Regulation of in vitro shoot and root formation and the histologicalorigin of newly formed shoots was studied in the apple cultivarÅkerö. Both composition of mineral elements and benzylaminopurine(BAP) concentration affected shoot multiplication. Similar numbersof shoots were obtained with Lepoivre and MS medium after twosucceeding subcultures, but Murashige and Skoog medium was preferabledue to production of longer shoots. The optimum BAP concentrationwax around 8.8 µM. Higher concentrations caused vitrifiedshoots. The rooting ability increased with numbers of subcultures.Also the concentration of indol-3-yl butyric acid (IBA) affectedrooting. A strong interaction between numbers of subculturesand IBA concentration was obtained. After insufficient numbersof subcultures, when shoots were still difficult to root, increasingIBA concentration exerted little effect on rooting. When shootshad reached an ‘easy-to-root condition’ root initiationdepended on IBA concentration, showing an optimum at 2.5 µM.Supraoptimal IBA concentrations delayed root initiation. Dark treatment of shoots during the root-initiation phase increasedrooting ability. The most positive effect was obtained at suboptimalIBA concentrations. Anatomical studies revealed both axillaryand adventitious shoots. Two kinds of adventitious structureswere demonstrated. Extracts of small and mature-size lupin pods yielded four substancesaffecting the growth of wheat-coleoptile sections: one acidpromotor (A), two acid inhibitors(B and X), and one neutralinhibitor(Y). Inhibitor B was extremely active, however, coleoptile sectionsshowed no signs of toxic effects; they resumed growth at a rapidrate after rinsing them and adding ß-indolylaceticand (IAA) to the medium. 1 µg of IAA was required to counteractthe effect of ‘B’ extracted from 230 mg. Of tissue.On an equal fresh weight basis the inhibiting action of ‘B’in lupin pods was 500–1,500 times more potent than thatof ‘inhibitor ß’ in etiolated pea seedlings. Small pods of plants infected with pea-mosaic virus yielded3 times the amount of ‘A’ of healthy plants (equivalentto 1 µg. IAA 0.3 µg. IAA per 25 g. of tissue respectively),and approximately the amount of ‘B’. Mature podsof virus-infected plants again yielded more‘A’,but also 2? times more ‘B’ than pods of healthyplants. Healthy pods yielded more ‘A’ than virus-infectedpods, and there was no difference in ‘X’. A lupin abscission test was developed and the effects of proximaland distal application of -naphthyl acetic acid (NAA) are presented,and discussed with respect to results of other abscission tests. ‘A’ accelerated abscission when applied proximally,and delayed or prevented it when applied distally. ‘B’strongly accelerated abscission when applied in either way.A possible mechanism explaining the abscission-inducing effectof developing pods on later flowers is discussed in terms ofthe substances ‘A’ and ‘B’. The partlyprevented abscission observed on virus-infected plants was foundto agree well with the proposed mechanism.  相似文献

High-frequency Organogenesis from Direct Seed Culture in Lathyrus   总被引:2,自引:0,他引:2  
Culture conditions were developed for inducing a high frequencyof direct shoot morphogenesis and whole plant regeneration incultures of intact seedlings of Lathyrus cicera L., L. ochrus(L.) DC., L. sativus L., and L. tingitanus L. The procedureof shoot regeneration involved culturing of whole, mature seedson MS medium containing cytokinins, or thidiazuron (TDZ), asubstituted phenylurea with cytokinin-like activity. Differentiationof shoots occurred without an intervening callus phase fromthe cotyledonary node and surrounding tissues of the intactseedlings developed from seeds germinated on media containingkinetin, BAP or TDZ. An average of 19·0, 15·8,28·8 and 43·0 shoots were regenerated at optimalconcentrations of 50·0, 50·0 and 80·0 µMBAP in L. cicera, L. tingitanus, L. ochrus and L. sativus, respectively.TDZ enhanced the shoot formation at the concentration of 10µM to an average of 33·1 and 33·7 shootsper seedling in L. cicera and L. tingitanus and at 50 µM,to 57·4 shoots per seedling in L. sativus. Regeneratedshoots developed roots on a modified MS medium containing 2·5µM NAA and the surviving plantlets were transferred tosoil. Histological studies revealed de novo formation of shoot budsfrom the epidermal or subepidermal cells of the basal and nodalregion of multiple epicotyls. Several meristematic centres consistingof actively-dividing cells developed in the subepidermal celllayer of the nodal tissue and adjacent areas within 7 d of seedculture on a cytokinin- or TDZ-supplemented medium. The morphogenic response of thin cell layers (TCLs) from vegetativetobacco (Nicotiana tabacum L.) plants can be directed very preciselyby varying the concentrations of benzyladenine (BA) and -naphthaleneacetic acid (NAA) in the culture medium. Medium containing 1·6µM BA and 0·5 µM NAA was optimal for shootformation, concentrations of 0·5 µM BA and 1·6µM NAA were optimal for the induction of shoots and rootson the same explant, whereas concentrations of NAA higher than16 µM resulted in callus proliferation only. Polarityin the distribution of the shoot buds was observed, i.e. a switchfrom basal to apical shoot formation occurred with increasingNAA concentrations, suggesting basipetal transport of NAA. Histologicalexamination of TCLs on shoot induction medium revealed thatfirst cell divisions occurred within 2 d in cortical cells whichwere directly in contact with the medium along the longitudinalcut surface, and after 2 d in subepidermal cells along the lateraledges of the explants. Individual lateral buds originated fromone subepidermal and one or more epidermal cells, while apicalbuds originated from single subepidermal or cortical cells locateddirectly at the apical end of the explant. After culture ofTCLs for 2-3 d on root/shoot induction medium cells in the regeneration-competentsubepidermis elongated, while on callus induction medium subepidermalcells elongated and dedifferentiated. Controlled environments were used to establish the base-linegrowth and to evaluate the photoperiodic response of two Tossajute (Corchorus olitorius) cultivars. Fresh and dry weightsof shoots and roots, leaf areas and stem lengths were much greaterat long day (LD) 12·5, 11·5 than LD 11·5,12·5 photoperiods for both cultivars. Root lengths weregreater at LD 12·5, 11·5 initially but were longestat LD 11·5, 12·5 at 35 days from emergence. Meanrelative growth rates (MRGR) were similar irrespective of photoperiodor cultivar but declined with plant age. The superior growthof cultivar ‘Yaya’ at LD 12·5, 11·5was due to more growth in the first 7 days, while, for cultivar‘Angbadu’ it was a combination of growth for 7 days,and higher MRGR up to 21 days. A comparison was made of effects of ‘low’ and ‘high’soil temperature (LST and HST, about 9 and 21·5 °Crespectively) on shoot growth of Norwegian and Portuguese populationsof Dactylis glomerata. In experiments lasting 8 short days (SD,8 h photoperiods) LST decreased leaf extension more markedlyin the Portuguese population. No differential effect of LSTon leaf growth was recorded in experiments lasting 20 or 21SD or in experiments of 8 d duration in long days (LD, 16 hphotoperiods). Since the meristem and region of cell extensionis close to the soil surface LST could directly influence bothroot and shoot growth. The application of gibberellic acid enhanced leaf extension,particularly in plants grown at HST. 6-Benzylaminopurine tendedto decrease leaf length. Extractable gibberellin levels wererelatively low after 8 SD in shoots of both races grown at LST.Cytokinin levels increased at LST, more so in shoots of Portugueseplants which sustained the greater reduction in leaf extension. The data suggest that LST may reduce the production of endogenousgibberellins important for leaf growth in Dactylis glomerata.Cytokinins are probably necessary for growth processes but theirlevel may reflect, rather than direct, the rate of leaf growth.  相似文献

In this further study of light as an environmental factor theeffects of shading on the growth of Helianthus annuus, Fagopyrumesculentum, Trifolium subterraneum, Tropaeolum majus, Lycopersicumesculentum, Vicia faba, Pisum sativum, Hordeum vulgare, Solanumdulcamara, and Geum urbanum have been investigated. It has been established that the reductions in the net assimilationcaused by shading are similar for all ten species. The net assimilationrate during the season of active growth is linearly relatedto the logarithm of the light intensity. When similar experimentsare conducted late in the autumn and the relative growth rateis very low, the logarithmic relationship no longer holds. From the data it is possible to obtain precise estimates ofthe compensation point. The mean values of the compensationpoint ranged from 0·06 to 0·09 of daylight foreight species, while for V. faba and H. vulgare somewhat higherfigures were obtained—0·14 and 0·18 of daylight. Flctuations in the net assimilation rate in full daylight showedno correlation with variations in the value of the compensationpoint. From these results it is concluded that species cannot be groupedinto ‘sun’ or ‘shade’ plants, eitheron the basis of differences in the value of the compensationpoint or on the grounds that there are large variations in theeffects of shading on net assimilation rate.  相似文献

Barley (Hordeum vulgare L., cvs Golf, Mette, and Laevigatum)was grown under nitrogen limitation in solution culture untilnear maturity. Three different nitrogen addition regimes wereused: in the ‘HN’ culture the relative rate of nitrate-Naddition (RA) was 0·08 d–1 until day 48 and thendecreased stepwise to, finally, 0·005 d–1 duringgrain-filling; the ‘LN’ culture received 45% ofthe nitrogen added in HN; the ‘CN’ culture was maintainedat RA 0·0375 d–1 throughout. Kinetics of net nitrateuptake were measured during ontogeny at 30 to 150 mmol m–3external nitrate. Vmax (which is argued to reflect the maximuminflux rate in these plants) declined with age in both HN andLN cultures. A pronounced transient drop was observed just beforeanthesis, which correlated in time with a peak in root nitrateconcentration. Similar, but less pronounced, trends were observedin CN. The relative Vmax (unit nitrogen taken up per unit nitrogenin plants and day) in all three cultures declined from 1·3–2·3d–1 during vegetative growth to 0·1–0·7d–1 during generative growth. These values are in HN andLN cultures 15- to more than 100-fold in excess of the demandset by growth rates throughout ontogeny. Predicted balancingnitrate concentrations (defined as the nitrate concentrationrequired to support the observed rate of growth) were below6·0 mmol m–3 in HN and LN cultures before anthesisand then decreased during ontogeny. In CN cultures the balancingnitrate concentration increased during grain-filling. Apartfrom the transient decline during anthesis, most of the effectof ageing on relative Vmax can be explained in terms of reducedcontribution of roots to total biomass (R:T). The loss in uptakeper unit root weight is largely compensated for by the declinewith time in average tissue nitrogen concentrations. The quantitativerelationships between relative Vmax and R:T in ageing plantsare similar to those observed for vegetative plants culturedat different RAs. The data support the contention that the capacity for nitrateacquisition in N-limited plants is under general growth control,rather than controlled by specific regulation of the biochemicalpathway of nitrate assimilation. Field experiments using two soybean (Glycine max L. Merrill)cultivars (‘Elgin 87’ and ‘Essex’) wereconducted for 2 years near Lexington, KY, USA to evaluate theeffect of source-sink alterations on seed carbohydrate statusand growth. Sucrose concentrations in developing cotyledonsof control plants were consistently low (<50 m M) early inseed development, but they increased to 100–150 m M byphysiological maturity. The concentrations increased in bothyears by 47 to 59% when 90% of the pods were removed from ‘Elgin87’, but the increase had no effect on individual seedgrowth rate (SGR). Shading (80%) reduced cotyledon sucrose levelsand SGR in both years. The critical cotyledon sucrose concentration(the concentration providing 80% of the maximum cotyledon growthrate) was estimated fromin vitro cotyledon growth at sucroseconcentrations of 0–200 m M. These critical concentrationsvaried from 72–124 m M;in planta control cotyledon sucroseconcentrations were below this critical level during the firsthalf of seed growth but exceeded it in the later stages of growthin all experiments. The estimated critical concentration wasconsistent with the failure of in planta SGR to respond to anincrease in assimilate supply and with the reduction in SGRassociated with a decrease in assimilate supply. The resultssuggest that soybean SGR is generally sink limited if photosynthesisincreases during seed filling, but source limited if photosynthesisis reduced. An investigation has been made into the growth regulators presentin ethanol extracts of the seedling roots of Vicia faba afterseparation on paper partition chromatograms, using segmentsof Avena coleoptiles and mesocotyls and of Pisum sativum.rootsas biological assay material. Acetonitrile purification shows the presence of at least threeauxins running in isobutanol: methanol: water, at Rfs of 0–0·25,0·4–0·6, and 0·65–0·95;the latter may represent two different auxins. A similar, butclearer, picture is shown by the ether-soluble acid fraction.Here an auxin at Rf 0–0·25 also stimulates rootgrowth and could be ‘accelerator ’. A second atRf 0–0·25 is an indole compound which inhibitsroot growth and does not seem to be be IAA. A third at Rf 0·8–1·0is also a root-growth inhibitor and gives no indole reaction.The ‘inhibitor ß’ complex was demonstrated(Rf 0·65–0·85) together with a number ofother inhibitors at lower Rf value. The ether-soluble neutral component also contains auxins orauxin precursors. The water-soluble, ether-insoluble fraction contains four readilyinterconvertible substances with auxin properties. The success of Triticum aestivumxZea mays crosses, used to producewheat doubled haploids, is influenced by light intensity. Toexamine the basis for this response, pollen tube growth, embryosurvival and indicators of photosynthetic rate were measuredin two wheat cultivars (‘Karamu’ and ‘Kotuku’)crossed with maize at two irradiance levels (250 or 750 µmolm-2s-1, PAR). Pollen tube growth was significantly affectedby light intensity in ‘Karamu’ plants but not in‘Kotuku’ plants, despite both cultivars being pollinatedby the same maize source. The percentage of pollen tubes reachingthe cavity between the ovarian wall and integuments, or in themicropyle of ‘Karamu’ plants at high light intensity(65%) was nearly three-times greater than that at low lightintensity (22%). Thus, either low light intensity can affectthe maternal wheat plant in a way that inhibits pollen tubegrowth and/or high light intensity may promote pollen tube growthin ‘Karamu’ plants. Significant differences in ratesof electron transport in plants grown at the two light intensitiesindicated that the rate of photosynthesis may also have an effecton pollen tube growth. These results have importance for improvingthe efficiency of wheat x maize crosses and other wide cerealcrosses. During the growth of Isochrysis galbana, several non-proteinamines may be detected in the growth medium. Of these, one (termed‘TTl’) accumulates in proportion to the numbersof cells present. The concentrations of ‘TTl’, andof another (termed ‘TA’), are 3–5 times higherin cultures in which Isochrysis is predated by Oxyrrhis marina.The lowest estimates of the concentration of extracellular ‘TTl’are an order of magnitude higher than those of any protein aminoacid. Of the protein amino acids, some like glycine are utilizedduring predation while others, like histidine, accumulate inthe medium Because of the unknown N-content and reactivity ofthe non-protein amines during HPLC, it is not possible to sayif these compounds (together with other components of dissolvedorganic N) form a significant proportion of the unaccountedfor N in the system after predatory activity. During predationin the absence of detectable free ammonium (when Isochrysismay be expected to be N-deprived), particles accumulate in themedium. Most of these are <2.5 µ.m in diameter andare suggested to be remains of digested prey. There is evidenceof a reassimilation of these particles by prey-deplete Oxyrrhis.  相似文献

Zhang, J. and Davies, W. J. 1987. Increased synthesis of ABAin partially dehydrated root tips and ABA transport from rootsto leaves.—J. exp. Bot. 38: 2015–2023. Isolated root tips of pea (Pisum sativum L. cv. Feltham First)and Commelina communis L. were air-dried until they lost between10% and 40% of their fresh weight, followed by a period of incubationat these reduced water contents. These treatments resulted inincreased ABA production, suggesting that root tips of bothspecies have the capacity to synthesize ABA in increased amountswhen water deficits develop in the root. The ABA concentrationin pea roots increased linearly as turgors fell below about0·15 M Pa and relative water contents (R WC) fell below90%. Commelina roots produced more ABA when RWC fell below asimilar value but the threshold turgor for increased ABA productionin Commelina roots was around 0·30 MPa. Roots of intact plants loaded with ABA as a result of incubationin solutions of varying concentrations provided ABA to leaveswhich resulted in increased ABA concentrations in the leaveswhen these were assayed several hours later. This occurred whenthese roots were not contributing substantially to transpirationalflux. Leaves on shoots that were enclosed and darkened and thereforenot transpiring, did not accumulate ABA from ‘loaded’roots. This review compares new developmental models on flowering andother vascular plants with evolutionary hypotheses formulatedby Agnes Arber (1879–1960) and like-minded botanists.Special emphasis is laid on philosophical basics such as perspectivism,pluralism about evolutionary modelling, continuum way of thinking,and fuzzy logic. Arber's perspective is best labelled as F uzzyA rberian M orphology (FAM Approach). Its proponents (‘FAMmers’)treat structural categories (e.g. ‘roots’, ‘shoots’,‘stems’, ‘leaves’, ‘stipules’)in vascular plants as concepts with fuzzy borderlines allowingintermediates (including transitional forms, developmental mosaics).The FAM Approach complements Cla ssical Plant M orphology (ClaMApproach), which is the traditional approach in botany. ClaMproponents (‘ClaMmers’) postulate that the structuralcategories of vascular plants are regarded as concepts withclear-cut borderlines and without intermediates. However, duringthe evolution of vascular plants, the root-shoot distinctionand the stem-leaf distinction have become blurred several timesdue to developmental changes, resulting in organs with uniquecombinations of features. This happened, for example, in thebladderworts (Utricularia, Lentibulariaceae). When focusingon the ‘leaf’, the FAM Approach is identical toArber's ‘partial-shoot theory of the leaf’ and Sinha's‘leaf shoot continuum model’. A compound leaf canrepeat the developmental pathway of the whole shoot, at leastto some degree. For example, compound leaves of Chisocheton(Meliaceae)with indeterminate apical growth and three-dimensional branchingmay be seen as developmental mosaics sharing some growth processeswith whole shoots! We focus here on the FAM Approach becausethis perspective is especially promising for developmental geneticistsstudying flowering and other vascular plants. The effect of phosphorus supply (1–15 µM) on protonrelease and the role of P in symbiotic nitrogen fixation inmedic (Medicago truncatula L. ‘Jemalong’) was investigated.As P concentration in the nutrient solution increased, shootand root growth increased by 19 and 15%, respectively by day35, with maximal growth at 4 µM P. A P concentration of15 µM appeared to be toxic to plants. Phosphorus supplyhad no influence on nodule formation by day 12 but increasednodule number by day 35. Nitrogenase activity was estimatedby in situ measurement of acetylene reduction activity (ARA)in an open-flow system. During the assay, a C2H2-induced declineof ARA was observed under all P concentrations except 4 µM.Specific ARA (per unit nodule weight) doubled when P supplywas increased from 1 to 8 µM. This effect of P was muchgreater than the effects of P on nodulation and host plant growth.Concentrations of excess cations in plants decreased with increasingP concentration in the nutrient solution. Phosphorus deficiencystimulated uptake of excess cations over anions by the plantsand hence enhanced proton release. The results suggest thatP plays a direct role in nodule functioning in medic and thatP deficiency increases acidification which may facilitate Pacquisition. Significant differences in response to soil moisture stress(SMS) and restricted root zone volume (RRZV) were found in twocultivars of soybean [Glycine max (L.) Merr.] (‘Forrest’and ‘Williams’) plants grown under controlled-environmentconditions. Leaf water potentials of SMS-treated plants were0·4-0·6 MPa lower than those of controls and stomata1conductances 23-56% lower. In the case of RRZV treatment, however,there were no differences in either parameter. Initiation ofnew leaves as reflected in the plastochron index was stronglyreduced by SMS but was unaffected by RRZV. Photosynthetic rates(CO2 fixation dm2 of leaf) of plants given SMS were reducedby 11-21% while those of RRZV-treated plants were unaffected.SMS caused a strong preferential allocation of dry matter tothe root at the expense of the shoot in both cultivars. RRZV,however, had no effect on assimilate distribution in ‘Forrest’and only slightly favoured root growth in ‘Williams.’Carbohydrate concentrations of both alcohol-soluble and insolublefractions were increased significantly by SMS, especially inthe leaves, but were little affected by RRZV. Nitrogen concentrationin the root fraction was reduced by 22-24% and that in the leafand stem fractions by 7-14% under SMS but was not affected appreciablyby RRZV. Phosphorus concentration in the leaf, stem, and rootfractions was reduced by 45-65% under SMS but was relativelyunaffected by RRZV. These findings suggest that SMS and RRZVare basically different in their mechanism of action and thatthe impairment of growth resulting from these two stresses mayinvolve different physiological processes. Winged bean (Psophocarpus tetragonolbtis) seed coat in the hilarregion consists of a double layer of sclereids, tracheid barand spongy parenchyma cells. This is contrasted to the seedcoat structure on either side of the hilar region, which hasa single layer of sclereids, columnar cells and crushed parenchymacells. Cotyledonary cells are large (50 to 100µm in diameter)and have cell walls 2·4 to 4·7 µm thickwith pit-pair structures. Protein bodies and lipid bodies arethe main structural components of the cytoplasm while only asmall number of starch granules are present in each cell. Themajor portion of the lipid can be removed by non-polar solventsand contains oleic and linoleic acids as the predominant unsaturatedfatty acids. High levels of behenic acid were present in both‘free’ and ‘bound’ lipids. Psophocarpus tetragonolobus (L.)
