The NMR structure of the Antheraea polyphemus pheromone-binding protein 1 at pH 4.5, ApolPBP1A, was determined at 20 degrees C. The structure consists of six alpha-helices, which are arranged in a globular fold that encapsulates a central helix alpha7 formed by the C-terminal polypeptide segment 131-142. The 3D arrangement of these helices is anchored by the three disulfide bonds 19-54, 50-108 and 97-117, which were identified by NMR. Superposition of the ApolPBP1A structure with the structure of the homologous pheromone-binding protein of Bombyx mori at pH 4.5, BmorPBPA, yielded an rmsd of 1.7 A calculated for the backbone heavy-atoms N, Calpha and C' of residues 10-142. In contrast, the present ApolPBP1A structure is different from a recently proposed molecular model for a low-pH form of ApolPBP1 that does not contain the central helix alpha7. ApolPBP1 exhibits a pH-dependent transition between two different globular conformations in slow exchange on the NMR chemical shift timescale similar to BmorPBP, suggesting that the two proteins use the same mechanism of ligand binding and ejection. The extensive sequence homology observed for pheromone-binding proteins from moth species further implies that the previously proposed mechanism of ligand ejection involving the insertion of a C-terminal helix into the pheromone-binding site is a general feature of pheromone signaling in moths.  相似文献

A novel olefinic analog, Z,Z-1,12,14-heptadecatriene, was synthesized from Z,Z-11,13-hexadecadienal, a major pheromone component of the navel orangeworm, Amyelois transitella (Walker) (Lepidoptera: Pyralidae), and evaluated as a potential disruptant of mating communication in commercial almond orchards. The effectiveness of the triene as a disruptant was compared to that of the aldehyde by evaluating the reduction in captures of feral males in female-baited traps and the reduction in mating success for females. The triene was highly effective in bringing about reductions in male captures in traps in all tests relative to controls. This effectiveness lasted as long as 5 weeks in one test and was influenced by type of dispenser, release rate and proximity of dispensers to female-baited traps. In none of the tests did the triene outperform the aldehyde in reducing male captures. The results of mating success tests showed the triene to be ineffective relative to controls (26% reduction) while the aldehyde yielded 100% reductions in matings during a 6-day period.
Résumé Le Z-Z-1,12,14-heptacécatriène, qui mime l'aldéhyde Z-Z-11,13-hexadécadénial, principal composé de la phéromone de A. transitella Walker, a été essayé pour interrompre les accouplements dans la nature. Un conditionnement avec plastique stratifié a mieux dispersé le triène que la présentation en capsules de polyéthylène. Comme dans le cas des autres produits examinés précédemment, ce triène a interrompu l'attraction sexuelle (réduction du nombre de mâles capturés dans les pièges, réduction du nombre d'accouplements) de A. transitella dans les vergers d'amandiers, pendant moins longtemps que l'aldéhyde. Placé dans des pièges contenant des femelles vierges, il est efficace comme l'aldehyde pendant 5 semaines, pendant 3 semaines lorsqu'il est placé autour des pièges et pendant 13 jours quand il est utilisé dans des parcelles de 9 arbres. Des essais ultérieurs avec d'autres présentations et des quantités plus élevées de triène devraient montrer si ce produit peut être utilisé pour l'interruption des accouplements. Les analogues de phéromones ont généralement une plus grande stabilité et un coût de synthèse moins élevé que les aldéhydes.

The effect of reduced application rate, soil temperature at shallow depth (2.5 cm), and soil type on the efficacy of Steinernema carpocapsae against the navel orangeworm, Amyelois transitella, was evaluated in six field trials employing 1 m2 plots conducted from November 2003 through December 2004 in Madera and Kern Counties, California. Nematodes were applied at a concentration of 100,000 infective juveniles (IJs)/m2 (109/ha) in a volume of 187 ml water/m2 (1870 L/ha) with a post-application irrigation in all trials. Mortality ranged from 7.9 to 64.9% in successful trials and percent reduction in live larvae per plot was as high as 74.6%. Percent reduction and mortality were highly correlated (r2 = 0.78) and larval reduction typically was 10–11% greater than mortality for any treatment. In one trial, although nematode treatment significantly increased mortality compared to the controls, the treatment was deemed unsatisfactory because mortality was <15%. Soil temperature in this trial rose to 39 °C within 5 h after application. Nematodes failed in two other trials when soil temperature fell below freezing (minimum temperatures −3.0, −5.5 °C, respectively) several times in a 5-day period. We conclude that a commercially feasible application volume of 1870 L water/ha followed by post-application irrigation at this same rate was effective, and that soil maximum temperature at or below 32 °C during the first 24 h after application is necessary for treatment success.  相似文献

The Bombyx mori pheromone-binding protein (BmorPBP) undergoes a pH-dependent conformational transition from a form at basic pH, which contains an open cavity suitable for ligand binding (BmorPBPB), to a form at pH 4.5, where this cavity is occupied by an additional helix (BmorPBPA). This helix α7 is formed by the C-terminal dodecapeptide 131-142, which is flexibly disordered on the protein surface in BmorPBPB and in its complex with the pheromone bombykol. Previous work showed that the ligand-binding cavity cannot accommodate both bombykol and helix α7. Here we further investigated mechanistic aspects of the physiologically crucial ejection of the ligand at lower pH values by solution NMR studies of the variant protein BmorPBP(1-128), where the C-terminal helix-forming tetradecapeptide is removed. The NMR structure of the truncated protein at pH 6.5 corresponds closely to BmorPBPB. At pH 4.5, BmorPBP(1-128) maintains a B-type structure that is in a slow equilibrium, on the NMR chemical shift timescale, with a low-pH conformation for which a discrete set of 15N-1H correlation peaks is NMR unobservable. The full NMR spectrum was recovered upon readjusting the pH of the protein solution to 6.5. These data reveal dual roles for the C-terminal tetradecapeptide of BmorPBP in the mechanism of reversible pheromone binding and transport, where it governs dynamic equilibria between two locally different protein conformations at acidic pH and competes with the ligand for binding to the interior cavity.  相似文献

The distribution of odorant-binding proteins among olfactory sensilla of three moth species was studied by immuno-electron microscopy. Two polyclonal antisera were used in a post-embedding labelling protocol on sections of cryo-substituted antennae. The first was directed against the pheromone-binding protein (PBP) of Antheraea polyphemus, the second against the general odorant-binding protein (GOBP) of the same species. Immunoblots showed that these antisera were highly specific; both antisera did, however, cross-react with related proteins in the related species A. pernyi, and in the bombycid moth B. mori. PBP and GOBP were localized only in olfactory sensilla trichodea and sensilla basiconica, the principal site being the sensillum lymph surrounding the sensory dendrites. In the males of all three species, the pheromone-sensitive long sensilla trichodea exclusively contained PBP. the majority of the sensilla basiconica in both sexes in these species contained GOBP; these sensilla are known to respond to plant and other general odours. Some sensilla were not labelled by either antiserum; presumably, these held an odorantbinding protein of a different subfamily. Never were PBP and GOBP co-localized in the same sensillum. Two observations deserve special attention: (1) PBP was also found in a few sensilla in females, and (2) in B. mori, where the long sensilla trichodea have a different functional specificity in males (pheromone) and females (plant odours), the expression of the odorant-binding protein (males: PBP; females: GOBP) is similarly different. Summary The antennal lobe of both sexes of the silk moth Bombyx mori contains 55–60 ventrally located antennal glomeruli; in addition, that of the male contains a dorsal macroglomerular complex (MGC). A group of identifiable glomeruli consisting of two lateral large glomeruli (LLG) and four medial small glomeruli (MSG) is present in both sexes, but the LLG are greatly enlarged in the female. A MGC is also present in the male gypsy moth Lymantria dispar and male giant silk moth Antheraea polyphemus. The MGC in all of these species is organized into 3–4 distinct levels of glomeruli. Antennal sensory fibers were stained by cobalt backfills in B. mori, A. polyphemus, and L. dispar. Most fibers stained from cut long hairs (sensilla trichodea) projected to MGC in males and LLG in both sexes of B. mori. The distribution of fibers in the MGC of B. mori was topographically biased in that a majority of fibers from anterior branches projected medially in MGC while most fibers from posterior branches projected laterally or anteriorly. Terminal arborizations of single fibers were each restricted to a single glomerular level of the MGC. Fibers projecting to the posterior antennal center were frequently stained in cut-hair and control preparations, apparently by uptake of cobalt through intact sensilla on flagellar branches.  相似文献

Summary The fine structure of the superposition eye of the Saturniid moth Antheraea polyphemus Cramer was investigated by electron microscopy. Each of the approximately 10000 ommatidia consists of the same structural components, but regarding the arrangement of the ommatidia and the rhabdom structure therein, two regions of the eye have to be distinguished. In a small dorsal rim area, the ommatidia are characterized by rectangularly shaped rhabdoms containing parallel microvilli arranged in groups that are oriented perpendicular to each other. In all other ommatidia, the proximal parts of the rhabdoms show radially arranged microvilli, whereas the distal parts may reveal different patterns, frequently with microvilli in two directions or sometimes even in one direction. Moreover, the microvilli of all distal cells are arranged in parallel to meridians of the eyes. By virtue of these structural features the eyes should enable this moth not only discrimination of the plane of polarized light but also skylight-orientation via the polarization pattern, depending on moon position. The receptor cells exhibit only small alterations during daylight within the natural diurnal cycle. However, under illumination with different monochromatic lights of physiological intensity, receptor cells can be unbalanced: Changes in ultrastructure of the rhabdomeres and the cytoplasm of such cells are evident. The effects are different in the daytime and at night. These findings are discussed in relation to the breakdown and regeneration of microvilli and the influence of the diurnal cycle. They are compared with results on photoreceptor membrane turnover in eyes of other arthropod species.  相似文献

Plasma hyaluronan-binding protein (PHBP), an activator of factor VII and prourokinase, is a serine protease circulating as a single-chain proenzyme (pro-PHBP). Pro-PHBP converts to the active two-chain form through autoproteolysis, and effectors that modulate autoactivation can regulate PHBP-mediated processes. Here, we show that histone promotes pro-PHBP autoactivation in vivo. Histone bound to pro-PHBP and promoted intermolecular pro-PHBP binding. Histone-mediated pro-PHBP activation in plasma leads to the formations of bradykinin and PHBP−α2-antiplasmin complex as well as histone degradation. Pro-PHBP activation was observed in the circulation of mice after injection of histone or lipopolysaccharide, which induced septic response accompanying extracellular histone release. Our results suggest pathophysiological relevance of histone-dependent pro-PHBP activation in hyperinflammatory process.  相似文献

Glycogen, a branched polymer of glucose, acts as an intracellular carbon and energy reserve in many tissues and cell types. An important pathway for its degradation is by transport to lysosomes in an autophagy-like process. It has been proposed that starch-binding domain-containing protein 1 (Stbd1) may participate in this mechanism by anchoring glycogen to intracellular membranes. In addition, Stbd1 has been reported to interact with a known autophagy protein, GABARAPL1, a member of the Atg8 family. Here, we confirm this interaction and identify an Atg8 interacting motif (AIM) in Stbd1 necessary for GABARAPL1 binding as judged by co-immunoprecipitation from cell extracts and co-localization in cells as evidenced by immunofluorescence microscopy. The AIM sequence of Stbd1 200HEEWEMV206 lies within a predicted disordered region of the molecule and fits the consensus of other AIM sequences in cargo-specifying proteins such as p62 and Nix. Mutation of the AIM, including single point mutations of either W203 or V206, eliminated the co-localization of Stbd1 with both over-expressed and endogenous GABARAPL1. Stbd1 may therefore function as a novel cargo binding protein that delivers glycogen to lysosomes in an autophagic pathway that could be termed “glycophagy”.  相似文献

Alagille syndrome (AGS) is a dominantly inherited disorder characterized by liver disease in combination with heart, skeletal, ocular, facial, renal, and pancreatic abnormalities. We have recently demonstrated that Jagged1 (JAG1) is the AGS gene. JAG1 encodes a ligand in the Notch intercellular signaling pathway. AGS is the first developmental disorder to be associated with this pathway and the first human disorder caused by a Notch ligand. We have screened 54 AGS probands and family members to determine the frequency of mutations in JAG1. Three patients (6%) had deletions of the entire gene. Of the remaining 51 patients, 35 (69%) had mutations within JAG1, identified by SSCP analysis. Of the 35 identified intragenic mutations, all were unique, with the exceptions of a 5-bp deletion in exon 16, seen in two unrelated patients, and a C insertion at base 1618 in exon 9, also seen in two unrelated patients. The 35 intragenic mutations included 9 nonsense mutations (26%); 2 missense mutations (6%); 11 small deletions (31%), 8 small insertions (23%), and 1 complex rearrangement (3%), all leading to frameshifts; and 4 splice-site mutations (11%). The mutations are spread across the coding sequence of the gene within the evolutionarily conserved motifs of the JAG1 protein. There is no phenotypic difference between patients with deletions of the entire JAG1 gene and those with intragenic mutations, which suggests that one mechanism involved in AGS is haploinsufficiency. The two missense mutations occur at the same amino acid residue. The mechanism by which these missense mutations lead to the disease is not yet understood; however, they suggest that mechanisms other than haploinsufficiency may result in the AGS phenotype.  相似文献

Presynaptic group III metabotropic glutamate receptors (mGluRs) and Ca2+ channels are the main neuronal activity-dependent regulators of synaptic vesicle release, and they use common molecules in their signaling cascades. Among these, calmodulin (CaM) and the related EF-hand Ca2+-binding proteins are of particular importance as sensors of presynaptic Ca2+, and a multiple of them are indeed utilized in the signaling of Ca2+ channels. However, despite its conserved structure, CaM is the only known EF-hand Ca2+-binding protein for signaling by presynaptic group III mGluRs. Because the mGluRs and Ca2+ channels reciprocally regulate each other and functionally converge on the regulation of synaptic vesicle release, the mGluRs would be expected to utilize more EF-hand Ca2+-binding proteins in their signaling. Here I show that calcium-binding protein 1 (CaBP1) bound to presynaptic group III mGluRs competitively with CaM in a Ca2+-dependent manner and that this binding was blocked by protein kinase C (PKC)-mediated phosphorylation of these receptors. As previously shown for CaM, these results indicate the importance of CaBP1 in signal cross talk at presynaptic group III mGluRs, which includes many molecules such as cAMP, Ca2+, PKC, G protein, and Munc18-1. However, because the functional diversity of EF-hand calcium-binding proteins is extraordinary, as exemplified by the regulation of Ca2+ channels, CaBP1 would provide a distinct way by which presynaptic group III mGluRs fine-tune synaptic transmission.  相似文献

【目的】为了更好地了解性信息素结合蛋白(pheromone binding proteins, PBPs)在桃蛀螟Conogethes punctiferalis (Guenée)嗅觉识别过程中的作用,明确其与配体化合物的结合特性。【方法】本研究利用RT-PCR结合RACE方法克隆了桃蛀螟一个性信息素结合蛋白基因;采用Real-time PCR方法分析了该蛋白在桃蛀螟不同发育阶段及雌雄蛾间的表达差异;利用荧光竞争结合实验对Cpun-PBP1蛋白与16种配基化合物的结合特性进行了分析。【结果】克隆了一个桃蛀螟性信息素结合蛋白基因,命名为Cpun-PBP 1(GenBank登录号:KP027486)。Cpun-PBP 1开放阅读框全长510 bp,编码 169个氨基酸,预测分子量为19.12 kDa,等电点为5.09,N-末端包括由起始位置开始的30个氨基酸组成的信号肽。蛋白特征分析显示,该氨基酸序列具有昆虫气味结合蛋白的典型特征,即含有6个保守的半胱氨酸残基。Cpun-PBP 1在桃蛀螟成虫阶段表达量最高,且几乎全部在触角中表达,卵期微量表达,幼虫期和蛹期均不表达。通过构建Cpun-PBP 1原核表达载体,诱导并获得Cpun-PBP 1重组蛋白。荧光竞争结合实验对2种性信息素组分和14种寄主植物挥发物的结合力发现,Cpun-PBP1不但能有效地与桃蛀螟性信息素组分(顺-10-十六碳烯醛和十六醛)结合,结合常数分别为7.32和9.39 μmol/L;还能与8种寄主植物挥发物有效结合;其中,与莰烯的结合能力最强,结合常数为3.76 μmol/L。【结论】根据这些结果,我们推测Cpun-PBP1在桃蛀螟感受性信息素和寄主植物挥发物的过程中发挥着双重作用。  相似文献

In this study, we report that a single mutation of cysteine 18 to isoleucine (C18I) in Escherichia coli Hha abolishes the repression of the hemolysin operon observed in the wild-type protein. The phenotype also includes a significant decrease in the growth rate of E. coli cells at low ionic strength. Other substitutions at this position (C18A, C18S) have no observable effects in E. coli growth or hemolysin repression. All mutants are stable and well folded and bind H-NS in vitro with similar affinities suggesting that Cys 18 is not directly involved in H-NS binding but this position is essential for the activity of the H-NS/Hha heterocomplexes in the regulation of gene expression.  相似文献

Metal binding to the C-terminal region of the copper-responsive repressor protein CopY is responsible for homodimerization and the regulation of the copper homeostasis pathway in Enterococcus hirae. Specific involvement of the 38 C-terminal residues of CopY in dimerization is indicated by zonal and frontal (large zone) size-exclusion chromatography studies. The studies demonstrate that the attachment of these CopY residues to the immunoglobulin-binding domain of streptococcal protein G (GB1) promotes dimerization of the monomeric protein. Although sensitivity of dimerization to removal of metal from the fusion protein is smaller than that found for CopY (as measured by ultracentrifugation studies), the demonstration that an unrelated protein (GB1) can be induced to dimerize by extending its sequence with the C-terminal portion of CopY confirms the involvement of this region in CopY homodimerization.  相似文献

This communication describes SAXS data based global structures of tetravalent antibody CD4–IgG2 and its dimeric to pentameric complexes with gp120s. Comparison of models brought forth that while the two CD4s grafted on each arm remain tightly packed in the unliganded antibody, they enable binding of first two gp120s preferentially to the same Fab arm in an asymmetric manner. Retention of residues in the CD4–Fab linker earlier reasoned to enable bi-fold collapse of gp120-bound soluble CD4, and observed asymmetry of the (CD4–IgG2)/(gp120)2 complex suggest that encoded flexibility in CD4–Fab linker is a critical structure–function factor for this broad spectrum neutralizing antibody.  相似文献

The Heterochromatin Protein 1 (HP1) is a conserved protein which is best known for its strong association with the heterochromatin of Drosophila melanogaster. We previously demonstrated that another important property of HP1 is its localization to the telomeres of Drosophila, a feature that reflects its critical function as a telomere capping protein. Here we report our analysis of the euchromatic sites to which HP1 localizes. Using an anti-HP1 antibody, we compared immunostaining patterns on polytene chromosomes of the Ore-R wild type laboratory strain and four different natural populations. HP1 was found to accumulate at specific euchromatic sites, with a subset of the sites conserved among strains. These sites do not appear to be defined by an enrichment of known repetitive DNAs. Comparisons of HP1 patterns among several Drosophila species revealed that association with specific euchromatic regions, heterochromatin and telomeres is a conserved characteristic of HP1. Based on these results, we argue that HP1 serves a broader function than typically postulated. In addition to its role in heterochromatin assembly and telomere stability, we propose that HP1 plays an important role in regulating the expression of many different euchromatic regions.  相似文献

Formylpeptide receptor (FPR1) is involved in inflammation, which is important in the pathogenesis of diverse conditions, including common diseases and cancers. To date, little is known about the relationships between FPR1 and such diseases, aside from the fact that FPR1 is related to periodontitis, which is implicated in systemic diseases such as stomach cancer. We hypothesized that FPR1 polymorphisms related to periodontal disease may confer susceptibility to stomach cancer. Two single nucleotide polymorphisms (SNPs) in the second extracellular region and C-terminus of the formylpeptide receptor gene were analyzed in 1531 consecutive autopsy cases in the Japanese elderly. The tri-allelic SNP of rs1042229 was detected by modified melting temperature analysis. Homozygous K alleles of rs1042229 were associated with stomach cancer (Odds ratio [OR] = 1.62, confidence interval [CI] = 1.05–2.48, p = 0.028). In the analysis of the recessive model of the K allele, FPR1 was associated with a high risk of stomach cancer (OR = 1.73, CI = 1.15–2.55, p = 0.0075). The risk allele for stomach cancer pointed in the same direction as periodontitis. This is the first study to evaluate polymorphisms of the FPR1 gene in stomach cancer to find a positive association between these polymorphisms and stomach cancer. Further studies on the relationship between stomach cancer and the FPR1 gene are warranted.  相似文献
