Carabidae (Coleoptera) are commonly used as indicators of ecosystem health due to their high diversity, conservation value and relative ease of identification. Carabids were monitored at Ahuriri Scenic Reserve, Port Hills, Canterbury, from August 2007 to September 2008 using pitfall traps. Carabid captures, species richness and distribution across the reserve were compared with those found in a similar study in 1977–1978. Monthly carabid captures were highly correlated with average temperature in both studies. There was little evidence of carabids responding to habitat factors, such as leaf litter depth, pH or ground cover. However, Megadromus antarcticus was positively associated with canopy openness and ground cover, whereas Holcaspis angustula was more abundant where canopy cover was extensive and there was a relatively high incidence of bare rock. Thirteen species of carabid were recorded in 1977–1978, of which six were absent, four had decreased, one was stable and two species increased in abundance from collections made in 2007–2008. The loss of one Banks Peninsula and five New Zealand endemic beetle species from this reserve is of concern, and future work should aim to gain more detailed information on the habitat requirements of these species to assist conservation management strategies.  相似文献

Petroleum ether, acetone, water and aqueous extract of Pedalium murex L. (Pedalaceae) leaves, root and fruit were tested against fourth instar larvae of the tobacco cutworm, Spodoptera litura (Fabr.) under laboratory conditions using leaf dip method. Larval mortality; larval, pupal and adult periods; pupal weight, pupation and adult emergence; larval and pupal deformities of S. litura were recorded. All the tested solvent extracts of P. murex were effective against S. litura life stages by causing mortality in a dose dependent manner. However, the efficacy was more significant with respect to acetone (leaf and root) and petroleum ether (root) at higher concentrations (0.8%) which leads to 100% larval mortality. The water and aqueous extracts of root caused 86.6 and 88.0% larval mortality respectively at 4% concentration. Among the plant parts tested, root had more importance followed by leaf and fruit. Plant extracts extended the larval, pupal and adult periods; reduced the pupal weight, pupal and adult emergence and caused larval–pupal deformities. Total life time was highly prolonged in acetone extracts of P. murex root. Though all the extracts were found to have insecticide activity, acetone and petroleum ether extracts of P. murex root can be used as an effective alternative to modern synthetic insecticides. Bioactive principles from these extracts can be isolated, identified and integrated in S. litura management.  相似文献

The susceptibility of Spodoptera litura to SlMNPV infection was markedly affected by phyto-chemicals ingested during the acquisition of viral inoculum on foliage of tomato and cauliflower. The LD50 values computed for second, third and fourth instar larvae assayed on tomato leaves were 254, 819 and 23395 PIBs/larva, respectively whereas, it was 326, 1719 and 43843 PIBs/larva for respective instars when assayed on cauliflower leaves. Thus LD50 values for second, third and fourth instar larvae were 1.28-, 2.09- and 1.87- fold lower, respectively in tomato leaves. Similarly, LT50 values for second, third and fourth instar larvae assayed on tomato leaves were 7.1 and 7.5 days, respectively at inoculum dose of 2.7×104 PIBs/larva whereas, it was 7.7 and 8.0 days for respective instars when assayed on cauliflower leaves at same inoculum. This result also showed that the S. litura were more susceptible on tomato leaves in comparison to cauliflower leaves as the time required for mortality was lower in tomato leaves. The possible biochemical bases for differential level of mortality of S. litura larvae on tomato and cauliflower crops needs to be investigated.  相似文献

  • The research conducted including its rationale: Spodoptera litura is the major pest of tomato causing significant reduction in tomato yield. Application of Plant growth promoting rhizobacteria(PGPR) prevent use of chemical fertilizer and synthetic pesticides through enhancement of plant growth and yield and induction of systemic resistance. Present investigation is an attempt to evaluate the role of PGPR, Pseudomonas putida and Rothia sp. on the physiology and yield of tomato fruit infested with the S. litura.
  • Central methods applied : The surface sterilized seeds of tomato were inoculated with 48 h culture of P. putida and Rothia sp. At 6–7 branching stage of the plant, the larvae of S. litura at 2nd in star was used to infect the tomato plant leaves.
  • Key results: The S. litura infestation decreased dry weight of shoots and roots by 46% and 22%, and significant reduction was recorded in tomato fruit yield. The P. putida and Rothia sp. inoculations alleviated the adverse effects of insect infestation and resulted in 60% increase in plant biomass and 40% increase in yield over infested plants.
  • Main conclusions including key points of discussion: PGPR: Defense appears to be mediated via increase in proline production, enhanced activities of antioxidant enzymes, stimulation in the activities of protease and polyphenol oxidases, increased contents of phenolics, protein and chlorophyll. The formulation of biopesticide involving PGPR comprise an environment friendly and sustainable approach to overcome insect infestation.

Profilicollis novaezelandensis n. sp. (Acanthocephala: Polymorphidae) is described from the South Island pied oystercatcher Haematopus ostralegus finschi Martens (Haematopodidae) and the intertidal crab Hemigrapsus crenulatus (Milne Edwards) (Brachyura: Grapsidae) from the South Island of New Zealand. The new species can be distinguished from all the other species of the genus by a combination of the following characters: long neck (13% of total body length for adults) and a subspherical proboscis with 14–16 longitudinal rows of 7–8 hooks. The mud crabs Helice crassa Dana (Grapsidae) and Macrophthalmus hirtipes (Heller) (Ocypodidae) were also harbouring cystacanths and the bar-tailed godwit Limosa lapponica (Linnaeus) (Scolopacidae) juveniles of P. novaezelandensis. This is the first record of brachyuran decapods as intermediate hosts of Acanthocephala from New Zealand. P. antarcticus is recorded from three crab species (Helice crassa, Hemigraspus crenulatus and Macrophthalmus hirtipes) and two bird species (Haematopus o. finschi and Limosa lapponica) in New Zealand. An unidentified species of Plagiorhynchus was also found in two bird species (H. o. finschi and H. unicolor Forster). P. antarcticus and P. novaezelandensis are the first records of Profilicollis from New Zealand.  相似文献

We studied the effects of natural wounding by insects and artificial wounding by clipping with scissors on the phenolic chemistry of two willows, Salix myrsinifolia and Salix pentandra. Half of the blade of a mature leaf was removed from each experimental plant either by allowing insects (chrysomellid beetles) to feed on the leaf or by clipping off half the blade of a leaf with scissors. We also examined the ability of wounded plants to warn neighboring plants of imminent wounding by an airborne signal by maintainign one set of control plants in the room containing the wounded plants and another set of control plants in a room hermetically sealed from the room containing the wounded plants. After 48 h, the experimental leaf and the fourth leaf and eighth leaf upwards in the leaf sequence from the experimental leaf were analyzed for phenols by high-pressure liquid chromatography. The same leaves in the leaf sequence from each control plant were similarly analyzed for phenols. Only one phenol, salicortin in leaves of S. myrsinifolia, increased in concentration in response to defoliation, and the observed response was small. The type of wounding affected this increase in salicortin, with natural wounding by insects causing a greater response than artificial wounding in one S. myrsinifolia clone, and artificial wounding causing a greater response than insect wounding in the other clone. This result indicates that S. myrsinifolia cannot control the effects of diffeeent types of wounding on its leaf secondary chemistry. We also found no indication of airborne warning signals between wounded and unwounded plants that trigger an elevation of leaf defenses in unwounded plants in anticipation of herbivore attack.  相似文献

Recently two species of carabid beetle were accidentally introduced onto the sub-Antarctic island of South Georgia. Both species are carnivorous and flightless. One of the species, Trechisibus antarcticus, is locally very abundant and in the process of invading the coastal lowland area, where the endemic herbivorous beetle Hydromedion sparsutum (Perimylopidae) is common. Field samples showed the abundance of the endemic species to be much lower, and its adult body size to be larger, in carabid-infested locations than in carabid-free locations. The sample data allowed us to estimate the growth rate of the H. sparsutum larvae and to reconstruct the most likely life-cycle of both species. A laboratory experiment showed a high mortality for the first three (out of six) larval instars of H. sparsutum in groups which had been subjected to predation by T. antarcticus. The duration of the period during which the larvae are vulnerable to predation was shown in a growth experiment to depend on food type. Quantitative and qualitative aspects of the interaction between the introduced predator and the endemic prey, and conditions which allowed the former to invade are discussed.  相似文献

Castor (cv. DCS-9) has been transformed through Agrobacterium-mediated and particle gun bombardment methods using appropriate vectors containing the Bt chimeric gene cry1EC driven by enhanced 35S promoter. About 81 and 12 putative transformants were regenerated following selection on hygromycin and kanamycin, respectively. Southern analysis of DNA extracted from T0 plants confirmed integration of the introduced gene in castor genome. The integration and inheritance of the introduced genes was demonstrated up to T4 generation by PCR and Southern analysis. Southern analysis of two events having single and two copies showed the same pattern of integration in the subsequent generations. Insect feeding experiments conducted in the laboratory by releasing neonate larvae of castor semilooper and S. litura on leaf tissues excised from transgenic and control plants showed varying degrees of larval mortality and slow growth in larvae fed on transgenic leaf tissue. Field bioassays against Spodoptera litura and castor semilooper conducted for eight events in T1–T4 generations under net confinement were more informative and events conferring resistance to the two major defoliators were identified.  相似文献

【目的】测定金龟子绿僵菌(Metarhizium anisopliae)对斜纹夜蛾(Spodoptera litura) 2龄幼虫的毒力,研究金龟子绿僵菌侵染后寄主体内抗氧化酶活性和肠道内细菌群落的变化,探讨斜纹夜蛾对金龟子绿僵菌侵染的防御机制。【方法】采用浸渍法测定不同浓度金龟子绿僵菌对斜纹夜蛾2龄幼虫的毒力;应用IlluminaMiSeq高通量测序技术测定肠道细菌群落。【结果】不同浓度的孢悬液对斜纹夜蛾2龄幼虫均有一定的毒力,处理7 d时半致死浓度(LC50)为3.944 107个孢子/mL;浓度为1.0×109个孢子/mL时,半致死时间最短(LT50)为4.6 d,校正后的死亡率为81.03%。处理后未致死的斜纹夜蛾幼虫体内抗氧化酶活性显著高于对照组。处理后致死的斜纹夜蛾幼虫肠道细菌群落多样性显著高于对照组;且处理后致死的斜纹夜蛾幼虫肠道细菌群落组成与对照组差异显著。【结论】金龟子绿僵菌对斜纹夜蛾幼虫的致死率和致死效率与金龟子绿僵菌的浓度呈正相关;斜纹夜蛾幼虫体内的抗氧化酶可能在抵抗金龟子绿僵菌侵染的过程中起重要作...  相似文献

茉莉酸甲酯(Methyl jasmonate, MeJA)与植物间的防御通讯密切相关,其挥发性强,可以在空气中传播而诱导邻近植物产生防御反应,为探明MeJA诱导邻近植株抗虫性的浓度和距离效应,本研究在大棚(长4 m×宽1 m×高1 m)进口处喷以不同浓度MeJA,检测离气味源60 cm、120 cm、180 cm、240 cm和300 cm远的番茄植株叶片多酚氧化酶(Polyphenol oxidase, PPO),过氧化物酶(Peroxidase, POD)和脂氧合酶(Lipoxygenase, LOX)的活性以及斜纹夜蛾幼虫取食不同处理番茄植株叶片后的体重增长率,研究结果表明1 mmol/L和10 mmol/L MeJA挥发物可以显著诱导增强邻近番茄植株叶片PPO,POD和LOX酶的活性,以离10 mmol/L MeJA气味源60 cm处植株叶片的酶活性最高,并随着距离的增加而呈降低的趋势;取食60 cm和300 cm对照组植株叶片的斜纹夜蛾幼虫其体重增长率没有显著差异,然而1 mmol/L和10 mmol/L MeJA处理下,取食300 cm处植株叶片的斜纹夜蛾幼虫其体重增长率显...  相似文献

White clover growing in New Zealand is experiencing increasing levels of ultraviolet-B (UV-B) radiation as a result of ozone depletion. We evaluated the effects of UV-B radiation on the foliar chemistry of two populations of white clover (Trifolium repens L.), ’Huia’ and ’Tienshan,’ and the consequences for the performance of armyworms (Spodoptera litura) and cutworms (Graphania mutans). Plants were grown in controlled environment rooms with and without supplemental UV-B radiation at a dose of 13.3 kJ m–2 day–1, corresponding to a 25% mid-summer ozone depletion above Palmerston North, New Zealand. In both white clover populations, UV-B radiation elicited changes in foliar chemistry, including slight increases in nitrogen concentrations and decreases in carbohydrate concentrations. In addition, the ’Huia’ population showed decreases in fiber concentrations and marked increases in cyanogenic activity. No change in UV-absorbing compounds was detected in either population. Long- and short-term feeding trials were conducted to assess dietary effects on insect growth, consumption, and food utilization. Changes in the performance of both insect species were generally small. The most pronounced effect was a 36% reduction in weight of S. litura after 2 weeks of feeding on Huia grown at high UV, but larval development times were only slightly prolonged and pupal weights were unaffected. S. litura short-term performance was affected by differences in white clover population. The long-term performance of G. mutans was not affected and its short-term performance (stadium duration and consumption rate) was only marginally affected by the high-UV treatment. We conclude that the effects of elevated UV-B radiation on white clover plant chemistry can be specific to certain plant populations. The differences in sensitivity of the two generalist insect species suggest that effects may also be specific to certain plant-herbivore associations. We have characterized, using several types of bioassays, the resistance induced in young tomato plants by feeding of the corn earworm, Helicoverpa zea. Beet armyworm larvae, Spodoptera exigua, and leafminers, Liriomyza trifolii, were used to assay the induced resistance. In whole-plant experiments, damage localized to a single leaflet of fourleaf tomato plants induced a systemic increase in resistance such that beet armyworm larvae confined to previously damaged (induced) plants grew at a rate about half that of larvae raised on control plants and consumed less leaf tissue from induced plants than from control plants. In experiments using excised leaves, beet armyworm larvae suffered increased mortality when reared on leaves from induced plants. The strength of this induced resistance varied spatially relative to the damaged position; moreover, the spatial distribution of induced resistance changed over a three-week period following damage. Other experiments demonstrated that the mechanisms of induced resistance in tomato foliage involves both a decrease in larval preference for and a decrease in the nutritional value of induced foliage. Induction also retarded the oviposition and/or early development of leafminers. Thus, induced resistance has relatively severe effects on the biology of subsequent herbivores. These data should allow us to begin to elucidate cause-effect relationships between induced resistance and induced chemistry in tomato plants.  相似文献

Bacillus thuringiensis (Bt) is a microbial pesticide widely used to control crop pests. Its strains have good biocontrol activity against crop insect pest, but lack some desirable characteristics that are found in Bacillus subtilis. An attempt has been made to combine those desirable characteristics; we used a highly effective biocontrol strain of B. thuringiensis in protoplast fusions with a strain of B. subtilis. The fusants were identified through cell culture and stained with crystal violet. The Bt and B. subtilis protoplasts were induced to fuse by PEG 6000. The fusants were produced almost 95% mortality in first instar larvae of Spodoptera litura. The lethal doses (The LC50 and LC90) for mortality of S. litura values were significantly in lower level in the fusant-treated larvae, when compared with Bt and B. subtilis individual treatment. The consumption and digestion of S. litura significantly decreased after treatment with fusant. Also the approximate digestibility of S. litura increased significantly.  相似文献

Nucleotide sequence encoding the truncated insecticidal Cry1Ca1 protein from Bacillus thuringiensis was extensively modified based on the codon usage of rice genes. The overall G + C contents of the synthetic cry1Ca1 coding sequence were raised to 65% with an additional bias of enriching for G and C ending codons as preferred by monocots. The synthetic gene was introduced into the Chinese japonica variety, Xiushui 11, by Agrobacterium-mediated transformation. Transgenic rice plants harboring this gene were highly resistant to Chilo suppressalis and Spodoptera litura larvae as revealed by insect bioassays. High levels of Cry1Ca1 protein were obtained in the leaves of transgenic rice, which were effective in achieving 100% mortality of S. litura and C. suppressalis larvae. The levels of Cry1Ca1 expression in the leaves of these transgenic plants were up to 0.34% of the total soluble proteins. The larvae of C. suppressalis and S. litura could consume a maximum of 1.89  and 4.89 mm2 of transgenic leaf area whereas the consumption of non-transgenic leaves by these larvae was significantly higher; 58.33 and 61.22 mm2, respectively. Analysis of R1 transgenic plants indicated that the cry1Ca1 was inherited by the progeny plants and provided complete protection against C. suppressalis and S. litura larvae.  相似文献

Spodoptera litura (Fabricius) (Lepidoptera: Noctuidae), known as common cutworm, is a worldwide pest that causes severe damage to various crops and vegetables in South Korea. In this study, we predicted the potential distribution of S. litura in South Korea in a climate change scenario by applying species distribution modeling. We used the CLIMEX software as a main tool and determined optimal parameter values to simulate the current distribution of S. litura in Asia. We then used these parameter values to predict the species' future distribution in South Korea. As a result, we prepared maps indicating areas with suitable climate for S. litura and showed that these areas gradually increased as a result of climate change. Approximately 98% of the areas in South Korea were predicted to have a favorable climate for S. litura in 2100; 63.2% of the area in South Korea is currently favorable. To the best of our knowledge, this is the first study to predict the potential distribution of S. litura in South Korea, and it provides the basic data necessary to establish an optimal control strategy of this species.  相似文献

Herbivore feeding may induce an array of responses in plants, and each response may have its own temporal dynamics. Precise timing of these plant responses is vital for them to have optimal effect on the herbivores feeding on the plant. This study measured the temporal dynamics of various systemically induced responses occurring in Brassica juncea (L.) Czern. (Brassicaceae) leaves after insect herbivory in India and The Netherlands. Morphological (trichomes, leaf size) and chemical (glucosinolates, amino acids, sugars) responses were analysed. The effects of systemic responses were assessed using a specialist [Plutella xylostella L. (Lepidoptera: Plutellidae)] and a generalist [Spodoptera litura Fabricius (Lepidoptera: Noctuidae)] herbivore. We tested the hypotheses that morphological responses were slower than chemical responses and that generalist herbivores would be more affected by induced responses than specialists. Glucosinolates and trichomes were found to increase systemically as quickly as 4 and 7 days after herbivore damage, respectively. Amino acids, sugars, and leaf size remained unaffected during this period. The generalist S. litura showed a significant feeding preference for undamaged leaves, whereas the specialist herbivore P. xylostella preferred leaves that were damaged 9 days before. Performance bioassays on generalist S. litura revealed that larvae gained half the weight on leaves from damaged plants as compared to larvae feeding on leaves from undamaged plants. These studies show that although morphological responses are somewhat slower than chemical responses, they also contribute to induced plant resistance in a relatively short time span. We argue that before considering induced responses as resistance factors, their effect should be assessed at various points in time with both generalist and specialist herbivores.  相似文献

A new species of Gladioferens is described from a freshwater lake in the Bunger Hills, Greater Antarctica. A key to the six species of Gladioferens is provided for both sexes. Most species of the subgenus containing G. antarcticus sp. nov. are remarkable in possessing asymmetries in the second legs (male) and fourth legs (female). This suggests that the mating behaviour of these species may be radically different from that in most heterarthrandrian calanoids and calls for further study. However, G. antarcticus is more primitive than its consubgeners in having, in the male, symmetrical second legs and 3-segmented endopods in the fifth legs. The genus shows a highly disjunct distribution (Australia, New Zealand and Bunger Hills) and its biogeography is now of outstanding interest.  相似文献

Transgenic cotton lines were developed for high-level expression of a synthetic cry1EC gene from a wound inducible promoter. The tobacco pathogenesis related promoter PR-1a was modified by placing CaMV35S promoter on its upstream in reverse orientation. The resultant chimeric promoter CaMV35S(r)PR-1a expressed constitutively and was further up-regulated at the site of feeding by insects. It was induced more rapidly by treatment with salicylic acid (SA). The CaMV35S(r)PR-1a cry1EC expressing transgenic lines of cotton showed 100% mortality of Spodoptera litura larvae. The tightly regulated low-level expression of PR-1a was modified to a highly expressing constitutive expression by CaMV35S placed in reverse orientation. Salicylic acid treatment and wounding enhanced the expression further by the chimeric promoter. The leaves expressed more δ-endotoxin around the sites of insect bites. The levels of expression and induction varied among different transgenic lines, suggesting position effect. Some of the transgenic lines that expressed Cry1EC from the chimeric promoter at a low level also showed 100% mortality when induced with salicylic acid. A highly expressing insect bite and wound inducible promoter is desirable for developing insect resistant transgenic plants.  相似文献

Four tomato accessions, namely Ac 238, Roma, Seijima Jeisei and Varushanadu Local selected from preliminary screening of 321 accessions and their hybrid derivatives, namely HY1F1 (Varushanadu Local × Ac 238), HY2F1 (Varushanadu Local × Roma), HY3F1 (Ac 238 × Roma) (first generation), HY1F2, HY2F2 and HY3F2 (second generation) were evaluated for their resistance in comparison with a susceptible check, 1979 against the leaf caterpillar, Spodoptera litura Fab. both under field and laboratory conditions at Annamalainagar, Tamil Nadu, India. In the field evaluation, accession Seijima Jeisei and hybrid HY3F1 recorded the minimum larval population. In glasshouse and laboratory studies, Seijima Jeisei was the least preferred by S. litura for both feeding and oviposition. The hybrid HY3F1 exerted pronounced antibiosis effect on S. litura. Among the various biophysical and biochemical bases of resistance, contents of reducing sugar and phenol were found negatively correlated with S. litura larval feeding. Among the hybrids, HY3F1 and HY3F2 and the parent Seijima Jeisei recorded higher fruit yield per plant in both seasons. Considerable variation in the resistance traits of the hybrids was observed when compared with the parents.  相似文献

The South American tree Solanum mauritianum Scopoli (Solanaceae), a major environmental weed in South Africa and New Zealand, has been targeted for biological control, with releases of agents restricted to South Africa. The leaf-sucking lace bug, Gargaphia decoris Drake (Tingidae), so far the only agent released, has become established in South Africa with recent reports of severe damage at a few field sites. To evaluate the insect’s suitability for release in New Zealand, host-specificity testing was carried out in South Africa in laboratory and open-field trials, with selected cultivated and native species of Solanum from New Zealand. No-choice tests confirmed the results of earlier trials that none of the three native New Zealand Solanum species are acceptable as hosts. Although the cultivated Solanum muricatum Aiton and S. quitoense Lam. also proved unacceptable as hosts, some cultivars of S. melongena L. (eggplant) supported feeding, development and oviposition in the no-choice tests. Although eggplant was routinely accepted under laboratory no-choice conditions in this and previous studies, observations in the native and introduced range of G. decoris, open-field trials and risk assessment based on multiple measures of insect performance indicate that the insect has a host range restricted to S. mauritianum. These results strongly support the proposed release of G. decoris in New Zealand because risks to non-target native and cultivated Solanum species appear to be negligible.
