喙核桃(Annamocarya sinensis)为国家二级重点保护野生植物和极小种群野生植物。阐明广西喙核桃野生种群遗传多样性和遗传变异,可为广西喙核桃遗传多样性保护及管理策略制定提供指导。该研究筛选出6对稳定性和多态性良好的SSR引物,对广西4个喙核桃野生种群(共47个个体)的遗传多样性和遗传变异进行了分析。结果表明,4个喙核桃野生种群的观测等位基因数平均为2.125,有效等位基因数平均为1.581,Shannon信息指数平均为0.477,观测杂合度平均为0.310,期望杂合度平均为0.286,等位基因丰富度平均为1.677,广西喙核桃的遗传多样性水平较低。群体间遗传呈中度分化(0.120),种群间遗传变异占20%,种群内遗传变异占80%。Structure和UPGMA分析表明,47份样品大致分为3类,样品间彼此相互混杂,遗传背景丰富。桂林市永福县种群的群体遗传多样性最高(He=0.339,I=0.598),应把该种群作为重点保护单元。 相似文献

麻竹EST-SSR标记开发及其对慈竹变异类型的分析研究 总被引:1,自引:0,他引:1
利用麻竹(Dendrocalamus latiflorus)已有的EST序列,开发了一批EST-SSR分子标记,并用于慈竹(Bambusa emeiensis)栽培变异类型的多态性研究。结果表明,在麻竹9574个EST中找到了331个EST,含有381个SSR位点,SSR出现的频率为3.98%。EST-SSR的重复类型共有59个,其中2个核苷酸的重复最多,占63.8%,其次是3个核苷酸的重复序列。根据含有SSR的EST序列设计出了44对引物,对慈竹及其变异类型进行了扩增效率、多态性及通用性检测,其中37对引物能够扩增出稳定且清晰的条带,且扩增具有多态性,扩增片段长度主要集中在100~1000 bp,引物有效率达84.1%。遗传相似性分析结果显示,所检测样品间遗传距离为0.263~0.840,平均为0.552,其中‘黑笋慈竹’与‘琴丝慈竹’‘、蛇头慈竹’与‘龙头慈竹’之间的遗传距离较近,而‘罗汉慈竹’与其他样品的遗传距离最远。 相似文献

为揭示紫花风铃木(Handroanthusimpetiginosus)的群体遗传变异特征,对广东省6市12个群体72份种质材料进行遗传多样性和群体遗传结构分析。结果表明,9对引物共扩增出123个等位基因位点,引物的平均多态信息量为0.754,具有较高的多态性。12个群体均具有较高的遗传多样性,群体间平均有效等位基因数为3.272个,平均Shannon指数为1.159。AMOVA分析表明群体间遗传分化程度相对较低,群体内遗传分化程度较高。群体的总体遗传分化系数为0.077,处于中等程度。基于Structure分析、主坐标分析和NJ聚类分析均可将12个群体分为2大类群,分组结果具有一定相似性,表明供试紫花风铃木群体遗传结构较为简单。这为紫花风铃木优良种质资源的利用、遗传变异和科学育种提供了理论参考。 相似文献

In this study, inter-simple sequence repeats (ISSR) markers were applied to assess genetic diversity and genetic relationships of 92 cultivars of sacred lotus (Nelumbo nucifera Gaertn.), one of the most famous flowers in China. Our results showed that sacred lotus exhibited a low level of genetic diversity (percentage of polymorphic bands, PPB = 55.8%), which may result from its asexual mode of reproduction and long-term artificial selection. Clustering analyses indicated that these cultivars could be divided into two clades. Most cultivars of Chinese lotus species origin were included in one clade, and one cultivar of American lotus species origin was nested in the other clade. The hybrid cultivars from hybridization between the two subspecies were interspersed in these two clades. Seven cultivars native to Thailand formed a distinct subclade among the cultivars of Chinese lotus species origin. Genetic differentiation between two subspecies, and between cultivars from Thailand and other cultivars could be attributed to geographic isolation. The monophyly of three cultivars of Sanshui Winter Lotus and their closest relationships to Chinese lotus species origin suggests that they might have a common origin and may consist completely or mainly of genetic material from N. nucifera subsp. nucifera. 相似文献

Summary Genetic variation in resistance to 16 species of herbivorous insects was studied in 18 clones of Solidago altissima growing in an old field near Ithaca, New York, USA. Resistance to each insect, defined as the abundance of a species attacking a particular host genotype relative to other genotypes, was measured in both the natural stand and in two experimental gardens. The heritability of resistance was estimated by parent-offspring regression and sibcorrelation. The primary result was that clones differed in resistance to 15 of 16 insect species. The resistance of genotypes to these insect species remained relatively constant over the four years of the study. However, for only 10 of these resistances were the heritability estimates significantly different from zero. Thus the common assumption of plant-insect studies — that phenotypic variation in insect abundance is closely correlated with underlying genetic variation — is only conditionally true. There is heritable variation in resistance to many insects, but not all. The insects for which we observed heritable variation in plant resistance represent five different orders and several functional groups, including leaf chewers, phloem and xylem feeders, and gall formers. There was no apparent pattern between the degree of heritability of plant resistance and the destructiveness, feeding method, breadth of host range, or taxonomic group of the insects. The lack of marked heritable variation in resistance to some insects may be the result of (a) reduced variation caused by strong selection during prolonged or repeated insect outbreaks, and (b) genotype-environment interactions that obscure differences among genotypes. 相似文献

Genetic variation in 55 accessions of Cynodon dactylon was estimated using inter-simple sequence repeat (ISSR) markers. The plant materials used in this study originated from 17 countries. A total of 236 ISSR fragments were generated with 14 primers. Fragment sizes ranged from 200 to 3000 bp. All scorable bands were polymorphic in nature and none of the primers used produced monomorphic bands, indicating a high level of genetic variation in this grass. The accessions were found to be clustered into eight major groups through the unweighted pair-group method with arithmetic averages. Genetic similarity coefficients (GSC) among the 55 accessions ranged from 0.52 to 0.95. The results clearly indicate that a high level of variation exists in Cynodon accessions. This study shows that the ISSR technique is a reliable tool for differentiating Cynodon accessions and for determining the genetic relationships among them. 相似文献

ep=0.054), but varied across populations in a clinal fashion such that genetic variation decreased with increasing latitude.

Russian apple R12740-7A is the designation for an accession grown from seed collected in Russia, which was found to be highly resistant to apple scab. The resistance has historically been attributed to a naturally pyramided complex involving three major genes: one race-nonspecific gene, Vr, conditioning resistance to all known races, plus two race-specific genes. The race-nonspecific gene was identified as an independently segregating gene by Dayton and Williams (1968) and is referred to in this paper as Vr-DW. The first researchers to study the scab resistance gene complex in Russian apple never described the phenotype conditioned by the race-nonspecific gene. Later, Aldwinckle et al. (1976) associated the name Vr with a scab resistance gene conditioning distinctive stellate necrotic reactions, which we refer to as Vr-A in order to distinguish it from Vr-DW. We show that the segregation ratios in progenies from the scab differential hosts 2 and 4 that are derived from Russian apple, crossed with susceptible cultivars were consistent with a single gene conditioning resistance in each host. The genes have been named Vh2 and Vh4, respectively. Resistant segregants from host 2 showed stellate necrotic reactions, while those from host 4 showed hypersensitive reactions. Both the phenotypes and the genetic maps for the genes in the respective hosts were very similar to those of the genes previously named Vr-A and Vx, respectively, in an F1 family of Russian apple. We showed that race 2 of V. inaequalis isolated from host 2 was able to infect resistant descendants of the non-differential accession PRI 442-23 as well as host 2. The descendants of PRI 442-23 were expected to carry the race-nonspecific Vr-DW gene, but in fact carry Vr-A. We conclude that the Vh2 gene in host 2 and Vr-A are the same, and that the Vh4 gene in host 4 and Vx are the same. However, a major finding of this study is that the latter gene mapped to linkage group 2 of apple instead of linkage group 10 as suggested from previous research. With the two race-specific genes from Russian apple defined now, we discuss the nature of the race-nonspecific Vr-DW gene in this accession. We also report the identification of a new scab resistance gene, VT57, from either Golden Delicious or Red Dougherty, which conditions chlorotic resistance reactions and is linked to Vh2. 相似文献

成团泛菌(Pantoea agglomerans)YS19和菠萝泛菌(Pantoea ananatis)YJ76是从水稻(Oryza sativa)“越富”品种中分离得到的2株优势内生固氮细菌。利用YS19和YJ76共培养方式模拟菌体栖息微环境,探索潜在的种间生物效应,发现适量的YJ76能够促进YS19共质体的形成并且YJ76细胞可以参与到YS19的共质体结构中,YS19?YJ76接种量比例为10?3时促进效果最为显著。相比之下,当使用吲哚产量显著下降的YJ76突变株Δzwf与YS19共培养时,上述促进效应均显著减弱。与单独培养相比,共培养显著提高了两个菌株对重金属Zn2+、四环素、紫外线以及干燥的耐受能力,YS19分别提高了63.9%、56.0%、57.4%、72.0%,YJ76分别提高了147.9%、172.7%、266.3%、348.6%。结果表明水稻的两种主要内生菌YS19和YJ76在共栖息时表现为强烈的协同作用,而YJ76产生的吲哚信号分子在这个过程中起到重要调节作用。 相似文献

A trypsin inhibitor locus (Tri) has been mapped close to Vc-2 on Pisum (pea) linkage group 5 using recombinant inbred lines derived from crosses of genotypes showing qualitative variation in seed trypsin inhibitors. F2 seed populations derived from crosses between lines showing qualitative variation in trypsin inhibitors as well as quantitative variation in inhibitor activity showed an association between the segregation of the structural variation and relative activity levels. Clones complementary to Pisum trypsin inhibitor mRNA were used in hybridization analyses which showed that the segregation of protein polymorphisms reflected directly the segregation of polymorphisms associated with the structural genes. 相似文献

Allozyme variation in viviparid snails from the genus Mekongia in Thailand were examined across the different species, subspecies and geographical locations (river drainage systems). Using horizontal starch gel electrophoresis, 11 presumed allozyme loci (eight polymorphic) from eight enzyme systems were screened. Heterozygosity was moderately low (Hexp = 0.000–0.109, mean = 0.037). One population of Mekongia pongensis (Nong Khai) was monomorphic at all 11 examined loci in contrast to the other two populations, suggesting bottleneck within this population. Populations were more differentiated in the Mekongia sphaericula complex (FST = 0.587) than in either the Mekongia swainsoni complex (FST = 0.161) or M. pongensis species (FST = 0.073). Mekongia sphaericula sphaericula and Mekongia sphaericula extensa exhibited fixed allele differences at two loci, a high genetic distance (D = 0.265–0.300) and a potential polyphyletic relationship, suggesting two distinct lineages (species). 相似文献

Vitex rotundifolia L.f. is a woody perennial and has sexual and asexual modes of reproduction. Allozyme study was conducted on 550 plants in
13 Korean populations. The levels of genetic variability and divergence within and among populations, respectively, are considerably
lower and higher than the mean values for woody plants with similar life history tralts. Mean percentage of polymorphic loci

(genotypes occurring in more than 75% of the populations) were detected. The mean number of multilocus genotypes per population

loci. The abundance of ramets of these genets is responsible for the low levels of expected heterozygosity within populations.

Ribes nigrum germplasm was screened for random amplified polymorphic DNA (RAPD) markers. Fiftyfour markers were identified which generated individual fingerprints for each of 21 cultivars. Genetic diversity of 23 populations of Carthamus tinctorius L. and two populations of Carthamus lanatus L. in China was investigated using Sequence-related Amplified Polymorphism (SRAP). All populations could be uniquely distinguished by 30 primer combinations with 483 bands and 274 polymorphic bands which generated 57% of polymorphic ratio. Unweighed pair-group method of with arithmetical averages (UPGMA) cluster analysis enabled construction of a dendrogram for estimating genetic distances among different populations. The extreme variation was observed when No. 4 cultivated and No. 13 wild population of C. lanatus were grouped at GS = 0.58, and separated from 23 populations of C. tinctorius at GS = 0.10. The result suggested that the cultivated and wild populations of C. lanatus had close relationship with each other and far relationship with C. tinctorius. Dendrogram also revealed a large genetic variation in 23 C. tinctorius populations; different primer combinations allowed them distinctly distinguished one from others with relatively low genetic similarity. Furthermore, five typical representative fragments in C. lanatus were obtained by four most informative primer combinations, which provided a possibility to distinguish C. lanatus from the C. tinctorius evidently. 相似文献

为选择红锥(Castanopsis hystrix)优良家系,对2–11年生红锥家系的生长性状和遗传参数进行了研究分析。结果表明,家系间在树高、胸径和单株材积上的差异达显著或极显著水平,家系的树高、胸径和单株材积受中等或中等以上遗传控制,具有很强的家系和家系内单株选择潜力;家系的单株材积变异幅度较大,胸径居中,树高的较小,显示出红锥家系间存在着丰富的变异;家系和单株遗传力随林龄的增大逐渐变小,到7–9年生时逐渐稳定。年度生长性状的相关分析表明,树高、胸径和单株材积的遗传相关系数随林龄逐渐增大,到7年生时最大,分别达到0.9602、0.9340和0.9849,之后趋于稳定,因此,7年生可作为早期选择的适合年龄。结合早期选择及形质指标,11年生时最终选择出优良家系10个,其平均树高、胸径和单株材积分别为13.95 m、14.34 cm和0.1229 m3,平均遗传增益分别为12.24%、18.69%和51.59%,其通直度、圆满度、分枝角度、最大分枝、冠幅、枝下高等形质指标也提高7.71%~12.94%。 相似文献

为筛选苗期表现优良的山鸡椒[Litsea cubeba(Lour.)Pers.]种源/家系,对其13个种源55个家系的苗高(h)、地径(bd)性状进行遗传变异分析。结果表明,山鸡椒1年生苗木生长期可分为生长初期、速生期和完全木质化期,苗高、地径生长不同步。山鸡椒不同种源、家系间的苗高、地径生长量均存在极显著差异(P0.01)。利用综合选择指数法建立综合选择式为I=0.007Xh+0.74Xbd,建阳2号、富阳1号、遂昌8号、遂昌13号和岳西1号等5个家系优良度为I级,入选率为9.1%。浙江富阳和福建建阳2个种源的苗高、胸径期望遗传增益值最高;建阳2号、遂昌8号、富阳1号、遂昌13号、岳西1号、建阳3号、岳西6号等7个家系的苗高、胸径期望遗传增益最高。因此,依据生长性状综合选择和遗传增益表现,选择出建阳2号、富阳1号、遂昌8号,遂昌13号,岳西1号5个家系以及浙江富阳和福建建阳2个种源为优良家系/种源。 相似文献

Père David’s deer is a severely bottlenecked species but without showing inbreeding depression, making it essential to develop
molecular markers to explore her genetic mechanism of population recovery. In this study, we isolated 18 novel polymorphic
microsatellite loci from a dinucleotide-enriched library. This suit of markers presented 2–3 alleles for each locus and their
observed and expected heterozygosities were 0.057–0.610 and 0.056–0.598, respectively. These new microsatellite loci had an
average of 2.12 alleles and thus contributed to relatively low exclusion probabilities of parentage and paternity testing
(0.768 and 0.921). However, when these loci were examined in combination with previous microsatellite markers, overall probabilities
of parentage and paternity exclusion went up to 0.905 and 0.990, respectively, showing that these 26 microsatellite loci should
be adopted together in future genetic analyses for this highly inbred species. 相似文献

We report in this study, the successful deployment of a double mutant acetolactate synthase gene (ALSdm, containing Pro 197 to Ser and Ser 653 to Asn substitutions) as an efficient in vitro selection marker for the development of transgenic plants in Brassica juncea (oilseed mustard). The ALS enzyme is inhibited by two categories of herbicides, sulfonylureas (e.g. chlorsulfuron) and imidazolinones (e.g. imazethapyr), while the mutant forms are resistant to the same. Three different selection agents (kanamycin, chlorsulfuron and imazethapyr) were tested for in vitro selection efficiency in two B. juncea cultivars, RLM198 and Varuna. For both the cultivars, higher transformation frequencies were obtained using chlorsulfuron (3.8 +/- 0.6% and 4.6 +/- 0.9% for RLM198 and Varuna, respectively) and imazethapyr (10.2 +/- 0.7% for RLM198 and 7.8 +/- 1.2% for Varuna) as compared to that obtained on kanamycin (3.1 +/- 0.2% and 2.8 +/- 0.5% for RLM198 and Varuna, respectively). Additionally, transformation frequencies were higher on imazethapyr than on chlorsulfuron for both the cultivars indicating that imidazolinones are better selective agents than sulfonylureas for the selection of mustard transgenics. 相似文献

Garrulax morrisonianus, an endemic avian species of Taiwan, inhabits evergreen forests at high elevations of 2,000–3,952 m. In this study, we developed
15 microsatellite primer pairs for genetic study. These markers were screened for 52 samples collected from wild populations
of different geographical regions. The number of alleles ranged from 4 to 13. The expected (H
E) and observed (H
O) heterozygosities were 0.430–0.725 and 0.000–0.500, respectively. All loci were significantly deviated from Hardy-Weinberg
equilibrium due to the heterozygote deficiency.
