AIMS: Studies were carried out to investigate the survival of Salmonella Typhimurium and Salmonella Derby in pig slurry during summer and winter seasons. METHODS AND RESULTS: Pig slurry samples collected from a commercial fattening house were inoculated with a broth culture of Salmonella Typhimurium and Salmonella Derby, each at a level of log(10) 5.0 CFU ml(-1) and log(10) 2.0 CFU ml(-1). At the higher inoculum level, S. Typhimurium and S. Derby survived for 34 and 23 days, respectively in the summer, and 58 and 46 days, respectively in the winter. Survival at the lower inoculum level for S. Typhimurium and S. Derby was 19 and 16 days, respectively, in the summer and 24 days for both in the winter. CONCLUSIONS: The survival of S. Typhimurium and S. Derby observed in this study indicates that a 2-month holding period of pig slurry, prior to land spreading, may be adequate if separate storage facilities are provided. SIGNIFICANCE AND IMPACT OF THE STUDY: Despite difficulties correlating laboratory studies with on-farm conditions, pig slurry may not represent a major source of transmission of Salmonella spp. in the farm environment in Ireland.  相似文献

AIMS: The aim of this study was to construct an advanced high-power pulsed light device for decontamination of food matrix and to evaluate its antibacterial efficiency. Key parameters of constructed device-emitted light spectrum, pulse duration, pulse power density, frequency of pulses, dependence of emitted spectrum on input voltage, irradiation homogenicity, possible thermal effects as well as antimicrobial efficiency were evaluated. METHODS AND RESULTS: Antimicrobial efficiency of high-power pulsed light technique was demonstrated and evaluated by two independent methods - spread plate and Miles-Misra method. Viability of Salmonella typhimurium as function of a given light dose (number of pulses) and pulse frequency was examined. According to the data obtained, viability of Salmonella typhimurium reduced by 7 log order after 100 light pulses with power density 133 W cm(-2). In addition, data indicate, that the pulse frequency did not influence the outcome of pathogen inactivation in the region 1-5 Hz. Moreover, no hyperthermic effect was detected during irradiation even after 500 pulses on all shelves with different distance from light source and subsequently different pulse power density (0-252 W cm(-2)). CONCLUSION: Newly constructed high-power pulsed light technique is effective nonthermal tool for inactivation of Salmonella typhimurium even by 7 log order in vitro. SIGNIFICANCE AND IMPACT OF THE STUDY: Novel advanced high-power pulsed light device can be a useful tool for development of nonthermal food decontamination technologies.  相似文献

1. Zooplankton are important in transferring dietary nutrients, including polyunsaturated fatty acids (PUFA), up through aquatic food webs. 2. We tested the hypothesis that the taxonomic composition of zooplankton affects the retention and subsequent transfer of PUFA from upwards through the food web. Using laboratory experiments, we investigated dietary PUFA accumulation and bioconversion capacities of six cladoceran species (Ceriodaphnia sp., Daphnia longispina, Daphnia magna, Daphnia pulex, Scapholeberis mucronata and Simocephalus vetulus) fed on two diets (Scenedesmus obliquus and Cryptomonas sp.) that differed in their PUFA profiles. We performed experiments at two different temperatures (14 and 20 °C) to assess the role of temperature in the trophic transfer of PUFA. 3. There was little variation in the concentrations of PUFA in these cladocerans which were controlled by dietary PUFA supply. Moreover, as expected, the concentrations of PUFA in all cladoceran species were higher at low temperature. 4. However, even if the composition of PUFA in the cladoceran species generally corresponded to that in their diet, preferential accumulation of some PUFA was recorded in all these taxa. When fed on a highly unsaturated fatty acid‐deficient diet, all the cladocerans showed some ability to convert C18‐PUFA into arachidonic acid and eicosapentaenoic acid. Interspecific variation in the ability to accumulate and bioconvert PUFA in cladocerans was more pronounced at low temperature (14 °C) for both diets. 5. Our results strongly suggest that in heterogeneous habitats with food partitioning between co‐existing cladocerans, foraging behaviour may affect the transfer of PUFA more strongly than interspecific variation in accumulating and/or bioconverting dietary PUFA.  相似文献

The temperature-size rule (TSR) is a well-established phenomenon to describe the growth response of ectotherms to temperature by which individuals maintained at low temperatures grow more slowly, but attain a larger size upon maturity. Although there are adaptive and non-adaptive theories about the plasticity of body size in response to temperature, these cannot be applied to all ectotherms, and little is known about the changes in growth and development rates through ontogeny. The ostracod species Heterocypris bosniaca, an inhabitant of freshwater temporary ponds, was used to examine the growth and development rates of its nine growth stages and female fecundity at four different temperatures (15 °C, 20 °C, 25 °C and 30 °C). The development rate of this species accelerates with increasing temperature, reaching a maximum value at 25 °C. The growth factor has a reverse-TSR in younger instars, and the typical TSR is followed only in the last two moults, resulting in non-monotonic response of adult size to temperature. Fecundity (total offspring per female) was not directly related to adult size and was generally higher at lower temperatures. Our results agree with recent research showing that the TSR may vary during ontogeny, and may not be a general trend in ostracod species from temporary waters. Indeed, adult carapace size seems to follow the pattern of a thermal reaction norm, probably influenced by the reduction of oxygen bioavailability at low temperature and the drastic increase in metabolic demand at the upper extreme of the thermal gradient.  相似文献

Aims:  To evaluate a novel secondary model structure ( Int J Food Microbiol 2008; 128: 67) that describes the effect of medium structure on the maximum specific growth rate ( μ max) of Salmonella Typhimurium on the growth of S. Typhimurium, Listeria innocua , Lactococcus lactis and Listeria monocytogenes .
Methods and Results:  In the present study, the novel secondary model is validated for S . Typhimurium in more realistic media, namely, pasteurized milk and a cheese mimicking medium. The predictions were accurate. Next, the secondary model structure was evaluated in a two step and a global regression procedure on literature data. On the one hand, the growth of two other micro-organisms, namely L. innocua and L. lactis , in monoculture for varying gelatine concentrations was tested and on the other hand the growth rate of L. monocytogenes was fitted in a broth of which the viscosity was altered with polyvinylpyrrolidone. The model was able to describe the effect of increasing gelatine concentration or viscosity accurately.
Conclusions:  The proposed secondary model structure is able to describe the effect of gelatine concentration on the μ max of the micro-organisms tested in this study.
Significance and Impact of the Study:  In predictive microbiology, much attention has been paid to the effect of food structure on the μ max of bacteria. However, to the authors' knowledge, a lack of secondary models still exists to describe this effect. Although the proposed model is empirical, the model parameters have clear biological meaning. The predictive power of the model to describe the effect of food structure is clearly illustrated.  相似文献

Bacterial microcompartments (MCPs) are subcellular organelles that are composed of a protein shell and encapsulated metabolic enzymes. It has been suggested that MCPs can be engineered to encapsulate protein cargo for use as in vivo nanobioreactors or carriers for drug delivery. Understanding the stability of the MCP shell is critical for such applications. Here, we investigate the integrity of the propanediol utilization (Pdu) MCP shell of Salmonella enterica over time, in buffers with various pH, and at elevated temperatures. The results show that MCPs are remarkably stable. When stored at 4°C or at room temperature, Pdu MCPs retain their structure for several days, both in vivo and in vitro. Furthermore, Pdu MCPs can tolerate temperatures up to 60°C without apparent structural degradation. MCPs are, however, sensitive to pH and require conditions between pH 6 and pH 10. In nonoptimal conditions, MCPs form aggregates. However, within the aggregated protein mass, MCPs often retain their polyhedral outlines. These results show that MCPs are highly robust, making them suitable for a wide range of applications.  相似文献

Early instar nymphs of the mayfly, Cloeon sp. were kept on an algal or a detrital diet at 20 ± 1 °C in the laboratory. A control group was not given any food. Nymphs kept on algae showed significantly (P = 0.001) higher growth both in terms of length and biomass than for those kept on detritus. None of the nymphs given algae or detritus died during the experiment, and 40 per cent of those fed algae and 20 per cent fed detritus reached maturity by the termination of the experiment. Food retention time as well as gill ventilation rate of nymphs feeding on algae were significantly (P = 0.001) higher than those feeding on detritus. It is probable that these two factors largely enabled the nymphs to survive as well as to grow and reach maturity, though relatively slowly, solely on a detrital diet. However, algae was clearly shown to be a superior food resource for Cloeon sp. nymphs.  相似文献

Summary Chaoborus crystallinus fourth-instar larvae were reared individually at 14°, 17° and 20° C under different food conditions. Daphnia magna of 1.25 mm average length served as prey. The following were measured: amount of prey ingested, larval weight gain, duration of fourth instar, body weight of the adults, and egg number per female. At a given temperature, the body weight, egg-number and developmental rate increased with food consumption. At a given food consumption, higher temperatures caused a decrease in body weight and egg number, and an increase in developmental rate. Gross production efficiencies for fourth-instar larvae were highest at temperatures around 17° C. The results clearly indicate that from an energetic point of view higher temperatures are disadvantageous. In C. crystallinus vertical migration is evidently a way of lowering the temperature to which the animals are exposed and hence optimizing food conversion into biomass and offspring production, especially if prey densities are below the saturation level.  相似文献

In this study, we investigated the effects of alternating tangential flow (ATF) cell separation on high-density perfusion cultures. We have developed methods to estimate theoretical residence times of cells in the ATF system and discovered that long residence times (above 75 s) correlate with decreased growth, metabolism, and productivity. We have calculated energy dissipation rates in the ATF transfer line and filter and empirically studied the impacts of increased exchange rates on cell culture, determining that increased hydrodynamic stress can lead to decreased cell size, lactate production, and specific productivity. Finally, we have conducted experiments to understand the relationship between filtration fluxes and ATF membrane fouling, finding that at fluxes above 60 L·m–2·day –1, protein sieving coefficients see significant rates of decrease (greater than 1% per day). While most of these studies have been conducted with one cell line at one target viable cell density (40 million cells/ml), the general, directional knowledge arising from this study should be applicable to other conditions and programs, ultimately leading to more robust and well-designed perfusion processes.  相似文献

Using museum data of adult specimens whose sex, age, and locality are known, we studied temporal and geographical body size trends among the otter, Lutra lutra, in Norway. We found that body size of the otters increased during the last quarter of the twentieth century, and suggest that this trend is related to increased food availability from fish farming and possibly also to energy saving due to elevated sea temperatures. Birth year and death year explained 38.8 and 43.5%, respectively, of the variation in body size. Body size of otters was positively related to latitude, thus conforming to Bergmann’s rule.  相似文献

报道了不同温度、盐度和饵料条件下西藏拟溞的摄食强度、西藏拟溞对不同饵料生物蛋白核小球藻(Chlorellapyrenoidesa)、牟氏角毛藻(Chaetoceros muelleri)、湛江等鞭金藻(Isochrysis zhanjiangensis)、亚心形扁藻(Platymonas subcordiformis)、盐藻(Dunaliella salina)、新月拟菱形藻(Nitzschiella closterium)的选择性以及摄食节律。结果表明,在适温范围(2 20℃)内,西藏拟溞的滤水率、摄食率随着温度的升高而增加,14 16℃为西藏拟溞的适宜温度。在盐度20时,西藏拟溞的摄食率最高,幼、成溞对等鞭金藻的摄食率分别为1.88 ng C/(ind.h)和3.46 ng C/(ind.h)。西藏拟溞对角毛藻的滤水率和摄食率均随着龄期增加而增加,而对等鞭金藻呈相反趋势,但二者的日粮均随体长增大而减小。幼、成溞的滤水率和摄食率随食物密度变化的趋势基本一致,均是在盐藻密度为1×106 ind./L时均最高,分别为0.194、0.221 mL/(ind.h)和0.030μgC/(ind.h)和0.034μgC/(ind.h)。幼溞对6种藻的食物选择性为角毛藻小球藻盐藻扁藻金藻拟菱形藻;成溞为扁藻角毛藻盐藻金藻小球藻拟菱形藻。幼溞、成溞均对角毛藻和盐藻的选择性较好。西藏拟溞的幼溞和成溞对牟氏角毛藻的摄食有相同的节律,在15:00 17:00和01:00 03:00时有两个摄食高峰,在09:00 11:00和23:00 01:00有两个低谷。  相似文献

AIMS: To comparatively evaluate the adaptive stationary-phase acid tolerance response (ATR) in food-borne pathogens induced by culturing in glucose-containing media, as affected by strain variability and antibiotic resistance, growth temperature, challenge pH and type of acidulant. METHODS AND RESULTS: Antibiotic resistant or sensitive strains of Listeria monocytogenes, Salmonella including S. Typhimurium DT104, and Escherichia coli O157:H7 were cultured (30 degrees C for 24 h; 10 degrees C for up to 14 days) in trypticase soya broth with yeast extract (TSBYE) with 1% or without glucose to induce or prevent acid adaptation, respectively. Cultures were subsequently exposed to pH 3.5 or 3.7 with lactic or acetic acid at 25 degrees C for 120 min. Acid-adapted cultures were more acid tolerant than nonadapted cultures, particularly those of L. monocytogenes and Salmonella. No consistent, positive or negative, influence of antibiotic resistance on the pH-inducible ATR or acid resistance (AR) was observed. Compared with 30 degrees C cultures, growth and acid adaptation of L. monocytogenes and S. Typhimurium DT104 at 10 degrees C markedly reduced their ATR and AR in stationary phase. E. coli O157:H7 had the greatest AR, relying less on acid adaptation. A 0.2 unit difference in challenge pH (3.5-3.7) caused great variations in survival of acid-adapted and nonadapted cells. CONCLUSIONS: Culturing L. monocytogenes and Salmonella to stationary phase in media with 1% glucose induces a pH-dependent ATR and enhances their survival to organic acids; thus, this method is suitable for producing acid-adapted cultures for use in food challenge studies. SIGNIFICANCE AND IMPACT OF THE STUDY: Bacterial pathogens may become acid-adapted in foods containing glucose or other fermentable carbohydrates. Low storage temperatures may substantially decrease the stationary-phase ATR of L. monocytogenes and S. Typhimurium DT104, but their effect on ATR of E. coli O157:H7 appears to be far less dramatic.  相似文献

Horizontal flow constructed wetlands are engineered systems capable of eliminating a wide range of pollutants from the aquatic environment. Nevertheless, poor hydrodynamic behavior is commonly found resulting in preferential pathways and variations in both (i) the hydraulic residence time distribution (HRTD) and, consequently, (ii) the wetland's treatment efficiency. The aim of this work was to outline a methodology for wetland design that accounts for the effect of heterogeneous hydraulic properties of the porous substrate on the HRTD and treatment efficiency. Biodegradation of benzene was used to illustrate the influence of hydraulic conductivity heterogeneity on wetland efficiency. Random, spatially correlated hydraulic conductivity fields following a log-normal distribution were generated and then introduced in a subsurface flow numerical model. The results showed that the variance of the distribution and the correlation length in the longitudinal direction are key indicators of the extent of heterogeneity. A reduction of the mean hydraulic residence time was observed as the extent of heterogeneity increased, while the HRTD became broader with increased skewness. At the same time, substrate heterogeneity induced preferential flow paths within the wetland bed resulting in variations of the benzene treatment efficiency. Further to this it was observed that the distribution of biomass within the porous bed became heterogeneous, rising questions on the representativeness of sampling. It was concluded that traditional methods for wetland design based on assumptions such as a homogeneous porous medium and plug flow are not reliable. The alternative design methodology presented here is based on the incorporation of heterogeneity directly during the design phase. The same methodology can also be used to optimize existing systems, where the HRTD has been characterized with tracer experiments.  相似文献

Testate amoebae are useful environmental indicators in ecological and palaeoecological studies from peatlands. Previous quantitative studies have focused on the peatlands of Northern and Central Europe, North America, and New Zealand and have considered a relatively restricted variety of peatland types, mostly ombrotrophic or Sphagnum-dominated while more minerotrophic fens have been less studied. Here we present the first quantitative ecological study of testate amoebae from four small mesotrophic fens (pH 5.5-8.1) in the Elatia Forest, northern Macedonia province, Greece. Relationships with the environmental data were investigated using redundancy analysis and mantel tests. Transfer function models were derived using a variety of techniques. Results demonstrate that as for Sphagnum-dominated mires hydrology is the most important control on amoebae community composition. Transfer function models should enable water tables to be predicted within 2.5 cm, when data selection is used this is reduced to less than 2 cm. pH is also an important environmental control on testate amoebae communities, a transfer function model enables pH prediction within 0.4 pH units. The hydrological transfer function is the best performing such model yet produced in terms of prediction error. This study provides new data on the ecology of testate amoebae in fens, and the transfer function models should allow quantitative palaeohydrological reconstruction.  相似文献

The effect of experiences, such as contact with honeydew, rejections of hosts, and ovipositions in hosts, and of temperature on the time allocation of individualEncarsia formosa female parasitoids on tomato leaflets have been studied. Behavioral records were analyzed by means of the proportional hazards model. Analyses were carried out at two levels: (1) the tendency of leaving and (2) the tendency of changing from one leaf side to another. The patch-leaving behavior ofE. formosa can be described by a stochastic threshold mechanism, which is characterized by a certain tendency (probability per time) to leave. The median time from being placed on the leaflet or, if it occurred, from the latest encounter with a host until leaving was 18.6 min. The median time for changing from one leaf side to the other was initially 11.6 min and dropped to 5.7 min after both leaf sides had been visited. The effect of temperature, ranging from 20 to 30°C, was negligible. The presence of honeydew as well as the first oviposition in an unparasitized host decreased the tendency to leave, thus increasing the giving up time (GUT) since the latest encounter with a host. Encounters with parasitized hosts did not affect the GUT since latest encounter; as a result, the total residence time increased. After the first oviposition in an unparasitized host the tendency of changing from the lower leaf side on which hosts were present to the upper side was decreased. The presence of honeydew did not affect the tendency of changing leaf sides.  相似文献
