Aims: To evaluate the mycobiota and natural levels of aflatoxins, fumonisins and zearalenone present in compound feed and home‐corn grains intended for fattening pigs. Methods and Results: Total fungi, Fusarium and Aspergillus species occurrence were examined. Aflatoxins and zearalenone were detected by thin‐layer chromatography and fumonisins by high‐pressure liquid chromatography. Fungal counts were generally higher than 1 × 105 colony forming units (CFU) ml?1. Aspergillus flavus, Aspergillus parasiticus and Fusarium verticillioides were the most prevalent species. FB1 and FB2 were detected in all feed and corn samples. Aflatoxin B1 was detected in 33·33% of initial and growing feed and in 44·44% of final feed samples. It was not detected in corn samples. All feed and corn samples were negative for AFB2, AFG1, AFG2 and ZEA presence during all growing stages tested. Conclusions: Fungal counts at all growing periods exceeded the levels proposed as feed hygienic quality limits. Aflatoxin levels in all feeds and fumonisin levels in many samples were higher than the established regulations. Significance and Impact of the Study: The presence of mycotoxins indicates the existence of contamination. This fact requires periodic monitoring to prevent the occurrence of mycotoxicosis in animal production, to reduce the economic losses and to minimize hazards to human health.  相似文献

Natural occurrence of fumonisins B1 (FB1) and B2 (FB2), a promoter for hepato-carcinogenesis, was investigated in corn and corn — based products sampled in Japan, Nepal, and China by high — performance liquid chromatographic method. From the 9 imported corn kernel and 6 gluten feed samples, FB1 was detected in 8 corn (0.6 ~ 4.1μg/g) and all gluten feed (0.3 ~ 2.4μg/g) samples, while FB2 was found in the same corn (0.3 ~ 10μg/g) and 3 gluten feed (0.8 ~ 8.5μg/g) samples. ELISA analysis also revealed the contamination of aflatoxin B1 in 2 corn and all gluten feed samples along with fumonisins. Of 17 corn grit samples, 14 and 5 samples were contaminated with fumonisin B1 and B2, with maximum levels of 2.6 and 2.8μg/g, respectively. As for corn-based foodstuffs marketed in Japan, no significant contamination of fumonisins was observed. Among 24 corn kernel samples in Nepal, 12 and 7 samples were positive for FB1 and FB2, and averaged to 0.6 and 1.6μg/g, respectively. One sample showed the highest fumonisin contents as 4.6 and 5.5μg/g, respectively. In corn samples harvested at Shanghai and Beijing, China, FB1 and FB2 were detected in various concentrations. Mycological survey has also revealed the presence of a fumonisin — producing fungus in a crop field of Japan. These findings have for the first time demonstrated high levels of contamination of fumonisins in corn and corn — based products in Asian countries. Natural co — occurrence of fumonisins and aflatoxin B1 was also detected in raw materials for mixed feed.  相似文献

A total of 404 samples of commercial swine feed from Portugal feed mills were analysed by HPLC methods for the presence of mycotoxins: 277 samples of feed for fattening pigs were analysed for ochratoxin A (OTA), zearalenone (ZEA), and deoxynivalenol (DON), and 127 samples of feed for sows were analysed for ZEA and fumonisins (FB1 + FB2). Concerning feed for fattening pigs, 21 (7.6%) samples were positive for OTA, (2–6.8 μg/kg), 69 (24.9%) were positive for ZEA (5–73 μg/kg), and 47 (16.9%) were positive for DON (100–864 μg/kg). In feed for sows, the results showed 29.9% of positive samples for ZEA (5–57.7 μg/kg) and 8.7% positive samples for FB1 and FB2 (50–391.4 μg/kg). Co-occurrence of DON/ZEA was found most frequently, but simultaneous contamination with OTA/ZEA and OTA/DON was also found.  相似文献

Twenty-one rice samples from field (ten), store (six) and market (five) from the traditional rice-growing areas of Niger State, Nigeria were analysed for aflatoxins (AFs), ochratoxin A (OTA), zearalenone (ZEA), deoxynivalenol (DON), fumonisin B1 (FB1) and B2 (FB2), and patulin (PAT) by thin-layer chromatography (TLC) and high-performance liquid chromatography (HPLC) respectively. T-2 toxin was determined using TLC only. AFs were detected in all samples, at total AF concentrations of 28–372 μg/kg. OTA was found in 66.7% of the samples, also at high concentrations (134–341 μg/kg) that have to be considered as critical levels in aspects of nephrotoxicity. ZEA (53.4%), DON (23.8), FB1 (14.3%) and FB2 (4.8%) were also found in rice, although at relatively low levels. T-2 toxin was qualitatively detected by TLC in only one sample. Co-contamination with AFs, OTA, and ZEA was very common, and up to five mycotoxins were detected in a single sample. The high AF and OTA levels as found in rice in this study are regarded as unsafe, and multi-occurrences of mycotoxins in the rice samples with possible additive or synergistic toxic effects in consumers raise concern with respect to public health.  相似文献

Peripheral blood lymphocytes were isolated from broiler chicks that had ingested feed amended with autoclavedFusarium proliferatum culture material containing fumonisin B1 (FB1), fumonisin B2 (FB2) and moniliformin. Lymphocyte viability was determined for birds that were placed on amended rations at day 1 or day 7 of age at three different levels of mycotoxins, ranging from 61–546 ppm FB1, 14–94 ppm FB2 and 66–367 ppm moniliformin. Reduction of the tetrazolium salt, MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide], to yield MTT formazan, based on mitochondrial metabolic activity, was used to assess cell viability. Lymphocyte cytotoxic effects were observed in all treatment groups on day 21; chicks that started on amended feed at day 1 of age were affected more than those that started at day 7. Abnormal erythrocytes resembling early stages of erythroblasts were observed in peripheral blood from test chicks. Abnormally shaped red cells (poikilocytes) having a spindle-shape with one or both ends pointed were present. Some red cells appeared to be undergoing mitosis. Both reduced lymphocyte viability and abnormal erythrogenesis occurred in chicks given feed amended withF. proliferatum culture material containing FB1, FB2 and moniliformin.  相似文献

The objective of this study was to monitor the occurrence and levels of fumonisin B1 (FB1) and fumonisin B2 (FB2) in animal feeds distributed in South Korea in 2011. The contamination levels of FB1 and FB2 were investigated in 150 samples of compound feeds and in 40 samples of feed ingredients. The contamination rate of feed ingredients with FB1 and FB2 was 50 and 40 %, respectively. FB2 was only found in samples contaminated with FB1. Of the compound feeds, 85 % were contaminated by FB1 and 47 % were contaminated by FB2. The highest contamination rate of FBs was observed in compound feeds for cattle (FB1: 100 %; FB2: 80 %), followed by poultry feed (FB1: 78 %; FB2: 40 %) and swine feed (FB1: 76 %; FB2: 22 %). The highest contamination level (14,600 ng/g) for FB1 were found in poultry broiler feed (early feeding period) samples, which had 82 % contamination rate (9/11), and the highest level of FB2 (2,280 ng/g) was found in feed for fatting calves,which had a contamination rate of 100 %.  相似文献

The natural occurrence of fumonisin B1 (FB1) and B2 (FB2), a promoter for hepatocarcinogenesis, was investigated in Korean corn kernels for feed by HPLC with fluorescence detection. From the 12 corn kernel samples, FB1 was detected in 5 samples at levels ranging from 53 to 1327ng/g, while FB2 was found in 4 samples from 69 to 680ng/g. In the positive samples, the average concentrations of FB1 and FB2 were 506 and 288ng/g, respectively. One sample (No. K3) showed the highest FB1 and FB2 contents as 1327 and 680ng/g, respectively. In the micrological survey on 5 positive samples for FB1 and FB2, 6 strains ofFusarium monlliforme were isolated, and all these isolates had a producibility of FB1 and FB1, with maximum levels of 80.7 to 180.9 g/g.Thisis the first report on the natural co-contamination of FB1 and FB2 in Korean corn kernels for feed, and on the ability ofF. moniliforme isolated from corn kernels for feed in Korea to produce FB1 and FB2.  相似文献

Fumonisins (Fums) are mycotoxins widely distributed in crops and feed, and ingestion of Fums‐contaminated crops is harmful to animal health. The purpose of this study is to explore the effect of Fum B1 (FB1) on barrier functions of porcine intestinal epithelial cells, IPEC‐J2, to clarify the intestinal toxicity of Fums in pigs. The results showed that the persistent treatment of FB1 significantly decreased the viability of IPEC‐J2. Moreover, the expressions of Claudin 1, Occludin, Zonula Occluden‐1 (ZO‐1) on the messenger RNA (mRNA), and protein levels and MUC1 on the mRNA level were significantly inhibited after FB1 treatment, while the mRNA relative expression level of MUC2 was clearly increased. FB1 also enhanced the monolayer cell permeability of IPEC‐J2. Importantly, FB1 promoted the expression of phosphorylated extracellular regulated protein kinase (p‐ERK1/2). These data suggest that long‐term treatment of FB1 can suppress IPEC‐J2 proliferation, damage tight junctions of IPEC‐J2, and regulate expression of mucins to induce the damage of barrier functions of porcine intestinal epithelial cells, which may be associated with the ERK1/2 phosphorylation pathway.  相似文献

Aflatoxins and fumonisins are important food-borne mycotoxins implicated in human health and have cytotoxic effects. The aims of the current study were to evaluate the protective role of Panax ginseng extract (PGE) against the synergistic effect of subchronic administration of aflatoxin B1 (AFB1) and fumonisin B1 (FB1) on DNA and gene expression in rat. Female Sprague–Dawley rats were divided into eight groups (ten rats/group) and treated for 12 weeks including the control group, the group having received AFB1 (80 µg/kg bw), the group having received FB1 (100 µg/kg bw), the group having received AFB1 plus FB1 and the groups having received PGE (20 mg/kg bw) alone or with AFB1 and/or FB1. At the end of experiment, liver and kidney were collected for the determination of DNA fragmentation, lipid peroxidation (LP), glutathione (GSH) contents and alterations in gene expression. The results indicated that these mycotoxins increased DNA fragmentation, LP and decreased GSH content in liver and kidney and down-regulated gene expression of antioxidants enzymes. The combined treatments with AFB1 and/or FB1 plus PGE suppressed DNA fragmentation only in the liver, normalized LP and increased GSH in the liver and kidney as well as up-regulated the expression of GPx, SOD1 and CAT mRNA. It could be concluded that AFB1 and FB1 have synergistic genotoxic effects. PGE induced protective effects against their oxidative stress and genotoxicity through its antioxidant properties.  相似文献

Fusarium species can produce fumonisins (FBs), fusaric acid, beauvericin (BEA), fusaproliferin (FUS) and moniliformin. Data on the natural occurrence of FBs have been widely reported, but information on BEA and FUS in maize is limited. The aims of this study were to establish the occurrence of Fusarium species in different maize hybrids in Mexico, to determine the ability of Fusarium spp. isolates to produce BEA, FUS and FBs and their natural occurrence in maize. Twenty-eight samples corresponding to seven different maize hybrids were analyzed for mycobiota and natural mycotoxin contamination by LC. Fusarium verticillioides was the dominant species (44–80%) followed by F. subglutinans (13–37%) and F. proliferatum (2–16%). Beauvericin was detected in three different hybrids with levels ranging from 300 to 400 ng g−1, while only one hybrid was contaminated with FUS (200 ng g−1). All samples were positive for FB1 and FB2 contamination showing levels up to 606 and 277 ng g−1, respectively. All F. verticillioides isolates were able to produce FB1 (13.8–4,860 μg g−1) and some also produced FB2 and FUS. Beauvericin, FUS, FB1 and FB2 were produced by several isolates including F. proliferatum and F. subglutinans and co-production was observed. This is the first report on the co-occurrence of these toxins in maize samples from Mexico. The analysis of the presence of multiple mycotoxins in this substrate is necessary to understand the significance of these compounds in the human and animal food chains.  相似文献

The intake of mycotoxin-contaminated feeds can lead to nutrient losses and may have adverse effects on animal health and on productivity. The aims of this study were (1) to determine the mycobiota present in poultry feed samples, and (2) to evaluate the natural occurrence of aflatoxin B1, fumonisin B1 and zearalenone. Fungal counts were similar between all culture media tested (103 CFU g−1). The most frequent genus isolated was Penicillium spp. (41.26%) followed by Aspergillus spp. (33.33%) and Fusarium spp. (20.63%). High precision liquid chromatography was applied to quantify aflatoxin B1 and fumonisin B1. Thin layer chromatography was used to determine zearalenone levels. Aflatoxin B1 values ranged between 1.2 and 17.5 μg kg−1. Fumonisin B1 levels ranged between 1.5 and 5.5 μg g−1. Zearalenone levels ranged between 0.1 and 7 μg g−1. The present study shows the simultaneous occurrence of two carcinogenic mycotoxins, aflatoxin B1 and fumonisin B1, together with another Fusarium mycotoxin (zearalenone) in␣feed intended for poultry consumption. Many samples contained AFB1 levels near the permissible maximum and it could affect young animals. A synergistic toxic response is possible in animals under simultaneous exposure.  相似文献

The fumonisin B mycotoxins (FB1 and FB2) have been purified and characterized from corn cultures of Fusarium moniliforme strain MRC 826. Fumonisin B1 (FB1, the major fumonisin produced in culture, has been shown to be responsible for the major toxicological effects of the fungus in rats, horses and pigs. Recent investigations on the purification of compounds with chromatographic characteristics similar to FB1 have led to the identification of two new fumonisins, FB3 and FB4. Fumonisins A1 and A2, the N-acetyl derivatives of FB1 and FB2 respectively, were also purified and shown to be secondary metabolites of the fungus. Short-term carcinogenesis studies in a rat liver bioassay indicated that over a period of 15 to 20 days, at dietary levels of 0.05–0.1%, FB2 and FB3 closely mimic the toxicological and cancer initiating activity of FB1 and thus could contribute to the toxicological effects of the fungus in animals. In contrast, no biological activity could be detected for FA1 under identical experimental conditions. These studies and others have indicated that the fumonisin B mycotoxins, although lacking mutagenicity in the Salmonella test or genotoxicity in the DNA repair assays in primary hepatocytes, appear to induce resistant hepatocytes similar to many known hepatocarcinogens.  相似文献

Fumonisins are mycotoxins produced primarily by Fusarium moniliforme and Fusarium proliferatum in corn. In liquid culture, production of fumonisin B1 (FB1), the most common moiety of the family of fumonisins, can be obtained using a defined medium that is nitrogen-limited. Under nitrogen-limited conditions both growth and the production of FB1 were greatly influenced by pH and aeration. At pH above 5.0, F. proliferatum grew normally but produced little FB1 (<100 μg m−1). At pH below 5.0, there was less growth but substantially more FB1. Below a pH of 2.5, both growth and metabolism were slower with very little FB1 produced. When the optimal pH range of between 3.0 and 4.0 under well-aerated conditions was used, both growth and FB1 production were high. This study aimed to establish the combined effect of aflatoxin B1 (AFB1) and fumonisin B1 (FB1) on wild Litopenaeus vannamei hepatopancreas alkaline phosphatase (AP) activity compared with that of farmed shrimp. AP activity in hepatopancreas extract was confirmed by several specific inhibitor assays. AP activity of wild shrimp was higher than that of farmed shrimp (p?<?0.05). However, AP activity from both wild and farmed shrimp was inhibited when incubated with AFB1 and FB1. The greatest inhibition occurred when AP was incubated with a mixture of AFB1 and FB1. The IC50 for AFB1 on AP activity of wild and farmed shrimp hepatopancreases was 0.790 and 0.398 μg/mL, respectively. The IC50 of FB1 was 0.87 μg/mL for wild shrimp and 0.69 μg/mL for farmed shrimp. These results suggest that, at the mycotoxins concentrations used in the study, AP from farmed L. vannamei was sensitive to the presence of both mycotoxins; however, AP is more sensitive to the combination of AFB1?+?FB1 suggesting a possible synergistic or potentiating inhibitory effect.  相似文献

Fumonisins are mycotoxins produced by Fusarium moniliforme, F. proliferatum, and related Fusarium species found on corn. They occur naturally in corn-based feeds and foods and are suspected human esophageal carcinogens. Fumonisin B1 (FB1), the most common homologue, causes the animal diseases associated with F. moniliforme. Hepato- and nephrotoxicities, disrupted sphingolipid metabolism, and liver cancer have been found in rats fed FB1. To determine the in vivo effects of diets containing fumonisins B2 (FB2) or B3 (FB3), male rats were fed culture materials (CM) of FB1 non-producing F. moniliforme isolates to provide low (4.6–6.7 ppm), mid (32–49 ppm) or high (219–295 ppm) dietary levels of either FB2 (FB2CM) or FB3 (FB3CM). Other groups were fed culture material of an FB1 producing isolate (FB1CM) providing 6.9, 53 or 303~ppm total fumonisins (FB1 : FB2 : FB3 = 1.0 : 0.38 : 0.15) and a tenth group was fed a control diet having no detectable fumonisins. One-half (n = 5/group) the animals were killed after three weeks, at which time the toxicological and histopathological effects of the three culture materials were similar, mimicked the effects of FB1, and included decreased body weight gains, serum chemical indicators of hepatotoxicity, decreased kidney weights, and apoptosis of hepatocytes and kidney tubular epithelium. FB1CM, FB2CM, and FB3CM affected sphingolipids, causing increased sphinganine to sphingosine ratios (Sa/So) in both liver and kidneys. The remaining animals (n = 5/group) were fed a control diet for three additional weeks. All body weight and tissue specific effects, including increased Sa/So, induced by the FB2CM, FB3CM and low level FB1CM diets were absent following the recovery period. Except for mild biliary lesions found in the high dose FB1CM group and a few apoptotic hepatocytes present in one mid- and two high-dose FB1CM rats, no evidence of toxicity remained in these groups following the recovery period.This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

Fumonisin B1 (FB1) is the most abundant of the fumonisin mycotoxins, mainly produced in maize by F. verticillioides and F. proliferatum. A total of 282 corn samples harvested in 2005 from six provinces, the main corn-producing areas of China, were analyzed for FB1 using high-performance liquid chromatography. All samples except one were (99.6%) positive for FB1 at levels varying from 3 to 71,121 ng/g with mean and median levels for all samples of 6,662 and 1,569 ng/g, respectively. During an analysis of the distribution pattern for FB1, it became apparent that 43.6% of tested samples had FB1 concentrations below 1,000 ng/g, while 25.2% contained in excess of 5,000 ng/g. The average exposure to FB1 (1.1 μg/kg body weight/day) is within the provisional maximum tolerable daily intake of 2 μg/kg body weight/day set by the Joint FAO/WHO Expert Committee on Food Additives.  相似文献

In a survey of the mycoflora and mycotoxins in foods and feeds, 66 samples of mixed poultry feeds and some component raw materials were investigated. Fungal counts ranged from < 102 to 1.3 × 106 CFU/g.Fusarium spp. counts ranged from 102 to 1.0 × 106 CFU/g. TheFusarium spp. strains isolated were screened for their potential to produce fumonisin B1 (FB1) and fumonisin B2 (FB2) in maize cultures. Samples and maize cultures were analysed for FB1 and FB2 using TLC and fluorescamine-derivative HPLC. No fumonisins were detected in the samples (<6 ppm).Fusarium moniliforme was isolated in 59.1% of samples, and 97.4% of the strains produced FB1 and 79.4% of strains produced FB2 in maize cultures. Dried Distiller’s Grains with Solubles (DDGS), a by-product of bio-ethanol production from maize and other cereals, is increasingly used as a feed additive. In this study, five Fusarium toxins, including fumonisin B1 (FB1), fumonisin B2 (FB2), deoxynivalenol (DON), zearalenone (ZEN) and beauvericin (BEA) were quantified by LC-MS/MS in 59 corn-DDGS samples. In addition, the fumonisin level in 30 randomly selected-samples was compared using an ELISA detection technique. No sample was free from mycotoxin contamination, and 50.8 % of the samples were co-contaminated with all five mycotoxins. Moreover, toxin levels were generally high, with mean levels of 9 mg kg?1 FB1, 6 mg kg?1 FB2, 1.2 mg kg?1 DON, 0.9 mg kg?1 ZEN, and 0.35 mg kg?1 BEA. Maximum levels for FB1 (143 mg kg?1) and FB2 (125 mg kg?1) are of acute toxicological relevance. The ELISA method had a tendency to underestimate the fumonisin content when compared with LC-MS/MS. Finally, this is the first reported beauvericin contamination in corn-DDGS.  相似文献

Fumonisin B1 (FB1) is a Fusarium mycotoxin frequently occurring in corn in combination with deoxynivalenol (DON) and zearalenone. The aim of this study was to determine if FB1, alone and combined with DON or α-zearalenol (ZEA), zearalenone major active metabolite, can affect granulosa cell proliferation, steroid production, and gene expression in swine. Porcine granulosa cells were cultured for 2 days in serum-containing medium followed by 1 or 2 days in serum-free medium with or without added treatments. Fumonisin B1 had inhibitory effects on granulosa cell proliferation. Deoxynivalenol strongly inhibited cell growth, and no significant difference was detected in combination with FB1. α-Zearalenol showed a stimulatory effect on granulosa cell numbers even in combination with FB1. Regarding steroid production, FB1 increased progesterone production, and FB1 had no effect on estradiol production. Deoxynivalenol strongly inhibited progesterone and estradiol production, and FB1 had no significant effect on this response. α-Zearalenol increased progesterone production, and its combination with FB1 produced additive effects. α-Zearalenol had no effect on estradiol production, whereas it decreased estradiol production when co-treated with FB1. Fumonisin B1 was found to decrease CYP11A1 messenger RNA abundance, and the stimulatory effect of FB1 on progesterone production was found to be not dependent on 3-hydroxy-3-methylglutaryl-coenzyme A reductase activity suggesting that FB1 increases progesterone production through a different mechanism. The results show that these Fusarium mycotoxins can influence porcine granulosa cell proliferation and steroid production, thereby demonstrating their potential reproductive effects on swine.  相似文献
