Fusarium wilt is an increasingly serious disease of watermelon that reduces crop productivity. Changes in microorganism populations and bacterial and fungal community structures in rhizosphere soil of watermelon cultivars resistant or susceptible to Fusarium oxysporum f. sp. niveum were investigated using a plate culture method and PCR-DGGE analysis. Plate culture showed that populations of culturable bacteria and actinomycetes were more abundant in the rhizosphere of the resistant watermelon cultivar than the susceptible cultivar, but the fungi population had the opposite pattern. Populations of Penicillium , Fusarium , and Aspergillus were significantly lower in the resistant cultivar than the susceptible cultivar at the fruiting and uprooting stages (p?< 0.05). Pattern matching analysis generated the dendrogram of the DGGE results indicating the relatedness of the different resistant watermelon cultivars and their corresponding rhizosphere microbial communities. Further sequencing analysis of specific bands from DGGE profiles indicated that different groups of bacteria and fungi occurred in the rhizosphere of different watermelon cultivars. Our results demonstrated that plant genotype had a significant impact on soil microbial community structure, and the differences in the rhizosphere microbial community may contribute to the differences in resistance to F. oxysporum f. sp. niveum.  相似文献

为研究青枯病易感和钝感桑树品种植株根际土壤真菌群落组成,该研究以ITS1F和ITS2R为引物,基于高通量测序技术对桑树青枯病易感品种(台湾长果桑,SM)和桑树青枯病钝感品种(桂桑12号,IM)植株根际土壤真菌群落结构进行分析。结果表明:(1)两个品种间指示真菌丰富度的ACE、Chao1指数及表征多样性的Shannon指数无显著差异,门分类水平,被孢霉门(Mortierellomycota)和球囊菌门(Glomeromycota)是青枯病钝感桑树品种植株根际土壤中特有的优势真菌门;而属分类水平,Apiotrichum、地丝菌属(Geotrichum)、足放线病菌属(Scedosporium)和腐质霉属(Humicola)等是青枯病易感桑树品种植株根际土壤中富集的特有优势真菌属。(2)青枯病易感桑树品种植株根际土壤中,缺失了被孢霉门、球囊菌门真菌,以及被孢霉属(Mortierella)、镰刀菌属(Fusarium)、曲霉菌属(Aspergillus)和毛壳菌属(Chaetomium)等具有生防功能的优势真菌门属,可能是其易感青枯病的重要原因。(3)根据真菌群落对同类环境资源的利用途径进行功...  相似文献

Action of some fungal antagonists in the rhizosphere of resistant and susceptible tomato plants in the greenhouse The quantitative presence of the rhizospheric mycoflora fungi: Aspergillus alutaceus, Paecilomyces lilacinus, Penicillium herquei, P. nigricans and Trichoderma viride known for its antagonistic action to many pathogens is essentially differentiated in the two cultivars – the sensitive ‘Early pack’ and the resistant to tracheomycosis ‘GC 204′. It seems that the presence of the cultivar ‘GC 204’ favours the growth of these fungi. The first establishment of the fungi Penicillium chrysogenum and P. funiculosum, which are isolated from both cultivars, in the ‘Early pack’ rhizosphere might facilitate the colonization of this zone by the above mentioned antagonist which is not favoured by the sensitive cultivar. Such a changein the rhizospheric mycoflora of the sensitive cultivar may introduce some resistant to soil borne fungi diseases and open new perspectives of the biological control of these diseases.  相似文献

Summary The influence of different levels of nitrogen on the incidence of downy mildew disease of pearl millet on different varieties was studied under field conditions. Nitrogen nutrition to the host did not have much influence on the disease incidence. Among the three hybrid varieties of pearl millet tested J 1270 exhibited resistance to downy mildew incidence while J 934 and HB3 were found to be highly susceptible.  相似文献

The phyllosphere is colonized by complex microbial communities, which are adapted to the harsh habitat. Although the role and ecology of nonpathogenic microorganisms of the phyllosphere are only partially understood, leaf microbiota could have a beneficial role in plant growth and health. Pesticides and biocontrol agents are frequently applied to grapevines, but the impact on nontarget microorganisms of the phyllosphere has been marginally considered. In this study, we investigated the effect of a chemical fungicide (penconazole) and a biological control agent (Lysobacter capsici AZ78) on the leaf microbiota of the grapevine at three locations. Amplicons of the 16S rRNA gene and of the internal transcribed spacer were sequenced for bacterial and fungal identification, respectively. Pyrosequencing analysis revealed that the richness and diversity of bacterial and fungal populations were only minimally affected by the chemical and biological treatments tested, and they mainly differed according to grapevine locations. Indigenous microbial communities of the phyllosphere are adapted to environmental and biotic factors in the areas where the grapevines are grown, and they are resilient to the treatments tested. The biocontrol properties of phyllosphere communities against downy mildew differed among grapevine locations and were not affected by treatments, suggesting that biocontrol communities could be improved with agronomic practices to enrich beneficial populations in vineyards.  相似文献

 在葡萄抗病育种中 ,幼苗期排除感霜霉病的后代具有特别重要的意义 .用 BSA,RAPD和SCAR方法研究了葡萄感霜霉病基因的分子标记 .分析了两个种间杂交组合 [毛葡萄 (抗病 )×欧洲葡萄 (感病 ) ]88- 1 1 0和 88- 84与 88- 1 1 0的 F1代自交或互交所得的 3个 F2 代 ,以及欧洲葡萄品种和中国野生葡萄种 .共筛选了 2 80个随机引物 .引物 OPO1 0产生了一个 RAPD标记 OPO1 0 - 80 0与葡萄感霜霉病主效基因紧密联锁 .将该 DNA片段克隆并测序 .OPO1 0 - 80 0的实际长度为 835bp,所以 OPO1 0 - 80 0应为 OPO1 0 - 835.据其两端序列 ,设计了一对长度为 2 6bp和 2 8bp的特异引物分别扩增上述试材 ,获得了与该 RAPD标记相同大小的一条带 ,将 RAPD标记转化为 SCAR标记SCO1 0 - 835.并证实了此 SCAR标记的通用性 ,该 SCAR标记可用于葡萄抗病育种中杂种后代对霜霉病的抗病与感病性鉴定 .  相似文献

52 entries including landraces, old cultivars and wild accessions of B. oleracea and closely related Brassica species were screened for resistance against downy mildew and clubroot. Several accessions resistant to downy mildew and a few to clubroot were found. Genetic inheritance of the resistance in downy mildew was investigated by screening F1 and BC1F1 offspring from three resistant landrace accessions crossed with both a resistant and a susceptible father. The seedling resistance against downy mildew was found to be inherited recessively. This is a bit surprising as earlier papers mostly report of inheritance controlled by a single dominant gene. Previous screenings of B. oleracea resistance against downy mildew at the cotyledon stage have been done with P. parasitica isolated from B. oleracea as the original host plant. The recessive nature of the cotyledon resistance found in this screening might be due to the fact that the P. parasitica isolate was collected from B. napus fields. The clubroot seedling resistance was found to be controlled by recessive inheritance after screening the F1 offspring, this in agreement with earlier results/reports.  相似文献

The barley net blotch agent Pyrenophora teres (Died) Drechs. is one of the dominant fungal pathogens in agricultural crops worldwide. Here we aim to study the aggressiveness and virulence of P. teres populations collected at different ontogenesis stages (BBCH 30 and BBCH 47) from winter barley cultivars of various resistance types: moderately resistant, moderately susceptible and highly susceptible. We observed a direct proportional relationship between cultivar resistance and the aggressiveness of P. teres populations collected in both growth phases of the host plant. The isolates collected at an early stage of host plant development have a large difference in aggressiveness criteria: colony growth rate, sporulation intensity, latency period, plant damage degree, and the number of identified races. At the BBCH 30 growth stage, the growth rate of fungus colonies selected from a resistant cultivar is 1.2 times higher than that of a susceptible cultivar. The growth rate of colonies selected from resistant and susceptible cultivars in the earlier BBCH 30 stage is 1.04 times higher than the growth rate of colonies selected from the later phase. The sporulation intensity of fungal populations selected from a resistant cultivar is higher than that of populations selected from a susceptible cultivar (for BBCH 30–5.4 times, for BBCH 47–4.0 times); and it is 1.3 times higher in an earlier phase of plant development. Correlation between colony growth rate and spore formation rate in the BBCH 30 is r = 0.4. A high correlation level (r = 0.9) and notable difference between the variants were revealed when studying the duration of the latent period. The average value of plant damage by the P. teres from resistant cultivar is 4 times higher than from the susceptible cultivar in the BBCH 30 stage; and 12 times – in the BBCH 47 stage. There is a moderate negative correlation between the plant damage degree and the number of races identified from the fungal population, r = ?0.59 for the BBCH 30, r = ?0.8 for the BBCH 47. The number of races identified from P. teres populations collected in the late phase of plant growth was one third less. Our study helped to acquire new knowledge about intrapopulation processes under the influence of various factors – plant growth stage and cultivar genotype. The results obtained are the basis for the development of adaptive-integrated techniques for managing populations of the hemibiotrophic pathogen, barley net blotch.  相似文献

本文通过传统微生物培养方法,研究了在土培和基质培条件下,抗感枯萎病西瓜不同生育期根际和非根际微生物数量的变化.结果表明,微生物数量在根际显著高于在非根际微生物,且随西瓜生长发育的阶段不同而变化,苗期根际微生物数量最少,以后随西瓜生长发育,根际微生物数量不断增加,至生长旺盛的开花结果期,微生物数量达到最高,在西瓜生长发育后期,根际微生物数量又有所回落.西瓜抗枯萎病性与根际细菌的数量具有相关性,在生长发育各个阶段,无论是土培还是基质培,均表现为抗病材料的根际细菌数量高于感病材料的根际细菌数量.根际真菌与放线菌数量与西瓜的抗感枯萎病性没有相关性.非根际微生物数量在整个生育期变化辐度较小.非根际细菌数量在土培条件下几乎保持在同一水平,在基质培条件下迅速增加,至生长后期有所回落.非根际真菌与放线菌数量在土培和基质培条件下均于生长后期达到最高.  相似文献

The aims of the present work were (1) to determine the influence of plant growth stages on the population size of culturable bacteria and fungi associated with rhizoplane and endo-rhizosphere of maize grown in field and (2) to establish the community structure of total culturable bacteria and fungi. Density, diversity and community structure of culturable rhizoplane and endo-rhizosphere populations at different maize plant growth stages were estimated. Plant development did not have influence on total culturable microflora density but it selectively influenced some bacterial and fungal groups present in the rhizosphere. However, the microbial community structure changed markedly over time. This knowledge is relevant for exploring endophytic rhizobacteria potential in the promotion of plant-growth, protection against pathogens and to detect perturbations in natural agro ecosystems.  相似文献

Plant Growth‐promoting Fungus (PGPF) Penicillium oxalicum was isolated from rhizosphere soil of pearl millet and was tested for its ability to promote growth and induce systemic resistance in pearl millet against downy mildew disease. The fungal isolate P. oxalicum UOM PGPF 16 was identified as P. oxalicum using ITS sequencing and morphological analysis and sequence was deposited at NCBI with accession number KF150220. Pearl millet susceptible seeds were treated with three different inducers (CS, CF and LCF) of PGPF P. oxalicum and all the inducers significantly reduced the downy mildew disease and enhanced plant growth. Among the inducers tested, CS treatment recorded highest seed germination of 91% and 1427 seedling vigour followed by LCF and CF treatments. The vegetative growth parameter and NPK uptake studies under greenhouse conditions revealed that the CS treatment of P. oxalicum remarkably enhanced the parameters tested when compared to control plants. A significant disease protection of 62% and 58% against downy mildew disease was observed in plants pretreated with CS of P. oxalicum under greenhouse and field conditions, respectively. The spatio‐temporal studies revealed that inducers P. oxalicum required a minimum of 3 days for developing maximum disease resistance which was maintained thereafter. The maximum Peroxidase (POX) activity (62.7 U) was observed at 24 h in seedlings treated with CS of PGPF P. oxalicum and the activity gradually reduced at later time points after pathogen inoculation. Chitinase (CHT) activity was significantly higher in inducer treated seedlings when compared to control seedlings inoculated with pathogen after 48 h and remained constant at all time points.  相似文献

【目的】了解不同品种酿酒葡萄根围、叶围细菌群落结构。【方法】以不同品种酿酒葡萄根围、叶围样品为分析材料,采用末端限制性片段长度多态性(T-RFLP)技术研究根围、叶围细菌群落结构,并对细菌群落多样性进行分析。【结果】不同品种葡萄根围微生物优势菌群为变形菌门,不同品种葡萄叶围微生物优势菌群为放线菌目。根围Simpson指数大小顺序为白福尔琼瑶浆白诗南赛美蓉白玉霓米勒长相思,Shannon指数与Simpson指数基本保持一致。叶围Simpson指数最大的为白诗南,最小的为米勒,与Shannon指数基本吻合。【结论】葡萄细菌群落结构综合多样性指数和品种以及定殖部位密切相关。  相似文献

During the pathogenesis of pearl millet by the downy mildew (Sclerospora graminicola [Sacc.] Schroet.), the activities of peroxidase (PO) and indoleacetic acid oxidase (IAAO) and their isozyme pattern determined by isoelectric focusing on polyacrylamide gels, were observed in extracts of leaves and ears at different stages of development. The PO activity in extracts from infected plants of a susceptible cultivar was found to be higher than in healthy plants. The higher activity was most probably due to the acceleration of host senescence by the pathogen. Quantitative differences in the isozyme patterns of PO and IAAO were found. In inoculated plants of the resistant cultivar, no symptoms developed under the conditions used for infection of the susceptible cultivar and the changes in enzyme activities after inoculation were not significant. The results indicated that different proteins are synthesized in the two cultivars.  相似文献

【背景】三七的连作障碍与微生物区系失衡有一定关联,了解种植年限对三七微生物区系的影响,判断根际微生物区系是否失衡,对三七连作障碍的防治具有重要意义。【目的】探究文山不同种植年限健康三七根际土壤细菌和真菌群落结构、多样性差异,以及与种植年限之间的关联,为后续缓解三七连作障碍提供理论依据。【方法】采集一年生、二年生和三年生健康三七根际土壤,采用16S rRNA基因和ITS序列进行高通量测序。对测序结果进行分析,比较一年生、二年生和三年生健康三七根际土壤细菌和真菌的多样性和群落分布规律及与种植年限的关联。【结果】一年生健康三七根际细菌和真菌群落的α指数均显著高于二年生和三年生。通过β多样性分析发现,一年生健康三七根际土壤中真菌群落结构与其他年限的真菌群落结构有较大差异。相关性分析结果表明,真菌的均匀度和香农指数与种植年限呈现显著的负相关性,其余指数不显著相关。三七根际丰富度前三的门和属,除Acidobacteria和Aquicella丰富度随着三七种植年限的增加而显著增加外,其余群落丰度都与种植年限呈负相关。细菌中Bacteroidetes、Proteobacteria和真菌中Ascomycota、Basidiomycota、Chytridiomycota的丰富度随着种植年限增加而降低。在属水平上,细菌中Opitutus和真菌中Mortierella、Clitopilus、Pholiota的丰富度随着三七种植年限的增加而降低。在二年生三七根际土壤中病原真菌Alternaria、Cylindrocarpon、Fusarium和Pestalotiopsis的丰富度低于一年生和三年生,而有益真菌Mucor和Bacillus的丰富度却呈相反趋势。同时,有益细菌Flavobacterium和有益真菌Myxocephala、Aspergillus的丰富度随着种植年限的增加呈下降趋势。【结论】三七种植年限的增加,会引起三七根际微生物区系失衡,增加根际病原真菌属的丰度,同时降低一些有益真菌属的丰度。  相似文献

Downy mildew is one of the most destructive diseases of maize in subtropical and tropical regions in Asia. As a prerequisite for improving downy mildew resistance in maize, we analyzed quantitative trait loci (QTLs) involved in resistance to the important downy mildew pathogens--Peronosclerospora sorghi (sorghum downy mildew) and P. heteropogoni (Rajasthan downy mildew) in India, P. maydis (Java downy mildew) in Indonesia, P. zeae in Thailand and P. philippinensis in the Philippines--using a recombinant inbred line population derived from a cross between Ki3 (downy mildew resistant) and CML139 (susceptible). Resistance was evaluated as percentage disease incidence in replicated field trials at five downy mildew 'hotspots' in the four countries. Heritability estimates of individual environments ranged from 0.58 to 0.75 with an across environment heritability of 0.50. Composite interval mapping was applied for QTL detection using a previously constructed restriction fragment length polymorphism linkage map. The investigation resulted in the identification of six genomic regions on chromosomes 1, 2, 6, 7 and 10 involved in the resistance to the downy mildews under study, explaining, in total, 26-57% of the phenotypic variance for disease response. Most QTL alleles conferring resistance to the downy mildews were from Ki3. All QTLs showed significant QTL x environment interactions, suggesting that the expression of the QTL may be environment-dependent. A strong QTL on chromosome 6 was stable across environments, significantly affecting disease resistance at the five locations in four Asian countries. Simple-sequence repeat markers tightly linked to this QTL were identified for potential use in marker-assisted selection.  相似文献

【目的】揭示葡萄生单轴霉(Plasmoparaviticola)菌围可培养细菌和真菌的多样性特征,筛选对葡萄霜霉病有较强稳定防治效果的生防菌。【方法】连续两年从我国南北方具有代表性的7个葡萄产区采集葡萄霜霉病叶,镊子夹取经保湿培养获得的新鲜霉层并配制孢子囊悬浮液,采用传统分离培养法,结合形态分类、BOX-PCR指纹图谱分析以及分子鉴定结果,对葡萄生单轴霉菌围的可培养细菌和真菌进行聚类分析;采用菌株及其发酵液与病原菌孢子囊悬浮液等体积混合培养测定其对孢子囊的抑制作用,离体叶片接种法检测该菌株及其发酵液对霜霉病的防治效果。【结果】分离获得了90株细菌和110株真菌,分别归属于8个细菌属和14个真菌属,且相同地区不同葡萄品种葡萄生单轴霉菌围的细菌和真菌在同年处于同一分支。假单胞菌属(Pseudomonas)和枝孢属(Cladosporium)稳定存在于各地区不同品种葡萄霜霉病叶上葡萄生单轴霉菌围;在两年间稳定存在的菌株占比多数在80.0%以上且均具有较高的生防作用;其中,广泛分布的6株枝顶孢属(Acremonium)真菌对葡萄霜霉病的防治效果均较好,最高可达100.0%;防治效果较高的11个菌株的无菌发酵液中,黑曲霉(Aspergillusniger) NX2F、苋楔孢黑粉菌(Thecaphora amaranthi) BJ1G和匍枝根霉(Rhizopus stolonifer) BM1L的无菌发酵液防治效果均为100.0%。【结论】葡萄生单轴霉菌围的可培养细菌和真菌群落主要受地区因素影响,有较高的稳定性和生防作用,揭示了枝顶孢属真菌在我国葡萄主要产区葡萄生单轴霉菌围附生的普遍性,为葡萄霜霉病的防治提供了丰富和宝贵的资源。  相似文献

The inheritance of resistance to downy mildew disease and the defense-related enzymes β-1,3-glucanase and peroxidase was studied in crosses of pearl millet using a generation-mean analysis. The study material comprised six generations (susceptible and resistant parents, F1, F2, BC1 and BC2) in three crosses. Seedlings from these generations were inoculated with the downy mildew pathogen Sclerospora graminicola and disease incidence was recorded. Analysis of constitutive levels of β-1,3-glucanase and peroxidase in the seedlings of different generations indicated that the resistant populations showed higher enzyme activities, while lower activities of the enzymes were recorded in the susceptible populations. In the generation-mean analysis, the significance of scaling tests revealed the existence of non-allelic interactions in the inheritance of resistance to downy mildew as well as with the enzymes. Among the gene effects, both additive and dominant effects were significant. All the non-allelic interaction effects were significant in the crosses. Studies on the isozyme patterns of the enzymes substantiated the results of the disease-incidence experiments in most of the generations. The results indicated that the inheritance of downy mildew disease resistance and the expression of β-1,3-glucanase and peroxidase in pearl millet is not only under the control of additive and dominant genes but are also governed by complex non-allelic interactions. Arachidonic acid (AA) induces hypersensitive response (HR) on coleoptile/root regions of two-day-old pearl millet seedlings. The response is comparable to the HR induced by the downy mildew pathogen, Sclerospora graminicola. A time gap in the appearance of cell necrosis among genotypes of pearl millet was related to the degree of resistance to downy mildew. Based on the time required for the development of necrotic spots induced by AA, the pearl millet genotypes were categorised as highly resistant/resistant (HR in 3–6 h), susceptible (HR in 7–12 h) and highly susceptible (HR in 13 h and above). The percentage disease incidence in each genotype was compared with the time required for the development of AA-induced HR. The appearance of hypersensitive cell necrosis was rapid in genotypes having high resistance to downy mildew and was slow in genotypes with high susceptibility. This simple method of screening various pearl millet genotypes in the absence of the pathogen aids in identifying the downy mildew resistant/susceptible host cultivars without the risk of introducing the virulent race of the pathogen.  相似文献
