Cytoplasmic DNA variation and relationships in cereal genomes   总被引:2,自引:0,他引:2  
Summary Chloroplast (cp) and mitochondrial (mt) DNAs were isolated from four cereal genomes (cultivated wheat, rye, barley and oats) and compared by restriction nuclease analysis. Cleavage of cp and mt DNAs by Sal I, Kpn I, Xho I and EcoR I enzymes indicated that each cereal group contains specific cytoplasmic DNAs. A phylogenetic tree of cereal evolution has been obtained on the basis of cp DNA homologies. It is suggested that wheat and rye diverged after their common ancestor had diverged from the ancestor of barley. This was preceded by the divergence of the common ancestor of wheat, rye and barley and the ancestor of oats.The molecular weight of the different cp DNAs was determined from the Sal I and Kpn I patterns. cp DNAs from wheat, rye, barley and oats appeared to be characterized by a very similar molecular weight of about 80–82.106 d.In the case of the mt DNAs, the great number of restriction fragments obtained with the restriction enzymes used prevented precise comparisons and determination of molecular weights.  相似文献

In the autumn of 1953 an experiment was begun to follow changes in the cereal-root eelworm population of small plots on a field in Shropshire. The plots were cropped with either oats, wheat, barley or rye, sown in the autumn and spring, and some plots had fertilizer. Each plot received the same treatment for 3 years; in the fourth year an indicator crop of spring oats was grown on all the plots.
Under rye and autumn-sown wheat the eelworm population fell to a level which permitted a good oat crop in 1957. Autumn-sown wheat, barley and rye generally produced lower eelworm populations than their spring-sown counterparts, but autumn-sown oats proved to be the most efficient host. The order of host efficiency was oats (best), barley, wheat, rye. The eelworm populations were generally higher on plots receiving fertilizer treatment. In this experiment all oat plots, and spring-sown barley plus fertilizer, produced populations which severely damaged the 1957 oat crop.  相似文献

THE INCIDENCE OF CERTAIN SEED-BORNE DISEASES IN COMMERCIAL SEED SAMPLES   总被引:1,自引:0,他引:1  
The incidence of ergot, Claviceps purpurea (Fr.) Tul., in commercial cereal seed samples submitted to the Official Seed Testing Station from 1918 to 1957 has been examined.
Rye samples were more frequently contaminated than wheat or barley and no cereal ergots were recorded in oats.
A relatively high proportion of ergot-contaminated rye samples occurred every 8–10 years. This appeared to be associated with high relative humidity and low maximum temperature during June. Similar trends were found for wheat and barley.
No varietal differences in susceptibility to ergot contamination were found for rye or barley, but there was some evidence that spring wheat varieties were more frequently contaminated than winter ones.  相似文献

Total globulins from embryos and endosperms of barley, wheat,rye, and oats were separated by SDS-PAGE under reducing andnon-reducing conditions. The preparations from embryos of allfour cereals contained major groups of bands with Mr's of 50-60000,which were not affected by reduction. These have been characterizedpreviously from oats and shown to correspond to subunits ofthe 7S storage globulin. Immunochemical relationships betweenthese bands (and others with Mr's between 40000 and 70000) weredemonstrated by immunodiffusion and ‘Western Blotting’using antiserum raised against the major subunits of the oat7S globulins. The 7S globulins were also prepared from hand-dissectedembryos of the four cereals using sucrose density ultracentrifugation.Their amino acid compositions were broadly similar, but differedfrom those of the 7S vicilins of legumes. An experiment examined the intake, growth responses and rumen digestion of young sheep fed ad libitum oat or barley straws alone or supplemented with approximately isonitrogenous amounts of barley grain and urea (Bar/N), safflower seed meal (SAF) or linseed meal (LIN) supplements provided at 3 day intervals. The supplements comprised 15–22% of total dry matter (DM) intake. Sheep offered either of the straws alone consumed 35.0–37.2 g DM/kg liveweight (LW0.75) per day of straw and an estimated 2.03–2.07 MJ metabolizable energy (ME) per day, and lost 85–97 g LW per day. Supplements increased (P<0.05 or <0.001) voluntary intake of straw and of total DM, and the organic matter (OM) digestibility of the entire diet. Each of the supplements increased (P<0.001) the estimated ME intake to a similar extent and changed the rapid LW loss of sheep fed straw alone to approximate LW maintenance. Rumen ammonia concentrations in sheep fed barley and oat straws alone (12 and 24 mg NH3/l, respectively) were expected to be deficient for microbial activity, but were increased (P<0.001) by provision of the supplements. Digestion of straw in synthetic fibre bags incubated in the rumen was markedly increased (P<0.01 or <0.001) when supplements were provided. Rumen pH was depressed briefly to pH <6.0 by the Bar/N, but not by the LIN or SAF, supplements. In young sheep fed cereal straws and losing LW rapidly the oilseed meal supplements increased wool growth more than the barley grain–urea supplements, but both types of supplement increased ME intake similarly and were equally effective to reduce the extent of LW loss.  相似文献

In a long-lasting field experiment at the Research Station Hanninghof at Dülmen (Westphalia), laid out in 1971, the inoculum potential (IP) of vesicular-arbuscular mycorrhizal (VAM) fungi of soil from winter barley in a continuous monoculture (CM) and in a four-years cereal crop rotation (CR: winter wheat, winter barley, winter rye, oats) was estimated from 1980–1982. The influence of green manure and increased mineral N-fertilization (100 and 200 kg ha–1) on VAM-IP was also assessed. VAM-IP was high at both levels of N-application, although the higher N-fertilization resulted in a reduction of VAM-IP in winter barley — in CR as well as in CM. No influence of N-fertilization on VAM-IP was observed with oats and winter rye, which were only cultivated in CR. Green manure had a negative effect on IP in CM barley, but not in CR barley. This may be due to the annual applications of green manure in CM, while green manure in CR was only applied prior to the cultivation of oats and winter wheat, but not to winter barley. Except for the effects of green manure remarkable differences in VAM-IP were found neither in CR nor in CM of winter barley. This is also reflected in the yields of barley in CM and indicates the relative self-tolerance of winter barley.  相似文献

A number of defined desiccation treatments without low temperature exposure were able to induce freezing tolerance in 20 cultivars of winter cereals. A maximal degree of freezing tolerance was induced in epicotyls at 24°C in 24 hours at 40% relative humidity in rye and wheat, 7 days at 54% RH in barley, and 4 days at 70% RH in oats. Freezing tolerance was not correlated to water content of the plants after desiccation treatment but was related to the genetic capacity of the cultivars to frost harden. Levels of freezing tolerance induced by desiccation were similar to those induced by cold acclimation in rye and wheat, but considerably less in barley and oats. This is associated with a more rapid desiccation injury in barley and oats, precluding the completion of the hardening process.  相似文献

The yield of wheat and the incidence of take-all were measured in crops grown in six different 4-year sequences, repeated in 3 successive years. The first crop of winter wheat grown after oats or beans yielded 13–23 cwt/acre (1632–2887 kg/ha) more grain than wheat after wheat or barley. Spring wheat after oats yielded 2–5 cwt/acre (250–625 kg/ha) more than spring wheat after wheat. The smaller yields of wheat after wheat or barley were caused mostly by greater prevalence of take-all. Regression analysis indicates that each 1 % increase in straws with take-all decreased yield of winter wheat by 0·6%. Take-all was more prevalent in the second and third successive wheat crops after oats than in the fourth crop.  相似文献

Ten isolates ofFusarium graminearum Schwabe originating from diseased cereal plants and kernels were tested for pathogenicity to various cultivars of wheat, rye, triticale and oats. The isolates varied greatly in their pathogenicity to the seedlings of the species, and were most pathogenic to rye and triticale, less pathogenic to barley and wheat and least pathogenic to oats.  相似文献

The 2-oxoglutarate carrier from the inner membrane of bovine heart mitochondria was purified by chromatography on hydroxyapatite / celite and reconstituted with egg yolk phospholipid vesicles by the freeze-thaw-sonication technique. In the reconstituted system the incorporated 2-oxoglutarate carrier catalyzed a first-order reaction of 2-oxoglutarate / 2-oxoglutarate exchange. The substrate affinity for 2-oxoglutarate was determined to be 65 ± 18 μM (15 determinations) and the maximum exchange rate at 25°C reaches 4000–22000 μmol / min per g protein, in dependence of the particular reconstitution conditions. The activation energy of the exchange reaction is 54.3 kJ / mol. The transport is independent of pH in the range between 6 and 8. When the first fraction of the hydroxyapatite / celite column eluate was used for reconstitution, besides the 2-oxoglutarate / 2-oxoglutarate exchange, a significant activity of unidirectional uptake was observed. This activity may be due to a population of the carrier protein which is in a different state.  相似文献

Newly germinated seedlings of susceptible cultivars of oats, wheat, barley and rye were inoculated with second-stage juveniles of Heterodera avenae in pots of sand. Subsequent examination showed oat root tips to be more commonly invaded, and by a greater range of nematode numbers than the other cereals. A comparison of oats and barley showed that lower nematode numbers in barley were not due to a higher emigration from barley; invasion, establishment and emigration by nematodes all being greater in oats. Second-stage juveniles were more likely to migrate prior to establishment in barley than in oats.  相似文献

We have found extensive nucleosomal fragmentation of native DNA extracted from leaves of healthy cereal plants, as indicated by ladder patterns on agarose gels and TUNEL staining. The time of first appearance of fragmentation differed among cereals. Native DNA from the first leaf of 10-day-old plants formed a clear ladder pattern of multiples of 180 bp fragments in wheat and triticale but not in barley and oats. In one cultivar of rye a weak ladder pattern occurred but not in another. Freezing and thawing of samples before DNA extraction resulted in much more extensive DNA fragmentation in wheat but not in rye and barley, indicating that DNA-degrading enzymes are present in the cytoplasm of wheat, but not in barley and rye, at this stage. In barley, nucleosomal fragmentation was first detected in 25-day-old plants. These results indicate that programmed cell death takes place in developing leaves of young cereal plants, but that the time of onset differs among cereal species.  相似文献

A family of related retroelements was characterized in the genomes of some Graminease species. The structure of these retroelements indicates that they are retrotransposons containing reading frames with sequence similarity to the polyproteins of copia and Ty. This family of retroelements (termed WIS-2) occurs in the genomes of barley, wheat, rye, oats, and Aegilops species. Ongoing genomic variation both within individual plants of a wheat variety and within and between varieties of wheat is associated with some members of the WIS-2 family.  相似文献

The effect of DNA fragment size on the extent of hybridisation that occurs between repeated sequence DNAs from oats, barley, wheat and rye has been investigated. The extent of hybridisation is very dependent on fragment size, at least over the range of 200 to 1000 nucleotides. This is because only a fraction of each fragment forms duplex DNA during renaturation. From these results estimates of the proportions of repeated sequences of each of the cereal genomes that are homologous with repeated sequences in the other species have been determined and a phylogenetic tree of cereal evolution constructed on the basis of the repeated sequence DNA homologies. It is proposed that wheat and rye diverged after their common ancestor had diverged from the ancestor of barley. This was preceded by the divergence of the common ancestor of wheat, rye and barley and the ancestor of oats. Once introduced in Gramineae evolution most families of repeated sequences appear to have been maintained in all subsequently diverging species. — The repeated sequences of oats, barley, wheat and rye have been divided into Groups based upon their presence or absence in different species. Repeated sequences of related families are more closely related to one another within a species than between species. It is suggested that this is because repeated sequences have been involved in many rounds of amplification or quantitative change via unequal crossing over during species divergence in cereal evolution.  相似文献

Prolamin mixtures were isolated from oats, rice, normal and high-lysine sorghum, two varieties of pearl millet, two strains of teosinte, and gamma grass and subjected to NH2-terminal amino acid sequence determinations. In each case (except for rice, whose prolamins apparently have blocked or unavailable NH2-terminal residues), primarily a single sequence was observed despite significant heterogeneity, suggesting that prolamin homology in each cereal arose through duplication and mutation of a single ancestral gene. Comparisons were then made to prolamin sequences previously determined for wheat, corn, barley, and rye. Within genera, different varieties or subspecies exhibited few differences, but more distantly related genera, subtribes, and tribes showed increasingly large differences. Within the subfamily Festucoideae, no homology was apparent between prolamins of oats and those of the subtribe Triticinae (including wheat, rye, and barley, for which prolamin homology was previously demonstrated). Within the subfamily Panicoideae, corn was shown to be closely related to teosinte but more distantly to Tripsacum. Sorghum was shown to have diverged less from corn than had millet. These comparisons demonstrate that prolamin sequence analyses can successfully predict and clarify evolutionary relationships of cereals.  相似文献

The saccharification of the polysaccharides of barley, oat, and wheat straws and Solka Floc was studied using the extracellular enzyme system synthesized by mutant strain NTG III/6 of the fungus Penicillium pinophilum 87160iii. The enzymes obtained in cultures containing Solka Floc or barley straw as the carbon source were compared. Solka Floc at 10% (w/v) concentration was hydrolyzed to the extent of 70% in 72 h at 50 degrees C using a reaction mixture containing 7 filter paper units/mL of cellulase induced on Solka Floc, but hydrolysis was increased to 90% when the enzyme induced on barley straw was used. Under the same conditions, the polysaccharides in barley, oat, and wheat straws were hydrolyzed, respectively, in 72 h, to the extent of 42-48%, 62%, and 52%, but hydrolysis was increased to 93%, 100%, and 92%, respectively, after treatment of the substrates with alkaline-H(2)O(2) reagent at room temperature.  相似文献

We investigated the kinetics of hot liquid water (HLW) hydrolysis over a 60-min period using a self-designed setup. The reaction was performed within the range 160–220 °C, under reaction conditions of 4.0 MPa, a 1:20 solid:liquid ratio (g/mL), at 500 rpm stirring speed. Xylan was chosen as a model compound for hemicelluloses, and two kinds of agricultural wastes–rice straw and palm shell–were used as typical feedstocks representative of herbaceous and woody biomasses, respectively. The hydrolysis reactions for the three kinds of materials followed a first-order sequential kinetic model, and the hydrolysis activation energies were 65.58 kJ/mol for xylan, 68.76 kJ/mol for rice straw, and 95.19 kJ/mol for palm shell. The activation energies of sugar degradation were 147.21 kJ/mol for xylan, 47.08 kJ/mol for rice straw and 79.74 kJ/mol for palm shell. These differences may be due to differences in the composition and construction of the three kinds of materials. In order to reduce the decomposition of sugars, the hydrolysis time of biomasses such as rice straw and palm shell should be strictly controlled.  相似文献

Twelve species of lacewings: Chrysopa altaica, Ch. commata, Ch. perplexa, Ch. phyllochroma, Ch. dasyptera, Ch. carnea, Ch. formosa, Ch. intima, Ch. perla, Ch. prasina, Ch. septempunctata, and Nineta inpunctata were found in cereal agroecosystems of the forest-steppe zone of Western Siberia. The dominant species were Ch. carnea and Ch. phyllochroma. The biological characteristics, seasonal dynamics of the abundance of lacewings in the agrocenoses of winter rye, spring wheat, and oats are given. The abundance of larval and adult lacewings in the spring wheat agrocenosis was not affected by the level of chemicalization (upon condition of the rational use of insecticides), tillage variant, and predecessor crop. In the years when the density of lacewings was low, no differences in their population were found between the agrocenoses of wheat (after fallow), winter rye, oats, vetch-oats, canola, barley, and barley with melilot. In the years characterized by relatively high abundance of lacewings, they occurred more frequently in the crop rotations with wheat after fallow and with oats. These plants were settled by cereal aphids to the greatest extent. In all the years studied, the density of lacewings on alfalfa was 2–2.8 times as great as that on wheat after fallow.  相似文献

Acylation reactions of naringin with palmitic acid were performed by a lipase after formation of highly concentrated homogeneous solutions. Their initial naringin concentration was 840–950 mM, which is 20–60 times greater than that in organic solvent media. The overall productivity of highly concentrated solutions was more than 15 times greater than those of organic phase media. The addition of DMSO (20–40%, w/w) to substrate mixtures lowered the melting temperature of a naringin–palmitic acid mixture (1:1 molar ratio) to about 40 °C. Reactions at 80 °C apparently followed Michaelis–Menten kinetics despite extremely high substrate concentrations. As the temperature increased from 60 °C to 80 °C, the apparent viscosity of the highly concentrated solution decreased remarkably from 4.31 Pa s to 0.063 Pa s. An activation energy of 7.65 kcal/mol obtained in a range of 60–75 °C suggests a diffusion-control. On the other hand, an activation energy of 17.09 kcal/mol in a range of 75–90 °C indicates a reaction-control. The highest product conversion yield of 33% (mol/mol) was obtained in a 10 h reaction at 80 °C. Addition of activated molecular sieves to the highly concentrated solution increased the product conversion yield by 7% (mol/mol), suggesting that the original equilibrium was disrupted by removing water and then a new equilibrium was reached.  相似文献

Assimilable nitrogen in various forms prolonged the life of Ophiobolus graminis in infected wheat straw, whether added directly to the straw or to the surrounding soil. When the infected straws were buried in washed quartz sand, 0.5 g. nitrogen per 100 g. air-dry straw was the optimum dressing for longevity of Ophiobolus. Addition of sodium phosphate did not significantly increase longevity.
Nitrogen is considered to prolong the life of Ophiobolus by enabling the mycelium to form new branch hyphae, which can explore unexhausted parts of the substrate; it is suggested that aged mycelium dies from carbohydrate starvation, through exhaustion of the zones of enzymic erosion around the hyphae. This hypothesis is supported by the extended life of the fungus in infected straws that were shaken twice weekly in 3 % dextrose solution.
Ophiobolus was found to survive longer in infected straws buried in a fallow soil than in the same soil under oats, mustard or trefoil; this finding suggests the use of catch crops as competitors with Ophiobolus for soil nitrogen.  相似文献
