The systematics of the Prasiolales was investigated by phylogenetic inference based on analyses of the rbcL and 18S rRNA genes for representatives of all four genera currently attributed to this order (Prasiococcus, Prasiola, Prasiolopsis, Rosenvingiella), including all type species. The rbcL gene had higher sequence divergence than the 18S rRNA gene and was more useful for phylogenetic inference at the ranks of genus and species. In the rbcL gene phylogeny, three main clades were observed, corresponding to Prasiola, Prasiolopsis, and Rosenvingiella. Prasiococcus was nested among species of Prasiola occurring in subaerial and supralittoral habitats. Trichophilus welckeri Weber Bosse, a subaerial alga occurring in the fur of sloths in Amazonia, was closely related to Prasiolopsis ramosa Vischer. The species of Prasiola were grouped into three well‐supported clades comprising (i) marine species, (ii) freshwater and terrestrial species with linear blades, and (iii) terrestrial species with rounded or fan‐shaped blades. Sequence divergence was unexpectedly low in the marine group, which included species with different morphologies. For the 18S rRNA gene, the phylogenetic analyses produced several clades observed for the rbcL gene sequence analysis, but, due to very little sequence variation, it showed considerably lower resolution for inference at the species and genus levels. Due to the low support of some internal branches, the results of the analyses did not allow an unambiguous clarification of the origin and the early evolution of the Prasiolales.  相似文献

Trebouxiophytes of the genus Prasiola are well known in Antarctica, where they are among the most important primary producers. Although many aspects of their biology have been thoroughly investigated, the scarcity of molecular data has so far prevented an accurate assessment of their taxonomy and phylogenetic position. Using sequences of the chloroplast genes rbcL and psaB, we demonstrate the existence of three cryptic species that were previously confused under Prasiola crispa (Lightfoot) Kützing. Genuine P. crispa occurs in Antarctica; its presence was confirmed by comparison with the rbcL sequence of the type specimen (from the Isle of Skye, Scotland). Prasiola antarctica Kützing is resurrected as an independent species to designate algae with gross morphology identical to P. crispa but robustly placed in a separate lineage. The third species is represented by specimens identified as P. calophylla (Carmichael ex Greville) Kützing in previous studies, but clearly separated from European P. calophylla (type locality: Argyll, Scotland); this alga is described as P. glacialis sp. nov. The molecular data demonstrated the presence of P. crispa in Maritime and Continental Antarctica. P. antarctica was recorded from the Antarctic Peninsula and Shetland Islands, and P. glacialis from the Southern Ocean islands and coast. Such unexpected cryptic diversity highlights the need for a taxonomic reassessment of many published Antarctic records of P. crispa. The results also indicate that marine species of Prasiola form a well‐supported monophyletic group, whereas the phylogenetic diversity of freshwater species is higher than previously suspected (at least three separate lineages within the genus include species living in this type of environments).  相似文献

Species of Prasiolales (Trebouxiophyceae, Chlorophyta) are among the most common terrestrial and freshwater algae in polar regions. Comprehensive molecular studies of this group are available for Antarctica, but not yet for Arctic regions. We examined the diversity of the Prasiolales in the Svalbard Archipelago combining morphological observations of field-collected material, culture studies, molecular data (plastid rbcL and tufA sequences) and literature records. We confirmed the widespread occurrence of Prasiola crispa and P. fluviatilis, species recorded from Spitsbergen since the 19th century. Molecular phylogenetic analyses led to the discovery of two new genera of Prasiolales. Prasionema payeri is morphologically identical to filamentous stages of P. crispa, but represents an early-diverging lineage in the order. Prasionella wendyae is a colonial alga reproducing by aplanospores; its phylogenetic position is among the basal lineages of the order, but it could not be reliably reconstructed due to weak statistical support. The inclusion of P. wendyae in the prasiolalean phylogeny determined the paraphyly of Rosenvingiella, requiring the establishment of the new genus Rosenvingiellopsis for R. constricta. A poorly known species described from Spitsbergen, Ulothrix discifera, is transferred here to Rosenvingiella. Whereas some species of Prasiolales have bipolar distribution (P. crispa), others appear to be restricted to one or other of the poles. Our results suggest that polar regions are still a major repository of unknown algal diversity and highlight the importance of continued field surveys and the use of molecular data.  相似文献

Molecular phylogenetic tools are often useful in distinguishing cryptic species with similar morphologies, but they can also be helpful in identifying morphotypes of a species, which displays completely different shapes. We performed molecular-phylogenetic analysis of supra-tidal green algae in the Kamchatka peninsula that belong to the order Prasiolales (Trebouxiophyceae, Chlorophyta). Based on rbcL sequences results, two new species were recorded for the first time in Kamchatka. Approximately 1.4% of the field-collected Rosenvingiella constricta had a unique uniseriate hood-like blade shape, although their rbcL sequences were 100% identical with typical multiseriate filamentous plants. Kamchatka’s population of R. constricta showed 99.4–99.6% identity in rbcL sequences with the populations from Canada and New Zealand. Another similar-looking Rosenvingiella species collected from the same locality had 93.5% identity of the rbcL gene sequence with R. constricta. Morphological and geographical analyses also suggested that this species might be a new species of the genus Rosenvingiella. Prasiola delicata was recorded for the first time in Kamchatka. The Kamchatka population of P. delicata showed 100% identity in rbcL gene sequence with the population from Vancouver, but differed from the Canadian population morphologically.  相似文献

Recent collections from marine and freshwater locations have enabled the investigation of diversity of Prasiola in Japan. Sequence data from the rbc L and tuf A markers revealed the presence of three marine species and one freshwater species. Prasiola delicata was confirmed to occur on Daikokujima, Prasiola calophylla was found for the first time in Japan from Hokkaido, and a species within the P. meridionalis/linearis/stipitata complex was found on both Hokkaido and Daikokujima. Collections from a range of populations of freshwater Prasiola, identified here as P. japonica, were found to be conspecific and identical in rbc L and tuf A sequences to freshwater collections from Nepal, Korea, and China.  相似文献

A molecular phylogenetic analysis of the little‐studied filamentous brown alga Discosporangium mesarthrocarpum (Meneghini) Hauck using rbcL and partial 18S rDNA sequences revealed that the species forms a monophyletic clade with Choristocarpus tenellus (Kütz.) Zanardini that is sister to all other brown algae. Although D. mesarthrocarpum has unique disk‐shaped plurilocular reproductive organs, D. mesarthrocarpum and C. tenellus share the following basic morphological features, which are considered to be plesiomorphic characters in the brown algae: (1) apical (and diffuse) growth; (2) uniseriate, subdichotomously branched filaments; (3) multiple chloroplasts per cell without pyrenoids; and (4) lack of heterotrichy and of phaeophycean hairs. The rbcL DNA sequence of an Australian D. mesarthrocarpum specimen showed considerable deviation from Mediterranean and Macaronesian specimens. Therefore, the presence of a second species in the genus is suggested; however, the taxonomic treatment of this putative species is not pursued in the present report. Regarding the higher‐ranking systematic position of D. mesarthrocarpum, reinstatement of Discosporangiaceae and Discosporangiales is proposed, and the inclusion of Choristocarpaceae in the order is also suggested. Under short‐day and long‐day culture conditions at 15°C–25°C, Mediterranean D. mesarthrocarpum exhibited a direct type of life history, with a succession of uniseriate filamentous thalli bearing characteristic disk‐shaped plurilocular zoidangia, but thalli did not survive at 10°C and below.  相似文献

A new species in the genus Prasionema (Prasiolales, Trebouxiophyceae) is described from Campbell Island, in the New Zealand subantarctic region, the first record of this genus in the southern hemisphere. Prasionema heeschiae sp. nov. is filamentous, uni- to predominantly biseriate, with disc-shaped cells, and an axial plastid with a central pyrenoid. It is anchored by an enlarged pigmented basal cell. There is evidence of reproduction by both spores and fragmentation. This species was found growing on a timber wharf above the high tide level. The only other species in the genus, P. payeri Heesch, M.Pazoutová & Rindi, was described from Spitzbergen growing on soil in a high nutrient environment. The bipolar distribution of Prasionema is discussed. Based on phylogenetic analyses that included sequence data from holotype material of southern filamentous Prasiolales, we reduce Rosenvingiella australis to synonymy with R. tasmanica.  相似文献

In their natural habitats, some representatives of Prasiolales are regularly, or often, washed by waves; others are sprayed with sea-water only occasionally, or not at all. In correspondance to this ecological grouping, species require marine or freshwater media respectively for optimal growth in cultures. The halophilic group consists ofPrasiola stipitata andRosenvingiella constricta. Though not analysed in detail, morphological characters and behaviour in cultures suggest that several different species are united under the namePrasiola stipitata. Rosenvingiella constricta, new for Helgoland and thus far reported only twice in Europe, is a dioecious haplont; both meiosis and genotypic sex-determination occur in the germinating zygote. The parenchymatous “Gayella-stage” ofRosenvingiella polyrhiza proved to be male and female gametangia, producing spermatozoids and non-motile eggs respectively. Fertile threads were extremely rare in natural material and — contrary toRosenvingiella constricta — did not occur in cultures, so the life cycle could not be elucidated.Prasiola calophylla is frequently associated withRosenvingiella polyrhiza. The life cycle includes heteromorphic asexual generations, the spores of the leafy thallus giving rise to unicellular aplanosporangia.Prasiola furfuracea reproduces only by spores. Samples ofRosenvingiella polyrhiza frequently containPrasiolopsis in addition to a number of other Chlorophyceae. Consequently, it is easy to appreciate that previous investigators came to erroneous conclusions about life histories and biological relations in studies based only on field material.  相似文献

Twenty-seven populations of Tuomeya, including the types of Tuomeya fluviatilis Harvey and Baileya americana Kützing, were analyzed from the entire known range of the genus: northern Newfoundland to northern Florida and east of the Mississippi River. Key morphological features were examined in detail since it has been recently proposed that Tuomeya should be reduced to a section of the genus Batrachospermum. Our observations confirmed the presence of several characteristics unique to Tuomeya: gametophyte development from a basal mass of undifferentiated cells rather than a chantransia stage, pseudoparenchymatous growth, and carpogonia with obliquely to perpendicularly attached trichogynes. Based on these findings, we conclude that the genus Tuomeya should be retained. Using multivariate morphometrics, two groupings were found that differed significantly in plant length (X = 14.1 and 24.1 mm, respectively). However, since there was no other morphometric, environmental, or geographic basis for separation of the groupings, only one species is recognized, T. americana (Kützing) Papenfuss. Populations tend to occur in large streams (>2.8 m wide) with low ion content (≤ 100 μS · cm?1).  相似文献

Green algae belonging to the order Prasiolales have been reported frequently in terrestrial, freshwater, and marine habitats, generally in situations in which they are not submerged continuously and often in association with deposits of avian feces or other rich sources of nitrogenous compounds. Terrestrial habitats in Galway City on the west coast of Ireland have large populations of four such algae: Rosenvingiella polyrhiza (Rosenvinge) P. C. Silva, Prasiola calophylla (Carmichael ex Greville) Kützing, P. crispa (Lightfoot) Kützing, and P. stipitata Suhr ex Jessen, of which only the last is in any way fully associated with a tidal regime. The other species are found in an extraordinary range of nonmarine terrestrial habitats in the center of Galway, a city that still retains its narrow, medieval street plan. Width of the streets, proportion of residences, substratum type, and aspect are shown to be the main factors affecting the distribution of these algae, although a combination of consistent dampness and nitrogenous-compound availability from animal wastes is likely to be the primary determining factor. Morphological and phenological data are presented for each entity, and it is concluded from field observations that R. polyrhiza should continue to be recognized as a separate entity, even though a close relationship with Schizogonium murale Kützing is probable.  相似文献

Generic concepts in the economically important agarophyte red algal family Gracilariaceae were evaluated based on maximum parsimony, Bayesian likelihood, and minimum evolution analyses of the chloroplast‐encoded rbc L gene from 67 specimens worldwide. The results confirm the monophyly of the family and identify three large clades, one of which corresponds to the ancestral antiboreal genera Curdiea and Melanthalia, one to Gracilariopsis, and one to Gracilaria sensu lato, which contains nine distinct independent evolutionary lineages, including Hydropuntia. The species currently attributed to Hydropuntia comprise a single well‐supported clade composed of two distinct lineages. The two most basal clades within Gracilaria sensu lato deserve generic rank: a new genus centered around G. chilensis Bird, McLachlan et Oliveira and G. aff. tenuistipitata Chang et Xia and a resurrected Hydropuntia encompassing primarily Indo‐Pacific (G. urvillei [Montagne] Abbott, G. edulis [S. Gmelin] P. Silva, G. eucheumatoides Harvey, G. preissiana [Sonder] Womersley, and G. rangiferina [Kützing] Piccone) and western Atlantic species (G. cornea J. Agardh, G. crassissima P. et H. Crouan in Mazé et Schramm, G. usneoides [C. Agardh] J. Agardh, G. caudata J. Agardh, and G. secunda P. et H. Crouan in Mazé et Schramm). Cystocarpic features within the Gracilaria sensu lato clades appear to be more phylogenetically informative than male characters. The textorii‐type spermatangial configuration is represented in two distinct clusters of Gracilaria. The rbc L genetic divergence among the Gracilariaceae genera ranged between 8.46% and 16.41%, providing at least 2.5 times more genetic variation than does the 18S nuclear rDNA. rbc L also resolves intrageneric relationships, especially within Gracilaria sensu lato. The current number of gracilariacean species is underestimated in the western Atlantic because of convergence in habit and apparent homoplasy in vegetative and reproductive anatomy.  相似文献

The freshwater green algal family Hydrodictyaceae (Sphaeropleales, Chlorophyta) has traditionally consisted of four coenobial genera, Pediastrum Meyen 1829, Hydrodictyon Roth 1797, Sorastrum Kützing 1845, and Euastropsis Lagerheim1894. Two recent molecular phylogenetic studies demonstrated the need for reevaluation of the generic and species boundaries in this morphology‐rich family. This study expands the previous work to include phylogenetic analyses of 103 ingroup isolates representing North America, Europe, and Australia, with an emphasis on the common and geographically widespread species Pediastrum duplex. Nucleotide sequence data were collected from the nuclear LSU (26S rDNA) and the chloroplast RUBISCO LSU (rbcL) genes, totaling >3,000 aligned characters. The 26S and rbcL data sets were analyzed using maximum‐likelihood (ML) and Bayesian phylogenetic methods. In addition, SEM was used to examine the wall morphology of a majority of the isolates. The results supported previous indications that the P. duplex Meyen 1829 morphotype is nonmonophyletic and resolved some previously ambiguous relationships recovered in earlier phylogenetic estimations using fewer isolates. These new data allowed testing of the recent taxonomic revisions of the family that split Pediastrum into five genera. Some of the previous revisions by Buchheim et al. (2005) were well supported (erection of Stauridium and Monactinus), while others were not (Pediastrum, Pseudopediastrum, Parapediastrum).  相似文献

The determinants of host specificity, which are poorly understood in red algal parasites, were studied in the red algal parasites Bostrychiocolax australis Zuccarello et West and Dawsoniocolax bostrychiae (Joly et Yamaguishi-Tomita) Joly et Yamaguishi-Tomita. Culture studies were performed to determine host range, sites of host resistance, and genetics of transmission of resistance. Both species parasitize Bostrychia radicans (Montagne) Montagne, whereas Bostrychiocolax australis also parasitizes Bostrychia moritziana (Sonder ex Kützing) J. Agardh and Stictosiphonia kelanensis (Grunow ex Post) R. J. King et Puttock. Isolates of B. radicans resistant to both parasites were found worldwide, often within the same population as susceptible isolates. On resistant Bostrychia species and isolates, specificity was manifested at three stages: 1) host penetration, in which the spore germ peg failed to penetrate the host cuticle/wall; 2) parasite–host cell fusion, in which the fusion cell died and the parasite died; and 3) growth, in which parasites grew but soon died; parasites rarely reproduced and infections did not continue in culture. Resistance to parasite infection was usually transmitted as a dominant trait and did not segregate as a single locus during meiosis. In certain crosses, transmission of resistance was non-mendelian.  相似文献

Bostrychia radicans(Montagne) Montagne is a pantropical/temperate red alga associated with mangroves and saltmarsh plants. Collections were made from a similar north-south geographic distribution along both the Pacific and Atlantic coasts of North America. Hybridization studies were performed with cultured isolates to assess the extent of interfertility and reproductive isolation along these two coastlines. All male and female gametophytes derived from single tetrasporophytes were intercompatible. Almost all isolates extending over 1500 km of coast line from northern Pacific Mexico are compatible, forming cystocarps that released viable carpospores. Even isolates which morphologically would be placed in two species [B. radicans and B. moritziana(Sender ex Kützing) J. Agardh], based on the presence or absence of monosiphonous branches, were capable of hybridizing. Crosses of isolates from the Atlantic USA showed a greater amount of incompatibility. Certain isolates were not compatible with any other isolates including isolates collected in close proximity (North Carolina isolates), while other isolates from the same locality were compatible (South Carolina). An isolate from South Carolina formed tetrasporophytes with isolates from Pacific Mexico but tetraspores were not viable. Certain incompatible crosses formed ‘pseudocystocarps’ but viable carposporophytes did not develop. Generalizations about reproductive isolation within a species must also consider differences between populations from different biogeographic regions that may reflect different paleoclimatological histories, founder effects and unique dispersal events.  相似文献

The complete rbcL gene was sequenced for 21 species and 32 strains of Vaucheria and for five other Xanthophyceae (Asterosiphon dichotomus (Kützing) Rieth, Botrydium becharianum Vischer, B. cystosum Vischer, B. stoloniferum Mitra, Tribonema intermixtum Pascher). The psbA‐rbcL spacer, upstream of the rbcL gene, and the RUBISCO spacer between the rbcL and rbcS genes were also completely sequenced for the Vaucheria strains and Asterosiphon. The psbA‐rbcL spacer was the most variable region that was sequenced, and only the 3′ end of the spacer could be aligned. Phylogenetic analyses (maximum parsimony, neighbor joining, and maximum likelihood) were conducted using the DNA sequence and the amino acid sequence for the rbcL gene, and a second analysis was conducted using a portion of the psbA‐rbcL spacer +rbcL gene + RUBISCO spacer. All analyses showed that Vaucheria species formed monophyletic clades that corresponded with morphologically based subgeneric sections, including the section Racemosae. Species producing a gametophore (= fruiting branch, bearing both an antheridium and oogonium) formed a monophyletic clade in all analyses. The nongametophore species sometimes formed a monophyletic clade but other times formed a basal grade. Pair‐wise comparisons of nucleotides and amino acids showed that for some species, numerous nucleotide changes resulted in relatively few amino acid changes. Consequently, phylogenetic analysis of the amino acids produced numerous trees, which in a strict consensus tree resulted in numerous polychotomies. An original strain of V. terrestris that was deposited in two culture collections over 25 years ago had identical sequences, suggesting no rapid change was occurring in the sequenced regions. Two strains of V. prona, isolated from Europe and North America, had identical sequences. Other species, for which two or more strains were examined, had different sequences. These results suggest that cryptic species complexes exist within Vaucheria because the rbcL gene is a conservative gene that is identical in other protists.  相似文献

The placement of Avrainvillea and Cladocephalus in the family Udoteaceae (order Bryopsidales) has been questioned on the basis of nuclear, plastid, and other ultrastructural characteristics unique to these genera. Bayesian analysis of the chloroplast‐encoded LSU RUBISCO (rbcL) gene showed that the Udoteaceae is paraphyletic. Cladocephalus luteofuscus (P. Crouan et H. Crouan) Børgesen, Avrainvillea nigricans f. floridana D. Littler et Littler, and A. mazei G. Murray et Boodle form a clade with the freshwater alga Dichotomosiphon tuberosus (A. Braun ex Kütz.) A. Ernst that is basal to a clade that includes other members of the Udoteaceae, the Halimedaceae, and the Caulerpaceae. The noncalcified species Boodleopsis pusilla (Collins) W. R. Taylor, A. B. Joly et Bernat. groups with species of the calcified Udoteacean genera Penicillus, Rhipocephalus, Udotea, and Halimeda.  相似文献

Ahnfeltia plicata (Hudson) Fries, the type species of Ahnfeltia Fries, is reported to be a widespread alga which is an important source of agar. However, after characterizing type material and representative populations of A. plicata, we found that several taxa usually considered to be synonyms of A. plicata instead represent separate species with differing geographic distributions. Ahnfeltia plicata sensu stricto is characterized by the development of external carposporophytes on female sori that are present only on mature axes. Pseudocarposporophytes bearing monosporangia and cortical monosporangial sori occur on both young and old axes. Characteristic vegetative features include long medullary cells and cortical growth rings. Ahnfeltia plicata s.s. is circumpolar in both hemispheres and is harvested for agar in the White Sea. Ahnfeltia setacea (Kútzing) Schmitz from the Falkland Islands, Gymnogongrus comosus Kützing from Chile and Gymnogongrus filiformis Kützing from southern Argentina appear to be synonymous with A. plicata. Ahnfeltia fastigiata (Postels et Ruprecht) Makienko, from the North Pacific, differs from A. plicata principally in the development of female sori and carposporophytes only near apices; medullary cells are shorter and cortical growth rings, cortical monosporangia and pseudocarposporophytes are not formed. It is dioecious and its life history involves the development from carpospores of a crustose Porphyrodiscus tetrasporophyte. We suggest that A. plicata var. tobuchiensis Kanno et Matsubara, the principal form of Ahnfeltia utilized for agar production in the North Pacific, represents an unattached ecad of A. fastigiata. Ahnfeltia elongata Montagne, known only from Chile and with carposporophytes restricted to main axes, is more closely related to A. plicata than to A. fastigiata. Thalli are large and regularly dichotomous, with long medullary cells, and do not form pseudocarposporophytes or monosporangia.  相似文献

对采自黑龙江省东南部兴凯湖湿地的100余号硅藻标本进行分类研究,共发现桥弯藻科、异极藻科(硅藻门)中国新记录植物5种2变种,分别为亚粗糙桥弯藻(Cymbella peraspera Krammer)、尖头弯肋藻[Cymbopleura cuspidata (Kützing) Krammer]、不显内丝藻高山变种(Encyonema obscurum var.alpina Krammer)、西里西亚内丝藻翼形变种(Encyonema silesiacum var.lata Krammer)、耳状异极藻(Gomphonema auritum Braun & Kützing)、棍棒异极藻(Gomphonema clava Reichardt)、极细异极藻[Gomphonema exilissimum (Grunow) Lange-Bertalot & Reichardt]。对其分类学特征进行了详细的描述,与同属中相似的种类进行了比较与讨论,为今后该区域的藻类研究提供了基础的资料。  相似文献

Species discrimination within the gigartinalean red algal genus Hypnea has been controversial. To help resolve the controversy and explore phylogeny within the genus, we determined rbcL sequences from 30 specimens of 23 species within the genus, cox1 from 22 specimens of 10 species, and psaA from 16 species. We describe H. caespitosa as a new species characterized by a relatively slender main axis; a pulvinate growth habit with entangled, anastomosing, and subulate uppermost branches; and unilaterally borne tetrasporangial sori. The new species occurs in the warm waters of Malaysia, the Philippines, and Singapore. The phylogenetic trees of rbcL, psaA, and cox1 sequences showed a distant relationship of H. caespitosa to H. pannosa J. Agardh from Baja California and the marked differentiation from other similar species. The rbcL + psaA tree supported monophyly of the genus with high bootstrap values and posterior probabilities. The analysis revealed three clades within the genus, corresponding to three sections, namely, Virgatae, Spinuligerae, and Pulvinatae first recognized by J. G. Agardh. Exceptions were H. japonica T. Tanaka in Pulvinatae and H. spinella (C. Agardh) Kütz. in Spinuligerae.  相似文献
