1. An important goal of conservation biology is to preserve the evolutionary potential of a species by maintaining natural levels of genetic diversity. Here, we assess the population differentiation in the Atlantic salmon, Salmo salar, listed in Annex II of the European Habitats Directive, to provide valuable information for its conservation in Normandy (France).
2. Samples collected from 10 natural sites revealed that 13 of 14 microsatellite loci were polymorphic. Significant differentiation among populations was detected ( F ST = 0.054, P  <   0.001), and all F ST pairwise comparisons except one were significant. A genetic split was observed between populations inhabiting streams with limestone geology compared to those inhabiting streams with siliceous geology, which could reflect adaptative differences.
3. Hatchery stocks used for the restocking of two rivers were genetically distinct from native stocks.
4. Analysis of three stream habitats restored in 1995 showed that all were recolonized naturally by wild salmon from geographically close populations and no founder effects were detected. Allelic richness was similar between recolonized and wild populations.
5. From a management perspective, our study revealed that restoration of habitat is very effective to recreate new populations in rivers from which salmon have disappeared and that natural recolonization can be fast and effective in terms of genetic diversity.  相似文献

The genetic variation and clonal diversity of two divergent types (grey-green and yellow-green) of clonal populations of Leymus chinensis Tzvel at 14 loci were compared. Total gene diversity (HT) and the coefficient of genetic differentiation (GST) were all higher for the yellow-green type (HT = 0.270; GST =0.186) than for the grey-green type (HT = 0.250; GST = 0.157) of L. chinensis. Rare alleles usually occurred as heterozygotes rather than homozygotes and significant deviations from Hardy-Weinberg equilibrium were found only at a few loci. This indicated that these two types of populations were mainly out-crossing. Clonal diversity, evenness of clones, and mean clone size were not significantly different between the two types. We found that differences between the clone size and genetic variation of the yellow-green type of populations occurred with different climate and habitat population groups. However, for the grey-green type of populations, these genetic variations decreased under conditions of different climate and habitat population groups.  相似文献

Allozyme Diversity in Populations of Cymbidium goeringii (Orchidaceae)   总被引:1,自引:0,他引:1  
Abstract: Using 14 allozyme loci, we investigated levels of genetic diversity within populations, and degree of genetic divergence among 24 populations of Cymbidium goeringii (Orchidaceae) in Korea and Japan. Cymbidium goeringii maintains high levels of genetic diversity both at population (mean expected heterozygosity, H e = 0.238) and species levels (0.260). Means of H e found in 24 populations were not significantly different from each other. About 90 % of the total variation in the species is common to all populations (mean G ST = 0.108). No unique allele was found in any population. The indirect estimate of gene flow based on the mean G ST was high ( Nm = 2.06). Nei's genetic identities for pairs of populations had high values (mean = 0.974 [SD = 0.013]). The Mantel-Z test showed a significant correlation between genetic distance and geographic distance. However, the mean G ST value between 17 populations in Korea and seven Japanese populations was relatively low (0.029), even though the land connection between the southern Korean peninsula and southern Japanese archipelagos has not existed since the middle Pleistocene. Large numbers of small seeds of C. goeringii might travel long distances by wind from populations to populations both in Korea and Japan, increasing genetic diversity within populations and maintaining low genetic differentiation among populations.  相似文献

Populations of the guppy Poecilia reticulata from six locations in N. Trinidad were examined by starch gel electrophoresis to estimate their degree of genetic divergence. Variability at seven enzyme-coding loci demonstrated that populations differed markedly in allele frequencies with some allelic substitution between sites (for 23 loci, Nei's mean genetic identity, Ī ranged from 0.869–1.00; the coefficient of gene differentiation, G ST= 0.086; and the absolute differentiation between populations, m= 0.044). There was a good correspondence between degree of physical isolation and extent of genetic differentiation, as exemplified by the partitioning of gene diversity (Nei's gene diversity analysis = 66% between river basins; 32% within river basins; 2% within rivers), though there was considerable variability in the contribution of individual loci. Most populations were in Hardy-Weinberg equilibrium, and any significant deviations were due exclusively to heterozygote deficiencies. The patterns of population differentiation are discussed in relation to probable past geological and historical events, and present-day evolutionary forces. Notable among these are previous continental land connections, periodic immigration from N.E. South America, and post-colonization events including differential predation, sexual selection, apparent restricted vagility and small effective population sizes.  相似文献

1. The great silver water beetle Hydrophilus piceus is one of the largest aquatic insects in Europe. In Britain it is rare and endangered, and confined to a small number of low-lying marshes. Very little is known about the beetle populations in any of these areas, or the connectivity between them.
2. To investigate the population structure of H. piceus in Britain, four polymorphic microsatellite loci were identified and characterized. The genome of this beetle seems to have few microsatellites but contains a high proportion of a larger repeated sequence.
3. All six of the main British populations (Somerset, Lewes, Pevensey, Romney, North Kent and Norfolk) showed substantial genetic diversity at the microsatellite loci. However, estimates of effective population size at one site (Pevensey) were remarkably low, at <10 adults for the period 2004–05.
4. Most of the genetic diversity was partitioned within rather than among the populations, although there was, nevertheless, significant genetic sub-structuring. Almost all population pairwise F st estimates were significantly different from zero, and there was a clear isolation-by-distance effect. Assignment tests and cluster analyses demonstrated interpopulation relationships largely consistent with their geographical separations.
5. Hydrophilus disperses by flight, and records from moth traps indicated that there was no month in which the beetles never flew, but that flight activity was highest in the spring.
6. The genetic data highlight the need to maintain or regenerate habitat connectivity within flying distance for H. piceus , and to sustain large areas of suitable breeding marshes.  相似文献

1. The recent arrival and explosive spread of the zebra mussel, Dreissena polymorpha (Pallas), in Ireland provided a rare opportunity to study the population genetics of an invasive species.
2. Eight polymorphic allozyme loci ( ACO-1, ACO-2 , EST-D, GPI, IDH-2, MDH, OPDH and PGM ) were used to investigate genetic diversity and population structure in five Irish populations, and the results were compared with those from a previous microsatellite study on the same samples.
3. The mean number of alleles per locus (2.7 ± 0.1) was similar to the mean for the same loci in European populations, suggesting that Irish founder populations were large and/or multiple colonization events took place after foundation. A deficiency of heterozygotes was observed in all populations, but was uneven across loci.
4. Pairwise comparisons, using Fisher's exact tests and F ST values, revealed significant genetic differentiation among populations. The overall multilocus F ST estimate was 0.118 ± 0.045, which contrasted with an estimate of 0.015 ± 0.007 from five microsatellite loci on the same samples in a previous study.
5. Assuming that microsatellites can be used as a neutral baseline, the discordant results from allozymes and microsatellites suggest that selection may be acting on some allozyme loci, specifically ACO-1, ACO-2 , IDH-2 and MDH, which contributed most to the significant differentiation between samples.  相似文献

Aim  In order to look for a possible centre of survival for the Norway spruce ( Picea abies Karst.) in the south-western Alps, six natural populations of this area were investigated by means of genetic markers in order to assess the degree and the distribution of genetic diversity within the species.
Location  Western and South-western Alps.
Methods  Populations were genotyped using seven simple sequence repeat (SSR) markers. Basic population genetics parameters were estimated and the amount of genetic differentiation calculated.
Results  A large amount of variability was found (0.59 <  H e < 0.67); genetic differentiation as measured by F ST was 0.05, close to other similar studies; no isolation by distance was detected by a Mantel test. Analysis of molecular variance confirmed a high degree of variability within populations and a low degree of variability among populations. Finally, the number of populations from which those observed could have arisen was estimated by Bayesian analysis.
Main conclusions  The results presented here suggest that the present populations derive their genetic make-up from three inferred clusters. The possible existence in this area of a relict/refuge population during the last glaciation is discussed.  相似文献

Aim  To explore the genetic and phylogeographic structure of a temperate forest species, Pinus strobiformis Englem., in a subtropical region in the context of climate change during the Pleistocene. It is expected that the colder conditions during glacial stages favoured range expansions of P. strobiformis , thus promoting gene flow.
Location  Mexico and the United States.
Methods  Estimates of genetic diversity and structure were obtained using chloroplast microsatellite loci of 23 populations of P. strobiformis across its entire range, seven neighbouring populations of Pinus ayacahuite Ehrenb. ex. Schtdl, and one population of Pinus flexilis James.
Results  The genetic diversity of P. strobiformis ( H e = 0.856) was found to be high, especially in western populations, whereas eastern populations were less variable and more genetically similar to P. ayacahuite of central Mexico. We found evidence of significant phylogeographic structure ( N ST = 0.444; P  =   0.026), high genetic structure ( R ST = 0.270), and isolation by distance. Pairwise R ST and samova (spatial analysis of molecular variance) results indicated an east–west partition of genetic variation, with populations within each group showing little differentiation and no isolation by distance.
Main conclusions  The phylogeographic structure of P. strobiformis across the entire range was pronounced, with two main genetic and geographic groups separated by the Chihuahuan Desert. However, within each of the two groups there was little population differentiation and no isolation by distance, suggesting genetic connectivity as a result of population expansions within these areas during glacial stages.  相似文献

Lychnis alpina is a perennial caryophyllaceous plant with an amphi-Atlantic distribution. 27 populations are described on the basis of allozyme data from 11 electro-phoretic loci. L. alpina has a relatively low level of genetic variation, 3 of the loci are polymorphic ( P.,%= 20.2%) and the mean expected heterozygosity per indivindual is H3= 0.233 the three polymorphic loci included (H = 0.064, 11 loci included). Despite relatively low level of genetic variation, the genetic diversity among populations is relatively high with FST= 0.51. A hierarchial analysis of genetic diversity demonstrates that most of the genetic diversity exists as interpopulational, intra-regional diversity. Genetic identity is not associated with the intercontinental geographical distribution of the populations, neither is the diversity correspondant to taxonomic relationships.  相似文献

Relationships of genetic diversity at microsatellite loci and quantitative traits were examined in hatchery-produced populations of Japanese flounder using a relatively straightforward experiment. Five hatchery populations produced by wild-caught and domesticated broodstocks were used to examine the effects of different levels (one to three generations) of domestication on the genetic characteristics of hatchery populations. Allelic richness at seven microsatellite loci in all hatchery populations was lower than that in a wild population. Genetic variation measured by allelic richness and heterozygosity tended to decrease with an increase in generations of domestication. In addition, the degree of genetic differentiation from a wild population increased with an increase in generations of domestication. Significant differences in three morphometric traits (dorsal and anal fin ray counts and vertebral counts) and three physiological traits (high temperature, salinity and formalin tolerance) were observed among the hatchery populations. The degree of phenotypic difference among populations was larger in morphometric traits than in physiological traits. The divergence pattern of some quantitative traits was similar to that observed at microsatellite loci, suggesting that domestication causes the decrease of genetic variation and the increase of genetic differentiation for some quantitative traits concomitantly with those for microsatellite loci. Significant positive correlation was observed between F ST and the degree of phenotypic difference in the three morphometric traits and formalin tolerance, indicating that genetic variation at microsatellite loci predicts the degree of phenotypic divergence in some quantitative traits.  相似文献

Abstract Dioscorea tokoro Makino is a herbaceous climber species widespread in East Asia. Genetic structure of a natural population of D. tokoro was examined employing starch gel electrophoresis of allozymes. Genotypes of seven loci were studied for 1,128 individuals. Twenty-six populations located mainly in the Kinki district of Japan were subgrouped into four large clusters by the geographical distribution of alleles. The D. tokoro population was revealed to contain greater total genetic diversity ( HT =0.282) and higher intrapopulational genetic diversity ( HS =0.258) than other outcrossing species for which data are available. On the other hand, interpopulational differentiation ( GST =0.096) was smaller than in other outcrossers. For the heterozygosity deficiency observed ( FIT =0.125), population subdivision ( FST =0.096) and inbreeding within the population ( FIS =0.067) were revealed to contribute to the same extent. From these F -statistics, the migration rate among subpopulations and the rate of between-relative matings were estimated. Overall results on the genetic structure of the D. tokoro population indicated a high gene flow among its subpopulations, and this may be the consequence of its life form as a climber and its habitat in a disturbed environment. During the study, the geographical cline of Pgi allele frequencies was observed. This finding was supposed to be the result of the selection imposed on Pgi by the temperature differences between localities.  相似文献

Wild plant species develop their own way of living to adapt to the specific environment of their habitats. Their life-history traits strongly affect the genetic structure of the population. The wild species Oryza glumaepatula Steud. growing in the Amazon basin seems to have characteristic life-history traits suited for the flood condition. At the vegetative growth stage, the culms frequently break at internodes. With no roots anchoring on the ground, plant bodies floating in the water move downriver by water current and wind. To examine the association between the life-history traits and genetic population structure of Amazonian O. glumaepatula , we analysed allozyme variability at 29 loci of 16 enzymes using 37 populations from five regions. Allozymes were not so variable (total gene diversity H E = 0.044) compared with Asian wild rice, O. rufipogon Griff. The bottleneck effect and rare opportunity of interspecies gene flow may prevent the development of allozyme variability. Population genotypes tended to be differentiated among geographically isolated regions. Observed heterozygosities were much lower than expected heterozygosities, or gene diversity ( H O = 0.003 for whole population) and F IS over polymorphic loci was 0.931, indicating that O. glumaepatula has developed an inbreeding system. But, the intrapopulation gene diversity ( H S) was higher than interpopulation gene diversity ( D ST), as generally observed in outbreeding populations. The migration ability of O. glumaepatula makes long-distance seed dispersal possible. This might have led to frequent gene flow among populations.  相似文献

1. Previously, the Yangtze River connected thousands of shallow lakes which together formed a potamo-lacustrine system capable of sustaining a rich variety of submerged macrophytes.
2.  Potamogeton malaianus is one of the dominant submerged macrophytes in many lakes of this area. Genetic variation and population structure of P. malaianus populations from ten lakes in the potamo-lacustrine system were assessed using inter-simple sequence repeat markers.
3. Twelve primer combinations produced a total of 166 unambiguous bands of which 117 (70.5%) were polymorphic. Potamogeton malaianus exhibited a moderate level of population genetic diversity ( P P = 70.5%, H E = 0.163 and I =  0.255), as compared with that of plants in the same habitat and range. The main factors responsible for this moderate value were the plant's mixed breeding system (both sexual and asexual) and the hydrological connectivity among habitats.
4.  F statistics, calculated using different approaches, consistently revealed a moderate genetic differentiation among populations, contributing about 20% of total genetic diversity. An estimate of gene flow (using F ST) suggested that gene flow played a more important role than genetic drift in the current population genetic structure of P. malaianus ( Nm  = 1.131).
5. The genetic diversity of P. malaianus did not increase downstream. A high level of linkage–disequilibrium at the whole population level suggested that metapopulation processes may affect genetic structure. The migration pattern of P. malaianus was best explained by a two-dimensional stepping stone model, indicating that bird-mediated dispersal could greatly influence gene movements among lakes.  相似文献

Comparative analyses of the genetic differentiation in microsatellite markers ( F ST) and leaf morphology characters ( Q ST) of Amphicarpaea edgeworthii Benth. were conducted to gain insight into the roles of random processes and natural selection in the population divergence. Simple sequence repeat analyses on 498 individuals of 19 natural populations demonstrate that a significant genetic differentiation occurs among populations (mean F ST = 0.578), and A. edgeworthii is a highly self-fertilized species (mean selfing rate s  = 0.989). The distribution pattern of genetic diversity in this species shows that central populations possess high genetic diversity (e.g. population WL with H E = 0.673 and population JG with H E = 0.663), whereas peripheral ones have a low H E as in population JD (0.011). The morphological divergence of leaf shape was estimated by the elliptical Fourier analysis on the data from 11 natural and four common garden populations. Leaf morphology analyses indicate the morphological divergence does not show strong correlation with the genetic differentiation ( R  = 0.260, P  = 0.069). By comparing the 95% confidence interval of Q ST with that of F ST, Q ST values for five out of 12 quantitative traits are significantly higher than the average F ST value over eight microsatellite loci. The comparison of F ST and Q ST suggests that two kinds of traits can be driven by different evolutionary forces, and the population divergence in leaf morphology is shaped by local selections.  © 2009 The Linnean Society of London, Biological Journal of the Linnean Society , 2009, 96 , 505–516.  相似文献

Although it is widely recognised that spatial subdivision of populations is common in nature, there is no consensus as to how metapopulation dynamics affect genetic diversity. We investigated the genetic differentiation of natterjack toads, Bufo calamita , in three regions of Britain where habitat continuity indicated the likely occurrence of extensive metapopulations. Our intention was to determine whether genetic analysis supported the existence of metapopulation structures, if so of what type, and to identify barriers to migration between subpopulations. Allele frequencies were determined across eight polymorphic microsatellite loci for a total of 24 toad subpopulations at three separate sites. Genetic differentiation was assessed using five measures of genetic distance, notably F ST , R ST , Nei's standard distance D s , Δμ2 and the Cavalli-Sforza chord distance D c . B. calamita exhibited small but significant levels of genetic differentiation between subpopulations in all three study areas, and genetic and geographic distance correlations indicated isolation-by-distance effects in all three cases. The effects on correlation strengths of compensation for positive (sea, rivers, urban development) and negative (pond clusters) barriers to toad migration between the subpopulations in each area were also determined. D c , a measure which assumes that differentiation is caused by drift with negligible mutation effect, yielded the most plausible interpretation of metapopulation structures. Overall the patterns of genetic variation suggested the existence of a mixed metapopulation model for this species, with high levels of gene flow compatible with one version of the classical model but often supported by particularly stable subpopulations as in the mainland-island model.  相似文献

Regional differentiation of North American Atlantic salmon at allozyme loci   总被引:1,自引:0,他引:1  
Allozyme variation was characterised by starch gel electrophoresis at 23 enzyme coding loci and one regulatory locus in Atlantic salmon from 53 rivers in Eastern Canada, encompassing the majority of the species' North American range. Variation among rivers was highly heterogeneous and eight of the 15 polymorphisms showed regionally restricted distributions. Nearest neighbour joining (NJ) analysis and multi‐dimensional scaling suggest six distinct regional groups; Labrador/Ungava, Gulf of Saint Lawrence, Newfoundland (excluding Gulf rivers), the Atlantic shore/Southern Uplands of Nova Scotia, the inner Bay of Fundy, and the outer Bay of Fundy. Approximately 25% of observed genetic variation was distributed among these regions with a weak though significant overall correlation of genetic and geographic distance (Mantel Test, r  = 0·255, P  = 0·005). Collectively, the rivers showed consistent divergence from European populations with strong bootstrap support for the two clusters across loci in the NJ analysis. Mean heterozygosity was 0·061 for both continental groups, but the European population showed more than twice the variation among populations. F ST values were 0·076 and 0·176 for North America and Europe, respectively, with an overall F ST of 0·330.  相似文献

1. Genetic variation at five microsatellite loci was investigated in six sea trout ( Salmo trutta ) populations to describe their spatio-temporal genetic variation in north-western Spain. We observed significant genetic variation between river basins, and isolation by distance with restricted gene flow between neighbouring rivers, which suggests an important homing behaviour.
2. Despite these populations suffering a serious demographic decline during 1998, we did not detect any reduction in their genetic variation, suggesting a reasonably high effective population size and temporal stability.
3. Genetic differences among rivers should be taken into account in future management activities. Given the high genetic variability and the temporal stability observed, we believe that no supportive breeding programmes are presently needed in these populations.  相似文献

Patterns of genetic diversity were examined in five endemic subspecies of the Lobularia canariensis complex from six of the Canary Islands. The taxa are interfertile, insect-pollinated outbreeders with wind dispersal. Electrophoretic analysis revealed a high level of genetic polymorphism at ten loci coding soluble enzymes, with a mean of 2.38 alleles per locus, 73.7 % polymorphic loci, and a mean heterozygosity of 0.279. Excesses of homozygotes, indicating inbreeding, were observed in small populations. The average total diversity was high, F.,=0.518. Among-population diversity, FST=0.318, contributed more to the total diversity than within-population diversity, FIS=0.222. Little geographic or taxonomic patterning of the allozyme variation was observed. The mean genetic identitity for pairwise comparisons of the 19 populations was 0.76, with a range of 0.51–0.96 and c. 17 % of the comparisons below 0.67 and c. 8 % above 0.90. The results contrast with the many cases of high genetic identities reported for populations of endemic plants on oceanic islands. High levels of allozyme divergence suggest a relatively old origin of the L. canariensis complex and a long period of isolation of some of the populations.  相似文献

Populations of the guppy, Poecilia reficulata , in N. Trinidad exhibit marked population differentiation in allozyme frequencies. Here we investigate six further populations electrophoretically at 25 enzyme-coding loci to examine patterns in geographical structuring, genotypic distributions and genetic diversity. With one exception, possibly related to an experimental introduction, populations divided broadly (dendrogram of Nei's mean genetic identity, Ī) in accordance with proposed ancestral colonization. Most populations were in Hardy Weinberg equilibrium, though some significant deficits in heterozygotes were detected. Incorporating information from published data, markedly hjgher levels of genetic diversity (mean observed heterozygosity, o) were recorded in lowland [o= 0.0382 ± k0406 ( s.e .), n = 9] compared with upstream populations [o= 0.0112 ± 00034 (S.E.), n = 9]. Patterns are discussed in relation to historical and present-day evolutionary forces.  相似文献

1. Generalized additive models (GAMs) were used to predict macroinvertebrate taxonomic richness and individual taxon diversity at the reach level across seven European glacier-fed river sites from a set of 11 environmental variables. Maximum water temperature and channel stability were found to explain the most deviance in these models.
2. Using this information, and data from other recent studies of glacier-fed rivers, a modified conceptual model based on Milner & Petts (1994) is presented which predicts the occurrence of macroinvertebrate families and subfamilies as determined by maximum water temperature ( T max) and channel stability. This deterministic model only applies to the summer meltwater period when abiotic variables drive community structure.
3. Where maximum water temperature is below 2 °C, Diamesinae chironomids are typically the sole inhabitants, but where T max >2 °C but <4 °C Orthocladiinae are found and, where channels are more stable, Tipulidae and Oligochaeta also occur. Above 4 °C Perlodidae, Taeniopterygidae, Baetidae, Simuliidae and Empididae can be expected to be part of the glacier-fed river community, particularly in Europe.
4. At other times of the year when environmental conditions ameloriate, glacial rivers support higher macroinvertebrate abundance and diversity, with a number of taxa present that are not found during the summer melt period.
5. Dispersal constraints influence macroinvertebrate assemblages of many glacier-fed rivers located on islands and in some alpine areas.  相似文献
