New Zealand white rabbits were immunized with partly fed Hyalomma dromedarii tick-derived midgut concealed antigens (supernate and pellet fractions) and Freund's complete adjuvant (FCA). The rabbits received three inoculations subcutaneously on days 0, 14 day 21 at a dose rate of 1 mg antigen per animal. The effects of the immunity induced was determined by infesting the rabbits with adult H. dromedarii ticks. In immunized rabbits a significant reduction in tick yield, engorgement weight, oviposition period, egg mass weight and percentage of egg hatchability was found. The gut supernatant antigen fraction induced the best protection in terms of reduced feeding and reproductive performance of the ticks.  相似文献

The effect of methyl jasmonate (MJ) on the water-soluble protein pattern of Ricinus communis leaves was analyzed. Several dynamic changes occurred after a period of 24 and 48h including six proteins (Mr 13,000, 15,000, 16,000, 27,000, 29,000 and 60,000) whose levels increased by 48h and seven others (Mr 11,000, 18,000, 20,000 30,000, 37,000, 40,000 and 58,000) whose levels decreased. Four proteins (Mr 24,000, 34,000, 64,000 and 66,000) were induced after 24h of treatment, but returned to control levels by 48h. On the other hand, the levels of three proteins (Mr 74,000, 84,000 and 88,000) decreased after 24h, but returned to control levels after 48h. One of the proteins that accumulated after 48 h had the 13 first residues sequenced. This polypeptide named MJRC-15, was identical to the C-terminal sequence of Rubisco-large chain polypeptide (position 337–350) from tobacco chloroplast. Western-blot analysis using polyclonal antibodies against Rubisco supports the hypothesis that MJRC-15 is a degradation product of Rubisco.  相似文献

The plasma membrane ATPase of mung bean (Phaseolus mungo L.) roots has been solubilized with a two-step procedure using the anionic detergent, deoxycholate (DOC) and the zwitterionic detergent, zwittergent 3-14 as follows: (a) loosely bound membrane proteins are removed by treatment with 0.1% DOC; (b) The ATPase is solubilized with 0.1% zwittergent in the presence of 1% DOC; (c) the solubilized material is further purified by centrifugation through a glycerol gradient (45-70%). Typically, about 10% of the ATPase activity is recovered, and the specific activity increases about 11-fold. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis shows that the peak fraction from the glycerol gradient contains three major polypeptides of Mr = 105,000, 67,000, and 57,000 daltons. The properties of the purified ATPase are essentially the same as those of membrane-bound ATPase, with respect to pH optimum, substrate specificity, inhibitor sensitivity, and ion stimulation.  相似文献

Promastigotes and amastigotes of Leishmania tropica were surface-radioiodinated using the lactoperoxidase technique. Detergent lysates of the labeled organisms were analyzed by two-dimensional gel electrophoresis. Analysis of radioiodinated promastigote membrane proteins revealed six major and some minor acidic polypeptides. Analysis of the amastigote membrane proteins revealed six major proteins, mostly acidic, and some poorly resolved basic proteins. Four of the major membrane proteins appeared to be common to the two parasitic forms (Mr 67,000, Mr 50,000, Mr 68,000, and Mr 80,000). These polypeptides were recognized by antipromastigote antibodies as well as antibodies from CBA/H mice that had recovered from infection. Peptide mapping confirmed their homology in the two parasite forms. One polypeptide appeared to be specific for the promastigote (Mr 50,000) and two polypeptides appeared to be specific for the amastigote form of the parasite (Mr 94,000 and Mr 43,000).  相似文献

The core structures of microvilli from absorptive cells of the intestinal epithelium are primarily composed of calmodulin (Mr 16,000), actin (Mr 43,000), villin (Mr 95,000) and a protein of Mr 110,000. We have isolated this protein and raised antibodies against it. The antibodies interact specifically with villin and Mr 110,000 polypeptides present in isolated microvilli or brush borders. However, after absorption on an immobilized villin preparation, these antibodies still immunoprecipitate the Mr 110,000 protein but not villin. Thus, these two proteins appear to share some antigenic determinants but also contain other determinants specific for each protein. Immunolocalization studies have been performed using specific antibodies against the Mr 110,000 protein. Immunofluorescent studies on thin frozen sections of intestinal cells show that this protein is located in the brush border and at the basolateral faces of these polarized cells. Immunoferritin studies on rat brush borders demembranated with the detergent Triton X-100 show the association of the Mr 110,000 protein with core filaments of microvilli, as well as with some filaments localized in the terminal web network.Using sealed, right-side-out vesicles prepared from pig intestinal mucosa in the presence of Ca2+ and Mg2+, a polypeptide of Mr 140,000 was found to be a major component of the Triton X-100 insoluble pellet. This protein is a minor component of an equivalent pellet obtained from isolated microvilli prepared in the presence of EDTA. The significance of this Mr 140,000 polypeptide associated with the core residue of intestinal microvilli is discussed.  相似文献

Summary Leaves and stems from endod (Phytolacca dodecandra L'Herit), known to produce the 29 kDa ribosome-inactivating protein (RIP) dodecandrin, were initiated into tissue culture. Callus and suspension cultures were maintained on modified Murashige and Skoog medium plus 1.0 mg/l 2,4-dichlorophenoxyacetic acid. Six callus and two suspension cell lines were screened for dodecandrin production by western blots with affinitypurified antiserum. Antiribosomal activity of culture extracts was tested by in vitro translation assays. One suspension cell line was found to be free of immunoreactive proteins and a ribosome inhibitor. All other cell lines contain a ribosome inhibitor, although only two callus cell lines show detectable amounts of immunoreactive proteins at the same Mr as dodecandrin. Other immuno-reactive proteins were detected in callus (Mr 31000, 33000, 41000 and 43000) and in suspension cells (Mr 23000 and 43000), and may be ribosome inhibitors related to dodecandrin—either other RIPs or dodecandrin at various stages of processing.  相似文献

We determined whether ambient temperature influences the proteins produced by Ixodes dammini ticks. Nonfed adult females were subjected to temperature pulses, and 35S-labeled methionine was injected into the hemocoel or used as an incubation medium for excised salivary glands. Heat-induced protein synthesis was observed in both whole-body and excised salivary gland preparations from nonattached ticks solely when ambient temperature was raised to 42°C. The approximate Mr of each protein was 88 kilodaltons (kDa), 75 and 74 kDa, and between 21 and 27 kDa for a group of lower molecular weight proteins. In another experimental series, ticks were allowed to attach to rabbits and then were subjected to temperature pulses. The 88 and 74 kDa proteins were present in preparations from nonheated ticks that had attached for 1 h. Only small amounts of these proteins were evident in tissues prepared from ticks attached for 2 days or more. Heat-induced proteins became apparent in ticks that were incubated at 42°C, regardless of time of attachment.  相似文献

Summary Extracellular laccase in cultures of Grifola frondosa grown in liquid culture on a defined medium was first detectable in the early/middle stages of primary growth, and enzyme activity continued to increase even after fungal biomass production had peaked. Laccase production was significantly increased by supplementing cultures with 100–500 (M Cu over the basal level (1.6 mM Cu) and peak levels observed at 300 mM Cu were ∼ ∼7-fold higher than in unsupplemented controls. Decreased laccase activity similar to levels detected in unsupplemented controls, as well as an adverse effect on fungal growth, occurred with further supplementation up to and including 0.9 mM Cu, but higher enzyme titres (2- to 16-fold compared with controls) were induced in cultures supplemented with 1–2 mM Cu2+. SDS-PAGE combined with activity staining revealed the presence of a single protein band (M r ∼ ∼70 kDa) exhibiting laccase activity in control culture fluids, whereas an additional distinct second laccase protein band (M r␣∼ ∼45 kDa) was observed in cultures supplemented with 1–2 mM Cu. Increased levels of extracellular laccase activity, and both laccase isozymes, were also detected in cultures of G. frondosa supplemented with ferulic, vanillic, veratric and 4-hydroxybenzoic acids, and 4-hydroxybenzaldehyde. The optimal temperature and pH values for laccase activity were 65 °C and pH 2.2 (using 2,2′-azinobis(3-ethylbenzthiazoline-6-sulfonate) {ABTS} as substrate), respectively, and the enzyme was relatively heat stable. In solid-state cultures of G. frondosa grown under conditions adopted for industrial-scale mushroom production, extracellular laccase levels increased during the substrate colonization phase, peaked when the substrate was fully colonized, and then decreased sharply during fruit body development.  相似文献

Trichosia pubescens larval midgut ceca cells display in their plasma membranes α-glucosidases (Mr 95,000; pHo 5.5; Km 5.7 mM; Ki for TRIS 8.9 mM), trehalases (Mr 69,000; pHo 5.3; Km 0.92 mM; Ki for TRIS 57 mM), and aminopeptidases (Mr 95,000; pHo 8.7; Km 0.19 mM) which are solubilized by Triton X-100. The enzymes were purified by electrophoresis and used to raise antibodies in a rabbit. T. pubescens imaginal midgut cells display in their plasma membranes an α-glucosidase (Mr 156,000; pHo 5.8; Km 2.3 mM; Ki for TRIS 0.2 mM), a trehalase (Mr 93,000; pHo 5.5; Km 0.72 mM; Ki for TRIS 45.5 mM), and an aminopeptidase (Mr 210,000; pHo 9.0; Km 0.47 mM). Antiserum produced against the larval enzymes shows no precipitation arc when tested by double immunodiffusion or by immunoelectrophoresis with Triton X-100-solubilized membrane proteins from imaginal midguts. Otherwise, a similar test showed that larval midgut cecal enzymes and larval ventriculus enzymes display complete immunological identity. The data suggest that, despite the fact the larval and imaginal aminopeptidase, α-glucosidase, and trehalase probably have similar functions, the genes coding for them in larvae and imagoes must differ.  相似文献

A thrombocyte-specific antigen was identified in two closely related catfish,Ictalurus punctatusandIctalurus furcatus,by monoclonal antibodies 4-20 and 7-2. The antibodies immunoprecipitate two noncovalently associated glycoprotein chains ofMr180,000 andMr95,000. Under reducing conditions theMr180,000 chain is resolved intoMr150,000 and 32,000 subcomponents. Analysis of N-terminal amino acid sequences indicates homology of theMr95,000 chain with the β3integrin subunit and homology of theMr150,000 chain with the αIIbintegrin subunit. These antibodies induce catfish thrombocyte aggregation and alteration of cell shape. The data indicate conservation of the megakaryocyte/platelet-restricted CD41/CD61 complex in bony fish.  相似文献

Demineralization (DM) from crab shell (CS) waste was carried out using a lactic acid-producing bacterium, Lactobacillus paracasei subsp. tolerans KCTC-3074 for 7 days at 25, 30, and 35°C. DM rates were 89∼92% and slightly affected by temperature. DM was also performed for four particle-sized shell samples (0.84∼3.35, 3.35∼10, 10∼20, and 20∼35 mm) with 10% inoculum, 5% shell, and 10% glucose at 30°C and 180 rpm for 7 days. It was found out that the shell size had a slight effect on the rate of DM. Negative relationships were found between DM and residual dry weight (r2 = 0.960), and between DM and pH (r2 = 0.906). Conversely, positive relationships were found between DM and medium protein (r2 = 0.696), and between DM and total titratable acidity (r2 = 0.630).  相似文献

The exopolysaccharides produced by Lactobacillus delbrueckii subsp. bulgaricus NCFB 2772 grown in defined medium were investigated. At equal cell densities, the strain produced 95 mg l−1 exopolysaccharides with glucose and 30 mg l−1 with fructose as the carbohydrate source. High-performance size-exclusion chromatography of the exopolysaccharides produced on glucose showed the presence of two fractions with relative molecular masses (M r) of 1.7 × 106 and 4 × 104 in almost equal amounts. The exopolysaccharides produced on fructose contained mainly a fraction of low M r of 4 × 104. The high-M r fraction of the purified exopolysaccharides produced on glucose appeared to have a sugar composition of galactose, glucose and rhamnose in the molar ratio of 5:1:1, whereas the low-M r weight fraction contained galactose, glucose and rhamnose in the molar ratio of approximately 11:1:0.4. The purified exopolysaccharide fractions produced on fructose showed comparable ratios. The high-molecular-mass fractions contained terminally linked galactose, 1,2,3-linked galactose, 1,3,4-linked galactose, 1,3-linked glucose and terminally linked rhamnose. The low-molecular-mass fractions contained mainly 1,3-linked galactose and 1,6-linked galactose and lower amounts of other sugar linkages. The production of the high-M r fractions appeared to be dependent on the carbohydrate source, whereas the low-M r fractions were produced more continuously. The major heat shock proteins of Staphylococcus aureus had apparent Mrs of 84,000, 76,000, and 60,000, and other prominent proteins of Mrs 66,000, 51,000, 43,000 and 24,000 were also induced. Staphylococcus epidermidis showed a similar response. These proteins were also induced by CdCl2, ethanol and apparently osmotic stress (1.71 M NaCl or 2.25 M sucrose). Most of the proteins sedimented with the membrane fraction, but the Mr 60,000 protein remained in the cytoplasm.  相似文献

Although Amblyomma brasiliense Aragão 1908 has been reported as one of the most aggressive ticks to humans in Brazil, information about the biology of this tick species is virtually inexistent. This work reports data on the life cycle of A. brasiliense fed on rabbits and pigs and maintained in an incubator at 20°C, 90% RH and 12 h of light for off-host development. Tick yield of adult females fed on pigs and rabbits was 81.2% and 58.3%, respectively. Females fed on pigs had mean engorgement weight of 862.3 mg and egg mass of 208 mg, while females fed on rabbits had mean engorgement weight of 606.1 mg and egg mass of 160 mg; these values did not differ statistically between host species. Feeding period of female ticks fed on pigs (10 days) was significantly shorter than that on rabbits (17 days). Mean preoviposition period was slightly longer (35.9 days) for ticks fed on pigs than on rabbits (30 days). The minimum incubation period of eggs of ticks from both host species was similar and over 100 days. Egg production efficiency was low for females fed on both hosts (less than 30% and 20% for ticks from pigs and rabbits, respectively). More than 55% of larvae and 79% of nymphs fed on rabbits, set free inside the feeding chambers, engorged successfully. These ticks attained an engorgement weight of 1.3 and 18.2 mg, respectively, and fed for approximately 5 days. The minimum pre-molt period was 30 days for engorged larvae and over 44 days for nymphs. Molting success was low, less than 50% in the case of larvae and less than 20% for nymphs. Further studies are required to better determine the off-host requirements of this tick species.  相似文献

Summary The junction of isolated triads can be mechanically broken by passage through a French press and subsequently reformed by incubation of the isolated organelles with certain salts of weak acids (e.g., K cacodylate. K propionate, and K butyrate). In contrast, other salts (e.g., KCl, K phosphate, and K benzoate) are ineffective in promoting triad formation. An endogenous factor obtained from a muscle homogenate acts in the same manner as these artificial compounds. When rabbit skeletal muscle is homogenized in a KCl solution and centrifuged to remove large cellular components and membrane fractions, an endogenous factor is extracted into the high speed supernatant which promotes the reformation of mechanically broken triads. A three-stage purification of this factor has been achieved using: (1) ammonium sulfate fractionation, (2) adsorption chromatography, and (3) molecular sieve chromatography. SDS-PAGE showed that the protein was purified to homogeneity and had a subunitM r of 34,000 daltons. This protein has the following characteristics: (1) it exists in 0.1m KCl as a polymeric substance with an estimatedM r =123,000 on molecular sieve chromatography and aM r =155,000 on sedimentation equilibrium; (2) it promotes the formation of triadic vesicles from isolated organelles in a low ionic strength medium; (3) Both this protein and cacodylate share the property of specifically catalyzing the association and aggregation of junctional proteins which had previously been dissolved by neutral detergent and salt; (4) it appears to be identical to an extrinsic constituent of terminal cisternae, which has been described as a protein ofM r =34K. It is not clear, however, whether this protein is a necessary and integral component of the junctional feet or whether it exerts predominantly a catalytic role in the formation of the triad junction.  相似文献

When L929 cells are cultured in the abssence of glucose the rate of synthesis of two polypeptides (95,000 and 82,000 Mr) is increased while that of an 85,000 Mr polypeptide is correspondingly decreased. It has been shown recently that the 85,000 Mr polypeptide is identical with the heat shock protein that is associated with the Rous sarcoma virus transforming gene product (pp60src). The present study shows that dexamethasone completely inhibits the alterations in the pattern of protein synthesis that would normally ensue following glucose deprivation. In fact, synthesis of the 85,000 Mr polypeptide and another polypeptide (69,000 M,) that is not glucose regulated are dramatically increased. In contrast, when L929 cells are maintained in ample glucose the effects of dexamethasone are much less dramatic with only one major SDS polyacrylamide gel band (90,000 Mr) being affected. This effect is specific for glucocorticoids. One-dimensional peptide mapping did not demonstrate any obvious structural relatedness among the 95,000, 90,000, 85,000, and 82,000 Mr polypeptides. Therefore, the observed changes appear to result from alterations in the rate of protein synthesis rather than from changes in precursor-product relationships. It is suggested on the basis of this data that glucocorticoids may play a role in modulating the response of cultured cells to glucose deprivation by eliciting a heat shock-like response.  相似文献

In contrast to mammalian transglutaminases (TGs), plant members of the superfamily are poorly characterized. In order to produce pure and active TG for its functional and structural studies, variants of maize chloroplast transglutaminase (TGZ, Patent WWO03102128) were sub-cloned into a pET28 vector and overexpressed in Escherichia coli BL21 (DE3) cells. The recombinant proteins were present mainly as insoluble inclusion bodies. The TGZ4p variant with four B-type repeats (M r∼55 kDa), was affinity purified from urea-solubilized inclusion bodies. TGZ4p was refolded by rapid dilution in a Ca2+- and guanidine-containing buffer. Active TGZ4p shows the general catalytic characteristics described for other TGs. Various stressors were used to induce stress proteins in Clostridium perfringens. Cultures of C. perfringens FD-1041 were subjected to cold shock (28°C for 1 h), acid shock (pH 4.5 for 30 min), or heat shock (50°C for 30 min). Cells were lysed and protein samples were analyzed by immunoblotting with antiserum derived from rabbits suffering from gas gangrene. Eight cold shock proteins (approximate Mr 101, 82, 70, 37, 22, 12, 10 and 6 kDa) and also eight heat shock proteins (approximate Mr 101, 82, 70, 27, 22, 16, 12 and 10 kDa) were immunoreactive with the serum. No immunoreactive proteins were detected in samples subjected to acid shock proteins and purified DnaK protein was also non-immunoreactive with the serum. These immunogenic stress proteins may be important in regulating diseases caused by C. perfringens. Such proteins could be involved in cell survival mechanisms, serve as targets during infection, or play a role in recognition of the bacteria by the host.  相似文献

Aminopeptidase (soluble form Mr 110,000), carboxypeptidase A (soluble form Mr 47,000), maltase (a dimer composed of two identical Mr 60,000 subunits) and trypsin (two charge isomers with Mr 34,000) are found in major amounts in the crop and midgut tissue, whereas amylase (a trimer of three identical Mr 18,000 subunits) and cellobiase (a trimer of three identical Mr 27,000 subunits) occur mainly in the crop and midgut contents. Subcellular fractions of midgut cells were obtained by conventional homogenization, followed by differential centrifugation or differential calcium precipitation. The results suggest that part of the aminopeptidase and carboxypeptidase A activity is bound to microvilli, that major amounts of trypsin and maltase are trapped in the cell glycocalyx and finally that soluble aminopeptidase, amylase and cellobiase occur in intracellular vesicles. The data support the hypothesis that most protein and carbohydrate digestion takes place in the crop under the action of enzymes passed forward from the midgut, after being secreted by exocytosis. Nevertheless, part of the intermediate and final digestion occurs at the surface of the midgut cells. The peculiar features of the digestion of P. aequinoctialis beetles, including their partly fluid peritrophic membranes, are thought to be derived from putative Coleoptera ancestors.  相似文献

The complete sequence of the 121 amino acid residues of piratoxin-I (PrTX-I), a phospholipase A2 (PLA2)-like myotoxin fromBothrops pirajai snake (Bahia jararacussu) venom, is reported. From the sequence, anM r of 13,825 and an approximatepI of 8.3 were calculated. PrTX-I shows a high sequence homology with Lys-49 myotoxins from other bothropic (∼95%) and nonbothropic (∼80%) venoms, but only 70–75% homology w hen aligned with the catalytically active Asp-49 PLA2s. When compared with bothropstoxin-I fromBothrops jararacussu, which is morphologically almost identical toB. pirajai, only two changes out of 121 total amino acid residues have been observed. The approximate minimal lethal doseLD 50 (mice, i.p., 24 hr) of PrTX-I was 8 (6.8–9.1) mg/kg, and the minimal edematogenic dose (MED) in a rat paw model was 39.5±1.8 ug. After alkylation of His-48 withp-bromophenacyl bromide, the MED was 40.1±1.9 ug, but up to 4LD 50 were unable to cause death in any of a group of eight mice after 72 hr. Therefore the edematogenic activity was retained and apparently did not involve His-48, suggesting that at least two biologically active sites are present in PrTX-I.  相似文献
