The family Percidae is among the most speciose families of northern hemisphere fishes with > 178 178 North American species and 14 Eurasian species. Previous phylogenetic studies have been hampered by a lack of informative characters, inadequate taxonomic sampling, and conflicting data. We estimated phylogenetic relationships among 54 percid species (9 of 10 genera and all but one subgenus of darters) and four outgroup taxa using mitochondrial DNA data from the 12S rRNA and cytochrome b genes. Four primary evolutionary lineages were consistently recovered: Etheostomatinae (Ammocrypta, Crystallaria, Etheostoma, and Percina), Perca, Luciopercinae (Romanichthys, Sander, and Zingel), and Gymnocephalus. Except Etheostoma and Zingel, all polytypic genera were monophyletic. The Etheostoma subgenus Nothonotus failed to resolve with other members of the genus resulting in a paraphyletic Etheostoma. The subfamily Percinae (Gymnocephalus and Perca) was not recovered in phylogenetic analyses with Gymnocephalus sister to Luciopercinae. Etheostomatinae and Romanichthyini were never resolved as sister groups supporting convergent evolution as the cause of small, benthic, stream-inhabiting percids in North American and Eurasian waters.  相似文献

Base compositions and differential melting rate profiles of genomic DNAs from twenty species of North American cyprinid fishes were generated via thermal denaturation. Base pair composition expressed as % GC values ranged among the twenty species from 36.1–41.3%. This range is considerably broader than that observed at comparable taxonomic levels in other vertebrate groups. Both the range and average difference in base pair composition between species in the diverse and rapidly evolving genus Notropis were considerably greater than those between species in other North American cyprinid genera. This may indicate that genomic changes at the level of base pair composition are frequent and possibly important events in cyprinid evolution. Compositional heterogeneity and asymmetry values among the twenty species were uniform and low, respectively, suggesting that most of the species lacked DNA components in their genomes which differed substantially from their main-band DNAs in base pair composition. The melting rate profiles revealed a prominent and distinct heavy or GC-rich DNA component in the genomes of three species belonging to the subgenus Cyprinella of Notropis. These and other data suggest that the heavy melting component may reflect a large, comparatively GC-rich family of highly repeated or satellite DNA sequences common to all three genomes.  相似文献

It has been hypothesized that the length of an exon tends to increase with the GC content because stop codons are AT-rich and should occur less frequently in GC-rich exons. This prediction assumes that mutation pressure plays a significant role in the occurrence and distribution of stop codons. However, the prediction is applicable not to all exons, but only to the last coding exon of a gene and to single-exon CDS sequences. We classified exons in multiexon genes in eight eukaryotic species into three groups-the first exon, the internal, and the last exon-and computed the Spearman correlation between the exon length and the percentage GC (%GC) for each of the three groups. In only five of the species studied is the correlation for the last coding exon greater than that for the first or internal exons. For the single-exon CDS sequences, the correlation between CDS length and %GC is mostly negative. Thus, eukaryotic genomes do not support the predicted relationship between exon length and %GC. In prokaryotic genomes, CDS length and %GC are positively correlated in each of the 68 completely sequenced prokaryotic genomes in GenBank with genomic GC contents varying from 25 to 68%, except for the wall-less Mycoplasma genitalium and the syphilis pathogen Treponema pallidum. Moreover, the average CDS length and the genomic GC content are also positively correlated. After correcting for genome size, the partial correlation between the average CDS length and the genomic GC content is 0.3217 ( p < 0.025).  相似文献

Base ratio of DNA from 21 bacteriophage of Pseudomonas was determined by chemical hydrolysis and paper chromatography. Obtained values of the GC pair content were compared with melting temperature of DNA in 0,1 X SSC. The content of GC pairs correlates with melting temperature by equation %GC = 2,53 (Tm - 53,4). The content of GC pair for DNA from 30 bacteriophages of Pseudomonas was calculated. Some speculations concerning the distribution in DNA base ratio of bacteriophages of Pseudomonas are discussed.  相似文献

Comparing variation across evolutionarily independent characters, notably nuclear and mitochondrial genes, yields a more robust estimate of diversification than is generally recovered from individual characters. Patterns of variation across multiple molecular markers from the mitochondrial ( 16SrRNA , cytochrome b ) and nuclear ( ldhA6 and aldB ) genomes were examined from six populations of Etheostoma collis and two populations of Etheostoma saludae , species aligned in the collis groups. Phylogenetic analyses revealed that sequence variation among individuals from the Roanoke, Tar and Neuse Rivers and the Catawba and Pee Dee Rivers, respectively, form highly supported, deeply divergent clades. Relationships of alleles sampled from Saluda River E. saludae and Cape Fear River E. collis to these lineages are unresolved, but all groups are reciprocally monophyletic for both nuclear and mtDNA loci. Phylogenetic analyses suggest that historical factors have had a strong influence on the distribution of genetic variation among populations. Genetic variation within the collis group is consistent with all previously proposed taxonomic hypotheses for the collis group, providing no taxonomic insights. From a conservation standpoint, each population of the collis group is an ESU, thereby warranting a drainage-specific management strategy.  相似文献

Hypotheses of relationship among genera of Percidae have been conflicting. Based on different phylogenetic premises, the evolution of small benthic forms in Percidae has been interpreted as resulting from either convergence or common ancestry. In order to assess various phylogenetic hypotheses of Percidae we collected complete sequences (1140 bp) of mitochondrially encoded cytochrome b for 21 species of percids. Seven species representing four additional families of Perciformes were used as outgroups. Maximum parsimony and minimum evolution analyses both recovered single shortest trees, and the results of these analyses were generally congruent with one another. All analyses consistently recovered three monophyletic groups in Percidae: Etheostomatinae (Ammocrypta, Crystallaria, Etheostoma, and Percina), Percinae (Perca and Gymnocephalus), and Luciopercinae (Stizostedion, Zingel, and Romanichthys). As a result of this analysis we present a revised classification of Percidae and discuss the phylogenetic evidence for the independent evolution of small benthic species within Etheostomatinae and Luciopercinae.  相似文献

Phenotypic characteristics of physiology and morphology of 71 strains belonging to the genus Dipodascus de Lagerheim were examined. The GC contents of genomic DNAs of 46 strains were calculated from the thermal denaturation curves using the spectrophotometric method. The first derivatives of the melting curves revealed that the DNAs of these strains are heterogeneous; four categories could be recognized. However, DNA similarity values calculated by using DNA-DNA reassociation kinetics showed that each category could be subdivided further. Two categories were separated into four subgroups each; the other two yielded five subgroups each. Strains belonging to the same subgroup exhibited high levels of DNA similarity ranging from 82 to 100%. The 18 subgroups represented 13 currently accepted Dipodascus species and five anamorphic Geotrichum species, four representing novel taxa. A phenotypic key to distinguish the taxa of Dipodascus, Galactomyces and Geotrichum is presented.  相似文献

MIDDLETON, D. J., A systematic survey of leaf and stem anatomical characters in the genus Gaultheria and related genera (Ericaceae). Leaf and stem anatomical characters of 123 species within the Gaultheria group of genera of the tribe Andromedeae are surveyed. A number of characters show considerable variation within a species which limits their taxonomic use. Other characters such as marginal sclerenchyma and, to a lesser extent, the hypodermis, the presence of adaxial stomata, the presence of free fibres and the pith type were of more taxonomic interest. These characters are discussed in relation to the generic and infrageneric classification of the group.  相似文献

Nucleosomal-length DNA was prepared from the genomic DNA of various prokaryotic and eukaryotic organisms by limited nuclease digestion after reconstitution with core histones. The DNAs ranged in base composition from 26.5% to 72% guanosine-plus-cytosine (%GC). The nucleosomal-length DNAs were then used in a competitive reconstitution assay in order to quantitatively determine their relative abilities to form nucleosomes. The results of the assay indicate a linear dependence of the free energy of nucleosome formation on base composition and, surprisingly, show that several prokaryotic DNAs form nucleosomes as well as or better than eukaryotic DNAs.  相似文献

本文对毛霉目中6科16属40个种共60株真菌DNA的G+C含量与分布作了系统的研究。在提取DNA时选用了Storck等人在Marmur提取细菌DNA方法基础上发展起来的真菌DNA提取法,经过一些修改后成功地提取了毛霉目真菌的DNA。经该法提取的DNA片段长、纯度高,在热变性时DNA的增色效应一般都大于35%。各个种的GC含量大致有一固定值。根据所测毛霉目16属各属真菌的平均GC含量,可将它们分为明显的三组:Gongronella, Haplosporangium, MortierellaSyncephalastrum为一组,GC含量最高为46.0-49.8%; Cunninghamella单独成一组,GC含量最低只有28.8%;其他各属包括Absidia, Mucor, Rhizopus等GC含量介乎这两组之间,分布于34.9-41.9%。这一次序除Helicostylum与Circinella的数值低于他人的报道外,其余和文献报道值一致。一般属内GC含量变化小于10%,种内变化小于2%。所测得的G+C mol%对分Syncephalastrum monosporum Zheng et al.和Mortierella ramanniana (Moeller) Linneman的合理归属提供了佐证。对某些所测结果与文献报道值有出入的原因作了讨论和分析。对采用Mandel等1970年建议的公式GC=(Tm 0.1×SSC/50.2)-0.990来计算GC含量的依据也作了讨论。  相似文献

The adjacent GN7-M-GN7 cross-linking and adjacent G-M-G sandwich-complex models for DNA metal ion binding were evaluated both with native DNAs differing in GC content as well as with the synthetic polymers poly [(dGdC)]2, poly[(dAdT)]2, and poly[(dAdC)(dGdT)]. The effect of Zn2+ was studied in depth, and limited studies were also performed with Co2+ and Mg2+. The results were compared to the extensive information available on Cu2+ binding to native DNAs and poly[(dAdT)]2. At high ratios of metal/base (R), Zn2+ caused all native DNAs to denature with the same melting temperature Tm, approximately 61 degrees C. A similar pattern was reported previously for Cu2+, but the typical Tm was approximately 35 degrees C. The extent of renaturation on cooling DNAs denatured in the presence of Zn2+ increased with GC content, as reported previously for Cu2+. These results, together with previously reported similarities, strongly indicate that the DNA binding characteristics of the two cations are similar. By comparison of the Tm values and hyperchromicity changes monitored at 260 and 282 nm, it is clear that, during thermal denaturation in the presence of Zn2+, both AT and GC regions were denatured, even at high R. The Tm vs R profile for the native DNAs was typical. The rise at low R and subsequent decrease at high R were inversely and directly related, respectively, to GC content. Except for poly[(dAdT)]2, where Tm increased with R, the other synthetic polymers exhibited the increase/decrease pattern. Poly[(dAdC)(dGdT)] gave a Tm value at high R of 54 degrees C. In the absence of Zn2+, this polymer exhibited little hypochromicity on cooling of denatured polymer. However, in the presence of Zn2+, nearly complete hypochromicity was observed, although the midpoint of the cooling curve was lower than the Tm value by approximately 15 degrees C at R = 10. These characteristics were similar to those with native DNAs, although viscosity and CD studies suggested that the "renatured" polymer was not identical to the unheated polymer. Furthermore, addition of Zn2+ after denaturation nearly completely reversed the absorption increase. This finding contrasts with those for native DNAs, where the Zn2+ must be present during denaturation in order to reverse the absorption increase nearly completely on cooling. With some caveats, poly[(dAdC)(dGdT)] appears to be a good model for native DNAs since its properties, including CD and uv changes on addition of Zn2+ to premelted and melted polymer, parallel those of the native polymers.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

Melting curves are calculated for infinitely long DNA-like random copolymers composed of AT and GC pairs of nucleotides. The entropy of random coil rings formed on melting is explicitly included through use of the Jacobson-Stockmayer ring-weighting factors. Transition curves are calculated for values of the cooperativity parameter σ in the range 10?2 ? σ ? 10?4. Ninety percent of the melting occurs in ca. 0.2°C for σ ? 10?3 regardless of the mole fraction of GC. We conclude that observed breadths of thermal denaturation curves for native DNAs result from a superposition of essentially all-or-none melting of various regions of the molecule. It is argued that refined approximations to the ring-weighting factors are probably not important when compared with the effects produced by long-range base sequence correlations which are known to occur in native DNA.  相似文献

Tandemly repeated DNAs, referred to as satellite DNAs, often occur in a genome in a genus-specific manner. However, the mechanisms for generation and evolution for these sequences are largely unknown because of the uncertain origins of the satellite DNAs. We found highly divergent genus-specific satellite DNAs that showed sequence similarity with genus-specific intergenic spacers (IGSs) in the family Solanaceae, which includes the genera Nicotiana, Solanum and Capsicum. The conserved position of the IGS between 25S and 18S rDNA facilitates comparison of IGS sequences across genera, even in the presence of very low sequence similarity. Sequence comparison of IGS may elucidate the procedure of the genesis of complex monomer units of the satellite DNAs. Within the IGS of Capsicum species, base substitutions and copy number variation of subrepeat monomers were causes of monomer divergence in IGS sequences. At the level of inter-generic IGS sequences of the family Solanaceae, however, genus-specific motif selection, motif shuffling between subrepeats and differential amplification among motifs were involved in formation of genus-specific IGS. Therefore, the genus-specific satellite DNAs in Solanaceae plants can be generated from differentially organized repeat monomers of the IGS rather than by accumulation of mutations from pre-existent satellite DNAs.  相似文献

The host range of ectomycorrhizal (ECM) fungi in Britain was examined by compilation of a data matrix from published literature sources, based primarily on accounts of sporocarp associations with particular host genera. Information was gathered for 577 species of ECM fungi belonging to 51 genera, and 25 genera of host trees, representing the majority of ECM fungal species and host genera recorded in Britain.
Pronounced variation was recorded in the number of ECM fungal species associated with different host genera, with over 200 species recorded with Betula , Fagus , Pinus and Quercus . There was a positive linear relationship ( r 2=0·47, P =0·007) between the number of species of ECM fungi associated with different host genera and the total area occupied by each tree genus in Britain (both values log-transformed). There was also variation in the number of species of ECM fungi which were apparently specific to particular host genera, values ranging from zero (in 15 genera) to >40 in the case of Betula and Fagus . In total, 233 fungal species appeared to be specific to a single host genus (i.e. 40% of those surveyed). Comparison of the ECM mycota associated with different host genera by PCA accounted for 17% of the total variation, with genera belonging to the Fagaceae ( Quercus , Fagus and Castanea ) tending to cluster together, indicating a degree of overlap in their ECM associates. Exotic conifer species, which displayed a lower ECM diversity than would be expected from their distributional areas, were characterized by a high degree of overlap with the ECM associates of Pinus and Betula .
These results indicate that the abundance of different genera of host trees and variation in host specificity could provide a basis for understanding patterns of diversity in ECM fungi within Britain.  相似文献

We have cloned and sequenced members of a family of satellite DNAs from three genera of the tilapiine tribe of fishes: Oreochromis, Sarotherodon, and Tilapia. The satellite DNAs, visualized as intensely staining bands following electrophoretic separation of EcoRI-digested genomic DNA, consist of three size variants differentially distributed in the various tilapiine species. The sizes of the monomers are approximately 237 bp (type I), 230 bp (type II), and 209 bp (type III). Several cloned monomers were sequenced from Oreochromis niloticus (type III), Oreochromis placidus (types I and II), Sarotherodon galilaeus (type I), Tilapia zillii (type I), and Tilapia rendalli (type I). Comparison of derived consensus sequences for the monomer units of the satellite DNAs revealed sequence identities within and between species that ranged from 89 to 96%. The type II and type III size variants appear to have arisen by deletions of 9 and 29 bp, respectively, within different regions of the type I satellite. Hybridization of a cloned monomer satellite from O. niloticus (type III) to PalI digests of genomic DNA from all three genera detected polymorphic, high molecular weight restriction fragments that produced fingerprint-like patterns. The complexity of these DNA fingerprints varied from one species to another, suggesting a markedly different genomic organization for these polymorphic satellite DNAs.  相似文献

In order to investigate a potential hybrid zone between the candy darter, Etheostoma osburni, and variegate darter, Etheostoma variatum, and examine population variation within E. osburni, a suite of primers for 15 polymorphic microsatellite loci were developed. The average number of alleles per locus was 5.5 in E. osburni and 7.6 in E. variatum, and the average observed heterozygosities were 62.5% and 71.4%, respectively. There were no deviations from Hardy-Weinberg equilibrium and no observed linkage disequilibrium after Bonferroni correction. The utility of these primers was also tested in 11 species of darters representing all four genera of darters. Success of cross-species amplification was largely consistent with phylogenetic relationships of darters.  相似文献

The DNAs of Cucurbita species were examined by several methods. All Cucurbita DNAs have a similar CsCl isopycnic banding pattern consisting of a major band at 1.695 g/cc and a well separated satellite band at 1.707 g/cc. Compared to other plant and animal genera, Cucurbita species have a large genomic proportion of rDNA; this value ranging from 1.4% to 3.1%. The genomic proportion of rDNA was found not to be useful as a characteristic indicating degree of relatedness of the various Cucurbita species. However, Cucurbita DNAs can be distinguished by the extent to which their repetitive sequences cross-hybridize to each other and an assessment of species relationships can be made on this basis.  相似文献

The rodent family Heteromyidae contains bipedal hoppers and quadrupedal runners. The possibility that bipedalism is associated with forelimb specialization for nonlocomotory functions, such as burrowing and seed-gathering, motivated a static functional-morphometric and interspecific allometric analysis of 18 metric characters of the forelimb skeleton. A principal-components analysis, across 28 species in six genera, showed that lengths of proximal (scapula, humerus) and distal (ulna, radius, metacarpal) elements were negatively allometric, and widths were positively allometric. Quadrupedal and bipedal species groups showed qualitatively similar allometric patterns, except that scapula width anterior to the spine was positively allometric in quadrupeds and negatively allometric in bipeds; scapula width posterior to the spine was positively allometric in bipeds and isometric in quadrupeds; and olecranon length was isometric in bipeds and positively allometric in quadrupeds. Most morphometric characters varied significantly among species within genera, even when effects of size variation were reduced by reconstructing all species to a common general size (as indicated by their score on the first principal component). These shape differences caused species to vary in the mechanical advantage of the forelimb, of possible importance for digging and seed-harvesting performance. Relative to quadrupeds, bipedal species tended to have greater mechanical advantage for proximal forelimb elements and smaller mechanical advantage for distal forelimb elements, but only the distal pattern remained in reconstructed forms, and no functional character was significantly different when tested over variation among genera nested within locomotion type. Cluster analysis confirmed that forelimb characters related to digging or seed-harvest are not coincident with mode of locomotion. Forelimb characters were, however, associated with digging or seed-harvest performance. Mechanical advantage of the proximal forelimb was positively related to an index of the compaction of soils with which 26 desert-dwelling species are associated, and also to relative use of heavy vs. light soils by nine species in the laboratory. Across 10 species, deviations in seed-harvest rate from expected allometric values were negatively correlated with mechanical advantage of the distal forelimb.  相似文献

为确定特化等级裂腹鱼类的遗传分化关系 ,对 7种 2 2个裂腹鱼个体进行随机扩增多态DNA(RAPD)研究。从所使用的 40个随机引物中 ,选择了 37个扩增带谱清晰的引物进行分析。根据构建的分子系统树显示 ,特化等级裂腹鱼类划分为 3个属级分类单元较为合适 ,这与采用形态学特征进行系统分析所得出的结论一致。在 2 2个个体之间遗传距离矩阵中 ,最大的遗传距离指数达到了 90 38% ,此外 ,还有较多的遗传距离指数也在 6 0 %~ 90 %之间。通过分析 ,认为用RAPD标记技术来分析属级或属级以上分类单元的亲缘关系 ,具有一定的适用性。对于一些遗传差异较大的类群 (如裂腹鱼类 ) ,应用属级或属级以上分类单元的亲缘关系 ,很可能会出现个体之间遗传距离接近或达到 10 0 %的情况 ,从而无法探讨其相互之间的亲缘关系。  相似文献
