棉铃虫单核衣壳核多角体病毒几丁质酶基因的克隆与表达   总被引:2,自引:0,他引:2  
根据棉铃虫单核衣壳核多角体病毒(HaSNPV)几丁质酶基因的序列,设计引物,引入适当的酶切位点,利用PCR扩增出不含N末端信号肽以及C末端内质网定位肽的几丁质酶基因片段.将该基因片断克隆至原核表达载体pProEXHTb,经IPTG诱导,在大肠杆菌DH5α中获得了高效表达,表达产物的大小为60kD,含量占菌体总蛋白量的40%.利用来源于AcMNPV 几丁质酶的抗体对表达蛋白进行检测,获得特异性的显色信号,证实所获原核表达产物与杆状病毒的几丁质酶具有同源性.  相似文献

昆虫几丁质酶是昆虫几丁质代谢不可或缺的关键酶类。本研究旨在构建美洲大蠊几丁质酶Pa Cht1基因原核表达载体,纯化并鉴定Pa Cht1重组蛋白,为后续酶活等测定奠定基础。定向克隆Pa Cht1基因成熟肽序列至原核表达载体pET32a(+)上,将构建成功的pET32a-Cht1重组表达质粒导入大肠埃希菌Rosetta中,分别对诱导剂IPTG浓度和诱导时间进行优化,确定最佳诱导浓度和诱导时间,并对表达产物进行可溶性分析。表达产物经镍离子柱纯化后通过Western Blot鉴定。结果表明,美洲大蠊几丁质酶Pa Cht1成熟肽基因编码区长1 083 bp,编码360个氨基酸。最佳诱导浓度和诱导时间分别为0.2 mmol/L和4 h。所得重组蛋白大小约60 k D,与预期结果大小一致,重组蛋白主要以包涵体的形式存在。Western Blot鉴定重组蛋白可与6×His-tag单克隆抗体特异性结合。说明成功构建了原核表达载体pET32a-Cht1,并诱导表达获得了重组几丁质酶蛋白。  相似文献

将天花粉蛋白、烟草几丁质酶和烟草β1,3葡聚糖酶的结构基因分别克隆到原核表达系统中进行表达,对三种基因的原核表达产物的粗提取物分别进行体外抗菌活性检测,发现三种蛋白质均有抗真菌活性。三种蛋白中任意两种蛋白的组合,其抗真菌活性显著高于单一组分的抗真菌活性。三种蛋白共同作用时,获得了更好的抗真菌效果  相似文献

目的:实现棘孢木霉(Trichoderma asperellum)几丁质酶基因tachi2的原核高效表达,研究几丁质酶Tachi2的酶学性质.方法:利用PCR技术扩增得到几丁质酶基因tachi2,将其克隆到原核表达载体pEHISTEV中,测序后,转化大肠杆菌BL21感受态细胞,经异丙基硫代-β-D-半乳糖苷(IPTG)诱导后进行Tachi2蛋白的纯化和复性.用纯化的目的蛋白Tachi2进行几丁质酶酶学性质的研究.结果:tachi2基因在重组大肠杆菌中正确表达,其主要以包涵体形式存在;重组蛋白Tachi2分子量约为44kDa,经过纯化和复性后得到的Tachi2有较高的几丁质酶活性.该酶的最适温度为40℃,最适pH值为7.0,几丁质酶在40℃以下比较稳定、pH 6~9时酶有较高活性,受Cu2和Zn2+的强烈抑制.结论:成功实现了棘孢木霉几丁质酶基因tachi2的原核高效表达,表达纯化了重组蛋白,明确了几丁质酶Tachi2的酶学性质,为该几丁质酶的进一步开发利用和深入研究奠定了基础.  相似文献

运用RT-PCR技术扩增编码烟夜蛾Helicoverpaassulta(Guen啨e)幼虫几丁质酶基因的cDNA片段,将其克隆至pMD18-T载体,获得该基因的成熟蛋白阅读框序列。将该基因重组到表达型质粒pGEX-4T-2中,并转化入原核细胞中表达,序列测定结果表明,烟夜蛾幼虫几丁质酶基因的成熟蛋白阅读框全长1338bp,编码445个氨基酸残基,预测分子量和等电点分别为50.1kDa和9.26;推导的氨基酸序列与其近缘种棉铃虫几丁质酶氨基酸序列的一致性达99%,与其他6种昆虫几丁质酶的氨基酸序列也高度一致(65%~76%),并具有几丁质酶的典型特征。将该基因克隆到原核表达载体pGEX-4T-2上并转化BL21,SDS-PAGE和Western印迹分析表明,经IPTG诱导,76kDa附近没有特异蛋白条带出现,表明烟夜蛾几丁质酶基因不能在原核表达载体pGEX-4T-2中表达。  相似文献

根据本研究组已克隆的大白菜ClassⅣ类几丁质酶基因序列设计引物,通过RT-PCR反应扩增得到该几丁质酶成熟肽基因CHB4,构建原核表达载体pET-CHB4,利用IPTG诱导表达并对诱导表达参数进行优化,SDS-PAGE分析表明,该基因表达的蛋白分子量为28 kD左右,其表达产物主要以包涵体的形式存在,25℃并没有改变表达蛋白的可溶性,而在pH9.5的培养基条件下,诱导表达产物的上清液中却有重组蛋白条带出现。以酵母菌为指示菌做抑菌圈实验,结果显示IPTG浓度为0.6 mmol/L时抑菌效果最明显。几丁质酶的活力测定结果表明,当IPTG浓度为0.6 mmol/L时,几丁质酶活力达到最大值2.8 U/mL。  相似文献

【目的】克隆耐冷菌假交替单胞菌(Pseudoalteromonas sp.DL-6)的几丁质酶基因并进行原核表达,纯化重组蛋白并研究其酶解产物。【方法】采用PCR扩增法从Pseudoalteromonas sp.DL-6中克隆几丁质酶基因(chi A),连接到表达载体p ET28a,导入Escherichia coli BL21(DE3)进行诱导表达。SDS-PAGE检测几丁质酶Chi A的分子量与纯度,4-甲基伞形酮荧光底物4MU-(Glc NAc)2测定酶活,电喷雾质谱(ESI-MS)检测酶解产物。【结果】chi A基因(Gen Bank登录号KF234015)在大肠杆菌中高效表达,Ni-NTA亲和层析柱纯化几丁质酶Chi A的总活力可达168.68 U。ESI-MS检测结果表明重组蛋白酶解1%胶体几丁质的产物为几丁寡糖。【结论】利用内切几丁质酶Chi A水解几丁质生产几丁寡糖,为其在食品、医药和农业等领域的潜在应用提供有利参考。  相似文献

为了探索杆状病毒几丁质酶对微生物杀虫剂的增效作用及其利用途径 ,分别在大肠杆菌和昆虫细胞中表达棉铃虫单粒包埋型核型多角体病毒 (HaSNPV)几丁质酶 .用PCR方法扩增出不含N端信号肽编码序列的几丁质酶基因片段 ,并分别克隆至原核表达载体pET2 8a和重组到杆状病毒BactoBac表达系统 ,在大肠杆菌 (E .coli)BL2 1和粉纹夜蛾 (Trichoplusiani)细胞系Tn 5B1 4中分别进行了表达 .在大肠杆菌中表达量约占细菌总蛋白 15 % ,在昆虫细胞中表达量约占细胞总蛋白10 % .将含有几丁质酶的大肠杆菌和昆虫细胞表达产物添加到苏云金杆菌 (Bt)菌液中一起喂食 2龄家蚕 .结果显示 ,HaSNPV几丁质酶基因的 2种表达产物和Bt杀虫剂的混合物使处理的家蚕的致死时间较对照处理均明显缩短 .昆虫细胞和大肠杆菌表达产物与Bt混合物处理的LT50 分别从 93 5h和 95 1h缩短到 5 6 2h及 6 7 2h ,并且供试家蚕的生长速度明显缓慢 .研究结果表明 ,重组的HaSNPV几丁质酶有望作为Bt杀虫剂的增效剂  相似文献

几丁质脱乙酰酶(Chitin deacetylase,CDA)是昆虫几丁质代谢酶系中的重要组分,是害虫防治的重要靶标。通过RT-PCR技术克隆得到编码甜菜夜蛾几丁质脱乙酰酶secda7基因(Gen Bank登录号为MG604929),该基因长1 431 bp,包含开放阅读框长1 134 bp,SeCDA7蛋白的预测分子量分别为43.156 k D。结构域分析显示,SeCDA7具有一个多聚糖乙酰基转移酶催化区,属于第Ⅴ类CDA蛋白。分别构建了原核和真核重组表达载体,利用大肠杆菌和Bac-to-Bac昆虫杆状病毒表达系统转染Sf9昆虫细胞,成功表达了SeCDA7蛋白,纯化SeCDA7蛋白并分析几丁质结合活性,结果表明SeCDA7蛋白具有几丁质结合活性;荧光定量PCR结果显示secda7基因主要在中肠组织表达。本研究实现了甜菜夜蛾几丁质脱乙酰酶基因secda7的外源表达,并鉴定出SeCDA7蛋白具有几丁质结合活性,为深入探究甜菜夜蛾几丁质脱乙酰酶的生理功能提供了理论依据。  相似文献

几丁质酶(Chitinase,Chi.)、β-1,3-葡聚糖酶(β-1,3-Glucanase,Glu.)和萝卜抗真菌蛋白(Rs-AFP2)是植物体内正常的表达产物,它们对防御植物的真菌病害具有重要的作用。基于它们在功能上具有协同作用,本研究利用基因工程技术构建了几丁质酶和抗真菌蛋白、几丁质酶和葡聚糖酶双价表达载体,通过农杆菌直接转化技术将双价表达载体转入农杆菌EHA105,最后采用PCR、DNA dot blotting技术对所获得的农杆菌工程菌株进行了鉴定分析。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
