were no qualitative differences in the metabolite profiles between the diploid S. commersonii and its tetraploids. By contrast, the results showed that the phenylpropanoid content was generally significantly higher in the tetraploids than in the diploid S. commersonii. Concerning the glycoalkaloids (GAs), the results provided evidence that the content of minor GAs (solanidenediol triose, solanidadienol lycotetraose, and solanidenol lycotetraose) was higher in tetraploids than in the diploid progenitor, while the content of major GAs (dehydrodemissine and dehydrocommersonine) was significantly higher in diploid S. commersonii than in its tetraploid genotypes. The results are discussed from the practical perspective of potato biodiversity enhancement.  相似文献

Detailed ecological, morphological and molecular analyses were performed in mixed populations of diploid and autotetraploid Dactylorhiza maculata s.l. in Scandinavia. Comparisons were made with pure populations of either diploid ssp. fuchsii or tetraploid ssp. maculata. It was shown that mixed populations are the result of secondary contact between ssp. fuchsii and ssp. maculata. No patterns of recent and local autopolyploidization were found. Morphology and nuclear DNA markers (internal transcribed spacers of nuclear ribosomal DNA) showed that diploids and tetraploids from mixed populations have similar levels of differentiation to diploids and tetraploids from pure populations. Vegetation analyses, as well as analyses of environmental variables, revealed that diploid and tetraploid individuals in mixed populations are ecologically well differentiated on a microhabitat level. Diploids and tetraploids in pure populations have wider ecological amplitudes than they do in mixed populations. Triploid hybrids grew in intermediate microhabitats between diploids and tetraploids in the mixed populations. Plastid DNA markers indicated that both diploids and tetraploids may act as the maternal parent. Based on morphology and nuclear markers triploids are more similar to tetraploids than to diploids. There were indications of introgressive gene flow between ploidy levels. Plastid markers indicated that gene flow from diploid to tetraploid level is most common, but nuclear markers suggested that gene flow in opposite direction also may occur. Similar patterns of differentiation and gene flow appeared in localities that represented contrasting biogeographic regions. Disturbance and topography may explain why hybridization was slightly more common and the differentiation patterns somewhat less clear in the Scandinavian mountains than in the coastal lowland. 以二倍体野生一粒小麦(Triticum boeoticum)、栽培一粒小麦(T. monococcum)、节节麦(Aegilops tauschii)和黑麦(Secale cereale)、四倍体野生二粒小麦(T. dicoccoides)、栽培二粒小麦(T. dicoccum)、硬粒小麦(T. durum)、六倍体普通小麦(T. aestivum)‘扬麦9号’和‘扬麦158’及八倍体小黑麦(Triticale)为材料,采用盆栽试验研究了不同小麦进化材料生育后期旗叶光合特性的演变及产量的差异。结果表明,与六倍体普通小麦和八倍体小黑麦相比,二倍体和四倍体材料在开花前具有较高的光合速率(Pn)、气孔导度(Gs)、最大光能转换效率(Fv/Fm)和实际光化学效率(ΦPSⅡ)。开花以后,二倍体和四倍体材料受非气孔因素的影响,光合能力下降较快;除黑麦外,旗叶光合速率在开花10 d后都低于普通小麦和小黑麦,胞间CO2浓度(Ci)迅速增加,Fv/FmΦPSⅡ和叶绿素含量快速下降。二倍体和四倍体材料开花前单株总叶面积和旗叶叶面积较大,花后下降迅速,功能期短;单株穗数也较多,但穗粒数、千粒重、产量和收获指数却显著低于普通小麦。因此,小麦长期进化过程中,普通小麦花后较高的光合能力及较长的光合持续期是提高千粒重,进而提高产量的重要生理基础。  相似文献

The influence of polyploidization on ribulose-1,5-bisphosphate carboxylase (RuBPCase), buffer-soluble protein (BSP), chlorophyll (Chl), and DNA was examined in fully expanded leaves of isogenic diploid-tetraploid (DDC 2X-4X) and tetraploid-octoploid (IC 4X-8X) sets of alfalfa (Medicago sativa L.). The concentration of RuBPCase in leaf extracts was determined by rocket immunoelectrophoresis. Activities of RuBPCase, expressed per milligram protein or per milligram Chl, and leaf tissue concentrations of RuBPCase, BSP, Chl, and DNA were similar between ploidy levels of the DDC 2X-4X set. Tetraploids and octoploids were similar in RuBPCase activities, expressed per milligram protein or per milligram Chl, and in leaf tissue concentrations of RuBPCase and DNA. Octoploids were significantly lower than tetraploids in concentrations of Chl and BSP.

When compared on a per leaf basis, tetraploids were 80% higher in BSP and essentially double comparable diploids in fresh weight, RuBPCase, Chl, and DNA. The observation that leaves of the DDC tetraploid population contain twice as much DNA as comparable diploids suggests that leaves of both ploidy levels contain similar numbers of cells. Leaves of the octoploid population were 33% to 80% higher than corresponding tetraploids in BSP, fresh weight, RuBPCase, Chl, and DNA. Ratios of RuBPCase to DNA and Chl to DNA were similar across ploidy levels of both isogenic sets suggesting that cellular content of Chl and RuBPCase increases proportionately with the amount of DNA per cell.

Current molecular characterization of ex situ plant germplasm has placed more emphasis on cultivated gene pools and less on exotic gene pools representing wild relative species. This study attempted to characterize a selected set of germplasm accessions representing various Avena species with the hope to establish a reference set of exotic oat germplasm for oat breeding and research. The amplified fragment length polymorphism (AFLP) technique was applied to screen 163 accessions of 25 Avena species with diverse geographic origins. For each accession, 413 AFLP polymorphic bands detected by five AFLP primer pairs were scored. The frequencies of polymorphic bands ranged from 0.006 to 0.994 and averaged 0.468. Analysis of molecular variance revealed 59.5% of the total AFLP variation resided among 25 oat species, 45.9% among six assessed sections of the genus, 36.1% among three existing ploidy levels, and 50.8% among eight defined genome types. All the species were clustered together according to their ploidy levels. The C genome diploids appeared to be the most distinct, followed by the Ac genome diploid A. canariensis. The Ac genome seemed to be the oldest in all the A genomes, followed by the As, Al and Ad genomes. The AC genome tetraploids were more related to the ACD genome hexaploids than the AB genome tetraploids. Analysis of AFLP similarity suggested that the AC genome tetraploid A. maroccana was likely derived from the Cp genome diploid A. eriantha and the As genome diploid A. wiestii, and might be the progenitor of the ACD genome hexaploids. These AFLP patterns are significant for our understanding of the evolutionary pathways of Avena species and genomes, for establishing reference sets of exotic oat germplasm, and for exploring new exotic sources of genes for oat improvement.  相似文献

Continuous leaf phenology, with sequential production of metamers, is usually found in pioneer species, but is rare in shade‐tolerant species. Because of the nature of ‘continuous’ growth, continuous phenology has seldom been related to meteorological factors. We studied the leaf demography of seedlings and tall trees of a shade‐tolerant emergent species, Dipterocarpus sublamellatus (Dipterocarpaceae), in Malaysia. Although leaf phenology of D. sublamellatus was classified as continuous, the rates of leaf production and leaf fall were not constant and greatly fluctuated during the census period. Seasonal patterns of leaf production and fall were not synchronized across tall trees in most combinations or between seedlings and tall trees. Mean number of leaves produced per month and its seasonal fluctuation were greater in tall trees than in seedlings. Moreover, relationships between leaf phenology and meteorological factors (monthly rainfall, net radiation, maximum and minimum temperature, and minimum relative humidity) differed among trees. This suggests that endogenous rhythms, i.e., differences in allocation of resources to growth and storage among trees, are important to fluctuations in leaf phenology. These patterns are in contrast to patterns found in various pioneer species with continuous phenology, in which rates of leaf production and fall are generally constant. Different patterns might be caused by differences in allocation to growth and storage between pioneer and shade‐tolerant species, and might be related to their regeneration strategies.  相似文献

Seventeen greenhouse-grown grasses from the Nebraska Sandhills region were surveyed for foliar stomatal density and distribution, closed guard cell lengths, open stomatal apertures, and surface characteristics (using scanning electron microscopy), surface conductance (using a steady-state porometer), and drought-induced leaf modifications. Leaves of C3 species exhibited a proclivity toward being amphistomatic or hyperstomatic, while C4 species tended to be more hypostomatic. Leaf modification, when it occurred, resulted in the enshrouding of the adaxial surface. Conductance data showed functional amphistomaty in most species, revealing differential functioning of adaxial and abaxial stomata. Conductance patterns were not closely related to stomatal aperture per unit area leaf surface or to stomatal distribution patterns. Lowered adaxial: abaxial conductance ratios, increased stomatal density, reduced stomatal size, and less drought-induced leaf modification were seen in C4 grasses as compared with C3 grasses. C3 range and C3 meadow species did not differ in conductance ratios, density ratios, or stomatal size, although meadow species exhibited much greater drought-induced leaf modification. Postulations involving correlation of adaxial: abaxial conductance ratios to stomatal distribution patterns, and assumptions of stomatal distribution based upon habitat and/or photosynthetic pathway may be erroneous. These characteristics may be of limited usefulness as morphological indicators in the search for drought-tolerant ecotypes of prairie grasses.  相似文献

Rhizomatosae is a taxonomic section of the South American genus Arachis, whose diagnostic character is the presence of rhizomes in all its species. This section is of particular evolutionary interest because it has three polyploid (A. pseudovillosa, A. nitida and A. glabrata, 2n?=?4x?=?40) and only one diploid (A. burkartii, 2n?=?2x?=?20) species. The phylogenetic relationships of these species as well as the polyploidy nature and the origin of the tetraploids are still controversial. The present study provides an exhaustive analysis of the karyotypes of all rhizomatous species and six closely related diploid species of the sections Erectoides and Procumbentes by cytogenetic mapping of DAPI/CMA heterochromatin bands and 5S and 18–26S rDNA loci. Chromosome banding showed variation in the DAPI heterochromatin distribution pattern, which, together with the number and distribution of rDNA loci, allowed the characterization of all species studied here. The bulk of chromosomal markers suggest that the three rhizomatous tetraploid species constitute a natural group and may have at least one common diploid ancestor. The cytogenetic data of the diploid species analyzed evidenced that the only rhizomatous diploid species—A. burkartii—has a karyotype pattern different from those of the rhizomatous tetraploids, showing that it is not likely the genome donor of the tetraploids and the non-monophyletic nature of the section Rhizomatosae. Thus, the tetraploid species should be excluded from the R genome, which should remain exclusively for A. burkartii. Instead, the karyotype features of these tetraploids are compatible with those of different species of the sections Erectoides and Procumbentes (E genome species), suggesting the hypothesis of multiple origins of these tetraploids. Allium przewalskianum, a wild onion species growing at altitudes ranging from 1800 to 4500 m, has long been commonly used as an important vegetable and/or condiment by Tibetans, Indians, and Nepalese in the highlands of the Himalayas and adjacent regions. This species comprises both diploids and tetraploids. In this study, we examined the nutritional content and biomass accumulation profiles of two cytotypes, collected from 29 sites, with different altitudinal origins but cultivated in a common garden. On an average, this species has superior qualities in the minerals and amino acids compared to other edible congeners. When compared with the diploids, the tetraploids grew faster and accumulated more biomass; in addition, the tetraploids had higher values of moisture and energy, higher contents of cystine and phosphorus, but lower fiber levels. Moreover, the tetraploids from the higher altitudes had greater biomasses than the other tetraploids, in addition to having increased levels of proteins, fats, and the minerals Mg, Fe, Mn, and Cu. These results illustrate the large variations in nutritional efficacy and growth within this single morphological species, and provide critical information for its effective consumption in the future.  相似文献

Aim  This study aimed to document precisely the patterns of DNA ploidy variation in the native and secondary ranges of Lythrum salicaria distribution. The hypothesis that species invasiveness had been induced by a switch in ploidy level was addressed.
Location  Europe, Middle East, North America.
Methods  DNA ploidy levels of 1884 progenies of 578+ plants collected at 124 localities were determined by DAPI flow cytometry.
Results  Large cytotype variation (2 x , 3 x , 4 x and 6 x ) was found across the native area of distribution (64 populations covering 12 European and two Middle Eastern countries). DNA hexaploids were detected for the first time, and rare DNA triploids were reliably confirmed. DNA tetraploids largely prevailed across the native range studied, while DNA diploids and DNA hexaploids were recorded only in Israel and Turkey, respectively. DNA triploid progenies occurred in one population from Hungary (together with DNA tetraploids). Sympatric growth of DNA tetraploids and DNA hexaploids was repeatedly encountered in Turkey. In contrast, cytotype uniformity was a typical feature of the invasive North American plants. Sixty populations, covering 13 states of the USA and provinces of Canada, were characterized by the presence of only DNA tetraploids.
Main conclusions  Several L. salicaria cytotypes (2 x , 3 x , 4 x , 6 x ) occur in the native range of distribution, with much variation concentrated in the Middle Eastern countries, whereas only DNA tetraploids appeared to occur in North America. Our data show that the invasive spread of North American populations was not triggered by differences in ploidy level. Alternative explanations should be sought.  相似文献

Cultivated peanut is an allotetraploid (genome type AABB) with a very narrow genetic base, therefore wild species are an attractive source of new variability and traits. Because most wild species are diploid, the first step of introgression usually involves hybridization of wild species and polyploidization to produce a synthetic allotetraploid (AABB) that is sexually compatible with peanut. This study investigates drought-related traits such as leaf morphology, transpiration profile, chlorophyll meter readings (SCMR), specific leaf area (SLA) and transpiration rate per leaf area for two wild diploids (Arachis duranensis and Arachis ipaënsis) that could be of interest for improvement of the peanut crop. Furthermore, the inheritance of the traits from the diploid to the tetraploid state was investigated. Results showed that whilst some diploid traits such as SCMR, are maintained through hybridization and polyploidization, most characters, such as the leaf area, stomata size, trichome density and transpiration profile, are substantially modified. The study concludes that direct evaluations of drought-related traits in wild diploids may be useful for evaluation of wild species to be used in introgression. However, evaluations on wild-derived synthetic tetraploids are likely to be more informative.  相似文献

The Leptynia hispanica stick insect species complex includes bisexuals, triploid and tetraploid parthenogenetic populations, suggesting that polyploidy has played a central role in the evolution of this complex. An analysis of karyotype, mitochondrial DNA (cox2) and nuclear DNA (ef1-alpha) markers was carried out to clarify phylogenetic relationships and microevolutionary/phylogeographical patterns of the L. hispanica complex. Our analyses suggested a subdivision of bisexual populations into four groups, tentatively proposed as incipient species. Moreover, triploids and tetraploids showed two independent origins, the latter being more ancient than the former. From ef1-alpha analysis, triploids showed hybrid constitution, while the hybrid constitution of tetraploids is likely, but more data are needed. We suggest that L. hispanica is a case of 'geographical parthenogenesis' with parthenogenetic strains colonizing large peripheral ranges, and bisexuals confined to glacial refuge areas. Moreover, the age, wide distribution and competitive advantage of polyploids over diploids, demonstrate their significance in the evolution of the L. hispanica species complex.  相似文献

Chromosome number determinations from 118 populations of eight species of the aster genus Virgulus are reported for the first time. Based on these counts and literature reports the geographic distributions of diploids and tetraploids in V. concolor, V. novae-angliae, V. oblongifolius, and V. patens have been determined. In the first three species, diploids were widely distributed, while tetraploids were more restricted. In V. patens, diploids were restricted to the southwestern portion of the species’ range, while tetraploids were found throughout the range. Both V. adnatus and V. walteri were found only at the tetraploid level throughout their ranges. Two other species with restricted distributions were consistently found to be high level polyploids: V. georgianus with 2n = 50, V. grandiflorus with 2n = 60. The following new combinations are listed: Virgulus georgianus (Alexander ex Small) Semple; Virgulus subsect. Brachyphylli (Torr. & Gray) Semple; and Virgulus sect. Polyliguli (Semple & Brouillet) Semple.  相似文献

Examining how both climate and species distribution patterns correlate with leaf morphology can give insights into the ecological and evolutionary patterns that drive adaptive selection of leaf form and function. Drip-tips are a common feature of leaves in rain forest tree species; they are thought to be an adaptation that aids leaf drying and maximizes photosynthesis in areas with high-rainfall climates. We tested whether this macroecological pattern holds true across the precipitation gradients in a non-rain forest region—the woodland savannas of Brazil known as the Cerrado—and compared our results with previous studies from Amazonia. Drip-tips were, as expected, less common overall in the drier Cerrado than in Amazonia. In addition, within the Cerrado, drip-tips were more prevalent in areas with higher rainfall as well as in Cerrado sites that were closer to Amazonia. Moreover, species that occurred across both the Cerrado and Amazonia had drip-tips more often than species that were found only in the Cerrado. These findings support the hypothesis that drip-tips are adaptive and that either the cost of retaining drip-tips is low or that in drier regions they have other benefits.  相似文献
