A population of the intertidal, ovoviviparous, cirolanid isopod Annina mesopotamica was studied at the Al-Chibassy tributary of the Shatt Al-Arab river from 14 May 1973 to 29 April 1974, and at the Garmat-Ali river of the Shatt Al-Arab, Basrah, Iraq from 5 November 1984 to 30 June 1986. Population density varied between 128 to 5760 individuals m–2 with a mean of 1218 m–2. Highest numbers occurred between June and September. Breeding extended for most of the year except November/January to February/March, although ovigerous females were present throughout the year. The incubation period varied with season, and was ca. 2 weeks during the warmest period, and about a month at lower temperatures. Egg numbers varied between 4 to 66 with mean 24. Brood size was linearly proportional to female size. The size frequency distribution of the population was polymodal with a predominance of females over males. Four different types of growth curves were found: exponential, concave, convex, and intermediate between concave and convex. Growth rate varied between 0.63 and 2.19 mm month–1. Mortality was higher among summer cohorts. Life span varied between 5 and 11 months.  相似文献

This study evaluated the use of egg ratio (eggs rotifer–1) and swimming speed (mm min–1) as prediction criteria for production and culture quality in mass cultures of the rotifer Brachionus plicatilis. Egg ratio was determined to be a suitable predictor of rotifer growth and production in the cultures. Low egg ratios (i.e., 0–0.17 eggs rotifer–1) indicate reduced rotifer population over time (i.e., negative net population growth rates). However, at this time egg ratio dynamics are not suitably understood to predict in advance a sudden population collapse.Swimming speed of reproductive, egg-carrying females in the exponential growth phase was 40–45 mm min–1. During exponential growth swimming speed was independent of the food used. Lower swimming speeds were obtained in late stationary phase (10–25 mm min–1) when yeast was used as a food source. Both environmental factors (e.g., accumulating metabolites) and changes in nutritional state of the rotifers may have affected the swimming speed, but environmental factors appear to be the most important. We believe that swimming speed has the potential of becoming an accurate predictor of culture quality in mass cultures of rotifers.  相似文献

Pinus taiwanensis is a widely distributed species in the southeastern China (Zhejiang, Anhui, Hubei, Human, Jiangxi, Fujian, Taiwan provinces) at an elevation of 700–2000 m. This pine is a pioneer in forest succession and is often used as a species for afforestation in this region at an elevation above 700 m. This study was carried out at Guanshanyuan, Zhejiang province, at a latitude of N 28° 18, a longitude of E 119° 16 and an altitude of 800–1502 m above sea level. On the basis of a census of all individuals of Pinus taiwanensis at different successional stages and various habitats, age structure, spatial pattern, density, biomass of population and their dynamics were described. Considering the population dynamics throughout the successional process, three phases could be recognized. Until about 9–10 years after Pinus taiwanensis invaded the stands, the density of population was increased by the recruitments along with increase of the mean tree weight and population biomass (phase I). Thereafter, the population was in full density state, the biomass of population and the mean tree weight increased exponentially, while the density was decreased drastically by the self-thinning and the invasion of other broad-leaved trees (phase II). The –3/2 power law of natural thinning was applicable to the populations in this phase. When the broad-leaved trees reached the canopy, although the mean tree weight increased slowly, the density and biomass of Pinus taiwanensis population decreased gradually (phase III) until the population senesced and retreated from the successional series completely. The population dynamics of Pinus taiwanensis during the successional process was in common with pioneer species in forest succession. At some special habitats such as rocky steep slopes and ridges, however, Pinus taiwanensis population could form such an edaphic climax community that the population density, biomass and the mean tree weight in phase III could be in a stable state for very long period.  相似文献

A local marine diatom, Nitzschia conspicua Grunow, was cultured in enriched synthetic seawater using flasks (agitated by magnetic stirring) and a 1.2 l fermenter. Lipids, fatty acids, proteins, carbohydrates and ash of the flask cultures were determined at various stages of growth (day 3, 5, 7, 10, 13, 15 and 17). The fermenter culture was harvested during the stationary phase for similar chemical analyses. N. conspicua attained a higher biomass concentration during the stationary phase when cultured in the fermenter (188 mg dry weight l–1) than in flasks (140–151 mg dry weight l–1). However, both systems showed similar specific growth rates based on chlorophyll-a concentration. Appreciable amounts of the essential fatty acids 20:4 (0.6–4.7% total fatty acids) and 20:5 (1.9–4.7% total fatty acids) are present in this diatom. Maximal amounts of these fatty acids were produced after 7 days' growth (i.e. 2 days after the end of the exponential phase). Mortierella alpina LPM 301, a producer of arachidonic acid (ARA), was found to possess a unique property of intense lipid synthesis in the period of active mycelium growth. Under batch cultivation of this strain in glucose-containing media with potassium nitrate or urea, the bulk of lipids (28–35% of dry biomass) was produced at the end of the exponential growth phase and remained almost unaltered in the stationary phase. The ARA content of lipids comprised 42–50% at the beginning of the stationary phase and increased continuously after glucose depletion in the medium due to the turnover of intracellular fatty acids; by the end of fermentation (189–210 h), the amount of ARA reached 46–60% of the total fatty acids (16–19% of dry mycelium). Plausible regulatory mechanisms of the growth-coupled lipid synthesis in microorganisms are discussed.  相似文献

Macrocystis pyrifera (L.) Ag.frond elongation rates were measured during autumn-winter, spring and summer in a shallow water (7.5 m depth) kelp bed in Bahía Papalote, northern Baja California. Frond elongation was maximum during spring and minimum during winter. Frond growth was significantly correlated with solar radiation, and was highest in the smallest size fronds (0–2 m). Average frond elongation rate varied between 0.3–11% d–1 during the study period. The relationship between average frond length and elongation rate followed an exponential curve with a negative slope during autumn-winter and summer, but was best described by a straight line during spring. Standard growth rates were obtained by a graphic method. Standard growth rates had intermediate values between those reported for southern California and southern Baja California.M. pyrifera growth cycle shows a different trend from what has been previously reported.  相似文献

The ability ofPseudomonas fluorescens, Escherichia coli andAcinetobacter radioresistenns to remove phosphate during growth was related to the initial biomass as well as to growth stages and bacterial species. Phosphate was removed by these bacteria under favourable conditions as well as under unfavourable conditions of growth. Experiments showed a relationship between a high initial cell density and phosphate uptake. More phosphate was released than removed when low initial cell densities (102–105 cells ml–1) were used. At a high initial biomass concentration (108 cells ml–1), phosphate was removed during the lag phase and during logarthmic growth byP. fluorescens. Escherichia coli. at high initial biomass concentrations (107 cells ml–1), accumulated most of the phosphate during the first hour of the lag phase and/or during logarithmic growth and in some cases removed a small quantily of phosphate during the stationary growth phase.Acinetobacter radioresistens, at high initial cell densities (106, 107 cells ml–1) removed most of phosphate during the first hour of the lag phase and some phosphate during the stationary growth phase.Pseudomonas fluorescens removed phosphate more thanA. radioresistens andE. coli with specific average ranges from 3.00–28.50 mg L–1 compared to average ranges of 4.92–17.14 mg L–1 forA. radioresistens and to average ranges of 0.50–8.50 mg L–1 forE. coli.  相似文献

The distribution and stage composition of Calanoidescarinatus (Kroyer) are described together with changes in protein and lipid content, respiration rate and gonad development in copepodite stages IV and V (CV) and adults in the Northern Benguela current (17°S, 23°S and 25°S).During active upwelling the population consisted of two parts: the surface part over the shelf was represented by all development stages, while the deep part offshore was dominated (90–95%) by diapausal CVs. In the surface CVs the surplus assimilated energy was allocated to structural growth and maturation or to synthesis of reserve lipids. CVs with large oil sacs and high lipid content descended into deeper layers and formed diapausal stock; they were characterized by a dramatic decrease of respiration rate. Increase of gonad size in association with decrease of oil sac volume in diapausal CV suggests that reserve lipids were expended not only for respiration but also for gonad development. The moulting of dispausal CVs into adults took place in deep water.These results are discussed in relation to the life cycle of the C. carinatus population and the factors causing the formation and termination of the diapausal phase.  相似文献

The studies of the cell cycle ofDiatoma hiemale var.mesodon have been continued on a population during the whole vegetative period (August–October 1976). The lapse of time between the maxima of the chromatophore division and the mitosis is 8 hours in the exponentional and stationary phase of growth of the population; during the decline and decay of the population it is shortening to 2–4 hours. The sequence of phases is maintained, but the stage of praemitotic stagnation is shortened to a minimum. During the stage of the chromatophore division a relationship was found between time lapse and chromatophore number per cell. The cell cycle (asexual life cycle) ofDiatoma hiemale var.mesodon has been schematically illustrated during optimal development and its different stages are discussed.

WhenCryptococcus neoformans was grown in yeast nitrogen base (YNB) supplemented with 0.5% glucose, the medium was acidified to below pH 3 during the exponential growth phase, which caused early growth-phase death in susceptible strains. Even in resistant strains, 30–70% cells died if incubated for 2 d in YNB supplemented with 1.5% glucose, whereas the remaining cells survived long. Two types of fatal alterations have been observed in dead cells. In the first type, release of cytoplasm occurred through weakened parts of the cell wall; structures attached to cell walls of dead cells were shown to be rich in proteins by FITC staining, indicating their cytoplasmic origin. In the second type, cells shrank distinctly with no sign of wall rupture. The shrinkage may be due to dysfunction of the plasma membrane at low pH. The mechanism of cell survival in medium below pH 3 was also examined. Aniline blue alone, or calcofluor together with methylene blue, allowed cell wall glucan or chitin and dead cell cytoplasm to be stained simultaneously. In the later stages of incubation, cells showing bright staining for cell wall glucan and chitin emerged. These changes in cell wall synthesis could be considered as an adaptation mechanism to acidification of the medium, because such cells survived longer than cells showing no change in the cell wall staining pattern.  相似文献

The diatomsChaetoceros sp.,Skeletonema costatum andThalassiosira pseudonana were grown with different irradiances of white and of blue-green light, and with a mixture of blue-green plus 6.5 mol m–2 s–1 of white light. Exponential growth rates were higher in mixed blue for the first two, whileT. pseudonana grew faster in white light but, in all cases, mean cell division rates did not differ with increasing irradiances. Harvesting in stationary, rather than in late exponential growth phase, resulted in higher protein contents forChaetoceros sp. andS. costatum, but forT. pseudonana the highest value was in the exponential phase. The highest protein content was in blue-green light for the three species and it increased with irradiance. As to other fractions, the three strains showed different responses, related to quality and quantity, as well as to culture ages.  相似文献

A new species of sporulating saccharolytic anaerobe, designated as Clostridium quinii sp. nov., is described. A gram-positive strain BS1, was isolated from the granular metanogenic sludge (UASB) from a waste-water treatment plant at a sugar refinery. The strain exhibits a series of morphological stages, developing from a spore to a small rod to a motile rod (peritrichous flagella) in the exponential growth phase, and then swelling to form cigar-shaped cells, exhibiting tumbling movements, in the late exponential growth phase before finally becoming large nonmotile ovoid cells in the stationary phase. Swelling occurs as a result of glucose being taken up and stored as a glycogen-like substance. The main fermentation products when growing on glucose is H2, CO2, formate, acetate and ethanol as well as small amounts of butyrate during exponential growth. Lactate is formed during the stationary phase, when glucose is abundant. Optimal conditions for growth is 40–45°C and pH of around 7.4. The type strain BS1 contains 28.0% mol G+C.  相似文献

The breeding cycle, recruitment, population dynamics, growth and production ofScrobicularia plana were studied at one station in the Bidasoa estuary (SE of the Biscay Bay, Spain) during a period of 3 or 4 years. To complete this study, the reproduction for a period of 1 year was analysed at two other sites of the estuary. At none of the stations important differences in spawning activity were observed between the different years. The animals reproduced for the first time when they reached the age of two years. The histological data and the recruitment analysis showed only one spawning period between June and mid-September, with a peak in July-August. This breeding pattern differs from those shown by other southern European populations. The recruitment of spat (0.25–0.50 mm) occurred from June–July to October–December in different years. The population showed different annual patterns of density and population structure, depending on recruitment success. Growth was most rapid in spring and summer. The bivalves reached a mean length of 21.8 mm at the age of approximately 16 months; in the following year they grew to 30 mm. The growth during the life span fitted the theoretical growth equation of Von Bertalanffy. Annual production was in the range of 59.0–83.4 g AFDW m–2 year–1. The P/B ratios of the population varied between 0.84 and 1.48 (1.06 ± 1.36; ¯x ± 95% C.I.).  相似文献

Characterization of common cell surface-bound antigens inCandida albicans strains, particularly those expressed in the walls of mycelial cells might be useful in the diagnosis of systemic candidiasis. Hence, antigenic similarities among wall proteins and mannoproteins fromC. albicans clinical serotype A and B isolates, were studied using polyclonal (mPAbs) and monoclonal (MAb 4C12) antibodies raised against wall antigens from the mycelial form of a commonC. albicans serotype A laboratory strain (ATCC 26555). Zymolyase digestion of walls isolated from cells of the different strains studied grown at 37°C (germination conditions), released, in all cases, numerous protein and mannoprotein components larger than 100 kDa, along with a 33–34 kDa species. The pattern of major antigens exhibiting reactivity towards the mPAbs preparation was basically similar for all the serotype A and B isolates, though minor strain-specific bands were also observed. The immunodeterminant recognized by MAb 4C12 was found to be absent or present in very low amounts inC. albicans isolates other than the ATCC 26555 strain, yet high molecular weight species similar in size (e.g., 260 kDa) to the wall antigen against which MAb 4C12 was raised, were observed, particularly in wall digests from serotype A strains. Cell surface hydrophobicity, an apparently important virulence factor inC. albicans, of the cell population of each serotype B strain was lower than that of the corresponding serotype A counterparts, which is possibly due to the fact that the former strains exhibited a reduced ability to form mycelial filaments under the experimental conditions used.Abbreviations CSH cell surface hydrophobicity - IIF indirect immunofluorescence  相似文献

Nannochloropsis oculata (strain CCAP 849/1) was sampled at least every 12 h over a 26-d period of batch culture growth in a 12 h/12 h light/dark illumination cycle. Exponential cell-specific growth rate was 0.5 d–1. Cell division occurred during the dark phase, while ammonium uptake, pigment synthesis and cell volume increase occurred mainly during the light. Stationary phase cells were on average larger that the largest exponentially growing cells. The lag phase prior to cell division was short with the C/N ratio returning to 6.25 (from 28) within 2 d of refeeding with ammonium. Significant Chl.a synthesis commenced after this period; net synthesis of Chl.a ceased on exhaustion of the N-source with a 40% fall in levels by the end of the stationary phase. Levels of carotenoids per cell also declined during N-deprivation although per ml of culture levels remained constant. Ammonium-refeeding of N-deprived cells resulted in a very rapid rise in glutamine (Gln) and very high ratios of glutamine/glutamate (Gln/Glu peaking at 35 within 1 h); peak Gln/Glu was lower in cells refed in the dark or after a shorter period of N-deprivation. The major intracellular amino acids during exponential phase were Glu, Gln, alanine and arginine, but on exhaustion of the N-source, levels of Gln fell rapidly (Gln/Glu falling to below 0.1 from 0.5–0.9 in the light and 0.3 in darkness during exponential growth). During N-deprivation tyrosine accumulated within the cells. To determine the underlying processes to population growth in the rotifer Brachionus plicatilis, we conducted an experiment using 1.5 ml cultures for 70 days. All individuals were transferred daily to culture media containing algae, and the number of individuals, clutch sizes and number of deaths were counted. The population dynamics showed a typical sigmoid curve. The population density increased exponentially from 10 to 682 individuals during the first 7 days (exponential growth phase), and gradually up to about 1500 individuals during the next 30 days (post-exponential growth phase). The population density then remained at a constant level with small fluctuations during the rest of the experimental period (stationary phase). Mortalities appeared from the post-exponential growth phase and were almost constant at about 2% throughout the experimental period. The clutch size decreased from 5 to 1 during the first 5 days, and afterwards females laid only one egg each. The proportion of non-reproductive females increased from 30% (exponential growth phase) to 80% (post-exponential growth phase) to 90% (stationary phase). These results suggest that the exponential growth phase resulted from the imbalance between a high birth rate and a low death rate, while the stationary phase was maintained by the compensation between low birth and death rates.  相似文献

The investigation of growth dynamics and protein content in a batch Pseudomonas fluorescens culture grown in a synthetic medium with glucose as the sole source of carbon and energy showed that cells reversibly adhere to the walls of the cultivation flask during the first 2–3 h of growth. Over this time period, the total protein content of free and bound cells increased exponentially at a rate of 0.25 h–1, the fraction of proteins in cells being almost the same (60–70%). The protein content in the medium increased from 3 to 50 mg/l, reaching about 30% of the total protein of the culture. The addition of the exponential culture liquid filtrate to the medium together with the inoculum led to the complete inhibition of cell adhesion and a drastic activation of proteolysis, with a concurrent release of more than 80% of cellular proteins into the medium. After 3–5 h of growth, the concentration of extracellular proteins decreased to the control level. Exogenously added proteinase K inhibited cell adhesion, the effect being more pronounced for R-type than for S-type cells. The hypothesis is discussed that the short-term reversible adhesion of cells is regulated with the involvement of a mixture of hydrocarbons, which inactivate the functional activity of bacterial adhesins, and proteases, which digest these adhesins.  相似文献

The study of the effect of periplasmic glucan isolated from the root-nodule bacterium Sinorhizobium meliloti CXM1-188 on the symbiosis of another strain (441) of the same root-nodule bacterium with alfalfa plants showed that this effect depends on the treatment procedure. The pretreatment of alfalfa seedlings with glucan followed by their bacterization with S. meliloti 441 insignificantly influenced the nodulation parameters of symbiosis (the number of root nodules and their nitrogen-fixing activity) but induced a statistically significant increase in the efficiency of symbiosis (expressed as the masses of the alfalfa overground parts and roots). At the same time, the pretreatment of S. meliloti 441 cells with glucan brought about a considerable decrease in the nodulation parameters of symbiosis (the number of root nodules and their nitrogen-fixing activity decreased by 2.5–11 and 7 times, respectively). These data suggest that the stimulating effect of rhizobia on host plants may be due not only to symbiotrophic nitrogen fixation but also to other factors. Depending on the experimental conditions, the treatment of alfalfa plants with glucan and their bacterization with rhizobial cells enhanced the activity of peroxidase in the alfalfa roots and leaves by 10–39 and 12–27%, respectively.  相似文献

Summary Candida utilis var. major NRRL-Y-1084 was grown in a defined medium without a phosphorous (P) source. During the exponential phase, cells divided according to a specific growth rate of 0.32 h-1, which is lower than the usual rate for a balanced medium (0.4–0.6 h-1). The relative P content of the biomass decreased from 2.70% to 0.75% over a period of 6 h, including 2 h of cell division arrest. At the end of this period there was another interruption of cell division. After that, multiplication restarted at a considerably lower rate and it deviated slightly from the exponential pattern. The stationary phase began when biomass P content reached 0.4%–0.5%, slowly decreasing afterwards to 0.25–0.20%. Biomass synthesis was less affected than cell division by the relative decrease of endogenous P, the two processes differing partially in their kinetics. Cell lysis started shortly before the stationary phase and affected about 20% of the population by the end of the assay. RNA and P content of the resulting biomass were 2.4% and 0.25% respecitvely, P being mainly incorporated to RNA.The relationship of biomass production to glucose uptake was very low, probably because the marked P deficiency called for an increase in energy consumption for growth and specially for maintenance. Information on the life cycle of individual species is vital to an understanding of their ecology, particularly their distribution. In this investigation, aspects of the Macrothrix flabelligera (Cladocera, Macrothricidae) life cycle were studied in laboratory cultures under controlled conditions. Neonates from parthenogenetic females previously acclimatized were isolated and grown at constant temperature, in a 12-h/12-h light/dark cycle. Organisms were fed with a suspension of Scenedesmus bijugus at a concentration of 105 cells ml–1. Daily measurements of body length and age were made, to obtain the growth rate, size and age of primipara. M. flabelligera had high fecundity rates, producing broods of 13.6 eggs female–1. At 23.0°C, the duration of embryonic and post-embryonic development was 35.9 h and 4.9 days, respectively. The mean longevity was 12.7 days. A length–weight relationship was established and used to convert measured length into weight, in order to plot weight growth curves, as subsidiary information for future secondary production calculations.  相似文献
