Summary Responses of pollen grains of Nicotiana tabacum to high humidity (95% RH, 4 h) and temperature (38°/45° C, 4 h) stresses were investigated. Pollen grains were subjected to only RH or only temperature, or to both of these stresses. Their viability was assessed on the basis of the fluorochromatic reaction (FCR) test, and vigour was assessed on the basis of the time taken for in vitro germination as well as on the emergence of pollen tubes through the cut end of semi-vivo implanted styles. None of the stress conditions affected pollen viability and high RH or high temperature stress did not individually affect pollen vigour. However, pollen vigour was markedly affected when both the stresses were given together. Pollen grains subjected to high RH at 38° C took a longer time to germinate in vitro and the pollen tubes emerged later from the cut end of the semi-vivo styles; division of the generative cell was also delayed. Pollen grains subjected to high RH at 45° C failed to germinate in vitro, but did germinate on the stigma. Many pollen tubes subjected to this treatment showed abnormalities, and the growth of pollen tubes in the pistil was much slower than that observed in other treatments. Pollen samples subjected to all of the stress conditions were able to induce fruit and seed set. The implications of these results on the relationship between the FCR test and viability, and between viability and vigour, especially in stressed pollen, are discussed.  相似文献

Declining viability of pollen during storage at 24° C in atmospheres of 40% relative humidity (RH) and 75% RH was studied, with special emphasis on lipid changes. Pollens of Papaver rhoeas and Narcissus poeticus, characterized by a high linolenic acid content, were compared with Typha latifolia pollen which has a low linolenic acid content. The rationale behind this was to answer the question of whether lipid peroxidation is involved in the rapid viability loss and reduced membrane integrity of, in particular, the unsaturated-lipid pollen types. Viability and membrane integrity degraded more rapidly at 75% RH than at 40% RH. All pollen species showed deesterification of acyl chains of lipids but no detectable peroxidation at both RH levels. Considerable amounts of lipid-soluble antioxidants were detected that did not degrade during storage. Free fatty acids and lysophospholipids were formed during storage, the effects of which on membranes are discussed. These degradation products were very prominent in the short-lived Papaver pollen. The loss of viability does coincide with phospholipid deesterification. A significant decrease of the phospholipid content occurred at 75% RH, but not at 40% RH. Based on compositional analyses of phospholipids and newly formed free fatty acids, it was concluded that the deesterification of acyl chains from the lipids occurred at random. We suggest that, due to the low water content of the pollen, free radicals rather than unspecific acyl hydrolases are involved in the deesterification process.  相似文献

High humidity (95 % RH) and temperature (38/45 °C) stress for 4h applied to pollen grains of Nicotiana tabacum did not affect pollen viability, assessed on the basis of the fluorochromatic reaction test, but affected in vitro germination; pollen grains treated at 38 °C showed marked delay in germination, while those treated at 45 °C failed to germinate in vitro. The major ultrastructural effect of the stress was on RER. Stacks of RER, characteristically present in fresh pollen, were largely dissociated in the stressed pollen. The extent of dissociation of RER was greater in pollen samples stressed at 45 °C than at 38 °C. The generative cell did not show any obvious change in the stressed pollen. RER was restored in pollen grains which showed germination following culture; but not in those which failed to germinate. Apart from affecting other RER-related functions the dissociation of RER is likely to result in the destruction of long-lived mRNA and thus affect the ability of pollen grains to initiate protein synthesis needed for germination.  相似文献

Pollen of the palm Trachycarpus fortunei was kept at 25°C and relative humidities (RH) of 20, 55 and 98%. Changes in viability, water content and carbohydrates were measured over 2–17 days. Water content remained almost constant at 20 and 50% RH and increased dramatically at 98%. Pollen viability and germination rate remained almost constant over 14 days at 20% RH and decreased to about 2% after 7–9 days at 55% and to even less at 98% RH. Although the three experimental conditions were constant, qualitative and quantitative variations in pollen carbohydrates were recorded, even after pollen had lost its viability. The quantities of mono-, di- and polysaccharides varied with the period of pollen storage at the various RH. The greatest changes in glucose, fructose and sucrose content were recorded at 55 and 98% RH. At these relative humidities, maximum glucose and fructose content and minimum sucrose content occurred at maximum water content. Starch was not present in mature pollen but appeared and peaked after 7–9 days of pollen storage at 55 and 98%. Appearance of starch coincided with an increase in pectin content. PAS-positive cytoplasmic polysaccharides showed an increasing trend at 20% RH. A relation was found between pollen viability, water content and monosaccharide content. Pollen viability and germination capacity remained high at 20% RH for 14 days. At this relative humidity, pollen water, glucose and fructose contents remained almost constant, while sucrose reached its maximum value. The fluctuations of more complex carbohydrates (starch, pectins and PAS-positive cytoplasmic polysaccharides) were less easy to interpret. Changes observed under experimental conditions could simulate processes occurring in nature during pollen presentation and dispersal.  相似文献

Petunia hybrida and Cucurbita pepo pollen was exposed to 30 and 75% relative humidity (RH). Water content, viability and carbohydrate content (glucose, fructose, sucrose and starch) were measured at fixed intervals over 6 h. Water content of C. pepo pollen decreased drastically at both RHs, while P. hybrida pollen dehydrated slightly at RH 30% and hydrated at RH 75%. The pollen of the two species also showed very different sensitivity to dehydration. P. hybrida pollen was resistant to desiccation, with viability remaining around 80% throughout the experiment, whereas C. pepo was very sensitive to desiccation, showing an abrupt decrease in viability when its water content reached about 13% (fresh weight basis) at both RHs. Carbohydrate content was also different in the two species. Sucrose content increased soon after dehydration of P. hybrida pollen at RH 30%, whereas it remained almost constant in C. pepo pollen at the same RH. The results are discussed in the framework of basic pollen physiology and of plant reproductive strategy.  相似文献

Background and Aims

Pollinator-limited seed-set in some terrestrial orchids is compensated for by the presence of long-lived flowers. This study tests the hypothesis that pollen from these insect-pollinated orchids should be desiccation tolerant and relatively long lived using four closely related UK terrestrial species; Anacamptis morio, Dactylorhiza fuchsii, D. maculata and Orchis mascula.

Methods

Pollen from the four species was harvested from inflorescences and germinated in vitro, both immediately and also after drying to simulate interflower transit. Their tolerance to desiccation and short-term survival was additionally assessed after 3 d equilibration at a range of relative humidities (RHs), and related to constructed sorption isotherms (RH vs. moisture content, MC). Ageing of D. fuchsii pollen was further tested over 2 months against temperature and RH, and the resultant survival curves were subjected to probit analysis, and the distribution of pollen death in time (σ) was determined. The viability and siring ability, following artificial pollinations, were determined in D. fuchsii pollen following storage for 6 years at –20 °C.

Key Results

The pollen from all four species exhibited systematic increases in germinability and desiccation tolerance as anthesis approached, and pollen from open flowers generally retained high germinability. Short-term storage revealed sensitivity to low RH, whilst optimum survival occurred at comparable RHs in all species. Similarly, estimated pollen life spans (σ) at differing temperatures were longest under the dry conditions. Despite a reduction in germination and seeds per capsule, long-term storage of D. fuchsii pollen did not impact on subsequent seed germination in vitro.

Conclusions

Substantial pollen desiccation tolerance and life span of the four entomophilous orchids reflects a resilient survival strategy in response to unpredictable pollinator visitation, and presents an alternative approach to germplasm conservation.  相似文献

A method is described which is suitable for assessing the effects of relative humidity (RH) on the viability of two unicellular algae in experimental aerosols. Viable cells of Nannochloris atomus collected from the airborne state were detected by plating onto agar surfaces of an appropriate growth medium, whereas viable airborne cells of Synechococcus sp., because of unreliable growth on solid media, were determined by a liquid assay system. The assays were performed at intervals during short-term and prolonged storage of algal aerosols in chambers preconditioned to a selected RH and temperature. Both species showed the greatest loss in viability during the first minute after atomization, and the extent of this inactivation, as a function of RH, reflected the subsequent long-term survival. The airborne eukaryotic alga was unable to survive at an RH below 91%, whereas the airborne prokaryotic alga was comparatively stable over a wide humidity range. Initial inactivation was least and long-term survival best, for both species, at 94% RH.  相似文献

Twenty five surface samples/moss cushions were collected for palynological analysis from open areas of Reasi District, Jammu and Kashmir (India). These samples were used to investigate the relationships between extant vegetation and modern pollen spectra, which serve as modern analogue for the reliable ecological interpretation of fossil pollen records. The present vegetation in the region comprises tropical dry deciduous forests and subtropical pine forests with scattered stands of oak. The pollen analysis reveals that Pinus sp. (average 69% in the pollen assemblages), amongst the conifers, dominates the pollen rain, which can be attributed to its high pollen productivity and exceptional pollen dispersal efficiency. Cedrus sp. and Podocarpus sp. pollen contribute with an average of 16 and 5% to the total pollen rain. Other conifers such as Picea sp., Abies sp., Juniperus sp. and Tsuga sp., as well as broad-leaved taxa such as Quercus sp., Alnus sp., Betula sp., Carpinus sp., Corylus sp., Juglans sp., Ulmus sp., Salix sp., Elaeocarpus sp., Mallotus sp. and Aesculus sp., have lower averages of 1 to 4.5% in the total pollen rain which could be either due to their poor pollen dispersal efficiency or to the poor preservation in the samples. Tubuliflorae (average 25%), Poaceae (average 6.26%), Cerealia and other crop plants (average 7.68%) are other prominent taxa in the pollen rain. The nearly complete absence of members of tropical dry deciduous forests in the pollen spectra likely is due to the fact that most species in this vegetation type are not wind pollinated.  相似文献

High temperature exposure is widely used as a physical mutagenic agent to induce 2n gametes in Populus. However, whether high temperature exposure affects induced 2n pollen viability remains unknown. To clarify whether high temperature exposure affected the induced 2n pollen viability, 2n pollen induced by 38 and 41 °C temperatures, pollen morphology, 2n pollen germination in vitro, and crossing induced 2n pollen with normal gametes to produce a triploid was, based on observations of meiosis, conducted in Populus canescens. We found that the dominant meiotic stages (F = 56.6, p < .001) and the treatment duration (F = 21.4, p < .001) significantly affected the occurrence rate of induced 2n pollen. A significant decrease in pollen production and an increase in aborted pollen were observed (p < .001). High temperature sometimes affected in ectexine deposition and some narrow furrows were also analysed via details of ectexine structure. However, no significant difference in 2n pollen germination rate was observed between natural 2n pollen (26.7%) and high‐temperature‐induced 2n pollen (26.2%), and 42 triploids were created by crossing high‐temperature‐induced 2n pollen, suggesting that 38 and 41 °C temperatures exposure will not result in dysfunctional induced 2n pollen.  相似文献

Seeds are usually stored in physiological conditions in which they gradually lose their viability and vigor depending on storage conditions, storage time, and genotype. Very little is known about the underlying genetics of seed storability and seed deterioration. We analyzed a mutant in Arabidopsis disturbed in seed storability. This mutant was isolated as a grs (green-seeded) mutant in an abi3-1 (abscisic acid 3) mutant background. Genetic and physiological characterization showed that the monogenic grs mutant was not visibly green seeded and mapped on chromosome 4. This enhancer mutation did not affect the ABA sensitivity of seed germination or seed dormancy but was found to affect seed storability and seedling vigor. Seed storability was assessed in a controlled deterioration test, in which the germination capacity of the mutant decreased with the duration of the treatment. The decrease in viability and vigor was confirmed by storing the seeds in two relative humidities (RHs) for a prolonged period. At 60% RH, the mutant lost germinability, but storage at 32% RH showed no decrease of germination although seed vigor decreased. The decrease in viability and vigor could be related to an increase in conductivity, suggesting membrane deterioration. This was not affected by light conditions during imbibition, expected to influence the generation of active oxygen species. During seed maturation, ABI3 regulates several processes: acquiring dormancy and long-term storability and loss of chlorophyll. Our results indicate that GRS is a common regulator in the latter two but not of dormancy/germination.  相似文献

Cistus incanus and Myrtus communis are two species commonly found in Mediterranean shrublands, flowering respectively in spring and summer. Pollen of single flowers of C. incanus is available for dispersal for 8-10 hours, during which time viability decreases from 91% to 61%. Field measurements of the variation in the percent pollen viability during the anthesis of single flowers of M. communis shows that, immediately after anthesis, pollen viability is very high; by the end of the first day it significantly declines and 36 hours after anthesis almost all grains are unviable. Experiments carried out under controlled environment showed that, in C. incanus, the greatest loss of viability occurs at 100% humidity and high temperatures, while under dry conditions pollen viability remains high throughout a wide temperature range. Similarly, pollen viability of M. communis remains high for several hours at high temperature and dry conditions, while it rapidly decreases at high humidity. In conclusion, the temperature experiments indicate that in C. incanus and M. communis, pollen viability does not appear to be drastically reduced if the relative humidity of the environment is low. Therefore, humidity is a far more important factor determining pollen viability loss in both species.  相似文献

Background and aimsThis paper is about pollen ecophysiology, anther opening, pollen dispersal and the timing of the male and female phases in Parietaria judaica (Urticaceae).MethodsEcophysiological (effects of different relative humidities (RHs) and osmotic relationships) and cytological methods (stigmatic receptivity, pollen viability, histology and histochemistry) were used to determine pollen and pollination features during the long blooming period of this species.Key resultsPollen is dispersed by rapid uncurling of the filament and anther opening. The filament and anther lack cells with lignified wall thickenings, which are usually responsible for anther opening and ballistic pollen dispersal. Instead, dispersal is the result of the sudden movement of the filament. Pollen is of the partially hydrated type, i.e. with a water content greater than 30% at shedding, and readily loses water, and hence viability, at low RH. Pollen carbohydrate reserves differ with season. Starchless grains germinate quickly and are less subject to water loss. Flowers are protogynous, pollen release occurring only after complete cessation of the female phase within an inflorescence.ConclusionParietaria has partially hydrated pollen which differs from typical pollen of this type because of its reduced size and the absence of callose. Because of its low water content at the time of shedding it survives better at higher RH. Dispersal and pollination are adapted to pollen features.  相似文献

The aim of this study was to determine if the loss of germinability and viability of beech (Fagus sylvatica L.) seeds stored at different variants of temperature (4, 20, and 30 °C) and relative humidity (RH: 45 and 75 %) is associated with a loss of membrane integrity and changes in lipid composition. Beech seeds stored for 9 weeks gradually lost viability at a rate dependent on temperature and humidity. The harmful effect of temperature increased with growing humidity. The loss of seed viability was strongly correlated with an increase in membrane permeability and with production of lipid hydroxyperoxides (LHPO), which was regarded as an indicator of peroxidation of unsaturated fatty acids. The condition of membranes was assessed on the basis of their permeability and the state of lipid components: phospholipids and fatty acids. During seed storage we observed a decline in concentration of individual phospholipids and fatty acids, proportional to the loss of seeds viability. We also detected a decrease in concentrations of α-tocopherol and sterols, which play an important role in protection of membranes against the harmful influence of the environment. Our results show that the germinability of beech seeds declines rapidly at temperature above 0 °C and growing humidity. This is due mainly to the loss of membrane integrity, caused by peroxidation of unsaturated fatty acids.  相似文献

Main conclusion

Pollen tube growth in styles was strongly inhibited by temperature above 35 °C, and the yield of cotton decreased because of the adverse effect of high temperatures during square development. High-temperature stress during flowering influences the square development of upland cotton (Gossypium hirsutum L.) and cotton yield. Although it is well known that square development is sensitive to high temperature, high-temperature sensitive stages of square development and the effects of high temperature on pollen tube growth in the styles are unknown. The effect of high temperature on anther development corresponding to pollen vigor is unknown during anther development. The objectives of this study were to identify the stages of square development that are sensitive to high temperatures (37/30 and 40/34 °C), to determine whether the abnormal development of squares influenced by high temperature is responsible for the variation in the in vitro germination percent of pollen grains at anthesis, to identify the effect of high temperature on pollen germination in the styles, and to determine pollen thermotolerance heterosis. Our results show that the stages from the sporogenous cell to tetrad stage (square length <6.0 mm) were the most sensitive to high temperature, and the corresponding pollen viability at anthesis was consistent with the changes in the square development stage. Pollen tube growth in the styles was strongly inhibited by temperature above 35 °C, and the yield of cotton decreased because of the effect of high temperature during square development. The thermotolerance of hybrid F1 pollen showed heterosis, and pollen viability could be used as a criterion for screening for high-temperature tolerance cultivars. These results can be used in breeding to develop new cotton cultivars that can withstand high-temperature conditions, particularly in a future warmer climate.

The development of the male reproductive structures of American chestnut (Castanea dentata) is described to advance our understanding of its reproductive behavior. This information has been vital in the development of a strategy to collect pollen grains from male catkins suitable for in vitro germination and transformation experiments. Cutting male catkins into small segments and rolling them over a culture plate resulted in evenly dispersed and large amounts of pollen with minimal unwanted accessory floral parts. To optimize pollen viability, the effect of various storage conditions on in vitro germination was examined. Our results showed that initial storage at 4°C for 2 weeks significantly increased percent germination as compared to freshly collected pollen and those stored directly at −20°C or −80°C. This also means that for long-term storage of American chestnut pollen, the catkins should first be kept at 4°C for a couple of weeks and then at −80°C. The use of pollen grains with high viability is necessary for the transformation of American chestnut pollen. To optimize pollen transformation via particle bombardment, the effects of target distance, target pressure, and pollen developmental stage were examined. Statistical analysis showed that bombardment of ungerminated pollen at 1,100 psi resulted in the highest percent transient GFP expression (4.1%).  相似文献

Aims: To assess the effectiveness of iodine-treated biocidal filter media against bacterial spore aerosols. Methods and Results: Bacillus subtilis spores were aerosolized and introduced into a filtration system. Both treated and untreated filters exhibited high viable removal efficiency (>99·996%) with negligible variation in pressure drop during the entire experiment. The viability of collected spores on the filter was investigated by enumeration of spores extracted from the filter by vortexing. At room temperature and low relative humidity (RH), the survival fraction of the treated filter was significantly lower than that of the untreated filter (P-value < 0·05). Meanwhile, at room temperature and high RH and at high temperature and high RH, the survival fractions on the treated medium were statistically the same as the untreated control at room temperature and low RH. Conclusions: Both treated and untreated filters achieved excellent viable removal efficiency for spores. The pressure drop of the treated filter was not affected by the iodine treatment. The viability of collected bacterial spores was decreased because of the exertion of iodine disinfectant. Significance and Impact of the Study: The evaluation demonstrates that the iodine-treated filter is a viable medium for respiratory protection against infectious spore aerosols. The results warrant further evaluation of smaller biological agents, which exhibit higher penetration.  相似文献

Engineered materials to improve the shelf-life of desiccated microbial strains are needed for cost-effective bioaugmentation strategies. High temperatures and humidity of legume-growing regions challenge long-term cell stabilization at the desiccated state. A thermostable xeroprotectant core and hydrophobic water vapour barrier shell encapsulation technique was developed to protect desiccated cells from the environment. A trehalose core matrix increased the stability of desiccated Bradyrhizobium by three orders of magnitude over 20 days at 32°C and 50% relative humidity (RH) compared to buffer alone; however, the improvement was not deemed sufficient for a shelf-stable bioproduct. We tested common additives (skim milk, albumin, gelatin and dextran) to increase the glass transition temperature of the desiccated product to provide further stabilization. Albumin increased the glass transition temperature of the trehalose-based core by 40°C and stabilized desiccated Bradyrhizobium for 4 months during storage at high temperature (32°C) and moderate humidity (50% RH) with only 1 log loss of viability. Although the albumin-trehalose core provided exceptional protection against high temperature, it was ineffective at higher humidity conditions (75%). We therefore incorporated a paraffin shell, which protected desiccated cells against 75% RH providing proof of concept that core and shell encapsulation is an effective strategy to stabilize desiccated cells.  相似文献

Gentiana leucomelaena manifests dramatic flower color polymorphism, with both blue‐ and white‐flowered individuals (pollinated by flies and bees) both within a population and on an individual plant. Previous studies of this species have shown that pollinator preference and flower temperature change as a function of flower color throughout the flowering season. However, few if any studies have explored the effects of flower color on both pollen viability (mediated by anther temperature) and pollinator preference on reproductive success (seed set) in a population or on individual plants over the course of the entire flowering season. Based on prior observations, we hypothesized that flower color affects both pollen viability (as a function of anther temperature) and pollen deposition (as a function of pollinator preference) to synergistically determine reproductive success during the peak of the flowering season. This hypothesis was tested by field observations and hand pollination experiments in a Tibetan alpine meadow. Generalized linear model and path analyses showed that pollen viability was determined by flower color, flowering season, and anther temperature. Anther temperature correlated positively with pollen viability during the peak of the early flowering season, but negatively affected pollen viability during the peak of the mid‐ to late flowering season. Pollen deposition was determined by flower color, flowering season (early, or mid‐ to late season), and pollen viability. Pollen viability and pollen deposition were affected by flower color that in turn affected seed set across the peak of the flowering season (i.e., when the greatest number of flowers were being pollinated). Hand pollination experiments showed that pollen viability and pollen deposition directly influenced seed set. These data collectively indicate that the preference of pollinators for flower color and pollen viability changed during the flowering season in a manner that optimizes successful reproduction in G. leucomelaena. This study is one of a few that have simultaneously considered the effects of both pollen viability and pollen deposition on reproductive success in the same population and on individual plants.  相似文献

Survival of two Lactobacillus kefir strains after spray drying in reconstituted skim milk with or without the addition of 12.5 g monosodium glutamate/l, 20 g sucrose/l, or 20 g fructo-oligosaccharides (FOS)/l and during subsequent storage under different conditions of temperature (20 and 30°C) and relative humidity (RH) (0, 11 and 23%) was evaluated. After being dried, L. kefir 8321 and L. kefir 8348 had a decrease in viability of 0.29 and 0.70 log cfu/ml respectively, while the addition of different protectants improved the survival of both strains significantly. During storage, bacterial survival was significantly higher under lower conditions of RH (0–11%), and monosodium glutamate and FOS proved to be the best protectants.  相似文献

Data on the survival of pollen ofTypha latifoliaL. stored forup to 261 d over seven different saturated salt solutions (providing0.5 to 66% relative humidity) and six different constant temperatures(from -5 to +45 °C) were analysed to quantify the effectof air-dry storage environment on pollen longevity. Pollen survivalcurves conformed much more closely to negative cumulative normaldistributions than to negative exponential relations. Estimatesofp50(storage period required to reduce pollen viability to50%), provided by negative cumulative normal distributions,were available from 37 different storage environments in whichpollen viability was reduced below 50%. Once observations at0.5% and 5.5% relative humidity were excluded from analysis,there was a negative logarithmic relation between these estimatesof longevity and pollen moisture content (%, wet basis) anda curvilinear semi-logarithmic relation between longevity andtemperature. When the negative logarithmic relation betweenlongevity and moisture content was replaced by a negative semi-logarithmicrelation between longevity and the relative humidity of thestorage environment the resultant model was less satisfactory,principally because pollen longevity over saturated solutionsof calcium nitrate (43–62% relative humidity) and sodiumnitrite (60–66% relative humidity) were consistently greaterand smaller, respectively, than fitted values. Notwithstandingthese errors, comparison between the fitted relations and observationsat the two lowest relative humidities provided estimates ofthe lower-relative-humidity limits to these relations. Theseprovisional estimates varied with storage temperature beinglowest at 25 °C (<5.5% relative humidity). However, therewas no linear trend to that variation (P>0.25): the meanestimate was 11.9 (s.e.=1.4)%. The considerable similaritiesamong models of pollen longevity in air-dry storage, and theirestimated lower limits, and those developed previously for orthodoxseeds and spores are discussed.Copyright 1999 Annals of BotanyCompany.
