In the following a method for the induction of pistil-like structures in wheat suspension cultures (Triticum aestivum L.) is described. In young influorescences of plants, which were artificially infected with the wheat bunt fungi (Tilletia controversa), organogenic calli with pistil-like structures could be induced on loblolly pine medium + 3 mg/l 3,6-dichloro-2-methoxy benzoic acid. Interference of phytohormones (jasmonic, gibberellic, and abscisic acids) and synthetic growth regulator melafen on Ca2+ translocation across the membrane of plasma membrane vesicles prepared from dormant potato (Solanum tuberosum L.) tubers was studied. The activity of plasma membrane Ca2+, Mg2+-ATPase was stimulated by melafen and jasmonic and gibberellic acids and suppressed by abscisic acid. These substrances did not change the passive membrane permeability for Ca2+. The pattern of the effect of melafen on the activity of Ca2+,Mg2+-ATPase depended on the presence of phytohormones in incubation medium. When melafen and each phytohormone were simultaneously added to incubation medium, their effects were not additive, which indicates that the effects of the tested compounds on the Ca2+ uptake into the plasma membrane vesicles are interdependent. Apparently, the interaction between the phytohormones and plasma membrane components modulates the response to melafen.  相似文献

Treating carrot (Daucus carota L.) discs with ice-cold NaCl solutions for 30 minutes caused three effects that appear to be functionally related: the exchange of tissue Ca2+ and Mg2+ for Na+, the release of protein, and the suppression of active uptake of glucose and orthophosphate. Cyclosis continued apparently unabated after treatment with NaCl at concentrations of up to 0.25 m, so the cells remained viable and energetically competent. The correlation between the release of Ca2+ and Mg2+ and release of protein, and between these effects and the suppression of glucose and orthophosphate uptake, supports the hypothesis that divalent cations maintain, and monovalent cations disrupt, linkages between the outer cell surface and proteins required for active solute uptake. Calcium preserved uptake activity only when it was added in time to prevent the release of protein. Cells gradually recovered some glucose uptake activity after it had been completely inactivated by treatment with 0.25 m NaCl. This recovery occurred in the absence of added Ca2+. It was inhibited by puromycin and so appears to require some protein synthesis. Beet (Beta vulgaris L.) discs were more resistant than carrot discs to treatment with NaCl solutions, thus reflecting the difference in tolerance of the two species to sodicity.  相似文献

Most measurements of nutrient uptake use either hydroponic systems or soil-grown roots that have been disturbed by excavation. The first objective of this study was to test how root excavation affects nitrate uptake. Rates of NO3? uptake by mycorrhizal loblolly pine (Pinus taeda L.) seedlings were measured in intact sand-filled columns, hydroponics, and disturbed sand-filled columns. Total nitrate uptake in intact sand-filled columns was higher than in disturbed columns, indicating that disturbance lowers uptake. Transferring plants from the sand-filled columns to hydroponics had little effect on NO3? uptake beyond delaying uptake for an hour. The second objective of this study was to determine whether NH4+, Ca2+, Mg2+ and K+ uptake could be studied using sand-filled columns, since previous studies had tested this method only for nitrate uptake. Uptake rates of NH4+ and K+ were positive, while Ca2+ and Mg2+ uptake rates were negative in intact sand-filled columns, indicating that net efflux may occur even without physical disturbance to the root system. The sand-filled column approach has some limitations, but holds promise for conducting nutrient uptake studies with minimal disturbance to the root system.  相似文献

A wheat (Triticum aestivum L.) cell line, derived from anther culture of an F1 hybrid, has exogenous Ca2+, to that of calcium-dependent cells grown on complete medium. The calcium-independent cell line has been grown in the absence of Ca2+ for more than 1.5 years. The cell line grew at a rate similar to that on complete medium for up to 12 weeks, if supplied with any one of the divalent cations, Ca2+, Mg2+, Mn2+, Zn2+, Cu2+ or Co2+, but declined and appeared necrotic when all 6 of these were removed from the medium. The calcium-independence trait, while identified in tissue culture, was also observed in germinated immature embryos of the same hybrid and one of its parental inbred lines.  相似文献

Protoplasts from dark-grown wheat (Triticum aestivum L.) maintained at a constant osmotic potential at 22°C, were found to swell upon red irradiation (R) and the effect was negated by subsequent far-red light (FR), indicating phytochrome involvement. Swelling only occurred when Ca2+ ions were present in the surrounding medium, or were added within 10 min after R. Furthermore, Mg2+, Ba2+ or K+ could not replace this requirement for Ca2+. The presence of K+ did not enhance the Ca2+-dependent swelling response. When the Ca2+-ionophore A 23187 was added to the medium, protoplasts swelled in the dark to the same extent as after R. Both the Ca2+-channelblocker Verapamil and La3+ inhibited R-induced swelling. It is proposed that R causes the opening of Ca2+-channels in the plasma membrane. The presence of Ca2+ ions in solution is vital for root growth. The plasma membrane is one of the first sites where competition between Ca2+ and other ions occurs. We studied the competition between Ca2+ and Na+ or Mg2+ for sorption sites on the plasma membrane of melon root cells.Sorption of 45Ca2+ to right-side-out PM vesicles of melon (Cucumis melo L.) roots (prepared by aqueous two-phase partitioning) was studied at various Ca2+ concentrations, in the presence of increasing concentrations of Na+ or Mg2+ chlorides. Experimentally determined amounts of Ca2+ sorbed to the plasma membrane vesicles agreed fairly well with those calculated from a competitive sorption model. The best fit of the model to the experimental data was obtained for an average surface area of 370 Å2 per charge, and binding coefficients for Na+, Mg2+ and Ca2+ of 0.8, 9 and 50 m -1, respectively.Our results suggest that nonphospholipid components in the plasma membrane contribute significantly to Ca2+ binding. The high affinity of Ca2+ binding to the plasma membrane found in this study might explain the specific role of Ca2+ in relieving salt stress in plant roots.This research was supported by the GIFRID German-Israel fund for research and international development.  相似文献

Summary The distribution of free cytosolic Ca2+ was studied during somatic embryogenesis of carrot using confocal scanning laser microscopy with fluo-3 as a fluorescent Ca2+ indicator. Chlorotetracycline fluorescence, antimonate precipitation and proton induced X-ray emission analysis were used as additional methods to confirm the results obtained with fluo-3. The process of embryogenesis was found to coincide with a rise in the level of free cytosolic Ca2+. The level of Ca2+ was low in proembryogenic masses and relatively high in later stages of embryogenesis. The highest signal was found in the protoderm of embryos from the late globular to the torpedo-shaped stage. A gradient in fluorescence intensity was often observed along the longitudinal axis of the embryos. The most conspicuous intracellular signal was found in the nucleus. Other organelles did not take up the dye and were always without fluorescence. The changes in [Ca2+]c are discussed in relation to physiological processes which are known to be important during somatic embryogenesis.This article is dedicated to the memory of the late Dr. Hans-Dieter Reiss.  相似文献

The light-dependent germination response of turions (resting fronds) is mediated by phytochrome and requires the presence of Ca2+ in the medium (K.-J. Appenroth and H. Augsten, 1990, Photochem. Photobiol. 52: 61–65). The Ca2+ requirement of germination is apparent only in the presence of exogenous Mg2+. A competitive ion antagonism was demonstrated between Ca2+ and Mg2+ in this physiological response; Mg2+ could also be replaced by Ba2+ or Sr2+. Without exog-enous Mg2+, a Ca2+ concentration as low as 0.9 μM fulfilled the Ca2+ requirement. This type of ion antagonism resembled the competitive Ca/Mg interaction reported previously for calcium-binding proteins. The physiological response was blocked by inhibitors of Ca2+ uptake (verapamil, La3+). It was concluded that uptake of Ca2+ from the external medium is an essential step in the phytochrome-mediated germination of turions. The results are in agreement with the assumption that the uptake of Ca2+ is blocked at the side of entry by other alkaline earth ions. Treatment of turions with Mg2+ (1 mM) for 24 h at varying times after the red light pulse in otherwise virtually Ca2+-free KNO3 solution resulted in a response similar to a Ca2+ step-down treatment. This is in agreement with the assumption that the Ca2+- and the Mg2+-sensitive periods coincide. The ion interaction described here represents the first photophysiological example in plants of an antagonistic effect between Ca2+ and Mg2+ similar to that which occurs in vitro with calmodulin. Summary Effects of divalent cations on oscillations of membrane potentials (i.e., spontaneous repetitive hyperpolarizing responses) and on hyperpolarizing responses induced by electrical stimuli as well as on resting potentials were studied in large nondividing L cells. Deprivation of Ca2+ from the external medium inhibited these hyperpolarizing responses accompanying slight depolarization of the resting potential. Sr2+ or Mn2+ applied to the external medium in place of Ca2+ was able to substitute for Ca2+ in the generation of hyperpolarizing responses, while Mg2+, Ba2+ or La3+ suppressed hyperpolarizing responses. The addition of A23187 to the bathing medium or intracellular injection of Ca2+, Sr2+, Mn2+ or La3+ induced membrane hyperpolarization. When the external Ca2+, Sr2+ or Mn2+ concentration was increased, the resting potential also hyperpolarized, in a saturating manner. The amplitude of maximum hyperpolarization produced by high external Ca2+ was of the same order of magnitude as those of hyperpolarizing responses and was dependent on the external K+ concentration. In the light of these experimental observations, it was deduced that the K+ conductance increase associated with the hyperpolarizing excitation is the result of an increase in the intracellular concentration of free Ca2+ mainly derived from the external solution.  相似文献

Levels of Mg2+, Ca2+ and Fe2+/Fe3+ were determined in roots and shoots of sugar beet seedlings (Beta vulgaris L. cv. Monohill) cultured for 5 weeks in a complete nutrient solution to which either Cd2+ (0, 5 or 50 μM), EDTA (0, 10 or 100 μM) or a combination of both was added. The plants subjected to the various treatments showed a variety of deficiency symptoms. Leaves of the Cd2+-treated plants became thin and chlorotic (Mg- and Fe-deficiency symptoms). The plants showed reduced growth and developed only a few brownish roots with short laterals (Ca-deficiency symptoms). EDTA treatment resulted in green, stunted, hard leaves and reduced growth (Ca-deficiency symptoms). The deficiency symptoms observed correspond well with the observed uptake rates and distributions of Mg2+, Ca2+ and Fe2+/Fe3+. Increases in either Cd2+, EDTA or a combination of both in the growth medium, were correlated with increasing Mg2+ levels in the roots and with decreasing Mg2+ levels in the shoots. Cd2+ alone or in combination with EDTA had little influence on Ca2+ levels in the shoots but decreased Ca2+ levels in the roots. Thus, Cd2+ affects Mg2+ and Ca2+ transport in opposite ways: Mg2+ transport to the shoots is inhibited while that of Ca2+ is facilitated. Treatment with EDTA alone did not affect Ca2+ concentrations in either the shoots or the roots. Treatment with Cd2+ lowered Fe2+ concentrations in both roots and shoots.  相似文献

Summary The effects of Mg+2 and Ca+2 ion concentrations onPichia stipitis growth and flocculation were studied by a 2×2 factorial design. Negative interaction was detected between these ions (p<0.01): maximum growth rate was attained with high Mg+2 (3 mM) and low Ca+2 (0.34 mM) concentration levels, whereas the inverse ionic ratio was required to reach the maximum flocculation extent. Neither stimulatory or inhibitory effects on growth or flocculation were observed by increasing other cations (Zn+2, Mn+2, Co+2, Fe+2) or vitamins concentration. To determine whether or not inositol trisphosphate (IP3) mobilizes calcium in higher plant cells, we investigated the effect of IP3 on Ca2+ fluxes in fusogenic carrot (Daucus carota L.) protoplasts. The protoplasts were incubated in 45Ca2+-containing medium and the 45Ca2+ associated with the protoplasts was monitored with time. Addition of IP3 (20 micromolar) caused a 17% net loss of the accumulated 45Ca2+ within 4 minutes. There was a reuptake of 45Ca2+ and the protoplasts recovered to their initial value by 10 minutes. Phytic acid (IP6), also stimulated 45Ca2+ efflux from the protoplasts. Both the IP3 and the IP6induced 45Ca2+ efflux were inhibited by the calmodulin antagonist, trifluoperazine.  相似文献

A procedure was developed which demonstrates the expression of differential resistance in pine callus tissues to the fungal pathogen Phytophthora cinnamomi Rands. Callus tissues were maintained on a modified Murashige and Skoog medium with 10–5M 2,4-D and inoculated with hyphae of P. cinnamomi at 26°C in the dark. The number of intracellular hyphae was used as an index of resistance. The effect of low pH on net H+ release and root growth of corn (Zea mays L.) and broad bean (Vicia faba L.) seedlings was investigated in short-term experiments at constant pH. Broad bean was more sensitive to low pH than corn: the critical values (pH values below which net H+ release and root growth ceased) were pH 4.00 (broad bean) and pH 3.50 (corn) at 1 millimolar Ca2+. Both proton release and root growth were progressively inhibited as the medium pH declined. Additional Ca2+ in the root medium helped to overcome the limitations of low pH for net H+ release and root growth. Potassium (for corn) and abscisic acid (for broad bean) increased both net H+ release and root growth rate at the critical pH value. It is concluded that poor root growth at low pH is caused by a lack of net H+ release that may decrease cytoplasmic pH values. Inhibited net H+ release at high external H+ activity is not due to a shortage of energy supply to the H+ ATPase. Instead, a displacement of Ca2+ by H+ at the external side of the plasmalemma may enhance reentry of H+ into root cells.  相似文献

–The effects of divalent cations on the initial rates of noradrenaline uptake by synaptosomes were determined using Millipore filtration to terminate the reaction. The removal of either Ca2+ or Mg2+ from the incubation medium had no effect on uptake, but when both Ca2+ and Mg2+ were removed, uptake was reduced. Uptake was also diminished when Ca2+ was absent and 1 mm -EGTA added to the medium. It appeared that Ca2+ was required for optimal uptake but that Mg2+ could partially substitute for Ca2+ in this regard. The reduction in the rate of uptake when both Ca2+0 and Mg2+ were absent could be rapidly and completely reversed by restoring Ca2+, Mg2+, or both Ca2+ and Mg2+ to the incubation medium. Of the divalent cations tested, Ca2+ and Mg2+, but not Mn2+, supported noradrenaline uptake. When the kinetics of uptake were examined, it was found that removing both Ca2+ and Mg2+ from the medium resulted in a reduction of the Vmax for noradrenaline uptake. It is apparent from these results that, in addition to facilitating the release of noradrenaline from noradrenergic terminals, Ca2+ may also play a role in the uptake of noradrenaline by presynaptic nerve-endings in the CNS.  相似文献

The rate of Ca2+ accumulation in plasmalemma vesicles isolated from quiescent and sprouting potato (Solanum tuberosum L.) tubers and the effect of 10?5–10?10 M jasmonic acid on the accumulation of Ca+2 in plasmalemma vesicles and its efflux were studied. It was found that potato tuber plasmalemma contains a Ca+2,Mg+2-ATPase whose activity decreases upon the transition from forced quiescence to growth. The direction of the effect of jasmonic acid on Ca+2,Mg+2-ATPase (stimulation or suppression) depends on the physiological state of tubers and the phytohormone concentration.  相似文献

The effects of Co2+ and Ni2+ on ethylene production and somatic embryogenesis by carrot (Daucus carota L.) cell cultures were studied. At concentrations of 10 M to 50 M, CoCl2 effectively inhibited ethylene production by embryogenic cultures and significantly stimulated somatic embryogenesis. The observed increase of embryo number was proportional to the inhibition level of ethylene production. However, CoCl2 had no effect when Ethephon was supplied. Nickel also reduced ethylene production, but to a slightly lesser extent than CoCl2, bringing about a lower increase in the number of somatic embryos. This paper summarizes the data on nutrient uptake and soil responses in opentop chambers planted with ponderosa pine (Pinus ponderosa Laws.) treated with both N and CO2. Based upon the literature, we hypothesized that 1) elevated CO2 would cause increased growth and yield of biomass per unit uptake of N even if N is limiting, and 2) elevated CO2 would cause increased biomass yield per unit uptake of other nutrients only by growth dilution and only if they are non-limiting. Hypothesis 1 was supported only in part: there were greater yields of biomass per unit N uptake in the first two years of growth but not in the third year. Hypothesis 2 was supported in many cases: elevated CO2 caused growth dilution (decreased concentrations but not decreased uptake) of P, S, and Mg. Effects of elevated CO2 on K, Ca, and B concentrations were smaller and mostly non-significant. There was no evidence that N responded in a unique manner to elevated CO2, despite its unique role in rubisco. Simple growth dilution seemed to explain nutrient responses in almost all cases.There were significant declines in soil exchangeable K+, Ca2+, Mg2+ and extractable P over time which were attributed to disturbance effects associated with plowing. The only statistically significant treatment effects on soils were negative effects of elevated CO2 on mineralizeable N and extractable P, and positive effects of both N fertilization and CO2 on exchangeable Al3+. Soil exchangeable K+, Ca2+, and Mg2+ pools remained much higher than vegetation pools, but extractable P pools were lower than vegetation pools in the third year of growth. There were also large losses of both native soil N and fertilizer N over time. These soil N losses could account for the observed losses in exchangeable K+, Ca2+, Mg2+ if N was nitrified and leached as NO 3 .  相似文献

Plant cells frequently and rapidly have to respond to environmental changes for survival. Regulation of transport and other energy-requiring processes in the plasmalemma of root cells is therefore one important aspect of the ecological adaptation of plants. Wheat (Triticum aestivum L. cv. Drabant) was grown hydroponically, with or without 50 nM benzyladenine in the medium, and plasma membranes from root cells of 8-day-old plants were prepared by aqueous polymer two-phase partitioning. The influence of Ca2+ and Mg2+ on the plasmalemma ATPase activities was investigated. The presence of benzyladenine during growth increased the ATPase activity, that dependent upon Ca2+ more than that elicited by Mg2+. As a general characteristic, ATP was the preferred substrate, but all nucleotide tri- and diphosphates could be accepted with activities in plasma membranes from control plants of 7-36% (Mg2+) and 40-86% (Ca2+) and in plasma membranes from benzyladenine-treated plants of 12-47% (Mg2+) and 53-102% (Ca2+) as compared with activities obtained with ATP. Nucleotidemonophosphates were not hydrolyzed by the preparations. In preparations from benzyladenine-treated plants one peak of Ca2+-ATPase at pH 5.2–5.6, with a tail from pH 6 and upwards, and one peak of Mg2+-ATPase at pH 6.0–6.5 were observed in the presence of EDTA in the assay media. In preparations from control plants, the addition of EDTA to the assays resulted in a wide optimum between pH 6 and 7 for Mg2+-ATPase and low Ca2+-ATPase activity with no influence of pH in the range 4.5 to 8. Analysis of the pH dependence in the presence of both Ca2+ and Mg2+ indicates that the control plants mainly contain Mg2+-ATPase corresponding to the proton pump. Preparations from benzyladenine-treated wheat roots show, in addition, activation by Ca2+, which, in the slightly alkaline pH range may correspond to a Ca2+-extruding (Ca2++ Mg2+)-ATPase. In the acidic range, the responses are more complicated: the Mg2+-ATPase is inhibited by vanadate, while the Ca2+-ATPase is insensitive, and benzyladenine added during growth influences the interaction between Ca2+ and Mg2+ in a way that parallels the effect of high salt medium.  相似文献
