Aldehyde dehydrogenase ST0064, the closest paralog of previously characterized allosteric non-phosphorylating glyceraldehyde-3-phosphate (GAP) dehydrogenase (GAPN, ST2477) from a thermoacidophilic archaeon, Sulfolobus tokodaii, was expressed heterologously and characterized in detail. ST0064 showed remarkable activity toward succinate semialdehyde (SSA) (K m of 0.0029 mM and k cat of 30.0 s?1) with no allosteric regulation. Activity toward GAP was lower (K m of 4.6 mM and k cat of 4.77 s?1), and previously predicted succinyl-CoA reductase activity was not detected, suggesting that the enzyme functions practically as succinate semialdehyde dehydrogenase (SSADH). Phylogenetic analysis indicated that archaeal SSADHs and GAPNs are closely related within the aldehyde dehydrogenase superfamily, suggesting that they are of the same origin.  相似文献

Seeds of two lettuce cultivars (Lactuca sativa L., cv. Meikoninginand cv. Grand Rapids) were hermetically stored with constantmoisture contents ranging between 3.6 and 17.9 per cent (freshweight basis) at constant temperatures ranging between 5 and75 °C. The decline with time in percentage germination andpercentage normal seedlings was determined for each storagetreatment. The data were fitted to an equation which containsthe constants: K1, the probit of the initial percentage germinationor normal seedlings; KE, a species constant; CW, the constantof a logarithmic moisture term; CH, the constant of a lineartemperature term and CQ, the constant of a quadratic temperatureterm. Regression analysis of data from storage periods up to5.5 years at temperatures of 5–75 °C and seed moisturecontents of 3.6–13.6 per cent yielded the following values:KE= 8.218, CW=4.797±0.163, CH=0.0489±0.0050 andCQ=0.000365±0.000056. Although this equation consistentlyprovided a better fit, simplified equations, assuming eithera log-linear relationship between seed longevity and temperature,or a log-linear relationship between seed longevity and bothmoisture content and temperature, accounted for more than 94per cent of the variation at the restricted temperature rangeof 5–40 °C. Longevity of the same seed lots at sub-zero temperatures (–5,–10 and –20 °C) was studied in separate tests.Freezing damage, resulting in abnormal seedlings in the germinationtest, occurred at –20 °C when the moisture contentof the seeds exceeded 12 per cent. No decline in percentagenormal seedlings was observed after a storage period of 18 monthsor longer at –20 °C, provided the seed moisture contentdid not exceed 9.5 per cent. For seeds stored at –5 and–10 °C with 9.6–12.5 per cent moisture content,the observed rate of decline of percentage normal seedlingswas adequately predicted by the viability equation, using theabove values for the constants. This suggests that for low moisturecontents the viability equation can be applied to estimate longevityat sub-zero temperatures. This research determined constants for a viability equationto predict the longevity of groundnut seeds and to improve themanagement of seedlot storage throughout the trading period.Seeds of the Brazilian cultivar ‘Tatu’ (Valenciabunch type) were tested. Nine moisture content levels (rangingfrom 2.4 to 12.8%) and three storage temperatures (40, 50 and65 °C) were used. Sub-samples for each moisture content-storagetemperature combination were sealed in laminated aluminium-foilpackets and stored in incubators until complete survival curveswere obtained. A simple method is presented for selecting individual seedsfor growth and vigour prior to germination. The selection isbased on the measurement of an electric current originatingduring the initial stage of seed imbibition. After selecting,seeds may be returned to the quiescent state without affectingviability. Results of both laboratory germination and fieldexperiments demonstrate that high germination rates, more extensivegrowth, higher yields, and fewer defective plants are associatedwith small currents. Seeds of low viability and high currentsmay be removed to upgrade the germination and vigour. Resultsare presented from several varieties of Phaseolus vulgaris andfrom three other plant species. Experiments are discussed whichindicate that seeds graded by current levels possess markedlydifferent respiration rates.  相似文献

Methanol dehydrogenase (MDH) and aldehyde dehydrogenase (ALDH) were purified to a homogenous state from Methylobacillus glycogenes, an obligate methylotroph. MDH (Mr 140,000) was composed of two different subunits (Mr 60,000 and 9,000) forming an α2β2 structure. MDH was indicated as a metalloquinoprotein containing one atom of calcium (Ca) per enzyme molecule. Binding of Ca was so tight that it was hard to remove Ca completely without denaturation of enzyme protein. A partially resolved enzyme resumed its original enzyme activity upon exogenous addition of Ca. Purified ALDH (Mr 144,000) was composed of two identical subunits of molecular mass of 72,000. ALDH was proved to be a quinoprotein in which PQQ is bound covalently.  相似文献

In an investigation of the seed of six rice (Oryza spp.) varietiesstored under identical conditions no significant differencein the period of viability was shown. This suggests that someequations previously derived from an experiment on a singlevariety to describe the relationship between temperature, moisturecontent, and the period of viability may be applied generallyto the seed of cultivated rice varieties. The fact that thevarieties used here differed markedly in dormancy period whilein all cases the viability period was the same is discussedwith other evidence to show that the commonly held view thatthere is an association between dormancy and viability shouldbe abandoned, at least in rice.  相似文献

Cancer stem cells (CSCs) are defined as a small population of cancer cells with the properties of high self-renewal, differentiation, and tumor-initiating functions. Recent studies have demonstrated that aldehyde dehydrogenase 1 (ALDH1) is a marker for CSCs in adult cancers. Although CSCs have been identified in some different types of pediatric solid tumors, there have been no studies regarding the efficacy of ALDH1 as a marker for CSCs. Therefore, in order to elucidate whether ALDH1 can be used as a marker for CSCs of pediatric sarcoma, we examined the characteristics of a population of cells with a high ALDH1 activity (ALDH1high cells) in rhabdomyosarcoma (RMS), the most common soft tissue sarcoma in children. We used the human embryonal RMS (eRMS) cell lines RD and KYM-1, and sorted the cells into two subpopulations of ALDH1high cells and cells with a low ALDH1 activity (ALDH1low cells). Consequently, we found that the ALDH1high cells comprised 3.9% and 8.2% of the total cell population, respectively, and showed a higher capacity for self-renewal and tumor formation than the ALDH1low cells. With regard to chemoresistance, the survival rate of the ALDH1high cells was found to be higher than that of the ALDH1low cells following treatment with chemotherapeutic agents for RMS. Furthermore, the ALDH1high cells exhibited a higher degree of pluripotency and gene expression of Sox2, which is one of the stem cell markers. Taken together, the ALDH1high cells possessed characteristics of CSCs, including colony formation, chemoresistance, differentiation and tumor initiation abilities. Differentiation of oligodendroglial precursor cells (OPCs), a crucial prerequisite for central nervous system (CNS) remyelination in diseases such as Multiple Sclerosis (MS), is modulated by a multitude of extrinsic and intrinsic factors. In a previous study we revealed that the chemokine CXCL12 stimulates rodent OPC differentiation via activation of its receptor CXCR7. We could now demonstrate that CXCR7 is also expressed on NogoA- and Nkx2.2-positive oligodendroglial cells in human MS brains and that stimulation of cultured primary fetal human OPCs with CXCL12 promotes their differentiation as measured by surface marker expression and morphologic complexity. Pharmacological inhibition of CXCR7 effectively blocks these CXCL12-dependent effects. Our findings therefore suggest that a specific activation of CXCR7 could provide a means to promote oligodendroglial differentiation facilitating endogenous remyelination activities.  相似文献

Germination and growth inhibiting activities of surface lipids from 54 Nicotiana species were investigated. Almost half of the extracts were found to have such activities. Among them the surface lipids of N. glutinosa, N. bigelovii, N. sylvestris, N. repanda, N. stocktonii, and N. nesophila were rather strong. Guided by a bioassay using the inhibitory effects on tobacco seed germination and growth, two types of sucrose esters were isolated and identified from the surface lipids of N. glutinosa. The ester positions of each compounds were identified by 13C-NMR using deuterium exchange of HO ? OD. The structures were (2,3,4-tri-O-acyl)-α-d-glucopyranosyl)-(3-O-acetyl)-β-d-fructofuranqside (M1) and (2,3,4-tri-O-acyl)-α-d-glucopyranosyl-β-d-fructofuranoside (M2). The main fatty acids of M1 and M2 were acetic (only in M1), propionic, 2-methylbutyric, 4-methylpentanoic, 4-methylhexanoic, 5-methylhexanoic, and octanoic acids. These sucrose esters obtained from N. glutinosa inhibited not only tobacco seed germination and growth but also other plants’ growth.  相似文献

The completion of the rice genome sequence has made it possible to identify and characterize new genes and to perform comparative genomics studies across taxa. The aldehyde dehydrogenase (ALDH) gene superfamily encoding for NAD(P)+-dependent enzymes is found in all major plant and animal taxa. However, the characterization of plant ALDHs has lagged behind their animal- and prokaryotic-ALDH homologs. In plants, ALDHs are involved in abiotic stress tolerance, male sterility restoration, embryo development and seed viability and maturation. However, there is still no structural property-dependent functional characterization of ALDH protein superfamily in plants. In this paper, we identify members of the rice ALDH gene superfamily and use the evolutionary nesting events of retrotransposons and protein-modeling–based structural reconstitution to report the genetic and molecular and structural features of each member of the rice ALDH superfamily in abiotic/biotic stress responses and developmental processes. Our results indicate that rice-ALDHs are the most expanded plant ALDHs ever characterized. This work represents the first report of specific structural features mediating functionality of the whole families of ALDHs in an organism ever characterized.  相似文献

甜菜碱醛脱氢酶(Betaine aldehyde dehydrogenase,BADH)对非生物胁迫下植物渗透调节物质的合成和积累具有重要作用。分别从异苞滨藜和鞑靼滨藜两种盐生植物中分离到了BADH基因。序列分析表明,BADH全长均为1 507bp,编码501个氨基酸,两种BADH序列具有较高的相似性。甜菜碱醛脱氢酶的克隆为植物的基因转化及其功能分析奠定了基础。  相似文献

Abstract: The nuclear mitochondrial and synaptosomal fractions of rat brain were each found to contain some 25–30% of the total aldehyde dehydrogenase activity. The cytoplasmic fraction had a very low total aldehyde dehydrogenase activity. There were differences in the distribution of the activity when different aldehydes were used as substrates, suggesting the presence of isoenzymes in the various subcellular compartments. When rats were treated intra-cisternally with 6-hydroxydopamine there was no change in brain aldehyde dehydrogenase activity, although the noradrenaline content and the activities of tyrosine hydroxylase and dopamine-β-hydroxylase were markedly decreased. Treatment with 6-hydroxydopamine also had no significant effect on the aldehyde dehydrogenase activity in retinal homogenates. The results suggest that the aldehyde dehydrogenase activity in rat brain is predominantly outside the catecholaminergic nerve terminals.  相似文献

Various physiological roles of mammalian aldehyde dehydrogenase had been anticipated because of its broad substrate specificity. In order to clarify roles of the enzyme and the regulation of aldehyde metabolisms in liver, the intracellular distribution and isozyme of beef liver aldehyde dehydrogenase were studied.

The presence of the mitochondrial, the microsomal and the cytoplasmic isozymes were proved by the isoelectric focusing. These isozymes were different from each other in pH-activity curve in the responces for steroid hormones and disulfiram.

It was suggested by comparing the reactivities of these isozymes for various aldehydes that particular aldehyde might be oxidized by a favorite isozyme at particular locality in the liver cells and that a share of physiological role among these isozymes is probable.  相似文献

亚洲人群中普遍存在突变型的乙醛脱氢酶2(ALDH2*2)。此酶突变后活性缺失,导致乙醛在肝脏内大量累积使突变携带者在喝酒后会有脸红等不适反应,因此这可能影响他们的饮酒行为。由于ALDH2*2等位基因与饮酒行为相关,它也可能与酒精引起的肝脏损伤及某些癌症密切相关,而且,它在不同的亚洲人群中有不同的频率分布。近年来对ALDH2*2等位基因的序列结构、表达及其重要功能等有了更深入的了解,对ALDH2的多态性在研究方法、研究群体分布范围等都有很大进展。本文还讨论了不同地理分布、不同年龄结构、性别差异条件下,中国人群中ALDH2基因型频率与饮酒行为的关系。 Abstract: An atypical allele (ALDH2*2) in low Km aldehyde dehydrogenase (ALDH2), which is highly prevalent in Asian, may influence drinking behavior because of higher production of acetaldehyde in the liver. High alcohol sensitivity such as flushing after drinking has been shown to be mainly due to the atypical ALDH2 genotypes. The atypical allele is associated with alcohol-induced liver injury and some cancers. Recently, the researches on the polymorphisms not only in the gene itself but also its frequencies in different Asian populations have been made great progress. Three factors, including different sex, age and geography, were also analyzed with the genotypes of ALDH2 in Chinese populations.  相似文献

Background

CCR7-mediated signalling is important for dendritic cell maturation and homing to the lymph nodes. We have previously demonstrated that Jak3 participates in the signalling pathway of CCR7 in T lymphocytes.

Methodology and Principal Findings

Here, we used Jak3−/− mice to analyze the role of Jak3 in CCR7-mediated dendritic cells migration and function. First, we found no differences in the generation of DCs from Jak3−/− bone marrow progenitors, when compared to wild type cells. However, phenotypic analysis of the bone marrow derived DCs obtained from Jak3−/− mice showed reduced expression of co-stimulatory molecules compared to wild type (Jak3+/+). In addition, when we analyzed the migration of Jak3−/− and Jak3+/+ mature DCs in response to CCL19 and CCL21 chemokines, we found that the absence of Jak3 results in impaired chemotactic responses both in vitro and in vivo. Moreover, lymphocyte proliferation and contact hypersensitivity experiments showed that DC-mediated T lymphocyte activation is reduced in the absence of Jak3.

Conclusion/Significance

Altogether, our data provide strong evidence that Jak3 is important for DC maturation, migration and function, through a CCR7-mediated signalling pathway.  相似文献
