The proton magnetic relaxation time, T1, has been measured at 29 MHz in 0.1M KH2PO4 and 0.1M NaCI (both pH 6) aqueous solutions of human ferrihaemoglobin, the protein concentrations ranging from 0.5 to 5 mM per haem. The linear dependence on protein concentration of the enhancement in relaxation rates, Δ(1/T1), due to the presence of the paramagnetic iron in haemoglobin was confirmed at 34°C and at ~10°C. In the middle temperature range there is a thermally activated process, whose energy of activation depends on protein concentration. This dependence is different for the two salt solutions; Ea increases with cHb for 0.1M KH2PO4 and decreases for 0.1M NaCI. The model of water-proton exchange between the bulk solvent and the sixth coordination site of the haem iron was used to calculate the distance from the “liganded” water protons to the haem iron. This yields distances much larger than those determined by X-ray crystal structure analysis. A model is proposed which reconciliates both types of data. The low-temperature relaxation rates cannot be used in deriving quantitative stereochemical data for the haem pocket because of its special shape. Irrespective of the molecular model adopted, the experimental results show clearly that, both at low (~10°C) and higher (>34°C) temperatures, the interaction of paramagnetic haem iron with water protons is practically the same for the two aqueous solutions. The dynamic state of the haemoglobin molecule, as indicated by the middle-temperature range, is completely different in 0.1M KH2PO4 and 0.1M NaCl, pH 6.  相似文献

The longitudinal proton magnetic relaxation times T1 were measured for ferri (met)-and carbonmonoxy-bovine haemoglobin and equine myoglobin in 0.1 M KH2PO4 aqueous solutions near pH 6 at 5°C and 35°C from 1.5- to 60-MHz Larmor frequencies. It is concluded that the correlation time τC for the dipole–dipole interaction of electron and nuclear spins is in fact the electron (ferric) spin relaxation time τS being close to 1.5 × 10?10 sec for both metHb and metMb at 5°C. At 35°C the paramagnetic relaxation rates are not determined solely by the relaxation of protons exchanging from the haem pocket with bulk solvent. Hence, τC at 35°C cannot be calculated from the dispersion data obtained at this temperature. The relevance of this for the determination of interspin distances r is discussed.  相似文献

The proton and deuterium longitudinal relaxation rates were Studied at room temperature up to the highest protein concentrations in oxyhaemoglobin solutions of different H2O/D2O composition. The deuterium relaxation rates followed the experimentally well known single linear dependence on protein concentration, the slopes being little influenced by solvent (D2O/H2O) composition. The proton ralaxation rates show two different liner dependences on haemoglobin concentration. The entire concentration range is described by two straight lines with the threshold concentration about 11 mM (in haem), The ratio of the slopes is 1.6 (high-to-low Hb-conc.). Only in the higher concentration range two T1's were observed if the solvent contained more than half of D2O. The slow relaxation phase of protons has T1's similar to those measured in solutions with less than half of D2O. The relaxation of the other phase was ten times faster. The ratio of the proton populations in these two phases was equal to 2 (slow-to-fast) and independent of protein concentration. The fast relaxing protons are attributed to water molecules encaged within two or more haemoglobin molecules which associate for times long enough on the PMR time-scale.  相似文献

The influence of added salts on the dynamic viscoelastic properties are investigated for aqueous solutions of alginates that have various molecular weights and mannuronate/guluronate (M/G) ratios. The dynamic moduli of the systems increase with increasing concentration of the added salt in the low-frequency region. The effect is notable in the order of KCl < NaCl < MgCl2 ? CaCl2. The values of the dynamic moduli in the rubbery plateau are independent of the addition of the salts, irrespective of the M/G ratio of the alginate. These facts strongly suggest that the structure that is formed by the interaction between the alginates and the metal ions does not work as cross-linking points but as heterogeneous relaxation units having a relatively long relaxation time from a rheological viewpoint.  相似文献

Preparations of radioactive lysosomes were obtained from mouse kidney after injection of radioactive iodine-labeled bovine ribonuclease. Stability of these lysosomes in various media was estimated from measurements of proteolytic activity towards the ribonuclease, and of ribonuclease retention in particles. The lysosomes were stable at 37 °C in isotonic, sucrose-free solutions of KCl, NaCl and potassium acetate, and in mixtures of these with MgCl2, showing that these salts are relatively impermeant through the lysosomal membranes. The membranes were less permeable to Na+ than to K+. Both KCl and NaCl exerted their optimal protective effects over a broad concentration range above 0.125 M in 0.025 M acetate buffer. Mg2+ enhanced the protective effect of both K+ and Na+; the osmotic effect of 0.075 M NaCl-0.05 M MgCl2 was indistinguishable during the entire course of ribonuclease digestion from that of isotonic sucrose. Osmotic protection by KCl-MgCl2 was demonstrated over the pH range 5.5–7.0. A marked alteration in membrane properties occurs at lower temperatures in 0.11 M KCl-0.01 M MgCl2 such that, at 0 °C, K+ permeability is much higher than at 37 °C, as shown by a several-fold decrease in stability at the lower temperature.  相似文献

Vanadate was a potent inhibitor of the membrane-bound (Ca+Mg)-ATPase from rat brain, the concentration required for 50% inhibition under conditions optimal for enzymatic activity being 3 M. Vanadate inhibition increased with the MgCl2 concentration, half-maximal inhibition occurring at 2 mM MgCl2, near the MgCl2 concentration required for half-maximal activation of the ATPase activity. MnCl2 could substitute for MgCl2, and at concentrations of 1 mM (Ca+Mn)-ATPase activity was greater than (Ca+Mg)-ATPase activity, although sensitivity to vanadate was less. Vanadate inhibition increased also with the KCl concentration, half-maximal inhibition occurring at 8 mM, again near the concentration required for half-maximal activation of ATPase activity. By contrast, NaCl stimulated (Ca+Mg)-ATPase activity without potentiating vanadate inhibition. These effects of cations on ATPase activity and vanadate inhibition resemble properties of certain transport ATPases and thus suggest mechanistic and functional similarities.  相似文献

The fine structure of constitutive heterochromatin and euchromatin was compared in electron microscope whole-mount preparations of Tenebrio molitor (Insecta, Coleoptera) spermatocyte nuclei. Tenebrio molitor pachytene chromosomes display extended segments of centromeric heterochromatin and thus are especially suitable for this purpose. When nuclei were incubated in solutions containing different concentrations of NaCl or of MgCl2, two levels of chromatin fine structures were observed in the euchromatic segments: nucleosome fibers (0.1 mM–20 mM NaCl) and supranucleosomal fibers with 28 nm in diameter (40 mM–100 mM NaCl, 0.2 mM–1.0 mM MgCl2). The fine structure in the heterochromatic segments was the same as that in the euchromatic segments in all NaCl concentrations and in MgCl2 concentrations up to 0.4 mM. In higher MgCl2 concentrations the heterochromatin remained more compact than the euchromatin and consisted of 37-nm-thick fibers in 0.6 mM MgCl2 and of 65-nm-thick fibers in 1.0 mM MgCl2. After the 37-nm and the 65-nm fibers had been dispersed in Mg2+-free solutions they could be recondensed by incubation in 0.6 mM and 1.0 mM MgCl2, respectively. It is concluded that a Mg2+-sensitive component of the heterochromatin is responsible for the folding of the nucleosome chain to heterochromatin-specific supranucleosomal structures.  相似文献

Abstract

Equilibrium binding is believed to play an important role in directing the subsequent covalent attachment of many carcinogens to DNA. We have utilized UV spectroscopy to examine the non-covalent interactions of aflatoxin B1 and B2 with calf thymus DNA, poly(dAdT):poly(dAdT), and poly(dGdC):poly(dGdC), and have utilized NMR spectroscopy to examine non-covalent interactions of aflatoxin B2 with the oligodeoxynucleotide d(ATGCAT)2. UV-VIS binding isotherms suggest a greater binding affinity for calf thymus DNA and poly(dAdT):poly(dAdT) than for poly(dGdC):poly(dGdC). Scatchard analysis of aflatoxin B1 binding to calf thymus DNA in 0.1 M NaCl buffer indicates that binding of the carcinogen at levels of bound aflatoxin ? 1 carcinogen per 200 base pairs occurs with positive cooperativity. The cooperative binding effect is dependent on the ionic strength of the medium; when the NaCl concentration is reduced to 0.01 M, positive cooperativity is observed at carcinogen levels ? 1 carcinogen per 500 base pairs. The Scatchard data may be fit using a “two-site” binding model [L.S. Rosenberg, M J. Carvlin, and T.R. Krugh, Biochemistry 25, 1002–1008 (1986)]. This model assumes two independent sets of binding sites on the DNA lattice, one a high affinity site which binds the carcinogen with positive cooperativity, the second consisting of lower affinity binding sites to which non-specific binding occurs. NMR analysis of aflatoxin B2 binding to d(ATGCAT)2 indicates that the aflatoxin B2/oligodeoxynucleotide complex is in fast exchange on the NMR time scale. Upfield chemical shifts of 0.1–0.5 ppm are observed for the aflatoxin B2 4-OCH3, H5, and H6a protons. Much smaller chemical shift changes ? 0.06 ppm) are observed for the oligodeoxynucleotide protons. The greatest effect for the oligodeoxynucleotide protons is observed for the adenine H2 protons, located in the minor groove. Nonselective T1 experiments demonstrate a 15–25 % decrease in the relaxation time for the adenine H2 protons when aflatoxin B2 is added to the solution. This result suggests that aflatoxin B2 protons in the bound state may be in close proximity to these protons, providing a source of dipolar relaxation. Further experiments are in progress to probe the nature of the aflatoxin B1 and B2 complexes with polymeric DNA and oligodeoxynucleotides, and to establish the relationship between the non-covalent DNA-carcinogen complexes observed in these experiments, and covalent aflatoxin B1,-guanine N7 DNA adducts.  相似文献

Poly(A) containing ribonucleoprotein particles were prepared from rat liver nuclei and polyribosomes. The particles have sedimentation coefficients of 14 S and 9 S, respectively. In Cs2SO4 density gradients the particles banded at densities of 1.28–1.29 g cm-3. Petunia (Petunia hybrida Vilm. cv. ‘Snowstorm') plants were grown in saline solution (NaCl, MgCl2, and/or CaCl2) of 0, 1, 2, and 3 bars osmotic pressures. Pollen viability was tested by tetrazolium chloride staining and by germination (by the hanging drop method, using 15 % sucrose and 0.01 % boric acid as the nutrient medium, at 27 ± 1 C). Pollen viability decreased with increased salinity. Pollen from plants grown in single salt solutions of NaCl, MgCl2, and CaCl2 (each at 0, 1, 2, or 3 bars osmotic pressure) was germinated in base culture medium. Pollen viability decreased more with NaCl than with MgCl2 or CaCl2. In vitro studies of the effects of three salts, viz., NaCl, MgCl2, and CaCl2, on pollen germination and tube growth showed that NaCl inhibited germination and pollen tube growth more than did MgCl2 or CaCl2. MgCl2 was least injurious, and even promoted tube growth at 0.5 and 0.75 bars osmotic pressure. Adding low concentrations of MgCl2 reduced the toxic effect of NaCl and increased the percentage of germination. CaCl2 reduced the effect of NaCl less than did MgCl2. We conclude that specific ion effects were more important than osmotic pressure.  相似文献

Turbidity measurements were made of dilute aqueous dispersions of 1,2-dipalmitoyl-l-lecithin as the temperature was varied. In the range from 24 to 33° a decrease in turbidity is associated with the penetration of water between the layers of lipid in the crystalline structure. At the transition to the liquid crystalline form (39–41°) a sharp decrease in turbidity occurs. In solutions of 0.06 M LiCl, 1 mM CaCl2, MgCl2 or 0.05 M PO43 an increase in turbidity resulted at the transition temperature, whereas in 0.1 M NaCl, or quarternary ammonium salts the turbidity decreased. The presence of small amounts of dicetylphosphoric acid mixed with the lecithin decreased the turbidity, and at elevated pH levels there was no change at the transition temperature.  相似文献

Glycerol formation ofDunaliella cells in non-growing media was investigated.Dunaliella tertiolecta andD. bioculata grew well in a NaCl medium but not at all in a LiCl or a MgCl2 medium. When the cells originally suspended in a medium containing 0.5 M NaCl were transferred to media which contained one of 1 M NaCl, 1 M LiCl or 0.7 M MgCl2, the intracellular glycerol content increased.D. tertiolecta cultured in either a 1 M LiCl or a 0.7 M MgCl2 medium did not multiply, but maintained abilities to evolve O2 in the light and absorb O2 in thedark even after about a 5 day culture. From these results, it can be concluded that the halotolerance ofDunaliella to different kinds of salts is not directly related to osmoregulation by the glycerol formation.  相似文献

The (Na+ + K+)-dependent ATPase exhibits substrate sites with both high affinity (K m near 1 µM) and low affinity (K m near 0.1 mM) for ATP. To permit the study of nucleotide binding to the high-affinity substrate sites of a canine kidney enzyme preparation in the presence as well as absence of MgCl2, the nonhydrolyzable - imido analog of ATP, AMP-PNP, was used in experiments performed at 0–4°C by a centrifugation technique. By this method theK D for AMP-PNP was 4.2 µM in the absence of MgCl2. Adding 50 µM MgCl2, however, decreased theK D to 2.2 µM; by contrast, higher concentrations of MgCl2 increased theK D until, with 2 mM MgCl2, theK D was 6 µM. The half-maximal effect of MgCl2 on increasing theK D occurred at approximately 1 mM. This biphasic effect of MgCl2 is interpreted as Mg2+ in low concentrations favoring AMP-PNP binding through formation at the high-affinity substrate sites of a ternary enzyme-AMP-PNP-Mg complex; inhibition of nucleotide binding at higher MgCl2 concentrations would represent Mg2+ acting through the low-affinity substrate sites. NaCl in the absence of MgCl2 increased AMP-PNP binding, with a half-maximal effect near 0.3 mM; in the presence of MgCl2, however, NaCl increased theK D for AMP-PNP. KCl decreased AMP-PNP binding in the presence or absence of MgCl2, but the simultaneous presence of a molar excess of NaCl abolished (or masked) the effect of KCl. ADP and ATP acted as competitors to the binding of AMP-PNP, although a substrate for the K+-dependent phosphatase reaction also catalyzed by this enzyme,p-nitrophenyl phosphate, did not. This lack of competition is consistent with formulations in which the phosphatase reaction is catalyzed at the low-affinity substrate sites.  相似文献

1. The question of the critical pore diameter for streaming potential is discussed. 2. The surface charge is calculated for cellulose in contact with solutions of K3PO4, K2CO3, K2SO4, KCl, and ThCl4. 3. The surface charge of cellulose in contact with a solution of 2 x 10–4 N NaCl is calculated as a function of temperature and is found to show a sharp break at 39°. This is interpreted in terms of the change of the specific heat of water. 4. A marked ion antagonism is found in NaCl:KCl, KCl:MgCl2, NaCl:MgCl2, NaCl:CaCl2, KCl:CaCl2, CaCl2:MgCl2 mixtures when the surface charge is calculated as a function of concentration.  相似文献

1.25 per cent gelatin solutions containing enough NaOH to bring them to pH 7.367 (or KOH to pH 7.203) were made up with various concentrations of NaCl, KCl and MgCl2, alone and in mixtures, up to molar ionic strength. The effects of these salts on the pH were observed. MgCl2 and NaCl alone lower the pH of the Na gelatinate or the K gelatinate, in all amounts of these salts. KCl first lowers the pH (up to 0.01 M K+), then raises the pH. Mixtures of NaCl and KCl (up to 0.09 M of the salt whose concentration is varied) raise the pH; then (up to 0.125 M Na+ or K+) lower the pH; and finally (above 0.125 M) behave like KCl alone. Mixtures of MgCl2 and NaCl raise the pH up to 0.10 M Na+, and lower it up to 0.15 M Na+ regardless of the amount of MgCl 2. Higher concentrations of NaCl have little effect, but the pH in this range of NaCl concentration is lowered with increase of MgCl2. Mixtures of MgCl2 and KCl behave as above described (for MgCl2 and NaCl) and the addition of NaCl plus KCl to gelatin containing MgCl2 produces essentially the same effect as the addition of either alone, except that the first two breaks in this curve come at 0.07 M and 0.08 M [Na+ + K+] and there is a third break at 0.12 M. In this pH range the free groups of the dicarboxylic acids and of lysine are essentially all ionized and the prearginine and histidine groups are essentially all non-ionized. The arginine group is about 84 per cent ionized. Hence we are studying a solution with two ionic species in equilibrium, one with the arginine group ionized, and one with it non-ionized. It is shown that the effect of each salt alone depends upon the effect of the cation on the activity of these two species due to combination. The anomalous effects of cation mixtures may be qualitatively accounted for if one or both of these species fail to combine with the cations in a mixture in proportion to the relative combination in solutions of each cation alone. Special precautions were taken to ensure accuracy in the pH measurements. The mother solutions gave identical readings to 0.001 pH and the readings with salts were discarded when not reproducible to 0.003 pH. All doubtful data were discarded.  相似文献

Abstract Ethylene and ethane production in mung bean hypocotyl sections were evaluated as possible indicators of stress due to contact with four salts that are common in natural sites. Ethylene production decreased with increasing concentrations of applied NaCl and KCl. When CaCl2 was applied, the ethylene evolution was greater. However, when MgCl2 was applied, ethylene evolution remained high then decreased and at higher salt concentrations again showed an increase. NaCl (up to 0.1 kmol m?1) and KCl (up to 0.5 kmol m?3) caused a concentration-dependent increase in ethane production. The ethane production with CaCl2 was the lowest among the salts tested and only a minute increase was noticed with the increase of concentration from 0.01 to 1 kmol m?3. Ethane production showed a distinct maximum at 0.2 kmol m?3 MgCl2. The introduction of 0.01 kmol m?3 CaCl2, as well as anaerobic conditions obtained by purging vials with N2, eliminated that high ethane production. Respiratory activity of the mung bean hypocotyl sections in MgCl2 concentrations from 0 to 0.5 kmol m?3 was correlated with ethane but not with ethylene production. The ethane/ethylene ratio showed three patterns for the four salts tested.  相似文献

During stationary phase of growth under low stress of iron in succinic acid medium, Alcaligenes feacalis BCCM ID 2374 produced microbial iron chelators. Increase in iron concentration supported bacterial growth but suppressed siderophores production, 1 μM and 2 μM of iron was optimum for maximum siderophore yield, i.e. 354 and 360 μg/ml in untreated and deferrated medium, respectively. Threshold level of iron, which suppressed siderophores production in A. feacalis BCCM ID 2374, was 20 μM. Ten micromoles and above concentration of CuCl2 and CoCl2, and 20 μM of MgCl2, MgSO4, ZnCl2 and ZnSO4 severely affected siderophores production.  相似文献

Addition of KCl to medium at 0.1 M or higher promoted the formation of conidial heads in Aspergillus oryzae. When higher concentrations of KCl were added, a larger number of conidial heads were formed. NaCl and MgCl2 were slightly less effective than KCl. The effect of salt on the formation of conidial heads on a minimal medium was as high as on a potato/dextrose medium but slightly higher than that on a complex medium when 1 M KCl was added.  相似文献

I. The Plasmalemma. 1. On the plasmalemma of amebæ CaCl2 antagonizes the toxic action of LiCl better than it does NaCl, and still better than it does KCl. MgCl2 antagonizes the toxic action of NaCl better than it does LiCl and still better than it does KCl. 2. CaCl2 antagonizes the toxic action of LiCl and of KCl better than does MgCl2: MgCl2 antagonizes NaCl better than does CaCl2. II. The Internal Protoplasm. 3. The antagonizing efficiency of CaCl2 and of MgCl2 are highest against the toxic action of KCl on the internal protoplasm, less against that of NaCl, and least against that of LiCl. 4. CaCl2 antagonizes the toxic action of LiCl better than does MgCl2: MgCl2 antagonizes the toxic action of NaCl and of KCl better than does CaCl2. 5. LiCl antagonizes the toxic action of MgCl2 on the internal protoplasm more effectively than do NaCl or KCl, which have an equal antagonizing effect on the MgCl2 action. III. The Nature of Antagonism. 6. When the concentration of an antagonizing salt is increased to a toxic value, it acts synergistically with a toxic salt. 7. No case was found in which a potentially antagonistic salt abolishes the toxic action of a salt unless it is present at the site (surface or interior) of toxic action. 8. Antagonistic actions of the salts used in these experiments are of differing effectiveness on the internal protoplasm and on the surface membrane.  相似文献

Permeability to water in unfertilized eggs of the sea urchin, Arbacia punctulata, is found to be greater in hypotonic solutions of dextrose, saccharose and glycocoll than in sea water of the same osmotic pressure. 2. The addition to dextrose solution of small amounts of CaCl2 or MgCl2 restores the permeability approximately to the value obtained in sea water. 3. This effect of CaCl2 and MgCl2 is antagonized by the further addition of NaCl or KCl. 4. It is concluded that the NaCl and KCl tend to increase the permeability of the cell to water, CaCl2 and MgCl2 to decrease it. 5.
