Excised watermelon (Citrullus vulgaris Schrad.) cotyledons were grown in the dark in the presence of 0.1 mM benzyladenine (BA). Under these conditions reserve breakdown and organelle differentiation progress very slowly. Treatment with BA accelerates, breakdown of reserves and stimulates development of organelles. Electron micrographs of cells from treated cotyledons show a larger number of plastids with a more developed inner membrane system. The levels of plastid pigments and enzymes are increased while starch content is reduced. Glyoxysomal enzyme levels are increased by BA during the first three days of development and their decline is accelerated thereafter. Also the activity of hydroxypyruvate reductase (EC 1.1.1.81.), a peroxisomal enzyme, is increased, but this increase is not followed by a decay phase. In water controls, hydroxypyruvate reductase bands together with glyoxysomal enzymes after equilibrium centrifugation in a sucrose gradient. In treated cotyledons the equilibrium position of glyoxysomal enzymes is uchanged while that of hydroxypyruvate reductase is shifted to a lower density.Abbreviations BA benzyladenine - RuDP ribulose-1,5-diphosphate - HPR hydroxypyruvate reductase  相似文献

Successful propagation of seedlings and mature trees of Sorbus domestica L. has been achieved by in vitro methods. Multiple shoot formation was obtained by placing shoot apices or nodal segments on a modified Schenck and Hildebrandt medium containing benzyladenine. Regenerated shoots were excised and induced to root on media with auxin. In the best treatments 75–85% of shoots from juvenile material rooted. Rooting capacity of shoots from mature explants was lower (30%) and was not improved by dipping the base of shoots in concentration solutions of indolebutyric or naphthaleneacetic acids. . In vitro-grown shoot tips excised from preconditioned stock shoots of 'Troyer' citrange were successfully cryopreserved by encapsulation-dehydration. Optimal survival of cryopreserved shoot tips was achieved when encapsulated shoot tips were dehydrated to 17.1% water content. The sucrose concentration in the preconditioning medium significantly influenced the growth and dry matter percentage of the stock shoots as well as subsequent survival of the cryopreserved shoot tips. Maximal growth of stock shoots was obtained in sucrose concentrations in the range of 0.15 M to 0.29 M, while the dry matter percentage increased as sucrose concentration increased up to 0.44 M. The survival of cryopreserved shoot tips increased from 40% to approximately 80% as the sucrose concentration for stock shoots increased from 0.09 M to 0.22 M or 0.29 M. The benzyladenine concentration in the post-culture medium significantly affected the survival and regrowth of the cryopreserved shoot tips. Survival of the shoot tips was lowest when they were post-cultured on benzyladenine-free medium. However, high benzyladenine concentrations (3-4 µM) induced callus formation. Optimal recovery was obtained in post-culture medium containing 2 µM benzyladenine and 0.05 µM !-naphthalene acetic acid. The extraction of shoot tips from alginate beads greatly improved the regrowth of cryopreserved shoot tips.  相似文献

The thidiazolylurea derivative thidiazuron has been reported to be considerably more effective than benzyladenine in promotion of in vitro shoot formation in a number of dicotyledonous species. In the present study, axillary shoots of Miscanthus sinensis (Thunb.) Anderss. Giganteus that had been subcultured four times on modified Murashige & Skoog medium with 20M benzyladenine were transferred to media with benzyladenine, kinetin, isopentenyladenine or thidiazuron at concentrations of 0.01, 0.1, 1, 10, 30 or 100M and grown over four subcultures. Shoot and root formation stabilized after the first subculture and results from the three subsequent subcultures are presented. The common effects of cytokinins, i.e., promotion of axillary bud growth, inhibition of root formation, reduced stem growth and delay of senescence, were observed for all four cytokinins. In a descending order regarding shoot formation, the four cytokinins at the optimum concentration could be ranked as follows: benzyladenine, thidiazuron, kinetin and isopentenyladenine. Benzyladenine and thidiazuron had optimum effects at the same concentration with regard to axillary shoot formation but thidiazuron induced a significantly lower number of shoots than benzyladenine. The number of roots, shoot size and percentage of chlorotic shoots were also the same for benzyladenine and thidiazuron. ABSTRACT

The rooting-recalcitrant rac tobacco mutant has been multiplied in vitro via outgrowth of axillary buds in parallel to the D8 wild-type. The mutant shoots grew at a lower rate and did not root whatever the treatments, whereas the wild-type shoots rooted spontaneously during the culture cycle without auxin treatment. The mutant and wild-type shoots showed similar peroxidase variations along the culture cycle (21 days) but with higher levels of activity for the rac mutant: minimum peroxidase activity occurred at day 14 in whole shoots of both tobacco genotypes, but already at day 7 in the basal parts of the stems (where roots appear) of the wild-type tobacco, while it was delayed in the mutant. Free and conjugated auxin and polyamine levels were also determined in whole shoots and basal parts of the stems. The rac mutant was characterised by higher auxin and polyamine contents. A peak of auxins and polyamines appeared at day 14 in the whole shoots whatever the tobacco genotype. This peak was delayed in the basal parts of the rac stems compared to the wild-type ones. The mutant shoots contained higher levels of benzyladenine and isopentenyladenosine at the end of the culture cycle, whereas zeatin riboside was more abundant in wild-type shoots. In response to increasing concentrations of indole-3-butyric acid (IBA) and 1-naphthaleneacetic acid (NAA), only the wild-type shoots responded by an increase in growth rate followed by inhibition at high concentrations. The rac shoot responses were very low or nonexistent. Peroxidase activity was also measured in the basal parts of tobacco stems grown in the presence of IBA. Results suggest growth inhibition related to auxin accumulation, possibly combined with elevated putrescine content. Second, rooting induction seems to take place in both tobacco genotypes; however, the process of root formation is blocked in the mutant. The lack of initiation and expression phases of rooting in relation to auxin content in the mutant is discussed.  相似文献

Seedling establishment is a critical process to crop productivity, especially under saline conditions. This work was carried out to investigate the hypothesis that reserve mobilization is coordinated with salt-induced inhibition of seedling growth due to changes in source-sink relations. To test this hypothesis, cashew nuts (Anacardium occidentale) were sown in vermiculite irrigated daily with distilled water (control) or 50mM NaCl and they were evaluated at discrete developmental stages from the seed germination until the whole seedling establishment. The salt treatment coordinately delayed the seedling growth and the cotyledonary reserve mobilization. However, these effects were more pronounced at late seedling establishment than in earlier stages. The storage protein mobilization was affected by salt stress before the lipid and starch breakdown. The globulin fraction represented the most important storage proteins of cashew cotyledons, and its mobilization was markedly delayed by NaCl along the seedling establishment. Free amino acids were mostly retained in the cotyledons of salt-treated seedlings when the mobilization of storage proteins, lipids and starch was strongly delayed. Proline was not considerably accumulated in the cotyledons of cashew seedlings as a response to NaCl salinity. According to these results it is noteworthy that the salt-induced inhibition of seedling growth is narrowly coordinated with the delay of reserve mobilization and the accumulation of hydrolysis products in cotyledons. Also, it was evidenced that free amino acids, especially those related to nitrogen transport, are potential signals involved in the regulation of storage protein hydrolysis during cashew seedling establishment under NaCl salinity.  相似文献

Methyl jasmonate (JA-Me) at a concentration of 0.5% in lanolin paste totally inhibited bulblets formation induced by benzyladenine in intactMuscari bulbs. Lower concentrations of JA-Me delayed development and growth of bulblets induced by benzyladenine. It seems that methyl jasmonate acts as a powerful inhibitor of cell division induced by cytokinin in used test. In comparison with methyl jasmonate, abscisic acid did not show an inhibitory effect on bulblets formation induced by benzyladenine, even in a higher concentration.  相似文献

Roots of Vitis vinifera L., were treated with benzyladenine when the plant shoots were 38 cm long. Seventy-two hours after benzyladenine treatment, apical or basal leaves on separate shoots were exposed to 14CO2. Control shoots received 14CO2 but no benzyladenine. Application of benzyladenine directed 14C-photosynthate to roots, but a small amount of radioactivity was detected in the shoot tip when 14CO2 was administered to an apical leaf. Distribution of radioactivity among the sugar, organic acid, and amino acid fractions was altered by benzyladenine treatment. In all parts of plants with roots treated with benzyladenine and apical leaf fed 14CO2, the percentage of the total label in the sugar fraction comprised of fructose was generally more than twice that in control plants.  相似文献

Shoot formation from seedling explants of Pinus heldreichii was induced by pulse treatment with benzyladenine at different concentration, followed by culture on medium lacking plant growth regulators. After treatment with 222 M benzyladenine (BA) an average of 4.6 shoots per explant was obtained. Shoots, detached from explants, rooted with a frequency of about 10 %, and rooted plantlets were successfully transferred to ex vitro conditions.  相似文献

The growth in the sapling stage and participation of reserve materials in the formation of annual new shoots were studied in the evergreen treeQuercus glauca. The growth and some allometric relationship were analyzed for 1-to 4-year-oldQ. glauca plants. Each individual was felled at the foot, or all the leaves were removed at the end of the growth season (December). Sprouts were formed on the remaining stump and new shoots were formed from winter buds under dark or light conditions. The dry weight of each plant part was measured before and afte,, the formation of these new shoots. The amount of reserve materials in each plant part was estimated from the difference in allometric relatationships before and after the formation of new shoots. Although the results showed that a small amount of reserve material existed in roots, the participation of reserve materials in the formation of annual new shoots was negligible inQ. glauca growing under usual conditions, and substrates for the formation of new shoots were derived from the products of photosynthesis in old leaves and developing new leaves. Some of the growth characteristics ofQ. glauca were compared with those of the deciduous speciesQuercus variabilis to explain ecological behavior of the two species in warmtemperate secondary forests.  相似文献

Summary A procedure is described to regenerate shoots and plants from the embryonic axis of avocado seedlings with benzyladenine or thidiazuron. Explants were grown in the dark for 10 d and then transferred to an 18-h photoperiod to induce multiple shoots. Increased concentrations of benzyladenine or thidiazuron resulted in increased shoot production. Shoots were transferred to shoot promoting media where additional shoots were formed. Under the best conditions, six rooted shoots were produced per original explant. About 80% of the shoots with roots survived in the glasshouse and produced normal phenotypic plants.  相似文献

 Adventitious shoot buds were induced from leaf and stem explants of Bacopa monnieri on Murashige and Skoog medium supplemented with benzyladenine or kinetin. The source of the explants as well as different gelling agents in the medium were found to influence shoot induction and eventual shoot growth. The best response was obtained in leaf explants taken from shoot cultures grown in medium supplemented with 2 μM benzyladenine and gelled with 0.2% gelrite. A transverse section of the leaf explant incubated in this medium showed several shoot primordia emerging from the leaf surface. This system exhibited a potential for repeated harvesting of the shoots from the original leaf explant as the latter continued to expand and regenerate new shoots, upon repeated periodical subculturing onto fresh medium. However, the callusing response of the plant was very low. Qualitative TLC studies of the regenerated shoots revealed a phytochemical profile similar to that of the field grown-plants. Summary Multiple shoots were induced from nodal segments of mature trees of Ulmus pumila L. on Murashige and Skoog (MS) medium supplemented with benzyladenine (BA). Further multiple shoots were obtained from nodal segments taken from in vitro proliferated shoots when cultured in MS medium containing 0.5 mg.l–1 BA. Rooting of the shoots was achieved on half or full strength MS medium or in MS medium supplemented with 0.1 mg.l–1 naphtaleneacetic acid (NAA). Rooted plantlets were able to resume independent growth after a short period of acclimatization.  相似文献

The metabolism of radioactive metabolites of BA, obtained from tomato shoots, and purified by chromatographic techniques, was studied in rootless tomato shoots and soybean callus. There is apparently a great deal of interconversion between BA, BAR and BARMP. From the present results it is not possible to determine whether BARMP is formed directly from BA or whether BAR serves as an intermediate in its production. Irrespective of the metabolite applied, the principal metabolite formed in all instances had a retention time of 51 min following HPLC separation. This metabolite was itself not metabolized to any great extent by either tomato shoots or soybean callus. The growth of axillary shoots was initiated on nodal stem segments, excised from aseptically grown seedlings of Gentiana acaulis L., G. cruciata L., G. lutea L. and G. purpurea L. In later subcultures, a basal callus tissue developed on the shoots, giving rise to de novo formed buds. Optimum benzyladenine and indoleacetic acid combinations for shoot development were established. They were slightly different in the four species. From 35-70% of shoots rooted spontaneously, except in G. lutea, in which adventitious roots were induced by applying naphthaleneacetic acid. It was conduded that the four Gentiana species were amenable to propagation in vitro. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献

An efficient protocol was achieved for rapid propagation of Wedelia chinensis (Osbeck) Merr. through axillary bud proliferation and ex vitro rooting. Murashige and Skoog (MS) medium supplemented with benzyladenine (BA; 8.87 microM) and indole-3-butyric acid (IBA; 2.46 microM) was optimal for axillary bud proliferation, which developed a mean of 8.3 shoots/node. Excision and culture of node segments from in vitro shoots on medium supplemented with the same concentration of growth regulators developed more than 30 shoots within 40 days. Excision and culture of nodes in succession enhanced the number of shoots. Shoot multiplication did not exhibit decrease in the number of shoots even at 10th subculture. Nevertheless, the shoots exhibited a tendency towards stunted nature. But reduction of BA to 4.44 or 2.22 microM resumed normal growth of shoots. Half strength MS medium fortified with IBA (2.46 microM) induced the highest number of roots. All in vitro rooted shoots survived in field. Dipping of the basal end of shoots collected from multiplication medium in IBA (2.46 microM) solution for 7 days induced roots and its transfer to small pots facilitated the survival of all rooted shoots (100%). Rooting ex vitro by direct transfer of shoots from multiplication medium exhibited 89.2 per cent survival. Use of commercial sugar and tap water and also the omission of in vitro rooting reduce the propagation cost 50-70 per cent. The protocol enables to harvest more than 50,000 plantlets within 150 days starting from a single node explant.  相似文献

The morphogenic capacity of Digitalis obscura leaf explants cultured in vitro has been studied, noting factors promoting the differentiation of roots, buds and shoots as well as those promoting callus proliferation. Callus cultures were established from seedling hypocotyls of the tropical pasture legume Stylosanthes humilis H.B.K., and from leaves of in vitro-grown regenerated plantlets and glasshouse-grown plants. Callus was induced on Murashige and Skoog medium, supplemented with 1.0 mg/1 each of benzyladenine and naphthaleneacetic acid, and subcultured on the same medium with 0.5 mg/1 each of the same plant growth regulators. Induction of shoot formation occurred with a number of benzyladenine/naphthaleneacetic acid combinations. With 1.0 mg/1 benzyladenine (no auxin) all hypocotyl-derived calli and 78% (in vitro-grown plantlets) and 56% (glasshouse-grown plants) of the leaf-derived calli could be induced to form shoots. Morphogenetic potential was maintained during five subcultures. The process of induction of shoot formation took generally longer in leaf-derived calli than in those derived from hypocotyls. Most regenerated plants survived transfer to soil and all tested plants nodulated if inocculated with Rhizobium . No morphological abnormalities were observed.  相似文献

A procedure for multiple shoot formation from somatic embryo explants of Eastern redbud (Cercis canadensis L.) cultured on DKW medium containing benzyladenine (BA) and thidiazuron (TDZ) was developed. TDZ in combination with BA produced more shoots than either treatment alone. The highest number of shoots (3.3 to 3.4 shoots per explant) was obtained from partially desiccated and wounded explants treated with a combination of 5 or 10 M BA and 0.5 or 1.0 M TDZ for 20 days before being transferred to the same medium without TDZ. The number of shoots formed was increased from 1.5 to 3.2 shoots per explant by cutting through the cotyledonary node prior to culture. Hypocotyl segments, 5 to 8 mm length from 4 to 7 day old seedlings, callused on B5 medium supplemented with Kn (0.5 ppm) and NAA (0.1 ppm). Even without transfer, shoots were formed in such cultures. About 20% of the cultures produced multiple shoots. In medium with 1 ppm each of Kn and NAA direct shoots were formed at one end of the hypocotyl segment and callusing was initiated at the other end.
