The present study investigated lead effects on gill morphology, hematocrit, blood sodium, glucose, lipids, protein, and cholesterol of Prochilodus lineatus exposed to two sublethal lead concentrations for 96 h. Preliminary series of short-term static toxicity tests were run to determine LC50 (96 h) of lead in P. lineatus, which was 95 mg Pb.L-1. Therefore, lead concentrations tested in the sublethal experiments were 24 and 71 mg Pb.L-1, which correspond to 25% and 75% of the LC50 (96 h), respectively. Gills of P. lineatus exposed to both lead concentrations during 96 h presented a higher occurrence of histopathological lesions such as epithelial lifting, hyperplasia, and lamellar aneurism. P. lineatus did not show significant alterations in hematocrit during exposure to both lead concentrations. Fish exposed to the highest lead concentration showed a significant decrease in Na+ plasma concentration after 48 h, possibly reflecting a sodium influx rate decrease. P. lineatus exposed to both lead concentrations presented a "classical general adaptation syndrome to stress", as hyperglycemia associated with lowered lipids and proteins was reported. Stress-response magnitude was dose-dependent. While the response to the lowest lead concentration might represent adaptation, the highest concentration seems to characterize exhaustion.  相似文献

The aim of this study was to determine the toxicity of the aqueous extract of neem leaves, a product extensively used in fish-farms as alternative for the control of fish parasites and fish fry predators, for the neotropical fish Prochilodus lineatus. The 24 h LC(50) of neem leaf extract for juveniles P. lineatus was estimated as 4.8 g L(-1); the fish were then exposed for 24 h to 2.5, 5.0 and 7.5 g L(-1) or only clean water (control). Plasma glucose levels were higher in fish exposed to 2.5 g L(-1) and 5.0 g L(-1) neem extract, relative to control, indicating a typical stress response. Neem extract did not interfere with the osmoregulating capacity of the fish, as their plasma sodium, chloride, total protein and osmolarity did not change. The presence of the biopesticide interfered with the antioxidant defense system of P. lineatus, as there was a decrease in liver catalase activity at all neem concentrations and the detoxifying enzyme glutathione-S-transferase was activated in fish exposed to 5.0 g L(-1). Fish exposed to all neem extract concentrations exhibited damaged gill and kidney tissue. These results indicate that although neem extract is less toxic to P. lineatus than other synthetic insecticides used in fish-farming it does cause functional and morphological changes in this fish species.  相似文献

The effects of diflubenzuron (DFB), an insecticide to control ectoparasites in fish farms, on muscle acetylcholinesterase (AChE), detoxifying and antioxidant enzymes, hematological and physiological parameters, and liver histopathology were evaluated in Prochilodus lineatus after 6, 24 and 96 h of exposure to 25 mg L(-1) of DFB. The insecticide caused a reduction in the number of erythrocytes and hemoglobin content after 96 h exposure, probably due to hemolysis. Hyperglycemic response indicated energy mobilization, and may have contributed to the increase in osmolarity after 96 h exposure to DFB. The induction of glutathione-S-transferase (GST) and catalase activities in liver pointed to the activation of xenobiotic metabolic pathways and antioxidant defenses. The decrease in muscle AChE at all experimental times showed that DFB is an AChE inhibitor. In addition, DFB induced hepatic alterations that might impair normal liver functions. These results show that DFB can cause health disorders in fish and further studies are required to better define its safe use in aquaculture.  相似文献

Glyphosate-based herbicides, such as Roundup, represent the most extensively used herbicides worldwide, including Brazil. Despite its extensive use, the genotoxic effects of this herbicide are not completely understood and studies with Roundup show conflicting results with regard to the effects of this product on the genetic material. Thus, the aim of this study was to evaluate the genotoxic effects of acute exposures (6, 24 and 96 h) to 10 mg L(-1) of Roundup on the neotropical fish Prochilodus lineatus. Accordingly, fish erythrocytes were used in the comet assay, micronucleus test and for the analysis of the occurrence of nuclear abnormalities and the comet assay was adjusted for branchial cells. The results showed that Roundup produces genotoxic damage in erythrocytes and gill cells of P. lineatus. The comet scores obtained for P. lineatus erythrocytes after 6 and 96 h of exposure to Roundup were significantly higher than respective negative controls. For branchial cells comet scores were significantly higher than negative controls after 6 and 24 h exposures. The frequencies of micronucleus and other erythrocyte nuclear abnormalities (ENAs) were not significantly different between Roundup exposed fish and their respective negative controls, for all exposure periods. In conclusion, the results of this work showed that Roundup produced genotoxic effects on the fish species P. lineatus. The comet assay with gill cells showed to be an important complementary tool for detecting genotoxicity, given that it revealed DNA damage in periods of exposure that erythrocytes did not. ENAs frequency was not a good indicator of genotoxicity, but further studies are needed to better understand the origin of these abnormalities.  相似文献

The toxicity of Roundup, a glyphosate-based herbicide widely used in agriculture, was determined for the Neotropical fish Prochilodus lineatus. The 96 h-LC(50) of Roundup was 13.69 mg L(-1), indicating that this fish is more sensitive to Roundup than rainbow trout (Oncorhynchus mykiss) and Atlantic salmon (Salmo salar). These differences should be considered when establishing criteria for water quality and animal well-being in the Neotropical region. Short-term (6, 24 and 96 h) toxicity tests were then performed to evaluate the effects of sub-lethal concentrations of the herbicide (7.5 and 10 mg L(-1)) to P. lineatus. Roundup did not interfere with the maintenance of the ionic balance and there was no significant alteration in plasma cortisol levels in Roundup-exposed fish. However an increase in plasma glucose was noted in fish exposed to 10 mg L(-1) of the herbicide, indicating a typical stress response. Catalase liver activity also showed an increase in fish exposed to 10 mg L(-1) of the herbicide, suggesting the activation of antioxidant defenses after Roundup exposure. In addition, Roundup induced several liver histological alterations that might impair normal organ functioning. Therefore, short-term exposure to Roundup at subletal concentrations induced biochemical, physiological and histological alterations in P. lineatus.  相似文献

The loss of oocyte viability after ovulation is one of the limiting factors in controlled reproduction of several fish species. Experiments were performed with 15 feral Prochilodus marggravii female fish induced to spawn with crude carp pituitary extract to evaluate the viability of oocytes retained within the ovarian cavity (in situ storage) and outside of the ovarian cavity (ex situ storage). Because fertility rates rapidly declined after ovulation, simultaneously with an increase in the number of deformed larvae, P. marggravii oocytes could only be successfully stored for 1 h ex situ at room temperature ( approximately 26 degrees C). There was a highly negative correlation (r = -0.82) between fertilization and deformed larvae during in situ storage at approximately 26 degrees C. Ex situ cooling (18 degrees C) caused a drastic reduction in fertilization rates as compared with storage at approximately 26 degrees C. Oocyte structure was preserved during 2 h storage and the cortical reaction was induced before spawning. Since the micropylar apparatus remained open, it was not the primary cause for the loss of oocyte fertility. The cytoskeleton of the oocyte appeared to be affected since ooplasmic segregation was altered after 2 h storage.  相似文献

This paper studies the migration pathways and ranges of a dominant detritivorous fish Prochilodus lineatus along pollution gradients in the Río de la Plata basin using multivariate analysis of biochemical and pollutant data. Biochemical composition (water, ash, lipids, proteins, carbohydrates, neutral lipids classes and fatty acids), aliphatic hydrocarbons (ALI), polychlorinated biphenyls (PCB), linear alkylbenzenes (LAB) and organochlorine pesticides (OClP) were determined in muscle samples of P. lineatus collected in Metropolitan Buenos Aires, the lower Paraná River (Paraná: 200-1000 km from Buenos Aires) and the middle Paraná and Paraguay Rivers (North: 1000-1400 km north). Biochemical variables and pollutants exhibited large variability [Lipids 1.1-89.5% wet mass; ALI 1.4-413; LABs not detectable (n.d.)-115.2; PCBs n.d.-27.9; OClPs n.d.-11.8 μg g(-1) dry mass], due to the contrast of Buenos Aires with North fish. Fish from Buenos Aires were fatty (lipids 24.7 ± 12.3% wet mass), enriched in 18 carbon fatty acids and severely contaminated (ALI 152.4 ± 72.3; LABs 65.1 ± 26.4; PCBs 15.2 ± 6.8; OClPs 1.8 ± 1.9 μg g(-1) dry mass mean ±S.D.). In contrast, fish from North were lean (4.1 ± 3.1% wet mass), enriched in long chain (>20 carbons) polyunsaturated fatty acids, with average one to two orders of magnitude lower pollutant levels (ALI 41.2 ± 51.9; PCBs 2.2 ± 3.5; LABs 8.8 ± 21.1; OClPs 0.67 ± 0.75 μg g(-1) dry mass mean ±S.D.). Paraná showed intermediate values in all variables, denoting the mixing of different fish stocks. Based on principal component analysis, 14 outliers from 60 North and Paraná samples (representing 26 from 108 individual fish) were identified as pertaining to the Buenos Aires group with very similar lipid and pollutant levels. Data suggest that P. lineatus migrates a highly variable distance, exceeding 800-1000 km in multiple spatial and temporal overlapping ranges. Chemometric analysis of biochemical and pollutant data effectively discriminates fish according to the chemical signature acquired by detritus feeding in pristine and contaminated urban or industrial areas.  相似文献

In order to evaluate the effects of copper and atrazine on the stress response of the freshwater fish Prochilodus lineatus, juvenile fish were pre-exposed to copper (20 μg L(-1)) or atrazine (10 μg L(-1)) for 24 h and then submitted to air exposure for 3 min. Simultaneously fish kept in dechlorinated water for 24 h were subjected to air exposure and a non-stress group was not subjected to air stress or any contaminants. Animals were sampled immediately (t0) and after 1, 3 and 6 h of air exposure (t1, t3 and t6 respectively) for the analysis of plasma cortisol, glucose and Na(+), hepatic glycogen, branchial Na(+)/K(+)-ATPase (NKA), number of red blood cells per cubic millimeter of blood (RBC), hematocrit (Hct) and hemoglobin content (Hb). In fish pre-exposed to copper the stress response was inhibited, and at t1 and t3 both cortisol and glucose remained significantly lower compared to fish subjected to air stress only. In fish pre-exposed to atrazine there was no rise in cortisol, but there was an increase in plasma glucose, RBC, Hct and Hb at t0 and a return of these parameters to basal levels at t1, as they did not differ significantly in relation to non-stressed fish. Animals pre-exposed to either Cu or atrazine showed a significant reduction in NKA activity at t1 and t3, in relation to air stressed fish. These results clearly indicate that copper and atrazine impair cortisol stress response of P. lineatus and that fish subjected to a contaminant-induced stress, either by copper or atrazine, may not be able to respond to any additional stressors.  相似文献

Food assimilation by the detritivorous fish Prochilodus lineatus, throughout a life cycle, was studied by means of fatty acid analysis. Fish length ranged from 2.7 to 55 cm and weight from 2 to 2670 g. Samples for fatty acid determination were taken from the mesenteric deposit in adults and from the whole fish in juveniles. Phyto and zooplankton, and superficial mud were also studied. Samples were taken from the principal channel of the Paraná River and lentic environments in its alluvial plain (60 ° 29W and 31 ° 40S). Fatty acids in the smaller fish (2.8–3.8 cm) indicated the assimilation of zoo and phytoplankton. Longer P. lineatus have a fat-deficient diet, particularly in polyunsaturated acids. At different stages of life, P. lineatus is adapted to different sources of food. When young, it eats zoo- and phytoplankton, becomes gradually detritivorous and has a series of adaptations of the digestive tract (morphological and histological) to enable it to assimilate detritus.  相似文献

We investigated the effect of copper on liver key enzymes of the anaerobic glucose metabolism (hexokinase, HK; phosphofructokinase, PFK; pyruvate kinase, PK; lactate dehydrogenase, LDH) as well as of the pentose pathway (glycose-6-phosphate dehydrogenase, G6PDH) from the fish Prochilodus lineatus. The fish were acclimated at either 20 degrees C or 30 degrees C at pH 7.0, transferred to water at pH 4.5 or 8.0, and exposed to 96 h-CL(50) copper concentrations. Copper accumulation in liver was higher in fish acclimated at 20 degrees C and maintained in water pH 8.0. Three-way analysis of variance revealed a significant effect of temperature on all enzymes, a significant effect of pH on all enzymes except for PK, and a significant effect of copper on only PFK, and LDH in pH 4.5 at 20 degrees C and, at 30 degrees C, on PFK and PK at pH 4.5 and 8.0, HK at pH 4.5 and G6PDH at pH 8.0. There were significant interactions between treatments for many enzymes. These changes suggest that the activity of enzymes in question is modified by a change in ambient water. At least at 30 degrees C, the overall reduction in the glycolytic enzyme activities of copper-exposed fish seems to reduce energy availability via glucose metabolism, thereby contributing to enhance copper toxic effects.  相似文献

We described five novel microsatellite loci for the piracema fish species Prochilodus lineatus (Characiformes), endemic to South America and of extreme importance to both commercial and artisanal fisheries across its occurrence area. A primary, unenriched genomic library was constructed and radioactively screened for repetitive motifs. Positive clones were automatically sequenced and based on the design of new primers, polymerase chain reaction assays were carried out to determine optimum reaction and electrophoretic conditions for each characterized locus. We evaluated its usefulness in population genetic studies by determining Hardy-Weinberg equilibrium, FIS and a jackknife estimate of the number of alleles for a sample of fish caught below the Funil Hydroelectric Power Plant dam (N = 95), Grande River, Brazil. The number of alleles varied from 3 to 21 and expected heterozygosities ranged from 0.58 to 0.91. Two of five loci were in Hardy-Weinberg equilibrium. Jackknife estimates of the number of alleles were higher than the observed number of alleles for three loci and could provide a measure of sampling bias. These markers should provide important tools for the determination of genetic structure, stock delimitation and reservoir fish management in the Grande River as well as to improve hatchery practices for environmental mitigation measures and to help sustain fisheries in the river.  相似文献

To broaden the understanding of physiological responses of tropical fish to environmental stressors, the effects of nitrite on haematological parameters and plasma and red blood cell ion regulation were studied in two neotropical fish species, Astyanax altiparanae and Prochilodus lineatus. Both fish species were exposed to NaNO2 (30 mg l(-1)) over a 96-h period and blood samples were taken for ion and haematological analyses. The results revealed that nitrite leads to a decrease in P. lineatus blood haematocrit and haemoglobin content and an increase in blood methaemoglobin. A. altiparanae did not exhibit any significant difference in these haematological parameters. During the exposure to NO2- both fish species had significantly reduced plasma Na+ concentration and red blood cell (RBC) K+ concentration, but only P. lineatus showed an increase in extracellular K+ concentration. When RBC volume was analyzed in vitro, after 2 min of exposure to NaNO2, a 36% shrinkage was observed in P. lineatus cells, while only a 10% shrinkage was observed in A. altiparanae cells. These results suggest that for P. lineatus, nitrite entrance into the cell leads to methaemoglobin formation and K+ efflux, causing red cell shrinkage and increased plasma K+. However, A. altiparanae proved to be a species more resistant to nitrite, exhibiting fewer responses to this compound.  相似文献

Prochilodus lineatus Valenciennes (curimbatá) is an important migratory Neotropical fish. It does not feed during spawning migration, and often survives after spawning. The mobilization of energy reserves and some effects of starvation (zero to eight weeks) on fish health were experimentally evaluated. Hepatic glycogen and lipids from the perivisceral fat bodies were the main reserves mobilized during the first four weeks of fasting. During this period, somatic indices and blood parameters showed that fish health was not significantly affected. However, after five weeks of food deprivation, the main energy reserves were depleted and the fish became anaemic. The loss of muscle mass indicates that protein breakdown was an important energy source after the reduction of hepatic and perivisceral reserves. Mortality was increasingly observed from seven weeks of starvation. Prior accumulation of high amounts of reserves is essential to allow movements for long distances during spawning migrations in this species.  相似文献

The lipid composition of muscle and liver of detritivorous fish Prochilodus lineatus, settling particles and sediments from reference and polluted areas of the Paraná-Rio de la Plata basin were analyzed to evaluate the impact of feeding on contaminated detritus. Overall, muscular lipids were highly variable (14 ± 13 % wet mass, ww) and increased with body mass trough a rapid triglyceride accumulation [82 ± 7.9 % neutral lipids (NL) for fish weighing less than 1 kg to 99 ± 0.51 % NL for fish weighing more than 4 kg] with a parallel decrease of free fatty acid (13 ± 6.0 to 0.11 ± 0.23 % NL). Liver lipids were more uniform (6.0 ± 2.1 % ww) and were dominated by triglycerides (40 ± 21 % NL) and free fatty acids (34 ± 19 % NL). Compared with fish from reference areas in the North, polluted fish from Buenos Aires presented higher muscular lipid contents (24 ± 13 vs. 3.9 ± 3.1 % ww) and triglyceride abundance (98 ± 3.5 vs. 84 ± 9.7 % NL), and enlarged livers (Hepatosomatic index 1.4 ± 0.4 vs. 0.7 ± 0.2) enriched with esterified cholesterol (20 ± 9.1 vs. 11 ± 9.9 % NL). These differences were consistent with the higher proportions of lipids, enriched with free fatty acids and triglycerides, in stomach contents, settling particles and sediments from Buenos Aires relative to the North of the basin. The change in Prochilodus lineatus diet from organic-poor vegetal detritus in the north to polluted but abundant and fresh anthropogenic matter at Buenos Aires resulted in multiple alterations of lipid metabolism.  相似文献

The goal of the work was a study of the effect of exhaustive weightlifting exercise on prolonged changes in the physiological and biochemical variables characterizing the functional state of skeletal muscles. An exercise accentuated at muscles of the hip surface gave rise to a significant increase of the blood lactate concentration, which indicated that aerobic metabolism was a predominant mechanism of energy supply for muscle contraction. A reduction of the m. rectus femoris EMG amplitude and frequency, a decrease in the tone of tension, and an increase in the tone of relaxation were found immediately after exercise. One day later, the amplitude and the frequency of the EMG signal increased. On day 3, the activity of creatine kinase (CK, a marker of muscle injury) considerably increased, while the amplitude and frequency of EMG decreased. By the ninth day of recovery, all measured variables with the exception of CK were normalized. A significant negative correlation was found between the blood serum’s lactate concentration and m. rectus femoris EMG activity at the same time points. Blood serum CK activity and m. rectus femoris EMG and tone parameters were significantly correlated on the third postexercise day. The data demonstrate that exhaustive exercise-induced muscle injury resulted in phasic alterations in the electrical activity and the tone of the muscle apparently related to a decrease in pH because of lactate accumulation in the sarcoplasm and the cascade of reactions leading to muscle tissue damage.  相似文献

The goal of the work was a study of exhaustive weightlifting exercise effect on prolonged changes in physiological and biochemical variables characterized functional status of skeletal muscles. An exercise gave rise to significant blood lactate concentration increase that was indicative of an anaerobic metabolism to be a predominant mechanism of muscle contraction energy supply. A reduction of m. rectus femoris EMG activity (amplitude and frequency), tonus of tension and an increase in tonus of relaxation were found immediately after exercise. Both EMG amplitude and frequency were increased 1 day post-exercise. However, after 3 days of recovery, EMG amplitude and frequency were decreased again and, in parallel, blood serum creatine kinase (CK) activity was significantly increased. After 9 recovery days, all measured variables with the exception of CK were normalized. A significant reverse correlation was found between blood serum lactate concentration and m. rectus femoris EMG activity at the same time points. Blood serum CK activity and m. rectus femoris EMG and tonus variables were observed to be significantly reversely correlated on the 3rd post-exercise day. Presented data demonstrate that exhaustive exercise-induced muscle injury resulted in phase alterations in electrical activity and tonus which correlated with lactate concentration and CK activity in blood serum.  相似文献

The spatial and seasonal variability of stable carbon isotopes in detritivorous Prochilodus lineatus , primary producers and particulate organic carbon (POC) were examined to promote a better understanding of the trophic dynamics of food webs in Neotropical ecosystems. Spatial variability in carbon isotope composition in the species showed a decreasing gradient in the Upper Paraná River system, from the Paraná River and Itaipu Reservoir subsystems to the Baía and Ivinheima subsystems. Fish δ13C followed the isotopic trend of POC along the Paraná River and Baía subsystem. Seasonal differences were not observed in fish. The analysis of percentile contribution of C3 and C4 sources in the species diet revealed the predominance of C3 plants (phytoplanktonic and periphytic algae and riparian vegetation) in most of the analysed subsystems and seasons. Nevertheless, the considerable participation of riparian vegetation was verified, especially in the Ivinheima subsystem in the dry season. C4 macrophytes presented a maximum contribution along the Upper Paraná River. These results demonstrate the existence of specific variability for the consumer and their food sources in different environments of the same ecosystem.  相似文献

This study investigated the application of intra- and extra-cellular cryoprotectant combinations on the quality of curimba Prochilodus lineatus semen subjected to cryopreservation. Semen treatments were tested with 8% DMSO or methanol as intracellular cryoprotectant, 5% egg yolk or lactose as extracellular cryoprotectant and 5% BTS. These cryoprotectant combinations are suitable for curimba but have not been tested at the lesser concentrations proposed or in combination with BTS. Semen samples collected from 19 curimbas were diluted into one of four cryoprotectant combinations: DMSO+yolk; DMSO+lactose; methanol+yolk; and methanol+lactose. After dilution, semen samples were cryopreserved in 0.5 mL straws for 10 days in a liquid nitrogen tank. Semen was thawed in a water bath at 60°C for 8s. We evaluated the quality of fresh, diluted (pre-freezing) and post-freezing semen according to sperm motility rate (%) and duration (s). Sperm morphology was also analyzed in thawed semen. Sperm motility rate decreased progressively after dilution and thawing. The motility rate in post-freezing semen was higher in the treatments using DMSO+lactose and methanol+yolk. Sperm motility duration in post-freezing sperm was greater in the treatments using methanol rather than DMSO as intracellular cryoprotectant, irrespective of the extracellular cryoprotectant used. Abnormality frequency in thawed sperm was less in semen treated with egg yolk than with lactose. Thus the use of methanol intracellular cryoprotectant is recommended along with yolk extracellular cryoprotectant in the cryopreservation process for curimba semen.  相似文献

Acid extracts of carefully dissected proadenohypophysis (PA) and metaadenohypophysis (MA) of the teleost Prochilodus platensis were subjected to chromatography in Sephadex G-50 after which several pro-opiomelanocortin (POMC) peptides were detected by means of three heterologous RIA systems: alpha-MSH, ACTH and beta-endorphin. Parallelism among extracts displacement curves ranged from 26% to 95% of those of the standard curves for the different systems employed. In PA chromatograms, peaks of ACTH immunoreactivity (IR) were detected at the positions of 30 kilodalton (K), 20K, 9K, a large 4.5K peak and 2K. Only one peak of beta-endorphin IR was detected at 30K. In MA chromatograms, ACTH IR detected similar peaks as in PA runs, but 4.5K peak was much smaller, whereas a large 2K peak roughly coincided with all alpha-MSH detected in the chromatograms. beta-Endorphin IR was detected mainly as a large peak coinciding with synthetic beta-endorphin in MA runs. Bioactivity was detected in both PA and MA 4.5K ACTH peaks, whereas little activity could be demonstrated associated with the 30K, 20K and 9K ACTH IR peaks. Prochilodus PAs and MAs were incubated with tritiated aminoacids and the extracts immunoprecipitated with ACTH, beta-endorphin and N-terminal POMC (N-POMC) antisera. The dissociated complexes were run in SDS polyacrylamide slab gel electrophoresis. The tritiated bands detected confirmed the results obtained with Sephadex chromatography. N-POMC immunoprecipitated peptides were located at 28K, 18K and 9K positions. The first two probably accounted for POMC and the N-POMC/ACTH intermediate respectively; the third corresponded to the mammalian 1-76N-POMC.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
