Fruit- and nectar-feeding bats have high energy demands because of the cost of flight, and sugar is a good fuel because it is easily digested and absorbed. This study investigated the digestive efficiency of different sugars at different concentrations in Wahlberg's epauletted fruit bat (Epomophorus wahlbergi). We predicted that the sugar type and concentration would affect the total amount of solution consumed, while the total energy gained and the apparent assimilation efficiency would be high, irrespective of sugar type or concentration. Equicaloric solutions of two sugar types, glucose and sucrose, at low (10%), medium (15%) and high (25%) concentrations were offered in separate trials to bats. Total amount of solution consumed, total energy gained from each solution, and apparent assimilation efficiency, were measured. Bats had higher total volumetric intake of glucose and sucrose at the low concentrations than at the higher concentrations. However, bats maintained similar total energy intake on the respective glucose and sucrose concentrations. Bats were found to have high assimilation efficiencies on both glucose and sucrose irrespective of concentration. As bats used both sugars efficiently to maximize and maintain energy gain, it is expected that they feed opportunistically on fruit in the wild depending on temporal and spatial availability to obtain their energy requirements. Furthermore, fruit with high sucrose or glucose content will be consumed.  相似文献

The objective of this study was to examine the potential toxicity of sucrose (Experiment 1) and of various cryoprotectants (Experiment 2) to porcine preimplantation embryos. In Experiment 1, 65 embryos, ranging from compact morulae to hatched blastocysts, were allocated within donor female across 5 concentrations of sucrose (0, 0.25, 0.50, 1.0, 2.0 M) to determine the highest concentration that would not inhibit subsequent embryo development. After a 48-h post-treatment culture period, the embryos were stained and cell nuclei were counted. The concentration of sucrose affected embryo development (P < 0.001) and embryo quality (P < 0.001). Embryos placed into 2.0 M sucrose exhibited poorer development and quality than embryos at the lower 4 concentrations, which were not different from one another. In Experiment 2, 182 embryos of the same developmental stages as in Experiment 1 were collected from 16 donors. Embryos were allotted within donor female to 2 of the 5 concentrations (10, 20, 30, 40, or 50%) of each of 3 cryoprotectants (ethylene glycol, propylene glycol, glycerol). After a 30-sec exposure to a cryoprotectant, the embryos were cultured and stained as in Experiment 1. As the concentration of an individual cryoprotectant increased beyond 30%, embryo development decreased. Embryos exposed to glycerol or propylene glycol exhibited poorer development than did embryos placed into ethylene glycol, especially at concentrations of 40% or higher.  相似文献

Inclusion of either 1 or 3% (w/v) bovine serum albumin (BSA) in 8.6, 10, or 12% sucrose enhanced the maintenance of equine sperm motility in vitro at 38 degrees C for 8 h. There was a trend toward higher percent motile spermatozoa (PMS) at 16 and 24 h of incubation in semen samples containing BSA than in those that did not. The highest concentration of sucrose (12%) was slightly less effective in supporting PMS than either of the lower concentrations. However, sucrose concentrations had no apparent effect on rate of forward movement (RFM) of spermatozoa. Pregnancy and foaling rates were similar for mares inseminated with semen extended in either cream gel or 3% BSA-10% sucrose.  相似文献

Although operant ethanol self-administration can increase accumbal dopamine activity, the relationship between dopamine and ethanol levels during consumption remains unclear. We trained Long-Evans rats to self-administer escalating concentrations of ethanol (with 10% sucrose) over 7 days, during which two to four lever presses resulted in 20 min of access to the solution with no further response requirements. Accumbal microdialysis was performed in rats self-administering 10% ethanol (plus 10% sucrose) or 10% sucrose alone. Most ethanol (1.6 +/- 0.2 g/kg) and sucrose intake occurred during the first 10 min of access. Sucrose ingestion did not induce significant changes in dopamine concentrations. Dopamine levels increased within the first 5 min of ethanol availability followed by a return to baseline, whereas brain ethanol levels reached peak concentration more than 40 min later. We found significant correlations between intake and dopamine concentration during the initial 10 min of consumption. Furthermore, ethanol-conditioned rats consuming 10% sucrose showed no effect of ethanol expectation on dopamine activity. The transient rise in dopamine during ethanol ingestion suggests that the dopamine response was not solely due to the pharmacological properties of ethanol. The dopamine response may be related to the stimulus properties of ethanol presentation, which were strongest during consumption.  相似文献

Nectarivorous bats include very dilute nectar in their natural diet, and recent work with Pallas's long-tongued bat Glossophaga soricina showed that sugar (energy) intake rate decreased at dilute sucrose solutions. However, chiropterophillous nectar is composed mainly of the hexoses glucose and fructose. Because bats fed hexose nectar would save the delay of hydrolyzing sucrose, we hypothesized that sugar intake rate should be higher on this diet than on sucrose nectar. We compared intake response in Pallas's long-tongued bats offered 1 : 1 glucose-fructose (hexose) and sucrose diets at 5%, 10%, 20%, 30%, and 40% (mass/volume) sugar solutions. We also tested the hypothesis that sucrose hydrolysis limits food intake in bats. Intake response was the same in bats fed both types of diet: sugar intake rate was lower in dilute solutions and then increased with sugar concentration. Similar intake responses in both diets indicate that sucrose hydrolysis alone does not limit food intake and support the idea that the burden of processing excess water in dilute solutions plays a major role.  相似文献

Weanling Sherman rats were pair-fed for 8 days on a control or a magnesium deficient diet containing 70.5% sucrose. After a 12-hour fast, the rats were injected intraperitoneally with glucose (250 mg/100 g body weight) and arterial blood was drawn at 0, 15, 30, 60, 90 minutes after injection. Before glucose loading, in magnesium deficient rats, plasma magnesium levels were significantly increased. The plasma triglyceride concentration was significantly higher in magnesium deficient rats compared to controls. After glucose loading, in the control group, the plasma insulin concentrations increased to 67.9 +/- 5.8 microU/ml at 15 minutes and returned to pretreatment levels by 30 minutes; in the magnesium-deficient rats, the plasma insulin levels were significantly lower at 15 minutes 32.9 +/- 5.6 microU/ml (P less than 0.01) and returned more slowly to the pre-challenge level. No significant differences were observed in plasma glucose levels between the two groups of rats.  相似文献

Plasma noncholesterol sterols in male distance runners and sedentary men   总被引:1,自引:0,他引:1  
Plasma lathosterol concentration is taken to be an index of the rate of cholesterol synthesis and plasma concentrations of plant sterols just as campesterol and betasitosterol are taken to be indeces of cholesterol absorption efficiency. These noncholesterol sterols were measured in plasma from 14 male distance runners and 10 sedentary men. Plasma lathosterol concentration was 30% lower (P less than 0.02) and plasma betasitosterol concentration was 33% higher (P less than 0.02) in the runners compared to the sedentary men. Plasma concentrations of lathosterol and plant sterols were inversely and significantly (P less than 0.05) correlated in both the runners and the sedentary men. Plasma plant sterol concentrations were correlated positively and significantly (P less than 0.01) with plasma high density lipoprotein cholesterol (HDL-C) concentrations in the runners and sedentary men combined. These findings suggest that more efficient cholesterol absorption may lead to higher plasma plant sterol concentrations and may contribute to lower cholesterol synthesis rates, reduced concentrations of plasma lathosterol and higher plasma HDL-C concentration in distance runners.  相似文献

Sugar content and freezing tolerance of protoplasts of Arabidopsis thaliana leaves were manipulated by incubating seedlings in a sucrose solution before protoplast isolation. Incubation in a 400 mM sucrose solution at 2 °C in the dark increased their freezing tolerance equivalent to that achieved after a conventional cold acclimation at 2 °C. The increased freezing tolerance was due to a decrease in the incidence of freeze‐induced lesions: expansion‐induced lysis (EIL) between ?2 and ?4 °C and loss of osmotic responsiveness (LOR) between ?5 and ?12 °C. The concentration of sucrose in the incubation medium required to minimize the incidence of the lesions was substantially different: 10–35 mM for EIL and 30–400 mM for LOR. Incubation in the sucrose solution at 23 °C decreased LOR only at ?5 and ?6 °C but less than that incubated at 2 °C, and there was no effect on EIL. Incubation in sorbitol solutions at 2 °C also decreased LOR at ?5 and ?6 °C but much less than in the sucrose solution. These results suggest that low concentrations of sucrose act as a metabolic substrate for the low‐temperature‐induced alterations required for the amelioration of EIL and, at higher concentrations, sucrose has a direct cryoprotective effect to minimize LOR.  相似文献

Preservation solutions for buffy coat-free red cell concentrates with sucrose concentrations from 234 decreasing up to 15 mmol per 1 solution were tested. The hemolysis rate increased from 0.5 up to 1.9% by decreasing the sucrose concentration. The red cell volume was unchanged at low sucrose concentrations. No differences were noticed in ATP content and morphological changes. A considerable extracellular pH shift at high sucrose concentration exists only at the beginning of storage. A sucrose concentration of 30-50 mmol/l solution (3-5 mmol per unit red cell concentrate) at an ionic strength of 0.16 proves to be most suitable.  相似文献

Both the lag period and the time required for the filament and sporoplasm to emerge from Nosema algerae spores were prolonged when germination occurred under hyperosmotic conditions. Polyethylene glycol (PEG) and sucrose inhibited germination, first by preventing eversion of the filament, and then at higher concentrations by preventing stimulation. The size of the spore cases decreased by about 21% following germination, indicating an elastic spore wall and turgor pressure in the dormant spores. Increased pressure during germination was indicated by less osmotically-induced shrinkage in stimulated than in dormant spores and by higher concentration of solutes in the homogenates of germinated than ungerminated spores. These results are consistent with the hypothesis of a pressure increase during germination that is caused by an endogenous increase in solute concentration.  相似文献

The production of ethanol and enriched fructose syrups from a synthetic medium with various sucrose concentrations using the mutant Saccharomyces cerevisiae ATCC 36858 was investigated. In batch tests, fructose yields were above 90% of theoretical values for the sucrose concentrations between 35 g/l and 257 g/l. The specific growth rates and biomass yields were from 0.218 to 0.128 h(-1) and from 0.160 to 0.075 g biomass/g of glucose and fructose consumed, respectively. Ethanol yields were in the range of 72 to 85% of theoretical value when sucrose concentrations were above 81 g/l. The volumetric ethanol productivity was 2.23 g ethanol/(l h) in a medium containing 216 g/l sucrose. Fructo-oligosaccharides and glycerol were also produced in the process. A maximum fructo-oligosaccharides concentration (up to 9 g/l) was attained in the 257 g/l sucrose medium in the first 7 h of the fermentation. These sugars started to be consumed when the concentrations of sucrose in the media were less than 30% of its initial values. The fructo-oligosaccharides mixture was composed of 6-kestose (61.5%), neokestose (29.7%) and 1-kestose (8.8%). The concentration of glycerol produced in the process was less than 9 g/l. These results will be useful in the production of enriched fructose syrups and ethanol using sucrose-based raw materials.  相似文献

Ants of the same size can vary their intake rate of a given sucrose solution depending on the colony's needs for carbohydrates. As this capacity has not yet been described for another insect, the question of how they can do that was the focus of our work. When viscosity and ant-morphometry remain constant, changes in intake rate can only be attributed to the sucking forces. The aim of this study was to analyze the nectar sucking activity in the ant Camponotus mus. Feeding behavior seems to be under motivational control; therefore, we developed a non-invasive experimental device. We recorded the electrical signal generated during nectar feeding by offering ants sucrose solutions of different concentrations (from 10%w/w to 70%w/w). The signal frequency was between 2 and 12 peaks/s. We could distinguish two different patterns of electrical signal during feeding depending on the solution concentration. Only the more concentrated solutions reached frequencies higher than 7 peaks/s and the signal performance was quite irregular. For the other concentrations (10%, 30% and 50%), signal frequencies were lower than 6 peaks/s and the signal pattern was sinusoidal, regular and decreased with intake in all cases. We discuss the possible implications of these two signal patterns.  相似文献

Summary Body temperature and duration of foraging activities were affected by the concentration of sucrose solution imbibed. When experienced foragers of Apis mellifera arrived at a gravity feeder from the hive, thoracic temperature (TTH) was independent of sucrose concentration (X = 36.3 °C). While imbibing 40% and 60% (g solute per g of solution) solutions bees maintained TTH at approximately the same high level as upon arrival, but those imbibing 10%, 20%, and 30% solutions regulated TTH lower (X = 33.5 °C). All bees departed the feeder for the hive at the same TTH (X = 36.1 °C). Bees that imbibed 40% and 60% solutions sometimes immediately took flight after imbibition and averaged less than 15 s to takeoff. Time to takeoff was 2–3 times longer for bees that had imbibed 10% and 20% solutions because warmup preceded takeoff. The rate of energy expenditure at TTH=36.3°C (at 40% and 60% solutions) was 20% greater than that at 33.3°C (at 10%, 20%, and 30% solution). Bees that fed on the highly concentrated solutions regulated TTH so that rate of net energy gain was enhanced, but bees that fed on less concentrated solutions could have increased rate of net gain by maintaining a higher TTH which would have reduced time required for takeoff. Experiments were conducted to develop a simple rapid-freezing protocol for mature mouse oocytes that would yield a high proportion of oocytes with developmental potential. The effects of concentration (3.5, 4.5 and 6.0 M dimethyl sulfoxide (DMSO) all with 0.5 M sucrose) and the duration of exposure (2.5 min vs 45 sec) of oocytes to the cryoprotectant and its extraction after thawing in 2, 3 or 4 steps of descending sucrose concentration were studied. The most effective of the rapid-freezing and thawing protocols (4.5 M DMSO; 45 sec exposure and 3-step thawing) was compared to slow freezing protocols using 1.5 M DMSO and 1.0 M 1,2 propanediol as cryoprotectants. The DMSO concentrations had an effect on survival, fertilization and embryo development using short (45 sec) but not long (2.5 min) exposure. The rate of morphological oocyte survival was significantly higher using 4.5 M DMSO than 3.5 or 6.0 M (92% vs 82 and 73%, respectively). The development of fertilized embryos to blastocysts was also significantly higher at 4.5 M than at 3.5 or 6.0 M (68% vs 42 and 53%, respectively). The extraction of cryoprotectant in 3 or 4 steps of descending sucrose concentration resulted in higher survival (P < 0.01) and fertilization than in 2 steps. The best survival, fertilization and development was achieved with the 3-step procedure. Optimal combinations of conditions were 4.5 M DMSO at 45 sec prefreeze exposure and 3-step extraction of the cryoprotectant. Oocytes frozen by conventional methods had a survival, fertilization and development to blastocyst rate significantly lower than those frozen under the optimal rapid conditions. Thus rapid freezing of mature mouse oocytes with 4.5 M DMSO + 0.5 M sucrose and short prefreeze exposure is effective and has the additional advantage of being less time-consuming than slow freezing methods.  相似文献

Roux-en-Y gastric bypass (RYGB) surgery has been shown to decrease consummatory responsiveness of rats to high sucrose concentrations, and genetic deletion of glucagon-like peptide-1 receptors (GLP-1R) has been shown to decrease consummatory responsiveness of mice to low-sucrose concentrations. Here we assessed the effects of RYGB and pharmacological GLP-1R modulation on sucrose licking by chow-fed rats in a brief-access test that assessed consummatory and appetitive behaviors. Rats were tested while fasted presurgically and postsurgically and while nondeprived postsurgically and 5 h after intraperitoneal injections with the GLP-1R antagonist exendin-3(9-39) (30 μg/kg), agonist exendin-4 (1 μg/kg), and vehicle in 30-min sessions during which a sucrose concentration series (0.01-1.0 M) was presented in 10-s trials. Other rats were tested postsurgically or 15 min after peptide or vehicle injection while fasted and while nondeprived. Independent of food-deprivation state, sucrose experience, or GLP-1R modulation, RYGB rats took 1.5-3× as many trials as sham-operated rats, indicating increased appetitive behavior. Under nondeprived conditions, RYGB rats with presurgical sucrose experience licked more to sucrose relative to water compared with sham-operated rats. Exendin-4 and exendin-3(9-39) impacted 0.3 M sucrose intake in a one-bottle test, but never interacted with surgical group to affect brief-access responding. Unlike prior reports in both clearly obese and relatively leaner rats given RYGB and in GLP-1R knockout mice, we found that neither RYGB nor GLP-1R blockade decreased consummatory responsiveness to sucrose in our less obese chow-fed rats. Collectively, these results highlight the fact that changes in taste-driven motivated behavior to sucrose after RYGB and/or GLP-1R modulation are very model and measure dependent.  相似文献

Cell cultures of Perilla frutescens, growing on Linsmaier and Skoog medium, released more anthocyanin with 40 to 50 g sucrose/l compared with the control of 30 g sucrose/l of medium. More proteins were also released into the medium with the higher sucrose concentration and the cell volume was smaller, suggesting that the higher osmotic pressure, caused by the high sucrose concentration, may cause these releases. The capacitance of the cultured cells indicated a difference in membrane structure between the cells cultivated with different sucrose concentrations, supporting the hypothesis that cell permeabilization is increased at higher sucrose concentrations.J.-J. In vitro germination of 20-day old immature ovules of Impatiens platypetala Lindl. was inhibited at concentrations as low as 50 mM sucrose or mannitol and 100 mM glucose. Younger ovules (12, 14, and 16 days old) were similarly inhibited at 100 mM sucrose.Inorganic nitrogen concentration did not affect germination regardless of ovule age, but seedling fresh weight was significantly less and abnormal development of seedlings was significantly increased by total inorganic nitrogen concentrations higher or lower than 30 mM (at a ratio of 20: 10 mM NO3 -: NH4 +) in the culture medium.  相似文献

提高解冻胚胎的发育能力和简化冷冻解冻程序是胚胎冷冻研究的两大永恒的主题。尽管乙二醇(EG)广泛用于家畜胚胎冷冻,但很少用于冷冻小鼠和人胚胎。为数很少的以EG慢冻小鼠或人胚胎的研究均采用较为复杂的人胚冷冻程序,未见简化程序和用EG冷冻小鼠桑椹胚的报道。采用简单的牛胚胎冷冻程序研究了发育时期、EG浓度、平衡方法、添加蔗糖以及解冻后脱除EG等对小鼠胚胎冻后发育能力的影响。结果显示:(1)致密晚期桑椹胚冻后体外培养囊胚发育率(81.92%±2.24%)和孵出率(68.56%±2.43%)显著(P<0.05)高于4-细胞、8-细胞胚胎和致密早期桑椹胚胎;(2)1.8mol/L EG冷冻小鼠致密晚期桑椹胚的囊胚发育和孵出率显著高于其它浓度;(3)在EG中平衡10min的冻后囊胚发育显著好于平衡5、20或30min;(4)两步平衡冷冻胚胎的囊胚发育率和孵出率显著高于一步平衡;(5)用EG冷冻小鼠胚胎无需添加蔗糖;(6)解冻后可不脱除EG;(7)冻后发育的早期囊胚和囊胚细胞数明显少于体内发育胚胎。因此,用EG冷冻小鼠胚胎的最佳方案为:致密晚期桑椹胚用1.8mol/L EG不添加蔗糖、两步平衡15min、以简单的牛胚胎冷冻程序冷冻解冻、解冻后不脱除EG直接培养或移植。  相似文献

A system consisting of a flow-through chamber connected to a commercial infrared gas analysis system was developed to measure homopteran respiration during feeding. Using this system, respiration rates of 202 and 206 μmol CO(2) h(-1) g(-1) (4.96 and 5.04 ml CO(2) h(-1) g(-1)) were determined for whiteflies and cotton aphids, respectively, at 25 degrees C on diets containing 15% sucrose. These rates were considerably higher than those of other stationary insects, indicating that whiteflies and aphids maintain a relatively high metabolic rate when feeding. Whitefly respiration increased with temperature from 25 to 46 degrees C with a Q(10) of about 2 on diets containing 10, 15 and 20% sucrose, but less than 2 on diets containing 2.5 and 5% sucrose. Respiration rates were similar on the diets containing >10% sucrose, but were generally lower on the diets containing <10% sucrose. Respiration rates decreased upon extended exposure to 47 degrees C; the rate of decrease was inversely related to the dietary sucrose concentration up to 15%. The results indicate that whiteflies require a sucrose concentration of between 5 and 10% (i.e. 0.15 and 0.3 M) for maximum rates of metabolism while feeding. Higher concentrations of sucrose in the diet delayed high-temperature mortality, possibly a reflection of the high sucrose requirement for sorbitol synthesis in whiteflies.  相似文献
