Summary Ginkgo biloba L. is an important landscape tree, is resistant to insect, fungi and other pests, and produces a number of chemicals that have pharmaceutical properties (termed ginkgolides). Studies were initiated to establish an in vitro culture protocol for Ginkgo. Explants (intact embryos, embryos with cotyledons removed, and cotyledon tissue) were removed from disinfested seeds and cultured on Murashige and Skoog minimal organics medium with various combinations of either 2,4-dichlorophenoxyacetic acid (2,4-D) or naphthaleneacetic acid (NAA) and either kinetin or benzyladenine (BA). Cultures were incubated in the light and morphological development was recorded. Both embryo and cotyledon explants produced callus (cotyledon tissue produced the most callus). Ginkgolides A and B were detected in callus tissue extracts. Intact embryo cultures initiated on media with 2,4-D plus NAA for 5 wk produced shoots and roots when transferred to media with 4.5 μM 2,4-D alone for an additional 5 wk. Plants were transferred from the 2,4-D media to pots and maintained in the greenhouse.  相似文献

Seed germination and micropropagation protocols of the medicinal species Maytenus canariensis (Loes.) G. Kunkel & Sunding were optimized. In vitro seed germination occurred (86 to 94.7 %) only after treatment of the seeds with H2SO4, followed by surface sterilization and culture on solid nutrient medium without any growth regulators. Micropropagation failed when explants were taken from mature trees, and browning of the nutrient medium frequently occurred despite testing many growth media. Nonetheless, adventitious shoot regeneration was achieved employing axillary or apical buds taken from 2–2.5 months old plantlets obtained after in vitro germination of seeds, following culture on nutrient media supplemented with benzylaminopurine, kinetin and naphthalenacetic acid (NAA), attaining up to 3.9 shoots per explant, after 4–6 months. Root induction was best on a medium containing 4.0 mg dm−3 NAA, achieving a 100 % induction. After hardening of rooted plants, survival after transfer to soil was 71.43 %.  相似文献

A micropropagation protocol through multiple shoot formation was developed for Thlaspi caerulescens L., one of the most important heavy metals hyperaccumulator plants. In vitro seed-derived young seedlings were used for the initiation of multiple shoots on Murashige and Skoog (MS) medium with combinations of benzylaminopurine (BA; 0.5–1.0 mg dm−3), naphthaleneacetic acid (NAA; 0–0.2 mg dm−3), gibberellic acid (GA3; 0–1.0 mg dm−3) and riboflavin (0–3.0 mg dm−3). The maximum number of shoots was developed on medium containing 1.0 mg dm−3 BA and 0.2 mg dm−3 NAA. GA3 (0.5 mg dm−3) in combination with BA significantly increased shoot length. In view of shoot numbers, shoot length and further rooting rate, the best combination was 1.0 mg dm−3 BA + 0.5 mg dm−3 GA3 + 1.0 mg dm−3 riboflavin. Well-developed shoots (35–50 mm) were successfully rooted at approximately 95 % on MS medium containing 20 g dm−3 sucrose, 8 g dm−3 agar and 1.0 mg dm−3 indolebutyric acid. As an initial step in establishing interspecific hybridization to broaden the genetic basis of lentils [Lens culinaris ssp.culinaris (Medikus) Williams], a set of experiments was carried out to produce an efficient in vitro protocol for propagation of lentil and two of its wild relatives (Lens ervoides andLens culinaris ssp.orientalis). The objective of the experiments was to optimize the media (Murashige and Skoog) to regenerate shootsin vitro from nodal segments without a callogenic phase. The number of shoots per explant, the number of nodes per shoot and shoot length showed that species differences, gibberellic acid and benzyladenine levels had the largest effects, with only minor interaction effects. The experiments therefore identified a standard protocol which gave the optimum levels of growth regulators, Murashige and Skoog (MS) salts and sucrose concentrations for maximum plant regeneration from the nodal segment of these species. The medium recommended for optimal shoot regeneration without a callogenic stage contained 2.89 μM GA3 in combination with 1.11 μM BA in MS medium lacking sucrose. The optimal medium for root induction on these shoots had the MS medium supplemented with 5.37 μM NAA. Final successful establishment of regenerated plants was completed by the transfer to a third medium containing half-strength MS salts.  相似文献

The present study involves in vitro propagation of Iphigenia indica (Kunth.) through multiplication of whole corms and corm buds. The whole corms produced very small micro-corms, which developed plants individually whereas corm buds multiplied to produce numerous shoots at variable rates in presence of -naphthaleneacetic acid (NAA) and 6-benzylaminopurine (BAP). The best response in corm and bud multiplication was obtained in Murashige and Skoog's basal medium (MS) supplemented with 2.69 M NAA and 8.88 M BAP. The shoots regenerated were further cultured on MS medium containing NAA and indole-3-butyric acid (IBA) for initiation of roots. MS medium with 5.38 M NAA and 4.92 M IBA induced highest percentage of roots (81%) within 2 weeks in culture.  相似文献

The micropropagation of neem (Azadirachta indica) was accomplished by culture of buds from crown branches of a mature tree, basal-sprouts of another mature tree and a single juvenile plant. Cultures derived from these three different sources showed significant variation in in vitro response. In case of the crown and basal-sprout explants, addition of 12.5 μM PVP-40 in the establishment medium controlled leaching of phenol growth inhibitors. Phenolic leaching was not observed in juvenile explants. DKW medium (with 0.22 μM BA) was significantly better than MS for shoot proliferation. Shoot cultures of crown branch origin did not elongate and eventually died after the third subculture. In the presence of 4.9 μM IBA in half-strength DKW, 90% of the shoots and 100% of basal-sprout and seedling explant origin, formed roots. Plantlets from both explant types showed 90% survival after acclimatization.  相似文献

In vitro propagation of a semi-dwarfing cherry rootstock   总被引:2,自引:0,他引:2  
A successful in vitro propagation system for the semi-dwarfing cherry rootstock Maxma-14 (Prunus avium L.) has been developed. Shoot tips and axillary buds were successfully established in vitro. Multiplication rate of about 6 was achieved over a 4-week period using Murashige and Skoog medium with 4.44 μM benzyladenine and 0.49 μM indole-3-butyric acid (IBA). Rooting occurred within 4 weeks on liquid and agar-gelled media containing 0.49 μM NAA or 0.49, 2.45 μM IBA. On liquid media, a maximum rooting efficiency of up to 100% was obtained. However, high concentrations of auxins delayed the time of root initiation for 3–5 days. Acclimatization was affected directly by rooting conditions. Survival was best when plantlets were transferred to pots after a short period of root emergence on rooting media. Multiplication medium was also important for successful acclimatization. Shoots transferred to rooting media from that with kinetin resulted in better acclimatization and survival than that derived from media with benzyladenine. Further, plantlets rooted on liquid media had better survival than that rooted on agar-gelled media. The effects of different growth regulators on induction and growth of callus ofAsparagus densiflorus cv. Sprengeri were studied. Calluses grew more rapidly on Murashige and Skoog basal medium supplemented with 5.4 μM p-chlorophenoxyacetic acid (pCPA) and 4.4 μM 6-benzylaminopurine (BA) (medium 1) as compared to the same medium with 11.3 μM 2,4-dichlorophenoxyacetic acid (2,4-d) and 4.6 μM kinetin (medium 2). Calluses on medium 1 were soft and friable, whereas, compact, hard calluses originated on medium 2. Different concentrations and combinations of BA and/or kinetin were also used to study their effects on shoot regeneration. Kinetin was found to be less effective than BA in the initiation of shoots (1.8 shoots/callus). High numbers of shoots were produced in the presence of 0.4 μM BA alone (3.3 shoots/callus). The addition of ancymidol (5 μM) in MS with 0.4 μM BA enhanced multiplication of shoots (9.8 shoots/explant) and also produced well-developed crowns.  相似文献

Three cultivars (cvs.) of Gladiolus hybridus Hort., namely ‘Her Majesty’, ‘Aldebaran’ and ‘Bright Eye’ were successfully micropropagated. The cultures were established using intact cormels or segments of cormels and inflorescence axes on Murashige and Skoog (1962; MS) medium. The response depended on media supplements; both callus formation or direct induction of shoot buds was observed. Shoot differentiation from callus could be obtained on MS medium containing 1.0 μM BA (6–benzyladenine) and 10.0 μM NAA (α-naphthalene acetic acid) in all three cultivars. The same could be achieved by giving a heat shock (HS; 50 °C, 1h) to callus cultures (in case of ‘Her Majesty’ and ‘Aldebaran’ only) maintained on the basal medium. In these two cultivars, high sucrose concentration (0.232, 0.290 or 0.348 M) also favoured growth and proliferation of shoot cultures on a plant growth regulator-free medium at 20 °C in comparison to the cultures kept at 25 °C. On the other hand, shoot cultures maintained on the basal medium at 25 °C containing normal (0.058 M, i.e., 2.0%, w/v) sucrose concentration responded similar to those maintained at 20 °C on a high sucrose medium; reduced response was observed on normal sucrose containing medium at 20 °C. Heat shock enhanced shoot proliferation in the cultures maintained on basal medium, but induced prolific rooting in shoot cultures, within 5 days of HS, on high sucrose (optimum 0.232 M) medium. While the number of roots increased at higher sucrose concentrations in the medium in case of cvs. ‘Her Majesty’ and ‘Aldebaran’, the same was found to be independent of sucrose concentration in cv. ‘Bright Eye’. Generally the rooted plants produced on high sucrose (0.232 M) medium in comparison to medium with normal sucrose concentration showed better survival (ca. 90% as against 40%) in the soil. Most climacteric and postmenopausal women appear to have vasomotor symptoms as well as a high risk of osteoporosis and cardiovascular disease. Although exogenous estrogens can reduce these symptoms, women are reluctant to use hormone replacement therapy (HRT) due to its undesirable side effects, such as irregular bleeding and an increased risk of breast cancer. A previous study suggested that Ginkgo biloba extracts (GBE) have estrogenic activity and might be suitable as an alternative to HRT. However, there are no reports of the preventive effect of GBE on breast cancer, which is the side effect of classical HRT. In this study, it was confirmed that GBE exhibits estrogenic and antiestrogenic activity depending on the E2 and GBE concentration, via estrogen receptor (ER)-dependent and ER-independent pathways. In addition, GBE reduced the E2 levels by stimulating the E2 metabolism and inhibiting E2 synthesis, which indicates that GBE can induce antiestrogenic activity via the depletion of E2. Furthermore, GBE might have similar action to selective arylhydrocarbon receptor modulators (SAhRMs), which induce antiestrogenic activity through cross-talk between the arylhydrocarbon receptor (AhR) and ER. In conclusion, GBE has a biphasic effect on estrogen, and can be considered as a potential alternative to HRT with chemopreventive effects on breast cancer. This experiment is designed to determine the most suitable conditions and media for propagating three selected fig (Ficus carica L.) clones through tissue culture. The clone 37 displayed a higher performance than clones 50 and 82. As the multiplication medium, the Murashige and Skoog (MS) medium containing 1 mg dm−3 α-indole-3-butyric acid (IBA), 1 mg dm−3 gibberellic acid and 5 mg dm−3 6-benzyladenine were the best, whereas, MS medium complemented with 1.2 and 2.5 μM IBA or 1-naphthalene acetic acid (NAA) were better in respect to rooting. Peat followed by volcanic tuff gave the best performance for acclimatization to outdoor conditions.  相似文献

Immature zygotic embryos were cultured on Murashige and Skoog's medium (MS) supplemented with various combinations of 2,4-dichlorophenoxyacetic acid (2,4-D), naphthaleneacetic acid (NAA), benzyladenine (BA) and zeatin or with various concentrations of 2,4-D alone. The maximum number (8 per embryo) of adventitious buds formed from cotyledons of heart stage embryos cultured on MS medium with 1 mg dm−3 BA and 0.01 mg dm−3 NAA. The adventitious buds originated from procambial strands of immature embryo cotyledons and then developed into adventitious bud primordia within 20 d. Adventitious buds transferred to hormone free MS medium grew into shoots, but did not produce plantlets because the shoots failed to root. An improved protocol for generation of viable cormlets from tissue culture derived shoots of saffron has been developed. Multiple shoots were generated from apical buds, small corms and in vitro developed single shoots. Bunches of two to three shoots when cultured on half strength Murashige and Skoog (MS) medium containing 3 mg dm−3 benzyladenine (BA) and 80 g dm−3 sucrose developed 1.89 cormlets per shoot bunch with an average fresh mass of 1.18 g. It took nine months from culture of apical buds to the harvest of cormlets but under field conditions 22 months. Sucrose appeared to be essential for cormlet induction as no cormlets were developed in the medium devoid of sucrose and only 0.29 per shoot in medium containing mannitol. In vitro derived cormlets sprouted from apical and axillary buds on MS medium containing 12 mg dm−3 BA, 3 mg dm−3 indolebutyric acid and 30 g dm−3 sucrose. Daughter cormlet formation from in vitro derived cormlets was also observed.  相似文献

The purpose of this study was to establish conditions for micropropagation of cloudberry (Rubus chamaemorus L.). Cultures were initiated from meristem cultures. When cultures were subcultured from clusters of 3–5 shoots, approximately 70 and 50 shoots were produced per cluster within 6 weeks at 8.9 μM BAP for the female cv. Fjellgull and the male cv. Apollen, respectively. Addition of 5.5 μM GA3 reduced the number of shoots. Auxins (IBA, NAA) promoted root development in vitro, but inhibited formation of new shoots. However, as much as 85% of shoots rooted without auxin treatment when planted in a peat:sand (80:20 v/v) mixture. Some of the male plants regenerated from shoot tip cultures flowered in the greenhouse within a year after transfer to soil.  相似文献

In vitro somatic embryogenesis and subsequent plant regeneration was achieved in callus cultures derived from immature zygotic embryos of Acacia arabica on semi-solid Murashige and Skoog (MS) basal salts and vitamins supplemented with 8.88 MBA, 6.78 M2,4-D and 30 g l–1 (w/v) sucrose. Somatic embryos proliferated rapidly by secondary somatic embryogenesis after transfer to MS medium supplemented with 6.66 M BA, 6.78 M 2,4-D. The maximum number of somatic embryos per callus was 72.6 after 8 weeks of culture on medium containing 6.66 M BA and 6.78 M 2,4-D. The isolated somatic embryos germinated on half-strength basal MS salts and vitamins supplemented with 0.04 M BA, 0.94 M ABA and 2% (w/v) sucrose. The embryo-derived plantlets were acclimatized in the greenhouse and subsequently showed normal growth.  相似文献

研究银杏叶提取物水溶液中多组分分子间的相互作用;采用动态光散射法和透射电镜扫描法,探讨银杏叶提取物水溶液是否存在分子聚集形态、分子聚集体粒径大小以及体外模拟不同胃肠pH环境条件下,银杏叶提取物水溶液分子聚集体的稳定性.实验数据证实银杏叶提取物水溶液存在纳米级分子聚集体,分子聚集体粒径在60 nm至100 nm之间;水溶液中1 nm以下的粒子大都是一些成分以单分子形式存在;随着溶液的浓度增加,分子聚集体的粒径也增大;在不同pH条件胃肠环境下,银杏叶提取物水溶液分子聚集体可以稳定存在.  相似文献

Ceropegiaevansii McCann (family: Asclepiadaceae), a critically endangered plant of Western Ghats has acquired significant importance due to its medicinal implications, edible tubers, and ornamental flowers. This study deals with the optimization of axillary bud proliferation using nodal explants followed by genetic stability analysis of regenerants. Maximum number of shoots (11.6 ± 1.1) was observed on the Murashige and Skoog (MS) medium supplemented with 6-benzylaminopurine (4.0 mg/l) and indole-3-acetic acid (0.3 mg/l) with 85% shoot multiplication frequency. In vitro-grown shoots were rooted best in 1/2 MS medium supplemented with indole-3-butyric acid (1.0 mg/l) with an average of 10.3 ± 0.9 roots per shoot and 92% rooting frequency. Plantlets were acclimatized best (90%) in a mixture of sterile soil, sand, and coco peat (1:2:1). Micropropagated plants were subjected to random amplified polymorphic DNA and inter simple sequence repeat markers analyses. Collectively, 759 bands were generated which were monomorphic and similar to the mother plant. Findings of this study are the first report on micropropagation and assessment of genetic stability of micropropagated plantlets in C. evansii which suggests that axillary shoot proliferation can safely be used as an effective tool for propagation and conservation of C. evansii.  相似文献

探讨利用银杏 (GinkgobilobaL .)细胞固定化培养方法 ,大规模生产药用成分银杏内酯类化合物的可能性。考察了植物生长调节剂对银杏固定化培养细胞生长及发酵液中银杏内酯类成分产生的影响。结果显示 ,最优培养基生长调节剂配比为 2 ,4 D 8 0mg L KT 0 0 4mg L NAA 0 4mg L。优化培养后 ,固定化细胞最高生物量出现在第 1 2d ,为 0 4gDW 瓶。发酵液中银杏内酯含量 :GKA在 2 2d时最高 (2 0 0 μg L) ,GKB在 1 8d时最高 (1 2 2 μg L) ,GKC在 1 8d时最高 (5 0 9μg L)。  相似文献

比较不同溶剂提取银杏(GinkgobilobaL.)叶黄酮类化合物的提取效率,从成本效益角度考虑,以70%乙醇作为提取溶剂更为有利。在分级沉淀中,黄酮含量与蛋白质含量呈正相关,在乙醇提取液中黄酮和蛋白质含量最高;蛋白质的存在有助于提高黄酮的溶解度。乙醇提取液用饱和(NH4)2SO4浓缩两次,可使醇相中的黄酮沉淀析出。根据试验结果,提出了银杏叶黄酮的优化提取方法。  相似文献
