The influence of exogenous triiodothyronine (0.25 ng/mL) on the activity glycosidases (maltase, amylolytic activity) and their sensitivity to Cu, Zn and Pb ions (25 mg/L) has been studied in the whole organism of juvenile blue bream Ballerus ballerus (L.). We have found that the treatment of fish with triiodothyronine resulted in an increase in amylolytic activity. Maltase activity is not affected. In addition, the exposure to exogenous triiodothyronine results in a decrease in the sensitivity of glycosidase to the in vitro action of the ions of biogenic metals Cu and Zn and in an increase in the sensitivity of the maltase to the action of Cu, Zn, and Pb ions.  相似文献

The separate and joint effect of Cu2+ and Zn2+ of 0.1–25 mg l−1 concentrations on the intestine carbohydrase activity were studied in vitro in 11 freshwater bone fishes. The total amylolytic activity of the fish intestinal mucosa decreased simultaneously with an increase in the Cu and Zn concentration. The maximal inhibitory effect varied from 15 to 46% and depended on the fish species. Copper had a more toxic effect on the amylolytic enzymes than zinc in planktivorous and bottom-feeding fish; the opposite pattern was observed in typical and facultative ichthyophagous species. The sucrase activity also decreased in the presence of Cu2+ and Zn2+, with minor exceptions; at the same time it increased by 18–50% in bream, ide, and burbot in the presence of Zn. The joint effect of Cu2+ and Zn2+ (1: 1) on the intestinal carbohydrase hydrolysis activity was significantly lower than when they were separate.  相似文献

The effects of in vitro exposure to the herbicide Roundup at concentrations of 0.1–50 μg/L on the activity of maltase and sucrase and the total amylolytic activity in the organism of invertebrates and fish fry have been investigated. Glycosidases in invertebrates are less sensitive to the herbicide than those in juvenile fish. Roundup has a greater inhibitory effect on glycosidase activity in the tissues of actual prey (roach recovered from pike stomach) than in potential prey (roach captured in the pond). The magnitude and direction of the effects depend on the animal species and the concentration of the toxicant.  相似文献

It was revealed that intestine and whole-body amylolytic activity (AA) in juvenile fish serving as potential feeding items for piscivores significantly decreases in the presence of Cu and Zn ions within a wide range (0.1–25 mg/l) of concentrations in vitro. In most of the studied fish species, Cu and Zn decrease the activities of carbohydrases in the intestine mucosa stronger than in the whole-body. On the other hand, in piscivore-facultative benthivore perch, which have the least AA, the inhibiting effect of Cu and Zn ions is 1.5 to 2 times higher in the whole body than in the intestine. The study data suggest that Cu and Zn ions at concentrations found in organisms that serve as fish food may not only reduce the rate of initial stages of carbohydrates hydrolysis in the juvenile fish intestine, but also considerably decrease the potential contribution of the food objects to the digestion processes in typical and facultative piscivores.  相似文献

Heavy metals are known to have adverse effects on soil ecosystems, while soil enzyme activities are sensitive to soil pollution. This study investigated the combined effects of Cu, Zn and Pb on the activities of invertase (IN), urease (U) and alkaline phosphatase (ALP) in soil obtained from the vicinity of a wellhead protection area via an orthogonal array (OA) design method. The experimental results showed the following: (1) Cu showed higher inhibition on the activities of all three enzymes than Zn and Pb when three metals were all present in the soil sample. IN activity, U activity and ALP activity decreased as the levels of Cu increased, and ranged from 15.9% to 55.7%, 3.57% to 78.6%, and 3.23% to 75.3%, respectively. Their lowest values were found in samples at 35 days with 400 mg/kg Cu. (2) Zn and Pb had different influences on the activities of the three enzymes. The lowest IN activity (the highest reduction 58.0%) and U activity (76.8%) were observed when Zn was at the concentration of 100 mg/kg after 35 days, whereas the highest inhibitory function of Zn on ALP activity (75.3%) was at 300 mg/kg after 7 days. When the concentration of Pb increased from 35 to 350 mg/kg, the activities of IN (62.5%) and U (69.6%) were most inhibited at 35 days and 14 days, respectively. However, when Pb was at the concentration of 500 mg/kg after 14 days, ALP activity (72.0%) showed the lowest value. (3) With respect to the three hydrolases in this study, ALP was the most sensitive to the two-variable interactive effects of Cu, Zn and Pb, especially Cu?×?Pb. It is concluded that the soil ALP activity may be a sensitive tool for assessing additive toxic effect on soil biochemical parameters. To provide more information about the potential ecological risk of chemicals on soil ecosystems, much more should be done to clearly determine the mechanisms of the combined effects of heavy metals in soil.  相似文献

研究了陕西省铜川煤矿矿区的重金属污染状况以及不同程度的重金属污染对土壤微生物代谢、微生物群落功能以及土壤酶活性的影响.结果表明: 铜川矿区土壤中重金属Cu、Zn、Cd、Pb全量及有效量均显著高于非矿区土壤,其中Cd污染最为严重.采用Biolog方法结合主成分分析和聚类分析发现,随着污染程度的增加,不同土壤微生物群落间的代谢特征发生显著变化,而且这种变化主要体现在糖类和氨基酸类碳源的利用差异.在轻度、中度污染情况下,土壤微生物群落对碳源的利用表现出激活效应;而在重度污染的情况下,土壤微生物群落对碳源的利用表现出抑制效应.随着污染程度的增加,脲酶、蛋白酶、碱性磷酸酶和过氧化氢酶的活性均呈现降低的趋势,矿区土壤脲酶、蛋白酶、碱性磷酸酶和过氧化氢酶活性分别是非矿区土壤中相应酶活性的50.5%~65.1%、19.1%~57.1%、87.2%~97.5%、77.3%~86.0%;蔗糖酶和纤维素酶在中等污染程度以下的土壤中表现为激活效应,而在重度污染的土壤中表现为抑制效应.  相似文献

The development of small intestinal enzymes (lactase, acid- and hetero beta-galactosidases, cellobiase, maltase, trehalase, and sucrase) was studied from 18 days after conception until birth in 24 rabbit fetuses, and during the postnatal period in 15 newborn, juvenile, and adult rabbits. Lactase, acid- and hetero beta-galactosidases, cellobiase, and trehalase activities increased significantly during the fetal stage, while changes in sucrase and maltase activities were not substantial. In the postnatal period, lactase and cellobiase activities decreased significantly whereas maltase, sucrase, and trehalase activities increased significantly to reach adult values by 30 days of age. The acid- and hetero beta-galactosidases remained unchanged.  相似文献

1. Intestinal disaccharidases were studied in nectarivorous (Leptonycteris curasoae and Glossophaga soricina), frugivorous (Artibeus jamaicensis and Sturnira lilium), and insectivorous (Pteronotus personatus) adult bats. 2. Adult bats lacked measurable lactase activity. With the exception of trehalase activity, which was present only in P. personatus, nectar- and fruit-eating bats exhibited higher disaccharidase activities standardized by intestinal nominal area than insect-eating P. personatus. 3. Maltase and sucrase activities were significantly linearly correlated. 4. Apparent affinity of sucrase varied almost 5-fold among species. This variation may reflect unstirred layer effects resulting from sucrase being a membrane bound enzyme rather than differences in the "true" affinity of sucrase in solution. 5. Passerine birds showed higher maltase activity per unit of sucrase activity than bats and hummingbirds. Maximal sucrase and maltase activities standardized per intestinal nominal area are 1.5-2 times higher in hummingbirds than in nectar-feeding bats.  相似文献

The chronic effects that polychlorinated biphenyls (PCBs) have on the activities of proteinases and carbohydrates in intestinal mucosa and chyme in juvenile roach Rutilus rutilus (L.) have been studied for the first time. Upon consuming food with PCB 50.8 ng/g wet weight for 218 days, the exposition of fingerlings in aquariums with dirt bottoms (contents PCB 425.6 ng/g dry weight) reduces the proteolytic activity of mucosa to a greater degree than amylolytic activity; the activity of sucrase changes in different directions. In 2-year-old fish receiving PCB only with food (50.8 ng/g wet weight bottom), proteolytic and amylolytic activities of the mucosa are reduced 18 and 35% in comparison with the control (52 days); the sucrose activity is reduced 13% (52 days) and 22% (169 days). In most cases the relation of amylolytic activity to proteolytic activity in fish of the experimental groups is less than in the control. The enzyme activities in chyme change in different directions, depending on the terms and conditions of the experiment.  相似文献

1. The maltase, isomaltase and invertase (sucrase) activities of solubilized mucosal preparations from human jejunum and ileum were studied with column chromatography on anion-exchange (diethylaminoethyl- and triethylaminoethyl-)cellulose and Sephadex G-200 gel. 2. On ion-exchange cellulose columns both kinds of enzyme preparations yielded two major disaccharidase peaks. The first peak contained maltase Ia (=isomaltase) and maltase Ib (=invertase). The second peak contained maltase II and maltase III. 3. On Sephadex G-200 gel columns jejunal preparations yielded the corresponding peaks as on ion-exchange columns, but the peaks appeared in the reverse order in the effluent. The ileal preparation studied yielded a single peak on gel columns, containing all the activities studied and eluted with the `void volume'. 4. Precipitation with ethanol did not affect the behaviour of the enzymes during ion-exchange chromatography. When gel filtration was performed after ethanol precipitation of the enzymes, however, two peaks were obtained also with the ileal preparation, and subfractionation of the invertase was obtained with both kinds of preparations. 5. The second peak from ion-exchange chromatograms, containing maltase II and maltase III, on concentration was found to have very weak isomaltase activity, probably exerted by these enzymes as such. This activity accounts for only about 1% of the total isomaltase activity of the mucosa. 6. The results support the concept of the specificity of the human small-intestinal disaccharidases previously described after heat-inactivation experiments. The subfractionation of the invertase that under certain conditions is seen on Sephadex G-200 columns appears most likely to be an artifact. Consequently the nomenclature for the human maltose-, isomaltose- and sucrose-splitting enzymes proposed by another research group after gel-filtration chromatography studies should be abandoned. It seems more logical to keep the nomenclature based on heat inactivation [maltase Ia (=isomaltase), maltase Ib (=invertase or sucrase), maltase II and maltase III] until increased knowledge about the specificity and structure of these enzymes makes possible a more rational nomenclature.  相似文献

The review summarizes for the first time the information on effects of Zn and Cu on various links of the fish exotrophy process. It has been shown that essential metals, like the nonessential ones, can produce negative effects on various aspects of the feeding behavior, on sensor systems providing feeding behavior, and on the digestive enzymes realizing digestion in fish of different ecological groups. A particular attention is paid to their different effects on proteinases functioning in stomach and intestine of various fish species as well as on hydrolase activities in their potential food objects (fish and invertebrate animals). Several mechanisms are considered which allow decreasing the negative action of Zn and Cu on the process of fish exotrophy.  相似文献

The protein content and the activities of alkaline phosphatase, maltase, and sucrase were measured at 0800, 1000, 1200, 1400, and 1600 in saline extracts of the proximal small bowels of germfree and of ex-germfree mice colonized with an indigenous microbiota. In extracts prepared from germfree mice, the total activities of all of the enzymes were relatively constant throughout the sampling period. Likewise, the total activity of alkaline phosphatase in extracts prepared from associated mice varied little as a function of time. By contrast, the total activities of maltase and sucrase in the extracts from these latter animals varied significantly from sample to sample. The total activity levels in extracts from germfree mice were approximately twofold greater than the levels in extracts from associated mice. The specific activities of alkaline phosphatase and sucrase did not vary from sample to sample in extracts prepared from either type of mouse. In contrast, the specific activity of maltase in extracts prepared from both germfree and associated mice differed significantly from sample to sample. The specific activities of all three enzymes were greater in extracts from germfree animals than in those from associated animals. The protein content of extracts prepared from germfree mice also was greater than that of extracts prepared from associated animals at every sampling time. The amount of protein extractable from the mucosa of the small bowels of the former animals varied significantly at different sampling times during the day, whereas the amount of protein extractable from the tracts of associated animals remained relatively constant throughout the day. The indigenous microbiota apparently stabilizes in some way the amount of protein extractable from the mucosa of the mouse small bowel.  相似文献

Hypoxia in the neonate is known to alter the activity of hepatic and pancreatic enzymes involved in lipid and carbohydrate metabolism. The purpose of this study was to evaluate the effect of neonatal hypoxia on the activity of intestinal enzymes, and to determine whether the administration of glucocorticoids to neonates can mimic the effects of hypoxia. Hypoxia in neonatal rats (0-7 days) increased protein content, and lactase and maltase activity in the duodenal and the jejunal segments of the small intestine compared with normoxic controls. Hypoxia in juvenile rats (28-35 days) did not change these enzymes. Two weeks after returning hypoxic (0-7 days) pups to normoxia, their body weight remained lower than the age-matched controls. In the group recovering from hypoxia, sucrase, maltase, and leucine aminopeptidase activities were lower in the duodenal and the jejunal segment. Compared with controls, LDH activity was lower only in the jejunal intestine in the group recovering from hypoxia. All enzyme activities returned to control levels 3 weeks after recovery. Neonatal rats treated with dexamethasone had a decrease in body weight, but increases in sucrase and maltase activity in both the duodenal and the jejunal segment. Hypoxia in newborn rats caused a delayed maturation of small intestinal enzymes. Increases in serum glucocorticoids after hypoxic exposure probably do not play a major role in the delayed maturation of the disaccharidase activity in the small intestine.  相似文献

1. The maltase, sucrase, isomaltase and palatinase activities of the chick small intestine are localized in particles that sediment when centrifuged at 100000g for 90min. 2. Solubilization of the particle-bound disaccharidases without loss of activity was achieved by digestion with papain. Trypsin was less effective and caused a preferential solubilization of the sucrase, isomaltase and palatinase activities. 3. On Sephadex G-200 columns, the solubilized preparations yielded two disaccharidase peaks. The first peak was eluted close to the void volume of the column and contained all the sucrase, isomaltase and palatinase activities and some of the maltase activity. The remainder of the maltase activity was eluted beyond the total volume of the column. 4. Precipitation with ethanol did not affect the behaviour of the disaccharidases of gel filtration. 5. The maltase activity of the second peak on rechromatography in a buffer containing 0.01m-maltose was eluted close to the void volume. 6. Similar pH optima but different K(m) values were obtained for the maltase activities of the two peaks. 7. Heat-inactivation studies showed that the first peak contained two disaccharidase enzymes; one hydrolysed sucrose and maltose and the other hydrolysed isomaltose, palatinose and maltose. The second peak contained three disaccharidase enzymes all specific for the hydrolysis of maltose. 8. It is proposed that the intestinal disaccharidases of the chick exist in the form of two complexes: a sucrase-isomaltase complex and a maltase complex.  相似文献

The activities of the digestive enzymes, maltase [EC 3.2.1.20], sucrase [EC 3.2.1.26], trehalase [EC 3.2.1.28], Leucine aminopeptidase [EC 3.4.11.1], and alkaline phosphatase [EC 3.1.3.1] were measured in various regions of the small intestine of rats. The activities of all these enzymes were much higher in the jejunum than in the ileum, and in the distal regions of the ileum no sucrase, trehalase or alkaline phosphatase activity was detected. In the jejunum, the activities of all the enzymes tested exhibited clear circadian variations with the highest activity at 0000-0400 h and the lowest at 1200 h when the rats were fed ad libitum. In the ileum, maltase and sucrase also exhibited circadian variations, but the amplitude of the rhythm was smaller than that in the jejenum. Trehalase and alkaline phosphatase did not show any circadian variation in the ileum. Leucine aminopeptidase showed a circadian variation in the ileum with the same amplitude as in the jejunum. The phase of the circadian variations shifted about half a day when the rats were fed in the daytime, but the amplitude of the rhythm did not change.  相似文献

The effects of phenol and its derivatives (4-chlorophenol, 4-nirophenol and 2,4-dinitrophenol) on the activity of peptidases in five species of fish differing in feeding modes have been studied. The power of the effects depends on the fish species and localization of the enzyme (mucosa or chyme). As a rule, in the in vitro conditions the phenols in concentrations of 0.06?0.5 μM/L considerably decreases activities of the intestinal peptidases in bream; silver bream; and, especially, in pike. In some cases, phenol and its derivatives induce an inconsiderable increase in peptidase activities. In zander and perch, in fact, no changes in the enzymatic activity under impact of phenols are observed. It is suggested that the effect of phenol and its derivatives depends to a high extent on the species specificity of peptidases: in the fish of fam. Percidae, the enzymes are relatively tolerant; in fam. Cyprinidae and Esocidae, they are sensitive to the studied toxicants.  相似文献

The effect of dexamethasone on the evolution pattern of brush border enzymes was examined in the rat jejunum cultured in vitro at different postnatal stages (4 to 21 days). Enzymic activities were analyzed in purified brush border membranes isolated from noncultured intestine and from explants cultured for 24 and 48 hr. The data obtained from this study indicated that dexamethasone exhibits two types of effects on the cultured intestinal tissue: (1) a nonspecific but protective effect against the drastic drop of all enzyme activities as well as against a loss of villus cells observed in control cultures, and (2) a direct and specific effect on precocious induction of sucrase and on stimulation of maltase activity. The SDS-polyacrylamide gel patterns of brush border membrane proteins showed that in the 6-day-old intestine, appearance of sucrase as well as stimulation of maltase activities elicited by dexamethasone were accompanied by a simultaneous appearance or enhancement of the corresponding protein bands. Furthermore, the radioactivity peaks on gels due to the incorporation of 14C-valine and of 14C-fucose indicated that dexamethasone induces the synthesis of new proteins or at least the glycosylation of preexisting proteins which may lead to the formation of active maltase and sucrase molecules.  相似文献

The activity of polyhydric alcohol dehydrogenases in Mycobacterium sp. 279 was studied under limitation of zinc in the growth medium. It was found that the activity of ribitol and sorbitol dehydrogenases were markedly lowered and that of D-arabinitol dehydrogenase remained unchanged in the Zn2+-deficient cells. Other ions tested i.e., Co2+, Cu2+, Ni2+ and Mn2+ failed to substitute Zn2+ ions in their effect on the enzyme activities. The Zn2+-responsive enzymes were sensitive to the chelating agents (1,10-phenanthroline, 2,2'-dipyridyl), whereas D-arabinitol dehydrogenase was insensitive. The results indicate possible existence of a zinc component in the ribitol and sorbitol dehydrogenases from Mycobacterium sp. 279.  相似文献

The study on remote consequences of hypomagnetic conditions during roach Rutilus rutilus embryogenesis (at the stages of embryo and prelarva) revealed multidirectional changes in body length/weight, activities of glycosidases (maltase, sucrase, and amylolytic activity), and kinetic characteristics of carbohydrates hydrolysis in the intestine of the yearlings. The exposure of embryos to hypomagnetic conditions leads to the increase in body length and weight in the yearlings; the exposure of prelarvae leads to the decrease in these parameters. The impact of hypomagnetic conditions at the stage of prelarva results in most pronounced changes in the physiological and biochemical parameters in the yearlings. Decline in the Michaelis constant values, reflecting affinity of an enzyme to substrate, may be considered as an adaptive response of the fish digestive system to the lack of magnetic field during embryogenesis.  相似文献
