Serine carboxypeptidases (SCPs) comprise a large family of protein hydrolyzing enzymes and have roles ranging from protein turnover and C-terminal processing to wound responses and xenobiotic metabolism. The proteins can be classified into three groups, namely carboxypeptidase I, II and III, based on their coding protein sequences and the fact that each family is characterized by a central catalytic domain of unique topology designated as the “α/β hydrolase fold”. The available SCP protein sequences have been utilized as datasets to build a HMM (hidden Markov model) profile, which is used to search the rice (Oryza sativa L. ssp. japonica) proteome. A total of 71 SCP and serine carboxypeptidase-like (SCPL) protein-coding genes exist in rice. The intron-exon structure, chromosome localization, expression and characteristics of encoded protein sequences of the 71 putative genes are reviewed.  相似文献

Plant morphogenesis is an intimate cooperation between environmental signals and a genetically-encoded developmental programme. Although all stages subsequent to floral induction may be influenced by daylength, the information available has come from diverse species during different morphogenetic events. This makes it difficult to investigate how photoperiodic signalling is integrated into various steps of the developmental programme. Here, we report that, in late japonica rice lines, morphogenetic events, including 12th leaf, axis of the main panicle, awn of the glum, anthers, elongation rate of internodes, and pollen fertility of the photoperiod-sensitive genic male-sterile (PGMS) rice, are significantly affected by daylength after panicle initiation. These data indicate that daylength affects many morphologic events, and that changes in morphology are mainly determined by the characteristics of the events themselves. These findings lay a foundation for future investigations into how potentially common photoperiodic signalling system(s) are integrated with diversified developmental events. In addition, we discussed the essential nature of PGMS rice.  相似文献

Genetic load in the genome of the model species, rice, was genetically dissected by mapping quantitative trait loci (QTLs) affecting the radiosensitivity of 226 recombinant inbred lines (RILs) to gamma-ray- and spaceflight-induced radiation. The parents and RILs varied considerably in their radiosensitivity to gamma-ray irradiation. A total of 28 QTLs affecting the two index traits, seedling height (SH) and seed fertility (SF), of radiosensitivity were identified. The japonica parent, Lemont, was much more sensitive to gamma-ray irradiation than the indica parent, Teqing, and its alleles at almost all QTLs were associated with increased radiosensitivity, suggesting a much higher genetic load in the japonica genome of rice. Six QTLs (QSh2a, QSh2b, QSh5a, QSh7, QSf3b, and QSf10b) were located in the genomic regions particularly sensitive to radiation and thus might represent possible 'mutation hot spots' in the japonica genome. Detailed characterization of these genomic regions may shed light on the evolution and subspecific differentiation of rice.  相似文献

Six diazotrophic bacteria were isolated from surface-sterilized roots of rice variety HUR-36, which is grown with very low or no inputs of nitrogen fertilizer. Out of six bacteria one isolate, RREM25, showed appreciable level of nitrogenase activity, IAA production, and Phosphate solubilization ability, and was further characterized with a view to exploiting its plant growth promoting activity. Based on 16S rRNA gene sequence analysis, this isolate was identified as Burkholderia cepacia. Diazotrophic nature of this particular isolate was confirmed by Western blot analysis of dinitrogenase reductase and amplification of nifH. Microscopic observation confirmed colonization of gfp/gusA-tagged RREM25 in the intercellular spaces of cortical as well as vascular zones of roots. Inoculation of RREM25 to rice plants resulted in significant increase in plant height, dry shoot and root weight, chlorophyll content, nitrogen content and nitrogenase activity. Plant growth promoting features suggest that this endophytic bacterium may be exploited in rice cultivation after a thorough and critical pathogenicity test.  相似文献

The origins of the Asian cultivated rice Oryza sativa from its wild ancestor O. rufipogon have been debated for decades. The question mainly concerns whether it originated monophyletically or polyphyletically. To shed light on the origins and demographic history of rice domestication, we genotyped a total of 92 individual plants from the two O. sativa subspecies and O. rufipogon for 60 microsatellites. An approximate Bayesian method was applied to estimate demographic parameters for O. rufipogon vs. O. sativa ssp. indica and O. rufipogon vs. O. sativa ssp. japonica. We showed that the japonica subspecies suffered a more severe bottleneck than the indica subspecies and thus a greater loss of genetic variation during its domestication. Across microsatellite loci there is a significant positive correlation in the reduction of genetic diversity between the two subspecies. The results suggest that completely independent domestication of indica and japonica subspecies may not explain our data and that there is at least partial sharing of their ancestral populations and/or recent gene flow between them.  相似文献

Embryogenic and non-embryogenic calluses were induced from 3,4,5 and 7d old coleoptile segments of indica rice (Oryza sativa L. cv. CH 1039). Compact, globular, yellow and creamy embryogenic and white friable non-embryogenic callus arose from the cut end and entire length of the coleoptile segments. Murashige and Skoog's (MS) medium supplemented with 2.5mg/1 2,4-D was used as callus induction medium. Plant regeneration from coleoptile segments occurred with the transfer of embryogenic callus to MS basal medium supplemented with 2.0mg/1 BAP and 0.5mg/1 NAA in combination. Lipases are useful enzymes that are primarily responsible for the hydrolysis of acylglycerides during lipid processing. We have cloned a lipase gene from a rice seed coat cDNA library (Oryza sativa cv. Dongjin). The cDNA was 1,445 bp in length and encoded 361 amino acid residues (GenBank accession No. AY580163). The deduced amino acid sequence had 82 and 56% identity to Oryza sativa (cv. Chuchung) and Arabidopsis thaliana lipase genes, respectively, and there was a GxSxG consensus motif near the catalytic triad at the active serine site. The deduced sequence had little homology to mammalian and microbial lipases. When the Oryza sativa lipase gene was expressed in Escherichia coli with the pET expression system, activity was found mainly in the pellet fraction. The purified product had lipolytic activity towards tributyrin and was about 40 kDa in size.  相似文献

太湖流域稻作历史悠久,是我国粳稻高产地区之一。但育成品种的遗传基础日趋狭窄,将会给农业生产带来潜在风险。为了有效利用地方水稻资源以扩大改良品种的遗传基础,作者通过调查19个农艺性状,对823个太湖流域粳稻地方品种的遗传多样性进行了研究。结果表明:15个数量性状的变异系数(CV)范围在4.6%(结实率)和33.7%(单株产量)之间。颖壳色、稃尖色、芒型和粒型4个质量性状分别有10、6、5、4种表现型。19个农艺性状的多样性指数(H′)变化范围在0.757(粒型)和1.930(每穗实粒数)之间,平均为1.540。9月10日以后抽穗的品种比9月10日及其以前抽穗的品种具有更丰富的表型变异。主成分分析表明,株高、单株有效穗、每穗总粒数、每穗实粒数、着粒密度、千粒重、单株产量和穗颈长等8个性状是解释太湖流域粳稻地方品种多样性的重要性状。  相似文献

用超级稻品种‘沈农606’和普通粳稻‘丽江新团黑谷’为亲本杂交获得的102份F_2代单株,通过SRAP分子标记遗传分析,构建了包含14个连锁群,由129个多态性位点组成的水稻连锁图谱,此图谱覆盖基因组长度1671.5 cM,平均图距13.0 cM。连锁群上有17.2%的多态性位点表现偏分离,偏分离标记在连锁群上存在热点区域。  相似文献

We present the first characterization of K+ optimization of N uptake and metabolism in an NH4+‐tolerant species, tropical lowland rice (cv. IR‐72). 13N radiotracing showed that increased K+ supply reduces futile NH4+ cycling at the plasma membrane, diminishing the excessive rates of both unidirectional influx and efflux. Pharmacological testing showed that low‐affinity NH4+ influx may be mediated by both K+ and non‐selective cation channels. Suppression of NH4+ influx by K+ occurred within minutes of increasing K+ supply. Increased K+ reduced free [NH4+] in roots and shoots by 50–75%. Plant biomass was maximized on 10 mm NH4+ and 5 mm K+, with growth 160% higher than 10 mm NO3‐grown plants, and 220% higher than plants grown at 10 mm NH4+ and 0.1 mm K+. Unlike in NH4+‐sensitive barley, growth optimization was not attributed to a reduced energy cost of futile NH4+ cycling at the plasma membrane. Activities of the key enzymes glutamine synthetase and phosphoenolpyruvate carboxylase (PEPC) were strongly stimulated by elevated K+, mirroring plant growth and protein content. Improved plant performance through optimization of K+ and NH4+ is likely to be of substantial agronomic significance in the world's foremost crop species.  相似文献

Proteomic approach is applied for the analysis of seed brans of 14 rice varieties (Oryza sativa L. ssp. indica) which can classify to five aromatic rice and nine nonaromatic rice. The two-dimensional electrophoresis (2-DE) protein patterns for 14 rice varieties were similar within pH ranges of 3-10 and 4-7. To characterize aromatic group-specific proteins, we compared 2-D gels of aromatic rice to nonaromatic rice using PDQUEST image analysis. Four out of six differential spots were identified as hypothetical proteins, but one (SSP 7003) was identified by matrix assisted laser desoption/ionization-quardrupole-time of fight (MALDI-Q-TOF) as prolamin with three matching peptides based on NCBI database. Prolamin is a class of storage proteins with three different polypeptides of 10, 13, and 16 kDa. Spot SSP7003 was identified as a 13 kDa polypeptide of prolamin by combination of mass spectroscopy and N-terminal sequence analyses. In contrast, one sulfur-rich 16 kDa polypeptide of prolamin was found in extremely high intensity in brans of deep-water rice compared to nondeep-water rice. Our results suggest that proteomics is a powerful step to open the way for the identification of rice varieties.  相似文献

More than 750 plants were regenerated from protoplasts isolated from microspore callus-derived cell suspensions of the Mediterranean japonica rice Miara, using a nurse-feeder technique and N6-based culture medium. The mean plating efficiency and the mean regeneration ability of the protocalluses were 0.5% and 49% respectively. Flow cytometric evaluation of the DNA contents of 7 month old-cell and protoplast suspensions showed that they were still haploid. Contrastingly, the DNA contents of leaf cell nuclei of the regenerated protoclones ranged from 1C to 5C including 60% 2C plants. This was consistent with the morphological type and the fertility of the mature plants. Summary In a humid tropical climate at Pattambi (10° 48 N, 76° 12 E), the evapotranspiration (ET) rates of rice (Oryza sativa L. cv. Thriveni) were 2.8–5.7 mm/day during the first crop season (May–September) and 6.2–9.1 mm/day during the second crop season (September–January). The crop was grown at the Station in the irrigated lowlands on sandy soils with average yields of 3025 and 2925 kg/ha in the first and second cropping seasons, respectively. The seasonal ET, water requirements, water use efficiency and field water use efficiency of the crop were 400 mm, 1150 mm, 7.56 and 2.63 kg/ha per mm in the first crop season and 650 mm, 1500 mm, 4.50 and 1.95 kg/ha per millimetre in the second crop season. Using the reference crop ET computed by Blaney-Criddle, Radiation, and Penman methods and measured evaporation from class A pan, Colorado and GGI 3000 pans, the crop coefficients were worked out. Correlations between weather parameters and the biomass of rice were obtained. The grain yield (Y, in kg/ha) of the crop was predicted using the equationY=1.71Y 0–56S+85F–2430 (N=8,r=0.920), whereY 0 is the sample of biomass of the rice at flowering in kg/ha,S andF are the duration of sunshine hours and maximum air temperatures (°C) between the 46th day of transplanting and maturity.  相似文献
