The changes in flax straw structure when treated with sulphur dioxide (SO2) and consequent effect on dew-retting of the treated straw were investigated. The active oxoanions released by sulfurous acid degraded phenolics bound to the polysaccharides and lipids leading to an increase in water-soluble carbohydrates and polyphenols. As a result, proportions of hemicellulose, nitrogen, sulphur, ash and caustic weight loss were higher in treated straw while lipid was reduced. The coarseness of treated fibre was possibly caused by in situ dryback of the hydrolysed polysaccharides. The deposition of salts containing high concentrations of sulphur, calcium and potassium in the vicinity of the fibre bundles and epidermal layer was observed and investigated. In a dew-retting trial comprising SO2-treated and untreated samples, the SO2 treatment delayed microbial colonisation and, consequently, retting by nearly 2 wk compared to the controls. The changes in pH, fungal population and polysaccharide degrading enzymes were monitored. The differences in fibre yield, fineness and strength are presented.  相似文献

Flax dew-retting is widely adopted in most flax-growing countries, but it does not represent a practical solution where dry weather conditions occur after harvest. A study of the local microbiological aspects was undertaken as a contribution to improve field-retting of flax under southern European climates. Fungi were isolated from soil and dew-retted flax in northern Italy, and 23 representative strains were chosen to test their ability to ret flax stems. Experiments were performed in vitro on flax stem pieces artificially inoculated with single fungal strains. Retting degree was assessed with a mechanical test, to evaluate the ease with which the bast was detached from the wood core, and by the analysis of the residual fibre pectins using uronic acid. Uronic acid dosage provided a better differentiation of the strains than the mechanical test. There was a large variability in retting ability among the species assayed and even among strains of the same species. The best results were obtained with all Aspergillus and Penicillium strains, while Mucor and Rhizopus strains showed a variable retting ability. Fusarium, Trichoderma strains and Epicoccum nigrum had the poorest retting abilities among all the fungal strains assayed.  相似文献

The suitability of ensilage as a means of preserving flax was investigated in a series of experiments in which 400 kg round bales of fresh flax were sealed in polyethylene film or plastic wrapped, with or without the addition of formic acid at 2.5 litre t-1 or formalin at 5.6 litre t-1 at the time of baling. Plastic wrapping provided a more effective seal than the bags which were easily punctured by the flax roots resulting in moulding and deterioration of the flax. Where the seal was not broken untreated flax underwent a clostridial fermentation and the pH fell to about 4.8. Cellulolytic activity degraded the flax fibre over a period of 3 to 6 months. The addition of formic acid reduced the cellulolytic activity provided the seal was not broken. In an experiment with 4 kg batches of flax of 65%, 40% or 25% MC sealed in polyethylene film, the addition of formic or propionic acids at 20 g kg-1 DM did not prevent moulding and deterioration, but both NH3 and SO2 at 40 g kg-1 DM preserved the physical structure of the flax. The NH3 darkened the flax and made it pliable and unscutchable while the SO2 bleached it and preserved the fibre without microbiological deterioration. The presence of acids on the moist stored flax appeared to inhibit the progress of normal water retting.  相似文献

Four methods of spray application were compared in 1986 with the recommended method of glyphosate application for pre-harvest retting of flax (Linum usitatissimum). Sampling individual plants immediately after application of glyphosate and dye demonstrated that spray penetration through the crop canopy, measured as dye deposition on the lower stem as a percentage of total deposition per plant, was the same for all methods of application. None of the methods of application improved the uniformity of dye deposition.
The following year, effects of deseeding prior to spraying and of using an air-assisted sprayer were investigated. Deseeding increased dye deposition on the lower stem but did not improve the efficacy of glyphosate. Application with an air-assisted sprayer increased the proportion of spray deposited on the lower stem but not the absolute amount deposited or the efficacy of glyphosate. Dye deposition on individual plants varied greatly for all methods of application and, when glyphosate was applied using the same methods, undesiccated plants occurred in all plots.
Considerable improvements in spray penetration and uniformity of spray deposition appear necessary for successful pre-harvest retting of flax with glyphosate.  相似文献

在研究天然水沤法脱胶的过程中,通过初筛、复筛,从沤麻主生物期的沤麻液中筛选出两株茵落周围产生透明圈较大、脱胶酶活较高的菌株。通过形态观察,并对其多项生理、生化指标进行了分析研究,初步鉴定并命名为枯草芽孢杆菌A1和B1。初步加茵脱胶实验表明:枯草芽孢杆菌A1产生果胶酶、木聚糖酶,而不产生纤维素酶,脱胶周期为72小时;枯草芽孢杆茵B1产生果胶酶、木聚糖酶和纤维素酶,脱胶周期为50小时。  相似文献

从亚麻种植土壤与沤麻水中分离出96株能分解果胶的菌株。通过初筛和复筛获得4株果胶酶高产菌株。其中,在果胶平板培养基上生长速度快、产果胶酶活力高的12号菌株被确定为优势菌,经鉴定其为枯草芽孢杆菌。最适脱胶条件为:麻水比1:15,pH7.5,温度32~33℃,预培养的优势菌接种量3%。结果表明,采用优势菌的脱胶周期比对照缩短50%左右,而且麻的纤维质量明显得以改善。  相似文献

The effect of temperature and humidity on SO2–induced photosynthetic depression was determined in gas exchange experiments with leaves of Vicia faba , L. Stomatal behaviour was sensitive to humidity resulting in higher uptake rates of SO2 and stronger reductions of photosynthesis at low VPD (vapour pressure deficit). After a fumigation period of 2 h, when the photosynthetic rate had stabilized, photosynthesis of leaves exposed to SO2 at 8°C was reduced much more than at 18°C at the same rate of SO2 uptake. Data analysis with a mechanistic model revealed that this effect was due to the slower rate of S(IV) oxidation at lower temperatures, resulting in higher accumulation of S(IV) and thus stronger reduction of photosynthesis. These results were confirmed by experimental analyses of the S(IV) concentration in leaves following fumigation, which showed that more S(IV) accumulated in leaves exposed at a lower temperature. This may explain the high sensitivity of plants exposed to SO2 under winter conditions, when both VPD and temperature are low.  相似文献

Hemp (Cannabis sativa) produces phloem fibres, which are formed in bundles towards the periphery of the stem. This study investigated one aspect of the retting process, the dissociation of these fibres from the core of the stem, which is influenced by the action of micro-organisms. Peel tests were used to investigate the work required to remove a tissue peel, rich in fibres, from the core of the stem. Two separate experiments studied (1) the effects of tissue dehydration as stems were dried out in the oven; and (2) the progress of retting in stems that were dew-retted in the field. A comparison between two different types of peel tests, where the stems were clamped either vertically or horizontally in the jaws of a tensile testing machine and the load applied vertically, showed that the work required to peel stems in the vertical position was over 20% greater than that for the horizontal position. Freshly harvested stems were dehydrated in an oven at 40°C over a period of 42 h and as the moisture content decreased there was a significant increase in the work to peel. In contrast, stems that had been dew-retted in the field showed a significant decrease in work to peel. When the moisture content of stems dried in the oven decreased below 12%, the work to peel rapidly decreased. As the plant tissues dehydrated their fracture characteristics changed and this was reflected in the increase in the work to peel seen in the oven-dried samples. This, however, was not evident in the field-retted samples, in which there was a decrease in work to peel. The field-retted samples were exposed to the activity of micro-organisms for a prolonged period of time compared with the oven-dried samples; this increased the potential degradation caused by microbial enzymes. Thus, the reduced work to peel in dew-retted stems is thought to be due to the colonisation of the stem by micro-organisms and the subsequent retting process. The dramatic decrease in work to peel at low moisture content in the oven-dried stems is thought to result from mechanical changes leading to brittle fracture characteristics. This study indicates that peel tests can be used to objectively monitor the reduction in work to peel for dew-retted stems and relate this reduction to the progress of retting. Thus, peel tests may be used to monitor the effects of different factors on the process of retting in hemp.  相似文献

AIMS: The objective of this study was to investigate the effects of free molecular and bound forms of sulphur dioxide and oxygen on the viability and culturability of a selected strain of Acetobacter pasteurianus and a selected strain of Brettanomyces bruxellensis in wine. METHODS AND RESULTS: Acetic acid bacteria and Brettanomyces/Dekkera yeasts associated with wine spoilage were isolated from bottled commercial red wines. One bacterium, A. pasteurianus strain A8, and one yeast, B. bruxellensis strain B3a, were selected for further study. The resistance to sulphur dioxide and the effect of oxygen addition on these two selected strains were determined by using plating and epifluorescence techniques for monitoring cell viability in wine. Acetobacter pasteurianus A8 was more resistant to sulphur dioxide than B. bruxellensis B3a, with the latter being rapidly affected by a short exposure time to free molecular form of sulphur dioxide. As expected, neither of these microbial strains was affected by the bound form of sulphur dioxide. The addition of oxygen negated the difference observed between plate and epifluorescence counts for A. pasteurianus A8 during storage, while it stimulated growth of B. bruxellensis B3a. CONCLUSIONS: Acetobacter pasteurianus A8 can survive under anaerobic conditions in wine in the presence of sulphur dioxide. Brettanomyces bruxellensis B3a is more sensitive to sulphur dioxide than A. pasteurianus A8, but can grow in the presence of oxygen. Care should be taken to exclude oxygen from contact with wine when it is being transferred or moved. SIGNIFICANCE AND IMPACT OF THE STUDY: Wine spoilage can be avoided by preventing growth of undesirable acetic acid bacteria and Brettanomyces/Dekkera yeasts through the effective use of sulphur dioxide and the management of oxygen throughout the winemaking process.  相似文献

The expansion of coal-fired power stations in South Africa has resulted in growing environmental concerns as they are the largest emitters of sulphur dioxide (SO2). Sulphur dioxide emissions from power plants pose a potential threat to avian populations. However, the effect of SO2 pollution on bird communities is poorly understood. Using point counts we investigated the relationships of bird species richness and species-specific density with SO2 concentrations around Matimba power station. Environmental parameters were derived from remotely sensed data and data reduction was performed using principal component analysis. Generalised linear mixed models were then used to infer the relationships of bird species richness and species-specific densities with SO2 concentrations and the environmental variables. Our results revealed that SO2-polluted air had no influence on bird species richness and densities in our study sites, but SO2 levels were below annual national ambient air quality standards. Vegetation productivity was found to have a greater influence on species densities than SO2 pollution. Monitoring of bird population changes around Matimba power station once neighbouring Medupi power station is fully operational is recommended, as SO2 content of the air will increase significantly, and species may be sensitive to this increase in SO2 levels.  相似文献

Ribulose bisphosphate (RuBP) carboxylase and glycollate oxidase were partially purified from jack pine (Pinus banksiana Lamb.) needles. Preincubation of RuBP carboxylase with HCO3? and Mg2+ markedly stimulated its activity. RuBP carboxylase showed hyperbolic reaction kinetics with respect to HCO3?, Mg2+, and RuBP. Both SO32- and SO42- inhibited RuBP carboxylase, but SO32- was more inhibitory than SO42-. The SO32- inhibition was competitive with respect to HCO3? (whether SO32- was present during activation or was added to the activated enzyme), while the SO42- inhibition was non-competitive with respect to HCO3?. Glycollate oxidase was inhibited more severely by low concentrations of SO32- than by SO42-. Fumigation of jack pine seedlings with 0.34 ppm sulphur dioxide for 24 and 48 hr produced a considerable decline in the activities of these enzymes, but 1 hr of fumigation produced no effect. During the longer exposures the sulphur content of the needles increased considerably, although the needles showed no visible injury. It is suggested that the accumulation of SO32- and SO42- in the needles following sulphur dioxide exposure influenced the enzyme activities.  相似文献

Sycamore (Acer pseudoplatanus L.) leaf litters from 15 woodlands exposed to a broad range of ambient sulphur dioxide (SO2) concentrations were fumigated with environmentally realistic concentrations (ll-20nmol mol?1) of SO2, for 166 d in an open-air fumigation experiment. Fumigation of the sycamore litters significantly increased sulphate-S and proton leaching, and decreased calcium, magnesium and potassium concentrations in leachates and leaf tissues. Leaf litters from relatively unpolluted woodlands showed a tendency to lose higher amounts of sulphate-S, calcium, magnesium and nitrate-N in leachates than litters from polluted sites when exposed to elevated levels of SO2 in treatment plots. Fumigation inhibited the decomposition rates (CO2 evolution) of the leaf litters. Marked changes in the composition of the saprotrophic fungal communities in SO2-fumigated leaf litters were also recorded, but fungal communities and responses to SO2, were similar between woodlands. There was no evidence from our data to suggest that resistance to SO2, was developed in decomposer mycofloras in woodlands more frequently polluted by the gas.  相似文献

The protein particles in soymilk were fractionated in size by differential centrifugation. Particles of more than 100 nm in diameter (LSP) constituted 40% of the total protein in raw soymilk, 70% of the protein components being 11S globulin. LSP was not formed in the presence of 2-mercaptoethanol and sodium ascorbate. LSP was decreased by heating, and particles of 100–40 nm in diameter (MSP) were increased. The formation of MSP was not due to any degradation of LSP but to the combination of supernatant proteins of less than 40 nm in diameter with each other. MSP formed by heating contained the β subunit of 7S and the basic subunit of 11S as main components. The particles of more than 40 nm in diameter (LSP + MSP) constituted 50% of the total protein in both raw soymilk and soymilk.  相似文献

An open reading frame (ORF) with 963 nucleotides from Paenibacillus campinasensis BL11 was cloned and expressed in Escherichia coli. It encodes a pectate lyase (EC 4.2.2.2) of 35.6 kDa, denominated Pel‐BL11. The recombinant Pel‐BL11 was fused with His‐tag and purified. An optimal activity of 1623 IU mg?1 was exhibited at 50°C, pH 10. Significant activities of Pel‐BL11 are demonstrated between 40 and 70°C and from a pH of 7–11. The observed half‐lives are 103 min at 70°C and 288 min at 40°C. Compared to other published acid and alkaline pectate lyases, Pel‐BL11 demonstrated exceptional thermostability and wider pH adaptability. Temperature effects on the cleavage of the pectate α‐1,4‐glycosidic bond by Pel‐BL11 were examined. Continuous cleavage occurred for the first 3 h at 30 and 50°C. However, at 70°C, the majority of the cleavage occurred during the first 10 min. Weight loss in gampi and paper mulberry fibres after enzyme treatment validate the potential of this treatment in fibre degumming.  相似文献

Serum Clara cell protein (CC16) and surfactant-associated protein D (SP-D) were measured in 161 workers exposed to sulphur dioxide (SO2) in a non-ferrous smelter. Seventy workers from a blanket manufacture served as referents. Exposure to SO2 and tobacco smoking were associated with a decrease of CC16 and an increase of SP-D in serum. Tobacco smoking and exposure SO2 interacted synergistically to decrease serum CC16 but not to increase serum SP-D. While further illustrating the potential of serum CC16 and SP-D, our study confirms that SO2 can cause airways damage at exposure levels below current occupational exposure limits.  相似文献
