During recording of impulse activity from single nerve fibers of electroreceptors of the ampullae of Lorenzini of skates, we studied the responses to electromagnetic radiation (EMR) at a frequency of 37–55 GHz and an intensity of 1–100 mW/cm2.Exposure of the ampullar canal pore to EMR at an intensity of 1–5 mW/cm2 and a distance of 1–10 mm evoked a transient increase in the frequency of low-threshold receptor activity (current threshold was 0.04–0.2 µA). An increase in EMR intensity by more than 8–10 mW/cm2 produced, together with elevation of receptor activity, an inhibition due to a rise in temperature of 1–3°C in the region exposed. The phase of increase in frequency of activity was absent in high current-threshold receptors (0.3–2.0 µA) when exposed to EMR. The receptors responded to irradiation of the ampullar canal pore at a distance of 15–20 mm by an increase in discharge frequency for 20 min. Direct irradiation of the ampullae of Lorenzini induced only inhibitory responses in receptor cells regardless of their excitability.The results obtained indicate that the sensory receptors of vertebrates are sensitive to EMR. The effect of extra-high frequency electromagnetic radiation (EHF EMR) on the development of organotypical culture of the spinal ganglia of a 9–10 day-old chick embryo was investigated. EMR with a wavelength of 5.6 mm and a rate of flow density <1.0, 4.0, and >100 mW/cm2 was used. The stimulating action of EMR at rate of flow density of 4.0 mW/cm2, manifested in intensification of the growth of neurites of sensory neurons and the proliferation of the peripheral glia, was observed. EHF EMR with a density >100 mW/cm2 exerted inhibitory influence. Summary The GABAergic innervation of the frog stomach was studied by means of an indirect immunohistochemical method. Whole mount preparations were obtained from frog stomachs after the animals had been perfused with a mixture of picric acid, glutaraldehyde and glacial acetic acid. Samples were incubated with an antiserum specific for GABA coupled to BSA with glutaraldehyde. Anti-rabbit IgG-HRP was processed by the two step method (Eckert and Ude 1983).GABA-positive varicose fibers and also nerve cell bodies were revealed within the myenteric plexus. The density of GABA-immunoreactive neurons was not higher than 4–8 cell/cm2, which is approximately 1% of the total nerve cell number in the myenteric plexus.  相似文献

Neuroinflammation is involved in the death of retinal ganglion cells (RGCs) after optic nerve injury. The purpose of this study was to determine whether systemic simvastatin can suppress neuroinflammation in the optic nerve and rescue RGCs after the optic nerve is crushed. Simvastatin or its vehicle was given through an osmotic minipump beginning one week prior to the crushing. Immunohistochemistry and real-time PCR were used to determine the degree of neuroinflammation on day 3 after the crushing. The density of RGCs was determined in Tuj-1 stained retinal flat mounts on day 7. The effect of simvastain on the TNF-α-induced NF-κB activation was determined in cultured optic nerve astrocytes. On day 3, CD68-positive cells, most likely microglia/macrophages, were accumulated at the crushed site. Phosphorylated NF-κB was detected in some astrocytes at the border of the lesion where the immunoreactivity to MCP-1 was intensified. There was an increase in the mRNA levels of the CD68 (11.4-fold), MCP-1 (22.6-fold), ET-1 (2.3-fold), GFAP (1.6-fold), TNF-α (7.0-fold), and iNOS (14.8-fold) genes on day 3. Systemic simvastatin significantly reduced these changes. The mean ± SD number of RGCs was 1816.3±232.6/mm2 (n = 6) in the sham controls which was significantly reduced to 831.4±202.5/mm2 (n = 9) on day 7 after the optic nerve was crushed. This reduction was significantly suppressed to 1169.2±201.3/mm2 (P = 0.01, Scheffe; n = 9) after systemic simvastatin. Simvastatin (1.0 µM) significantly reduced the TNF-α-induced NF-κB activation in cultured optic nerve astrocytes. We conclude that systemic simvastatin can reduce the death of RGCs induced by crushing the optic nerve possibly by suppressing astroglial NF-κB activation.  相似文献

The effects of severing the spinal trigeminal tract and its caudal nucleus on high-threshold jaw-opening reflex elicited by tooth pulp stimulation were investigated during experiments on cats under chloralose-Nembutal anesthesia. Low-threshold jaw-opening reflex produced by stimulating the A--infraorbital nerve at an intensity 2–3 thresholds in relation to the most excitable fibers on this nerve was also observed, as well as suppression of these reflexes induced by central gray matter stimulation. It was found that spinal trigeminal tract section produces a 8–52% increase in high-threshold reflex. The amplitude of low-threshold reflex either remained unchanged or showed a slight tendency to rise or fall. Brief stimulation of the central gray matter produced a 100% decrease in high-threshold reflex in intact animals compared with a 40–60% decrease after section of the trigeminal tract. Protracted stimulation of the central gray brought about an 80% decline in high-threshold reflex in intact animals as against 25–30% after section. The degree to which brief stimulation of the central gray produced depression of low-threshold stimulation remained unchanged by trigeminal tract section. Protracted stimulation of the central gray matter brought about a 25–50% reduction in low-threshold reflex in intact animals and a reduction of 75% in three animals and 15–20% in four animals. This implied that the caudal nucleus of the spinal trigeminal tract exerts a more substantial influence on the process of high- than low-threshold reflex inhibition when the central gray matter is stimulated.A. Hygrophilic soil animals, like enchytraeids, overwintering in frozen soil are unlikely to base their cold tolerance on supercooling of body fluids. It seems more likely that they will either freeze due to inoculative freezing, or dehydrate and adjust their body fluid melting point to ambient temperature as has been shown for earthworm cocoons and Collembola. In the present study we tested this hypothesis by exposing field-collected adult Fridericia ratzeli from Disko, West Greenland, to freezing temperatures under various moisture regimes. When cooled at –1 °C min–1 under dry conditions F. ratzeli had a mean temperature of crystallisation (Tc) of –5.8 °C. However, when exposed to temperatures above standard Tc for 22 h, at –4 °C, most individuals (90%, n= 30) remained unfrozen. Slow cooling from –1 °C to –6 °C in vials where the air was in equilibrium with the vapour pressure of ice resulted in freezing in about 65% of the individuals. These individuals maintained a normal body water content of 2.7–3.0 mg mg–1 dry weight and had body fluid melting points of about –0.5 °C with little or no change due to freezing. About 35% of the individuals dehydrated drastically to below 1.1 mg mg–1 dry weight at –6 °C, and consequently had lowered their body fluid melting point to ca. –6 °C at this time. Survival was high in both frozen and dehydrated animals at –6 °C, about 60%. Approximately 25% of the animals (both frozen and dehydrated individuals) had elevated glucose concentrations, but the mean glucose concentration was not increased to any great extent in any group due to cold exposure. The desiccating potential of ice was simulated using aqueous NaCl solutions at 0 °C. Water loss and survival in this experiment were in good agreement with results from freezing experiments. The influence of soil moisture on survival and tendency to dehydrate was also evaluated. However, soil moisture ranging between 0.74 g g–1 and 1.15 g g–1 dry soil did not result in any significant differences in survival or frequency of dehydrated animals even though the apparent wetness and structure of the soil was clearly different in these moisture contents.Abbreviations DW dry weight - FW fresh weight - MP melting point - RH relative humidity - Tc crystallisation temperatures - WC water contentCommunicated by I.D. Summary The neuropeptide content of nerve fibers associated with submucosal arteries in the small intestine of guinea pigs was studied in whole-mount preparations using immunohistochemical methods. Tissues were obtained from normal animals or animals in which the small intestine had been extrinsically denervated. In normal animals, submucosal arteries are innervated by extrinsic sensory nerve fibers which contain both substance P and calcitonin gene-related peptide, and by sympathetic noradrenergic nerve fibers. In preparations obtained from animals 5–9 days after denervation, nerve fibers which contained substance P without detectable calcitonin gene-related peptide were associated with a few submucosal arteries. Nerve fibers which contained vasoactive intestinal peptide were also associated with some arteries. By 42–48 days after extrinsic denervation, substance P-containing fibers (without calcitonin gene-related peptide) and vasoactive intestinal peptide-containing fibers were associated with nearly every blood vessel. The extrinsic sympathetic nerve fibers did not regenerate during the course of this study. The nerve fibers associated with submucosal arteries in denervated tissues were not sensitive to capsaicin treatment.The alteration in the innervation of submucosal arterioles that follows extrinsic denervation of the gut may reflect either an increase in the neuropeptide content of the fibers, synthesis of a new peptide, or an increase in the number of fibers as a result of axonal sprouting.  相似文献

The intracarotid injection method has been utilized to examine blood-brain barrier (BBB) glucose transport in normal mice, and after a 2-day fast. In anesthetized mice, cerebral blood flow (CBF) rates were reduced from 0.86 ml·min–1·gm–1 in control to 0.80 ml·min–1·gm–1 in fasted animals (p>0.05). Brain Uptake Indices were significantly (p<0.05) higher in fasted (plasma glucose =4.7 mM) than control (plasma glucose = 6.5 mM) mice, while plasma glucose was significantly lower. The maximal velocity (Vmax) for glucose transport was 1562±303 nmoles·min–1·g–1, and the half-saturation constant (Km =) 6.67±1.46 mM in normally fed mice. In fasted mice the Vmax was 2053±393 nmoles·min–1·g–1 (p>0.05), and the half-saturation constant (Km =) 7.30±1.60 mM (not significant, P>0.05). A rabbit polyclonal antiserum to a synthetic peptide encoding the 13 C-terminal amino acids of the human erythrocyte glucose transporter (GLUT-1) immunocytochemically confirmed that the mouse brain capillary endothelial glucose transporter is a GLUT-1 transporter, and immunoreactivity was similar in brain endothelia from fed and fasted animals. In conclusion, after a 2-day fast in the mouse, we saw significant reductions in forebrain weight (7%), and plasma glucose levels (27%). Increased brain glucose extraction (25%, p<0.05), and a 22% increase in the unsaturatedpermeability-surface area product (p<0.05) was also observed.  相似文献

The problem of resonance effects of electromagnetic radiation (EMR) on biological objects remained unsolved till now. Previously we demonstrated that low-intensity amplitude-modulated EMR of extremely high frequencies (EHF) modified the activity of mouse neutrophils in the synergistic reaction of calcium ionophore A23187 and phorbol ester PMA. The EHF EMR influence on the neutrophils was significant at the carrier frequencies of radiation within a narrow range of 41.8–42.05 GHz and at the modulation frequency of 1 Hz. The purpose of the work was the analysis of frequency-dependent modification of intracellular free calcium concentration ([Ca2+]i) by modulated EHF EMR on the basis of a special model for [Ca2+]i oscillations in the neutrophils. The calcium channels of plasma membrane were chosen as the action target of external modulation in the model. The computer simulation demonstrated the rise in [Ca2+]i at the influence of the external field with a threshold dependence on the modulation amplitude. The effect depended heavily on a sequence of delivery of the chemical and electromagnetic stimuli. The narrow-band rise in [Ca2+]i had a phase-frequency dependence. With the modulation amplitudes exceeding the threshold value, the rise in [Ca2+]i of more than 50% of the initial level was observed at the frequency of about 1 Hz and in the phase range of 0.3–2.5 radians. The results of the model analysis are in good correspondence with the experimental data obtained before, namely, with the resonance modification of the neutrophil activity at the modulation frequency of 1 Hz and with the presence of the effect only at high concentrations of calcium ionophore.  相似文献

The effect of exogenous acetylcholine (ACh) on potassium currents in the motor nerve ending (NE) has been studied in neuromuscular preparations of the frog cutaneous-sternal muscle by extracellular recording of evoked electrical potentials from the NE. The investigation was performed during inhibition of acetylcholinesterase (AChE) activity by specific inhibitors and AChE removal from the synaptic cleft by collagenase. After AChE inhibition by either armine or proserine, or after treatment of the preparation with collagenase, no effect of exogenous ACh in concentrations of 1·10–4–6·–4 mole/liter was observed, in contrast to results from preparations with intact AChE. However, under the same conditions, as in the case of active AChE, ACh in concentrations of 7·10–4–2·10–3 mole/liter inhibited Ca-activated potassium current of the NE membrane. Experiments with dipyroxim, a synaptic AChE reactivator, have shown that the ACh effect on the potential-dependent potassium current is mediated by specific AChE. Summary The efflux of sodium ions from the loggerhead turtle,Caretta caretta, was investigated when animals were acclimated to seawater, freshwater, and during the acclimation period after transfer from seawater to freshwater. The rate of sodium loss in animals acclimated to seawater was found to be 3 M Na · g–1 · hr–1. Cannulation of the cloaca showed that only 5% of the gross efflux of sodium was via the cloaca and it was calculated that 60% of the gross efflux was via nasal gland secretion and 35 % via integumentary diffusional loss. Cannulation experiments indicated that a significant amount of sodium may enter the body via the cloaca. Transfer of animals from seawater to freshwater resulted in a decline of sodium efflux by 90–99 % within 6 hr, and a further decline to only approximately 0 01 M Na · g–1· hr–1 after 1–2 days in freshwater. Cannulation experiments of individuals acclimated to freshwater indicated that the role of the cloaca in sodium loss in freshwater was minimal. It was calculated that sodium loss in freshwater is so small that survival in freshwater for at least 20 days is possible without active extraction of sodium from the medium. Summary The effect of cultivation (ploughing followed by rotavation) on the mineralization of soil nitrogen was measured at 2 sites on a silt loam soil. Both sites had a predominantly arable cropping history but one had been under grass for the previous 2 years and the other had carried wheat. Mineralization of N was slightly faster in cultivated soil but the difference was only significant at the site previously under grass. At this site cultivated soil contained 7 kg ha–1 more mineral N than uncultivated soil 2 weeks after treatment, and 9 kg ha–1 after 6 weeks. The corresponding figures for the site that had grown wheat were 4 and 6 kg N ha–1.  相似文献

Results of electron microscopy and electrophysiological studies of the frog optic nerve are presented. The nerve contains 96% unmyelinated (about 210,000) and 4% myelinated (8700–14,800) fibers. The peripheral zone of the nerve (20–30 µm) has relatively few myelinated fibers, whereas in other zones these fibers are distributed uniformly (counting area 300–400 µm2). The curve of the distribution of the diameters of myelinated fibers has a number of peaks: a main peak at 1 µm and additional peaks at 0.6 and 1.6 µm (the latter is more prominent). Individual fibers have a diameter of 0.4–3.9 µm. The diameter of the unmyelinated fibers are 0.1–0.4 µm; 64% of these fibers have a diameter of 0.2 µm. Most fibers at a temperature of 18–20° conduct at 0.3–0.4 m/sec and a few (myelinated with a diameter of 1.0–1.6 µm) at 3 and 6 m/sec. After enucleation the myelinated fibers degenerate at first and are phagocytized by neuroglia; the ultrastructure and function of the unmyelinated fibers at 18–20° remain unchanged up to 100 days postoperation.A. Electromagnetic radiation (EMR) of cellular phones may affect biological systems by increasing free radicals and changing the antioxidant defense systems of tissues, eventually leading to oxidative stress. Green tea has recently attracted significant attention due to its health benefits in a variety of disorders, ranging from cancer to weight loss. Thus, the aim of the present study was to investigate the effect of EMR (frequency 900 MHz modulated at 217 Hz, power density 0.02 mW/cm2, SAR 1.245 W/kg) on different oxidative stress parameters in the hippocampus and striatum of adult rats. This study also extends to evaluate the therapeutic effect of green tea mega EGCG on the previous parameters in animals exposed to EMR after and during EMR exposure. The experimental animals were divided into four groups: EMR-exposed animals, animals treated with green tea mega EGCG after 2 months of EMR exposure, animals treated with green tea mega EGCG during EMR exposure and control animals. EMR exposure resulted in oxidative stress in the hippocampus and striatum as evident from the disturbances in oxidant and antioxidant parameters. Co-administration of green tea mega EGCG at the beginning of EMR exposure for 2 and 3 months had more beneficial effect against EMR-induced oxidative stress than oral administration of green tea mega EGCG after 2 months of exposure. This recommends the use of green tea before any stressor to attenuate the state of oxidative stress and stimulate the antioxidant mechanism of the brain.  相似文献

Mycological analyses of 855 samples of stored grains and dried meat collected in period 1980–1987 from individual households in the nephropathic and wider non-nephropathic area in SR Croatia in Yugoslavia showed 10% of samples to be contaminated with Aspergillus ochraceus. Ability to produce ochratoxin A (OA) was tested in 70 samples (27 from nephropathic areas and 43 from non-nephropathic areas). The detection was carried out under UV-light (365 nm) (light blue fluorescence) and 6 OA-producers were found.A biosynthetic procedure on liquid nutritional substrate with sacharose and yeast extract as well as a method using wet crushed wheat revealed that 37% of the samples from a nephropathic area, and 35% of the samples from a non-nephropathic area produce OA.In the nephropathic area 1/10 strains was a strong producer of OA (concentration crushed wheat 135 mg/kg, and 240 mg/l on YES liquid substrate), 1/10 strains was a moderate producer (concentration 16.6 mg/l and 0.07–7.0 mg/l and 0.1–10.4 mg/kg).Among the strains isolated from a wider non-nephropathic area no strong producers of OA were found, but 2/15 strains were moderate producers of OA (concentration of OA 20.4–27.0 mg/l and 15.0–33.7 mg/kg). The other strains, 8/10 on the crushed wheat and 13/15 on the liquid substrate, were weak producers of OA with concentrations of OA between 0.2–9.0 mg/l and 0.2–10.0 mg/kg with the two methods respectively.  相似文献

The respective effects of aldosterone and arginine vasopressin (AVP) were examined on the number of active Na+-K+-ATPase and their pumping activity in nonperfused microdissected mouse cortical collecting tubules (CCD) by measuring specific 3H-ouabain binding and ouabain-sensitive 86Rb uptake. In adrenalectomized (ADX) animals, incubation of CCD with AVP (10–8 m for 5 min) had no effect on the number of pumps. In contrast, in ADX animals replete with aldosterone, AVP induced a 40% increase in the number of pumps. This was accompanied by a 60–65% increase in ouabain-sensitive Rb uptake. AVP effect was dose-dependent (10–10–10–8 m) and was reproduced by dDAVP, forskolin and 8-Br cAMP, indicating a V2 pathway. It was inhibited by amiloride 10–5 m, and did not occur in CCD incubated in hyperosmotic solution, suggesting that the signal was transmitted via apical sodium entry and cell swelling. Finally, the AVP-dependent increase in the number of pumps was rapid (within 5 min) and transient (<25 min).These results demonstrate that, in the CCD, aldosterone and AVP act synergistically to increase not only the apical sodium entry but also the basolateral Na+-K+-ATPase transport capacity: AVP allows a rapid recruitment and/or activation of an aldosterone-dependent pool of latent Na+-K+-ATPase.  相似文献

The effects of applying 4-aminopyridine (10–2 M), aminooxyacetic acid (AOAA — 10–4–10–3 M), -alanine (10–3–10–2 M), and bicuculline (10–5, 10–4 M) to the intact frog olfactory bulb were investigated. Having measured inhibition of orthodromic potential postsynaptic components produced either by a puff of air on the olfactory mucosa (OB input inhibition) or by single electrical stimulation of the olfactory nerve (postsynaptic inhibition) or by single electrical stimulation of the olfactory nerve (postsynaptic inhibition), it was found that 4-aminopyridine greatly intensified postsynaptic inhibition but strongly reduced that of OB input; inhibition of the latter was raised by AOAA or bicuculline and decreased by -alanine. These substances failed to exert any consistent, clear-cut effects on postsynaptic inhibition. Findings would support the hypothesis that OB input inhibition produced by a puff of air on the olfactory mucosa could occur as a result of GABA release from glial cells and subsequent binding of GABA to presynaptic GABAB-receptors in glomeruli.M. We have previously reported that the isolated frog corneal epithelium (a Cl-secreting epithelium) has a large diffusional water permeability (Pdw 1.8×10–4 cm/s). We now report that the presence of Cl in the apical-side bathing solution increases the diffusional water flux, Jdw (in both directions) by 63% from 11.3 to 18.4 l min–1 · cm–2 with 60 mm [Cl] exerting the maximum effect. The presence of Cl in the basolateral-side bathing solution had no effect on the water flux. In Cl-free solutions amphotericin B increased Jdw by 29% but only by 3% in Cl-rich apical-side bathing solution, suggesting that in Cl-rich apical side bathing solution, the apical barrier is no longer rate limiting. Apical Br (75 mm) also increased Jdw by 68%. The effect of Cl on Jdw was observed within 1 min after its addition to the apicalside bathing solution. HgCl2 (0.5 mm) reduced the Cl-increased Pdw by 31%. The osmotic permeability (Pf) was also measured under an osmotic gradient yielding values of 0.34 and 2.88 (x 10–3 cm/s) in Cl-free and Cl-rich apical-side bathing solutions respectively. It seems that apical Cl, or Cl secretion into the apical bath could activate normally present but inactive water channels. Factors influencing the species composition, distribution and abundance of benthic invertebrates were determined in a eutrophic subarctic lake from April 1978 to April 1979. Collections were made at five stations located at depths of 4 to 13 m. The largest populations of up to 5 × 103 animals m–2 were found in the deepest part of the lake. of the 24 species recorded in this area, the chironomidsProcladius denticulatus, Dicrotendipes modestus, Chironomus decorus andGlyptotendipes barbipes were most common. The strong development of benthos in the profundal zone was attributed to a consistently large supply of food and warm (4 °C) winter temperatures on bottom. Slightly smaller populations (up to 4 × 103 animals, m–2), composed of 19–23 species, occurred in shallower water, a reflection of lower (1.5 °C) winter temperatures. In the anoxic northern part of the lake, only 4–8 species were found in low numbers (400–1 000 animals m–2). This was likely due to low (<5% saturation) oxygen levels in water and high organic content (18.5%) of the sediments.  相似文献
