Mortar panels painted with three different white acrylic coatings were exposed to the environment in urban (São Paulo) and rural (Pirassununga) sites in Brazil for 7 years. After this time, all panels were almost equally discoloured, and paint detachment was observed to only a small degree. The biofilms were composed mainly of cyanobacteria and filamentous fungi, principal genera being Gloeocapsa and Chroococcidiopsis of the cyanobacteria, and Cladosporium and Alternaria of the fungi. Two of the three paints in Pirassununga became covered by a pink film that contained red-encapsulated Gloeocapsa and clay particles. The third, an 800% elastomeric matt formulation, became discoloured with a grey, only slightly pink, film, although the same cyanobacteria were present. The levels of paint detachments from all films in both locations were low, with rating range of 0–1 of a maximum 5 (100% detachment). After high-pressure water jetting, paint detachments increased at both locations, up to 2 in Pirassununga and 3 in São Paulo. Discoloration decreased; L*A*B* analysis of surface discoloration showed that ΔE (alteration in colour from the original paint film) changed from 28–39 before cleaning to 13–16 afterwards. The pink coloration was not entirely removed from Pirassununga samples, suggesting that cyanobacterial cells are difficult to detach, and microscopic analysis of the biofilms confirmed that Gloeocapsa was still present as the principal contaminant on all surfaces, with Chroococcidiopsis being present as the second most common. The brachyuran community of the coast of São Paulo State is represented by about 188 species of crabs inhabiting different kinds of coastal marine environments. The biodiversity of brachyurans found on non-consolidated sublittoral bottom was investigated. The Ubatuba region (Ubatumirim, Ubatuba and Mar Virado bays, Couves and Mar Virado Islands, offshore region) was sampled for 3 years (1998–2000), at depths of 2–40 m. All sampling was performed using a fishing boat equipped with two double-rig nets. We collected 79 brachyuran species representing 9 superfamilies (4 Dromioidea, 1 Homoloidea, 2 Calappoidea, 5 Leucosioidea, 20 Majoidea, 7 Parthenopoidea, 17 Portunoidea, 18 Xanthoidea, and 5 Pinnotheroidea) and 41 genera. Ubatuba bay showed the greatest species richness with 50 species, followed by Ubatumirim with 45 and Mar Virado with 29. The number of species collected represents about 57% of the known species of crabs already reported for the shore of São Paulo State. It is worth noticing that this percentage is restricted only to non-consolidated sublittoral bottom. This fact indicates a great biodiversity of the habitat in this studied region, probably one to the diversity of habitat types present in the bays.  相似文献

The decrease in viscosity is widely used to estimate the activity of pectinolytic enzymes. The partially-purified, thermally-stable trehalase from conidia of Humicola grisea was highly specific for trehalose and was free of potentially interfering activities. The enzyme was fully stable when stored in solution at -15°C for at least 6 months. A combination of experimental and theoretical approaches was used to investigate the role of nutrient starvation as a potential trigger for biofilm detachment. Experimental observations of detachment in a variety of biofilm systems were made with pure cultures of Pseudomonas aeruginosa. These observations indicated that biofilms grown under continuous-flow conditions detached after flow was stopped, that hollow cell clusters were sometimes observed in biofilms grown in flow cells, and that lysed cells were apparent in the internal strata of colony biofilms. When biofilms were nutrient starved under continuous-flow conditions, detachment still occurred, suggesting that starvation and not the accumulation of a metabolic product was responsible for triggering detachment in this particular system. A cellular automata computer model of biofilm dynamics was used to explore the starvation-dependent detachment mechanism. The model predicted biofilm structures and dynamics that were qualitatively similar to those observed experimentally. The predicted features included centrally located voids appearing in sufficiently large cell clusters, gradients in growth rate within these clusters, and the release of most of the biofilm with simulated stopped-flow conditions. The model was also able to predict biofilm sloughing resulting solely from this detachment mechanism. These results support the conjecture that nutrient starvation is an environmental cue for the release of microbes from a biofilm.  相似文献

The abundance and ecological distribution of the shrimp Pleoticus muelleri as a function of certain environmental factors were investigated from January 1998 through December 1999 in the region of Ubatuba, São Paulo, Brazil. The collections were made monthly in the bays of Ubatumirim (UBM), Ubatuba (UBA) and Mar Virado (MV). Each bay was divided into six sampling transects, four transects parallel to the shoreline and two near the rocky shores on the opposite side. We employed a commercial shrimp boat equipped with two double-rig nets. A total of 6252 shrimp were collected, including 3321 from MV, 1467 from UBM, and 1464 from UBA. Most of the shrimp were caught in the deeper, higher-salinity areas. The high abundance of P. muelleri observed in the southern part of the region studied was related to a sediments with a higher percentage of silt and clay. The numbers of P. muelleri were positively correlated with periods of cooler temperatures. Thus, temperature and the type of sediment were determinant factors in the distribution of P. muelleri in this region.  相似文献

The floristic composition (trees) of 26 forests in the state of São Paulo, Brazil, was compared using cluster analysis and Principal Coordinates Analysis (PCO) with simple Euclidean distance. The results obtained indicate the existence of two floristically distinct groups of forests. One group contains forests from higher areas (above 700 m) with a colder climate (Cfa and Cfb) and includes surveys from Angatuba, Atibaia, Guarulhos, Jundiaí, São José dos Compos and São Paulo. The second group is floristically less homogeneous and includes forests of the central and western parts of the state, usually at lower altitudes (500–700 m) and subject to hotter climatic conditions (Cwa).  相似文献

A freshly-prepared yeast extract at 30 or 50 g/l improved the growth of Bradyrhizobium japonicum SEMIA 587 in a 5-l stirred fermenter. Microsporum gypseum is not a common agent of human dermatophytosis. To the best of our knowledge, this fungus has not been described in human immunodeficiency virus (HIV)-infected patients. Upper Paleozoic sedimentary sequences from several localities of the northeastern Paraná Basin were sampled for paleobotanical and palynological studies. Well-preserved terrestrial assemblages from samples of Campinas, São Paulo State, from an outcrop at the 97 km mark on the Bandeirantes Highway, located at the Municipality of Campinas, São Paulo State were recorded where fertile bryophytes and megaspores were previously also found. The Campinas palynoflora is composed of 23 species of bryophytic and pteridophytic spores, 16 species of pollen grains, and one algae (Botryococcus braunii Kützing). Among the sporomorphs, Reticulatisporites asperidictyus Playford and Helby is described for the first time from the Paraná Basin. Assemblages bear diagnostic species of the Ahrensisporites cristatus Interval Zone, such as Anapiculatisporites argentinensis Azcuy, Cristatisporites inordinatus (Menéndez and Azcuy) Playford, and Psomospora detecta Playford and Helby, of Late Carboniferous age (Pennsylvanian). Palynological content suggests a glacio-deltaic/lacustrine environment of deposition. This is supported by lithological data and by the absence of marine palynomorphs.  相似文献

In order to study isolation within the salians subgroup, pair mating crosses involving its 7 species were performed. The results of testing different interspecific combinations varied from complete isolation at the insemination level to the production of fertile hybrids. Some isolation indices are proposed: Ii (isolation index at the insemination level), Fi (isolation index at the fertilization level), Ti (total isolation index), and Ai (average isolation index). A schema of the biological relationships on the basis of the isolation degree is also presented for the 7 species of the subgroup.Part of this study was included in the thesis submitted for the PH.D. degree at the University of São Paulo and was performed during the tenure of a fellowship supported by the Fundação de Amparo à Pesquisa do Estado de São Paulo.  相似文献

Detachment from biofilms was evaluated using a mixed culture biofilm grown on primary wastewater in a tube reactor. The growth of biofilms and the detachment of biomass from biofilms are strongly influenced by hydrodynamic conditions. In a long-term study, three biofilms were cultivated in a biofilm tube reactor. The conducted experiments of biofilm growth and detachment can be divided into three phases: 1) an exponential phase with a rapid increase of the biofilm thickness, 2) a quasi-steady-state with spontaneous fluctuation of the biofilm thickness between 500 and 1,200 microm in the investigated biofilm systems, and 3) a washout experiment with increased shear stress in three to four steps after several weeks of quasi-steady-state. Whereas the biofilm thickness during the homogeneous growth phase can be regarded constant throughout the reactor, it was found to be very heterogeneous during the quasi-steady-state and the washout experiments. Growth and detachment during all three phases was simulated with the same one-dimensional biofilm model. For each of the three phases, a different detachment rate model was used. During the homogeneous growth phase, detachment was modeled proportional to the biofilm growth rate. During the quasi-steady-state phase, detachment was described by random detachment events assuming a base biofilm thickness. Finally, the washout experiment was simulated with detachment being a function of the biofilm thickness before the increase of the shear stress.  相似文献

The process of detachment, through which bacteria use active mechanisms to leave biofilms and return to the planktonic (free-living) state, is perhaps the least understood aspect of the biofilm life cycle. Like other stages of biofilm development, detachment is a dynamic, regulated process, controlled by specific genes, and induced by particular environmental cues. In previous work we discovered Pseudomonas aeruginosa variants that exhibit accelerated biofilm detachment. These hyper-detaching variants arise spontaneously from biofilms at a high frequency, and they exhibit robust detachment under different biofilm growth conditions. Here we show that these variants detach by a mechanism requiring the biosurfactant rhamnolipid and that this detachment mechanism rapidly restores antibiotic sensitivity to separating bacteria. We also show that rhamnolipids can bring about detachment in wild-type P. aeruginosa biofilms. These findings raise the possibility that this detachment mechanism may be useful as a treatment to disrupt established biofilms. Interestingly, the rhamnolipid-mediated detachment mechanism involves the formation of cavities within the centre of biofilm structures. Our data suggest a model to explain detachment that occurs via this pattern.  相似文献

SUMMARY 1. Colonisation, species composition, succession of microalgae and nutrient dynamics in biofilms grown under light and dark conditions were examined during the initial phases of biofilm development in a lentic freshwater environment.
2. Biofilms were developed on inert (perspex) panels under natural illuminated and experimental dark conditions and the panels were retrieved for analysis after different incubation periods. Analysed parameters included biofilm thickness, algal density, biomass, chlorophyll a , species composition, total bacterial density and nutrients such as nitrite, nitrate, phosphate and silicate.
3. Biofilm thickness, algal density, biomass, chlorophyll a and species richness were significantly higher in light-grown biofilms, compared with dark-grown biofilms. The light-grown biofilms showed a three-phased succession pattern, with an initial domination of Chlorophyceae followed by diatoms (Bacillariophyceae) and finally by cyanobacteria. Dark-grown biofilms were mostly dominated by diatoms.
4. Nutrients were invariably more concentrated in biofilms than in ambient water. Nutrient concentrations were generally higher in dark-grown biofilms except in the case of phosphate, which was more concentrated in light-grown biofilms. Significant correlations between nutrients and biofilm parameters were observed only in light-grown biofilms.
5. The N : P ratio in the biofilm matrix decreased sharply in the initial 4 days of biofilm growth; ensuing N-limitation status seemed to influence biofilm community structure. The N : P ratios showed significant positive correlations with the chlorophycean fraction in both light and dark-grown biofilms, and low N : P ratio in the older biofilms favoured cyanobacteria. Our data indicate that nutrient chemistry of biofilm matrix shapes community structure in microalgal biofilms.  相似文献

The effects of u.v. light and of several chemical agents on spores of the tetracycline producer Streptomyces aureofaciens MT1 were studied using survival curves and induction of histidine prototrophic revertants (his +). Spores were highly resistant to u.v.; NTG induced most his + revertants. 4-Nitroquinoline-1-oxide and methyl methanesulphonate also gave good yields of revertants.  For a stable and reliable operation of the biofilm airlift suspension reactor (BAS reactor) means to control biomass concentration, biofilm thickness and biofilm morphology are required. For this reason, the influence of applied detachment forces and surface substrate loading on the formation of heterotrophic biofilms in laboratory-scale BAS reactors was studied. Detachment forces were altered by variation of the initial bare carrier concentration or the superficial air velocity. In addition, the dynamics of biofilm formation during start-up of a full scale BAS reactor (300 m3) was monitored and compared with the laboratory-scale start-up (3 l). This study shows that the biofilm morphology and strength were influenced to a large extent by the surface substrate loading and applied detachment forces. A moderate surface substrate loading and a high detachment force yielded smooth and strong biofilms. The combination of a high surface substrate loading and low detachment forces did lead to rough biofilms, but did not lead to the expected high amount of biomass on the carrier, apparently because of the formation of weaker biofilms. The strength of the bio-films appeared to be related to the detachment forces applied during biofilm formation, in combination with the surface substrate loading. The biofilm morphology and biomass on carrier in the BAS reactor can be controlled using the carrier concentration, substrate loading rate and the superficial air velocity as parameters. The dynamics of biofilm formation during the start-up of a full-scale BAS reactor proved to be similar to heterotrophic biofilm formation in laboratory-scale reactors. This indicates that a model system on the laboratory scale can successfully be applied to predict dynamic phenomena in the full-scale reactor. A suite of techniques was utilized to evaluate the correlation between biofilm physiology, fluid‐induced shear stress, and detachment in hollow fiber membrane aerated bioreactors. Two monoculture species biofilms were grown on silicone fibers in a hollow fiber membrane aerated bioreactors (HfMBR) to assess detachment under laminar fluid flow conditions. Both physiology (biofilm thickness and roughness) and nutrient mass transport data indicated the presence of a steady state mature biofilm after 3 weeks of development. Surface shear stress proved to be an important parameter for predicting passive detachment for the two biofilms. The average shear stress at the surface of Nitrosomonas europaea biofilms (54.5 ± 3.2 mPa) was approximately 20% higher than for Pseudomonas aeruginosa biofilms (45.8 ± 7.7 mPa), resulting in higher biomass detachment. No significant difference in shear stress was measured between immature and mature biofilms of the same species. There was a significant difference in detached biomass for immature vs. mature biofilms in both species. However, there was no difference in detachment rate between the two species. Biotechnol.
