Selenium (Se) is an essential trace element for humans and livestock. In the UK, human Se intake and status has declined since the 1980s. This is primarily due to the increased use of wheat (Triticum aestivum L.) grown in UK soils which are naturally low in Se. The aim of this study was to determine the potential for increasing grain Se concentration in a high-yielding UK wheat crop using fertilisers. The crop response of winter-wheat to Se fertilisation was determined under standard field conditions in two consecutive years at up to 10 sites. Selenium fertilisers were applied as high-volume drenches of sodium selenate solution, or as granular Se-containing products. Yield and harvest index were unaffected by Se fertilisation. Under all treatments, grain Se concentration increased by 16–26 ng Se g?1 fresh weight (FW) per gram Se ha?1 applied. An application of 10 g Se ha?1 would thereby increase the Se concentration of most UK wheat grain 10-fold from current ambient levels and agronomic biofortification of UK-grown wheat is feasible. Total recovery (grain and straw) of applied Se was 20–35%. The fate of Se in the food-chain and in the soil must be determined in order to optimize the efficiency of this process.  相似文献

Selenium (Se) has been proved to be an essential element for humans and animals. However, less is known about its effects on plants. Pea plants were treated foliarly once (OT) and twice (TT) with Se solution during their flowering period. Seeds obtained from these plants contained 383 and 743 ng Se g(-1), respectively, and, together with control seeds from untreated plants (UT) containing 21 ng Se g(-1), were sown in soil in a greenhouse. Se content and its chemical form in young plants were studied, and its impact on plant respiratory potential, measured as terminal electron transport system (ETS) activity, determined. ETS activity was highest in young pea leaves with the highest Se content. Higher ETS activity possibly reflected increased glutathione peroxidase (GSH-Px) activity in mitochondria. The Se content of leaves and stems of plants grown from control seeds was similar to that in the seed, being around 40 ng Se g(-1). Se concentration in leaves of young plants grown from OT and TT seeds was 605%, and 1340% higher, respectively, than the control, and in their stems 355%, and 680% higher, respectively. The ratio of Se concentrations in OT and TT seeds was the same as in the leaves and stems in the young plants grown from them. SeMet was the major Se compound in Se-rich pea seeds and leaves, comprising 49% and 67% of the total Se content in OT and TT seeds, respectively, and 85% and 79% in the corresponding leaves.  相似文献

Selenium (Se) is an essential trace element for humans and other mammals. Most dietary Se is derived from crops. To develop a Se biofortification strategy for wheat, the effect of selenate fertilization and bacterial inoculation on Se uptake and plant growth was investigated. YAM2, a bacterium with 99% similarity to Bacillus pichinotyi, showed many plant growth promoting characteristics. Inoculation with YAM2 enhanced wheat growth, both in the presence and absence of selenate: YAM2-inoculated plants showed significantly higher dry weight, shoot length and spike length compared to un-inoculated plants. Selenate also stimulated wheat growth; Un-inoculated Se-treated plants showed a significantly higher dry weight and shoot length compared to control plants without Se. Bacterial inoculation significantly enhanced Se concentration in wheat kernels (167%) and stems (252%), as well as iron (Fe) levels in kernels (70%) and stems (147%), compared to un-inoculated plants. Inoculated Se-treated plants showed a significant increase in acid phosphatase activity, which may have contributed to the enhanced growth. In conclusion; Inoculation with Bacillus sp. YAM2 is a promising Se biofortification strategy for wheat and potentially other crops.  相似文献

Interactions between Se (as selenate) and I (as iodate) uptake by spinach plants (Spinacia oleracea L.) were studied under controlled conditions using solution culture. Spinach readily accumulated both Se and I in the edible parts, the leaves, with solution-to-leaf transfer factors ranging from 3.5 to 13.4. The distribution coefficients between leaves and roots ranged from 4.07 to 5.66 for I and 4.51 to 8.59 for Se. Selenium concentrations in plant tissues were unaffected by addition of I to the nutrient solution. Similarly, plant I concentrations were unaffected by addition of Se to the nutrient solution, except in nutrient solution with I at a concentration of 50 μM, in which addition of Se lowered shoot I concentrations significantly, but the effect was of low magnitude. These results indicate the possible feasibility of dual supplementation of plant growth substrates with Se and I to improve human nutrition where these two elements are deficient in the diet. The data also indicate the involvement of a positive feedback mechanism in the uptake of Se by spinach plants, since Se concentrations in leaves increased disproportionately with increasing Se concentration in the nutrient solution.  相似文献

BackgorundCowpea is a crop widely used in developing countries due its rusticity. Besides its rich genotypic variability, most breeding programs do not explore its potential to improve elements uptake. Selenium (Se) is a scarce element in most soils, resulting in its deficiency being common in human diets. This study aimed to evaluate the interaction between biofortification with Se and genotypic variation in cowpea, on the concentrations of Se in roots, leaves + stem and grains.MethodsTwenty-nine cowpea genotypes were grown in a greenhouse in the absence (control) and presence of Se (12.5 μg Se kg−1 soil) as sodium selenate, in fully randomized scheme. The plants were cultivated until grains harvest. The following variables were determined: roots dry weight (g), leaves + stems dry weight (g), grains dry weight (g), Se concentration (mg kg−1) in roots, leaves + stems and grains, and Se partitioning to shoots and grains.ResultsSelenium application increased the Se concentration in roots, leaves + stems and grains in all genotypes. At least twofold variation in grain Se concentration was observed among genotypes. Selenium application did not impair biomass accumulation, including grain dry weight. Genotype “BRS Guariba” had the largest Se concentration in grains and leaves + stems. Genotype MNC04-795 F-158 had the largest partitioning of Se to shoots and grain, due to elevated dry weights of leaves + stems and grain, and high Se concentrations in these tissues.ConclusionThis information might be valuable in future breeding programs to select for genotypes with better abilities to accumulate Se in grain to reduce widespread human Se undernutrition.  相似文献

Selenium (Se), which has antioxidant, anticancer, and antiviral properties, is an essential micronutrient for humans and animals. This micronutrient is found in high quantity in legumes. Peas have an ever-increasing importance in Spain, and to increase their nutritional value, two foliar Se fertilizers: sodium selenate and sodium selenite, at five different rates: 0, 10, 20, 40, 80 g?ha?1, were studied during the 2010/2011 crop season on semiarid Mediterranean conditions. Sodium selenate was much more effectively taken up by plants compared to sodium selenite. There was a strong linear relationship between the total Se content and Se rate in both sodium selenate and selenite. For each gram of Se fertilization as either sodium selenate or sodium selenite, the increase of total Se concentration in the grain was 148 and 19 μg Se?kg?1 dry weight, respectively. Ingestion of 100 g of peas previously fertilized with 10 g of sodium selenate per hectare would result in an intake of 179 μg of Se. This is almost 90 % of the daily recommended dose needed to reduce the chance of some cancers and about 179 % of the minimum concentration required to prevent Se deficiency diseases in animals. The pea has shown to have a strong ability to uptake and accumulate Se under Mediterranean conditions; therefore, this would make it a very strong candidate for inclusion in biofortification programs aiming to increase Se in the food chain.  相似文献

Mineral nutrient malnutrition, especially deficiency in selenium (Se), affects the health of approximately 1 billion people worldwide. Wheat, a staple food crop, plays an important role in producing Se-enriched foodstuffs to increase the Se intake of humans. This study aimed to evaluate the effects of different Se application methods on grain yield and nutritional quality, grain Se absorption and accumulation, as well as 14 other trace elements concentrations in wheat grains. A sand culture experiment was conducted via a completely randomized 3 × 2 × 1 factorial scheme (three Se levels × two methods of Se application, foliar or soil × one Se sources, selenite), with two wheat cultivars (Guizi No.1, Chinese Spring). The results showed that both foliar Se and soil Se application methods had effects on wheat pollination. Foliar Se application resulted in early flowering of wheat, while soil Se application caused early flowering of wheat at low Se levels (5 mg kg−1 ) and delayed wheat flowering at high selenium levels (10 mg kg−1 ), respectively. For trace elements, human essential trace elements (Fe, Zn, Mn, Cu, Cr, Mo, Co and Ni) concentrations in wheat grains were dependent of Se application methods and wheat cultivars. However, toxic trace elements (Cd, Pb, Hg, As, Li and Al) concentrations can be decreased by both methods, indicating a possible antagonistic effect. Moreover, both methods increased Se concentrations, and improved grain yield and nutritional quality, while the foliar application was better than soil. Accordingly, this study provided useful information concerning nutritional biofortification of wheat, indicating that it is feasible to apply Se to conduct Se biofortification, inhibit the heavy metal elements concentrations and improve yield and quality in crops, which caused human health benefits.  相似文献

The objectives of this study were to determine the effect of selenium (Se) from Se-enriched kale sprout (Brassica oleracea var. alboglabra L.) on the performance and Se concentrations in tissues of growing Japanese quails. Two hundred quails were divided into five treatments. Each treatment consisted of four replicates and each replicate contained ten quails in a completely randomize design. The experiment was conducted for 5?weeks. The treatments were T1, control diet; T2, control diet plus 0.2?mg Se/kg from sodium selenite; T3, T4, and T5, control diet plus 0.2, 0.5, and 1.0?mg Se/kg from Se-enriched kale sprout. We know very little about how soil-borne pollutants such as selenium (Se) can impact pollinators, even though Se has contaminated soils and plants in areas where insect pollination can be critical to the functioning of both agricultural and natural ecosystems. Se can be biotransferred throughout the food web, but few studies have examined its effects on the insects that feed on Se-accumulating plants, particularly pollinators. In laboratory bioassays, we used proboscis extension reflex (PER) and taste perception to determine if the presence of Se affected the gustatory response of honey bee (Apis mellifera L., Hymenoptera: Apidae) foragers. Antennae and proboscises were stimulated with both organic (selenomethionine) and inorganic (selenate) forms of Se that commonly occur in Se-accumulating plants. Methionine was also tested. Each compound was dissolved in 1 M sucrose at 5 concentrations, with sucrose alone as a control. Antennal stimulation with selenomethionine and methionine reduced PER at higher concentrations. Selenate did not reduce gustatory behaviors. Two hours after being fed the treatments, bees were tested for sucrose response threshold. Bees fed selenate responded less to sucrose stimulation. Mortality was higher in bees chronically dosed with selenate compared with a single dose. Selenomethionine did not increase mortality except at the highest concentration. Methionine did not significantly impact survival. Our study has shown that bees fed selenate were less responsive to sucrose, which may lead to a reduction in incoming floral resources needed to support coworkers and larvae in the field. If honey bees forage on nectar containing Se (particularly selenate), reductions in population numbers may occur due to direct toxicity. Given that honey bees are willing to consume food resources containing Se and may not avoid Se compounds in the plant tissues on which they are foraging, they may suffer similar adverse effects as seen in other insect guilds.  相似文献

The fertilisation of wheat crops with Se is a cost-effective method of enhancing the concentration of organic Se in grain, in order to increase the Se intake of animals and humans. It is important to avoid phytotoxicity due to over-application of Se. Studies of phytotoxicity of Se in wheat grown in Australia, where rainfall and grain yield are usually relatively low, have not been reported previously, and overseas studies have had varied results. This study used trials conducted in the field, glasshouse and laboratory to assess Se phytotoxicity in wheat. In field trials that used rates of up to 120 g ha–1Se as selenate, and in pilot trials that used up to 500 g ha–1 Se soil-applied or up to 330 g ha–1 Se foliar-applied, with soils of low S concentrations (2–5 mg kg–1), no Se toxicity symptoms were observed. In pot trials of four weeks duration, the critical tissue level for Se toxicity was around 325 mg kg–1 DW, a level attained by addition to the growth medium of 2.6 mg kg–1 Se as selenate. Solution concentrations above 10 mg L–1 Se inhibited early root growth of wheat in laboratory studies, with greater inhibition by selenite than selenate. For selenite, Se concentrations around 70 mg L–1 were required to inhibit germination, while for selenate germination % was unaffected by a solution concentration of 150 mg L–1 Se. Leaf S concentration and content of wheat increased three-fold with the addition of 1 mg kg–1 Se as selenate to the growth medium. This effect is probably due to the induction of the S deficiency response of the main sulphate transporter. This study found wheat to be more Se-tolerant than did earlier studies of tobacco, soybeans and rice. We conclude that Se phytotoxicity in wheat will not be observed at the range of Se application rates that would be used to increase grain Se for human consumption (4–200 g ha–1 Se as selenate, which would result in soil and tissue levels well below those seen in the above studies), even when – as is common in Australia – soil S concentration and grain yield are low.  相似文献

A hydroponic experiment was carried out in a growth chamber to investigate the impact of Selenium (Se) levels on physiological and biochemical characteristics of a barley cultivar. Membrane lipid peroxidation (LPO), proline accumulation and antioxidant activities of some enzymes of barley seedlings under Se toxicity were investigated. Significant increase in thiobarbituric acid reactive substance (TBARS) content, and a stimulation of catalase (CAT, 1.11.1.6), ascorbate peroxidase (APX, 1.11.1.11), glutathione reductase (GR, 1.6.4.2), and glutathione S-transferase (GST, 2.5.1.18) activities were recorded in barley seedlings subjected to 2, 4, 8, 16 ppm Se. Superoxide dismutase (SOD, EC 1.15.1.1) activity was not altered significantly. Plant height and chlorophyll content of the seedlings were also affected significantly in a dose dependent manner by Se treatment. Considerable amount of proline accumulation was also observed in response to Se treatment. The results indicated that increases in the activities of the antioxidant enzymes were not sufficient to protect cell membrane against Se toxicity.  相似文献

We made the INAA of hair of the common hare (Lepus europaeus Pall.) and of the common vole (Microtus arvalis Pall.) living in the emission zones. Both the hare and the vole are almost exclusively herbivores; they consume relatively large amounts of contaminated food and mirror reliably the contamination degree of the respective ecosystem. The use of free-living animals for the control of the environmental quality may complete effectively the information obtained by examination of habitants. Though the free-living animals lead a rather different way of existence it has been found that the analyses of their hair correlate very well with the analyses of human hair. It may be expected that the changes of concentrations of heavy metals will manifest themselves earlier in animals than in men, because the animals are strictly tied to the local food sources. The hair samples of hares contained increased concentrations of Sm, La, Au, As, Se, Cr, Sc, Fe, Ce, Th and Co. The hair of voles showed increased concentrations of Sm, La, Zn, As, Se, Cr, Sc, Fe, Sb, Ce, Cs and Co. A conspicuous trend towards the cumulation in hair was observed in the following elements: As, Se, Sc and Fe. The increase of their concentrations in the hair of animals from the emission regions amounted as such as to 10(3)%.  相似文献

This study aimed to determine the effectiveness of Se from hydroponically produced Se-enriched kale sprout (HPSeKS) on productive performance, egg quality, and Se concentrations in egg and tissue of laying quails. Two-hundred quails, 63 days of age, were divided into four groups. Each group consisted of five replicates and each replicate had ten birds, according to a completely randomized design. The experiment lasted for 6 weeks. The dietary treatments were T1 (control diet), T2 (control diet plus 0.2 mg Se/kg from sodium selenite), T3 (control diet plus 0.2 mg Se/kg from Se-enriched yeast), T4 (control diet plus 0.2 mg Se/kg from HPSeKS). The findings revealed that productive performance and egg quality of quails were not altered (p?>?0.05) by Se sources. Whole egg Se concentrations of quails fed Se from HPSeKS and Se-enriched yeast were higher (p?<?0.05) than that of quails fed the control diet. Breast muscle Se concentrations in quails fed Se from HPSeKS were higher (p?<?0.05) than that of quails fed Se from sodium selenite and Se-enriched yeast. Heart tissue Se concentrations of quails fed Se from Se-enriched yeast and HPSeKS were similar (p?>?0.05), but higher (p?<?0.05) than that of quails fed Se from sodium selenite. The results reveal that Se from HPSeKS did not change the performance and egg quality of quails. The effectiveness of Se from HPSeKS was comparable to that of Se-enriched yeast, which was higher than that of Se from sodium selenite.  相似文献

This study assessed the interactive effect of selenium (Se) and farmyard manure (FYM) on soil microbial activities, growth, yield, and Se accumulation by wheat grains. Preliminarily, the effect of Se (0–250 µg kg?1 soil) and FYM (0–12.5 g kg?1 soil) was assessed on soil microflora. Selenium exhibited an adverse impact on soil microflora; respiration was decreased at?≥?10 µg kg?1 soil while dehydrogenase and urease activities were decreased at?≥?125 µg kg?1 soil. At 250 µg Se kg?1 soil, respiration, dehydrogenase and urease activities were decreased by 81, 40 and 35%, respectively, on unamended soil, and by 9, 47 and 22%, respectively, on FYM-amended soil. The subsequent plant experiments were conducted with same Se and FYM rates; one was harvested 42 days after sowing and other at crop maturity. The application of 125 µg Se kg?1 and 12.5 g FYM kg?1 soil improved seedling biomass by 12.6 and 22%, respectively, while their combined use lacked synergistic effect. Similarly, at maturity Se and FYM increased grain yield while their combined effect was not synergistic. The Se-induced suppression in microbial activities was not related to yield which was improved (11% at the highest rate in unamended soil) by Se application. Selenium application increased grain Se content in a rate-dependent manner, it increased from 0 to 1025 µg kg?1 by applying 250 µg Se kg?1 soil. Moreover, FYM application decreased Se accumulation in grains. It is concluded that FYM application increased soil microbial activities and yield but reduced grain Se accumulation in wheat on Se-applied soil.

The mungbean plants were grown hydroponically in the absence (control) or presence of 0.1, 0.25, 0.50 and 0.75 ppm selenium (as sodium selenate) for 10 days. The growth of shoots and roots increased with application of selenium with greater extent in shoots. With 0.5 and 0.75 ppm Se levels, the shoot growth was stimulated by 24% to 27% over control, respectively, while the roots showed a corresponding increase of 18-19%, respectively. The shoot-to-root ratio was enhanced significantly with Se application and maximum effects occurred at 0.75 ppm Se. A significant increase was observed in chlorophyll and cellular respiration ability with 0.5 and 0.75 ppm selenium. The increase in growth by selenium was accompanied by elevation of starch, sucrose and reducing sugars. The activity of starch hydrolysing enzymes--amylases and sucrose hydrolysing enzyme--invertase was stimulated significantly with selenium. This was associated with elevation of activities of sucrose synthesising enzymes--sucrose synthase and sucrose phosphate synthase. It was concluded that increase in growth of shoots and roots by application of Se was possibly the result of up-regulation of enzymes of carbohydrate metabolism thus providing energy substrates for enhanced growth.  相似文献

Two cultivars of common buckwheat (Fagopyrum esculentum), Pyra and Siva, were exposed to three treatments: water deficit (WD), foliar spraying by selenium (as Na2SeO4) (Se), and the combination of both. In WD-plants the stomatal conductance (g s) was significantly lower, while WD+Se-plants of Siva had significantly higher g s. None of the treatments resulted in significant differences of potential photochemical efficiency of photosystem 2 (PS2). A significantly higher actual photochemical efficiency of PS2 was obtained in Siva WD-plants and in Pyra Se-and WD-plants which was possibly due to improvement of plant water management during treatment. A significant interaction was observed between the effects of WD and Se on respiratory potential in Pyra. WD, Se, and the WD+Se combination resulted in shorter Pyra and Siva plants, with a reduced number of nodes. WD slightly negatively affected the yield per plant. The yield was highest in plants exposed to Se only. In Siva the number of seeds was triple while the average seed mass remained unchanged.  相似文献

Accumulation of different metals and metalloids was assessed in two vegetables radish (Raphanus sativus L.) and spinach (Spinacea oleracea L.) irrigated with domestic wastewater in the peri-urban areas of Khushab City, Pakistan. In general, the metal and metalloid concentrations in radish and spinach were higher at site-II treated with sewage water than those found at site-I treated with canal water. In case of radish at both sites the levels of metals (Cr, Mn, Fe, Co, Ni, Cu, Zn, As, Se, Mo, Cd, and Pb) were below the permissible level except those of Mn, Ni, Mo, Cd, and Pb. At both sites, the transfer factor ranged from 0.047–228.3 mg kg?1 with Cr having the highest transfer factor. The metal pollution index in soil was in the following order: As > Fe > Ni > Zn > Cd > Mo > Se > Co > Pb > Mn > Cr > Cu, respectively. While in case of spinach at both sites, the concentrations of metals and metalloids in vegetable samples irrigated with canal and sewage water were observed below the permissible level except Mn, Ni, Zn, Mo, and Pb. At both sites, the transfer factor ranged from 0.038–245.4 mg kg?1 with Cr having the highest transfer factor. The metal pollution index in soil was in the following order: Cd > Ni > Co > Se > Mn > Zn > Mo > Pb > Fe > Cr > As > Cu, respectively.  相似文献

Bean (Phaseolus vulgaris L.) seedlings were subjected to varying selenium levels (1, 2, 4, and 6 ppm) in a hydroponic culture. The germination reached 100% in 48 h in all Se levels except 6 ppm, where it took 72 h. The root and shoot growth was stimulated at 1 and 2 ppm Se levels that was commensurate with increase in chlorophyll content, leaf water content, and cellular respiration. At 4 and 6 ppm Se levels, the growth was inhibited appreciably, which was associated with increase in stress injury measured as damage to membranes and decrease in cellular respiration, chlorophyll, and leaf water content. The oxidative injury as elevation of lipid peroxidation was larger compared to hydrogen peroxide accompanied by reduced levels of enzymatic (superoxide dismutase, catalase, ascorbate peroxidase, and glutathione reductase) and non-enzymatic (ascorbic acid and glutathione) antioxidants. Proline content was significantly higher at 1 and 2 ppm Se but diminished considerably at 4 and 6 ppm levels concomitant with the reduced growth. Exogenous application of proline (50 μM) resulted in substantiation of its endogenous levels that antagonised the toxic effects of Se by improving the growth of seedlings. The stress injury was reduced significantly with simultaneous increase in enzymatic and non-enzymatic antioxidants. Especially the components of ascorbate–glutathione cycle showed larger stimulation with proline application. The role of proline in mitigating the toxic effects of Se is discussed.  相似文献

Gamma-aminobutryric acid (GABA) accumulation in response to diverse stresses is well known in plants; however, the comparative effects of different environmental stresses on GABA accumulation was not addressed in the same plant system. Here, we investigated GABA accumulation comparatively in sesame (Sesamum indicum L. cv. Cumhuriyet) plant under drought, salt, heavy metal (Se) and high-temperature stresses. Plants were stressed by application of 5% PEG-6000, 150 mM NaCl, 100 μg g−1 Se and high temperature (50°C for 2 h). Root and shoot growth reduced after PEG, NaCl, Se and high-temperature treatments. Among these, heavy metal treatment had the highest and earliest effect on growth. GABA accumulation could be related to stress perception rather than protection in sesame plant if we consider that the adverse effects of different abiotic stresses on growth were not elevated by GABA.  相似文献
