The underside of Arctic sea ice is inhabited by several autochthonous amphipod species (Apherusa glacialis, Onisimus spp., Gammarus wilkitzkii). The amphipods graze on ice-bound organic matter, such as ice algae, detritus and ice fauna, and release faecal pellets into the underlying water column, thus forming a direct link between the sea ice and the pelagic ecosystems. Experiments on faecal pellet production rates showed species-specific differences, which were related to size of the animals. The smallest species, A. glacialis, produced the highest mean number of pellets (15.4 pellets ind.-1 d-1), followed by Onisimus spp. (2.7 pellets ind.-1 d-1) and the largest species, G. wilkitzkii (1.1 pellets ind.-1 d-1). Relative carbon content of the pellets was very similar in all species (21.2–22.6% dry mass). Juvenile amphipods (Onisimus spp., G. wilkitzkii) produced more pellets with less POC than adults. Based on field determinations of the POC concentration in the lowermost 2 cm of the sea ice (mean: 36.4 mg C m-2) and mean amphipod abundances (A. glacialis: 33.8 ind. m-2, Onisimus spp.: 0.5 ind. m-2, G. wilkitzkii: 9.4 ind. m-2) in the Greenland Sea in summer 1994, the amount of POC transferred from the ice to the water by faecal pellet production was estimated (0.7 mg C m-2 d-1 or almost 2% of ice-bound carbon). Since this process probably takes place in all ice-covered Arctic regions as well as during all seasons, grazing and pellet production by under-ice amphipods contributes significantly to matter flux across the ice/water interface.  相似文献

The abundance of small faecal pellets is high in marine waters. Little is known, however, about the processes governing their production and fate in the water column. We investigated faecal pellet production and flux in relation to the phytoplankton and copepod assemblages present in Ellis Fjord, Antarctica. Results show that the phytoplankton community shifted from a dominance of diatoms to that of a cryptomonad species during late January. This coincided with an increase in abundance of the small copepods Paralabidocera antarctica and Oithona similis, although Oncaea curvata was still the dominant species. The mean faecal pellet flux was 9943 pellets m–2 d–1. Only 37% of the faecal pellet flux at 5 m sedimented to 10 m depth, 15% to 20 m, and 12% to 40 m depth. Our results suggest that recycling of faecal pellets by copepods contributes to this decreased flux with increasing depth, which concurs with results from large scale oceanic studies. Additionally, we propose that the summer ice melt changes the physical characteristics of the water column and the phytoplankton species abundance and distribution; both of which potentially impact on the distribution and abundance of copepods, thereby regulating faecal pellet flux.  相似文献

Transaldolase (TAL) is expressed at selectively high levels in oligodendrocytes and targeted by autoreactive T cells of patients with multiple sclerosis (MS). Among 14 TAL peptides with predicted HLA-A2 binding, TAL 168-176 (LLFSFAQAV, TALpep) exhibited high affinity for HLA-A2. Prevalence of HLA-A2-restricted CD8+ T cells specific for TALpep was increased in PBMC of HLA-A2+ MS patients, as compared with HLA-A2- MS patients, HLA-A2+ other neurological disease patients, and HLA-A2+ healthy donors. HLA-A*0201/TALpep tetramers detected increased frequency of TAL-specific CD8+ T cells, and precursor frequency of TAL-specific IFN-gamma-producing T cells was increased in each of seven HLA-A2+ MS patients tested. Stimulation by TALpep or rTAL of PBMC from HLA-A2+ MS patients elicited killing of TALpep-pulsed HLA-A2-transfected HmyA2.1 lymphoma cells, but not HLA-A3-transfected control HmyA3.1 targets. Without peptide pulsing of targets, HLA-A2-transfected, but not control MO3.13 oligodendroglial cells, expressing high levels of endogenous TAL, were also killed by CD8+ CTL of MS patients, indicating recognition of endogenously processed TAL. TCR Vbeta repertoire analysis revealed use of the TCR Vbeta14 gene by T cell lines (TCL) of MS patients generated via stimulation by TAL- or TALpep-pulsed APCs. All TAL-specific TCL-binding HLA-A*0201/TALpep tetramers expressed TCR Vbeta14 on the cell surface. Moreover, Ab to TCR Vbeta14 abrogated cytotoxicity by HLA-A2-restricted TAL-specific TCL. Therefore, TAL-specific CTL may serve as a novel target for therapeutic intervention in patients with MS.  相似文献

We conducted the most intensive estimate of the endangered Lower Keys marsh rabbit (Sylvilagus palustris hefneri) metapopulation to date using pellet surveys and capture–recapture methodology. We livetrapped 83 rabbits, evaluated 5 closed population models, and selected the model that best represented the data. We considered the variation in behavioral response model the best model and correlated (r2 = 0.913) its patch population estimates to patch pellet densities. From the prediction equation, we generated a range-wide metapopulation estimate of 317 rabbits, a western clade population of 257 rabbits, an eastern clade population of 25 rabbits, and translocated marsh rabbit populations of 35 and zero on Little Pine and Water keys, respectively. A subset of patches whose marsh rabbit subpopulations were last estimated in 1993 exhibited a 46% decline in abundance over 15 yr. Due to the low estimate of the eastern clade population, special effort should be initiated to avoid loss of genetic diversity. The prediction equation suffers from limited data at high pellet densities, patches with ≥5 pellets/m2. Future studies should investigate if the slope of the regression is indeed near 1 by sampling patches across the range of pellet densities, especially those with ≥5 pellets/m2. The equation provides managers a quick, efficient, and noninvasive method to estimate marsh rabbit abundance from pellet counts but the confidence of predicted rabbit densities from high pellet density patches is low. © 2011 The Wildlife Society  相似文献

Abstract: In the southern Rocky Mountains, USA, snowshoe hares (Lepus americanus) and mountain cottontails (Sylvilagus nuttallii) are syntopic. Previous researchers used size of fecal pellets to identify leporid species in the Southern Rockies, a common criterion for identifying species of cervids. We measured 655 fecal pellets from 10 mountain cottontails and 2,374 fecal pellets from 23 snowshoe hares. We found no relationship between the body weight of mountain cottontails and the size of their fecal pellets (r = 0.04, F = 0.01, P = 0.91) but found one for snowshoe hares (r = 0.48, F = 9.3, P = 0.005). Although the 2 species differed in the size of their fecal pellets, the difference between means (1.2 mm) was sufficiently small to require measuring individual pellets and is only applicable to individuals of adult size. Although fecal pellet counts may be used to estimate presence and relative abundance of snowshoe hares in the absence of syntopic leporids, where multiple species of leporids are syntopic this method may yield misleading results.  相似文献

‘Giant race’ stem nematode (Ditylenchus dipsaci) was well controlled in spring beans (Vicia faba) by up to 5 kg aldicarb or carbofuran ha-1 applied to the seed furrows at sowing. Carbofuran was rather more effective in the clay loam soil used than was aldicarb. The best treatments almost eliminated injury to the stems and nematode infestation in the harvested seed. Similarly applied, oxamyl and fenamiphos were less effective and phorate, dimethoate and disulfoton were ineffective. Applying part of the dosage of an effective nematicide to the seed furrows and part along the plant rows mid-season was no more effective and was sometimes less effective than applying the whole dose to the seed furrows. Treating the plant rows mid-season with aldicarb or phoxim sometimes enhanced control but thiabendazole applied thus did not. Seed furrow applications of aldicarb or carbofuran were much less effective in controlling the nematodes in winter beans and seed dressings were less effective than seed furrow treatments. In one experiment, in plots in which aldicarb or oxamyl had been applied to the seed furrows, phoxim or thiabendazole applied over the rows of plants, enhanced nematode control. In two other experiments, thiabendazole was ineffective when applied in this way or when applied as a combined soil and plant treatment.  相似文献

Upland birds that display grit ingestion behavior are potentially at risk of detrimental effects and death from lead poisoning at trap and skeet ranges and other areas where vast quantities of spent lead shot pellets abound. Because commonly cited force-feeding pellet exposure studies deviate from true field conditions, their results may not reflect true risks faced by upland birds. In particular, studies that use new shot pellets and administer more pellets than would be reasonably ingested, critically interfere with the understanding of actualized pellet exposures. In this study, northern bobwhites (Colinus virginianus), a frequent test species in shot pellet research, were dosed in an ecologically-relevant manner (i.e., with spent shot and with no more than three pellets). Notably, the 56-day post-dosing observation period, during which a battery of physiological measures were recorded, exceeded that of related studies. Despite a sustained suppression of a lead poisoning indicator, the data suggest upland birds can withstand spent shot pellet exposures. Data detected a survivorship ≥95%, absence of illness, demonstrated tolerance for extremely high blood lead concentrations, and unaffected blood parameters. In conjunction with ecological considerations (e.g., spatial scale and animal behavior), concern about bird population losses from the incidental ingestion of spent shot pellets is potentially overstated.  相似文献

Fungus‐growing termites (subfamily Macrotermitinae) cultivate the symbiotic basidiomycete fungus Termitomyces in their fungus comb to digest cellulosic materials and to supply nitrogen‐rich fungal diet. In Japan, the fungus‐growing termite Odontotermes formosanus is found on the Yaeyama Islands and Okinawa Island, Okinawa Prefecture. Odontotermes formosanus is thought to have been recently and artificially introduced to Okinawa Island as its distribution is discontinuous and restricted to small areas. Previous DNA analyses revealed that two types of Termitomyces, namely Termitomyces sp. Type A and Termitomyces sp. Type B, whose fruiting bodies correspond to Termitomyces microcarpus‐like pseudorhiza‐lacking small mushroom and Termitomyces intermedius, respectively, are cultivated by O. formosanus on the Yaeyama Islands. However, information about the Termitomyces types cultivated by O. formosanus on Okinawa Island is limited. To define the fungal types cultivated by O. formosanus on Okinawa Island, I developed a diagnostic polymerase chain reaction method using primer sets specific to the nuclear ribosomal DNA sequences consisting of the internal transcribed spacers (ITS1 and ITS2) and 5.8S rDNA of Termitomyces not using fungal mycelium, but using the termite gut metagenome including fungal DNA as a template. The results indicated that the same two Termitomyces types from Iriomote Island are cultivated by O. formosanus on Okinawa Island. The distribution pattern of Termitomyces types on Okinawa Island showed that Termitomyces sp. Type A is limited to the mountainous side of Sueyoshi Park, despite Termitomyces sp. Type B being widely distributed in the area in which O. formosanus is found. This finding implies that O. formosanus on Okinawa Island was recently introduced from Iriomote Island to Sueyoshi Park.  相似文献

Dispersed tricolpate pollen of Pseudointegricorpus clarireticulatum (Samoil.) Takah. from the Upper Maastrichtian in Zeya-Bureya Basin, Amur (Heilongjiang) River area, Russian Far East/China has been studied with light and electron microscopy. Pollen size, pole outlines and the shape of equatorial projections show some variation within the species. The exine is striate-reticulate, semitectate and columellate. The species is characterised by highly complex structures that have harmomegathic function and include equatorial projections, endexinous thickenings, difference in the thickness of the infratectum, foot layer and endexine throughout the pollen grain, and equatorial furrows. Exine layers taper towards colpi regions while they break abruptly in furrow regions. The furrows could have helped to shed the exine quickly and enabled pollen germination. A non-extended region with a small cavity in the ectexine was observed in the equatorial region. We think that this region is characteristic of most Triprojectate species.  相似文献

During the last decades wild rabbit (Oryctolagus cuniculus) populations have been progressively declining in the Iberian Peninsula as a result of several factors, namely habitat deterioration. Hence, habitat management has become one of the most commonly used management techniques to restore wild rabbit populations. To test the efficacy of some of these measures (creation of pastures, opening firebreaks) in rabbit populations, two managed (M1 and M2) and two control areas (C1 and C2) were selected in the Portuguese Southwest (SW) Coast. In each study area, the influence of habitat management was tested assessing temporal changes in rabbit abundance obtained from pellet counts and in rabbits’ diet through microhistological analysis of fecal pellets. Rabbit densities were higher in managed than in control areas (mean annual values: M1–1.08 pellets/m2; M2–1.60 pellets/m2; C1–0.69 pellets/m2; C2–0.40 pellets/m2). In general, Gramineae was the most consumed plant group throughout the year and in all study areas. In control areas, consumption of alternative species with low nutritive value (e.g., Cistus ladanifer) was observed, especially in summer. Our data suggest a positive influence of habitat management on rabbit populations since in managed areas individuals presented an overall higher abundance and a more nutritive diet. In addition, there was no evidence that sown species were consumed in detriment to naturally occurring grasses, suggesting that in Mediterranean semi-arid regions clearing vegetation inside the scrubland might be sufficient to improve habitat conditions for rabbits.  相似文献

Aflatoxins (AFTs) are hepatocarcinogens, mutagens, teratogens and toxins. Isolates of AFTs-producing strains of Aspergillus flavus can grow upon both autoclaved soybeans and to a lesser extent field-grown beans. Besides inhibiting the elongation of excised, in vitro cultured soybean roots, the AFTs can impair the roots' ability to remove [14C]-leucine from a culture medium and to incorporate the amino acid into acidinsoluble, cytoplasmic protein. In this connection, exogenous AFTs once taken-up and compartmentalized by the excised roots can reduce the acid-insoluble protein content of what appears to be a non-enriched plasmalemma fraction. Here, we report a combined biochemical and microscopical attempt to determine whether the protein reduction occurs throughout the excised, in vitro-cultured root and whether the plasmalemma proteins which are affected by AFTs can be both solubilized and characterized. Histochemistry of Carnoy-fixed roots revealed a reduction in Ninhydrin-Schiff-stainability throughout the AFTs-treated root. Transmission electron microscopy of 80 000 × g pellets derived from homogenates of both non-treated (NT) and treated (T) roots provided further evidence to our previously reported marker enzyme analyses for the occurrence of the plasmalemma in the 80 000 × g pellet. Treatment of the latter with the Laemmli SDS procedure released >85% of the protein associated with the pellets obtained from homogenates of either NT or T roots. Gel permeation chromatography on Biogel-100 of released proteins from 80 000 × g pellets of both NT and T roots yielded both void volume and retarded peaks. Both the amplitude and the quantities of protein recovered within the void volume peak were less within the void volume of the T than the NT root. Polyacrylamide tube gel electrophoresis of G-100 void volume peaks of chromatographed, SDS-released proteins from the pellets revealed quantitative but not qualitative differences in Coomassie Blue-gel staining patterns between T and NT roots. These results suggest that the SDS methods which we employed could solubilize the 80 000 × g pellet proteins but that combined gel permeation chromatography and tube gel electrophoresis were not sensitive enough to reveal whether AFTs could alter the types of proteins associated with the 80 000 × g pellet (purported plasmalemma).  相似文献

In soil samples taken with a shovel from 97 potato maincrops in east Scotland during 1978 to 1981, wheat bulb fly (Delia coarctata) eggs were more numerous on ridges than in furrows. The ratio of the number of eggs on the ridge to the number of eggs in the furrow was higher in Lothian Region (3·25) than in Fife Region (1·59) but differed significantly (P < 0·05) from 1·0 in both Regions. To obtain maximum precision of the estimate of the total wheat bulb fly egg population in a potato field, more samples should be taken from the ridge than from the furrow, in approximate proportion to the egg numbers.  相似文献

During 2,3,5,6-tetramethylpyrazine production from glucose by Bacillus strains, a novel product was detected and identified as 2,4,5-trimethylimidazole (TMI) by GC/MS. TMI appeared in the culture medium only after glucose had been depleted and then increased to 0.25–0.31 g l−1 in 90–120 h. When the ammonium source was changed from (NH4)2SO4 to (NH4)2HPO4, only about one tenth of TMI was detected. Although the mechanistic events largely remain unclear, both microbial strains tested demonstrated similar dynamic processes of TMI production, suggesting that TMI formation is a genuine feature of Bacillus species. Feeding strategies of copepods were studied during a dinoflagellate-dominated bloom in the North Sea in August 2001. The aim of this study was to evaluate the importance of mesozooplankton grazing as a biological loss factor of harmful algal blooms under natural conditions. Therefore, ingestion, egestion and egg production experiments were performed with the most abundant copepod species Calanus helgolandicus, Temora longicornis and Acartia sp. feeding on the natural phytoplankton community. Dinophysis norvegica and Ceratium furca were the most abundant dinoflagellate species at the time of the experiments. Grazing experiments as well as examination of fecal pellet content revealed C. helgolandicus fed efficiently on D. norvegica. Ingestion rates up to 47 cells female−1 h−1 were measured and a large proportion of the C. helgolandicus fecal pellets contained intact D. norvegica cells. Dinophysis cells were rarely seen in fecal pellets produced by T. longicornis, and never observed in pellets produced by Acartia sp. The ingestion rate of C. furca, which was the dominating Ceratium species, mimicked that of D. norvegica. C. helgolandicus grazed significantly on C. furca (16 cells female−1 h−1), while the ingestion rate of T. longicornis was low and Acartia sp. was not able to graze on C. furca. Egg production experiments revealed that 92% of the C. helgolandicus females produced eggs. The specific egg production rate and the proportion of females producing eggs among T. longicornis were low. This field experiment clearly shows that some copepod species feed efficiently on D. norvegica and C. furca under natural conditions, which may affect the bloom development of these dinoflagellates.  相似文献

Cytohesin Arf-GEFs are conserved plasma membrane regulators. The sole Drosophila cytohesin, Steppke, restrains Rho1-dependent membrane cytoskeleton activity at the base of plasma membrane furrows of the syncytial embryo. By mass spectrometry, we identified a single major Steppke-interacting protein from syncytial embryos, which we named Stepping stone (Sstn). By sequence, Sstn seems to be a divergent homologue of the mammalian cytohesin adaptor FRMD4A. Our experiments supported this relationship. Specifically, heterophilic coiled-coil interactions linked Sstn and Steppke in vivo and in vitro, whereas a separate C-terminal region was required for Sstn localization to furrows. Sstn mutant and RNAi embryos displayed abnormal, Rho1-dependent membrane cytoskeleton expansion from the base of pseudocleavage and cellularization furrows, closely mimicking Steppke loss-of-function embryos. Elevating Sstn furrow levels had no effect on the steppke phenotype, but elevating Steppke furrow levels reversed the sstn phenotype, suggesting that Steppke acts downstream of Sstn and that additional mechanisms can recruit Steppke to furrows. Finally, the coiled-coil domain of Steppke was required for Sstn binding and in addition homodimerization, and its removal disrupted Steppke furrow localization and activity in vivo. Overall we propose that Sstn acts as a cytohesin adaptor that promotes Steppke activity for localized membrane cytoskeleton restraint in the syncytial Drosophila embryo.  相似文献

Fungus‐growing termites of the subfamily Macrotermitinae together with their highly specialized fungal symbionts (Termitomyces) are primary decomposers of dead plant matter in many African savanna ecosystems. The termites provide crucial ecosystem services also by modifying soil properties, translocating nutrients, and as important drivers of plant succession. Despite their obvious ecological importance, many basic features in the biology of fungus‐growing termites and especially their fungal symbionts remain poorly known, and no studies have so far focused on possible habitat‐level differences in symbiont diversity across heterogeneous landscapes. We studied the species identities of Macrotermes termites and their Termitomyces symbionts by excavating 143 termite mounds at eight study sites in the semiarid Tsavo Ecosystem of southern Kenya. Reference specimens were identified by sequencing the COI region from termites and the ITS region from symbiotic fungi. The results demonstrate that the regional Macrotermes community in Tsavo includes two sympatric species (M. subhyalinus and M. michaelseni) which cultivate and largely share three species of Termitomyces symbionts. A single species of fungus is always found in each termite mound, but even closely adjacent colonies of the same termite species often house evolutionarily divergent fungi. The species identities of both partners vary markedly between sites, suggesting hitherto unknown differences in their ecological requirements. It is apparent that both habitat heterogeneity and disturbance history can influence the regional distribution patterns of both partners in symbiosis.  相似文献

White-rot fungi are extensively used in various submerged biotechnology processes to produce ligninolytic enzymes. Transfer of the process from the laboratory to the industrial level requires optimization of the cultivation conditions on the laboratory scale. An interesting area of optimization is pellet growth since this morphological form solves problems such as the decreased oxygen concentration, limited heat, and nutrient transport, which usually occur in dispersed mycelium cultures. Many submerged fermentations with basidiomycetes in pellet form were done with Phanerochaete, Trametes, and Bjerkandera species, among others. In our study, another promising basidiomycete, D. squalens, was used for ligninolytic enzyme production. With the addition of wood particles (sawdust) as a natural inducer and optimization of mixing and aeration conditions in laboratory stirred tank (STR) and bubble column (BCR) reactors on pellet growth and morphology, the secretion of laccase and the manganese-dependent peroxidase into the medium was substantially enhanced. The maximum mean pellet radius was achieved after 10 days in the BCR (5.1 mm) where pellets were fluffy and 5 days in the STR (3.5 mm) where they were round and smooth. The maximum Lac activity (1,882 U l−1) was obtained after 12 days in the STR, while maximum MnP activity (449.8 U l−1) occurred after 18 days in the BCR. The pellet size and morphology depended on the agitation and aeration conditions and consequently influenced a particular enzyme synthesis. The enzyme activities were high and comparable with the activities found for other investigations in reactors with basidiomycetes in the form of pellets.  相似文献

A “precocious” cleavage furrow develops and ingresses during early prometaphase in Mesostoma ehrenbergii spermatocytes (Forer and Pickett-Heaps Eur J Cell Biol 89:607-618, 2010). In response to chromosome movements which regularly occur during prometaphase and that alter the balance of chromosomes in the two half-spindles, the precocious furrow shifts its position along the cell, moving 2–3 μm towards the half cell with fewer chromosomes (Ferraro-Gideon et al. Cell Biol Int 37:892-898, 2013). This process continues until proper segregation is achieved and the cell enters anaphase with the cleavage furrow again in the middle of the cell. At anaphase, the furrow recommences ingression. Spindle microtubules (MTs) are implicated in various furrow positioning models, and our experiments studied the responses of the precocious furrows to the absence of spindle MTs. We depolymerized spindle MTs during prometaphase using various concentrations of nocodazole (NOC) and colcemid. The expected result is that the furrow should regress and chromosomes remain in the midzone of the cell (Cassimeris et al. J Cell Sci 96:9-15, 1990). Instead, the furrows commenced ingression and all three bivalent chromosomes moved to one pole while the univalent chromosomes, that usually reside at the two poles, either remained at their poles or moved to the opposite pole along with the bivalents, as described elsewhere (Fegaras and Forer 2018). The microtubules were completely depolymerized by the drugs, as indicated by immunofluorescence staining of treated cells (Fegaras and Forer 2018), and in the absence of microtubules, the furrows often ingressed (in 33/61 cells) at a rate similar to normal anaphase ingression (~?1 μm/min), while often simultaneously moving toward one pole. Thus, these results indicate that in the absence of anaphase and of spindle microtubules, cleavage furrows resume ingression.  相似文献

Pheromones provide an important source of communication during social interactions of caudate amphibians. To further examine their use in territorial defense, we performed a laboratory experiment to test the hypothesis that non-courting female red-backed salamanders (Plethodon cinereus) deposit pheromones in or on fecal pellets, as males are known to do during territorial advertisement. Four conditions were tested: (1) a burrow marked with a female's own pellet vs. a burrow marked by a conspecific female's pellet, (2) own vs. unmarked burrows, (3) conspecific vs. unmarked burrows, and (4) paired unmarked burrows as a control. Females nose-tapped (for olfactory cues) their own and conspecific pellets about equally. However, they spent significantly more time in both threat and submissive behavior toward the conspecific pellets and spent significantly more time in their own marked burrows. We infer that female P. cinereus do deposit pheromones in or on fecal pellets and that these pellets may be used to advertise territories. The behavioral responses of females toward pellets of other females were more aggressive than those of males (in a previous study) toward pellets of other males.  相似文献
