Excised barley roots accumulated 40 to 50% more K(+) from 0.04 mm than from 0.06 mm KCl when incubated for 24 hours in KCl solutions containing 0.2 mm CaSO(4). This phenomenon was not markedly influenced by the rate of absorption of the counteranion. The presence of Na(+) in the treatment solutions decreased total K accumulation but did not alter the K(+) concentration at which the accumulation peak occurred. Short interval studies indicated that this phenomenon is easily observable after 4 hours and begins to become apparent within 2 hours. In comparison with barley, accumulation of K(+) by excised wheat roots decreased as KCl concentration was increased from 0.02 to 0.06 mm; but K(+) accumulation curve for corn roots showed no peaks or depressions in the concentration range of 0.01 to 0.1 mm. A normal hyperbolic curve was noted for the accumulation of Na(+) from 0.01 to 1 mm NaCl by barley roots.  相似文献

Fusarium oxysporum grown in a low phosphate medium was found to take up several times as much K from KH2PO4 as from KCI solutions. Large amounts of phosphate also were taken up from KH2PO4. Similar large uptakes of Na and phosphate took place from solutions of NaH2PO4. Substantial quanties of phosphate were taken up from solutions of Ca(H2PO4)2 in the absence of any appreciable Ca uptake. When the fungus was grown in a medium containing high phosphate, little or no uptake of phosphate from KH2PO4 solutions occured and the K Uptake was at the same level as from KCI solutions. During large phosphate uptake sizable reductions in the organic acid content of the fungal cells were observed. Much, but not all, of the data could be explained on the basis of maintenance of charge balance within the cells. – The respiratory rate of fungus, grown in a low P medium, was markedly increased in KH2PO4 solution. Fungus, grown in a medium with high phosphate, had a higher respiratory rate which showed only a slight response to KH2PO4 solution. The effect of addition of Ca salts on accumulation of K from 5 mN KCl or K2SO4 solutions was found to depend on whether Ca was added as Cl or SO4 salt. Chloride as well as K uptake was increased when Ca and Cl concentrations in culture solutions were increased. Pre-treatment of roots with CaCl2 stimulated subsequent K uptake from K2SO4 solutions as compared to pre-treatment with distilled water but pre-treatment with CaSO4 did not. The results indicate that addition of Ca salts to KCl or K2SO4 solutions increased anion uptake and the effect of the addition of the Ca salts on K uptake was in part the result of increased anion uptake and not entirely a direct effect of Ca.  相似文献

The mechanism of iron transport into erythroid cells was investigated using rabbit reticulocytes and mature erythrocytes incubated with 59Fe-labelled Fe(II) in isotonic sucrose or in solutions in which the sucrose was replaced with varying amounts of isotonic NaCl or KCl. Iron uptake was inhibited at all concentrations of NaCl, in a concentration-dependent manner, but with KCl inhibition occurred only at concentrations up to 10 mM. Higher KCl concentrations stimulated iron uptake to the cytosol of the cells, but inhibited its incorporation into heme. This effect became more marked as the iron concentration was raised. It was found that KCl inhibits iron incorporation into heme and stimulates iron uptake by mature erythrocytes, as well as by reticulocytes. It is concluded that erythroid cells can take up nontransferrin-bound Fe(II) by two mechanisms. One is a high-affinity mechanism that is limited to reticulocytes, saturates at a low iron concentration, and is inhibited by metabolic inhibitors. The other is a low-affinity process that is found in both reticulocytes and erythrocytes, becomes more prominent at higher iron concentrations, and is stimulated by KCl, as well as RbCl, LiCl, CsCl, and choline Cl. The KCl stimulation is inhibited by amiloride, but not by metabolic inhibitors, and its operation is not dependent on changes in cell volume or membrane potential, but it does require the presence of a permeant extracellular anion. Iron uptake by this process appears to occur by facilitated transport and is possibly assoicated with exchange of Na+. A further aspect of this study was a comparison of iron uptake by reticulocytes from Fe(II)-sucrose and Fe(II)-ascorbate using a variety of incubation conditions. The addition of 1 micromolar 2,3,5-triiodobenzoic acid (TIBA) to solutions containing KCl resulted in the inhibition of K and Cl uptake in excised barley roots. The effectiveness of TIBA as an inhibitor increased as the pH of the treatment solution decreased and approached the pKa of TIBA. A lag period of approximately 20 minutes existed prior to the onset of TIBA induced inhibition of ion uptake. Respiratory activity was also inhibited by TIBA. The data suggest that in this material, TIBA functions by entering the cytoplasm and inhibiting metabolism. Comparisons made on the effect of added Ca, showed that at pH 5.7 and higher, Ca had no effect on ion uptake whereas at lower pH values the presence of Ca enhanced uptake by offsetting the deleterious effects of H+.  相似文献

The basic objective of this work was to study the effect of model cationic drug metformin HCl on swelling and erosion and, in turn, the release of KCl and drug itself, from the κ-carrageenan matrices. Water uptake by the matrix up to 2 hours was found to increase with KCl concentration from the plain matrix. Erosion was not affected by concentration of KCl. Incorporation of drug favors water uptake, but in presence of KCl it was found to be reduced. Drugcontaining matrices have shown higher release of KCl as compared with plain batches. Drug release was retarded as KCl concentration increased up to 5%, above which the reduced cohesivity of the matrix caused increase in drug release.  相似文献

Nectria sp.JZ6是从浙贝母新鲜鳞茎中首次分离到的一株内生真菌,其发酵液具有抑菌活性。为建立该真菌稳定分泌抑菌活性成分的发酵体系,本研究利用单因素实验和正交实验确定了发酵培养基的配方并初步优化了发酵条件。结果表明,将活化后的菌种接种于改良查氏培养基(8%葡萄糖,0.5%蛋白胨,0.05%KCl,0.1%K2HPO4,0.15%MgSO4,pH6.5)中,28℃、150r/min振荡培养6d后获得的发酵液抑菌活性比优化前提高了26%。该发酵液经100℃水浴30min不失活,在pH1-5时抑菌活性相对稳定,其后逐渐减弱,并在pH9.0及以上时丧失。Nectriasp.JZ6发酵液的乙酸乙酯浸膏抑菌活性最强,对金黄色葡萄球菌、藤黄八叠球菌、枯草芽孢杆菌、绿脓杆菌和大肠杆菌的最低抑菌浓度(MIC)分别为0.625、0.625、1.25、1.25和1.25mg/mL。  相似文献

Summary The rate of potassium uptake by two varieties of burley tobacco (Nicotiana tabacum) differed by as much as two-fold when excised roots were absorbing K from solutions of 2.5 × 10–4 M KCl. With increasing substrate levels of KCl the difference between varieties decreased and no difference was observed at a KCl concentration of 1.6 × 10–2 M. The K-absorption by excised roots of several varieties from solutions of 5 × 10–4 M KCl was compared.Contribution of the Department of Agronomy, Kentucky Agr. Exp. The influence of NO3 uptake and reduction on ionic balance in barley seedlings (Hordeum vulgare, cv. Compana) was studied. KNO3 and KCl treatment solutions were used for comparison of cation and anion uptake. The rate of Cl uptake was more rapid than the rate of NO3 uptake during the first 2 to 4 hours of treatment. There was an acceleration in rate of NO3 uptake after 4 hours resulting in a sustained rate of NO3 uptake which exceeded the rate of Cl uptake. The initial (2 to 4 hours) rate of K+ uptake appeared to be independent of the rate of anion uptake. After 4 hours the rate of K+ uptake was greater with the KNO3 treatment than with the KCl treatment, and the solution pH, cell sap pH, and organic acid levels with KNO3 increased, relative to those with the KCl treatment. When absorption experiments were conducted in darkness, K+ uptake from KNO3 did not exceed K+ uptake from KCl. We suggest that the greater uptake and accumulation of K+ in NO3-treated plants resulted from (a) a more rapid, sustained uptake and transport of NO3 providing a mobile counteranion for K+ transport, and (b) the synthesis of organic acids in response to NO3 reduction increasing the capacity for K+ accumulation by providing a source of nondiffusible organic anions.  相似文献

Plectonema boryanum requires approximately 5 days of exposure to a culture medium lacking phosphorus to induce the "polyphosphate overplus" phenomenon. At pH9, phosphate uptake is greatest both from normal culture medium and from dilute salt solutions. Phosphate uptake from dilute salt solutions was greatest when Na+ or K+ are combined with Ca2+ or Ca2+ and Mg2+. Cells starved of phosphorus in the presence of a high concentration of K+ or Ca2+ in the medium, and then allowed to take up phosphorus under the same conditions, assimilate more phosphorus than with other major ions.  相似文献

Pathotoxin preparations were obtained from either axenic culture filtrate of race T of Bipolaris maydis (Nisikado) Shoemaker (new culture media and toxin purification procedures are described) or extracts of maize leaves infected with the fungus. The toxins (10(-6) to 10(-8)m) caused inhibition of [(86)Rb]K(+) uptake in leaf discs and apical root segments of Zea mays L. cv W64A Texas (Tcms) and normal (N) cytoplasms. Significant inhibition was measurable as early as 5 min after adding toxin. In Tcms per cent inhibition was increased by increasing toxin concentration and time in toxin, by using solution at pH 5 rather than pH 7, by decreasing external KCl concentration over the range 50 to 0.1 mm (in the presence of 0.5 mm CaSO(4)), or by exposing leaf discs to light rather than dark during the uptake period in toxin. Root uptake of (22)Na(+) and (36)Cl(-) was inhibited to a lesser extent than K(+). Inhibition of (32)PO(4) (3-) uptake occurred after 40 min when cyclosis had ceased.When combined with data in the literature, our data indicate that the plasmalemma is the probable primary site of toxin action in N and Tcms maize. Comparison of the effects of toxin on K(+) uptake in N and Tcms maize suggests the existence of more than one mode of toxin action: a weak disruptive effect in N and Tcms, and in addition, specific membrane sites in Tcms involved in monovalent ion uptake.Six genotypes in N or Tcms cytoplasm which exhibited different degrees of disease susceptibility in the field showed a corresponding gradation of susceptibility to the toxin when a K(+) uptake bioassay was used. This correlation is strong evidence that the sites of toxin action affecting K(+) transport have characteristics closely related to cellular factors regulating susceptibility to fungal attack.  相似文献

Freshly harvested mycelium of the filamentous ascomycete Neocosmospora vasinfecta accumulated C1- against a concentration gradient by a process probably requiring the expenditure of metabolic energy. When mycelium, washed free of growth medium, was incubated in deionized water or tris (hydroxymethyl) aminomethane sulfate at pH 7.5 for 4 h and then transferred to K36C1 solutions, the C1- uptake rate was, on the average, 3.77 +/- 0.26 (+/-SE, N = 20) times the uptake rate exhibited by freshly harvested mycelium. This development of an increased rate of Cl- uptake could be blocked by the presence of an inhibitor of ribonucleic acid synthesis (azaguanine) or of protein synthesis (cycloheximide, fluorophenylalanine, or puromycin). The combined presence of glucose and a potassium salt in the preincubation solution virtually arrested the development of enhanced Cl- uptake. The rate of Cl- uptake by freshly harvested mycelium did not vary greatly with the age of the culture on harvest but the ability to develop an increased rate declined with age. The fact that it is possible to obtain mycelium possessing widely different capacities for Cl- uptake should assist in biochemical characterization of the Cl- uptake system.  相似文献

Growth retardation, measured as internode shortening, was observedin bean and tomato plants when grown in culture solutions towhich Rb had been added. The Rb effect was more pronounced inthe presence of low (0.5 meq/liter) K than high (8 meq/liter)K. Gibberellic acid, added to the solutions, ameliorated thegrowth retardation due to Rb. Scutella separated from grains of Himalaya barley after germination for 3 days rapidly took up l-leucine from aerated incubation media; with 1 millimolar leucine the rate varied between 4 and 14 micromoles per gram per hour and the pH optimum was at 3.5 to 5, both depending on buffer composition and prewashing time. The rate of the uptake increased with increasing concentration of leucine in a complex manner, which could be interpreted as multiphasic kinetics with apparent K(m) values of 3.4 and 15.5 millimolar below and above 3 millimolar leucine, respectively. The uptake took place against a concentration difference (highest estimated ratio 270: 1) and was strongly inhibited by dinitrophenol. Uptake was apparently due to active transport requiring metabolic energy.The development of the uptake activity during germination was studied using Pirkka barley. A low activity was present in the scutella of ungerminated grains. It began to increase after 6 hours imbibition, and the increase was biphasic, the major changes occurring during days 0 to 3 and 4 to 6. The total increase was about 20-fold.The regulation of the development was studied by allowing separated embryos to germinate on agar gel. The increase of uptake activity was strongly inhibited by inhibitors of RNA or protein synthesis. Increase did not require the presence of the embryo proper, and was not affected by gibberellic or abscisic acid. Removal of the endosperm greatly accelerated the increase of uptake activity, and the presence of 5 or 20 millimolar glutamine counteracted the removal of the endosperm. The results suggest that the availability of glutamine or amino acids in general in the endosperm may regulate the development or the activity of the transport system.  相似文献

In this study, evidence is given that a number of isolated coupled plant mitochondria (from durum wheat, bread wheat, spelt, rye, barley, potato, and spinach) can take up externally added K(+) ions. This was observed by following mitochondrial swelling in isotonic KCl solutions and was confirmed by a novel method in which the membrane potential decrease due to externally added K(+) is measured fluorimetrically by using safranine. A detailed investigation of K(+) uptake by durum wheat mitochondria shows hyperbolic dependence on the ion concentration and specificity. K(+) uptake electrogenicity and the non-competitive inhibition due to either ATP or NADH are also shown. In the whole, the experimental findings reported in this paper demonstrate the existence of the mitochondrial K(+)(ATP) channel in plants (PmitoK(ATP)). Interestingly, Mg(2+) and glyburide, which can inhibit mammalian K(+) channel, have no effect on PmitoK(ATP). In the presence of the superoxide anion producing system (xanthine plus xanthine oxidase), PmitoK(ATP) activation was found. Moreover, an inverse relationship was found between channel activity and mitochondrial superoxide anion formation, as measured via epinephrine photometric assay. These findings strongly suggest that mitochondrial K(+) uptake could be involved in plant defense mechanism against oxidative stress due to reactive oxygen species generation.  相似文献

Evidence for a transport system for glycolate in Chlamydomonas was obtained. [14C]Glycolate was taken up rapidly, reaching an equilibrium in less than 2 s at 4 degrees C. Glycolate uptake was stimulated by valinomycin and high KCl or high KCl alone and inhibited by N-ethylmaleimide. This uptake was not dependent on temperature or pH in contrast to uptake of benzoate by diffusion which decreased by orders of magnitude with increasing external pH. Based on these data, a transporter for glycolate is proposed.  相似文献

Summary Pseudomonas sp. EPS-5028 was examined for the ability to accumulate uranium from solutions. The uptake of uranium by this microorganism is very rapid and is affected by pH but not by temperature, metabolic inhibitors, culture time and the presence of various cations and anions. The amount of uranium absorbed by the cells increased as the uranium concentration of the solution increased up to 55 mg uranium/g cell dry weight. Electron microscopy indicated that uranium accumulated intracellularly as needle-like fibrils. Uranium could be removed chemically from the cells, which could then be reused as a biosorbent. Ursodeoxycholic acid dissolves cholesterol gallstones in humans. In the present study optimum conditions for ursodeoxycholic acid production by Fusarium equiseti M41 were studied. Resting mycelia of F. equiseti M41 showed maximum conversion at 28 degrees C, pH 8.0, and dissolved oxygen tension of higher than 60% saturation. Monovalent cations, such as Na+, K+, and Rb+, stimulated the conversion rate more than twofold. In the presence of 0.5 M KCl, the initial uptake rate and equilibrium concentration of lithocholic acid (substrate) were enhanced by 5.7- and 1.7-fold, respectively. We confirmed that enzyme activity catalyzing 7 beta-hydroxylation of lithocholic acid was induced by substrate lithocholic acid. The activity in the mycelium was controlled by dissolved oxygen tension during cultivation: with a dissolved oxygen tension of 15% and over, the activity peak appeared at 25 h of cultivation, whereas the peak was delayed to 34 and 50 h with 5 and 0% dissolved oxygen tension, respectively. After reaching the maximum, the 7 beta-hydroxylation activity in the mycelium declined rapidly at pH 7.0, but the decline was retarded by increasing the pH to 8.0. Several combinations of operations, such as pH shift (from pH 7 to 8), addition of 0.5 M KCl, and dissolved oxygen control, were applied to the production of ursodeoxycholic acid in a jar fermentor, and a much larger amount of ursodeoxycholic acid (1.2 g/liter) was produced within 96 h of cultivation.  相似文献
