The cytoprotective effect of a highly potent somatostatin (SRIF) analog, RC-121 (H-D-Phe-Cys-Tyr-D-Trp-Lys-Val-Cys-Thr-NH2), was examined in the absolute ethanol-induced gastric erosion model in rat. This analog diminished the degree of gastric erosion by 50-55% when administered in i.p. doses of 2 x 10(-10)-10(-8) g/100 g body weight, or in oral doses of 10(-8)-2 x 10(-7) g/100 body weight. The orally active, highly potent SRIF analogs may be useful as therapeutic agents in the treatment of human peptic ulcer.  相似文献

Subcutaneous administration of cholecystokinin-8 (CCK-8, 10-100 micrograms/kg) reduces in a dose-dependent manner gastric lesions induced by 96% ethanol in rats, and CCK-4, CCK-7, and the CCK-8 nonsulfated form (all up to 100 micrograms/kg sc) were inactive. The presence of the entire molecule and sulfation of the tyrosine in position 2 are necessary for the mucosal protective properties of CCK-8 against 96% ethanol-induced gastric lesions. These effects are probably at least in part, due to a sulfhydryl-sensitive process.  相似文献

The effect of copper(II) complexes on glucose metabolism was studied in normal and streptozotocin-induced diabetic rats. The copper(II) complexes used were bis(acetato)tetrakis(imidazole) copper (II), [Cu(OAc)2(Im)4], bis(acetato)bis(2-methylimidazole) copper(II), [Cu(OAc)2(1,2dmIm)2], and bis)acetato)bis(μ-acetato)tetrakis(N-methylimidazole) copper(II) hexaaquo, [Cu2(OAc)4-(NmIm)4]·6H2O. Intramuscular administration of various doses of Cu(OAc)2(Im)4 ranging from 10 to 100 mg/kg body mass to overnight fasted rats decreased blood glucose levels in a dose-dependent manner. Maximum hypoglycemic effect was observed 3 h after administration and lasted for at least 6 h. Treatment with 100 mg/kg body mass of Cu(OAc)2(Im)4 caused hypoglycemic shock, which was irreversible and even lethal. Blood insulin levels were reduced sharply during this hypoglycemic shock. Similar changes in blood glucose level were achieved using Cu(OAc)2)2mIm)2. The same pattern of hypoglycemia, although less pronouned, was observed for Cu2(OAc)4(NmIm)4·6H2O and Cu (OAc)2(1,2dmIm)2. Binary copper(II) acetate complex, the ligand imidazole, and the inorganic form of copper, such as copper(II) chloride, had no significant effect on blood glucose level. These results indicate that the hypoglycemic activity of these complexes varies with the imidazole ligand and structure of the complex. Intramuscular administration of Cu(OAc)2(Im)4 to diabetic rats caused a reduction in blood glucose levels and improved their tolerance for glucose.  相似文献

In this study, we investigated the protective effects of capsaicin on gastric mucosal oxidative damage induced by ethanol. Sprague Dawley rats intragastrically received 0.5-10 mg/kg, BW capsaicin or vehicle; 30 min later gastric lesions were induced by intragastric administration of absolute ethanol. Lipid peroxidation was estimated by measuring thiobarbituric acid reactive substances in gastric mucosa. Myeloperoxidase activity, a marker enzyme of polymorphonuclear leukocytes for tissue inflammation, was also measured in the gastric mucosa. The expression level of cyclooxygenase-2, which increases in inflammatory region, was determined by Western blot analysis. Capsaicin significantly suppressed gastric haemorrhagic erosions induced by ethanol. Capsaicin inhibited lipid peroxidation and myeloperoxidase activity in ethanol-induced gastric mucosal lesion in a dose-dependent manner. Capsaicin also inhibited the expression of cyclooxygenase-2 in the gastric mucosal lesion. The gastroprotective activity of capsaicin on the ethanol-induced oxidative damage may be important for chemoprevention.  相似文献

Copper(II) complexes with thiosemicarbazones have been shown to be more active in cell destruction, in the inhibition of DNA synthesis than the uncomplexed ligand. Several derivatives of thiosemicarbazones and their iron and copper complexes have been studied for their cytotoxicity and inhibiting activity against DNA synthesis. In the present work complexes formed in H2O-DMSO solution between copper(II) and the acetophenone thiosemicarbazone (ATSC) and the o-aminobenzaldehyde thiosemicarbazone (o-NH2TSC) have been studied. EPR studies have been performed at different pH values and metal-to-ligand ratios. The spectra have been recorded at both room (298 K) and low temperatures (120 K). A possible relationship between structure and activity is attempted on the basis of the EPR data.  相似文献

Cashew nut-shell liquid and the contained anacardic acids (AAs) have been shown to possess antioxidant, lipoxygenase inhibitory, anti-Helicobacter pylori and antitumor properties. Despite these known effects, hitherto there were no published reports on their likely gastroprotective effects. The present study was designed to verify whether AAs afford gastroprotection against the ethanol-induced gastric damage and to examine the underlying mechanism(s). Gastric damage was induced by intragastric administration of 0.2 mL of ethanol (96%). Mice in groups were pretreated orally with AAs (10, 30 and 100 mg/kg), misoprostol (50 μg/kg), or vehicle (2% Tween 80 in saline, 10 mL/kg), 45 min before ethanol administration. They were sacrificed 30 min later, the stomachs excised, and the mucosal lesion area (mm2) measured by planimetry. Gastroprotection was assessed in relation to inhibition of gastric lesion area. To study the gastroprotective mechanism(s), its relations to capsaicin-sensitive fibers, endogenous prostaglandins, nitric oxide and ATP-sensitive potassium channels were analysed. Treatments effects on ethanol-associated oxidative stress markers GSH, MDA, catalase, SOD, and total nitrate/nitrite levels as an index of NO were measured in gastric tissue. Besides, the effects of AAs on gastric secretory volume and total acidity were analysed in 4-h pylorus-ligated rat. AAs afforded a dose-related gastroprotection against the ethanol damage and further prevented the ethanol-induced changes in the levels of GSH, MDA, catalase, SOD and nitrate/nitrite. However, they failed to modify the gastric secretion or the total acidity. It was observed that the gastroprotection by AAs was greatly reduced in animals pretreated with capsazepine, indomethacin, l-NAME or glibenclamide. These results suggest that AAs afford gastroprotection principally through an antioxidant mechanism. Other complementary mechanisms include the activation of capsaicin-sensitive gastric afferents, stimulation of endogenous prostaglandins and nitric oxide, and opening of K+ATP channels. These combined effects are likely to be accompanied by an increase in gastric microcirculation.  相似文献

Copper(II) cations coordinated with PMDTA (pentamethyldiethylenetriamine) and TMEDA (tetramethylethylenediamine) possess a high synthetic potential. The synthesis of these cations was carried out by metathesis reactions with silver salts. The cationic copper(II) complexes, [Cu(PMDTA)(Me2CO)Cl]+, [Cu(PMDTA)(H2O)Cl]+, [Cu(PMDTA)(DMF)]+, [Cu(PMDTA)Cl]+, [Cu(PMDTA)OAc]+, [Cu(PMDTA)(MeCN)2]2+, [Cu2(TMEDA)2Cl3]+ and [Cu(TMEDA)(MeCN)3]2+ were synthesised as PF6 salts, crystallised and characterised by single-crystal X-ray diffraction.  相似文献

The present study determined the participation of different endogenous mediators in adaptive cytoprotection against gastric gland damage caused by ethanol in rabbits. Using the isolated gland preparation, pretreatment with 10−5M of either indomethacin, Nw-nitro-L-arginine methyl ester (L-NAME) or N-ethylmaleimide (NEM), but not of substance P antagonist, intensified the 10% (v/v) ethanol-induced gastric gland damage and lessened the degree of cytoprotection evoked by 2% (v/v) ethanol to a significant level. Co-administration with 10−4M of prostaglandin E2, L-arginine or glutathione to the respective groups completely reversed the above adverse effects. These results demonstrate the involvement of endogenous prostaglandins, nitric oxide and glutathione in gastric adaptive cytoprotection against the damaging action of ethanol in the rabbit gastric glands.  相似文献

The kinetics of substitution reactions of [η-CpFe(CO)3]PF6 with PPh3 in the presence of R-PyOs have been studied. For all the R-PyOs (R = 4-OMe, 4-Me, 3,4-(CH)4, 4-Ph, 3-Me, 2,3-(CH)4, 2,6-Me2, 2-Me), the reactions yeild the same product [η5-CpFe(CO)2PPh3]PF6, according to a second-order rate law that is first order in concentrations of [η5-CpFe(CO)3]PF6 and of R-PyO but zero order in PPh3 concentration. These results, along with the dependence of the reaction rate on the nature of R-PyO, are consistent with an associative mechanism. Activation parameters further support the bimmolecular nature of the reactions: ΔH = 13.4 ± 0.4 kcal mol−1, ΔS = −19.1 ± 1.3 cal k−1 mol−1 for 4-PhPyO; ΔH = 12.3 ± 0.3 kcal mol−1, ΔS = 24.7 ±1.0 cal K−1 mol−1 for 2-MePyO. For the various substituted pyridine N-oxides studied in this paper, the rates of reaction increase with the increasing electron-donating abilities of the substituents on the pyridine ring or N-oxide basicities, but decrease with increasing 17O chemical shifts of the N-oxides. Electronic and steric factors contributing to the reactivity of pyridine N-oxides have been quantitatively assessed.  相似文献

目的:探讨蓝莓提取物对过氧化氢致大鼠海马神经元氧化应激损伤的减缓作用。方法:将培养7d的海马神经细胞分为8组:①过氧化氢组(H2O2):培养液中加入50μmol/L的H2O2,作用24h。②不同剂量蓝莓提取物预处理组(BE+H2O2):在加入H2O2前24h,分别加入0.01、0.1、1.0、10.0、20.0和40.0μg/mlBE。③空白对照组(Control):处理步骤同上组,但每次处理物质均为等量的培养液。通过测定细胞存活率和上清液中乳酸脱氢酶(LDH)的活性确定减轻海马神经元损伤的蓝莓提取物的适宜浓度。并检测细胞内丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性以及细胞凋亡率的变化。结果:①蓝莓提取物组(0.1,1.0和10.0μg/mlBE)LDH活性显著低于H2O2组,细胞存活率由H2O2组的57.44%分别上升至78.42%、87.71%、72.40%;1μg/ml蓝莓提取物组对H2O2诱导的海马神经细胞氧化应激损伤的保护作用最好。②1μg/ml蓝莓提取物组海马神经细胞培养上清液中MDA含量及细胞凋亡率显著低于H2O2组,SOD活性显著高于H2O2组。结论:适宜剂量的蓝莓提取物对氧化应激损伤的海马神经元有一定的保护作用,其机制可能与抑制海马神经元凋亡、增强神经细胞的抗氧化功能有关。  相似文献

A new series of dinuclear squarato-bridged nickel(II) and copper(II) complexes [Ni2(2,3,2-tet)21,3-C4O4)(H2O)2](ClO4)2 (1), [Ni2(aepn)21,3-C4O4)(H2O)2](ClO4)2 (2), [Cu2(pmedien)21,3-C4O4)(H2O)2](ClO4)2.4H2O (3) and [Cu2(DPA)21,2-C4O4)(H2O)2](ClO4)2 (4) where is the dianion of 3,4-dihydroxycyclobut-3-en-1,2-dione (squaric acid), 2,3,2-tet = 1,4,8,11-tetraazaundecane, aepn = N-(2-aminoethyl)-1,3-propanediamine, pmedien = N,N,N′,N″,N″-pentamethyldiethylenetriamine and DPA = di(2-pyridylmethyl)amine were synthesized and structurally characterized by X-ray crystallography. The spectral and structural characterization as well as the magnetic behaviour of these complexes is reported. In this series, structures consist of the groups as counter ions and the bridging the two M(II) centers in a μ-1,3- (1-3) and in a μ-1,2-bis(monodentate) (4) bonding fashions. The coordination geometry around the Ni(II) ions in 1 and 2 is six-coordinate with distorted octahedral environment achieved by N atoms of the amines and by one or two oxygen atoms from coordinated water molecules, respectively. In the Cu(II) complexes 3 and 4, a distorted square pyramidal geometry is achieved by the three N-atoms of the aepn or DPA and by an oxygen atom from a coordinated water molecule. The electronic spectra of the complexes in aqueous solutions are in complete agreement with the assigned X-ray geometry around the M(II) centers. The complexes show weak antiferromagnetic coupling with ∣J∣ = 1.8-4.2 cm−1 in the μ-1,3- bridged squarato compounds 1-3, and J = −16.1 cm−1 in the corresponding μ-1,2- bridged squarato complex 4. The magnetic properties are discussed in relation to the structural data.  相似文献

The preparation and properties of the copper(II) halide complexes CuX2(NIDOL)2 (where X = Cl, Br) are reported for the anti-inflammatory drug nictindole (NIDOL). The diffuse reflectance spectra, magnetic moments and electron spin resonance spectra are consistent with a tetragonally distorted pseudo-octahedral environment around the copper(II) ions. The infrared spectra indicate monodentate coordination of the neutral drug to the central metal ion via the nitrogen atom of the pyridine ring.  相似文献

Redox properties of copper complexes are important for their catalytic functions in vitro and in biological systems, and can contribute to their reactivity toward selected targets. In order to evaluate the influence of different ligands on the reactivity of copper ions, comparative studies were carried out with some copper(II) complexes containing a tridentate imine, or a tetradentate di-Schiff base ligand with a mixed pyridine, pyrazine, or imidazole donor set, acting as catalysts in the oxidation of 2-deoxy-D-ribose. Addition of the reducing agent glutathione (gamma-glutamylcysteinylglycine; GSH), which can also act as a good ligand for copper(I), mediated the oxidation of the substrate. For some of these compounds, a reductive activation followed by competition for the metal ion was verified, with formation of copper(I)-glutathione complex monitored by fluorescence measurements. For others, however, the reduction of the metal by the glutathione seems to not occur. In the presence of hydrogen peroxide, the oxidative damage is significantly enhanced for all the complexes tested. Redox potential measurements by cyclic voltammetry corroborated partially these results, indicating that the most reactive complexes are those with more positive redox potential. Evidence for site-specific attack to 2-deoxy-D-ribose was also observed, consistent with the intermediary formation of a copper-hydroxyl species, [LCu(II)(*OH)], rather than 'free' hydroxyl radical.  相似文献

Cu2+ and Co2+ complexes of adriamycin (ADM) in aqueous solutions have been examined using EPR spectroscopy. An appreciable amount of Cu2+ and Co2+ complexes formed in the solutions were found to be in the EPR silent associated form, where the metal ions are antiferromagnetically coupled. The associated form of the Cu2+ complex may be neither a simple dimer nor coordination polymer but aggregates of a stacked type. Formation of a complex having Cu2+-ADM stoichiometry of 1:2 was observed for the solutions containing excess of ADM as an EPR observable species. The complex having Cu2+-ADM stoichiometry of 1:1 was not observed directly by EPR, but the presence of the complex is undeniable, especially at low pH range so far as large excessive ADM is not present. The Co2+ complex of ADM observed by EPR is in the high-spin (S = 3/2) state and may have a coordination structure of tetragonal symmetry. The EPR spectra of these complexes apparently show that the Cu2+ and Co2+ ions are bound at the carbonyl and phenolate oxygen in the 1,4-dihydroxyanthraquinone moiety and the amino nitrogen in the sugar part does not seem to participate in the coordination to the metal ions.  相似文献

Characteristic resonance Raman spectra are observed on ionization of the phenolic groups in adriamycin. On the basis of these results, vibrational assignments for the Raman bands of adriamycin are reported. Distinct Raman spectra are observed for Cu(II)-adriamycin complexes at pH approximately 5 and pH approximately 13. The data indicate that at lower pH a bis complex of Cu(II) is formed, which transforms to a polymeric Cu(II) chelate at higher pH. Upon interaction of the metal-drug complex with calf thymus DNA at pH approximately 5, a ternary complex is formed in which the Cu(II)-complexed adriamycin is intercalated into DNA.  相似文献

Protonation and Cu(II) coordination of kasugamycin were studied by potentiometry, UV-vis, CD, EPR, 13C NMR, and 1H NMR. Mononuclear complexes with stoichiometries ranging from CuHL to CuH(-1)L were found. The aminoamidine moiety provides the coordination site in the CuHL species. The additional axial coordination of the amino nitrogen of the aminosugar ring is present in CuL. Finally, the CuH(-1)L complex is formed as a result of a deprotonation and coordination of the hydroxyl group of the inositol ring. The non-planar arrangement of the chelate rings results in the relative stabilization of a Cu(I) species. As a consequence, Cu(I) and superoxide radicals are involved in the redox mechanism of H(2)O(2) activation by the Cu(II) complex of kasugamycin.  相似文献

A spectrophotometric study of cobalt(II), nickel(II) and copper(II) thiocyanato complexes was carried out in methanol at 25 degrees C and at a constant ionic strength of 1 M. Under the experimental conditions, two mononuclear complexes are identified with each of the three metal ions. Their stability constants are determined with a recent PC program SIRKO and the calculated values are: for cobalt, log beta 1 = 1.6, log beta 2 = 2.7; for nickel, log beta 1 = 1.8, log beta 2 = 3.0; and for copper, log beta 1 = 3.0, log beta 2 = 3.6.  相似文献
