A Gram-negative, non-mobile, polar single flagellum, rod-shaped bacterium WZBFD3-5A2T was isolated from a wheat soil subjected to herbicides for several years. Cells of strain WZBFD3-5A2T grow optimally on Luria-Bertani agar medium at 30?°C in the presence of 0–4.0?% (w/v) NaCl and pH 8.0. 16S rRNA gene sequence analysis revealed that strain WZBFD3-5A2T belongs to the genus Pseudomonas. Physiological and biochemical tests supported the phylogenetic affiliation. Strain WZBFD3-5A2T is closely related to Pseudomonas nitroreducens IAM1439T, sharing 99.7?% sequence similarity. DNA–DNA hybridization experiments between the two strains showed only moderate reassociation similarity (33.92?±?1.0?%). The DNA G+C content is 62.0?mol%. The predominant respiratory quinine is Q-9. The major cellular fatty acids present are C16:0 (28.55?%), C16:1ω6c or C16:1ω7c (20.94?%), C18:1ω7c (17.21?%) and C18:0 (13.73?%). The isolate is distinguishable from other related members of the genus Pseudomonas on the basis of phenotypic and biochemical characteristics. From the genotypic, chemotaxonomic and phenotypic data, it is evident that strain WZBFD3-5A2T represents a novel species of the genus Pseudomonas, for which the name Pseudomonas nitritereducens sp. nov. is proposed. The type strain is WZBFD3-5A2T (=CGMCC 1.10702T?=?LMG 25966T).  相似文献

A Gram-stain negative, motile, rod-shaped bacterium, designated strain WM-2T, was isolated from a forest soil in Sihui City, South China, and characterized by means of a polyphasic approach. Growth occurred with 0–5 % (w/v) NaCl (optimum 0–1 %) and at pH 5.0–10.5 (optimum pH 8.5) and 4–40 °C (optimum 30 °C) in Luria–Bertani medium. Comparative 16S rRNA gene sequence analyses showed that strain WM-2T is a member of the genus Pseudomonas and most closely related to P. guguanensis, P. oleovorans subsp. lubricantis, P. toyotomiensis, P. alcaliphila and P. mendocina with 97.1–96.6 % sequence similarities. In terms of gyrB and rpoB gene sequences, strain WM-2T showed the highest similarity with the type strains of the species P. toyotomiensis and P. alcaliphila. The DNA–DNA relatedness values of strain WM-2T with P. guguanensis and P. oleovorans subsp. lubricantis was 48.7 and 37.2 %, respectively. Chemotaxonomic characteristics (the main ubiquinone Q-9, major fatty acids C18:1 ω7c/C18:1 ω6c, C16:0 and C16:1 ω7c/C16:1 ω6c and DNA G+C content 65.2 ± 0.7 mol%) were similar to those of members of the genus Pseudomonas. Polar lipids consisted of diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, an unknown aminophospholipid, an unknown phospholipid and five unknown lipids. According to the results of polyphasic analyses, strain WM-2T represents a novel species in the genus Pseudomonas, for which the name Pseudomonas sihuiensis sp. nov. is proposed. The type strain is WM-2T (=KCTC 32246T=CGMCC 1.12407T).  相似文献

A Gram-staining-negative, rod-shaped and motile with several polar flagellums bacterium, designated WM-3T, was isolated from a rice paddy soil in South China. Growth occurred with 0–3.0 % (w/v) NaCl (optimum 2.0 %), at pH 5.5–9.0 (optimum pH 7.0) and at 25–42 °C (optimum 30–37 °C) in liquid Reasoner’s 2A medium. Analysis of the 16S rRNA gene and gyrB gene sequences revealed that strain WM-3T was most closely related to the type strains of the species Pseudomonas linyingensis and Pseudomonas sagittaria. Its sequence similarities with P. linyingensis CGMCC 1.10701T and P. sagittaria JCM 18195T were 97.4 and 97.3 %, respectively, for 16S rRNA gene, and were 94.1 and 94.2 %, respectively, for gyrB gene. DNA–DNA hybridization between strain WM-3T and these two type strains showed relatedness of 35.6 and 30.9 %, respectively. G+C content of genomic DNA was 69.4 mol%. The whole-cell fatty acids mainly consisted of C16:0 (30.0 %), C16:1 ω6c and/or C16:1 ω7c (19.3 %) and C18:1 ω6c and/or C18:1 ω7c (16.3 %). The results of phenotypic, chemotaxonomic and genotypic analyses clearly indicated that strain WM-3T belongs to genus Pseudomonas but represents a novel species, for which the name Pseudomonas oryzae sp. nov. is proposed. The type strain is WM-3T (=KCTC 32247T =CGMCC 1.12417T).  相似文献

A Gram-negative, rod-shaped, slightly halophilic and facultatively anaerobic bacterium, designated strain D15-8WT, was isolated from the sediment of the South China Sea. Growth was found to occur optimally at 25 °C, between pH 7.0 and 8.0 and with 1–5 % (w/v) NaCl. The strain was observed to utilize a variety of organic substrates and polycyclic aromatic hydrocarbons as sole carbon sources. The G+C content of the genomic DNA was determined to be 58.7 %. The predominant respiratory quinone was found to be Q-9. The significant fatty acids were determined to be C16:0, C16:1 ω9c, C18:1 ω9c, C12:0 and C14:0 3OH. Analysis of 16S rRNA gene sequences showed that strain D15-8WT fits within the phylogenetic cluster of the genus Marinobacter and is most closely related to Marinobacter segnicrescens CGMCC 1.6489T, Marinobacter bryozoorum DSM 15401T, Marinobacter lacisalsi CECT 7297T and Marinobacter daqiaonensis CGMCC1.9167T. The DNA–DNA hybridization values between strain D15-8WT and the type strains of the most closely related species were 42.3 % (CGMCC 1.6489T), 39.8 % (DSM 15401T), 37.3 % (CECT 7297T) and 35.2 % (CGMCC1.9167T). The results of this polyphasic study indicate that strain D15-8WT represents a novel species of the genus Marinobacter, for which the name Marinobacter nanhaiticus sp. nov. is proposed. The type strain is D15-8WT (=CGMCC 1.11019T=KCTC 23749T).  相似文献

A Gram-stain positive, aerobic, non-motile, endospore-forming and rod-shaped strain (THG-NT9T) was isolated from a green tea sample. Growth occurred at 20–45 °C (optimum 28–35 °C), at pH 6.0–8.0 (optimum 7.0) and at 0–2.0% NaCl (optimum 0%). Based on 16S rRNA gene sequence analysis, the near phylogenetic neighbours of strain THG-NT9T were identified as Scopulibacillus daqui DSM 28236T (98.6%), Scopulibacillus darangshiensis DSM 19377T (97.4%), Pullulanibacillus pueri CGMCC 1.12777T (96.7%) and Pullulanibacillus camelliae CGMCC 1.15371T (96.3%). The DNA G?+?C content of strain THG-NT9T was determined to be 47.5 mol %. DNA–DNA hybridization values between strain THG-NT9T and S. daqui DSM 28236T, S. darangshiensis DSM 19377T, P. pueri CGMCC 1.12777T, P. camelliae CGMCC 1.15371T and Pullulanibacillus naganoensis DSM 10191T were 41.3?±?0.1 (39.4?±?0.4% reciprocal analysis), 39.1?±?0.1 (37.3?±?0.1%), 21.4?±?0.7 (20.1?±?0.3%), 20.7?±?0.1 (20.1?±?0.4%) and 12.1?±?0.2% (8.3?±?0.2%). The polar lipids were identified as diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine and three unidentified lipids. The quinone was identified as MK-7. The major fatty acids were C18:3 ω7c, iso-C15:0, iso-C16:0, iso-C17:0 and anteiso-C17:0. The cell wall type was determined to be A1γ peptidoglycan with meso-diaminopimelic acid as the diagnostic diamino acid plus alanine and glutamic acid and glucose as the cell wall sugar. On the basis of the phylogenetic analysis, chemotaxonomic data, physiological characteristics, and DNA–DNA hybridization data, strain THG-NT9T represents a novel species of the genus Scopulibacillus, for which the name Scopulibacillus cellulosilyticus sp. nov. is proposed. The type strain is THG-NT9T (=?KCTC 33918T?=?CGMCC 1.16305T).  相似文献

Strains VGXO14T and Vi1 were isolated from the Atlantic intertidal shore from Galicia, Spain, after the Prestige oil spill. Both strains were Gram-negative rod-shaped bacteria with one polar inserted flagellum, strictly aerobic, and able to grow at 18–37 °C, pH 6–10 and 2–10% NaCl. A preliminary analysis of the 16S rRNA and the partial rpoD gene sequences indicated that these strains belonged to the Pseudomonas genus but were distinct from any known Pseudomonas species. A polyphasic taxonomic approach including phylogenetic, chemotaxonomic, phenotypic and genotypic data confirmed that the strains belonged to the Pseudomonas pertucinogena group. In a multilocus sequence analysis, the similarity of VGXO14T and Vi1 to the closest type strain of the group, Pseudomonas pachastrellae, was 90.4%, which was lower than the threshold of 97% established to discriminate species in the Pseudomonas genus. The DNA–DNA hybridisation similarity between strains VGXO14T and Vi1 was 79.6%, but below 70% with the type strains in the P. pertucinogena group. Therefore, the strains should be classified within the genus Pseudomonas as a novel species, for which the name Pseudomonas aestusnigri is proposed. The type strain is VGXO14T (=CCUG 64165T = CECT 8317T).  相似文献

A novel actinomycete, designated strain NEAU-GH7T, was isolated from a lake sediment and characterized using a polyphasic approach. Strain NEAU-GH7T was Gram-stain positive, aerobic, non-spore-forming and produced spherical sporangia. Phylogenetic analysis based on the 16S rRNA gene sequence indicated that strain NEAU-GH7T formed a monophyletic clade with the closest relative Streptosporangium longisporum DSM 43180T (99.0 %), an association that was supported by a bootstrap value of 74 % in the neighbour-joining tree and also recovered with the maximum-likelihood algorithm. However, the low level of DNA–DNA relatedness allowed the strain to be differentiated from its closest relative. Moreover, strain NEAU-GH7T could also be differentiated from S. longisporum DSM 43180T and other Streptosporangium species showing high 16S rRNA gene sequence similarity (>98.0 %) by morphological and physiological characteristics. On the basis of phylogenetic analysis, DNA–DNA hybridization and phenotypic characteristics, strain NEAU-GH7T should be classified as a new species of the genus Streptosporangium, for which the name Streptosporangium shengliensis sp. nov. is proposed. The type strain is NEAU-GH7T (=CGMCC 4.7105T=DSM 45881T).  相似文献

A novel aerobic, bacteriochlorophyll-containing bacteria strain JLT1202rT was isolated from Beibu Gulf in the South China Sea. Cells were gram-negative, non-motile, and short-ovoid to rod-shaped with two narrower poles. Strain JLT1202rT formed circular, opaque, wine-red colonies, and grew optimally at 3–4?% NaCl, pH 7.5–8.0 and 28–30?°C. The strain was catalase, oxidase, ONPG, gelatin, and Voges–Proskauer test positive. In vivo absorption spectrum of bacteriochlorophyll a presented two peaks at 800 and 877?nm. The predominant cellular fatty acid was C18:1 ω7c and significant amounts of C16:0, C18:0, C10:0 3-OH, C16:0 2-OH, and 11-methyl C18:1 ω7c were present. Strain JLT1202rT contained Q-10 as the major respiratory quinone and the genomic DNA G+C content was 76.3?mol%. Phylogenetic analysis based on 16S rRNA gene sequences of various species with validly published names showed that strain JLT1202rT fell within the genus Roseibacterium, family Rhodobacteraceae, sharing the highest similarity with Roseibacterium elongatum OCh 323T (97.9?% similarity), followed by Dinoroseobacter shibae DFL 12T (95.4?% similarity). The phylogenetic distance of pufM genes between strain JLT1202rT and R. elongatum OCh 323T was 9.4?%, suggesting that strain JLT1202rT was distinct from the only strain of the genus Roseibacterium. Based on the variabilities of phylogenetic and phenotypic characteristics, strain JLT1202rT stands for a novel species of the genus Roseibacterium and the name R. beibuensis sp. nov. is proposed with JLT1202rT as the type strain (=JCM 18015T?=?CGMCC 1.10994T).  相似文献

In this study we analysed three bacterial strains coded L10.10T, A4R1.5 and A4R1.12, isolated in the course of a study of quorum-quenching bacteria occurring in Antarctic soil. The 16S rRNA gene sequence was identical in the three strains and showed 99.7% pairwise similarity with respect to the closest related species Pseudomonas weihenstephanensis WS4993T. Therefore, the three strains were classified within the genus Pseudomonas. Analysis of housekeeping genes (rpoB, rpoD and gyrB) sequences showed similarities of 84-95% with respect to the closest related species of Pseudomonas, confirming its phylogenetic affiliation. The ANI values were less than 86% to the closest related species type strains. The respiratory quinone is Q9. The major fatty acids are C16:0, C16:1 ω7c/ C16:1 ω6c in summed feature 3 and C18:1 ω7c / C18:1 ω6c in summed feature 8. The strains are oxidase- and catalase-positive. Growth occurs at 4–30 °C, and at pH 4.0–10. The DNA G+C content is 58.2–58.3 mol %. The combined genotypic, phenotypic and chemotaxonomic data support the classification of strains L10.10T, A4R1.5 and A4R1.12 into a novel species of Pseudomonas, for which the name P. versuta sp. nov. is proposed. The type strain is L10.10T (LMG 29628T, DSM 101070T).  相似文献

The halophilic archaeal strain GX71T was isolated from the Gangxi marine solar saltern near the Weihai city of Shandong Province, China. Cells of the strain were pleomorphic and lysed in distilled water, stained Gram-negative and formed red-pigmented colonies. Strain GX71T was able to grow at 25–45 °C (optimum 30 °C), in the presence of 1.7–4.8 M NaCl (optimum 2.6 M NaCl), with 0.005–0.7 M MgCl2 (optimum 0.05 M MgCl2) and at pH 5.5–9.5 (optimum pH 7.0–7.5). Cells lysed in distilled water and the minimal NaCl concentration to prevent cell lysis was 10 % (w/v). The major polar lipids of the strain were phosphatidylglycerol, phosphatidylglycerol phosphate methyl ester, phosphatidylglycerol sulfate, one major glycolipid chromatographically identical to sulfated mannosyl glucosyl diether (S-DGD-3) and an unidentified lipid was also detected. The 16S rRNA gene sequence of strain GX71T showed 94.0–97.0 % similarity to members of the genus Halorubrum of the family Halobacteriaceae. The rpoB′ gene sequence of strain GX71T was 87.3–93.4 % similarity to current members of the genus Halorubrum. The DNA G+C content of GX71T was 67.1 mol%. Strain GX71T showed low DNA–DNA relatedness with Halorubrum lipolyticum CGMCC 1.5332T, Halorubrum saccharovorum CGMCC 1.2147T, Halorubrum kocurii CGMCC 1.7018T and Halorubrum arcis CGMCC 1.5343T, the most closely related members of the genus Halorubrum. The phenotypic, chemotaxonomic and phylogenetic properties suggest that strain GX71T represents a novel species of the genus Halorubrum, for which the name Halorubrum salinum sp. nov. is proposed. The type strain is GX71T (= CGMCC 1.10458T = JCM 17093T).  相似文献

Two gram-positive, aerobic, spore-forming, rod-shaped bacteria, designated HB09003T and HB12160, were isolated from seawater and sediment in the northern South China Sea, respectively. Cells were found to be motile by means of peritrichous flagella. The strains were found to grow with 0–15 % (w/v) NaCl, at 10–45 °C and pH 5.0–10.7, with an optimum of 3 % NaCl, 28 °C and pH 8.5, respectively. The predominant isoprenoid quinone of strain HB09003T, selected as the representative strain, was identified as MK-7. This strain was found to possess anteiso-C15:0, iso-C15:0, anteiso-C17:0 and C16:0 as the major fatty acids. The G+C contents of strain HB09003T and HB12160 were determined to be 34.1 and 34.3 mol%, respectively. Analysis of the 16S rRNA gene sequences of the two strains showed an affiliation with the genus Gracilibacillus, with Gracilibacillus kekensis CGMCC 1.10681T (similarity of 97.4, 98.0 %, respectively) and Gracilibacillus ureilyticus CGMCC 1.7727T (similarity of 97.1, 97.8 %, respectively) as their closest relatives. The DNA–DNA hybridization values between strain HB09003T and the two type strains were 42.2 and 54.1 %, respectively. On the basis of phenotypic and genotypic data, strain HB09003T and HB12160 are proposed to represent a novel species of the genus Gracilibacillus, for which the name Gracilibacillus marinus sp. nov. is proposed. The type strain is HB09003T (=CGMCC 1.10343T = DSM 23372T).  相似文献

A Gram-negative, polar flagella, rod-shaped bacterium LV T was isolated from a soil sample subjected to long-term manganese pollution in Hunan Province, China. Cells grow optimally on Luria–Bertani agar medium at 30 °C in the presence of 0–5.0 % (w/v) NaCl and pH 78. 16S rRNA gene sequence analysis revealed that strain LV T belonged to the genus Pseudomonas, with sequence similarity values of 98.6, 98.2, 98.7, and 97.3 % to Pseudomonas monteilii BCRC 17520 T , Pseudomonas putida BCRC 10459 T , Pseudomonas plecoglossicida BCRC 17517 T , and Pseudomonas asplenii BCRC 17131 T , respectively. The level of DNA–DNA relatedness between the five strains was <30 %. The DNA G+C content of strain LV T is 68.8 mol%. Chemotaxonomic data revealed that the strain LVT possesses ubiquinone Q-9. The polar lipid profile of strain LV T contained diphosphatidylglycerol, phosphatidylglycerol, phosphatidylcholine, and phosphatidylethanolamine. The major cellular fatty acids present are C10:03-OH (12.33 %), C16:0 (23.99 %), summed feature 3(C16:1ω7c and/or C16:1ω6c), and summed feature 8(C18:1 ω7c and C18:1 ω6c). Based on the genotypic, chemotaxonomic and phenotypic data, strain LV T is distinguishable from related members of the genus Pseudomonas. Thus, strain LV T represents a novel species of the genus Pseudomonas, for which the name Pseudomonas hunanensis sp. nov. is proposed. The type strain is LV T (=CICC 10558T = NCCB 100446T).  相似文献

A Streptomyces strain isolated from a hyper-arid Atacama Desert soil was characterised using a polyphasic taxonomic approach. The strain, designated C2T, had chemical and morphological properties typical of the genus Streptomyces. The isolate formed a branch in the Streptomyces 16S rRNA gene tree together with the type strain of Streptomyces chromofuscus and was also loosely related to Streptomyces fragilis NRRL 2424T. DNA:DNA relatedness values between the isolate and its two phylogenetic neighbours showed that it formed a distinct genomic species. The strain was readily distinguished from these organisms using a combination of morphological and phenotypic data. Based on the genotypic and phenotypic results, isolate C2T represents a novel species in the genus Streptomyces, for which the name Streptomyces bullii sp. nov. is proposed. The type strain is C2T (=CGMCC 4.7019T = KACC 15426T).  相似文献

A novel actinomycete, designated strain NEAU-st4T, was isolated from a soil sample collected from Shaanxi province, Northwest China and characterized using a polyphasic approach. 16S rRNA gene sequence analysis revealed that strain NEAU-st4T has the highest sequence similarities with Actinomycetospora rishiriensis RI109-Li102T (99.4 %), Actinomycetospora corticicola 014-5T (99.1 %), Actinomycetospora chiangmaiensis YIM 0006T (98.8 %) and Actinomycetospora iriomotensis IR73-Li102T (98.2 %). However, the low level of DNA–DNA relatedness differentiated strain NEAU-st4T from its closest phylogenetic neighbours. The main chemotaxonomic properties of strain NEAU-st4T, such as the diamino acid of the peptidoglycan, the whole-cell hydrolysates, the predominant menaquinones and the phospholipid profile, supported its classification within the genus Actinomycetospora. The distinctive morphology of this strain compared with that of other members in the genus Actinomycetospora is the formation of sporangia directly on the substrate hyphae. Phenotypic and genotypic differences also allowed the distinction of the strain from closely related species. Consequently, strain NEAU-st4T represents a new species of the genus Actinomycetospora, for which the name Actinomycetospora atypica sp. nov. is proposed. The type strain is NEAU-st4T (=CGMCC 4.7093T = DSM 45873T).  相似文献

A collection of Pseudomonas strains was isolated in different regions of Tunisia in the period 2016–2017 from the fruits and leaves of Citrus sinensis cv. ‘Valencia Late’ and Citrus limon cv. ‘Eureka’ plants with symptoms of blast and black pit disease. A phylogenetic analysis of the housekeeping gene rpoD was used for strain identification at the species level. The results demonstrated the affiliation of these strains with the genus Pseudomonas and revealed the presence of 11 strains representing two putative new species in two monophyletic branches. These strains were analyzed morphologically and genotypically by multilocus sequence analyses of the rpoD, gyrB and 16S rRNA (rrs) gene sequences, and their phenotypic characteristics by API 20NE and Biolog GEN III. Plant pathogenic properties were confirmed on fruits and detached leaves of C. limon cv. ‘Eureka’. Fatty acids and WC MALDI-TOF MS major protein profiles were determined. The genomes of both representatives were sequenced. The average nucleotide index and genome-to-genome distance from KC12T and E10BT are below the cut-off established for a described species. These results support the conclusion that the strains KC12T, KC17, KC20, KC22, KC24A, KC25 and KC26 represent a novel species of Pseudomonas, for which the name of Pseudomonas kairouanensis is proposed. The type strain is KC12T (=CECT9766 and CFBP 8662). The strains E10BT, E10AB, E10CB1 and Iy3BA represent another novel species of Pseudomonas for which the name of Pseudomonas nabeulensis is proposed; the type strain is E10BT (=CECT9765 and CFBP 8661).  相似文献

A Gram-positive, catalase and oxidase positive, rod-shaped bacteria, and spore-forming, designated as J20-3T was isolated from a peat soil, collected near a coal mine at Prokopyevsk, (GPS; N53°52′51″, E86°43′39″) Kemerovo Oblast, Russia. A polyphasic taxonomy study using phenotypic, phylogenetic, and genotypic method was performed to characterize strain J20-3T. Comparative 16S rRNA gene sequence analysis indicated that strain J20-3T represented a novel subline within the genus Cohnella in the family Paenibacillaceae. According to 16S rRNA gene sequence, strain J20-3T showed 93.7–97.2 % similarity levels with other Cohnella species. Strain J20-3T exhibited relatively low level of DNA–DNA hybridization value with type strains KACC 11643T (40 %), KACC 11771T (37.5 %), and KACC 15372T (30.5 %). The strain showed typical chemotaxonomic characteristic of the genus Cohnella, with the presence of predominant respiratory quinone MK-7; major fatty acids are C15:0, C16:0, iso, and C16:0. The DNA G+C content of the strain J20-3T was 56.3 mol%. The polar lipid profile of the strain J20-3T included major amount of diphosphatidylglycerol, phosphatidylglycerol, and phosphoatidylethanolamine. On the basis of its phenotypic and genotypic properties, and its phylogenetic distinctiveness, strain J20-3T should be classified as a novel species in the genus Cohnella, for which the name Cohnella humi sp. nov. is proposed.  相似文献

Strains V113T, V92 and V120 have been isolated from sand samples taken at the Atlantic intertidal shore in Galicia, Spain, after the Prestige oil spill. A preliminary analysis of the 16S rRNA and the partial rpoD gene sequences indicated that these strains belonged to the Pseudomonas genus, but they were distinct from any known Pseudomonas species. They were extensively characterized by a polyphasic taxonomic approach and phylogenetic data that confirmed that these strains belonged to the Pseudomonas pertucinogena group. Phylogenetic analysis of 16S rRNA, gyrB and rpoD gene sequences showed that the three strains were 99% similar and were closely related to members of the P. pertucinogena group, with less than 94% similarity to strains of established species; Pseudomonas pachastrellae was the closest relative. The Average Nucleotide Index based on blast values was 89.0% between V113T and the P. pachastrellae type strain, below the accepted species level (95%). The predominant cellular fatty acid contents and whole cell protein profiles determined by MALDI-TOF mass spectrometry also differentiated the studied strains from known Pseudomonas species. We therefore conclude that strains V113T, V92 and V120 represent a novel species of Pseudomonas, for which the name Pseudomonas gallaeciensis is proposed; the type strain is V113T (= CCUG 67583T = LMG 29038T).  相似文献

A Nocardia-like actinobacterial strain, designated YIM TG2190T, was isolated from rhizosphere soil of Psammosilene tunicoides collected from Gejiu, Yunnan province, China. The cells of strain YIM TG2190T were observed to be Gram-stain positive and non-motile. The strain forms extensively branched substrate mycelia that fragments into rod-shaped elements. The 16S rRNA gene sequence analysis showed that strain YIM TG2190T is closely related to Nocardia nova (97.5%), Nocardia jiangxiensis (97.1%) and Nocardia miyunensis (96.8%). Growth occurs at 4–30?°C (optimum 28?°C), pH 6.0–8.0 (optimum pH 7.0) and the strain can tolerate NaCl (w/v) up to 3% (optimum 0–1%). The cell walls were found to contain meso-diaminopimelic acid. The whole-cell sugars were identified as glucose, mannose, ribose, galactose, arabinose and fucose. The polar lipids were identified as diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylinositol mannosides, phosphatidylglycerol and an unidentified phospholipid. The menaquinones detected were MK-9 (H2) and MK-8 (H4). The major fatty acids (>?5%) were found to be C16:0 (33.9%), summed feature 3 (21.7%), C18:0 10-methyl TBSA (13.7%) and C18:1ω9c (7.0%). The DNA G+C content was determined to be 61.1 mol%. DNA-DNA relatedness between the strain YIM TG2190T and N. nova CGMCC 4.1705T, N. jiangxiensis CGMCC 4.1905T and N. miyunensis CGMCC 4.1904T were 46.9?±?2.6, 36.8?±?1.3, and 35.7?±?2.6%, respectively, values which are less than the threshold value (70%) for the delineation of prokaryotic genomic species. The phenotypic, chemotaxonomic and phylogenetic data indicates that strain YIM TG2190T represents a novel species of the genus Nocardia, for which the name Nocardia zhihengii sp. nov. is proposed. The type strain is YIM TG2190T (=KCTC 39596T?=?DSM 100515T).  相似文献

An alkaliphilic, moderately halophilic, bacterium, designated strain X10-1T, was isolated from saline-alkaline soil inDaqing, Heilongjiang Province, China. Strain X10-1T was determined to be a Gram-positive aerobe with rod-shaped cells. The isolate was catalase-positive, oxidase-negative, non-motile, and capable of growth at salinities of 0–16% (w/v) NaCl (optimum, 3%). The pHrange for growth was 7.5–11.0 (optimum, pH 10.0). The genomic DNA G+C content was 47.7 mol%. Itsmajor isoprenoid quinone was MK-7 and its cellular fatty acid profile mainly consisted of anteiso-C15:0, anteiso-C17:0, iso-C15:0, C16:0, and iso-C16:0. The peptidoglycan contained meso-diaminopimelic acid as the diagnostic diamino acid. The predominant polar lipids were diphosphatidylglycerol, phosphatidylethanolamine, and phosphatidylglycerol. Phylogenetic analysis based on 16S rRNA gene sequences showed that X10-1T is a member of the genus Bacillus, being most closely related to B. saliphilus DSM15402T (97.8% similarity) and B. agaradhaerens DSM 8721T (96.2%). DNA-DNA relatedness to the type strains of these species was less than 40%. On the basis of the phylogenetic, physiological, and biochemical data, strain X10-1T represents a novel species of the genus Bacillus, for which the name Bacillus daqingensis sp. nov. is proposed. The type strain is X10-1T (=NBRC 109404T = CGMCC 1.12295T).  相似文献
