Coenagrion mercuriale (Charpentier) (Odonata: Zygoptera) is one of Europe's most threatened damselflies and is listed in the European Habitats directive. We combined an intensive mark-release-recapture (MRR) study with a microsatellite-based genetic analysis for C. mercuriale from the Itchen Valley, UK, as part of an effort to understand the dispersal characteristics of this protected species. MRR data indicate that adult damselflies are highly sedentary, with only a low frequency of interpatch movement that is predominantly to neighbouring sites. This restricted dispersal leads to significant genetic differentiation throughout most of the Itchen Valley, except between areas of continuous habitat, and isolation by distance (IBD), even though the core populations are separated by less than 10 km. An urban area separating some sites had a strong effect on the spatial genetic structure. Average pairwise relatedness between individual damselflies is positive at short distances, reflecting fine-scale genetic clustering and IBD both within- and between-habitat patches. Damselflies from a fragmented habitat have higher average kinship than those from a large continuous population, probably because of poorer dispersal and localized breeding in the former. Although indirect estimates of gene flow must be interpreted with caution, it is encouraging that our results indicate that the spatial pattern of genetic variation matches closely with that expected from direct observations of movement. These data are further discussed with respect to possible barriers to dispersal within the study site and the ecology and conservation of C. mercuriale. 1. Genetic techniques are being used increasingly to address questions about dispersal and gene flow of freshwater invertebrates. However, population genetic structure can be affected by factors other than dispersal. Many stream insects have long life cycles that result in the simultaneous existence of multiple cohorts throughout the larval development period. If larval development is fixed, successive cohorts may be reproductively isolated and, as a result, genetically distinct. In such cases, significant levels of genetic differentiation between cohorts could confound estimates of dispersal based on population genetic structure.
2.  Peltoperla tarteri is a stonefly that can be abundant in Appalachian headwater streams. Although P. tarteri is univoltine at the type locality (Big Paint Hollow, WV, U.S.A.), the study populations in southwestern Virginia, U.S.A., were semivoltine. This semivoltine life cycle results in the simultaneous existence of multiple cohorts with the potential for significant genetic differentiation among them.
3. Levels of genetic differentiation among P. tarteri cohorts were analysed with mitochondrial DNA (mtDNA) sequence data from the non-coding origin of replication or `control' region from 93 individuals from two successive cohorts (collected in 1998 and 1999).
4. Analysis of molecular variance ( AMOVA ) indicated no genetic differentiation among cohorts ( F ST=0.0), and gene flow among cohorts was very high ( Nm =∞).
5. High levels of gene flow among cohorts suggest that larval development of P. tarteri is not fixed. Gene flow among cohorts most likely occurs as a result of a cohort split in which some individuals complete development in one or three years instead of two.  相似文献

Genetic and demographic estimates of dispersal are often thought to be inconsistent. In this study, we use the damselfly Coenagrion mercuriale (Odonata: Zygoptera) as a model to evaluate directly the relationship between estimates of dispersal rate measured during capture-mark-recapture fieldwork with those made from the spatial pattern of genetic markers in linear and two-dimensional habitats. We estimate the 'neighbourhood size' (Nb) - the product of the mean axial dispersal rate between parent and offspring and the population density - by a previously described technique, here called the regression method. Because C. mercuriale is less philopatric than species investigated previously by the regression method we evaluate a refined estimator that may be more applicable for relatively mobile species. Results from simulations and empirical data sets reveal that the new estimator performs better under most situations, except when dispersal is very localized relative to population density. Analysis of the C. mercuriale data extends previous results which demonstrated that demographic and genetic estimates of Nb by the regression method are equivalent to within a factor of two at local scales where genetic estimates are less affected by habitat heterogeneity, stochastic processes and/or differential selective regimes. The corollary is that with a little insight into a species' ecology the pattern of spatial genetic structure provides quantitative information on dispersal rates and/or population densities that has real value for conservation management.  相似文献

1.  Governmental authorities in many countries financially support the implementation of habitat connectivity measures to enhance the exchange of individuals among fragmented populations. The evaluation of the effectiveness of such measures is crucial for future management directions and can be accomplished by using genetic methods.
2.  We retraced the population history of the European tree frog in two Swiss river valleys (Reuss and Thur), performed comprehensive population sampling to infer the genetic structure at 11 microsatellite markers, and used first-generation migrant assignment tests to evaluate the contemporary exchange of individuals.
3.  Compared with the Thur valley, the Reuss valley has lost almost double the number of breeding sites and exhibited a more pronounced genetic grouping. However, similar numbers of contemporary migrants were detected in both valleys. In the Reuss valley, 81% of the migration events occurred within the identified genetic groups, whereas in the Thur valley migration patterns were diffuse.
4.  Our results show that the connectivity measures implemented in the Reuss valley facilitated effective tree frog migration among breeding sites within distances up to 4 km. Nevertheless, the Reuss valley exhibited high genetic differentiation, which reflected the impact of barriers to tree frog movement such as the River Reuss. By contrast in the Thur valley, a larger number of breeding sites have been preserved and high admixture indicated exchange of individuals at distances up to 16 km.
5.   Synthesis and applications . We show that genetic methods can substantiate the effectiveness of connectivity measures taken in conservation management at the landscape scale. We urge responsible authorities from both river valleys to continue implementing connectivity measures and to create a dense network of breeding sites, as spatial gaps of 8 km are rarely traversed by tree frogs.  相似文献

Many species, particularly insects, pass through a series of distinct phases during their life history, with the developmental timing directed towards appropriate resources. Any factor that creates variation in developmental timing may partition a population into discrete populations-or 'cohorts'. Where there is continued failure to recruit outside the natal cohort then alternate cohorts will have their own internal dynamics, eventually leading to independent demographic and evolutionary trajectories. By contrast, continued variation in development rates within a cohort-cohort splitting-may homogenise otherwise independent demographic units. Using a panel of 14 microsatellite loci, we quantify the genetic signature of apparent demographic isolation between coexisting, but alternate, semivoltine cohorts of the damselfly Coenagrion mercuriale at locations that span its distribution in the UK. We find consistently low levels of genetic divergence between sympatric cohorts of C. mercuriale, indicative of developmental plasticity during the larval stage (unregulated development) whereby some individuals complete their development outside the predominant 2-year (semivoltine) period. Thus, individuals that alter their developmental rate successfully recruit to a different cohort. Despite maintaining contrasting population sizes, gene flow between alternate cohorts broadly is sufficient to place them on a similar evolutionary trajectory and also buffers against loss of genetic diversity. Such flexible larval development permits a response to local conditions and may facilitate response to environmental change.  相似文献

1.  Detailed studies of population ecology are usually carried out in relatively restricted areas in which emigration and immigration play a role. We used a modelling approach to explore the population consequences of such dispersal and applied ideas from our simulations to the conservation of wild birds.
2.  Our spatial model incorporates empirically derived variation in breeding output between habitats, density dependence and dispersal. The outputs indicate that dispersal can have considerable consequences for population abundance and distribution. The abundance of a species within a patch can be markedly affected by the surrounding habitat matrix.
3.  Dispersal between habitats may result in lower population densities at the edge of good quality habitat blocks and could partially explain why some species are restricted to large habitat fragments.
4.  Habitat deterioration may not only lead to population declines within that habitat but also in adjacent habitats of good quality. This may confound studies attempting to diagnose population declines.
5.  Although mobile species have the advantages of colonizing sites within metapopulations, dispersal into poorer quality territories may markedly reduce total populations.
6.  There are two main approaches to conservation: one is to concentrate on establishing and maintaining protected areas, while the other involves conservation of the wider countryside. If dispersal is an important process then protecting only isolated areas may be insufficient to maintain the populations within them.  相似文献

Dispersal has a potentially profound effect on the dynamics of populations especially when a population occupies a patchy habitat. Ponds surrounded by terrestrial landscape are an example of patchy distribution of physical conditions and constitute "islands" for odonates. Few studies have focussed on dispersal in odonates. We have used the direct method of dispersal observing (capture-mark-recapture technique) in order to estimate the degree of linkage in three patchy populations of odonate localised on three ponds. We also examined the differences in dispersal ability within and among three species ( Coenagrion puella , Coenagrion scitulum and Libellula depressa ). The ponds were situated in southwest France on a limestone plateau. In this arid area, these ponds constitute the only surface water available and are relatively sparsely distributed. The size of the ponds ranged from 48 to 79 m2 and they were 200 and 775 m apart. We demonstrated that three factors influence the dispersal ability of these odonates. The first is represented by the abiotic factors and especially weather conditions. This determines the number of days that dispersal is possible. The second is interspecific differences. We showed that sensitivity to weather conditions, species size and species behaviour influence dispersal ability. The third factor is the intraspecific characteristics. We demonstrated that there are differences in dispersal ability according to sex and age. To conclude, we discuss the importance of pond management to maintain the existing odonate populations and to facilitate introduction of new populations in this region where little exchange occurs between ponds.  相似文献

Abstract. 1. Dispersal of adult aquatic insects between streams may have important consequences for local and regional population dynamics, but little is known about how dispersal is affected by weather conditions.
2. The influence of meteorological variables on flight activity of adult stoneflies (Plecoptera: Leuctridae, Nemouridae, and Chloroperlidae) was investigated using Malaise traps adjacent to three upland streams in the Plynlimon area of mid Wales, U.K.
3. Numbers of adult stoneflies captured weekly in the traps were related positively to air temperature and related negatively to wind speed. Meteorological conditions during daylight showed stronger relationships with flight activity than did conditions at night.
4. There was inter-site variation in the strength of weather effects on stonefly flight. Wind speed was significant at only one site, which had higher average wind speed than the other sites.
5. Annual variation in weather conditions during adult flight periods may result in varying extent of dispersal between sites, influencing community dynamics over a wide area.  相似文献

1. River corridors are well-known for their role in plant dispersal. The buoyancy of seeds, the possibility of dispersal by vegetative fragments, and the frequency and efficiency of dispersal among different river catchments determine linear distribution patterns. Little is known about the relative importance of these factors to observed patterns of genetic variation.
2. One hundred and fifty-six Nuphar lutea individuals from forty-four sampling sites in the river catchments of the Cidlina River, the Mrlina River and the Labe River (Czech Republic) were studied using ten microsatellite markers. Interpretation of patterns in genetic variation allowed several conclusions about dispersal mechanisms.
3. Vegetative long-distance dispersal is probably very limited in this species. Only one multilocus genotype was found in more than one sampling site. The distance between the sites was about 75 km.
4. To explain the distribution of Bayesian based clusters of related multilocus genotypes, both along-river and inter-river long-distance dispersals have to be invoked.
5. A marginally significant tendency for higher genetic diversity in the lower part of the river Cidlina was detected. Continuous downstream dispersal of seeds by water currents could be a valid explanation.
6. Significant positive autocorrelation was found among individuals at within-river distances of up to 25 km. Repeated dispersal of seeds over distances in the range of tens of kilometers is common.  相似文献

Wallacean island populations of two Vespertilioninae bats, Myotis muricola and Scotophilus kuhlii , which have similar geographical ranges, showed marked contrast in the amount and pattern of genetic variation. Within islands, genetic variation was on average much higher in M. muricola but declined from west to east, whereas all populations of S. kuhlii had uniformly low levels of genetic variation by mammalian standards. S. kuhlii showed little genetic differentiation between islands and estimates of gene flow were substantial whereas island populations of M. muricola differed markedly and there was a strong isolation-by-distance effect associated with the extent of the sea crossing between islands. Furthermore, the lower mean heterozygosity and small genetic distances between eastern island populations of M. muricola is evidence that there has been a bottleneck associated with the colonization of this area. The attenuation of genetic diversity to the east is also seen in some other mammalian species and may indicate limits to dispersal and have implications for species management. Abstract.  1. To persist in a highly fragmented environment, such as the intensively used agricultural landscapes of Europe, species may need to successfully disperse between sub-populations. This paper describes a capture – mark – resight experiment to investigate movement and dispersal patterns of the flightless bush cricket species Pholidoptera griseoaptera .
2. Movement parameters showed little difference between juveniles and imagos as well as between males and females. Both developmental stages and sexes moved considerable distances, were cap\able of crossing habitat boundaries, and showed successful inter-patch dispersal through the agricultural matrix.
3. The results suggest that P. griseoaptera exhibits movement behaviours that compensate for its inability to fly. It was concluded that the good dispersal ability exhibited by animals of both sexes and developmental stage is an important factor allowing P. griseoaptera to persist in fragmented agricultural landscapes.  相似文献

The level of gene flow is an important factor influencing genetic differentiation between populations. Typically, geographic distance is considered to be the major factor limiting dispersal and should thus only influence the degree of genetic divergence at larger spatial scales. However, recent studies have revealed the possibility for small-scale genetic differentiation, suggesting that the spatial scale considered is pivotal for finding patterns of isolation by distance. To address this question, genetic and morphological differentiation were studied at two spatial scales (range 2–13 km and range 300 m to 2 km) in the perch ( Perca fluviatilis L.) from the east coast archipelago of Sweden, using seven microsatellite loci and geometric morphometrics. We found highly significant genetic differentiation between sampled locations at both scales. At the larger spatial scale, the distance per se was not affecting the level of divergence. At the small scale, however, we found subtle patterns of isolation by distance. In addition, we also found morphological divergence between locations, congruent with a spatial separation at a microgeographic scale, most likely due to phenotypic plasticity. The present study highlights the importance of geographical scale and indicates that there might be a disparity between the dispersal capacity of a species and the actual movement of genes. 1. River systems offer special environments for the dispersal of aquatic plants because of the unidirectional (downstream) flow and linear arrangement of suitable habitats.
2. To examine the effect of this flow on microevolutionary processes in the unbranched bur-reed ( Sparganium emersum ) we studied the genetic variation within and among nine (sub)populations along a 103 km stretch of the Niers River (Germany–The Netherlands), using amplified fragment length polymorphisms.
3. Genetic diversity in S. emersum populations increased significantly downstream, suggesting an effect of flow on the pattern of intrapopulation genetic diversity.
4. Gene flow in the Niers River is asymmetrically bidirectional, with gene flow being approximately 3.5 times higher in a downstream direction. The observed asymmetry is probably caused by frequent hydrochoric dispersal towards downstream locations on the one hand, and sporadic zoochoric dispersal in an upstream direction on the other. The spread of vegetative propagules (leaf and stem fragments) is probably not an important mode of dispersal for S. emersum , suggesting that gene flow is mainly via seed dispersal. Realized dispersal distances exceeded 60 km, revealing a potential for long-distance dispersal in S. emersum .
5. There was no correlation between geographical and genetic distances among the nine S. emersum populations (i.e. no isolation by distance), which may be due to the occurrence of long-distance dispersal and/or colonization and extinction dynamics in the Niers River.
6. Overall, the genetic population structure and regional dispersal patterns of S. emersum in the Niers River are best explained by a linear metapopulation model. Our study shows that flow can exert a strong influence on population genetic processes of plants inhabiting stream systems.  相似文献

Abstract 1. The high-altitude wind-borne migration of the diamondback moth Plutella xylostella in the U.K. in 2000 was investigated (a) by direct monitoring of insect flight by vertical-looking radar and by aerial netting, and (b) through evidence of temporal variation in P. xylostella abundance deduced from a network of light traps.
2. Migrating P. xylostella were identified by a unique combination of size and shape data derived from the continuously operating vertical-looking radar.
3. Radar-detected migratory overflights correlated significantly with associated peaks in abundance of P. xylostella estimated by catches in a U.K.-wide light trap network; however the correlation was stronger when light trap catches were lagged by 1 day.
4. The first notable catches of P. xylostella in the U.K. occurred in early May, and were accompanied by migrations over the radar from the east.
5. Radar data and back-tracking indicated that a major wind-borne migration of P. xylostella from The Netherlands to southern England took place in early May, and that this was responsible for the establishment of the U.K. population.
6. The origin of early-season P. xylostella occurring in Britain is discussed.  相似文献

Migratory movements of the endangered stag beetles Lucanus cervus (18 males, 38 females) were monitored radio-telemetrically for three reproductive periods (2003–2005). The aim of the study was to estimate the migratory range of free-ranging individuals as a measure of connectivity among neighbouring populations for future conservation measures. Miniature transmitters of c . 350 mg (battery life: 10–15 days) were attached externally to the pronotum. Transmitter/beetle mass ratio was 12.8% on average (7.1–28.0%). Male dispersal behaviour consisted of frequent flights directed to sites with reproductive females and rarely of on-ground movement. Total displacement distance recorded was up to 2065 m, the maximum distance of a single flight being 1720 m. Flights always began at elevated structures such as trees and shrubs and took place in an air temperature range of 11–27°C. Within this range, temperature did not influence flight distance. Female dispersal behaviour consisted mostly of a single flight, followed by mating and consequent ground movements towards oviposition sites. Total displacement distance recorded was up to 762.6 m, the maximum distance of a single flight being 701 m. Climatic constraints of flights were the same as in males. Modelling the dispersal behaviour suggests that about 1% of males are capable of maintaining gene flux among nest sites within a radius of about 3 km. However, the colonization of new nest sites depends on the dispersal ability of females and amounts to less than 1 km. Thus, isolated populations (distance to the next population greater than 3 km) have an increased probability of local extinction.  相似文献

Abstract.  1. Species that undertake regular two-way migration may be expected, through population connectivity, to exhibit some level of genetic similarity over broad spatial scales. Although seldom following two-way migration, highly mobile insect species tend to exhibit either low haplotype diversity and no phylogeographic structuring, or relatively high haplotype diversity and pronounced phylogeographic structuring.
2. This study reveals the first wide-scale genetic characterisation of a migratory dragonfly, the common green darner Anax junius Drury. Unusually for insects, north-south two-way migration is common in this species, although not obligatory. In at least part of its range, some individuals follow an extended developmental period and overwinter in a state of diapause.
3. Mitochondrial sequence data were obtained from 92  A. junius individuals collected from 35 sites across Canada, U.S.A., and Mexico. These revealed 38 haplotypes, some of which were extremely widespread, although the majority (27 haplotypes) was found in only one individual. In contrast to previous studies on mobile insects, the overall pattern was of relatively high haplotype diversity in the absence of phylogeographic structuring.
4. Migrants and non-migrants, which sometimes shared haplotypes, were distributed across multiple genetic lineages. This suggests that, contrary to some earlier assertions, developmental pathways in this species may be plastic. Such plasticity would allow highly mobile species to adapt to a range of environmental conditions, and may be key to the widespread distribution of multiple haplotypes.  相似文献

Abstract.  1. Aquatic macro-invertebrates have frequently been used as biological indicators in lotic environments but much less commonly so in lentic habitats. Dragonflies and damselflies (Order Odonata) satisfy most selection criteria for lentic bioindicators of grazing impacts.
2. Intensive cattle grazing affects most of the Canadian prairie pothole region but the effects of grazing on wetlands are poorly understood.
3. Here the vegetation structure and invertebrate community composition of 27 prairie potholes in Alberta, Canada were studied and compared. Wetlands were evenly divided into three treatments of different grazing regimes.
4. Removal of emergent vegetation by cattle grazing decreased odonate abundance and reproductive effort. Shorter Scirpus acutus stems resulted in significantly fewer damselflies (Suborder Zygoptera) and lower reproductive efforts.
5. Overall odonate diversity was affected by the height of key plant species, highlighting the importance of the vegetation structure of both emergent vegetation for breeding and adjacent upland vegetation for nocturnal roosts. Wetland vegetation structure was more important than vegetation composition to the life history of odonates.
6. Wetland water quality parameters of nitrogen, phosphorus, total dissolved solids (TDS), and chlorophyll-a concentration did not change due to the presence of grazing cattle at wetlands so water quality influences were rejected as mechanisms of change.
7. Larval odonate diversity and abundance was positively correlated with overall aquatic macro-invertebrate diversity and abundance, hence it was concluded that the larval odonate community can be an accurate bioindicator of intactness and diversity of overall aquatic macro-invertebrate communities in Canadian prairie wetlands.  相似文献

Abstract. 1. The grasshopper species Chorthippus brunneus and C. jacobsi form a complex mosaic hybrid zone in northern Spain. Two mark–release–recapture studies were carried out near the centre of the zone in order to make direct estimates of lifetime dispersal.
2. A model framework based on a simple random walk in homogeneous habitat was extended to include the estimation of philopatry and flying propensity. Each model was compared with the real data, correcting for spatial and temporal biases in the data sets.
3. All four data sets (males and females at each site) deviated significantly from a random walk. Three of the data sets showed strong philopatry and three had a long dispersal tail, indicating a low propensity to move further than predicted by the random walk model.
4. Neighbourhood size estimates were 76 and 227 for the two sites. These estimates may underestimate effective population size, which could be increased by the long tail to the dispersal function. The random walk model overestimates lifetime dispersal and hence the minimum spatial scale of adaptation.
5. Best estimates of lifetime dispersal distance of 7–33 m per generation were considerably lower than a previous indirect estimate of 1344 m per generation. This discrepancy could be influenced by prezygotic isolation, an inherent by-product of mosaic hybrid zone structure.  相似文献

Landscape genetics aims to assess the effect of the landscape on intraspecific genetic structure. To quantify interdeme landscape structure, landscape genetics primarily uses landscape resistance surfaces (RSs) and least-cost paths or straight-line transects. However, both approaches have drawbacks. Parameterization of RSs is a subjective process, and least-cost paths represent a single migration route. A transect-based approach might oversimplify migration patterns by assuming rectilinear migration. To overcome these limitations, we combined these two methods in a new landscape genetic approach: least-cost transect analysis (LCTA). Habitat-matrix RSs were used to create least-cost paths, which were subsequently buffered to form transects in which the abundance of several landscape elements was quantified. To maintain objectivity, this analysis was repeated so that each landscape element was in turn regarded as migration habitat. The relationship between explanatory variables and genetic distances was then assessed following a mixed modelling approach to account for the nonindependence of values in distance matrices. Subsequently, the best fitting model was selected using the statistic. We applied LCTA and the mixed modelling approach to an empirical genetic dataset on the endangered damselfly, Coenagrion mercuriale. We compared the results to those obtained from traditional least-cost, effective and resistance distance analysis. We showed that LCTA is an objective approach that identifies both the most probable migration habitat and landscape elements that either inhibit or facilitate gene flow. Although we believe the statistical approach to be an improvement for the analysis of distance matrices in landscape genetics, more stringent testing is needed.  相似文献

All extant populations of Hebe speciosa (Plantaginaceae), a threatened endemic New Zealand shrub, were investigated using the amplified fragment length polymorphism technique (AFLP). Genetic diversity indices varied significantly among geographical regions and were positively correlated with population size. Among-population genetic differentiation was high (mean pairwise ΦST = 0.47), implying complex historical relationships between disjunct populations and negligible contemporary gene flow. Southern populations exhibited extremely low genetic diversity relative to those found in Northland, suggesting that these populations may be more recent in origin. Patterns of genetic relationship among some populations indicate pre-European Māori dispersal and cultivation. The three northernmost populations were found to contain the majority of the species' remaining genetic diversity and thus, should be a focus for future conservation management.
